The interaction between plants and insects is dynamic, and may favour either the plant or the insect. Plant chemicals are deeply implicated in this relationship and influence insect behaviour. Here, we investigated the oviposition behaviour response of Culex quinquefasciatus mosquitoes based on the colour cues produced by Ipomoea cairica leaves extract. In this study, two sets of oviposition choice experiments were conducted: (1) single solution in a cage; and (2) multiple concentration solutions in a cage. In the single solution experiment, only 1 available oviposition site was offered to 5 gravid females and in the multiple concentration tests, 4 available oviposition sites were offered to 20 gravid females. The tested concentrations were set up at 100 mL of: (1) control (distilled water only); (2) 50 ppm; (3) 150 ppm; and (4) 300 ppm of I. cairica plant extracts. The highest concentration of 300 ppm appeared to show the highest intensity with the darkest colour followed by 150 ppm and 50 ppm concentrations. More gravid females were found drowned in the highest concentration, 300 ppm of acethonilic leaves extract, compared to 150 ppm and 50 ppm of the tested extract. No eggs were found in all tested solutions. The studied extract was found to effectively attract gravid Cx. quinquefasciatus females and subsequently cause mortality and inhibit egg deposition. The interference caused by the acethonilic extract of I. cairica on the oviposition activity of Cx. quinquefasciatus can result in better control of the vector insect.
Probiotics are live microorganisms that can be found in fermented foods and cultured milk, and are widely used for the preparation of infant food. They are well-known as "health friendly bacteria", which exhibit various health beneficial properties such as prevention of bowel diseases, improving the immune system, for lactose intolerance and intestinal microbial balance, exhibiting antihypercholesterolemic and antihypertensive effects, alleviation of postmenopausal disorders, and reducing traveller's diarrhoea. Recent studies have also been focused on their uses in treating skin and oral diseases. In addition to that, modulation of the gut-brain by probiotics has been suggested as a novel therapeutic solution for anxiety and depression. Thus, this review discusses on the current probiotics-based products in Malaysia, criteria for selection of probiotics, and evidences obtained from past studies on how probiotics have been used in preventing intestinal disorders via improving the immune system, acting as an antihypercholesterolemic factor, improving oral and dermal health, and performing as anti-anxiety and anti-depressive agents.
Intestinal parasitic infections are one of the most common causes of human diseases that result in serious health and economic issues in many developing and developed countries. Raw vegetables and fruits play an important role in transmitting parasites to humans. Hence, the aim of this study was to investigate the parasitological contamination of select commonly consumed local leafy vegetables and fruits in Kuantan, Malaysia. One kilogram of locally consumed raw vegetables and fruits were collected randomly from the Kuantan wet market (Pasar Tani) during the monsoon season (November 2014-January 2015) and the dry season (February 2015-April 2015). A standard wet mount procedure and modified Ziehl-Neelsen staining were used for the detection of parasites. In the present study, the examination of vegetables revealed five different parasite species. The vegetable samples collected from Kuantan's wet market were positive for both helminthes and protozoa. However, the fruits samples were negative for parasitic contamination. Pegaga was the most contaminated leafy vegetable in this study, and Strongyloides was the parasite found most frequently. Furthermore, there was a high diversity in the type of parasites observed during the dry season compared to the monsoon season. Therefore, further action should be taken to reduce the occurrence of parasitic contamination in vegetables by implementing the principles of good agricultural practice and improving water treatment efficacy.
Mangroves support diverse macroalgal assemblages as epibionts on their roots and tree trunks. These algae provide nutrients to the primary consumers in the aquatic food web and have been reported to be substantial contributors to marine ecosystems. The species diversity, biomass, and habitat characteristics of mangrove macroalgae were investigated at three stations in the Sibuti mangrove estuary, Sarawak, Malaysia, from November 2012 to October 2013. Three groups of macroalgae were recorded and were found to be growing on mangrove prop roots, namely Rhodophyta (Caloglossa ogasawaraensis, Caloglossa adhaerens, Caloglossa stipitata, Bostrychia anomala, and Hypnea sp.), Chlorophyta (Chaetomorpha minima and Chaetomorpha sp.), and Phaeophyta (Dictyota sp.). The biomass of macroalgae was not influenced (p>0.05) by the season in this mangrove forest habitat. The macroalgal species Hypnea sp. contributed the highest biomass at both Station 1 (210.56 mg/cm(2)) and Station 2 (141.72 mg/cm(2)), while the highest biomass was contributed by B. anomala (185.89 mg/cm(2)) at Station 3. This study shows that the species distribution and assemblages of mangrove macroalgae were influenced by environmental parameters such as water nutrients, dissolved solids, and salinity in the estuarine mangrove habitats of Sibuti, Sarawak.
Plants have been a good source of therapeutic agents for thousands of years;
an impressive number of modern drugs used for treating human diseases are derived from
natural sources. The Theobroma cacao tree, or cocoa, has recently garnered increasing
attention and become the subject of research due to its antioxidant properties, which are
related to potential anti-cancer effects. In the past few years, identifying and developing
active compounds or extracts from the cocoa bean that might exert anti-cancer effects
have become an important area of health- and biomedicine-related research. This review
provides an updated overview of T. cacao in terms of its potential anti-cancer compounds
and their extraction, in vitro bioassay, purification, and identification. This article also
discusses the advantages and disadvantages of the techniques described and reviews the
processes for future perspectives of analytical methods from the viewpoint of anti-cancer
compound discovery.
This study analyses the sampling performance of three benthic sampling tools
commonly used to collect freshwater macroinvertebrates. Efficiency of qualitative D-frame
and square aquatic nets were compared to a quantitative Surber sampler in tropical
Malaysian streams. The abundance and diversity of macroinvertebrates collected using
each tool evaluated along with their relative variations (RVs). Each tool was used to
sample macroinvertebrates from three streams draining different areas: a vegetable farm,
a tea plantation and a forest reserve. High macroinvertebrate diversities were recorded using the square net and Surber sampler at the forested stream site; however, very low
species abundance was recorded by the Surber sampler. Relatively large variations in the
Surber sampler collections (RVs of 36% and 28%) were observed for the vegetable farm
and tea plantation streams, respectively. Of the three sampling methods, the square net
was the most efficient, collecting a greater diversity of macroinvertebrate taxa and a
greater number of specimens (i.e., abundance) overall, particularly from the vegetable
farm and the tea plantation streams (RV
Endophytic fungi inhabit apparently healthy plant tissues and are prevalent in
terrestrial plants, especially root tissues, which harbour a wide assemblage of fungal
endophytes. Therefore, this study focused on the isolation and characterisation of
endophytic fungi from the roots of wild banana (Musa acuminata). A total of 31 isolates of
endophytic fungi were isolated from 80 root fragments. The endophytic fungi were initially
sorted according to morphological characteristics and identified using the sequences of
the translation elongation factor-1α (TEF-1α) gene of Fusarium spp. and the Internal
Transcribed Spacer (ITS) regions of other fungi. The most common fungal isolates were
species of the genus Fusarium, which were identified as F. proliferatum, Fusarium sp.,
F. solani species complex, and F. oxysporum. Other isolated endophytic fungi included
Curvularia lunata, Trichoderma atroviride, Calonectria gracilis, Rhizoctonia solani,
Bionectria ochroleuca, and Stromatoneurospora phoenix (Xylariceae). Several of the
fungal genera, such as Fusarium, Trichoderma, Rhizoctonia, and Xylariceae, are among
the common fungal endophytes reported in plants. This study showed that the roots of wild
banana harbour a diverse group of endophytic fungi.
Genetic engineering is a powerful tool for the improvement of plant traits. Despite reported successes in the plant kingdom, this technology has barely scratched the surface of the Melastomataceae family. Limited studies have led to some optimisation of parameters known to affect the transformation efficiency of these plants. The major finding of this study was to optimise the presence of selected enhancers [e.g., monosaccharides (D-glucose, D-galactose and D-fructose), tyrosine, aluminium chloride (AICI3) and ascorbic
acid] to improve the transformation efficiency of Tibouchina semidecandra. Agrobacterium tumefaciens strain LBA4404 harbouring the disarmed plasmid pCAMBIA1304 was used to transform shoots and nodes of T. semidecandra. Different concentrations of the transformation enhancers were tested by using green fluorescent protein (GFP) as a reporter. The results obtained were based on the percentage of GFP expression, which was observed 14 days post-transformation. A combination of 120 µM galactose and 100
µM tyrosine supplemented with 600 µM AICI3 in the presence of 15 mg/l ascorbic acid gave the highest percentage of positive transformants for T. semidecandra shoots. Whereas 60 µM galactose and 50 µM tyrosine with 200 µM AICI3 in the presence of 15 mg/l ascorbic acid was optimum for T. semidecandra nodes. The presence of the hygromycin phosphotransferase II (hptII) transgene in the genomic DNA of putative
T. semidecandra transformants was verified by PCR amplification with specific primers.
Sixty scavenging chickens from villages in Penang were examined for their state of nutrition or body condition and the presence of helminthic parasites. The majority of the chickens were of poor body condition. More than 66.7% (females) and 73.3% (males) belonged to the ‘below normal’ category of general body condition. A total of eight different helminth species were recovered from the trachea and the gastrointestinal tract of the various chickens. Four of the species were nematodes and the rest were cestodes. No trematodes were recovered from the chickens. The highest mean worm burden was recorded for the cestode Raillietina echinobothrida while the lowest was the nematode, Ascaridia galli.
Heavy metal accumulation and depuration may alter the effectiveness of Meretrix meretrix as a biomonitoring organism for water quality assessment. Therefore, this study was conducted to evaluate the effects of heavy metal accumulation and depuration on M. meretrix, by immersing it in Copper (Cu), Zinc (Zn), and Lead (Pb)
solutions under laboratory conditions. The results showed that M. meretrix is able to accumulate Cu, Zn, and Pb at the rate of 0.99, 21.80, and 0.57 μg/g per day, respectively, and depurates at the rate of 0.42, 23.55, and 1.01 μg/g per day, respectively. These results indicate that M. meretrix could be effectively used as a biomonitoring organism for Cu because the accumulation rate is significantly (p ≤ 0.05) higher than the depuration rate. However, this was not the case for Zn because the accumulation rate was almost similar to the depuration rate, while for Pb, accumulation or depuration did not occur in M. meretrix.
An early step in the Agrobacterium-mediated transformation of Phalaenopsis violacea orchid was investigated to elucidate the plant-bacterium interaction. Directed movement in response to chemical attractants is of crucial importance to Agrobacterium tumefaciens strains. Chemotaxis of A. tumefaciens strains (EHA 101 and 105) towards wounded orchid tissues has been studied by using swarm agar plates. The results obtained indicate a minor role for chemotaxis in determining host specificity and suggest that it could not be responsible for the absence of tumourigenesis in P. violacea orchid under natural conditions. The spectrometric GUS and green fluorescent protein (GFP) assays provided information on the amount of inoculated A. tumefaciens that effectively bound to various orchid tissues. It can be concluded that, at least during the two early steps of interaction, A. tumefaciens appears to be compatible with P. violacea, indicating a potential basis for genetic transformation.
A new species of small, insular, forest floor skink, Sphenomorphus perhentianensis sp. nov., is described from Pulau Perhentian Besar of the Perhentian Archipelago, Peninsular Malaysia. This species is differentiated from all other 36 Sundaland species of Sphenomorphus based on a unique collection of morphological and colour pattern characteristics. These unique characteristics include a snout-vent length of 30.0 mm, 29 midbody scale rows, smooth as opposed to striated dorsal scales, 65 paravertebrals, 61 ventrals, 4 supraoculars, parietals contacting the posterior-most supraocular, 1 medially projecting superciliary scale, 2 loreals, 6 supralabials and infralabials, 10 lamellae beneath the fourth toe, smooth subdigital lamellae, enlarged preanal scales, no body bands, a dark brown, diffuse, dorsolateral stripe extending to just past the axilla, a cream coloured dorsolateral stripe on the nape and anterior-most portion of the body, and no cream coloured postorbital stripe. The discovery of a second endemic reptile in the Perhentian Archipelago underscores the unrealized biodiversity of its herpetofauna. Additional works will describe two additional species from the Perhentian Archipelago.
A survey was carried out at the Pasoh Forest Reserve from 13–19 December, 2007 and 20–24 January, 2008, to inventory the herpetofauna contained therein. A total of 23 species of reptiles and amphibians were collected during this survey, five of which have not previously been recorded in the Pasoh Forest Reserve. An updated checklist which incorporates findings from previous studies is presented.
A herpetological survey was conducted at Pulau Besar, Melaka from 10 to 12 June 2008, resulting in the discovery of five species of amphibians, nine species of lizards (including one new species of the gekkonid Cyrtodactylus) and one snake species. This report constitutes the first checklist of the herpetofauna of Pulau Besar and the Water Islands Archipelago.
A comparison study was conducted to determine the bird species composition, relative abundance, species diversity and feeding guilds based on point count (PC) and mist netting (MN) at the Paya Indah Wetland Reserve (PIWR), Peninsular Malaysia. A total of 13872 bird observations belonging to 100 species and 38 families were recorded using the PC method over 15 consecutive months, and a total of 1478 bird individuals
belonging to 65 species and 33 families were captured using the MN method over 1260 netting hours. The results showed that Treron vernans (1723 observations; 12.42%) was the most abundant bird species using the PC method, whereas Pycnonotus goiavier (378 individuals; 25.64%) was the most abundant bird species using the MN method. The Ardeidae (9 species; 23.68%) was the most dominant family using the PC method, but the Rallidae (6 species; 18.18%) was the most dominant family using the MN method. The PC method produced higher species diversity (Shannon’s N1 = 31.22) and richness (Margalef’s R1 = 10.42) than MN, whereas the MN method produced higher species evenness (McIntosh’s E = 0.86) than the PC method. Frugivore/insectivore comprised of bulbuls, orioles, pigeons and starlings was the most dominant feeding guild in both methods (PC = 27.81% and MN = 32.88%). In contrast, carnivore was the rarest feeding guild in both methods (i.e. PC = 0.17% and MN = 0.20%). These findings indicate that the PC method is more efficient and produces better results than the MN method.
Weedy rice (WR) is found in many direct-seeded rice fields. WR possesses morphological characteristics that are similar to cultivated rice varieties in the early stage of growth, making them more difficult to control than other weeds. A comparative morphological study was conducted by collecting WR accessions from four sites within the Pulau Pinang rice growing areas. The objective of the study was to characterise WR accessions of the Pulau Pinang rice granary by comparing their morphological characteristics to those of commercially grown rice in the area. Their morphometric relations were established by comparing 17 morphological characteristics of the WR accessions and the commercial varieties. A total of 36 WR morphotypes were identified from these 4 sites based on 17 characteristics, which included grain shattering habit and germination rate. The Principal Component Analysis (PCA) showed that 45.88% of the variation observed among the WR accessions and commercial varieties were within the first 3 axes. PB6, PP2 and SGA5 WR accessions had a higher number of tillers and longer panicle lengths, culm heights and leaf lengths compared to the commercial rice. The grain
sizes of the commercial varieties were slightly longer, and the chlorophyll contents at 60–70 days after sowing (DAS) were higher than those of the WR accessions. Results from this study are useful for predicting potential WR accession growth, which might improve WR management and agriculture practices that control WR in the future.
Algal communities possess many attributes as biological indicators of spatial and temporal environmental changes. Algal parameters, especially the community structural and functional variables that have been used in biological monitoring programs, are highlighted in this document. Biological indicators like algae have only recently been included in water quality assessments in some areas of Malaysia. The use of algal parameters in identifying various types of water degradation is essential and complementary to other environmental indicators.
There is not much information available on the distribution of the Sunda colugo (Galeopterus variegates) in Malaysia, despite it being one of only two known species in the order Dermoptera. Data on the presence of the Sunda colugo and the vernacular names used by various ethnic groups throughout Malaysia were collected and compiled from various primary and secondary sources. There were 27 locations from Peninsular, 11
locations from Sabah and 34 locations from Sarawak that reported the presence of the Sunda colugo throughout Malaysia. The various ethnic groups of Malaysia adopted 37 different vernacular names to describe the Sunda colugo. This baseline data can be useful for the management authorities in conducting periodic monitoring and will enhance our knowledge of the population dynamics of the Sunda colugo in Malaysia.
Research on natural products has been widely used as a strategy to discover new drugs with potential for applications in complementary medicines because they have fewer side effects than conventional drugs. The aim of the present study was to evaluate the in vitro cytotoxic effects of crude aqueous Catharanthus roseus extract on Jurkat cells and normal peripheral blood mononuclear cells (PBMCs). The aqueous extract was
standardised to vinblastine by high performance liquid chromatography (HPLC) and was used to determine cytotoxicity by the MTS [3-(4,5-dimethylthiazol-2-yl)-5(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium] assay. DNA fragmentation assay was employed to determine if cell death was due to apoptosis. The results showed that the aqueous extract induced cell death of Jurkat cells at 24, 48 and 72 hours posttreatment in a time- and dose-dependent manner. However, cells treated at 48 and 72 hours produced higher cytotoxic effects with half maximal inhibitory concentration (IC50)values of 2.55 µg/ml and 2.38 µg/ml, respectively. In contrast, the extract induced normal PBMC proliferation, especially after 24 hours treatment with 1000 µg/ml. This result indicates that the C. roseus crude aqueous extract showed differential effects of inhibiting the proliferation of the Jurkat cell line and promoting the growth of PBMCs. These data suggest that the extract may be applicable for modulating the normal and transformed immune cells in leukaemia patients.
The distribution and abundance of zooplankton species of Harapan and Aman
Lakes were investigated in relation to physical parameters and chlorophyll-a content. Both
lakes were characterised by the occurrence of algal bloom problem. The composition of
zooplankton was collected at monthly intervals from November 2013 to February 2014.
The total number of taxa in Harapan and Aman Lakes were 23 and 27, respectively.
Rotifera was the highest abundance group represent 64% of the total species recorded
followed by Copepoda (29%) and Cladocera (7%). Three dominant zooplankton that been
recorded in both the lakes are Brachionus forficula, Brachionus nilsoni, and Trichocerca
sp. High abundance of these species indicates that the lakes are eutrophic water bodies.
Overall, zooplankton species distribution and abundance in the study sites are influenced
by various environmental factors such as water transparency and chlorophyll-a content.
