    Introduction: Caregivers of children with speech-language pathology experience psychological distress as speech impedes communication and interferes children’s development. This study aimed to determine the effects of a one- day mindfulness-based intervention (MBI) in reducing depression, anxiety, and stress for caregivers of children with speech-language pathology in a Malaysian hospital. Methods: Caregivers (n = 62) who fulfilled the selection criteria were recruited and randomly assigned to either MBI or treatment-as-usual control groups. The caregivers completed self-rated questionnaires, namely the Depression Anxiety Stress Scale 21 (DASS 21) and Mindful Attention Aware- ness Scale (MAAS) before and at one-month post-intervention. Results: Hierarchical multiple regression analyses demonstrated significant improvement in DASS 21 and MAAS scores in the intervention group compared to control group.     Introduction: Mental problems have become increasingly important public health issue globally. This study aimed to determine the prevalence and factors associated with depression, anxiety, and stress among medical students in a public university in Malaysia. Methods: A total of 447 medical students were sampled by stratified proportion- ate to size sampling according to year of study. Validated self-administered questionnaire was used consisted of 5 sections namely, socio-demographic factors, social support received from family members and friends, presence of co-morbidity, previous history of resolved mental problem and Depression Anxiety Stress Scale DASS- 21. Data was analysed by using the statistical computer software ‘Statistical Package for Social Science (SPSS)’ version 22 involving three levels of analysis (descriptive, chi square and multiple logistic regression). Results: Only 31.1% of the respon- dents were found to be depressed with mainly within the moderately depressed category, 53.9% had anxiety and 26% were having stress. Perceived social support received from family members was a significant predicting factor for depression, anxiety and stress. Conclusion: High proportion of depression, anxiety and level of stress among medical students were found. The stressful medical program is putting medical students at risk of mental problems. Parents and family members should be encouraged to be more involved in any related programs to promote better mental health and well-being of medical students.

    Introduction: Children with visual impairment are reported to be at higher risk of poor oral health. They faced dif- ficulties in basic skills including oral care. For this reason, they may develop dental caries and periodontal disease. The aim of this study was to assess the oral health status of a group of visually impaired school children in Kuala Lumpur. Methods: A cross-sectional study was carried out on students from a Special Education School in Kuala Lumpur. Oral examination was conducted to determine caries experience and oral hygiene status. Caries was mea- sured using DMFT index and simplified oral hygiene index (OHI-S) for oral hygiene status. Oral examination was based on modified World Health Organization (WHO) oral health assessment criteria for children. Chi-square test was used to determine associations between demographic variables and oral health status. Results: A total of 91 visually impaired students (41 blind, 50 low vision) within the age group of 13 to 17 years old participated in this study. The mean OHI-S score was 1.68 (SD 0.87) with majority of students had fair oral hygiene (39.6%) and 29.7% with poor and very poor oral hygiene. The mean DMFT score of total students were 0.80 (SD 1.62). Male students demonstrated significantly better oral health status than female in relation to prevalence and mean score of dental caries. Conclusion: Most of the visually impaired children in this study showed fair oral health status. Regular oral health education is recommended to improve the oral hygiene especially in the totally blind group.

    Objectives: This study was carried out to determine the nutritional status of primary
    school children aged 6 to 11 years in Sepang District and to compare the results with a
    similar study carried out in 1999. Methods: This cross-sectional study was carried out in
    Sepang District using stratified random cluster sampling technique. A pre-tested, structured
    questionnaire was used to collect the data. Weight was recorded using a digital bathroom
    scale TANITA model HD-309 and height was measured by using a SECA Body meter
    Model 208. Body mass index (BMI) was calculated as weight in kilograms divided by the
    square of height in meters. Classifications of the nutritional status of respondents was
    based on WHO classification 1983 and 1995. Data were analysed using SPSS. Results:
    The overall mean BMI for the 1,910 respondents was 16.8 (95% CI= 16.67 - 16.99;
    SE=0.08). The median BMI was 15 .7 ranging from 9.51 to 36.75 kg/m2. The majority
    (84.4%) had normal weight-for-age status, 7.6% respondents had weight-for-age status
    below -2 SD of the NCHS-WHO median and 8.0% had weight-for-age status above +2 SD
    of the NCHS-WHO median. The prevalence of underweight and overweight was significantly
    higher in the males compared to the females (p< 0.05). There was a significant association
    between underweight and ethnicity (p< 0.05). The majority (90.3%) had normal heightfor-
    age status, 8.8% had height-for-age below - 2 SD of the NCHS-WHO standard median
    and 0.9% had height-for-age above 2 SD from the NCHS median. The prevalence of
    stunting was significantly higher in the male (10.3%) as compared to 7.4% in the females
    (X2 = 5.11, P = 0.02). The majority (81.4%) of the respondents were within the normal
    weight-for-height, 3.4% were wasted (weight-for-height was below - 2 SD of the NCHSWHO
    median) and 15.2% of the respondents were obese (weight-for-height that is more
    than +2 SD of the NCHS-WHO median). The prevalence of wasting was higher in the
    females (4.1 %) compared to males (2.8%). The prevalence of obesity was slightly higher
    in the males (15.4%) compared to females (14.9%). Conclusion: The overall nutritional
    status of primary schools children is good. However, there still exists pockets of population
    in Sepang where chronic and acute forms of undernutrition persist in these children.

    Overweight and obesity have become a serious health issue in worldwide and Malaysia is no exception. In view that obesity is one of the leading causes of non-communicable diseases, this systematic review aimed to explore whether Malaysian are getting fatter (overweight or obesity) and rounder (abdominal obesity). Searches of the published literature were conducted using 3 databases including EMBASE, MEDLINE, and PubMed. These were limited to cross-sectional or cohort studies in Malaysia. The databases provided the prevalence of overweight, obesity, and abdominal obesity among 15 years old and above Malaysian population from 2009 to 2015. In total, 47 articles were included in this review. Findings exhibit that the prevalence rates of overweight, obesity, and abdominal obesity range from 9.5% to 43.5%, 1.77% to 26.4% and 11.4% to 57.4%, respectively. Overweight is more predominant among males. However, obesity and abdominal obesity are more predominant among females. This means that more females are getting fatter and “rounder” as compared to males in Malaysia. The findings imply a lack of intervention programmes based on proper randomized controlled trials which are needed to adequately address the problem of obesity in Malaysia.

    Introduction: Neurodegenerative diseases such as Alzheimer’s disease (AD) and Parkinson’s disease (PD) have become increasingly prevalent in recent years. Microglia, the resident myeloid cells of the Central Nervous System (CNS) are primarily responsible for the production of inflammatory mediators that accumulate and become toxic during a chronic inflammatory response. The accumulation of inflammatory mediators over time inadvertently contributes to the functional impairment of surrounding neurons. Hence, suppressing microglial cell activation can be a solution to control the progress of neurodegenerative disorders. Symptomatic treatments are available, but no curative treatment is currently available, and some are linked to several side effects associated with their use. Honey is a natural product derived from the nectar harvested and modified by honeybees. Its therapeutic effects are widely documented, have been tested and verified extensively in literature. Honey is recognized in modern medicine for its varied pharmacological activities. While the medicinal properties of honeys such as Manuka honey are well established, further investigation is required to elucidate the medicinal properties of locally sourced honeys, namely Tualang (TH) and Kelulut (KH) honeys. In this study, we investigated the immunomodulatory effects of local TH and KH honey on microglial cell activities. Methods: BV2 cells, an immortalized microglial cell line was used in this in vitro study to assess the cell survival when treated with the TH and KH honey. Expression of CD40, CD11b and CD86 were measured using flowcytometry. Results: BV2 cells incubated with TH at concentrations of 0.1% and 0.5%, and KH at concentrations of 0.1% and 0.25% for 24 and 48 hours showed cell survivability above 75%. Both TH and KH decreased ROS production significantly on LPS-induced BV2 cells, but increased ROS production on unstimulated BV2 cells. Additionally, the expression levels of CD40, CD11b and CD86 were also reduced on honey- treated LPS-induced BV2 cells. Conclusion: These results have demonstrated that both TH and KH are capable of suppressing microglial activation. Therefore, we propose the idea of utilizing these honeys as a complementary treatment to suppress the progression of neurodegenerative diseases.

    Objectives: This study aimed to evaluate the UBI MAGIWELTM ζ-GLOBIN ELISA Kit for the presumptive diagnosis of αo-thalassaemia. The ELISA results obtained were confirmed by molecular characterisation of αo-thalassaemia using a Duplex-PCR. Methods: Routine peripheral blood counts and red cell indices were determined in 94 blood samples sent for Hb analysis. Hb subtypes were quantified by high performance liquid chromatography (HPLC) and Hb electrophoresis conducted on agarose gel at pH 8.5. Zeta-globin chain levels were determined using the UBI MAGIWELTM ζ-GLOBIN ELISA Kit. Molecular analysis was performed using a duplex-PCR which simultaneously amplifies
    a normal 136 bp sequence between the ψα−α2-globin genes and a 730 bp Southeast Asian deletion-specific sequence (–SEA) between the ψα2−θ1-globin genes.     Beta (β)- thalassaemia is a public health problem in Malaysia. The carrier rate is estimated to be 4.5% by micro-mapping studies particularly among Malays who comprise 53.5% of the population in Malaysia. The common diagnostic method in Malaysia for mutation detection is by amplification refractory mutation system (ARMS). It allows single mutation detection in each reaction but is labour intensive and time consuming when many mutations need to be identified. The purpose of this study was to develop a diagnostic tool for effective mutation detection of beta thalassaemia in Malay patients and compare its efficacy with ARMS-PCR, the current method in use. Methods: Reverse dot blot hybridization (RDBH) technique was incorporated in the development of two strip assays [RDBH-Strip M(6) and RDBH-Strip C(6)] to identify common beta thalassaemia mutations in the Malays. The panels of selected mutations were based on the mutation frequencies in Malaysia reported in previous studies. RDBH-Strip M(6) was applied as step 1 and RDBH-Strip C(6) was applied as step 2 for unidentified mutations. The strips were validated with the gold standard method, ARMS- PCR. Results: One hundred and thirty seven Malay patients with 274 alleles were studied. In Step 1 mutation detection, 238 alleles (86.9%) were identified in 119 of patients by RDBH-Strip M(6). Step 2 resulted in a further detection of 20 alleles in another 10 patients by RDBH-Strip C(6). The combination of both strips resulted in the identification of 258 alleles in 129 (94.6%) of 137 Malay patients. The strip assays were 100% sensitive and specific when compared with ARMS-PCR method. Conclusion: Two strip assays utilising the RDBH technique were developed to identify common β-thalassaemia mutations in Malays. The RDBH Strip M(6) identified 86.9% of the mutations and the RDBH-Strip C (6) detected further 7.3% alleles. This two step strategy was found to be rapid and cost effective for the direct diagnosis of β-thalassaemia mutations in the Malays. The remaining unidentified mutations would require DNA sequencing. It can serve as a specific molecular diagnostic tool for effective diagnosis of
    β-thalassaemia mutations in this ethnic group.

    Introduction: Type 2 diabetes is increasing at an alarming rate worldwide. One of the leading factors to this condition is obesity. Low glycemic index (GI) and glycemic load (GL) diets have been proposed as lifestyle changes to address obesity, however, there is a lack of consensus on the optimal approach for weight loss, glycemic control and improving insulin sensitivity. In addition, the outcome of these diets are equivocal, with some studies suggesting beneficial outcomes and others suggesting otherwise. Furthermore, discrepant study designs have led to divergent conclusions. In order to provide a comprehensive overview of the low GI and low GL diets, a systematic review of literature on relevant observational studies and randomised control trials was performed on these databases:- The Cochrane Library, Medline, PubMed, Embase, Cinahl and Web of Science. Methods: The review was conducted based on the methodological standards for the conduct and reporting of Cochrane intervention reviews, Version 1.07, November 2018. Population, Intervention, Comparison and Outcomes (PICO) tool was used as the organising framework to define key elements of the review question. Results: Pertinent outcome variables include body weight, insulin resistance, HbA1c, fasting serum glucose, BMI, waist-to-hip ratio, triglyceride, HDL and LDL cholesterol. Our current understanding of these diets has been complicated by the reports that were based on different study designs and study populations. This review defines the issues, gaps in the research, study design, and evidence that is needed to inform practice, policy making and future research. There is also a dearth of information on the effect of low GI and GL diets on the Asian populations, specifically on improving insulin resistance. High carbohydrate diets are a mainstay of Asian societies. Conclusion: As cases of obesity and type 2 diabetes surge, there is an urgent need for research on low GI and GL dietary modifications among the Asian populations.

    Introduction: Despite advancement of treatment modalities, Tuberculosis (TB) treatment interruption rate has glob- ally accelerating, calling for greater framework shifting towards psychosocial intervention. Similarly, Selangor state had reported the perturbing TB treatment interruption rate, which was figured persistently above 10% in the interval year of 2014 to 2018, thus signifies an empirical assessment on Information-Motivation-Behavioural skills (IMB) determinants of TB intensive phase treatment. This study aims to determine the time to intensive phase TB treatment interruption and its prognostic factors among newly diagnosed pulmonary Tuberculosis (PTB) smear positive patients in urban district Selangor. Methods: A multi-centric prospective cohort study will recruit 695 newly diagnosed PTB smear positive patients at treatment centres in urban districts, Selangor. This study will utilize validated self-admin- istered questionnaire and standardised data collection form (PROFORMA). At baseline, we will elicit information on IMB models constructs, additionally on socio-demographics, health service factors and clinical characteristics. Meanwhile, four points follow up will be executed to retrieve information on treatment status and time varying effects of body weight, treatment side effects, symptoms improvement and internalised stigma. Finally, survival analysis will be computed to identify the time to intensive phase treatment interruption and its prognostic factors. Conclusion: This study will enlighten IMB model determinants of intensive phase treatment interruption, hence to endeavour psychosocial elements in designing time relevant public health strategies in TB case management.

    Introduction: Diabetic control depends partially on self-management. Diabetes self-care is reported to be a strong predictor of glycemic control in Type 2 Diabetes Mellitus (T2DM) patients. Factors contributing to better self-care include self-efficacy and social support. We aim to determine effects of these factors on glucose control in T2DM patients in a primary care clinic. Methods: This study utilised data from a bigger study looking into factors affecting social support and self-efficacy and their correlation in T2DM patients. This study was done among 329 T2DM pa- tients attending a primary care clinic in Selangor, Malaysia between November 2013- January 2014. Systematic ran- dom sampling was used. Instruments used were the MOS Social Support Survey and the Diabetic Management Self Efficacy Scale (DMSES). Patients’ socio-demographic data and glucose monitoring records were obtained. The asso- ciation between social support and self-efficacy with blood glucose control was determined using logistic regression. Results: The mean age of the respondents is 54.6 (+11.2), mostly female (55.3%) and Malay ethnicity (63.5%). Being older and retired were predictors for good glucose control (OR=1.05,p=0.01 and OR=2.2,p=0.023) respectively. There is also significant correlations between number of close friends/relatives with social support and self-efficacy (r=0.162,p=0.030 and r=0.190,p=0.01) respectively. The correlation of social support and self-efficacy with HbA1c levels are r= -0.044(p=0.426) and r= -0.08(p= 0.744) respectively. Conclusion: There were no correlations between social support and self-efficacy with the level of HbA1c among patients with T2DM in this study. We found older aged and retired patients to have better HbA1c levels.

    With the availability of the Internet, pornographic materials are readily accessible. Pornographic materials may have a link to addiction and other deviant sexual behaviour. In this case report, we highlighted a link between voyeuristic disorder and internet sex addiction. Mr. SK is a 22-year-old single gentleman who was referred for psychiatric assessment due to voyeuristic acts. He had been watching pornographic materials since 16 years old and became addicted to it.

    The National Health and Morbidity Survey 2011 was a nationally representative household survey of non-institutionalized Malaysian population who were residing in Malaysia for at least 2 weeks prior to data collection. The aim of the survey was to provide health related community–based data and information to support Ministry of Health, Malaysia, in reviewing health priorities, programme strategies and activities, and planning for allocation of resources. There were twelve research scopes included in the survey. The sample size was calculated based on the requirement for each scope. A two-stage stratified sampling was adopted in the survey. The methods for data collection were via the questionnaire, clinical examination, and biochemical analysis. Quality controls were also instituted to ensure collection of high quality data. The National Health and Morbidity Survey 2011 (NHMS 2011) adopted an appropriate methodology for a population survey and all the necessary steps were taken to ensure valid and reliable findings.

    Introduction:The increasing prevalence of childhood obesity among primary school children is a growing public health concern worldwide. Previous study in West Malaysia showed a prevalence of obesity with an estimated of 6.6% among 7-year-olds children and rising to 13.8% among 10-year-olds children. However, no study has ever been conducted in East Malaysia on childhood obesity. This study was aimed to determine the prevalence of child-hood obesity and to investigate the association of physical activity levels with obesity and its intervention among primary school children in Kota Kinabalu. Methods: A cross-sectional study involving 484 students (aged 7-12 years old) was conducted in five primary schools at Kota Kinabalu, Sabah. The schools were selected based on cluster sampling method and simple random sampling method. Primary school children with a known medical condition such as bronchial asthma, type-1 diabetes mellitus or those with a prolonged use of oral steroids was excluded from the study. Self developed questionnaires were used to record sociodemographic details and anthropometric mea-sures both parents and children, and physical activity was measured using a Children Physical Activity Questionnaire (C-PAQ). The intervention involved 3 components; I) motivational interviewing, ii) brief and focused advice, iii) ac-tivity session. Follow up was done 3 months after the intervention was conducted. Results: The prevalence of child-hood obesity among primary school children is 13.2%. There is a negative correlation between children’s BMI and amount of physical activity (-0.347) and positive correlation between children’s BMI and amount of non-physical activity (0.339). Little changes in the BMI of the children after intervention, t(127)= 2.866, p=0.005. Conclusion: This study provides evidence that childhood obesity is not a prevalent health problem in Kota Kinabalu and has proved that physical activity levels have significant associations with children’s body mass indices.

    Introduction: Role of nurses is very important in raising awareness, motivation level and increasing rate of organ donation subsequently. There are many studies on critical care nurses’ roles and education affect organ donation. Methods: A narrative literature review article of published research. Twelve published articles from ten different countries were found using different renowned online search engines. Results: Knowledge about organ donation among nursing students almost all over the world is inadequate than expectation and requirement in respective country. Marlene R et al showed that scores on the knowledge, attitudes, and beliefs subscales were positively cor- related with the number of requests for tissue and organs and consents obtained (0.76); and consents obtained for tissues and consents obtained for organs (0.72). Professional education was indicated in 43% of nurses surpassing family beliefs, religion and mass media. The cognitive part (knowledge) is directly depends on non-cognitive (Aware- ness and Attitude). Findings from almost all articles suggest the need of adding organ donation topic in the nursing and medical curriculum to prepare competent nurses and health care providers for future. Conclusion: This study suggests that there is a need to integrate organ donation aspects into the educational program to prepare nurses for their roles in organ and tissue procurement as well as awareness among policy makers in this regard for the well-be- ing of our patients.

    Introduction: Cholera is an infection caused by the bacterium Vibriocholerae that causes severe watery diarrhoea, which can lead to dehydration. Beet-roothas been shown to induce favourable outcomes and holds promise as an economic, practical natural dietary intervention in cholera. Methods: Systematic search of peer-reviewed literature on beet-root ORS and carrot ORS use for acute diarrhoea in below 6-year-old. We identified 3 studies for abstraction. Results: A study had compared a commercial carrot/rice-based ORS A (Na 52 mmol/L) and two glucose-based ORS B (Na 55 mmol/L) and C (Na 90 mmol/L). Fluid intake, faecal and urine output and absorption of fluid was measured in 161 infants and children (3-48 months old) during the first 48 hours after admission. The number of stools (p < 0.01) and the mean faecal output (p < 0.05) per kg body weight were significantly lower in group A. Children in group A also had significantly (p < 0.01) greater fluid absorption (mean 464 ml/kg) than in groups C (312 ml/kg) and B (140 ml/kg). Conclusions: While there are some promising results, this analysis indicates that the need for further investi- gation into approaches to increasing beet-root ORS and carrot ORS use.

    Introduction: Recent Coronavirus outbreak has raised concern among student nurses who are doing their clinical posting tenure regarding the risk ofpatient acquired infection. This study examined perceived readiness to pro- vide coronavirus patient careduring clinical posting among student nurses in UMS. Method: This study deployed a cross-section survey design using a self-rated questionnaire to evaluate respondents’ level of readiness towards Coronavirus patient care. A total of 177 (N) respondents comprising of nursing students from UMS (year 1 n=55, year 2 n=56, year 3 n=66) participated in the study. The study questionnaire captured demographics and comprised of 15Likert -scale items that assessed the level of perceived readiness to provide Coronavirus patient care. The ques- tionnaire was adapted from a previous SARs pandemic studyand revalidated within the local context (α = 0.78). Data analysis used descriptive statistics by frequency counts and Fisher exact test for demographic correlates with the level of readiness. Results: Among 177 student nurses in this study68% (n= 121) were willing and ready to provide Coronavirus patient care and 32% (n = 56) were hesitant or not willing. Regarding confidence of safety using PPE to provide patient care, 34 % (n = 61) of respondents rated not confident and deemed it hazardous even with full PPE use. On the issue of if legally mandated to care for Coronavirus patients, 22% were hesitant to provide care and will considerleaving nursing training if compelled to do so. There was a significantly lower perceived level of readiness towards Coronavirus patient care among 1st-year student nurses and those who had not attended any Coronavirus educational session (p= 0.06). Conclusion: The findings indicate although the majority of student nurses are willing to provide care for Coronavirus infected patients during their clinical posting tenure, a small proportion of student’s nurses were hesitant.

    Introduction: Malaria is a public health concern and still endemic in Malaysia particularly in Sabah. Malaria is due to plasmodium parasite which can be further divided into human malaria and zoonotic malaria. Plasmodium knowlesi, a zoonotic malaria species, is increasing in trend and alarming despite of ongoing prevention and control measures especially in Sabah. Methods: This is a case-control study involving a total of 169 respondents which comprises of 43 cases and 126 controls from Mukim Paginatan and Mukim Nalapak, Ranau. A simple random sampling were used to select subjects. Results: This study found male, forest related and doing outdoor activity has higher risk of contracting malaria. Village house is another significant risk factor. The specific risk factors with the strongest association with malaria is the presence of monkey near housing or farm area (OR5.667; 95% CI: 2.429, 13.220), live near forest (OR 5.330; 95% CI: 1.209, 23.495), presence of pond or pool or river near house (OR 4.222; 95% CI: 1.552, 11.490), doing outdoor activity after 5 PM (OR 3.434; 95% CI: 1.522, 7.748) and ever overnight in the hut (OR 2.724; 95% CI: 1.334, 5.561). Conclusion: This study shows several risk factors that associated with malaria knowlesi. Based on the identified risk factors, preventive and control measure can be improved and optimized.

    Introduction: Food poisoning usually occurs with the consumption of contaminated food. Some related factors are unsafe water supply, poor sanitation, unhygienic waste disposal and unhygienic practices or poor personal hygiene by food handlers. The purpose of this study is to describe the spatial epidemiology of food poisoning cases in the four districts of Sabah. Methods: This review consists of all food poisoning cases reported from 2011 to 2014 from Kota Kinabalu, Penampang, Putatan and Papar, Sabah. The coordinates used for locations of cases are based on home addresses. Tools such as SPSS v20, ArcGIS v10 and CrimeStat IV were used for data analysis and mapping. Results: A total of 1,787 cases of food poisoning were reported during this review period. In 2011, only Kota Kinabalu and Pa-par illustrated significant food poisoning clusters. Meanwhile, in the year 2012 to 2014, Kota Kinabalu, Penampang and Putatan had clustering of cases. Analysis of nearest neighbour hierarchical clustering analysis showed 32 food poisoning clusters. There were 4 food poisoning points at 500 meters radius around a market place, 2 food poisoning points near a sewage plant and 1 food poisoning point near a water treatment plant. No cases were near a municipal landfill. For rivers and coastline, there were 37 points of food poisoning cases in the proximity of 500 meters. Con-clusion: Food poisoning usually occurs in clusters with possible associated environmental factors.

    CD4+CD25+ Foxp3+ T regulatory cell (Tregs) represents approximately 8-10% of the total CD4+ T cell population and are important for immune homeostasis and preventing autoimmune development. Thus, harnessing their functions as immune modulator may be coupled with the rapid advancement of nanotechnology development. Plant-mediated biosynthesis of silver nanoparticle (AgNP) is noteworthy due to simplicity, rapid rate and potentially render more biocompatibility with biomolecules. This study identified the effect of biosynthesized-AgNPs from Garcinia atroviridis (GA) in modulating inflammatory properties of Treg cells in Non-Obese Resistant (NOR). GA extract was used to biosynthesized AgNPs and was tested on the effect of inducing inflammatory properties in CD4+IL17Rhigh cells following 72hr in vitro treatment. Methods: Conventional CD4+CD25-Foxp3- cells from female NOR mice were sorted using magnetic separation and cultured in RPMI in the presence of anti-CD3/CD28 antibodies, TGF-β and IL-2 cytokines. Cells were then treated with or without GA-AgNPs for 48hr of iTreg cell induction and then re-cultured with new media treated with respective treatments received. After 72hr in vitro culture, cells were stained with fluorochrome-conjugated antibodies for flow cytometry. Results: Current result showed that AgNPs suppress CD4 expression in CD4+IL17Rhigh population. MAPK pathway proteins remain unchanged in both control and AgNP-treated groups. Conclusions: The preliminary findings may suggest the properties of GA-AgNPs in modulating CD4+ T cell population in normal condition. Further studies are necessary to elucidate the molecular mechanisms involve in such interaction. Current findings serve as basis in further identifying the immunomodulatory profile of nanoparticle for potential therapeutic use.
