    This study was aimed to identify and optimize the culture conditions for gamma-aminobutyric acid (GABA) production by a lactic acid bacterium strain isolated from mam nem, a fermented fish sauce. Among the six isolates obtained from mam nem, the MN12 had the most potent GABA-producing capability. The strain was then identified to be Pedioccocus pentosaceus by employing MALDI-TOF-MS and phenylalanyl-tRNA synthase sequencing methods. The initial cell density of 5.106 CFU/mL, monosodium glutamate concentration of 60 mM, initial pH of 7, temperature of 45°C and cultivation time of 72 h were found to be the optimal culture conditions for highest production of GABA, reaching 27.9 ± 0.42 mM, by this strain. The cultivation conditions for GABA production by P. pentosaceus MN12 have been successfully optimized, providing a foundation for the development of fermented foods enriched with GABA.

    Psychosocial safety climate (PSC) is a facet-specific climate for psychological health and safety which constitutes an important organizational resource for creating a conducive work environment. The process to regain/restore energy expended at work, known as "recovery," also plays a pivotal role for individuals; however, this process, together with PSC, remains largely underexplored. The main purpose of this study is to investigate the role of PSC in enhancing the moderating role of individuals' psychological detachment and relaxation during weekends on the relationship between daily job demands in Week 1 and daily emotional exhaustion in Week 2. Data from a shortitudinal study of 178 teachers (in total, 534 diaries) in Terengganu, Malaysia, over two consecutive weeks, were operationalized at the individual level and daily level using Hierarchical Linear Modeling (HLM) software. The analysis showed that the daily experience of job demands in the prior week led to an increment of emotional exhaustion in the following week. Interestingly, the interaction role of PSC*psychological detachment and PSC*relaxation moderated the relationship between daily job demands and daily emotional exhaustion. Overall, this study highlights the importance of individual off-job recovery time and that building good PSC at work is fundamental in compensating for the adverse relationships between job demands and its consequences.

    BACKGROUND: The purpose of this study is to uncover the effect of psychological safety climate (PSC) on employees' job satisfaction and organisational climate mediating processes explaining that association. It is posited that the four PSC aspects (management commitment, management priority, organisational participation, and organisational communication) are important for employees' job satisfaction and organisational climate act as resources to facilitate the enactment of managerial quality.

    METHODS: This study uses a quantitative approach through a questionnaire survey method involving 340 Kota Kinabalu City Hall employees who were selected through simple random sampling.

    RESULTS: The results of linear regression analysis found that organisation participation has a positive significant relationship with job satisfaction. Organisational communication also showed a negative and significant relationship with job satisfaction. Meanwhile, both management commitment and management priority are statistically insignificant. When the organisational climate is included in the relationship as a mediator through Structural Equation Modelling (SEM) to reinforce the role of psychological safety climate in increasing job satisfaction, such mediating role can only strengthen the relationship between management commitment and organisational participation with job satisfaction.

    CONCLUSION: Despite the study being cross-sectional, it contributes to knowledge on the resources facilitating PSC, which is important for employees' psychological health. From a practical viewpoint, this study contributes to the literature showing that organizations with good PSC should have policies and practices directed towards employee well-being. The implications of the study for DBKK management are to providing knowledge on the types of psychosocial safety climate domains that plays a crucial role in improving the job satisfaction of DBKK employees.

    OBJECTIVE: To compare catheter related blood stream infection (CRBSI) rate between cuffed tunnelled and non-cuffed tunnelled PICC.

    METHODS: We prospectively followed 100 patients (50:50 cuffed and non-cuffed PICC) and compared CRBSI rate between these groups. Daily review and similar catheter care were performed until a PICC-related complication, completion of therapy, death or defined end-of-study date necessitate removal. CRBSI was confirmed in each case by demonstrating concordance between isolates colonizing the PICC at the time of infection and from peripheral blood cultures.

    RESULTS: A total of 50 cuffed PICC were placed for 1864 catheter-days. Of these, 12 patients (24%) developed infection, for which 5 patients (10%) had a CRBSI for a rate of 2.7 per 1000 catheter-days. Another 50 tunnelled non-cuffed PICCs were placed for 2057 catheter-days. Of these, 7 patients (14%) developed infection, for which 3 patients (6%) had a CRBSI. for a rate of 1.5 per 1000 catheter-days. The mean time to development of infection is 24 days in cuffed and 19 days in non-cuffed groups. The mean duration of utilization was significantly longer in non-cuffed than in cuffed group (43 days in non-cuffed vs 37 days in cuffed group, p = 0.008).

    CONCLUSIONS: Cuffed PICC does not further reduce the rate of local or bloodstream infection. Tunnelled non-cuffed PICC is shown to be as effective if not better at reducing risk of CRBSI and providing longer catheter dwell time compared to cuffed PICC.

    Algae biomass comprises variety of biochemicals components such as carbohydrates, lipids and protein, which make them a feasible feedstock for biofuel production. However, high production cost mainly due to algae cultivation remains the main challenge in commercializing algae biofuels. Hence, extraction of other high value-added bioproducts from algae biomass is necessary to enhance the economic feasibility of algae biofuel production. This paper is aims to deliberate the recent developments of conventional technologies for algae biofuels production, such as biochemical and chemical conversion pathways, and extraction of a variety of bioproducts from algae biomass for various potential applications. Besides, life cycle evaluation studies on microalgae biorefinery are presented, focusing on case studies for various cultivation techniques, culture medium, harvesting, and dewatering techniques along with biofuel and bioenergy production pathways. Overall, the algae biorefinery provides new opportunities for valorisation of algae biomass for multiple products synthesis.

    Induced pluripotent stem cells (iPSCs) have great potentials for regenerative medicine. However, serious concerns such as the use of the viral-mediated reprogramming strategies and exposure of iPSCs to animal products from feeder cells and serum-containing medium have restricted the application of iPSCs in the clinics. Therefore, the generation of iPSCs with minimal viral integrations and in non-animal sourced and serum-free medium is necessary. In this report, a polycistronic lentiviral vector carrying Yamanaka's factors was used to reprogram mouse fibroblasts into iPSCs in feeder- and xeno-free culture environment. The generated iPSCs exhibited morphology and self-renewal properties of embryonic stem cells (ESCs), expression of specific pluripotent markers, and potentials to differentiate into the three-major distinct specialized germ layers in vitro. The iPSCs were also shown to have the potential to differentiate into neural precursor and neurons in culture, with greater than 95% expression of nestin, Pax6 and βIII-tubulin. This body of work describes an alternative method of generating iPSCs by using polycistronic lentiviral vector that may minimize the risks associated with viral vector-mediated reprogramming and animal derived products in the culture media.

    PURPOSE: This study aimed to assess the effect of nitrogen, salt and pre-culture conditions on the production of lovastatin in A. terreus ATCC 20542.

    METHODS: Different combinations of nitrogen sources, salts and pre-culture combinations were applied in the fermentation media and lovastatin yield was analysed chromatographically.

    RESULT: The exclusion of MnSO4 ·5H2O, CuSO4·5H2O and FeCl3·6H2O were shown to significantly improve lovastatin production (282%), while KH2PO4, MgSO4·7H2O, and NaCl and ZnSO4·7H2O were indispensable for good lovastatin production. Simple nitrogen source (ammonia) was unfavourable for morphology, growth and lovastatin production. In contrast, yeast extract (complex nitrogen source) produced the highest lovastatin yield (25.52 mg/L), while powdered soybean favoured the production of co-metabolites ((+)-geodin and sulochrin). Intermediate lactose: yeast extract (5:4) ratio produced the optimal lovastatin yield (12.33 mg/L) during pre-culture, while high (5:2) or low (5:6) lactose to yeast extract ratio produced significantly lower lovastatin yield (7.98 mg/L and 9.12 mg/L, respectively). High spore concentration, up to 107 spores/L was shown to be beneficial for lovastatin, but not for co-metabolite production, while higher spore age was shown to be beneficial for all of its metabolites.

    CONCLUSION: The findings from these investigations could be used for future cultivation of A. terreus in the production of desired metabolites.

    The application of pultruded glass fiber-reinforced polymer composites (PGFRPCs) as a replacement for conventional wooden cross-arms in transmission towers is relatively new. Although numerous studies have conducted creep tests on coupon-scale PGFRPC cross-arms, none had performed creep analyses on full-scale PGFRPC cross-arms under actual working load conditions. Thus, this work proposed to study the influence of an additional bracing system on the creep responses of PGFRPC cross-arms in a 132 kV transmission tower. The creep behaviors and responses of the main members in current and braced PGFRPC cross-arm designs were compared and evaluated in a transmission tower under actual working conditions. These PGFRPC cross-arms were subjected to actual working loads mimicking the actual weight of electrical cables and insulators for a duration of 1000 h. The cross-arms were installed on a custom test rig in an open area to simulate the actual environment of tropical climate conditions. Further creep analysis was performed by using Findley and Burger models on the basis of experimental data to link instantaneous and extended (transient and viscoelastic) creep strains. The addition of braced arms to the structure reduced the total strain of a cross-arm's main member beams and improved elastic and viscous moduli. The addition of bracing arms improved the structural integrity and stiffness of the cross-arm structure. The findings of this study suggested that the use of a bracing system in cross-arm structures could prolong the structures' service life and subsequently reduce maintenance effort and cost for long-term applications in transmission towers.

    The phenomenon of 'lost fetus' to orang bunian is quite commonly heard of in our culture. It may present in psychiatric settings and may potentially create confusion and difficulties in diagnostic and management aspects. A case of feigned full-term pregnancy followed by a pregnancy loss is described. This review emphasizes the need to recognize this phenomenon of 'lost advanced pregnancy' in context of the Malay belief. Possible etiological factors, diagnostic indicators of factitious disorder and management strategies are discussed.     This study determined the effect of growth media and culture concentration on the growth, proximate, and microelement composition of Ankistrodesmus falcatus. The culture of A. falcatus was done using three media, namely Modified COMBO Medium (COMBO), Bold's Basal Medium (BBM), and Bristol, at two concentrations (50% and 100%). The results obtained show that the cell density (>3.5 × 107 cells/mL), optical density (>0.24), and specific growth rate (>0.429%/day) were significantly higher (p ≤ 0.05) in BBM and COMBO than in Bristol (<3.1 × 107 cells/mL; <0.23; <0.416%/day, respectively) at both concentrations. However, biomass was higher in BBM (>2.20 g/L) than in COMBO (1.87-2.13 g/L), while Bristol had the lowest value observed (1.70-1.73 g/L). Biochemical and microelement composition showed variations between media and at the different concentrations, with higher values observed in BBM and COMBO. Based on the growth parameters and nutritional composition, it was concluded that BBM and COMBO were better media for the propagation of A. falcatus growth than Bristol. The study also demonstrated that the microalgae can be cultured using half of the media's concentration to lower production costs.

    For people with chronic illnesses in low-and-middle-income countries, access to enabling resources that contribute to health, economic and social resilience such as continued employment, often fall outside the health sector's remit or delivery of national structural protection. In the absence of sufficient laws and policies that mitigate discrimination and enhance reasonable work modifications, private employers have a high degree of agency and discretion in how they hire, manage, or terminate employees with chronic illnesses (ECI). There is a scarcity of research on how employers make decisions under these conditions. Using a constructivist grounded theory approach, we interviewed and analysed data from 30 human resource (HR) professionals and decision-makers within private organisations in Klang Valley, Malaysia (June 2015-September 2016). In this paper, we use 'ethics of care' as an analytic, and moral lens to present HR's decision-making rationales in caring for and managing ECI. Respondents described the positive influence of international practices, including through parent company policies, as a reference for best practice. While overt bias and discriminatory perceptions were predictably described, participants also discussed care as relational organisational culture, and strategy, albeit selectively. Apart from illness factors such as duration and severity, descriptions of 'selective caregiving' included considerations of an employee's duration in organisations, the perceived value of the employee to employers, organisation size, ethos, resources and capabilities, and how organisations managed the uncertainty of illness futures as a potential risk to organisation outcomes. Selective caregiving can contribute to social, economic and health inequalities in populations with chronic illness. Nevertheless, global health actors can use the problems identified by participants, as entry points to engage more closely with employers and the broader private and commercial sectors in LMICs, to facilitate more inclusive care, and care-based intersectoral work to address the social and economic determinants of health.

    Our work cautions against the use of serum-supplemented culture media in a transwell migration assay when using chemoattractants other than FBS. At 24 h, a 5% foetal bovine serum (FBS) gradient caused BV2 microglia to migrate toward the lower compartment of the transwell apparatus. Interestingly, FBS-supplemented media in the absence of a gradient also resulted in notable microglia migration. Serum can therefore confound the interpretation of a transwell migration assay when another chemoattractant is used.

    Objective. Our innovation was to propose a multilevel model to explain how an organizational factor, psychosocial safety climate (PSC) - the climate for worker psychological health - related to work investment (work engagement and workaholism) and, in turn, psychological distress. Methods. Longitudinal data were collected in Peninsular Malaysia across 26 police departments from 392 police personnel, matched across 4 months, and were tested using hierarchical linear modeling. Results. The analysis revealed between-group effects linking PSC to job resources, to work engagement and to workaholism. When PSC operated by improving job resources, aside from increased work engagement, it could unwittingly boost workaholism. However, this only existed under low PSC conditions. The secondary function of PSC buffered the impact of job resources on workaholism and psychological distress. When PSC was high, job resources reduced both workaholism and psychological distress, suggesting that PSC enabled resources to do their job of mitigating unfavorable conditions. Conclusions. Results support a multilevel PSC-extended job demands-resources motivational path with cross-links, and PSC's moderation function, as an explanation of worker psychological health. Confirming PSC as a leading indicator and the importance of a motivational path, this article presents new evidence in support of targeting PSC to improve worker psychological health.

    A review of valorization of oyster mushroom species and waste generated in the mushroom cultivation is presented, with a focus on the cultivation and valorization techniques, conditions, current research status and particularly the hazard mitigation and value-added recovery of the waste mushroom substrate (WMS) - an abundant waste in mushroom cultivation industry. Based on the studies reviewed, the production rate of the present mushroom industry is inadequate to meet market demands. There is a need for the development of new mushroom cultivation methods that can guarantee an increase in mushroom productivity and quality (nutritional and medicinal properties). This review shows that the cylindrical baglog cultivation method is more advantageous compared with the wood tray cultivation method to improve the mushroom yield and cost efficiency. Approximately 5 kg of potentially hazardous WMS (spreading diseases in mushroom farm) is generated for production of 1 kg of mushroom. This encourages various valorization of WMS for use in agricultural and energy conversion applications, mainly as biocompost, plant growing media, and bioenergy. The use of WMS as biofertilizer has shown desirable performance compared to conventional chemical fertilizer, whilst the use of WMS as energy feedstock could produce cleaner bioenergy sources compared to conventional fuels.

    A thermostable bacterial lipase from Geobacillus zalihae was expressed in a novel yeast Pichia sp. strain SO. The preliminary expression was too low and discourages industrial production. This study sought to investigate the optimum conditions for T1 lipase production in Pichia sp. strain SO. Seven medium conditions were investigated and optimized using Response Surface Methodology (RSM). Five responding conditions namely; temperature, inoculum size, incubation time, culture volume and agitation speed observed through Plackett-Burman Design (PBD) method had a significant effect on T1 lipase production. The medium conditions were optimized using Box-Behnken Design (BBD). Investigations reveal that the optimum conditions for T1 lipase production and Biomass concentration (OD600) were; Temperature 31.76 °C, incubation time 39.33 h, culture volume 132.19 mL, inoculum size 3.64%, and agitation speed of 288.2 rpm with a 95% PI low as; 12.41 U/mL and 95% PI high of 13.65 U/mL with an OD600 of; 95% PI low as; 19.62 and 95% PI high as; 22.62 as generated by the software was also validated. These predicted parameters were investigated experimentally and the experimental result for lipase activity observed was 13.72 U/mL with an OD600 of 24.5. At these optimum conditions, there was a 3-fold increase on T1 lipase activity. This study is the first to develop a statistical model for T1 lipase production and biomass concentration in Pichia sp. Strain SO. The optimized production of T1 lipase presents a choice for its industrial application.

    Spirulina biomass accounts for 30% of the total algae biomass production globally. In conventional process of Spirulina biomass production, cultivation using chemical-based culture medium contributes 35% of the total production cost. Moreover, the environmental impact of cultivation stage is the highest among all the production stages which resulted from the extensive usage of chemicals and nutrients. Thus, various types of culture medium such as chemical-based, modified, and alternative culture medium with highlights on wastewater medium is reviewed on the recent advances of culture media for Spirulina cultivation. Further study is needed in modifying or exploring alternative culture media utilising waste, wastewater, or by-products from industrial processes to ensure the sustainability of environment and nutrients source for cultivation in the long term. Moreover, the current development of utilising wastewater medium only support the growth of Spirulina however it cannot eliminate the negative impacts of wastewater. In fact, the recent developments in coupling with wastewater treatment technology can eradicate the negative impacts of wastewater while supporting the growth of Spirulina. The application of Spirulina cultivation in wastewater able to resolve the global environmental pollution issues, produce value added product and even generate green electricity. This would benefit the society, business, and environment in achieving a sustainable circular bioeconomy.

    Body measurement is the first process that must be encountered before any
    custom-made compression garment can be designed. The current practice of
    obtaining the measurement is by traditional methods using tools like
    measuring tape. However, this method is considered to be time-consuming
    and usually not accurate. The most popular solution to the problem is by using
    non-contact measurement. The development of the 3D whole body scanner
    has made non-contact body measurement become a reality due to its capacity
    to capture a vast amount of information. However, the cost to buy the whole
    body scanner is quite expensive. Moreover, their sizes are also bulky which
    make them less portable. Thus, a handheld body scanner provides a solution
    to the problem. Despite that, current handheld scanner only provide image
    and visualization aspect, but not the measurement aspect. This paper reports
    the development of a method to acquire body data from a 3D handheld
    scanner. In this new method, the point cloud of a body part was collected
    using the handheld scanner. Then, the data was transformed into point
    coordinates. Several processes were developed to filter the number of points
    to allow for faster processing time and increasing the measurement accuracy.
    In the first process, only points at specific height/layers are selected. In the
    second process, the remaining points are rearranged according to their height
    and angle. In the last process, the number of points is further reduced. In this
    process, the number of points per layer is limited to 72 points. Results show
    that the method can be used to determine body measurement.

    Ectoine production using inexpensive and renewable biomass resources has attracted great interest among the researchers due to the low yields of ectoine in current fermentation approaches that complicate the large-scale production of ectoine. In this study, ectoine was produced from corn steep liquor (CSL) and soybean hydrolysate (SH) in replacement to yeast extract as the nitrogen sources for the fermentation process. To enhance the bacterial growth and ectoine production, biotin was added to the Halomonas salina fermentation media. In addition, the effects addition of surfactants such as Tween 80 and saponin on the ectoine production were also investigated. Results showed that both the CSL and SH can be used as the nitrogen source substitutes in the fermentation media. Higher amount of ectoine (1781.9 mg L-1) was produced in shake flask culture with SH-containing media as compared to CSL-containing media. A total of 2537.0 mg L-1 of ectoine was produced at pH 7 when SH-containing media was applied in the 2 L batch fermentation. Moreover, highest amount of ectoine (1802.0 mg L-1) was recorded in the SH-containing shake flask culture with addition of 0.2 μm mL-1 biotin. This study demonstrated the efficacy of industrial waste as the nutrient supplement for the fermentation of ectoine production.

    The massive success of blockchain technology in Bitcoin is taking the world by storm by attracting vast acceptance from both the public and private sectors. Blockchain allows digital transactions between two parties without a third party as a broker. Blockchain is now applicable beyond fintech to various other industries. Among these, Hyperledger fabric has emerged as the most popular blockchain-based open-source permissioned platform targeting business applications. It has been used in over 400 proofs-of-concept blockchain and is well proven in applications, such as supply chain, healthcare, and so on. Despite the many obvious benefits observed in blockchain-enhanced platforms, there still exist technical challenges in scalability, causing performance deficiency, which includes latency and throughput. There is an exigent need to improve the current blockchain-based applications to have the blockchain nodes be scalable without compromising the blockchain performance. In this study, we present the impact of workload variance of up to 1000 transactions with the setup of 20 blockchain nodes in the Hyperledger LTS platform. The evaluation metrics are transaction success and failure rate, throughput, and latency in the blockchain. The transaction throughput was found to be consistent with the increasing workload on a constant number of nodes. However, it showed a declining trend with an increasing number of nodes. As far as the latency, it was in tandem with the increased workload and the number of nodes. We, therefore, conclude that the LTS version is suitable for small and medium enterprises that do not scale up.
