    Cruciferous vegetable consumption correlates with reduced risk of cancer. This chemopreventative activity may involve glucosinolates and their hydrolysis products. Glucosinolate-derived isothiocyanates have been studied for their toxicity and chemopreventative properties, but other hydrolysis products (epithionitriles and nitriles) have not been thoroughly examined. We report that these hydrolysis products differ in their cytotoxicity to human cells, with toxicity most strongly associated with isothiocyanates rather than epithionitriles and nitriles. We explored mechanisms of this differential cytotoxicity by examining the role of oxidative metabolism, oxidative stress, mitochondrial permeability, reduced glutathione levels, cell cycle arrest and apoptosis. 2-Propenylisothiocyanate and 3-butenylisothiocyanate both inhibited cytochome P450 1A (CYP1A) enzyme activity in CYP expressing MCL-5 cells at high cytotoxic doses. Incubation of MCL-5 cells with non-cytotoxic doses of 2-propenylisothiocyanate for 24h resulted in a dose-dependent inhibition of ethoxyresorufin O-deethylase, yet failed to affect CYP1A1 mRNA expression indicating interference with enzyme activity rather than inhibition of transcription. Increased reactive oxygen species (ROS) production was observed only for 2-propenylisothiocyanate treatment. 2-Propenylisothiocyanate treatment lowered reduced glutathione levels whereas no changes were noted with 3,4-epithiobutylnitrile. Cell cycle analysis showed that 2-propenylisothiocyanate induced a G2/M block whereas other hydrolysis products showed only marginal effects. We found that 2-propenylisothiocyanate and 3-butenylisothiocyanate induced cell death predominantly via necrosis whereas, 3,4-epithiobutylnitrile promoted both necrosis and apoptosis. Thus the activity of glucosinolate hydrolysis products includes cytotoxicity that is compound-class specific and may contribute to their putative chemoprotection properties.

    Peat moss (PM) is the most widely used growing substrate for the pot culture. Due to diminishing availability and increasing price of PM, researchers are looking for viable alternatives for peat as a growth media component for potted plants. A pot study was conducted with a view to investigate the possibility of using spent mushroom waste (SMW) for Kai-lan (Brassica oleracea var. Alboglabra) production replacing peat moss (PM) in growth media. The treatments evaluated were 100% PM (control), 100% SMW, and mixtures of SMW and PM in different ratios like 1 : 1, 1 : 2, and 2 : 1 (v/v) with/without NPK amendment. The experiment was arranged in a completely randomized design with five replications per treatment. Chemical properties like pH and salinity level (EC) of SMW were within the acceptable range of crop production but, nutrient content, especially nitrogen content was not enough to provide sufficient nutrition to plant for normal growth. Only PM (100%) and SMW and PM mixture in 1 : 1 ratio with NPK amendment performed equally in terms of Kai-lan growth. This study confirms the feasibility of replacing PM by SMW up to a maximum of 50% in the growth media and suggests that NPK supplementation from inorganic sources is to ensure a higher productivity of Kai-lan.

    Although Ca transport in plants is highly complex, the overexpression of vacuolar Ca(2+) transporters in crops is a promising new technology to improve dietary Ca supplies through biofortification. Here, we sought to identify novel targets for increasing plant Ca accumulation using genetical and comparative genomics. Expression quantitative trait locus (eQTL) mapping to 1895 cis- and 8015 trans-loci were identified in shoots of an inbred mapping population of Brassica rapa (IMB211 × R500); 23 cis- and 948 trans-eQTLs responded specifically to altered Ca supply. eQTLs were screened for functional significance using a large database of shoot Ca concentration phenotypes of Arabidopsis thaliana. From 31 Arabidopsis gene identifiers tagged to robust shoot Ca concentration phenotypes, 21 mapped to 27 B. rapa eQTLs, including orthologs of the Ca(2+) transporters At-CAX1 and At-ACA8. Two of three independent missense mutants of BraA.cax1a, isolated previously by targeting induced local lesions in genomes, have allele-specific shoot Ca concentration phenotypes compared with their segregating wild types. BraA.CAX1a is a promising target for altering the Ca composition of Brassica, consistent with prior knowledge from Arabidopsis. We conclude that multiple-environment eQTL analysis of complex crop genomes combined with comparative genomics is a powerful technique for novel gene identification/prioritization.

    "Cauliflower Ears" describes the appearance of swelling of the auricular cartilage resulting in gross distortion of the auricle. The underlying pathology is inflammation of the auricular cartilage. We report a case of a patient who presented to the ophthalmology clinic with an ocular inflammation, whose subsequent diagnosis of replapsing polychondritis was clinched with the finding of "Cauliflower Ears". This case emphasizes the importance of an appropriate general examination of ophthalmic patients for early diagnosis and prompt institution of medical treatment, which may prevent life-threatening complications to the patient.

    A developed colorimetric pH sensor film based on edible materials for real-time monitoring of food freshness is described. The mixed natural dyes from edible plants Clitoria sp and Brassica sp were extracted and incorporated into ι-carrageenan film as a colorimetric pH sensor film for monitoring food spoilage and its freshness. The color changes of the developed colorimetric sensor film were measured with chromametry and UV-vis spectroscopy, respectively. Experimental results show that colorimetric pH sensor film demonstrated statistically significant differences (p < 0.05) between CIE-L*a*b* coordinates color system indicated that the developed colorimetric sensor film was able to give a gradual change in color over a wide pH range. The color of the colorimetric sensor film also changes discretely and linearly with factors that contribute to food spoilage using shrimp and durian samples. Moreover, the developed colorimetric pH sensor film has the potential to be used as a safe, non-destructive testing and also a flexibly visual method for direct assessment of food freshness indicator during storage.

    Ammonium-enriched skim latex serum - used for absorption of contaminating ammonia gas - when composted with other rubber tree wastes, is promising as a good compost. The objective of this research was to utilize ammonium-enriched skim latex serum (S) as a raw composting ingredient after being combined with para sawdust (W1) and para rubber leaves (W2). Several ratios of S, W1 and W2 were experimented in a 15L composting vessel to determine the most effective compost. The best ratio was found to be 3:1:3 by weight at 12-day retention. The modified 30 L composting reactor employed with the derived optimum mixing conditions yielded N, P and K of 2.40, 1.51 and 14.84 %w/w. The growth of Brassica alboglabra after application of this compost combined with a chemical fertilizer generated the highest fresh weight (4.48 g/plant). Thus, compost from these wastes could be used as a fertilizer and logically should contribute to cost saving of waste disposal.

    The effects of zeolite and zinc foliar applications on the biochemical characteristics of canola cultivars under different moisture regimes were investigated in a study conducted during the 2010 and 2011 growing seasons. The study was completed using a factorial split-plot experiment based on randomized complete block design (RCBD) with three replications at the Seed and Plant Improvement Institute (SPII), Karaj, Iran. The treatments were: irrigation (I): complete (I1) and restricted (I2); zeolite (Z): 0 (Z1) and 15 ton ha-1 (Z2) and Zn: 0, 0.1 and 0.2% concentrations of zinc sulfate (Zn1, Zn2, and Zn3) at the pod formation stage. These treatments were applied during the pod formation stage to the Licord, RGS003 and Opera cultivars. This study showed that although applying Z and Zn had positive effects on the quality of canola, the highest performance and the best results were obtained using a combination of Z and Zn. The combined application of Z and Zn decreased the proline and carbohydrate contents to 44.35 and 34.42%, respectively. Therefore, with the low cost of natural Z and moderate Zn intake, these treatments can be used to enhance the performance of canola, especially in regions frequently subjected to water stress.

    Resistance of diamondback moth (DBM), Plutella xylostela (L), to coventional pesticides and concerns about environmental quality have lead to increased worldwide efforts to develop viable biocontrol methods for DBM. The success of using parasitoids, especially larval parasitoids like Diadegma, Cotesia and Microplitis species for controlling DBM have been reported in several countries. These larval parasitoids of DBM are commonly found in the field. Diadegma semiclausum (=eucerophaga) Hellen is the major parasitoid of DBM in Europe and Asia, while Diadegma insulare (Cresson) are predominates in the Americas. To date, the bacterium, Bacillus thuringiensis Berliner subspecies kurstaki, has been the most widely used DBM pathogen. Although it is environment friendly pesticides, its effectiveness always vary with weather, field location and frequency of application per season. These are the main factors that contribute to the slow acceptance of B.thuringiensis by the cabbage growers worldwide. Because of this many studies have been done to improve its efficacay and persistence in the field. This paper provides an overview of the current status of these agents for use in controlling DBM and suggest research that is needed to improve the usefulness of these biocontrol agents and to maximize their impact on the DBM management in the future.
    Ketahanan rama-rama belakang-intan (diamondback moth), Plutella xylostella (L.), kepada semua racun-racun perosak yang biasa digunakan untuk mengawalnya dan keprihatinan terhadap kualiti alam sekitar telah menyebabkan bertambahnya usaha mencari beberapa kaedah kawalan biologi rama-rama ini di seluruh dunia. Kejayaan menggunakan parasitoids, terutamanya parasitoid larva seperti spesies Diadegma, Cotesia dan Microplitis bagi kawalan serangga ini telah dilaporkan oleh beberapa negara. Parasitoid-parasitoid larva ini mudah didapati di lapangan. Diadegma semiclausum (=eucerophaga) Hellen adalah merupakan parasitoid utama di Eropah dan Asia, sementara Diadegma insulare (Cresson) predominen di benua America. Sehingga kini, bakteria Bacillus thuringiensis Berliner subspesies kurstaki, adalah sejenis patogen serangga yang digunakan secara meluas untuk mengawal rama-rama tersebut. Walaupun B. thuringiensis adalah sejenis racun makhluk perosak yang tidak mencemari alam sekitar, tahap keberkesanan tindakannya adalah kerap berubah-ubah mengikut perubahan cuaca, lokasi lapangan dan kekerapan penggunaannya. Ini adalah merupakan faktor-faktor penting yang menyebabkan kelambatan penerimaannya oleh penanam-penanam kobis di merata dunia. Oleh sebab inilah banyak kajian telah dan sedang dijalankan bagi memperbaiki tahap keberkesanan dan persistentnya di lapangan. Kertas ini disedia bagi memperihalkan status penggunaan kedua-dua jenis agen kawalan biologi disamping mengesyurkan penyelidikan yang perlu dibuat untuk mempertingkatkan kegunaan dan memaksimumkan dampaknya dalam pengurusan rama-rama ini masa akan datang.

    Kajian yang dijalankan adalah berkaitan dengan penentuan model yang sesuai serta analisis data penyerapan logam berat oleh sayuran berdaun yang terpilih iaitu kangkung (Ipomea aquatica), sawi bunga (Brassica chinensis var parachinensis), bayam (Amaranthus oleraceus L) dan sawi putih (Brassica chinensis L.). Kajian ini bertujuan untuk menentukan dan membandingkan kandungan serta corak pengambilan logam berat yang diserap oleh sayuran dan juga bahagian-bahagiannya yang meliputi daun, batang dan akar. Penentuan model yang dibuat bertujuan bagi melihat corak penyerapan logam berat oleh sayuran atau bahagian sayuran tertentu. Logam berat yang dikaji terdiri daripada kadmium , kromium, kuprum, ferum , mangan, plumbum dan zink. Plot serakan digunakan bagi menentukan corak pengambilan logam berat dalam sayuran dan bahagian-bahagiannya. Selain itu ujian Kruskal-Wallis digunakan bagi membuat perbandingan median di antara logam berat yang diserap oleh sayuran yang dikaji. Nilai khi-kuasa dua dan juga nilai-p digunakan bagi menentukan sama ada sesuatu logam berat yang diserap itu berkait rapat dengan jenis sayuran secara signifikan. Secara umum bolehlah dikatakan bahawa logam Fe, Mn dan Zn adalah dominan dalam semua bahagian sayuran yang dikaji. Selain itu, melalui ujian Kruskal-Wallis didapati penyerapan kesemua logam berat pada setiap bahagian sayuran adalah berbeza secara signifikan. Penyuaian model regresi linear, kuadratik, kubik atau eksponen telah dilakukan terhadap data ini dan didapati kebanyakan data dapat disuaikan dengan baik oleh model kuadratik dan kubik berdasarkan nilai pekali penentuan (R2).

    The present study investigates heavy metal uptake and protein expression by different vegetables collected from various districts of Khyber Pakhtunkhawa province of Pakistan. Statistical analysis of the data showed that maximum concentration of Cd, Cr, Ni, Zn were found in radish and spinach, respectively, collected from Peshawar. Maximum Pb and Mg accumulation were found in cauliflower and pea at Swat followed by coriander at Haripur and minimum Pb uptake was noticed in radish taken from Nowshehra. Highest Cu uptake was detected in spinach at Nowshehra. Data regarding Cd, Cr, Pb, Cu, Ni, Mg and Zn concentration in water samples gathered from different sites of KPK indicated that maximum concentration of Cd was observed in Swat. Maximum Cr and Cu concentration were measured in water samples from Peshawar while maximum concentration of Pb and Ni were detected in water samples from Haripur. In case of soil samples, maximum Cd, Cr, Mg and Zn uptake was observed in soil sample at Nowshehra. Maximum Pb and Ni concentration was found in soil samples collected from Peshawar. Cu concentration was observed to be the highest in soil at Swat. Protein profile of different vegetables i.e. cauliflower, radish, carrot, turnip, pea, spinach, coriander and garlic sampled across five different sites showed that uptake of Cd, Cr, Pb, Cu, Ni, Mg and Zn by these vegetables caused the expression of numerous polypeptides.

    The study was conducted to determine the best model suitable for the determination of ferrum uptake in Brassica chinensis (flowering white cabbage). A nonlinear regression model was selected to determine the amount of ferrum absorbed by each part of the Brassica chinensis plant namely the leaves, stems and roots. The Levenberg-Marquardt method was used to perform the nonlinear least square fit. This method employs information on the gradients and hence requires specification of the partial derivatives. A suitable model was obtained from the exponential regression model. The polynomial model was found to be appropriate for leaves, the mono-exponential model was suitable for stems and the simple exponential model for roots. The residual plots and the normal probability plots from each of the models indicated no substantial diagnostic problems, so it can be concluded that the polynomial and exponential regression models provide adequate fit to determine data on heavy metal uptake by the flowering white cabbage.

    Superhydrophilic graphene oxide/electrospun cellulose nanofibre (GO/CNF) was synthesized, characterized and successfully used in a solid-phase membrane tip adsorption (SPMTA) as an adsorbent towards a simultaneous analysis of polar organophosphorus pesticides (OPPs) in several food and water samples. Separation, determination and quantification were achieved prior to ultra-performance liquid chromatography coupled with ultraviolet detector. The influence of several parameters such as sample pH, adsorption time, adsorbent dosage and initial concentration were investigated. SPMTA was linear in the range of 0.05 and 10 mg l-1 under the optimum adsorption conditions (sample pH 12; 5 mg of adsorbent dosage; 15 min of adsorption time) for methyl parathion, ethoprophos, sulfotepp and chlorpyrifos with excellent correlation coefficients of 0.994-0.999. Acceptable precision (RSDs) as achieved for intraday (0.06-5.44%, n = 3) and interday (0.17-7.76%, n = 3) analyses. Low limits of detection (0.01-0.05 mg l-1) and satisfactory consistency in adsorption (71.14-99.95%) were obtained for the spiked OPPs from Sungai Pahang, Tasik Cheras, cabbages and rice samples. The adsorption data were well followed the second-order kinetic model and fits the Freundlich adsorption model. The newly synthesized GO/CNF showed a great adsorbent potential for OPPs analysis.

    Extruded canola meal (ECM) was included in diet of broiler chickens at 0, 10, 20, and 30% (wt/wt) from 1 to 35 days of age. A total of 240 day-old male chicks were assigned in groups of 5 to 48 battery cages in environmentally controlled chambers and diets were replicated with 12 cages/treatment. From d 29 to 35, birds from each dietary group were exposed to either thermoneutral (23 ± 1°C; unheated) or high (36 ± 1°C; heated) temperature conditions. High ambient temperature, irrespective of ECM inclusion, depressed the growth performance of birds. Inclusion of ECM increased feed conversion ratio (FCR) linearly in unheated birds during d 1 to 28 (P < 0.001) and d 29 to 35 (P = 0.001). However, no adverse effects of ECM inclusion were observed on the growth performance of heated birds. The absence of these detrimental effects could be associated with the lack of triiodothyronine (T3) elevation by ECM inclusion in heated birds. In conclusion, ECM can be fed, at least, up to 30%, without any adverse effect on growth performance of broiler chickens raised under chronic high ambient temperature.

    A diverse range of monocot and dicot grains and their by-products are commonly used in the animal feed industry. They all come with complex and variable cell wall structures which in turn contribute significant fiber to the complete feed. The cell wall is a highly interconnected matrix of various polysaccharides, proteins and lignin and, as such, requires a collaborative effort of different enzymes for its degradation. In this regard, we investigated the potential of a commercial multicomponent carbohydrase product from a wild type fermentation of Trichoderma reesei (T. reesei) (RONOZYME® MultiGrain) in degrading cell wall components of wheat, barley, rye, de-oiled rice bran, sunflower, rapeseed and cassava. A total of thirty-one different enzyme proteins were identified in the T. Reesei carbohydrase product using liquid chromatography with tandem mass spectrometry LC-MS/MS including glycosyl hydrolases and carbohydrate esterases. As measured by in vitro incubations and non-starch polysaccharide component analysis, and visualization by immunocytochemistry and confocal microscopy imaging of immuno-labeled samples with confocal microscopy, the carbohydrase product effectively solubilized cellulolytic and hemicellulolytic polysaccharides present in the cell walls of all the feed ingredients evaluated. The T. reesei fermentation also decreased viscosity of arabinoxylan, xyloglucan, galactomannan and β-glucan substrates. Combination of several debranching enzymes including arabinofuranosidase, xylosidase, α-galactosidase, acetyl xylan esterase, and 4-O-methyl-glucuronoyl methylesterase with both GH10 and GH11 xylanases in the carbohydrase product resulted in effective hydrolyzation of heavily branched glucuronoarabinoxylans. The different β-glucanases (both endo-β-1,3(4)-glucanase and endo-β-1,3-glucanase), cellulases and a β-glucosidase in the T. reesei fermentation effectively reduced polymerization of both β-glucans and cellulose polysaccharides of viscous cereals grains (wheat, barley, rye and oat). Interestingly, the secretome of T. reesei contained significant amounts of an exceptional direct chain-cutting enzyme from the GH74 family (Cel74A, xyloglucan-specific β-1,4-endoglucanase), that strictly cleaves the xyloglucan backbone at the substituted regions. Here, we demonstrated that the balance of enzymes present in the T. reesei secretome is capable of degrading various cell wall components in both monocot and dicot plant raw material used as animal feed.

    Brassica napus (canola/oilseed rape/rapeseed) is an economically important crop, mostly found in temperate and sub-tropical regions, that is cultivated widely for its edible oil. Major diseases of Brassica crops such as Blackleg, Clubroot, Sclerotinia Stem Rot, Downy Mildew, Alternaria Leaf Spot and White Rust have caused significant yield and economic losses in rapeseed-producing countries worldwide, exacerbated by global climate change, and, if not remedied effectively, will threaten global food security. To gain further insights into the host-pathogen interactions in relation to Brassica diseases, it is critical that we review current knowledge in this area and discuss how omics technologies can offer promising results and help to push boundaries in our understanding of the resistance mechanisms. Omics technologies, such as genomics, proteomics, transcriptomics and metabolomics approaches, allow us to understand the host and pathogen, as well as the interaction between the two species at a deeper level. With these integrated data in multi-omics and systems biology, we are able to breed high-quality disease-resistant Brassica crops in a more holistic, targeted and accurate way.

    The main goal of this publication is an overview of the biotechnological achievements concerning in vitro cultures and transformation of Brassica oleracea var. capitata. Faced with the requirements of the global food market, intensified work on the genetic transformation of economically important plants is carried out in laboratories around the world. The development of efficient procedures for their regeneration and transformation could be a good solution for obtaining, in a shorter time than by traditional methods, plants with desirable traits. Furthermore, conventional breeding methods are insufficient for crop genetic improvement not only because of being time-consuming but also because they are severely limited by sexual incompatibility barriers. This problem has been overcome by genetic engineering, which seems to be a very good technique for cabbage improvement. Despite the huge progress that has been made in the field of plant regeneration and transformation methods, up to now, no routine transformation procedure has been developed in the case of cabbage. This problem stems from the fact that the efficiency of cabbage transformation is closely related to the genotype and some varieties are recalcitrant to transformation. It is obvious that it is not possible to establish one universal regeneration and transformation protocol for all varieties of cabbage. Therefore, it seems fully justified to develop the above-mentioned procedures for individual economically important cultivars. Despite the obstacles of cabbage transformation in laboratories of many countries, especially those where this vegetable is extremely popular (e.g., China, India, Korea, Malaysia, Pakistan), such attempts are made. This article reviews the achievements in the field of tissue culture and cabbage transformation from the last two decades.

    Bok choy (Brassica chinensis L.) is a temperate vegetable grown in the cool highland areas of Malaysia. In June 2010, vegetable growing areas of the Cameron Highlands, located in Pahang State, Malaysia, were surveyed for the prevalence of anthracnose disease caused by Colletotrichum species. Diseased samples were randomly collected from 12 infested fields. Anthracnose incidence on bok choy varied from 8 to 36% in different nursery fields. Disease symptoms initially appeared as small water-soaked spots scattered on the leaf petioles of young plants. As these spots increased in size, they developed irregular round spots that turned to sunken grayish brown lesions surrounded by brownish borders. When the lesions were numerous, leaves collapsed. Pale buff to salmon conidial mass and acervuli were observed on well-developed lesions. The acervuli diameter varied in size from 198 to 486 μm, averaging 278.5 μm. Morphological and cultural characteristics of the fungus were examined on potato dextrose agar incubated for 7 days at 25 ± 2°C under constant fluorescent light. Vegetative mycelia were hyaline, septate, branched, and 2 to 7 μm in diameter. The color of the fungal colonies was grayish brown. Conidia were hyaline, aseptate, falcate, apices acute, and 21.8 to 28.5 × 2.6 to 3.4 mm. Setae were pale brown to dark brown, 75 to 155 μm long, base cylindrical, and tapering towards the acute tip. Appressoria were solitary or in dense groups, light to dark brown, entire edge to lobed, roundish to clavate, 6.5 to 14 × 5.8 to 8.6 μm, averaging 9.2 × 6.8 μm, and had a L/W ratio of 1.35. Based on the keys outlined by Mordue 1971 (2) and Sutton 1980 (3), the characteristics of this fungus corresponded to Colletotrichum capsici. Sequence analysis of the ITS-rDNA obtained from the Malaysian strain CCM3 (GenBank Accession No. JQ685746) using primers ITS5 and ITS4 (1) when aligned with deposited sequences from GenBank revealed 99 to 100% sequence identity with C. capsici strains (DQ286158, JQ685754, DQ286156, GQ936210, and GQ369594). A representative strain CCM3 was used for pathogenicity testing. Four non-infected detached leaves of 2-week-old B. chinensis were surface-sterilized and inoculated by placing 10 μl of conidial suspension (106 conidia ml-1) using either the wound/drop or non-wound/drop method, and distilled water was used as a control (1). Leaves were incubated at 25°C, 98% RH. The experiment was repeated twice. Five days after inoculation, typical anthracnose symptoms with acervuli formation appeared on the surface of tissues inoculated with the spore suspension, but not on the water controls. A fungus with the characteristics of C. capsici was recovered from the lesions on the inoculated leaves. Anthracnose caused by C. capsici has been reported on different vegetable crops, but not on bok choy (3).     Soft rot of cabbage (Brassica rapa) occurs sporadically in Malaysia, causing economic damage under the hot and wet Malaysian weather conditions that are suitable for disease development. In June 2011, 27 soft rotting bacteria were isolated from cabbage plants growing in the Cameron Highlands and Johor State in Malaysia where the economic losses exceeded 50% in severely infected fields and greenhouses. Five independent strains were initially identified as Pectobacterium wasabiae based on their inability to grow at 37°C, and elicit hypersensitive reaction (HR) on Nicotiana tabaccum and their ability to utilize raffinose and lactose. These bacterial strains were gram-negative, rod-shaped, N-acetylglucosaminyl transferase, gelatin liquefaction, and OPNG-positive and positive for acid production from D-galactose, lactosemelibiose, raffinose, citrate, and trehalose. All strains were negative for indole production, phosphatase activity, reducing sucrose, and negative for acid production from maltose, sorbitol, inositol, inolin, melezitose, α-methyl-D-glucoside, and D-arabitol. All the strains exhibited pectolytic activity on potato slices. PCR assays were conducted to distinguish P. wasabiae from P. carotovorum subsp. brasiliensis, P. atrosepticum, and other Pectobacterium species using primers Br1f/L1r (2), Eca1f/Eca2r (1), and EXPCCF/EXPCCR, respectively. DNA from strains did not yield the expected amplicon with the Br1f/L1r and Eca1f/Eca2r, whereas a 550-bp amplicon typical of DNA from P. wasabiae was produced with primers EXPCCF/EXPCCR. ITS-RFLP using the restriction enzyme, Rsa I, produced similar patterns for the Malaysian strains and the P. wasabiae type strain (SCRI488), but differentiated it from P. carotovora subsp. carotovora, P. atrosepticum, P. carotovorum subsp. brasiliensis, and Dickeya chrysanthemi type strains. BLAST analysis of the 16S rRNA DNA sequence (GenBank Accession No. KC445633) showed 99% identity to the 16S rRNA of Pw WPP163. Phylogenetic reconstruction using concatenated DNA sequences of mdh and gapA from P. wasabiae Cc6 (KC484657) and other related taxa (4) clustered Malaysian P. wasabiae strains with P. wasabiae SCRI488, readily distinguishing it from other closely related species of Pectobacterium. Pathogenicity assays were conducted on leaves and stems of four mature cabbage plants for each strain (var. oleifera) by injecting 10 μl of a bacterial suspension (108 CFU/ml) into either stems or leaves, and incubating them in a moist chamber at 80 to 90% relative humidity at 30°C. Water-soaked lesions similar to those observed in the fields and greenhouses were observed 72 h after injection and bacteria with similar characteristics were consistently reisolated. Symptoms were not observed on water-inoculated controls. The pathogenicity test was repeated with similar results. P. wasabiae was previously reported to cause soft rot of horseradish in Japan (3).     The Brassica napus secondary embryogenesis system requires no exogenous growth regulator to stimulate embryo development. It is stable embryogenically over a long period of culture and has a distinct pre-embryogenic stage. This system was used to investigate the morphological and cellular changes occurring in the embryogenic tissue compared to non-embryogenic tissue using various microscopy techniques. A unique ultrastructural feature designated the extracellular matrix (ECM) was observed on the surface of pre-embryogenic embryoids but not on the non-embryogenic individuals. The ECM layer was found to be dominant in the pre-embryogenic stage and reduced to fragments during embryo growth and development in mature embryogenic tissue. This is a novel aspect of the phenotype previously unreported in the Brassica system. This structure might be linked to acquisition of embryogenic competence.

    Antioxidants are substances that may protect cells from the damage caused by unstable molecules such as free radicals. Flavonoids are phenolic substances widely found in fruits and vegetables. The previous studies showed that the ingestion of flavonoids reduces the risk of cardiovascular diseases, metabolic disorders, and certain types of cancer. These effects are due to the physiological activity of flavonoids in the reduction of oxidative stress, inhibiting low-density lipoproteins oxidation and platelet aggregation, and acting as vasodilators in blood vessels. Free radicals are constantly generated resulting in extensive damage to tissues leading to various disease conditions such as cancer, Alzheimer's, renal diseases, cardiac abnormalities, etc., Medicinal plants with antioxidant properties play a vital functions in exhibiting beneficial effects and employed as an alternative source of medicine to mitigate the disease associated with oxidative stress. Flavonoids have existed over one billion years and possess wide spectrum of biological activities that might be able to influence processes which are dysregulated in a disease. Quercetin, a plant pigment is a potent antioxidant flavonoid and more specifically a flavonol, found mostly in onions, grapes, berries, cherries, broccoli, and citrus fruits. It is a versatile antioxidant known to possess protective abilities against tissue injury induced by various drug toxicities.
