    INTRODUCTION: Kidney disease is a worldwide health concern with an increasing mortality in the past 10 years. The Kidney Disease Improving Global Outcomes (KDIGO) guideline advocates the use of estimated glomerular filtration rate equation (eGFR) to estimate renal function. We evaluated the performance of Cockroft Gault (CG), Modified Diet of Renal Disease (MDRD), and Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) equations to measured GFR 99mTc- DTPA taking into account body mass index (BMI) and age group.

    MATERIALS AND METHODS: This is a cross-sectional study of patients referred for 99mTc-DTPA scan at the Nuclear Medicine Centre of International Islamic University Malaysia. The record was taken from patients visiting the centre from January 2016 to December 2019.

    RESULTS: The mean measured GFR by 99mTc-DTPA scan was 42.2 ± 20.38 ml/min. These were lower than that estimated by CG, MDRD, and CKD-EPI equations. CKD-EPI had the highest correlation of 0.72, least bias (mean bias of 11.08 ± 23.08) and was more precise (r2 = 0.4) as compared to MDRD and CG. In patients < 65 years old, CKD-EPI had the highest correlation; however, MDRD had the least bias and highest accuracy. In terms of BMI, CKD-EPI had the least bias and highest accuracy for BMI >30 and with the highest correlation for all classes of BMI.

    CONCLUSION: CKD-EPI has the best estimation of GFR taking into account the effect of BMI and age. A further study can be done to determine the correlation of estimated GFR equations with different ethnicity in Malaysia.

    Mandarin fish (Siniperca chuatsi) fed with an artificial diet is progressively gaining popularity, which is important for reducing product prices and resource consumption. However, food is the decisive factor of intestinal microbes, and the profound effects of change in their feeding habit on intestinal microbes of mandarin fish have not been revealed. In the present study, live bait fish and artificial diet were used to feed mandarin fish for 8 weeks to study the effect of different feeding habits on the histology, microbiota structure and dominant bacteria of gut in mandarin fish. The results indicated that feeding with the artificial diet significantly increased the intestinal villi height and muscular thickness in the hindgut of mandarin fish. In addition, the microbiota results showed that there were significant differences of beta diversity of gut microbiota in mandarin fish fed with different diets. At the genus level, feeding artificial diets significantly increased the abundance of Weissella in the mandarin fish gut. Furthermore, two Weissella bacteria were identified and characterized from the midgut contents of mandarin fish fed with artificial diet. Based on 16S rRNA gene sequencing, nine strains were assigned as Weissella confusa (RM125), and one as Weissella cibaria (SJ548). Biochemical analyses based on the VITEK2 method revealed a pattern of metabolic activities against W. confusa RM125 and W. cibaria SJ548, with 13 positive and 29 negative results, respectively. W. confusa RM125 and W. cibaria SJ548 exhibited sensitivities toward a variety of pathogens, including V. harveyi, S. aureus and V. parahaemolyticus, E. coli, A. hydrophila, S. enterica, V. anguillarum, and V. alginolyticus, indicating potential probiotics. Therefore, our results confirmed that the transformation of feeding habit altered the structure, microbial composition and dominant bacteria in gut of mandarin fish, and provided evidence that Weissella might play a crucial role in the digestion and absorption of artificial diet in mandarin fish.

    Gills are the main respiratory organs of fish and bear important physiological and immunological functions, but the functional heterogeneity of interlamellar cell mass (ILCM) at the single-cell level has rarely been reported. Here, we identified 19 cell types from the gills of crucian carp (Carassius auratus) by single-cell RNA sequencing (scRNA-seq) in combination with histological analysis. We annotated ILCM and analyzed its functional heterogeneity at the single-cell level for the first time. Functional enrichment analysis and cell cycle analysis identified ILCM as a type of metabolically active cells in a state of constant proliferation, and identified the major pathways responsible for ILCM immunoregulation. Histological analysis revealed the morphology and positional relationships of 6 cell types. Meanwhile, the gene regulatory network of ILCM was established through weighted gene co-expression network analysis (WGCNA), and one transcription factor and five hub genes related to immunoregulation were identified. We found that pyroptosis might be an important pathway responsible for the immune response of ILCM. Our findings provide an insight into the physiological and immune functions of gills and ILCM at the single-cell level and lay a solid foundation for further exploration of the molecular mechanism of ILCM immunity functions.
