    The occurrence of Leptospirosis and Escherichia coli coinfection in the post-partum period is a novel case. This report illustrated a previously well woman from a suburban area presented with acute neurological deterioration following a two days history of fever during her puerperal period. Early interventions with fluids, broad spectrum antibiotics and intensive supportive care were given. Despite that, she deteriorated rapidly and developed pulmonary hemorrhage, disseminated intravascular coagulopathy, and multi-organ failure. She succumbed within 12 hours of admission. The knowledge about such fatal co-infections should be disseminated to medical practitioners encountering Leptospirosis infection and general public.

    The safety level of microwaved foods remains at vague as this subject was less addressed
    scientifically. A study was initiated to address the matter by investigating on the
    survivability of Salmonella and Shiga-toxigenic Escherichia coli (STEC) O157 in
    microwave heated ready-to-eat (RTE) foods using the Most Probable Number coupled
    Polymerase Chain Reaction (MPN-PCR) technique. A total of 329 samples of various
    ready-to-eat (RTE) convenience meals were collected around Wilayah Persekutuan Kuala

    Setiap kejadian semula jadi merupakan rahmat dan memberi kemaslahatan kepada manusia termasuklah pendarahan haid dalam kalangan wanita. Haid merupakan suatu proses yang unik dan terancang yang melibatkan tiga fasa utama iaitu fasa haid, fasa folikel dan fasa luteal. Objektif penyelidikan ini ialah untuk mengkaji dan menghubungkaitkan kebaikan haid daripada perspektif sains dan maqasid syariah. Kajian ini dijalankan dengan menggunakan ulasan literatur secara deskriptif. Kajian mendapati bahawa perubahan hormon, warna darah dan faktor fiziokimia lain memberi kesan terhadap kenormalan kitaran haid. Darah haid juga dikenal pasti mempunyai agen antimikrob terutamanya terhadap bakteria E. coli dan bakteria Gram-negatif lain. Hal ini bertepatan dengan maqasid syariah memelihara jiwa kerana haid mampu juga memelihara kesihatan wanita. Selain itu, kajian lepas menemukan bahawa darah haid terdiri daripada sel stem yang boleh digunakan dalam aktiviti klinikal pada masa akan datang. Maqasid syariah melindungi keturunan juga dapat dilihat dengan kehadiran haid yang sering digunakan bagi menjangkakan waktu subur bagi merancang kehamilan. Hikmah Islam melarang mendekatkan diri (bersetubuh) dengan wanita yang sedang haid adalah satu rahmat yang besar, kerana wanita yang sedang haid biasanya mempunyai kelaziman gejala prahaid (PMS) yang melibatkan isu kesihatan. Oleh itu, haid daripada perspektif sains adalah bertepatan dengan maqasid syariah.

    Kawasan Pentadbiran Putrajaya merangkumi 70% daripada kawasan tadahan Tasik Putrajaya. Kerja-kerja pembangunan yang dijalankan di dalam kawasan Putrajaya kebiasaannya dapat dikawal dengan merujuk kepada peraturan-peraturan dan garis panduan yang telah disediakan oleh pihak Perbadanan Putrajaya dalam memastikan kualiti air Tasik dan Tanah Bencah Putrajaya sentiasa mematuhi piawaian yang telah ditetapkan. Seluas 30% daripada kawasan tadahan Tasik dan Tanah Bencah Putrajaya terletak di luar pentadbiran dan kawalan pihak berkuasa Putrajaya. Kawasan tadahan Utara Putrajaya adalah kawasan tadahan yang luas yang menyumbang sebahagian besar air yang memasuki kawasan Tasik dan Tanah Bencah Putrajaya. Berasal dari anak Sg. Chuau kini ia telah membentuk satu kawasan tasik dan tanah bencah tersebut. Data kualiti air yang dilakukan oleh pihak berkuasa Putrajaya iaitu Perbadanan Putrajaya dari tahun 2002 sehingga 2005 dianalisis bagi mengenalpasti masalah utama kualiti air dari kawasan Tadahan Utara Tanah Bencah Putrajaya. Data kualiti air yang dicatat sepanjang tahun 2002 mendapati purata peratusan parameter yang melebihi standard Putrajaya bagi ammoniakal nitrogen (NH3-N) ialah 43.7%, E. coli 31.3%, TSS 12.5% manakala DO dan COD masing-masing 6.2%. Untuk sepanjang tahun 2003, purata peratusan parameter bagi NH3-N ialah 23.7%, E. coli 18.4%, jumlah koliform 18.4%, TSS 2.6%, DO 13.2%, COD 13.2%, dan BOD 10.5%. Untuk sepanjang tahun 2004, purata peratusan parameter bagi NH3-N ialah 35.5%, E. coli 22.6%, jumlah koliform 12.9%, TSS 9.7%, COD 3.2%, dan BOD 16.1%. Untuk tahun 2005, purata peratusan parameter bagi E. coli 36.4%, NH3-N ialah 22.7%, jumlah koliform 18.2%, BOD 13.6%, DO 4.5%, dan COD juga 4.5%. Secara keseluruhan, analisis bagi tempoh empat tahun ini mendapati parameter NH3-N dan E. coli sering kali menunjuk bacaan yang tidak mematuhi Piawaian Putrajaya. Punca utama masalah kualiti air di kawasan Tadahan Utara Tanah Bencah Putrajaya adalah kegagalan mengurus kawasan tadahan secara bersepadu dan sikap pihak berkepentingan di kawasan kajian tidak mengurus dan mengawal secara menyeluruh masalah air larian, kawasan terbuka dan kelodakan ketika membangunkan kawasan.

    Stool samples obtained from 120 Malay children, aged below 6 years, in Mukim Ijok, Kuala Selangor, were tested. Enteric pathogens were found in 7.5% of specimens. Enteropathogenic Escherichia coli (EPEC) was the most common enteric pathogen isolated (6.7%) followed by Salmonella spp (0.83%). The rate of isolation was independent of age and sex.

    This investigation of the temperature dependence of DppA interactions with a subset of three dipeptides (AA. AF and FA) by isothermal titration calorimetry has revealed the negative heat capacity ([Formula: see text]) that is a characteristic of hydrophobic interactions. The observation of enthalpy-entropy compensation is interpreted in terms of the increased structuring of water molecules trapped in a hydrophobic environment, the enthalpic energy gain from which is automatically countered by the entropy decrease associated with consequent loss of water structure flexibility. Specificity for dipeptides stems from appropriate spacing of designated DppA aspartate and arginine residues for electrostatic interaction with the terminal amino and carboxyl groups of a dipeptide, after which the binding pocket closes to become completely isolated from the aqueous environment. Any differences in chemical reactivity of the dipeptide sidechains are thereby modulated by their occurrence in a hydrophobic environment where changes in the structural state of entrapped water molecules give rise to the phenomenon of enthalpy-entropy compensation. The consequent minimization of differences in the value of ΔG0 for all DppA-dipeptide interactions thus provides thermodynamic insight into the biological role of DppA as a transporter of all dipeptides across the periplasmic membrane.

    Escherichia coli strain USML2 was originally isolated from the inner leaf tissues of surface-sterilized phytopathogenic-free oil palm (Elaeis guineensis Jacq.). We present here the whole-genome sequence of this plant-endophytic strain. The genome consists of a single circular chromosome of 4,502,758 bp, 4,315 predicted coding sequences, and a G+C content of 50.8%.

    The production of heat-labile (LT) and heat-stable (ST) enterotoxins, colonisation factor antigens (CFAs) and haemagglutinins was investigated amongst 310 Escherichia coli (E. coli) isolates obtained from 62 children under the age of five, with diarrhoea. Twenty-one isolates were found to produce enterotoxins, of which fifteen (71%) isolates produced ST only, 2 (10%) produced LT only and 4 (19%) produced both LT and ST. However, none of the isolates demonstrated any of the common CFAs identified to date, but 8 out of the 21 isolates demonstrated haemagglutination with rabbit, sheep or human group A erythrocytes, suggesting the presence of putative CFAs, yet unidentified.

    Glaciozyma antarctica PI12 is a psychrophilic yeast isolated from Antarctica. In this work, we describe the heterologous production, biochemical properties and in silico structure analysis of an arginase from this yeast (GaArg). GaArg is a metalloenzyme that catalyses the hydrolysis of L-arginine to L-ornithine and urea. The cDNA of GaArg was reversed transcribed, cloned, expressed and purified as a recombinant protein in Escherichia coli. The purified protein was active against L-arginine as its substrate in a reaction at 20 °C, pH 9. At 10-35 °C and pH 7-9, the catalytic activity of the protein was still present around 50%. Mn2+, Ni2+, Co2+ and K+ were able to enhance the enzyme activity more than two-fold, while GaArg is most sensitive to SDS, EDTA and DTT. The predicted structure model of GaArg showed a very similar overall fold with other known arginases. GaArg possesses predominantly smaller and uncharged amino acids, fewer salt bridges, hydrogen bonds and hydrophobic interactions compared to the other counterparts. GaArg is the first reported arginase that is cold-active, facilitated by unique structural characteristics for its adaptation of catalytic functions at low-temperature environments. The structure and function of cold-active GaArg provide insights into the potentiality of new applications in various biotechnology and pharmaceutical industries.

    Enteropathogenic Escherichia coli (EPEC) is a leading cause of diarrhea among infants in developing countries. A total of 38 EPEC isolates, obtained from diarrhea patients of Hospital Miri, Sarawak, were investigated through plasmid profile, antibiotic resistance and randomly amplified polymorphic DNA (RAPD) analysis. From the 8 types of antibiotics used, all isolates were 100% resistant to furoxime, cephalothin and sulphamethoxazole and showed high multiple antibiotic resistant (MAR) indexes, ranging from 0.5 to 1.0. In plasmid profiling, 22 isolates (58%) showed the presence of one or more plasmids in the range 1.0 to 30.9 mDa. The dendrogram obtained from the results of the RAPD-PCR discriminated the isolates into 30 single isolates and 3 clusters at the level of 40% similarity. The EPEC isolates were highly diverse, as shown by their differing plasmid profiles, antibiotic resistance patterns and RAPD profiles.

    BACKGROUND: The significant role of retail poultry meat as an important exposure pathway for the acquisition and transmission of extended spectrum β-lactamase-producing Escherichia coli (ESBL-EC) into the human population warrants understanding concerning those operational practices associated with dissemination of ESBL-EC in poultry meat retailing. Hence, the objective of this study was to determine the prevalence, spatial distribution and potential risk factors associated with the dissemination of ESBL-EC in poultry meat retail at wet-markets in Selangor, Malaysia.

    METHODS: Poultry meat (breast, wing, thigh, and keel) as well as the contact surfaces of weighing scales and cutting boards were sampled to detect ESBL-EC by using culture and disk combination methods and polymerase chain reaction assays. Besides, questionnaire was used to obtain data and information pertaining to those operational practices that may possibly explain the occurrence of ESBL-EC. The data were analysed using logistic regression analysis at 95 % CI.

    RESULTS: The overall prevalence of ESBL-EC was 48.8 % (95 % CI, 42 - 55 %). Among the risk factors that were explored, type of countertop, sanitation of the stall environment, source of cleaning water, and type of cutting board were found to be significantly associated with the presence of ESBL-EC.

    CONCLUSIONS: Thus, in order to prevent or reduce the presence of ESBL-EC and other contaminants at the retail-outlet, there is a need to design a process control system based on the current prevailing practices in order to reduce cross contamination, as well as to improve food safety and consumer health.

    INTRODUCTION: Uropathogenic virulence factors have been identified by comparing the prevalence of these among urinary tract isolates and environmental strains. The uropathogenic-specific protein (USP) gene is present on the pathogenicity island (PAI) of uropathogenic Escherichia coli (UPEC) and, depending on its two diverse gene types and the sequential patterns of three open reading frame units (orfUs) following it, there is a method to characterize UPEC epidemiologically called PAIusp subtyping.
    METHODOLOGY: A total of 162 UPEC isolates from Sabah, Malaysia, were tested for the presence of the usp gene and the sequential patterns of three orfUs following it using polymerase chain reaction (PCR). In addition, by means of triplex PCR, the prevalence of the usp gene was compared with other two VFs of UPEC, namely alpha hemolysin (α-hly) and cytotoxic necrotizing factor (cnf-1) genes encoding two toxins.
    RESULTS: The results showed that the usp gene was found in 78.40% of UPEC isolates, indicating that its prevalence was comparable to that found in a previous study in Japan. The two or three orfUs were also associated with the usp gene in this study. All the PAIusp subtypes observed in Japan were present in this study, while subtype IIa was the most common in both studies. The usp gene was observed in a higher percentage of isolates when compared with α-hly and cnf-1 genes.
    CONCLUSIONS: The findings in Japan and Sabah, East Malaysia, were similar, indicating that PAIusp subtyping is applicable to the characterization of UPEC strains epidemiologically elsewhere in the world.

    Escherichia coli isolates resistant to ceftazidime isolated in the University Malaya Medical Center (UMMC) Kuala Lumpur, Malaysia, between the years 1998 and 2000 were studied for extended-spectrum beta-lactamase (ESBL) production. All strains were analysed phenotypically and genotypically and found to be ESBL-producing organisms harbouring SHV-5 beta-lactamase. This was confirmed by PCR-SSCP and nucleotide sequencing of the blaSHV amplified gene. As there was no evidence of ESBL activity in E. coli prior to this, coupled with the fact that there was a predominance of SHV-5 beta-lactamases in Klebsiella pneumoniae isolates in UMMC, we postulate that the E. coli obtained the SHV-5 beta-lactamase genes by plasmid transfer from the ESBL-producing K. pneumoniae.

    Strains of Escherichia coli that are non-typeable by pulsed-field gel electrophoresis (PFGE) due to in-gel degradation can influence their molecular epidemiological data. The DNA degradation phenotype (Dnd(+)) is mediated by the dnd operon that encode enzymes catalyzing the phosphorothioation of DNA, rendering the modified DNA susceptible to oxidative cleavage during a PFGE run. In this study, a PCR assay was developed to detect the presence of the dnd operon in Dnd(+) E. coli strains and to improve their typeability. Investigations into the genetic environments of the dnd operon in various E. coli strains led to the discovery that the dnd operon is harboured in various diverse genomic islands.

    The prevalence of Enterotoxigenic Escherichia coli (ETEC) in 433 stool samples from diarrhoeal cases of all ages was studied using two commercially available test kits for the detection of heat labile toxin (LT) and the infant mouse assay for the heat stable toxin (ST). 16 samples (3.7%) were positive for ETEC, of which nine were producing ST alone, six LT alone and only one was producing both LT and ST. Although the percentage of isolation rate was low, its occurrence was almost as common as the Shigella spp and Salmonella spp in the same study. Of the two test kits examined, the Phadebact ETEC-LT Test 50 (Pharmacia Diagnostics, Uppsala, Sweden) was found to be more suitable for use in a routine diagnostic laboratory. Ten out of 12 (83%) of the strains tested were resistant to one or more antibiotics.

    Plasmids of incompatibility group A/C (IncA/C) are becoming increasingly prevalent within pathogenic Enterobacteriaceae They are associated with the dissemination of multiple clinically relevant resistance genes, including blaCMY and blaNDM Current typing methods for IncA/C plasmids offer limited resolution. In this study, we present the complete sequence of a blaNDM-1-positive IncA/C plasmid, pMS6198A, isolated from a multidrug-resistant uropathogenic Escherichia coli strain. Hypersaturated transposon mutagenesis, coupled with transposon-directed insertion site sequencing (TraDIS), was employed to identify conserved genetic elements required for replication and maintenance of pMS6198A. Our analysis of TraDIS data identified roles for the replicon, including repA, a toxin-antitoxin system; two putative partitioning genes, parAB; and a putative gene, 053 Construction of mini-IncA/C plasmids and examination of their stability within E. coli confirmed that the region encompassing 053 contributes to the stable maintenance of IncA/C plasmids. Subsequently, the four major maintenance genes (repA, parAB, and 053) were used to construct a new plasmid multilocus sequence typing (PMLST) scheme for IncA/C plasmids. Application of this scheme to a database of 82 IncA/C plasmids identified 11 unique sequence types (STs), with two dominant STs. The majority of blaNDM-positive plasmids examined (15/17; 88%) fall into ST1, suggesting acquisition and subsequent expansion of this blaNDM-containing plasmid lineage. The IncA/C PMLST scheme represents a standardized tool to identify, track, and analyze the dissemination of important IncA/C plasmid lineages, particularly in the context of epidemiological studies.

    Local Thai and imported Malaysian beef in southern Thailand area carry
    several Shiga toxin-producing Escherichia coli (STEC) serotypes. STEC O104 is an
    important pathogen capable of causing outbreaks with considerable morbidity
    and mortality. This study investigated the presence of E. coli O104 from local Thai
    and imported Malaysian beef obtained from markets in Hat Yai City, Songkhla
    Province during August 2015 - February 2016. Thirty-one E. coli O104 strains
    were isolated from 12 beef samples (16% and 23% Thai and imported Malaysian,
    respectively). Thirty strains possessed aggA (coding for a major component of
    AAF/I fimbriae), a gene associated with enteroaggregative E. coli (EAEC) pathotype,
    and all strains carried fimH (encoding Type 1 fimbriae). Thirty strains
    belonged to phylogenetic group B1 and one strain (from Malaysian beef) to group
    A. Agglutination of yeast cells was observed among 29 E. coli O104 strains. Investigation
    of stx2 phage occupancy loci demonstrated that sbcB was occupied in 12
    strains. Antimicrobial susceptibility assay revealed that 7 strains were resistant
    to at least one antimicrobial agent and two were multi-drug resistant. One strain
    carried extended spectrum β-lactamase gene blaCTX-M and three carried blaTEM. PFGE-generated DNA profiling showed identical DNA pattern between that of
    one EAEC O104 strain from Thai beef and another from Malaysian beef, indicating
    that these two strains originated from the same clone. This is the first report
    in Thailand describing the presence of EAEC O104 from both Thai and imported
    Malaysian beef and their transfer between both countries. Thorough surveillance
    of this pathogen in fresh meats and vegetables should help to prevent any possible
    outbreak of E. coli O104.
