    A study was conducted at five selected rivers around Bukit Merah Reservoir,
    Perak, Malaysia for eight weeks in order to determine the fish diversity and distribution. A
    total of 28 species comprised of 9 families were identified. The study depicted that there
    were significant changes to the fish composition when compared to previous study which
    had captured 36 species due to different areas covered and different types of sampling
    gear used between both studies.

    Alpinia conchigera Griff. is a plant species from the family Zingiberaceae. Coloquially known as wild ginger, Alpinia conchigera Griff. is used as food condiment and for traditional treatment of skin diseases. Isolation studies to identify bioactive compounds of rhizomes of Alpinia conchigera yielded seven compounds; 1'S-1'-acetoxychavicol acetate (1), trans-p-coumaryl diacetate (2), p-hydroxycinnamyl acetate (3), 1'S-1'-hydroxychavicol acetate (4) p-hydroxybenzaldehyde (5), stigmasterol (6) and β-sitosterol (7). Compounds 1, 2 and 5 were evaluated for antimicrobial activity against methicillin-resistant Staphylococcus aureus (MRSA). Among the compounds tested, Compound 1 showed good antimicrobial activity against the strain of MRSA with minimum inhibition concentration (MIC) value of 0.5 mg/mL. Meanwhile, Compounds 2 and 5 exhibited moderate activity with MIC value between 1.0 and 2.0 mg/mL. These findings indicate antimicrobial potential of 1'S-1'-acetoxychavicol acetate (1), compound derived from rhizome of Alpinia conchigera Griff. against MRSA, which warrant further investigation.

    An early step in the Agrobacterium-mediated transformation of Phalaenopsis violacea orchid was investigated to elucidate the plant-bacterium interaction. Directed movement in response to chemical attractants is of crucial importance to Agrobacterium tumefaciens strains. Chemotaxis of A. tumefaciens strains (EHA 101 and 105) towards wounded orchid tissues has been studied by using swarm agar plates. The results obtained indicate a minor role for chemotaxis in determining host specificity and suggest that it could not be responsible for the absence of tumourigenesis in P. violacea orchid under natural conditions. The spectrometric GUS and green fluorescent protein (GFP) assays provided information on the amount of inoculated A. tumefaciens that effectively bound to various orchid tissues. It can be concluded that, at least during the two early steps of interaction, A. tumefaciens appears to be compatible with P. violacea, indicating a potential basis for genetic transformation.

    The (R)-3-hydroxyacyl-ACP-CoA transferase catalyses the conversion of (R)-3-hydroxyacyl-ACP to (R)-3-hydroxyacyl-CoA derivatives, which serves as the ultimate precursor for polyhydroxyalkanoate (PHA) polymerisation from unrelated substrates in pseudomonads. PhaG was found to be responsible for channelling precursors for polyhydroxyalkanoate (PHA) synthase from a de novo fatty acid biosynthesis pathway when cultured on carbohydrates, such as glucose or gluconate. The phaG gene was cloned from Pseudomonas sp. USM 4-55 using a homologous probe. The gene was located in a 3660 bp Sal I fragment (GenBank accession number EU305558). The open reading frame (ORF) was 885 bp long and encoded a 295 amino acid protein. The predicted molecular weight was 33251 Da, and it showed a 62% identity to the PhaG of Pseudomonas aeruginosa. The function of the cloned phaG of Pseudomonas sp. USM 4-55 was confirmed by complementation studies. Plasmid pBCS39, which harboured the 3660 bp Sal I fragment, was found to complement the PhaG-mutant heterologous host cell, Pseudomonas putida PhaGN-21. P. putida PhaGN-21, which harboured pBCS39, accumulated PHA that accounted for up to 18% of its cellular dry weight (CDW). P. putida PhaGN-21, which harboured the vector alone (PBBR1MCS-2), accumulated only 0.6% CDW of PHA.

    We report on the cloning of the lipase gene from Bacillus licheniformis IBRLCHS2
    and the expression of the recombinant lipase. DNA sequencing analysis of the
    cloned lipase gene showed that it shares 99% identity with the lipase gene from
    B. licheniformis ATCC 14580 and belongs to subfamily 1.4 of true lipases based on amino
    acid sequence alignment of various Bacillus lipases. The 612 bp lipase gene was then
    cloned into the pET-15b(+) expression vector and the construct was transformed into
    E. coli BL21 (DE3) for bulk expression of the lipase. Expression was analysed by SDSPAGE
    where the lipase was found to have a molecular weight of about 23 kDa.

    The occurrence of major outbreaks of dengue, and other vector borne diseases such as chikungunya and zika in tropical and subtropical regions has rendered control of the diseases a top-priority for many affected countries including Malaysia. Control of the mosquito vectors Aedes aegypti and Aedes albopictus through the reduction of breeding sites and the application of insecticides to kill immature forms and adults are the main control efforts to combat these diseases. The present study describes the association between Ae. albopictus and Ae. aegypti in shared breeding sites. This study is important given that any measure taken against one species may affect the other. A yearlong larval survey was conducted in four dengue endemic areas of Penang Island. Sorenson's coefficient index indicated that no association between number of the immatures of the two species regardless of container size and study location. Therefore, the mean number Ae. albopictus immature was not decreased in the presence of Ae. aegypti in shared breeding container. However Ae. aegypti appeared to prefer breeding in habitats not occupied by Ae. albopictus, the two species sharing breeding sites only where available containers were limited. In control efforts, eliminating the preferred breeding containers for one species might not affect or reduce the population of the other species.

    Land reclamation in Penang began two decades ago and is still rampant with
    large reclamation projects planned to be executed in the near future. The present study
    provides the first information on effects of land reclamation in Penang towards the coastal
    macroinvertebrates. This study assessed the abundance, diversity, and evenness of
    coastal invertebrates assumed to be the foremost affected when land is reclaimed. Three
    kinds of areas were focused on: reclaimed, unclaimed (adjacent to reclaimed), and
    undisturbed. A total of 53 species of macroinvertebrates from 10 classes (Gastropoda,
    Bivalvia, Polychaeta, Malacostraca, Maxillopoda, Echinoidea, Polyplacophora,
    Branchiopoda, Scaphopoda, and Holothuroidea) were sampled. Reclaimed areas were
    moderately rich in species averaging 11 species compared to 7 species in adjacent and
    14 in undisturbed areas. Species richness was the highest in Teluk Aling (an undisturbed
    area) with 22 species, and was the lowest in Gurney Drive (an adjacent area) with 2
    species. The average species diversity and evenness on reclaimed land was the lowest
    with values of 1.9974 and 0.5787, respectively. The diversity was higher by 5.07% in
    adjacent areas and by 22.92% in undisturbed areas compared to reclaimed areas.
    Species evenness was 29.75% higher in unreclaimed areas and 17.87% higher in
    undisturbed areas compared to reclaimed areas. Land reclamation reduces species
    diversity and evenness, and to a lesser extent, species richness.

    The interaction between plants and insects is dynamic, and may favour either the plant or the insect. Plant chemicals are deeply implicated in this relationship and influence insect behaviour. Here, we investigated the oviposition behaviour response of Culex quinquefasciatus mosquitoes based on the colour cues produced by Ipomoea cairica leaves extract. In this study, two sets of oviposition choice experiments were conducted: (1) single solution in a cage; and (2) multiple concentration solutions in a cage. In the single solution experiment, only 1 available oviposition site was offered to 5 gravid females and in the multiple concentration tests, 4 available oviposition sites were offered to 20 gravid females. The tested concentrations were set up at 100 mL of: (1) control (distilled water only); (2) 50 ppm; (3) 150 ppm; and (4) 300 ppm of I. cairica plant extracts. The highest concentration of 300 ppm appeared to show the highest intensity with the darkest colour followed by 150 ppm and 50 ppm concentrations. More gravid females were found drowned in the highest concentration, 300 ppm of acethonilic leaves extract, compared to 150 ppm and 50 ppm of the tested extract. No eggs were found in all tested solutions. The studied extract was found to effectively attract gravid Cx. quinquefasciatus females and subsequently cause mortality and inhibit egg deposition. The interference caused by the acethonilic extract of I. cairica on the oviposition activity of Cx. quinquefasciatus can result in better control of the vector insect.

    In present study, Water Quality Index (WQI) has been assessed of the Rawal Lake which is a major source of drinking water for people in the Federal Capital, Islamabad, and its adjacent city Rawalpindi in Pakistan. For this, the principal component analysis (PCA) and WQI were applied as an integrated approach to quantitatively explore difference based on spatial variation in 11 water quality parameters of the five major feeding tributaries of the Rawal Lake, Pakistan. The results of temperature in water, total dissolved solids, pH, electrical conductivity, chlorides and sulfates were well within the allowable World Health Organisation's (WHO) limits. However, the heavy metals like cadmium and lead were above permissible limits by the WHO in tributaries of Bari Imam and Rumli. Moreover, this has been proven by the Pearson correlation which suggested strong positive correlation (0.910*) between lead and cadmium. The results of present study were subjected to statistical analysis, i.e., PCA which gave three major factors contributing 96.5% of the total variance. For factor 1, pH, TDS, alkalinity, chlorides, sulfates and zinc have highest factor loading values (>0.60) and presented that these parameters were among the most significant parameters of first factor. As per the WQI results, the water was categorised in two major classes indicating that water of Bari Imam and Rumli is highly contaminated with heavy metals and totally unsuitable for drinking purposes. Based on the results of the present study, it is suggested to make heavy metals consideration as an integrated component in future planning for maintaining water quality of the Rawal Lake and its tributaries.

    Pembangunan yang semakin pesat di Malaysia telah meningkatkan bilangan aktiviti antropogenik, sekaligus menyebabkan kemerosotan alam sekitar. Ini menunjukkan bahawa perlunya inventori hidupan liar dan sumber alam sekitar dijalankan di dalam kawasan hutan yang sedia ada, untuk mempromosikan peningkatan yang seimbang dalam pelan pemuliharaan dan pengurusan yang sedia ada, terutamanya untuk haiwan yang terancam seperti spesies mamalia kecil. Kajian mamalia kecil telah dijalankan di Tasik Bera, Hutan Simpan Ulu Gombak, Pusat Konservasi Hidupan Liar Sungkai, Pulau Pinang, dan Taman Negeri Wang Kelian. Harp trap, mist net, cage trap dan pitfall trap telah dipasang untuk kajian. Spesies Rhinolophus affinis (N = 61) merupakan spesies yang paling banyak ditangkap, diikuti dengan R. Lepidus (N = 27). Manakala untuk mamalia kecil tidak terbang, species Leopoldamys sabanus merupakan spesies paling banyak (N = 33) diikuti dengan Maxomys rajah (N = 25) dan Tupaia glis (N = 22). Dua spesies cencurut, Suncus etruscus dan Crocidura monticola masing-masing telah ditangkap dengan jumlah satu individu bagi setiap satu spesies. Hutan Simpan Ulu Gombak merekodkan kepelbagaian spesies yang paling tinggi (H’ = 2.754), manakala Pulau Pinang merekodkan nilai yang paling rendah (H’ = 2.245). Senarai mamalia kecil yang telah direkodkan melalui kajian ini merupakan maklumat yang signifikan bagi tujuan pemantauan dan konservasi biodiversiti.

    Aquaculture is still in early development in Madagascar and Tanzania, while in Indonesia, aquaculture has a long history. In Madagascar, villagers are farming seaweed and sea cucumbers, as part of small-scale community-based aquaculture (CBA). They followed a contractual model between a private farming company and farmers. Local non-governmental organisations (NGOs) and public institutions in Madagascar jointly strive to reverse the trend of ongoing anthropogenic coastal degradation. In Tanzania, the cultivation of red seaweeds has been established for over 30 years, with declining production attributed to climate change. While shrimp farming still involves, to some extent, clearing of mangroves in Tanzania, seaweed culture has only mild impact on coastal ecosystems. Farming areas provide shelter and habitat for juvenile fish, crabs and other organisms. Therefore, NGOs ask for support to improve culture methods. Various problems and shortcomings in Indonesia have been clearly identified, including issues related to new aquaculture areas, pollutants, emerging diseases, insufficient broodstock and fry supply, as well as a lack of technology and manpower. To address these challenges and ensure the growth of aquaculture production, the government has implemented national policies and established training and broodstock centers throughout the country. In Madagascar, the CBA programme stands out as a success story and can serve as a template for other coastal regions and countries. In Tanzania, the adoption of CBA model for co-culture could be the future. In Indonesia, due to a very long coastlines and complicated legislation, IMTA seems to be particularly suitable, as successfully tested in model regions.

    Amyema are epiphytic hemiparasitic plants on different types of woody host plants and are abundant in temperate, subtropical, and tropical regions. In Marilog Forest Reserve, Southern Philippines, two Philippine endemic species of Amyema were recorded, viz., Amyema curranii (Merr.) Danser and A. seriata (Merr.) Barlow. In this study, these two species were compared and examined for their morphology and anatomy. Data revealed that the two Amyema species are morphologically distinct, with A. curranii having lanceolate leaves, pink flowers, and red fruits, whereas A. seriata has obovate leaves, red flowers, and yellow fruits. For the morpho-anatomy, A. curranii has a single-layered epidermis, paracytic stomata, collateral open vascular bundles, the Eustele type of stele with pith at the center, and the inferior free central type with a hairy ovary wall. Meanwhile, A. seriata has a pinkish, single-layered epidermis, paracytic stomata, collateral open vascular bundles, a eustele type of stele with the presence of pith at the center, and an inferior free central ovary type. As a result, employing these species' gross morphology and anatomy could scrutinise future evaluations and taxonomic placements.

    This study was conducted to examine the variabilities in the chronology of vegetative and reproductive development of weedy rice (Oryza spp.) in comparison with commercial varieties. Data at different growth stages of 14 weedy rice morphotypes and 4 commercial rice varieties were recorded and analysed. Plant height of all weedy rice morphotypes were observed to be significantly higher compared to the commercial varieties at every growth stages; increase in height was between 10-37 cm for weedy rice morphotype, for every 2 weeks. Initial tillering ability at 14 days after planting (DAP) was higher in weedy morphotypes, however all the commercial rice varieties produced significantly higher number of tillers throughout the rest of the vegetative phases. Correlation between plant height and tiller number detected that taller plants produce fewer tillers than shorter plants. Higher leaf area index (LAI) of all weedy morphotypes except PWR01 at early growth stages indicated the vigorous growth of the morphotypes. Weedy rice morphotypes showed a wide range of anthesis and maturity duration. Accessions from the same weedy rice morphotypes were more heterogeneous in the flowering, anthesis and maturity period than the commercial varieties. These traits enables identification of weedy rice morphotypes at their different growth stages in the field.

    The objective of this study was to describe the structure and histochemistry of the systemic organs in the female marine water-strider Asclepios annandalei from Pranburi river estuary, Thailand. Results from this study revealed for the first time that the integumentary system of this species consisted of three layers including epicuticle, exocuticle and endocuticle. The muscular system apparently contained only skeletal muscle along the body. In the urinary system, we observed well-developed Malpighian tubules, each of which was covered with the simple cuboidal epitheliums. These epitheliums also contained the secretory granules that were reacted positively with periodic acid Schiff (PAS). The digestive system of this species was composed of three distinct parts including foregut, midgut and hindgut. The respiratory system was composed of the respiratory organ, which was rarely found near the integument system. This organ was lined with a simple squamous epithelium. Two regions of nervous system, i.e., frontal ganglion connected to the eye structure and ventral nerve cord, were found. Each ganglion basically consisted of two layers, outer cortex and inner medullae. The outer cortex contained three types of cells, including neurosecretory cells, neuroglial cells and neurons. The cytoplasmic inclusion of neurosecretory cells contained secretory granules, which reacted positively with PAS, indicating the presence the glycoprotein. The neuroglia and neuron were also observed in the inner medullae layer. The female reproductive system (the ovarian structure, the reproductive tract and the accessory organ) of this gerrid species was seen under histological sections. The well-developed integument system and Malpighian tubule as well as the abundant respiratory organ is a characteristic of this species, which might be useful for the adaption to the estuarine condition.

    Pachychilidae is one of the freshwater gastropod family which was previously known under the Potamididae and Thiaridae families. Studies on freshwater gastropods especially on conchcology examinantions are still inadequate compared to marine gastropods. Morphological and morphometric studies of gastropods are practically used to identify and differentiate between species and necessary to complement molecular studies due to its low cost and tolerable resolving power of discrimination. The aim of the current study is to provide information on morphological and morphometric characteristics of Pachychilidae in Bintulu, Sarawak stream. A total of 20 individuals from each species of Sulcospira testudinaria, Sulcospira schmidti, Brotia siamensis, and Tylomelania sp. from Pachychilidae familiy were collected at three different sites from a small stream within the Bintulu area. Fourteen measurement of shell morphometrics were converted into proportioned ratios and analysed for univariate and multivariate analysis. Three shell morphometric (Aperture width, AW; Whorl width, WW2; and, Interior anterior length, AINL) of Pachychilidae indicated significant differences (P < 0.05) between species. However, multivariate analysis revealed that these shell morphometrics are pre-eminent factors to discriminate genus Sulcospira, Brotia and Tylomelania, as well as between Sulcospira species. This current study also suggests that these three characteristics are unique to Sulcospira species due to strong distinction among species. Findings on these three characteristics are significant for Sulcospira spp. as this study is the first shell morphometric report on the Pachychilidae species in Sarawak.

    Condition index, reproduction and feeding of three non-obligatory riverine Mekong cyprinids namely Hampala dispar, Hampala macrolepidota and Osteochilus vittatus were examined. The samples were from the Nam Ngiep (NN) River and Bueng Khong Long (BKL) Swamp, which are the representative of the lotic- and lentic-environments, respectively. These two habitats lay in the same geographical area but on the opposite banks of the Mekong mainstream. The samplings were conducted between May 2017 and April 2018. There were 365 H. dispar, 259 H. macrolepidota and 298 O. vittatus samples in this study. The condition index of all three species were beyond 90% implying they can live well in both lotic and lentic environments. Reproductions of all three species were taken place in both environments with two peaks at the onset and end of rainy season. The samples from BKL showed early maturation than NN samples in all three (3) species. Feeding plasticity, though dominant by insects, was observed in Hampala spp., while O. vittatus can utilise any available detritus in both environments. Results clearly show that all the three selected non-obligatory riverine fish species can live very well in either lotic or lentic environments and imply that they can adjust themselves to reservoir environment.

    Simian betaretrovirus serotype-2 (SRV-2) is an important pathogenic agent in Asian macaques. It is a potential confounding variable in biomedical research. SRV-2 also provides a valuable viral model compared to other retroviruses which can be used for understanding many aspects of retroviral-host interactions and immunosuppression, infection mechanism, retroviral structure, antiretroviral and vaccine development. In this study, we isolated the gene encoding reverse transcriptase enzyme (RT) of SRV-2 that infected Indonesian cynomolgus monkey (Mf ET1006) and predicted the three dimensional structure model using the iterative threading assembly refinement (I-TASSER) computational programme. This SRV-2 RT Mf ET1006 consisted of 547 amino acids at nucleotide position 3284-4925 of whole genome SRV-2. The polymerase active site located in the finger/palm subdomain characterised by three conserved catalytic aspartates (Asp90, Asp165, Asp166), and has a highly conserved YMDD motif as Tyr163, Met164, Asp165 and Asp166. We estimated that this SRV-2 RT Mf ET1006 structure has the accuracy of template modelling score (TM-score 0.90 ± 0.06) and root mean square deviation (RMSD) 4.7 ± 3.1Å, indicating that this model can be trusted and the accuracy can be seen from the appearance of protein folding in tertiary structure. The superpositionings between SRV-2 RT Mf ET1006 and Human Immunodeficiency Virus-1 (HIV-1) RT were performed to predict the structural in details and to optimise the best fits for illustrations. This SRV-2 RT Mf ET1006 structure model has the highest homology to HIV-1 RT (2B6A.pdb) with estimated accuracy at TM-score 0.911, RMSD 1.85 Å, and coverage of 0.953. This preliminary study of SRV-2 RT Mf ET1006 structure modelling is intriguing and provide some information to explore the molecular characteristic and biochemical mechanism of this enzyme.

    Increased consumption of xylose-glucose and yeast tolerance to lignocellulosic hydrolysate are the keys to the success of second-generation bioethanol production. Candida tropicalis KBKTI 10.5.1 is a new isolated strain that has the ability to ferment xylose. In contrast to Saccharomyces cerevisiae DBY1 which only can produce ethanol from glucose fermentation. The research objective is the application of the genome shuffling method to increase the performance of ethanol production using lignocellulosic hydrolysate. Mutants were selected on xylose and glucose substrates separately and using random amplified polymorphic DNA (RAPD) analysis. The ethanol production using lignocellulosic hydrolysate by parents and mutants was evaluated using a batch fermentation system. Concentrations of ethanol, residual sugars, and by-products such as glycerol, lactate and acetate were measured using HPLC machine equipped with Hiplex H for carbohydrate column and a refraction index detector (RID). Ethanol produced by Fcs1 and Fcs4 mutants on acid hydrolysate increased by 26.58% and 24.17% from parent DBY1, by 14.94% and 21.84% from parent KBKTI 10.5.1. In contrast to the increase in ethanol production on alkaline hydrolysate, Fcs1 and Fcs4 mutants only experienced an increase in ethanol production by 1.35% from the parent KBKTI 10.5.1. Ethanol productivity by Fcs1 and Fcs4 mutants on acid hydrolysate reached 0.042 g/L/h and 0.044 g/L/h. The recombination of the genomes of different yeast species resulted in novel yeast strains that improved resistance performance and ethanol production on lignocellulosic hydrolysates.

    The pteridophyte flora of Langkawi Archipelago consists of 130 species, 1 subspecies and 12 varieties in 68 genera and 27 families. This value represents 22.1% of the 647 taxa at the species level and below reported for Peninsular Malaysia. Of the 143 recorded taxa of pteridophytes at the species level and below, 8 species in 2 genera and 2 families are lycophytes and the other 135 taxa in 66 genera and 25 families are monilophytes or ferns.
