The objective of this research is to examine the relationship between the color changes of phenol red and the growth of cancer cells, i.e., HeLa and DU145 cells, over a specific period of time. Normal mouse skin fibroblasts (L929 cells) were used as a reference. In this research, the color changes of phenol red due to the acidification of the cell culture medium from the growth of the cells over a period of nine hours showed potential colorimetric characteristics of cancer cells. The color changes of phenol red were observed using visible absorbance spectroscopy. The transformation of the absorbance spectra into coefficients of determination against the examined range of wavelengths created a distinctive spectral signature that signifies phenol red discoloration in cancer and normal cell culture lines.
Cresol Red, a commercial dye that used widely to color nylon, wool, cotton, and polyacrylonitrile-modified nylon in the massive textile manufacture is toxic recalcitrant. Absidia spinosa M15, a novel fungal strain isolated from a tropical rain forest, was found to decolorize Cresol Red 65% within 30 d under agitation condition. UV-Vis spectroscopy, TLC analysis and mass spectra of samples after decolorization process in culture medium confirmed final decolorization of Cresol Red. Two metabolites were identified in the treated medium: benzeneacetic acid (tR 9.6 min and m/z 136) and benzoic acid (tR 5.7 min and m/z 122). Laccase showed the significant activity (133.8 U/L) in biomass obtained at the end of experiment demonstrates role of the enzyme in the decolorization process.
Endophytes are novel sources of natural bioactive compounds. This study seeks endophytes that produce the anticancer enzyme l-asparaginase, to harness their potential for mass production. Four plants with anticancer properties; Cymbopogon citratus, Murraya koenigii, Oldenlandia diffusa and Pereskia bleo, were selected as host plants. l-Asparaginase-producing endophytes were detected by the formation of pink zones on agar, a result of hydrolyzes of asparagine into aspartic acid and ammonia that converts the phenol red dye indicator from yellow (acidic condition) to pink (alkaline condition). The anticancer enzyme asparaginase was further quantified via Nesslerization. Results revealed that a total of 89 morphotypes were isolated; mostly from P. bleo (40), followed by O. diffusa (25), C. citratus (14) and M. koenigii (10). Only 25 of these morphotypes produced l-asparaginase, mostly from P. bleo and their asparaginase activities were between 0.0069 and 0.025 μM mL(-1) min(-1). l-Asparaginase producing isolates were identified as probable species of the genus Colletotrichum, Fusarium, Phoma and Penicillium. Studies here revealed that endophytes are good alternative sources for l-asparaginase production and they can be sourced from anticancer plants, particularly P. bleo.
OBJECTIVE: Asians are known to have different tear characteristics compared to Caucasians that may affect contact lens wear. There are scanty research studies that have evaluated tears during continuous wear contact lens in Asia. The present study aims to evaluate changes in tears in subjects wearing continuous wear rigid gas permeable contact lens (CWRGP) for 6 months.
MATERIALS AND METHODS: Thirty five neophyte subjects (21 females, 14 females) were recruited for this study. Subjects were fitted with CWRGP lenses with Dk of 163 on both eyes. Tear was evaluated using Phenol red thread test (PRT), tear break up time (TBUT) test and tear meniscus height (TMH) measurement. Non parametric and parametric analyses were used to compare the parameters.
RESULTS: Values at baseline (BL) and six months (6M) were as follow: PRT, BL=19.10 ± 3.86 mm, 6M= 21.02 ± 4.27 mm, TBUT, BL= 8.58 ± 4.90 sec, 6M=8.08 ± 5.32 sec, TMH, BL= 0.38 ± 0.12 mm, 6M= 0.34 ± 0.07 mm. Statistical analysis showed significant difference in tear volume for PRT only at 6 months (p=0.007).
CONCLUSION: Our analysis showed minimal change in the tear characteristics after six months of CWRGP lens wear, which indicated low impact of CWRGP contact lens on tears characteristics of Asian eyes. However, careful monitoring is required to prevent development of adverse events during contact lens wear.
In this work, zinc chromite (ZnCr2O4) nanostructures have been synthesized through co-precipitation method. The effect of various parameters such as alkaline agent, pH value, and capping agent type was investigated on purity, particle size and morphology of samples. It was found that particle size and morphology of the products could be greatly influenced via these parameters. The synthesized products were characterized by field emission scanning electron microscopy (FESEM), X-ray diffraction (XRD), fourier transform infrared (FT-IR) spectra, X-ray energy dispersive spectroscopy (EDS), photoluminescence (PL) spectroscopy, diffuse reflectance spectroscopy (DRS) and vibrating sample magnetometry (VSM). The superhydrophilicity of the calcined oxides was investigated by wetting experiments and a sessile drop technique which carried out at room temperature in air to determine the surface and interfacial interactions. Furthermore, the photocatalytic activity of ZnCr2O4 nanoparticles was confirmed by degradation of anionic dyes such as Eosin-Y and phenol red under UV light irradiation. The obtained ZnCr2O4 nanoparticles exhibit a paramagnetic behavior although bulk ZnCr2O4 is antiferromagnetic, this change in magnetic property can be ascribed to finite size effects.
