AIM: To assess the salivary levels of Copper (Cu), Zinc (Zn), Manganese (Mn) and Iron (Fe) obtained from children of different ethnic backgrounds in Shah Alam, Malaysia and investigate the possible relationships with caries.

MATERIALS AND METHODS: One hundred and twenty primary school children were included. They were divided into caries and caries-free groups. Unstimulated whole saliva was collected from each participant using spitting method. The salivary elements were measured using an Atomic Absorption Spectrophotometer. Descriptive statistics, bivariate and Pearson's correlation analysis were performed.

RESULTS: Salivary Cu and Zn levels were significantly higher in children with dental caries compared to those caries-free (p < 0.05). Moreover, these elements had a positive correlation with dental caries (Cu: r=0.698, p<0.001; Zn: r=0.181, p<0.05). No significant variations in Mn and Fe were observed between caries and caries-free group (p>0.05). Additionally, there were significant differences in salivary Zn and Fe among different age groups (p<0.05) and highly significant differences in salivary Cu, Mn and Fe among different ethnic groups (p<0.001). However, all elements exhibited no significant differences between males and females.

CONCLUSION: The salivary Cu and Zn levels showed significant differences between caries and caries-free groups. The findings also revealed significant variations in the levels of salivary Cu, Mn and Fe among different ethnic groups and salivary Zn and Fe among different age groups.

The normally high concentration of zinc in normal prostate gland is significantly reduced in malignant prostate tissues, but its precise role in prostate tumorigenesis remains unclear. The present study investigates the growth and transcriptional responses of LNCaP prostate cancer cells to prolonged high Zn2+ treatment. Restoration of high intracellular Zn2+ to LNCaP cells significantly reduced the cell proliferation rate by 42.2+/-7.4% at the exponential growth phase and the efficiency of colony formation on soft agar by 87.2+/-2.5% at week 5 post-treatment. At least 161 LNCaP cell genes responded to the high intracellular Zn2+, including approximately 10.6% genes that negatively regulate cell growth and approximately 16.1% genes that promote cancer cell proliferation. Inhibition of cell growth was transient as normal proliferation rate and colony formation efficiency were restored later even in the continuous presence of high intracellular Zn2+. RT-qPCR showed constitutively higher expression levels of FBL, CD164 and STEAP1 in LNCaP cells. FBL and CD164 were responsive to the treatment with Zn2+ in PNT2 prostate normal cells and were further overexpressed in the prolonged Zn2+-treated LNCaP cells. These observations suggest that in general high Zn2+ has suppressive effects on prostate cancer cell growth but continuous exposure to an environment of high Zn2+ can lead to the overexpression of cancer promoting genes such as FBL and CD164. This could be the antagonistic mechanism used to overcome the initial cell growth inhibitory effects of high Zn2+. These findings support a potential detrimental role of Zn2+ in prostate cancer.

The distribution of benthic Foraminifera throughout the coastal waters of Taman Negara Pulau Pinang (Penang National Park), Malaysia was studied to assess the impact of various anthropogenic activities, such as fishing, ecotourism and floating cage culture. Samples were obtained at 200 m intervals within the subtidal zone, extending up to 1200 m offshore at Teluk Bahang, Teluk Aling, Teluk Ketapang and Pantai Acheh. The depth within coastal waters ranged between 1.5 m and 10.0 m, with predominantly muddy substrate at most stations. Water quality analysis showed little variation in micronutrient (nitrite, NO2; nitrate, NO3; ammonia, NH4 and orthophosphate, PO4) concentrations between sampling stations. Temperature (29.6±0.48°C), salinity (29.4±0.28 ppt), dissolved oxygen content (5.4±0.95 mg/l) and pH (8.5± 0.13) also showed little fluctuation between stations. A total of nine genera of foraminifera were identified in the study (i.e., Ammonia, Elphidium, Ammobaculites, Bigenerina, Quinqueloculina, Reopax, Globigerina, Textularia and Nonion). The distribution of benthic foraminifera was dominated by opportunistic groups that have a high tolerance to anthropogenic stressors. Ammonia had the highest frequency of occurrence (84.7%), followed by Bigenerina (50%), Ammobaculites (44.2%) and Elphidium (38.9%). The Ammonia-Elphidium Index (AEI) was used to describe the hypoxic condition of benthic communities at all sites. Teluk Bahang had the highest AEI value. The foraminiferal assemblages and distribution in Teluk Bahang, Teluk Aling, Teluk Ketapang and Pantai Acheh showed no correlation with physical or chemical environmental parameters.

An instrumental neutron activation analytical (INAA) technique is used for the determination of thirty elements in five coal samples collected from Kapar power station, imported from Indonesia and Australia. Analyses of the samples are being associated with standards. All irradiations were performed in the nuclear reactor of Malaysia Nuclear Agency (MNA). Samples were counted by Hyper Pure Germanium (HPGe) detector for short period irradiations at MNA, while for the long period irradiations the samples were counted at Universiti Kebangsaan Malaysia (UKM). The concentrations of thirty elements have been determined: The major components are Cl, Ca, Mg, K, Fe, Ti and Na with the mean concentrations in the range between 70±69 ppm- 6100±1639 ppm; and the trace elements are Zr, V, Mn , Sc, Cr, Co, As ,Br ,Rb ,Sb ,Ba , La, Ce, Nd, Sm, Eu, Tb, Yb, Lu, Hf, Th, U and Ta with the mean concentrations in the range between 0.1381±0.0202 - 69.0±2.8 ppm. The results have been compared to the reported data of eight coal samples from the United States and the reported data of Australian bituminous coal.

The rapid growth of industrial and agricultural activities in Malaysia are leading to the impairment of most of the rivers in recent years through realising various trace metals. This leads to toxicity, particularly when the toxic has entered the food chain. Perak River is one of the most dynamic rivers for the Malaysian population. Therefore, in consideration of the safety issue, this study was conducted to assess the concentration of such metals (Cd, Cu, Zn, Fe, and Pb) in the muscles of most widely consumed fish species (Barbonymus schwanenfeldii, Puntius bulum, Puntius daruphani, Hexanematichthys sagor, Channa striatus, Mystacoleucus marginatus, and Devario regina) from different locations of Perak River, Malaysia by employing inductively coupled plasma optical emission spectroscopy (ICP-OES). Among the trace metals, Fe and Cd were found to be the highest (29.33-148.01 μg/g) and lowest (0.16-0.49 μg/g) concentration in all of the studied species, respectively. Although the estimated daily intakes (μg/kg/day) of Cd (0.65-0.85), Fe (79.27-352.00) and Pb (0.95-12.17) were higher than their reference, the total target hazard quotients values suggested that the local residents would not experience any adverse health effects from its consumption. In contrast, the target cancer risk value suggested that all fish species posed a potential cancer risk due to Cd and cumulative cancer risk values, strongly implying that continuous consumption of studied fish species would cause cancer development to its consumers.

The composite nature of Ennore Creek, receiving polluted waters from Buckingham canal, River Kortalaiyar, and the presence of numerous industries rapidly degrade the coastal environment. In this study, multivariate statistical technique was used to assess the nature of pollution and to identify the factors responsible for the enrichment of trace metals in the creek sediments. Forty samples were collected during pre- and post-monsoon periods to evaluate the seasonal variations on the concentration of trace metals in the sediments. The results indicate that not much seasonal variation exists in the concentration of trace metals in the sediments of Ennore creek. Results of the cluster analysis illustrate that the enrichment of trace metals was mainly from anthropogenic sources. Meanwhile, correlation coefficient among the metals reveals that some of the metals (Fe and Al) derived were of natural origin. The complex data matrix of the sediment were interpreted after reduction to three factors and the results illustrate the extent of the influence of anthropogenic activities. The spatial distribution diagrams demonstrate and demarcate the region of enrichment of metals in the sediments of Ennore creek.

Kidney stone samples of the types calcium oxalate, uric acid, and xanthine were analyzed for their elemental contents by neutron activation analysis to study both the elemental correlation and influence of element on stone precipitation processes. Elements, such as Al, Au, Br, Ca, Cl, Co, Cr, Fe, Hg, I, K, Mg, Na, Sb, Se, Sr, and Zn, were determined quantitatively. Calcium oxalate stones contained higher concentration of all the elements analyzed compared to uric acid or xanthine stones. The concentrations of Cl, Fe, K, Na, Sr, and Zn were relatively higher than Au, Co, Cr, and Sb. A positive correlation exists between Ca and Zn, whereas a negative correlation exists between Sr and Ca. Zinc may play an important role in the formation of calcium oxalate stone.

Trace metals are required in many cellular processes in bacteria but also induce toxic effects to cells when present in excess. As such, various forms of adaptive responses towards extracellular trace metal ions are essential for the survival and fitness of bacteria in their environment. A soil Pseudomonas putida, strain S13.1.2 has been isolated from French vineyard soil samples, and shown to confer resistance to copper ions. Further investigation revealed a high capacity to tolerate elevated concentrations of various heavy metals including nickel, cobalt, cadmium, zinc and arsenic. The complete genome analysis was conducted using single-molecule real-time (SMRT) sequencing and the genome consisted in a single chromosome at the size of 6.6 Mb. Presence of operons and gene clusters such as cop, cus, czc, nik, and asc systems were detected and accounted for the observed resistance phenotypes. The unique features in terms of specificity and arrangements of some genetic determinants were also highlighted in the study. Our findings has provided insights into the adaptation of this strain to accumulation and persistence of copper and other heavy metals in vineyard soil environment.

This study aimed to determine the spatial distribution of PM2.5 and PM10 collected in four regions (North, Central, South and East Coast) of Peninsular Malaysia during the southwest monsoon. Concurrent measurements of PM2.5 and PM10 were performed using a high volume sampler (HVS) for 24 h (August to September 2018) collecting a total of 104 samples. All samples were then analysed for water soluble inorganic ions (WSII) using ion chromatography, trace metals using inductively coupled plasma-mass spectroscopy (ICP-MS) and polycyclic aromatic hydrocarbon (PAHs) using gas chromatography-mass spectroscopy (GC-MS). The results showed that the highest average PM2.5 concentration during the sampling campaign was in the North region (33.2 ± 5.3 μg m-3) while for PM10 the highest was in the Central region (38.6 ± 7.70 μg m-3). WSII recorded contributions of 22% for PM2.5 and 20% for PM10 mass, with SO42- the most abundant species with average concentrations of 1.83 ± 0.42 μg m-3 (PM2.5) and 2.19 ± 0.27 μg m-3 (PM10). Using a Positive Matrix Factorization (PMF) model, soil fertilizer (23%) was identified as the major source of PM2.5 while industrial activity (25%) was identified as the major source of PM10. Overall, the studied metals had hazard quotients (HQ) value of <1 indicating a very low risk of non-carcinogenic elements while the highest excess lifetime cancer risk (ELCR) was recorded for Cr VI in the South region with values of 8.4E-06 (PM2.5) and 6.6E-05 (PM10). The incremental lifetime cancer risk (ILCR) calculated from the PAH concentrations was within the acceptable range for all regions.

Wilson's disease (WD), also known as hepatoleticular degeneration (HLD), is a rare autosomal recessive genetic disorder of copper metabolism, which causes copper to accumulate in body tissues. In this study, rats fed with copper-laden diet are used to render the clinical manifestations of WD, and their copper toxicity-induced organ lesions are studied. To investigate metabolic behaviors of 'decoppering' process, penicillamine (PA) was used for treating copper-laden rats as this chelating agent could eliminate excess copper through the urine. To date, there has been limited metabolomics study on WD, while metabolic impacts of copper accumulation and PA administration have yet to be established.

The concentration of nine trace elements were analyzed in the different tissue organs of commonly available crabs (Portunus sanguinolentus, Portunus pelagicus and Scylla serrate) and bivalve (Polymesoda erosa) species collected from the Miri coast, Borneo in order to evaluate the potential health risk by consumption of these aquatic organisms. Among the analyzed organs, metal accumulation was higher in the gill tissues. The essential (Cu and Zn) and non-essential (Pb and Cd) elements showed the highest (i.e. Zn) and lowest concentrations (i.e. Cd) in their tissue organs, respectively. The estimated daily intake and hazard indices of all metals in the muscle indicate that the measured values were below the provisional tolerable daily intake suggested by the joint FAO/WHO Expert Committee on Food Additives. Compared to Malaysian and international seafood guideline values the results obtained from the present study are lower than the permissible limits and safe for consumption.

This study focused on the influence of ultramafic terrains on soil and surface water environmental chemistry in Peninsular Malaysia and in the State of Sabah also in Malaysia. The sampling included 27 soils from four isolated outcrops at Cheroh, Bentong, Bukit Rokan, and Petasih from Peninsular Malaysia and sites near Ranau in Sabah. Water samples were also collected from rivers and subsurface waters interacting with the ultramafic bodies in these study sites. Physico-chemical parameters (including pH, EC, CEC) as well as the concentration of major and trace elements were measured in these soils and waters. Geochemical indices (geoaccumulation index, enrichment factor, and concentration factor) were calculated. Al2O3 and Fe2O3 had relatively high concentrations in the samples. A depletion in MgO, CaO, and Na2O was observed as a result of leaching in tropical climate, and in relation to weathering and pedogenesis processes. Chromium, Ni, and Co were enriched and confirmed by the significant values obtained for Igeo, EF, and CF, which correspond to the extreme levels of contamination for Cr and high to moderate levels of contamination for Ni and Co. The concentrations of Cr, Ni, and Co in surface waters did not reflect the local geochemistry and were within the permissible ranges according to WHO and INWQS standards. Subsurface waters were strongly enriched by these elements and exceeded these standards. The association between Cr and Ni was confirmed by factor analysis. The unexpected enrichment of Cu in an isolated component can be explained by localized mineralization in Sabah.

A study was carried out on the distribution and enrichment of trace elements in the core marine sediments of East Malaysia from three stations at South China Sea and one station each at Sulu Sea and Sulawesi Sea. Five stations of sediment cores were recovered and the vertical concentration profiles of six elements namely Br, Cs, Hf, Rb, Ta, and V were determined using the instrumental neutron activation analysis. The enrichment factor, geoaccumulation index and the modified degree of contamination were used to calculate the anthropogenic and pollution status of the elements in the samples. Except for Cs and Hf, which by the enrichment factor are categorized from minimum enrichment to moderate enrichment in all stations and for V and Rb in Sulu Sea and Sulawesi Sea, which are categorized minimum enrichment, other elements are found to be no enrichment at all stations. The geoaccumulation index of Hf in one station shows moderately polluted and for other elements are unpolluted. However, the modified degree values of all samples are less than 1, suggesting very low contamination of elements found in all the stations.

A study was conducted in an oil palm plantation in Peninsular Malaysia to elucidate the effects of applying Magnesium Rich Synthetic Gypsum (MRSG), a by-product of chemical plant, on the chemical properties of soil, the uptake of heavy metals by the palm trees, the oil quality and its impact on the surrounding environment. The results showed that MRSG application onto soil cropped to oil palm could bring positive impact in terms of soil chemical properties and oil palm production. The quality of the oil was not significantly affected by the continuous MRSG application as shown by the low heavy metals and trace elements of concern content (Cu: 0.062 mg/kg; Fe: 2.10 mg/kg; Mn: 1.93 mg/kg; Pb: 0.006 mg/kg; Zn: 0.103 mg/kg; Cr: 0.354 mg/kg; Ni: 0.037 mg/kg). From the I-geochem index, the soil was found to have values ranging from -3.81 to -1.03 which is considered as uncontaminated. Further, its application did not result in negative impact on the surrounding environment; hence, the quality of the soil and surface water in the plantation and/or the surrounding area remained intact. Phytotoxic elements in the oil palm tissue (As: 0.12 mg/kg; Se: 0.05 mg/kg; Zn: 1.48 mg/kg; Ce: 0.47 mg/kg; La: 0.26 mg/kg; Sr: 3.03 mg/kg) and cytotoxic elements in the oil were below the acceptable limit. Based on the results of the Environmental Monitoring out during the period of the study, it was concluded that application of the by-product of the chemical plant as a source of Mg to enhance soil fertility in the oil palm plantation was considered safe and sustainable. The effects of applying MRSG and Chinese kieserite was almost similar. So, MRSG can be used as a possible source of Mg to replace Chinese kieserite for oil palm production on the Ultisols in Peninsular Malaysia.

This study was conducted to determine the proximate composition and four micronutrients (Cd, Cu, Mn and Zn) of Indian Mackerel (Rastrelliger kanagurta), Eel (Monopterus albus) and Cockle (Anadara granosa). All fish and shellfish were purchased from local fish market in Kuantan city. All samples of each species were mixed and divided into two groups based on random selection. Each group were again divided into 3 sub-groups which were considered as replications. The first group were kept uncooked. The second group were fried in a beaker of 400 mL palm cooking oil capacity at a temperature approximately of 180 degrees C for a 15 min period. Both raw and fried samples were analysed following standard methods to determine protein, lipid, ash, moisture, carbohydrate, Cd, Cu, Mn and Zn contents. Results showed that protein content was higher in Indian mackerel and eel than cockle while overall Cd, Cu, Mn and Zn contents were higher in cockle than Indian mackerel and eel. Therefore, fish is better than shellfish in the nutritional point of view. Fried fish and shellfish had very high fat content. Therefore, frying cannot be recommended to prepare a healthy diet. More research is needed including all cooking methods of fish to know the nutritional changes by each cooking method. Fish contains many important fatty acids and amino acids which might be lost during frying. Therefore, future study should include the effects of different cooking methods on amino acids and fatty acids compositions of fish and shellfish.

N-Acetyl-p-Aminophenol (APAP), also known as acetaminophen, is the most commonly used over-the counter analgesic and antipyretic medication. However, its overdose leads to both liver and kidney damage. APAP-induced toxicity is considered as one of the primary causes of acute liver failure; numerous scientific reports have focused majorly on APAP hepatotoxicity. Alternatively, not many works approach APAP nephrotoxicity focusing on both its mechanisms of action and therapeutic exploration. Moringa oleifera (MO) is pervasive in nature, is reported to possess a surplus amount of nutrients, and is enriched with several bioactive candidates including trace elements that act as curatives for various clinical conditions. In this study, we evaluated the nephro-protective potential of MO leaf extract against APAP nephrotoxicity in male Balb/c mice. A single-dose acute oral toxicity design was implemented in this study. Group 2, 3, 4 and 5 received a toxic dose of APAP (400 mg/kg of bw, i.p) and after an hour, these groups were administered with saline (10 mL/kg), silymarin-positive control (100 mg/kg of bw, i.p), MO leaf extract (100 mg/kg of bw, i.p), and MO leaf extract (200 mg/kg bw, i.p) respectively. Group 1 was administered saline (10 mL/kg) during both the sessions. APAP-treated mice exhibited a significant elevation of serum creatinine, blood urea nitrogen, sodium, potassium and chloride levels. A remarkable depletion of antioxidant enzymes such as SOD, CAT and GSH-Px with elevated MDA levels has been observed in APAP treated kidney tissues. They also exhibited a significant rise in pro-inflammatory cytokines (TNF-α, IL-1β, IL-6) and decreased anti-inflammatory (IL-10) cytokine level in the kidney tissues. Disorganized glomerulus and dilated tubules with inflammatory cell infiltration were clearly observed in the histology of APAP treated mice kidneys. All these pathological changes were reversed in a dose-dependent manner after MO leaf extract treatment. Therefore, MO leaf extract has demonstrated some therapeutic effectiveness against APAP-induced nephrotoxicity through enhancement of the endogenous antioxidant system and a modulatory effect on specific inflammatory cytokines in kidney tissues.

Dental enamel, an avascular, irreparable, outermost and protective layer of the human clinical crown has a potential to withstand the physico-chemical effects and forces. These properties are being regulated by a unique association among elements occurring in the crystallites setup of human dental enamel. Calcium and phosphate are the major components (hydroxyapatite) in addition to some trace elements which have a profound effect on enamel. The current review was planned to determine the aptitude of various trace elements to substitute and their influence on human dental enamel in terms of physical and chemical properties.
