Kajian kelakuan harian diurnal telah dijalankan ke atas beberapa ekor cimpanzi (Pan troglodytes) di Zoo Negara. Objektif kajian ialah untuk mencerap peruntukan aktiviti harian cimpanzi di dalam kurungan dan mengkaji kesan pengkayaan terhadap kelakuannya. Kaedah persampelan yang digunakan ialah persampelan fokus dengan pengrekodan berterusan. Pemerhatian dijalankan selama 32 hari dari pukul 9.00 pagi hingga 5.00 petang dengan satu jam berehat. Fasa tanpa pengkayaan berlangsung selama 16 hari (80 jam 45 min). Fasa pengkayaan juga dilakukan selama 16 hari (86 jam 15 min). Hasil aktiviti harian menunjukkan cimpanzi banyak berehat (45.2%) dan memerhati persekitaran (16.81%) diikuti dengan aktiviti pergerakan (11.33%), makan (8.38%), berdandan (5.27%), mencari makanan (4.81%), tidur (3.40%), agresif (0.38%), kelakuan lain-lain yang tak tertakrif dalam skop kajian ini seperti kelakuan stereotipik (0.28%) dan kelakuan seksual (0.14%). Fasa pengkayaan memberi hanya perbezaan yang bererti kepada kelakuan mencari makanan (p<0.001), membina sarang (p<0.05) dan kelakuan memerhati persekitaran (p<0.05).
The effects of various concentrations of honey and sucrose on the longevity and fecundity of Apanteles metesae (Nixon), an endoparasitoid of oil palm bagworm, Metisa plana (Walker) (Lepidoptera: Psychidae) were studied in the laboratory. Our results showed that there were a significant difference (p<0.05) in the longevity of A. metesae fed on various concentrations of honey solutions. Longevity of A. metesae was significantly longer when fed on 50% honey than on pure honey. Nonetheless, longevity of A. metesae females when fed on 50% honey was relatively longer than other honey concentrations. The parasitoid lived significantly longer when fed on 20% sucrose than fed on 50% sucrose and distilled water. However, the parasitoid fed on 50% honey had a longevity that was statistically similar to 20% sucrose and 50% sucrose. Fecundity of A. metesae differed significantly (p<0.05) when fed on 50% honey, 20% sucrose and 50% sucrose. The highest and lowest fecundity was when the parasitoids fed on 20% sucrose and distilled water, respectively. Overall, the result showed that 20% sucrose was a more suitable diet for A. metesae females as compared with other diets.
Plutella xylostella (L.) (Lepidoptera: Plutellidae), the major insect pest of cruciferous crops worldwide shows significant
resistance to almost all classes of insecticides. In order to effectively prevent and manage the insecticidal resistance,
it is crucial to understand the physiological adaptation of insects against insecticides. Identification of insect protein
that interacting with insecticides and characterization of their modification in resistant strains can be done by using
differential proteomics approach. This study focuses on optimizing a sensitive and rapid method for the extraction of
high quality protein of both larva and adult tissues of P. xylostella to be used in two-dimensional gel electrophoresis.
Five extraction methods were evaluated for protein concentration, yields and resolving patterns of one-dimensional
and two-dimensional electrophoresis. The results showed that trichloroacetic acid/acetone extraction methods with
two different concentrations of 2-mercaptoethanol produced the highest protein concentration and yield for both adult
and larva tissues, respectively. Meanwhile, trichloroacetic acid/acetone with dithiothreitol extraction method gave
better separation of spots and intensity for both larva and adult tissues compared to other methods tested. As such, we
concluded that trichloroacetic acid/acetone with dithiothreitol successfully yielded high total protein concentration and
good separation of two-dimensional electrophoresis gel spots in both adult and larva P. xylostella.
Ficus species are characterized by their unusual enclosed inflorescences (figs) and their relationship with obligate pollinator fig wasps (Agaonidae). Fig trees have a variety of growth forms, but true epiphytes are rare, and one example is Ficus deltoidea of Southeast Asia. Presumably as an adaptation to epiphytism, inflorescence design in this species is exceptional, with very few flowers in female (seed-producing) figs and unusually large seeds. Figs on male (pollinator offspring-generating) trees have many more flowers. Many fig wasps pollinate one fig each, but because of the low number of flowers per fig, efficient utilization by F. deltoidea's pollinators depends on pollinators entering several female figs. We hypothesized that it is in the interest of the plants to allow pollinators to re-emerge from figs on both male and female trees and that selection favors pollinator roaming because it increases their own reproductive success. Our manipulations of Blastophaga sp. pollinators in a Malaysian oil palm plantation confirmed that individual pollinators do routinely enter several figs of both sexes. Entering additional figs generated more seeds per pollinator on female trees and more pollinator offspring on male trees. Offspring sex ratios in subsequently entered figs were often less female-biased than in the first figs they entered, which reduced their immediate value to male trees because only female offspring carry their pollen. Small numbers of large seeds in female figs of epiphytic F. deltoidea may reflect constraints on overall female fig size, because pollinator exploitation depends on mutual mimicry between male and female figs.
