Known as ‘belangkas’ in Malaysia, horseshoe crabs have been used by locals for food, bait and as fertilizer. Currently, these ancient mariners are gaining a lot of attention as the amoebocyte from their blood are harvested by the biomedical industry for use to detect human pathogens in patients, injectable drugs and intravenous devices.

FcgammaRIIs are the most widely distributed of the Fcgamma receptor family and play an important role in the clearance of immune complexes. Evidence that the FcgammaRIIa-R131 allotype is less able to process and clear immune complexes effectively suggests that this may be a disease susceptibility factor for systemic lupus erythematosus (SLE). Data from studies published thus far do not agree on the potential role of FcgammaRIIa polymorphism in the genetics of SLE. Most studies in fact show no evidence for any correlation between polymorphism of FcgammaRIIa and risk for SLE. However, it remains to be determined whether FcgammaRIIa polymorphism may play a critical role in certain groups of patients, especially in those of differing ethnic background. Polymorphism of FcgammaRIIa may also be important in determining disease phenotype, and identification of this influence may have important implications in patient care and in identifying patients for more aggressive therapy.

Pogostemon cablin (patchouli) is a medicinal herb well known for its
essential oil derived from the leaves. Patchouli oil shows excellent base note
in fragrance industries for its fixatives properties and its patchouli alcohol
(patchoulol) is used as quality indicator for its oil. However, the P. cablin is
the only commercial source of patchoulol and cannot be obtained
synthetically in the laboratory. Higher demand in the production of its
essential oil gave a significant contribution for in vitro grown P.cablin to
meet the market supply for industries. Hence, in this study, the essential oil
in both in vitro and ex vitro P.cablin were extracted from its leaves by means
of hydrodistillation method and its phytochemical constituents were
identified and compared using Gas Chromatography Mass Spectrometry.
The yield and quality of its essential oil from both in vitro and ex vitro
P.cablin’s leaves were investigated. In vitro patchouli essential oil extraction
gaves higher yield (40 ml) than the ex vitro patchouli essential oil (26 ml)
under similar condition for hydrodistillation. Six major components were
identified through GC-MS and was compared between two samples which
are β- patchoulene, Caryophyllene, α- guaiene, α- cedrene, α- bulnesene and
Patchouli alcohol. The patchoulol, which is the main constituents that is
important in fixative had doubled (42.18 %) in the in vitro P.cablin essential
oil compared to ex vitro (29.24%). This finding was reflected based on the
peak area percentage of each substance through GC-MS. Other constituents
in the in vitro P.cablin were found still competitive to the ex vitro in slightly
lower values. Overall, in vitro P.cablin showed higher yield and quality
compared to the ex vitro grown P.cablin.

This study aims to evaluate the antioxidant activity (total antioxidant and free radical scavenging activities) of seaweeds commercially available in the Malaysian supermarket. Four types of seaweeds namely Nori (Porphyra sp.), Kumbu (Laminaria sp.), Wakame (Undaria sp.) and Hijiki (Hijikia sp.) were used in the study. The extracts were prepared with water and ethanol, respectively. The b-carotene bleaching and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assays were used to determine antioxidant properties of seaweeds by measuring the decrease in absorbance at 470 and 517 nm. In water extract, Kumbu showed the highest total antioxidant activity of 63% compared with other samples. Kumbu, Nori and Hijiki exhibited higher radical scavenging activity than Wakame when extracted with water. Wakame exhibited the highest antioxidant and free radical scavenging activities in ethanolic extract with 58% and EC50 = 0.42 mg/ml respectively. The results of ANOVA analysis show significant differences (p<0.05) in the means of total antioxidant and free radical scavenging activities of the seaweeds. The results showed that processed commercial seaweeds exhibited varying degrees of antioxidant properties.

Microsatellites or simple sequence repeats (SSRs) are tandem repeats of DNA of 1-6 bp long. They ubiquitously occur in both eukaryotic and prokaryotic genomes. Because of their abundance,
they have widespread applications in both animal and plant sciences; such as varietal identification, genetic mapping, QTL mapping, phylogenetic and diversity studies. Thus, SSRs have become valuable DNA markers for molecular biologists and geneticists. Microsatellites are markers
of choice for many molecular geneticists because of their hypervariability, codominant
inheritance, multi-allelism and PCR-based assaying of variations that are amenable to automation and high throughput assay. However, the utilization of microsatellite markers in the past was
hampered by its laborious de novo isolations and species-specific nature.

The blood of the Painted Storks (Mycteria leucocephala) and the Milky Storks (M. cinerea) from Malaysia were collected
invasively from the breeding site. The blood was dropped on to FTA® cards and stored at room temperature. DNA was isolated from
the FTA® cards through a modification of the Wizard DNA Purification kit (Promega) procedure and PCR was performed with 11 pairs
of microsatellite primers of the American Wood Stork (M. americana). The collection of a drop of blood onto the card is superior to the
usual practice of collecting about five ml of blood into a vacuum tube as it causes fewer traumas to these sensitive birds. Moreover, this
collection procedure can be adopted for use in various wild animal species which are usually found in the remote areas of Malaysia as
the sample collection cards can be transported back to the laboratory at room temperature. Our procedure allows the typing of several
molecular genetic markers from just a drop of blood collected in the field and stored at room temperature alleviating the need for storage
in expensive deep freezers or liquid nitrogen tanks.

Electrophoresis is a crucial step for the studies of proteins, allozymes, DNAs and RNAs. Two commonly used electrophoresis systems are agarose gel and polyacrylmide gel. Agarose gel is frequently used for DNAs and RNAs studies whereas polyacrylmide gel is widely used for the studies of other macromolecules such as proteins, allozymes (isozymes), DNAs and RNAs. The banding patterns of the gels, rather than the numbers of bands appearing on the gels are important for scoring in fingerprinting, footprinting and in population genetic studies.

AIM: To describe the clinical characteristic of hepatitis C (HCV) patients and the results of pegylated interferon and ribavirin (PegIFN/RBV) therapy in a routine clinical practice.

METHODS: A retrospective review of consecutive HCV patients treated with PegIFN/RBV in 2004 to 2012.

RESULTS: A total of 273 patients received treatment. The mean age was 44.16 ± 10.5 years and 76% were male. The top 2 self-reported risks were blood or blood product transfusion before 1994 and injection drug use, found in 57.1% of patients. The predominant HCV genotype (GT) was 3 at 60.6%, second was GT1 at 36.1% and other GTs were uncommon at about 1% or less. About half of our patients have high baseline viral load (>800,000 iu/ml), 18.3% had liver cirrhosis and 22.3% had HIV co-infection. Co-morbid illness was found in 42.9%, hypertension and type 2 diabetes were the two most common. The overall sustained virological response (SVR) by intention-to-treat analysis were 54.9% (n=150/273), 41.2% (40/97) for GT1, 100% (5/5) for GT2 and 62% (101/163) for GT3. Subgroup analysis for HCV monoinfected, treatment naïve showed SVR of 49.2% (31/63) for GT1, 100% (5/5) for GT2 and 67% (69/103) for GT3. In HCV mono-infected and treatment experienced (n=29), the SVR was 28.6% (4/14) for GT1, 21.4% (69/103) for GT3. In the HIV/HCV co-infected, treatment naïve (n=56), the SVR was 28.6% (4/14) for GT1 and 64.3% (27/42) for GT3. Treatment naïve GT3 mono-infected patients had a statistically significant higher SVR compared to treatment experienced patients (P=0.001). In GT3 patients who achieved rapid virological response, the SVR was significantly higher at 85.2% (P< 0.001). The SVR for cirrhotics were low especially for GT1 at 21% (4/19) and 31% (4/13) based on all patients and treatment naïve HCV monoinfected respectively. In GT3 cirrhotics the corresponding SVR were 57.1% (16/28) and 60.9% (14/23). Premature discontinuation rate was 21.2% with the majority due to intolerable adverse events at 12.1%.

CONCLUSIONS: In our routine clinical practice, the HCV patients we treated were young, predominantly of GT3 and many had difficult-to-treat clinical characteristics. The SVR of our patients were below those reported in Asian clinical trials but in keeping with some "real world" data.
