    Fig, or Ficus carica, is a fruit tree from the Moraceae family and is widely grown in tropical and subtropical regions of the world. Fig plants are mainly propagated through grafting, air layering, and hardwood cutting whereby these methods were found to be less efficient. Plant tissue culture is efficient method to propagate plants, particularly to produce true-to-type platelets for mass multiplication. The aim of this study is to induce multiple shoot formation on Ficus carica cv. Japanese BTM 6 through identifying and optimising the concentrations of 6-Benzylaminopurine (BAP) and Zeatin suited for shoot formation. The axillary shoot tip explants were cultured in MS media supplemented with different concentrations of BAP and Zeatin (0, 0.5, 1.0, 1.5 and 2.0 mg/L) to determine the optimal concentration for the formation of multiple shoots. Number of shoots per explants and the differences in shoot height of explants were calculated after 8 and 12 weeks of culture respectively. Of all the treatments of BAP, MS media containing with 2 mg/L BAP marked the highest number of shoots per explant with the average value of 1.67 ± 0.33 while 1.5 and 2 mg/L of BAP produced the highest differences in shoot height with 0.51 ± 0.08 cm and 0.51 ± 0.07 cm after 12 weeks respectively. Murashige and Skoog (MS) media supplemented with 2 mg/L Zeatin showed the highest production of multiple shoots and differences in shoot height with the average of 0.83 ± 0.219 and 0.32 ± 0.04 cm respectively among all the different treatments of Zeatin. In this study, BAP performed better in shoot induction and elongation as compared to Zeatin for the cultivar Japanese BTM 6.

    Solanum melongena L. commonly known as the eggplant or brinjal comes from the family of Solanaceae, sharing the same ancestor with the tomato and potato. It is an economically important crop worldwide, being well studied for its medicinal properties, nutritional values and its role as an alternative model plant. The eggplant fruit has been previously used for treatments of various diseases such as bronchitis, asthma, arthritis and diabetes as well as its nutritive properties that are beneficial to the human diet. Plant transformation studies on the eggplant have been widely done for the production of transgenic eggplants harbouring genes that are beneficial for optimal plant growth and fruit production. Shoot induction is an essential step required for the successful regeneration of transformed plant tissues and therefore is an essential pre-requisite in Agrobacterium-mediated transformation. The local eggplant cv. Bulat Putih is a local cultivar of eggplant in Malaysia with white and round fruits making it a potential model plant colour pigment accumulation studies in fruit crops. The current work aims to investigate the shoot induction potential of 6-benzylaminopurine (BAP) and Kinetin from cotyledon explants of eggplant cv. Bulat Putih. Results indicated that both BAP and Kinetin were able to induce the regeneration of callus from cotyledon explants. On the other hand, Kinetin at the concentration of 2.0 mg/L successfully induced shoots at the value of 1.50 ± 0.22 shoots per explant, whereas BAP alone did not trigger any formation of shoots. This study indicated that kinetin alone is sufficient to induce shoots in eggplant cv. Bulat Putih without the presence of BAP.

    The metabolism of alcohol involves cytochrome P450 2E1 (CYP2E1)-induced oxidative stress, with the association of phosphatidylinositol-3-kinases (PI3K) and nuclear factor kappa B (NFκB) signalling pathways. CYP2E1 is primarily involved in the microsomal ethanol oxidising system, which generates massive reactive oxygen species (ROS) and ultimately leads to oxidative stress and tissue damage. Lauric acid, a major fatty acid in palm kernel oil, has been shown as a potential antioxidant. Here, we aimed to evaluate the use of lauric acid as a potential antioxidant against ethanol-mediated oxidative stress by investigating its effect on CYP2E1 mRNA expression and the signalling pathway in ethanol-induced HepG2 cells. HepG2 cells were firstly treated with different concentrations of ethanol, and subsequently co-treated with different concentrations of lauric acid for 24 h. Total cellular RNA and total protein were extracted, and qPCR and Western blot was carried out. Ethanol induced the mRNA expression of CYP2E1 significantly, but lauric acid was able to downregulate the induced CYP2E1 expression in a dose-dependent manner. Similarly, Western blot analysis and densitometry analysis showed that the phosphorylated PI3K p85 (Tyr458) protein was significantly elevated in ethanol-treated HepG2 cells, but co-treatment with lauric acid repressed the activation of PI3K. However, there was no significant difference in NFκB pathway, in which the normalised NFκB p105 (Ser933) phosphorylation remained constant in any treatment conditions in this study. This suggests that ethanol induced CYP2E1 expression by activating PI3K p85 (Tyr458) pathway, but not the NFκB p105 (Ser933) pathway in HepG2 cells.

    Rodents belong to the order Rodentia, which consists of three families in Borneo (i.e., Muridae, Sciuridae and Hystricidae). These include rats, mice, squirrels, and porcupines. They are widespread throughout the world and considered pests that harm humans and livestock. Some rodent species are natural reservoirs of hantaviruses (Family: Bunyaviridae) that can cause zoonotic diseases in humans. Although hantavirus seropositive human sera were reported in Peninsular Malaysia in the early 1980s, information on their infection in rodent species in Malaysia is still lacking. The rodent populations in residential and forested areas in Sarawak were sampled. A total of 108 individuals from 15 species of rodents were collected in residential (n = 44) and forested ( n = 64) areas. The species diversity of rodents in forested areas was significantly higher (H = 2.2342) compared to rodents in residential areas (H = 0.64715) (p < 0.001 of Zar-t test based on the Shannon index). Rattus rattus and Sundamys muelleri were present at high frequencies in both localities. An enzyme-linked immunosorbent assay (ELISA) showed that hantavirus-targeting antibodies were absent from 53 tested serum samples. This is the first report of hantavirus seroprevalence surveillance in rodent populations in Sarawak, East Malaysia. The results suggested that hantavirus was not circulating in the studied rodent populations in Sarawak, or it was otherwise at a low prevalence that is below the detection threshold. It is important to remain vigilant because of the zoonotic potential of this virus and its severe disease outcome. Further studies, such as molecular detection of viral genetic materials, are needed to fully assess the risk of hantavirus infection in rodents and humans in this region of Malaysia.

    The Trigonopoma pauciperforatum or the redstripe rasbora is a cyprinid commonly found in marshes and swampy areas with slight acidic tannin-stained water in the tropics. In this study, the complete mitogenome sequence of T. pauciperforatum was first amplified in two parts using two pairs of overlapping primers and then sequenced. The size of the mitogenome is 16,707 bp, encompassing 22 transfer RNA genes, 13 protein-coding genes, two ribosomal RNA genes and a putative control region. Identical gene organisation was detected between this species and other family members. The heavy strand accommodates 28 genes while the light strand houses the remaining nine genes. Most protein-coding genes utilise ATG as start codon except for COI gene which uses GTG instead. The terminal associated sequence (TAS), central conserved sequence block (CSB-F, CSB-D and CSB-E) as well as variable sequence block (CSB-1, CSB-2 and CSB-3) are conserved in the control region. The maximum likelihood phylogenetic tree revealed the divergence of T. pauciperforatum from the basal region of the major clade, where its evolutionary relationships with Boraras maculatus, Rasbora cephalotaenia and R. daniconius are poorly resolved as suggested by the low bootstrap values. This work contributes towards the genetic resource enrichment for peat swamp conservation and comprehensive in-depth comparisons across other phylogenetic researches done on the Rasbora-related genus.

    The nesting of green turtle (Chelonia mydas) was monitored from 1998 untill 2013 along the beaches of Pasir Panjang, Segari, Perak. The objective of the study is to assess the nesting status of green turtles in Perak, Peninsular Malaysia in terms of total nests, eggs, survival hatchings, and density of visitors. A total number of green turtle nests found for 16 years were 1,019 nests and varied from 10 to 220 nests per year. Meanwhile, the sum of eggs collected for 16 years were 107,820 eggs, and varied from 553 to 20,881 eggs per year. The temporal pattern of nesting indicates year-round nesting in Perak in most years within the 16 years period. The peak season of nesting was estimated to occur between May and June. Survival hatchlings varied from 23.33% (2,071 hatchlings) to 55.03% (5,018 hatchlings) from 1998 to 2013. The density of visitors was not uniformly distributed among the years, and shows a sign of decline especially from 2006 onwards. This publication provides basic knowledge of green turtle nesting population in Perak, and would be helpful in upgrading the conservation program in Malaysia. In future, we hope 1) for an increase in manpower to obtain accurate nesting records along the nesting beaches during nocturnal survey and, 2) to include the breeding biology data such as nest placement, emergence hour, and morphological characteristics of green turtle.

    Ten nests were collected from Kerachut and Teluk Kampi, Penang Island between 2 August 2009 and 9 December 2009, and each one nest was split into three small clutch sizes for incubation at three nesting depths (45 cm, 55 cm and 65 cm), with a total of 30 modified nests for this experiment. Three important objectives were formulated; to observe on the survival hatchings among the three nesting depths, to study on the effects of sand temperature on incubation period among the three nesting depths, and to investigate the influence of sand temperature on hatchling's morphology. Main result shows that the mean survival of the hatchlings was 25.40% at 45 cm nesting depth, followed by mean 17.60% at 55 cm nesting depth, and lastly, the mean was 21.50% at 65 cm nesting depth. Overall, there are 56.63% survival hatchlings, 10.97% dead hatchlings and 32.40% unhatched eggs were produced. The incubation period was also found to be significantly correlated with sand temperature, p > 0.001, and nesting depth, p < 0.001. The hatchling's length and weight varies is sizes across the nesting depths, p < 0.001. However, the small difference in hatchling sizes per nesting depths are not strong enough to prove the significant correlation with sand temperature, p > 0.05. This article provides a basic knowledge from the splitting clutch design method. A sum of 50%-60% survivals hatchlings produced were incubating under small range of clutch sizes, 29 to 49 eggs. This article provides basic result on the survival hatchlings, eggs survivorship, incubation period, temperature, hatchling's morphology and discussion on implication of this method on conservation in Malaysia.

    Keradangan hati berkait rapat dengan perubahan dalam ekspresi lipoprotein dan apolipoprotein. Interferon-γ (IFN-γ), wakil tunggal jenis kedua IFN, memainkan peranan yang penting dalam memodulasi dan mempergiatkan tindak balas keradangan. Justeru itu, kajian ini direka untuk mengenal pasti kesan IFN-γ terhadap apolipoprotein A-I (APOA-I) dan penglibatan nuclear factor–kappa B (NF-κB) dalam laluan isyarat tersebut. Tindak balas rantai polymerase transkripsi berbalik kuantitatif (qRT-PCR) dan analisis blot western telah dilaksanakan untuk menguantifikasi ekspresi APOA-I dalam sel-sel HepG2 selepas dirawat dengan IFN-γ. Kajian ini menunjukkan bahawa 50 ng/mL IFN-γ merangsangkan ekspresi mRNA dan protein APOA-I. Walau bagaimanapun, pra-rawatan sel dengan inhibitor laluan isyarat NF-κB mengurangkan tahap ekspresi APOA-I. Kajian ini juga mendemonstrasikan penglibatan langsung isyarat NF-κB dalam ekspresi APOA-I akibat rangsangan IFN-γ, di mana IFN-γ meningkatkan tahap fosforilasi NF-κB p65 Ser468 dan Ser536 kepada 2.59-ganda and 1.63-ganda. Namun demikian, pra-rawatan sel dengan perencat laluan isyarat NF-κB melumpuhkan peningkatkan tersebut dan kemudian mengurangkan ekspresi APOA-I dalam sel HepG2. Sebagai rumusan, kajian ini berjaya mengenalpasti peranan isyarat NF-κB dan pengaktifan p65 Ser468 dan Ser536 sebagai pengantara IFN-γ ke atas induksi APOA-I di dalam sel-sel HepG2.

    Cyanobacteria are one of the most important groups of photoautotrophic organisms, contributing to carbon and nitrogen fixation in mangroves worldwide. They also play an important role in soil retention and stabilisation and contribute to high plant productivity through their secretion of plant growth-promoting substances. However, their diversity and distribution in Malaysian mangrove ecosystems have yet to be studied in detail, despite Malaysia hosting a significant element of remaining mangroves globally. In a floristic survey conducted in Penang, peninsular Malaysia, 33 morphospecies of periphytic cyanobacteria were identified and described for the first time from a mangrove ecosystem in Malaysia. Sixteen genera, comprising Aphanocapsa, Chroococcus, Chroococcidiopsis, Cyanobacterium, Desmonostoc, Geitlerinema, Leptolyngbya, Lyngbya, Microcystis, Myxosarcina, Oscillatoria, Phormidium, Pseudanabaena, Spirulina, Trichocoleus and Xenococcus, were obtained from field material growing on diverse natural and artificial substrata. Oscillatoriales was the dominant order with Phormidium the dominant genus at nine of the 15 sampling sites examined. Three of the morphospecies, Aphanocapsa cf. concharum, Xenococcus cf. pallidus and Oscillatoria pseudocurviceps, are rare and poorly known morphospecies worldwide. Chroococcus minutus, Phormidium uncinatum, P. amphigranulata, and some species of Oscillatoriales are considered as pollution indicator species. This study provides important baseline information for further investigation of the cyanobacterial microflora present in other mangrove areas around Malaysia. A complete checklist will enhance understanding of their ecological role and the potential for benefits arising from useful secondary metabolites or threats via toxin production to the ecosystem.

    Species identification is important for epidemiological, clinical and treatment purposes. The aim of this study was to find out whether hippurate hydrolysis is a reliable test for differentiating between Campylobacter coli and Campylobacter jejuni. To achieve this, hippurate hydrolysis test was compared with multiplex Polymerase Chain Reaction (mPCR) for their ability to speciate C. coli and C. jejuni. Eighteen Campylobacter strains from poultry samples were used for this study. The results from 17 of the 18 strains were in agreement with both methods. Thus, the hippurate hydrolysis test can be used for distinguishing C. jejuni from C. coli although occasionally some strains of C. jejuni may be mis-identified as C. coli.

    The status and distribution of Mimosa pigra L., a semi-aquatic invasive species in Peninsular Malaysia, were continuously assessed between 2004 and 2007. This assessment investigated its population stand density and related weed management activities. In total, 106 sites of 6 main habitat types i.e., construction site (CS), dam/ reservoir (DM), forest reserve (FR), plantation (PL), river bank/waterway (RB) and roadside (RD) were assessed, and 55 sites were recorded with M. pigra populations. A CS is the most likely habitat to be infested with M. pigra (16 out of 18 assessed sites have this weed), whereas none of the FR visited were found to harbour M. pigra. In terms of population stand density, 41 populations were in the low range of stand density (individual plant of ≤5 m(-2)), compared to only 9 populations in the high range of stand density (individual plant of >10 m(-2)). In general, the current impact of M. pigra infestation on natural habitats is relatively low, as its distribution is only confined to disturbed areas. However, continuous monitoring of this weed species is highly recommended, especially in the riparian zone and wetland habitats.

    Mitragyna speciosa (Korth.) Havil. or locally known as ketum/daun sebiak/biak-biak belongs to Rubiaceae family and generally occurs in secondary forest or disturbed areas in tropical and subtropical region. This research enumerated the characterisation of Mitragyna speciosa leaf anatomy and micromorphology features which is still not well documented. This medium to large sized tree species characterised with opposite arrangement, ovate-acuminate leaf and with 12-17 pairs of veins. Transverse sections of petioles showed that this species has petiole outlines with slightly convex at the middle of the adaxial part and 'U'-shaped on abaxial side. Results also showed that this species has paracytic and hypostomatic stomata, combination of non-glandular (majority) and glandular trichomes (minority), with observation on the secretory cells present in petiole and midrib parenchyma cells. Cuticle on the abaxial and adaxial epidermal surfaces showed the presence granule and wax films with periclinal and anticlinal walls can be differentiated clearly. The results obtained in this study can be used to providing additional systematics information of Mitragyna speciosa with the documentation of the leaf anatomy and micromorphology characters.

    The pteridophyte flora of Langkawi Archipelago consists of 130 species, 1 subspecies and 12 varieties in 68 genera and 27 families. This value represents 22.1% of the 647 taxa at the species level and below reported for Peninsular Malaysia. Of the 143 recorded taxa of pteridophytes at the species level and below, 8 species in 2 genera and 2 families are lycophytes and the other 135 taxa in 66 genera and 25 families are monilophytes or ferns.

    Cross-breeding is a method of producing progeny with better resistance to pathogens. Resistance to pathogens usually involves pathogenesis-related (PR) proteins. Class II chitinase is an example of a defensive PR protein in plants. The class II chitinase in chilli is coded by the CaChi2 gene. In this study, we crossed susceptible with resistant chilli cultivars, analysed the F1 resistance response against pathogenic F. oxysporum, and analysed the level of CaChi2 gene expression in the F1. Data were collected using disease severity index (DSI) determination and gene expression analysis by qRT-PCR (quantitative Reverse Transcriptase Polymerase Chain Reaction). Results showed that the DSI of F1 was not significantly different from the resistant ancestor. The relative CaChi2 expression level of F1 was higher than the susceptible ancestor but not significantly different from the resistant ancestor. We concluded that the F1 can be categorised as resistant to F. oxysporum, and the CaChi2 gene is involved in the molecular defense response.

    Data on nutrient contents of freshwater crabs are important for ecological studies and species conservation assessments, especially when the species concerned is threatened among others by habitat destruction and uncontrolled resources utilisation. Indeed comprehensive biological information is required to reconcile the needs between sustainable resources utilisation and conservation of the species. This study documents the nutrient contents of a freshwater crab, Isolapotamon bauense which is listed as 'Vulnerable' in the IUCN Red List of Threatened Species and also being harvested by local community for dietary supplement. Results show that muscles of the freshwater crab contain a substantial amount of nutrients in particular water content (male = 79.31 ± 2.30 %, female = 77.63 ± 0.56 %), protein (male = 77.47 ± 6.11 %, female = 63.28 ± 3.62 %), magnesium (male = 51.48 ± 16.10 mg/g, female = 39.73 ± 6.99 mg/g) and calcium (male = 25.50 ± 6.98 mg/g, female = 39.73 ± 6.99 mg/g). Means of nutrient contents between male and female crabs are not significantly different. It is estimated that an individual of I. bauense with weight range of 56-139 g contained on average of 0.35 ± 0.15 g of protein. Our estimation also shows that the number of individuals of the freshwater crab required to meet the recommended daily protein intakes of the community concerned is in the range 35-96 individuals for children, 130-188 individuals for adolescents, 171-179 individuals for men and 149-159 individuals for women. The results imply that harvesting of wild I. bauense as a source of protein supplement naturally may not be practical because of its relatively low population abundance, and conservation of the species for its ecological roles may thus be preferred.

    Staphylococcus kloosii, an orange pigment-producing bacterium, was isolated from the respiratory tree of Holothuria (Mertensiothuria) leucospilota (Brandt 1835) from Teluk Nipah, Pangkor Island, Perak, Malaysia. This report is the first documentation of this Gram-positive strain, referred to as Strain 68 in Malaysia. A partial 16S ribosomal RNA gene sequence of the mesophilic strain has been registered with GenBank (National Center for Biotechnology Information, US National Library of Medicine) with accession number JX102547. Phylogenetic analysis using the neighbour-joining method further supported the identification of Strain 68 as S. kloosii. The circular strain produced orange pigments on tryptone glucose yeast extract agar (TGYEA) and in nutrient broth (NB) at approximately pH 7. The visible spectra of ethanolic and methanolic pigment extracts of the bacterial strain were considered identical with λmax at 426, 447 and 475 nm and λmax at 426, 445 and 473 nm, respectively. Both visible spectra resemble the visible spectra of lutein, which is a commercial carotenoid; however, further analyses are required to confirm the identity of this pigment. The methanolic extracts of the intracellular pigments comprised at least three pigment compounds: an orange pigment compound (major compound), a yellow pigment compound (the least polar) and a pink pigment compound (the most polar). These findings are the first documentation of the pigment composition of S. kloosii as no such record could be found to date.

    Banana peel flour (BPF) prepared from green or ripe Cavendish and Dream banana fruits were assessed for their total starch (TS), digestible starch (DS), resistant starch (RS), total dietary fibre (TDF), soluble dietary fibre (SDF) and insoluble dietary fibre (IDF). Principal component analysis (PCA) identified that only 1 component was responsible for 93.74% of the total variance in the starch and dietary fibre components that differentiated ripe and green banana flours. Cluster analysis (CA) applied to similar data obtained two statistically significant clusters (green and ripe bananas) to indicate difference in behaviours according to the stages of ripeness based on starch and dietary fibre components. We concluded that the starch and dietary fibre components could be used to discriminate between flours prepared from peels obtained from fruits of different ripeness. The results were also suggestive of the potential of green and ripe BPF as functional ingredients in food.

    This study utilised a bio-sorbent from exhausted coffee husk (ECHBS) for the removal of ion Pb2+ from an aqueous solution. Four different activation methods were conducted by chemical activation with KOH, H3PO4, ZnCl2, and without chemical activation. In addition, the influence of process parameters such as heating temperature, heating time and heating gradient were investigated. Based on the experimental results, ECHBS without chemical activation (biochar) had the highest Pb2+ ion removal efficiency. The results showed that the heating temperature of 500°C, the heating time of 60 min and the heating rate of 15°C/min were optimum for preparation of the biochar. Under the optimum conditions, the removal efficiency and adsorption capacity reached 99% and 3.3 mg/g, respectively. The experimental data indicated that the adsorption isotherms are well fitted with the Langmuir Equilibrium isotherm model. Furthermore, the adsorption of the biochar follows the pseudo-second-order model. The result obtained from the present study confirmed that exhausted coffee husk is a suitable low-cost bio-sorbent for removing ion Pb2+.

    Helopeltis antonii is the major pest affecting cashew plants in Indonesia and causes potential damage to the plant. The development of the population was influenced by the fecundity and fertility of their eggs. The effect of mating on the eggs’ fecundity and the fertility of H. antonii was studied. Laboratory studies at Wonogiri Estate Service in Ngadirojo District, Wonogiri, Indonesia investigated the sexual maturity, the influence of female to male sex ratio on the females’ fecundity and longevity, and the influence of mating frequency to fecundity and the eggs hatchability of H. antonii. The study reveals that H. antonii females and males are ready to mate when they are two days old. The number of eggs laid and the longevity of the females’ lives were not significantly affected by the sex ratio; however, female to male sex ratios of 2:1 and 1:2 tended to produce a greater number of eggs. The fecundity of the female was not significantly influenced by the number of times the female mated. Unmated females laid fewer eggs than females paired with a mature male. The study shows that females need to mate to produce fertile eggs.
