    The ctxB gene, the causative agent of cholera epidemic was successfully cloned from V. cholerae in E. coli. The insertion of the gene was confirmed by PCR as well as restriction digestion analyses. The sequencing results for the gene confirmed that the insert was in the correct orientation and in-frame with the P(BAD) promoter and it showed that the gene was 99% homologous to the published ctxB sequence. The CTB protein was successfully expressed in E. coli using the pBAD/His vector system. The expected protein of approximately 14 kDa was detected by SDS-PAGE and Western blot. The use of pBAD/His vector to express the cholera toxin gene in E. coli would facilitate future study of toxin gene products.

    Regeneration potentials in Gerbera jamesonii Bolus ex. Hook f. from tissues culture system was studied using leaf, petiole and root explants. In vitro regeneration, callus induction and root formation were optimized by manipulation of growth regulators during organogenesis. Various kinds of plant growth regulators such as 6-Benzylaminopurine (BAP), alpha-Naphthalene acetic acid (NAA), 2, 4-Dichlorophenoxyacetic acid (2,4-D), Indole-3-acetic acid (IAA), Indole-3-Butyric acid (IBA), N6-[2-Isopentenyl]adenine (2iP), Kinetin and Zeatin were used to initiate cultures. These plant growth regulators were added to Murashige and Skoog medium in different combinations and concentrations. Adventitious shoots were obtained from petiole explants cultured on Murashige and Skoog (MS) medium supplemented with 2.0 mg L(-1) BAP and 0.5 mg L(-1) NAA. Effectiveness of shoot regeneration medium, type of growth regulator used and duration of induction period were investigated. Leaf explants cultured on MS medium supplemented with 1.0 mg L(-1) BAP and 2.0 mg L(-1) 2, 4-D showed the best results for callus induction. Root explants were found to be non-regenerative in all experiments conducted. Petiole segment was identified as the best explant for regeneration of this species. Regenerated plants were rooted on Murashige and Skoog basal medium. Plantlets were then transferred to field with 75% survival rate.

    The present study was aimed to identify the gene of drug resistance betalactamase producing bacteria and clinical features of the infected patients at Hospital University Kebangsaan Malaysia. Blood samples from the patients were collected, processed and betalactamase producing drug resistance bacteria were identified by antibiotic sensitivity testing. Genes of the drug resistance bacteria were detected and characterized by polymerase chain reaction. A total of 34 isolates of drug resistance Betalactamase producing E. coli and Klebsiella spp. were isolated from 2,502 patients. Most common drug resistance gene TEM was found in 50% of the isolates. 11% was found positive for both TEM and SHV. Next 11% of the isolates expressed only SHV genes. Clinical features of the patients were recorded from where the bacteria isolated. Regarding community affiliations 70.5% of the infected patients were Malay 17.6% were Indian and 11.7% were Chinese. Majority of the patients has an underlying pre-morbid condition as reflected by their diagnosis. Better infection control and hygiene in hospitals, plus controlled and prudent use of antibiotics, is required to minimize the impact of drug resistance betalactamase producing bacteria and the spread of infections.

    BACKGROUND: Orange mud crab Scylla olivacea is one of the most important fisheries resources. A new development in ageing technique of crustaceans has been introduced. The detection of growth band deposited in hard structure of gastric mill in the cardiac stomach are found retained after moulting process can be used as age indicator and growth estimation.

    OBJECTIVE: This study was carried out to determine the comparison between carapace width and growth band count of S. olivacea in Malaysia.

    MATERIALS AND METHODS: Samples were collected from Setiu Wetlands, Terengganu, Malaysia from February until August, 2016. Samples were categorized based on their morphological measurements. The mesocardiac and zygocardiac ossicles in the gastric mill of S. olivacea was dissected out and preserved in solutions and underwent a cross sectioning process. A total of 76 of wild S. olivacea ranging from 6.56 to 12.84 cm in carapace width were analysed. The growth band counts were examined for each individual and ranging from 1 to 3 band counts.

    RESULTS: A positive linear relation was observed between CW and GBC with r2 = 0.5178, p<0.01. Overall, there was a strong, positive correlation between CW and GBC. Increase in CW were correlated with increases in GBC respectively for this species.

    CONCLUSION: Therefore, the carapace width, growth band counts and body weight can be used to improve data on growth, recruitment, maturation and mortality. Thus, this study would able to improve new ageing technique and contribute greatly to improve the conservation and management of S. olivacea in Setiu Wetlands, Terengganu, Malaysia.

    Effective waste management has been greatly restricted by insufficient statistical data on the generation, processing and waste disposal. This study was undertaken in the municipality of Tehran. A total of 6,060 samples were compared by statistically comparing source generation, destination and intermediate stations. The results from these analyses showed that the average per capita waste generation in Tehran was 589 g day(-1). It was also observed that, of the total amount of waste generated in the municipality of Tehran, 73% was domestic waste and 27% was non-domestic waste. In addition, 68% of total household waste was organic waste, while 41% of non-domestic waste was organic waste. Furthermore, 61% of waste in Tehran was generated at the source, while 72% of the waste coming into the Aradkoh disposal and processing center was organic waste. The physical analysis was showed that there was no significant difference between the wastes generated in 2004 and those generated in 2009 and that there was not equal percentage of wet waste coming into the disposal center with urban service stations. This indicates that active source separation programs in metropolitan Tehran.

    The mango (Mangifer indica L.) is an important species of the family Anacardiaceae and is one of the most important crops cultivated commercially in many parts of the world. Hence, a better understanding of the phylogeny in this species is crucial as it is the basis knowledge of improving its genetic resources which is beneficial for breeding programs. Phylogenetic relationships among 13 mango cultivars from Indonesia, Malaysia and Taiwan were carried out by comparing DNA sequence data sets derived from the Internal Transcribed Spacer (ITS) region pfnuclear ribosomal DNA (nrDNA). Analysis using parsimony method showed that the cultivars were classified into three major groups. The first group composed almost Malaysian cultivars although with low bootstrap value, the second group consisted of mainly Taiwan cultivars and the last group included mostly Indonesia one. The results indicated that some cultivars have a close relationships with each other even it is originated from different countries. With regards to the relationship among these cultivars, this gives better insight for generating new cultivar.

    During the production of palm sugar, the palm sap (Arenga pinnata) is heated up to 150 degrees C. Besides the hydrolysis of carbohydrate to generate reducing sugars and degradation of amino acid, many physicochemical changes produced at all these temperatures, having a significant impact on the overall quality of palm sugar. In this study, changes in physico-chemical properties of the palm sap due to heat processing were investigated. Analysis of colour, soluble solid, pH, temperature, sugar and amino acid concentration was determinant. The results showed clearly that the heating process at these high temperatures was necessary to create an environment which was rich in essential precursors for subsequent reactions such as Maillard reaction. Chemical compounds that showed drastic changes in concentration were polar side chain amino acids especially glutamine, asparagine and arginine as well as sucrose and pH value. Other quality characteristics of palm sugar based on colour and soluble solids (Brix) shared an increase in concentration as a function of time.

    Rice is one of the most important cereal crops with great potential for biotechnology progress. In transformation method, antibiotic resistance genes are routinely used as powerful markers for selecting transformed cells from surrounding non-transformed cells. In this study, the toxicity level of hygromycin was optimized for two selected mutant rice lines, MR219 line 4 and line 9. The mature embryos were isolated and cultured on an MS medium with different hygromycin concentrations (0, 20, 40, 60, 80 and 100 mg L(-1)). Evidently, above 60 mg L(-1) was effective for callus formation and observed completely dead. Further there were tested for specific concentration (0-60). Although, 21.28% calli survived on the medium containing 45 mg L(-1) hygromycin, it seemed suitable for the identification of putative transformants. These findings indicated that a system for rice transformation in a relatively high frequency and the transgenes are stably expressed in the transgenic plants. Green shoots were regenerated from the explant under hygromycin stress. RT-PCR using hptII and gus sequence specific primer and Southern blot analysis were used to confirm the presence of the transgene and to determine the transformation efficiency for their stable integration in regenerated plants. This study demonstrated that the hygromycin resistance can be used as an effective marker for rice transformation.

    Nitrate-nitrogen leaching from agricultural areas is a major cause for groundwater pollution. Polluted groundwater with high levels of nitrate is hazardous and cause adverse health effects. Human consumption of water with elevated levels of NO3-N has been linked to the infant disorder methemoglobinemia and also to non-Hodgkin's disease lymphoma in adults. This research aims to study the temporal patterns and source apportionment of nitrate-nitrogen leaching in a paddy soil at Ladang Merdeka Ismail Mulong in Kelantan, Malaysia. The complex data matrix (128 x 16) of nitrate-nitrogen parameters was subjected to multivariate analysis mainly Principal Component Analysis (PCA) and Discriminant Analysis (DA). PCA extracted four principal components from this data set which explained 86.4% of the total variance. The most important contributors were soil physical properties confirmed using Alyuda Forecaster software (R2 = 0.98). Discriminant analysis was used to evaluate the temporal variation in soil nitrate-nitrogen on leaching process. Discriminant analysis gave four parameters (hydraulic head, evapotranspiration, rainfall and temperature) contributing more than 98% correct assignments in temporal analysis. DA allowed reduction in dimensionality of the large data set which defines the four operating parameters most efficient and economical to be monitored for temporal variations. This knowledge is important so as to protect the precious groundwater from contamination with nitrate.

    Blue swimming crab, Portunus pelagicus is widely study and research throughout the Indo-West Pacific, but little is known of its reproductive biology in Malaysia. The present study describes the fecundity, embryonic development and ovarian development stages of the P. pelagicus from Johor coastal water, Malaysia. Carapace width range of berried crabs sampled was from 9.64 to 13.32 cm, while the body weight range was from 75 to 235 g. The mean number of egg produced by females in different sizes ranged from 105443.333 +/- 35448.075 per eggs batch. Mean egg size during embryonic development at stage 1 was 0.307 +/- 0.037, while 0.386 +/- 0.039 and 0.396 +/- 0.033 for stage 2 and stage 3, respectively. Study showed that there was significant (p < 0.05) relationship between the number of eggs and carapace width/body weight. Mean diameter oocyte during ovarian development at stage 1 was 97.732 +/- 12.391 while for stage 2 was 149.516 +/- 23.287. Stage 3 showed increasingly of size with mean diameter was 158.506 +/- 27.616 and 181.013 +/- 24.339 for stage 4.

    BACKGROUND AND OBJECTIVE: Welding fume exposure has led to the respiratory problems among welders including cough, phlegm, chest illnesses, nausea and fatigue. Inadequate ventilation during welding works causes the situation to worsen. Welding fumes can cause a decrease in lung function among welders. Chronic exposure will lead to other health effects especially COPD (Chronic Obstructive Pulmonary Disease). The objective of this study is to determine the exposure of welding fumes (Cd, Fe, Pb and Zn) towards respiratory health including lung function test (FEV1, FVC, FEV1/FVC, PEFR) of workers in Lumut shipyard, Perak.

    MATERIAL AND METHODS: This research study the relationship between exposures of welding fumes towards lung function test among workers in Lumut shipyard, Perak. Lung function test was measured by spirometry among 30 welders and 31 non-welders. The concentration welding fume exposure was measured using OSHA ID-121 method. Sociodemographic data, respiratory symptoms and smoking habit data was analyzed based on the ATS 1987 questionnaire.

    RESULTS: The mean concentration for Pb in welding fumes was 2.752 mg m-3 which is above 0.5 mg m-3 PEL-TWA. The FEV1 and FVC readings showed significant different between welders and non-welders (p = 0.001). Cough and phlegm symptoms showed significant different between welders and non-welders (p = 0.001). Welders had higher prevalence in smoking habit than the non-welders. Chest illnesses symptom showed an association with the smoking habit (p = 0.01).

    CONCLUSION: There is relationship between welding fumes exposure on lung function test of workers in Lumut shipyard. Pb in welding fumes has high concentration and exceeded PEL-TWA level. The FEV1 and FVC in welders are lower than non-welder due to the fumes exposure. Welders showed higher respiratory symptoms than non-welders. Smoking habit is a contributing factor towards respiratory problem.

    Photo fermentation is a biological process that can be applied for hydrogen production. The process is environmental friendly which is operated under mild conditions using renewable resources. In order to increase yield of H2 produced by Rhodobacter sphaeroides, some experimental factors that may enhance H2 production were studied. The effect of operating parameters including agitation, aeration and light on hydrogen production using R. sphaeroides NCIMB 8253 was investigated. Rhodobacter sphaeroides NCIMB 8253 was grown in 100 mL serum bottle containing growth medium with maliec acid as the sole organic carbon source. The cultures were incubated anaerobically at 30 degrees C with tungsten lamp (100 W) as the light source (3.8 klux) and argon gas was purged for maintaining anaerobic condition. The results show that maximum hydrogen produced was higher (54.37 mL) in static culture with 69.98% of H2 in the total gas compared with shake culture (11.57 mL) with 57.86% of H2. By using static culture, H2 produced was five times higher compared with non-static in both aerobic and anaerobic condition. It was found that growth and H2 production with fluorescent lamp showed better results than growth and H2 production with tungsten light.

    This research was conducted in order to investigate rumen degradability of some factors includes; Dray Matter (DM), Organic Matter (OM), Crude Protein (CP), Acid Detergent Fiber (ADF) in three different plant protein supplements includes; soybean, canola and cottonseed meal. The experiment was carried out using in three castrated and fistulated male Zel sheep. Each feedstuff was weighed into duplicate nylon bags and incubated in each of the three rumen fistulated sheep for 0, 2, 4, 8, 16, 24 and 48 h. Results revealed that effective degradability of DM of soybean, canola and cottonseed meal were 55.8, 73.8 and 48.5%, respectively. Effective degradabilities of the CP in feedstuffs were 55.8, 62 and 48.3% for the respective feedstuffs. Effective degradabilities of the OM were 55.7, 56.4 and 47.4%, respectively. Results also showed that effective degradabilities of the ADF were 55, 56.4 and 37.6, respectively. According to the results the researchers concluded that canola and soybean were more degradable in the rumen of the sheep while cottonseed meal were less degradable and, hence resulted in higher rumen undegradable protein.

    A study on diversity and distribution of fish communities and water qualities were carried out from January 2009 to December 2010 to cover monsoon and non-monsoon at Kuantan estuary, Pahang, Malaysia. A total of 19 species of primary marine fish belong to 12 families were recorded. Out of 311 individuals the fish fauna was dominated by Ariidae followed by Lutjanidae and Lactaridae. As such Ariidae contributes 50% of the fish caught in the study area and its diversity index (H') was 0.97. A The Ariidae family consist of four (4) species; Arius maculatus, Arius sumatranus, Arius tenuispinis and Arius thalassinus. The Ariidae family can be found in all stations as they are euryhaline (highly tolerant to salinity) and this fish family are known to be a hardy estuarine catfish. Among all species in family Ariidae, Arius thalassinus was the most dominant (23%) among all species. As such collected species showed highest species diversity (0.34) followed by Arius tenuispinis (0.25) compared to other species. Arius tenuispinis alone contributed 11.90% among the samples caught from all stations. The fishes were caught and recorded highest in September-December. Pseudorhombus quinque ocellatus, Nibea soldado, Sardinella fimbriata, Toxotes jaculatrix, Dasyatis ushiei, Setipinna taty were the least dominant in the Kuantan estuary with 9.33% of total abundance. Physico-temperatures, such as temperature (22.03-30 degrees C), Conductivity (10.342.43 mS cm(-1)), TDS (0.06-26.34 mg L(-1)), salinity (0.05-29.09 ppt), DO (6.37-8.38 mg L(-1)), pH (4.97-8.03), Chl a (0.01-1.33 microg L(-1)), nitrite (0.01-0.08 mg L(-1)), nitrate (0.60-0.88 mg L(-1)), phosphate (0.24-0.40 mg L(-1)). Nevertheless, the study envisages that the water quality and fish diversity are still conducive in the Kuantan estuary. The fish diversity of Pahang estuary was high monsoon compared to non-monsoons. The station 4 (LKIM fishing boat jetty and adjacent Hospital Kuantan) is the most polluted area due to the presence of several outskirts could be alarming for the sustainable development of fish and other aquatic organisms in Kuantan estuary in the long run.

    Investigation on copepod communities in Perai river estuary was conducted from November 2005 to May 2006. Five stations were established for monthly sampling and were located from the river mouth to the upper reaches of the river. Copepod samples were collected from vertical tows using a standard zooplankton net. The Perai river estuary was slightly stratified and salinity decreases significantly from the mouth of the river towards the upper reaches of the river. A total of 28 species of copepods were recorded and comprised of 14 families, Paracalanidae, Oithonidae, Corycaeidae, Acartiidae, Calanidae, Centropagidae, Eucalanidae, Pontellidae, Pseudodiaptomidae, Tortanidae, Ectinosomatidae, Euterpinidae, Clausidiidae and Cyclopidae. A total of 10 species showed high positive affiliation towards salinity (R > 0.60), Acartia spinicauda, Euterpina acutifrons, Microsetella norvegica, Oithona nana, Oithona simplex, Paracalanus crassirostris, Paracalanus elegans, Paracalanus parvus, Pseudodiaptomus sp. and Hemicyclops sp. The copepod species Pseudodiaptomus dauglishi were negatively affiliated towards salinity (R = -0.71). The copepod assemblages classified into two distinct groups according to salinity regimes, euryhaline-polyhaline group (25 marine affiliated species) and oligohaline-mesohaline group (3 freshwater affiliated species).

    In order to propose a prospective candidate for novel complementary phytopharmaceuticals, one of Zingiberaceae family plant, Etlingeraelatior or torch ginger, was being evaluated. The aim of this review was to provide a comprehensive literature research focused on the botanical aspects, nutritional quality, phytoconstituents and pharmacological activities of E. elatior. Researches on this particular plant were conducted in Malaysia (55.5%), Indonesia (33.3%), Thailand (8.3%) and Singapore (2.7%). This review article has revealed that the most prominent pharmacological activities were anti-microbial, anti-oxidant and anti-tumor activities in consistent with the dominated levels of flavonoids, terpenoids and phenols. However, extended and integrated research should be converged towards intensive investigations concerning to isolated phytoconstituents and its bioactivities, pharmacokinetics, bioavailability, molecular mechanism of its specific pharmacological activities, safety and efficacy studies for further development.

    Organotin complexes are recognized as the biologically active compounds in inducing cancerous cells death at very low doses. To date, organotin compounds currently appear among the most potent candidates in research related to the new anticancer drugs. In this study, new organotin(IV) N-butyl-N-phenyldithiocarbamate compounds have been successfully synthesized between the reaction of N-butylaniline amine with organotin(IV) chloride in 1:2/1:1 molar ratio. All compounds were characterized using the elemental analysis, FT-IR and NMR spectroscopy. The single crystal structure was determined by X-ray single crystal analysis. The elemental analysis showed good agreement with the suggested formula (C4H9)2Sn[S2CN(C4H9)(C6H5)]2 (Compound 1 and 2), (C6H5)2Sn[S2CN(C4H9)(C6H5)]2 (Compound 3) and (C6H5)3Sn[S2CN(C4H9)(C6H5)] (Compound 4). The important infrared absorbance peaks, v (C = N) and v(C = S) were detected in range between 1457-1489 cm(-1) and 951-996 cm(-1), respectively. The chemical shift of carbon in NCS2 group obtained from 13C NMR was found in range 198.86-203.53 ppm. The crystal structure of compound 4 showed that the dithiocarbamate ligand coordinates in a monodentate fashion. It crystallized in monoclinic P2(1)/n space group with the crystal cell parameter: a = 10.0488(1) angstroms, b = 18.0008(2) angstroms, c = 15.2054(2) angstroms, beta = 102.442(1) degrees and R = 0.044. The cytotoxicity (IC50) of these compounds against Jurkat E6.1 and K-562 leukemia cells were in the range between 0.4-0.8 and 1.8-5.3 microM, respectively as assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazholium bromide (MTT) assay. In conclusion, our study demonstrate that all compounds showed potent cytotoxicity towards both cell lines tested with the triphenyltin(IV) compound displayed the greatest effect.

    Muscle and feather in tissue of 40 juveniles and 40 adult green-lipped mussel Perna veridis (L.) collected from Muar Estuary, Johor were analyzed for copper (Cu), cadmium (Cd), lead (Pb) and zinc (Zn) concentration using a fast and sensitive Inductively Coupled Plasma Mass Spectrometer (ICP-MS). In this study, the average concentration of Cu was 8.96 microg g(-1) dry weights, Cd with 0.58 microg g(-1) dry weight, Pb averaging 2.28 microg g(-1) dry weights and Zn averaged to 86.73 microg g(-1) dry weight. The highest accumulation of metal studied was found in feather sample compared to the muscle. The positive relationship of Cu, Cd, Pb and Zn with P. virdis length suggesting that the accumulation of these metals were formed in the mussel. In all cases, metal levels found were lower than the guideline of international standards of reference and the examined bivalve were not associated with enhanced metal content in their tissues and were safe within the limits for human consumption.

    Muscle, stomach and gill from four dominant fish species, Mytus nemurus, Pristolepis fasciata, Ompok bimaculatus and Osteochilus hasseltii, caught from Bebar peat swamp forest river were analyzed for mercury (Hg). The concentration of Hg was measured with a fast and sensitive Flow Injector Mercury Spectrometer (FIMS). The average Hg concentration of all species caught was 0.169 microg g(-1) dry weights, lower than a limit for human consumption recommended by the World Health Organization, 0.5 microg g(-1) dry weights. The mean concentration of Hg was relatively high in stomach (0.28 +/- 0.12 microg g(-1) dry weights) followed by gill (0.17 +/- 0.06 microg g(-1) dry weights) and lowest in muscle (0.05 +/- 0.02 microg g(-1) dry weights). The positive relationship of Hg with fish length and weight suggesting that the accumulation of Hg were formed in the fish.

    An effort to analyze selected heavy metal accumulation by the razor clam (Solen brevis) from Tanjung Lumpur was conducted on January to April 2010. A total of fifty individuals of Razor clam Solen brevis were sampled and metals such as Iron (Fe), Zinc (Zn), Copper (Cu), Manganese (Mn), Lead (Pb) and Cadmium (Cd) Concentrations were determined using Inductively Coupled Plasma Mass Spectrometry (ICP-MS). Among the metals Fe occurred in elevated concentration in the soft tissue of razor clam followed by Zn. Cd was found to be in least concentration in the sample. Mean concentration of Fe, Zn, Mn, Cu, Cd and Pb in the soft tissue were 415.2 +/- 56.52, 87.74 +/- 11.85, 18.71 +/- 2.10, 8.64 +/- 1.75, 0.67 +/- 0.29 and 1.61 +/- 0.45 microg g(-1) dw, respectively indicating that the bioaccumulation of essential metals in the soft tissue was greater than the non essential heavy metals. Metal accumulation in the soft tissue of razor clam followed Fe > Zn > Mn > Cu > Pb > Cd order in present study. The observed concentration of acute toxicity of metals in Solen brevis (Family: Solenidae) from Tanjung Lumpur Coastal waters was lower than the permissible limit recommended by National and international standards proved that this species could be utilized for human consumption.
