    Sinigrin, a precursor of allyl isothiocyanate, present in the Raphanus sativus exhibits diverse biological activities, and has an immense role against cancer proliferation. Therefore, the objective of this study was to quantify the sinigrin in the R. sativus roots using developed and validated RP-HPLC method and further evaluated its' anticancer activity. To achieve the objective, the roots of R. sativus were lyophilized to obtain a stable powder, which were extracted and passed through an ion-exchange column to obtain sinigrin-rich fraction. The RP-HPLC method using C18 analytical column was used for chromatographic separation and quantification of sinigrin in the prepared fraction, which was attained using the mobile phase consisting of 20 mM tetrabutylammonium: acetonitrile (80:20%, v/v at pH 7.0) at a flow rate of 0.5 mL/min. The chromatographic peak for sinigrin was showed at 3.592 min for pure sinigrin, where a good linearity was achieved within the concentration range of 50 to 800 µg/mL (R2 > 0.99), with an excellent accuracy (-1.37% and -1.29%) and precision (1.43% and 0.94%), for intra and inter-day, respectively. Finally, the MTT assay was performed for the sinigrin-rich fraction using three different human cancer cell lines, viz. prostate cancer (DU-145), colon adenocarcinoma (HCT-15), and melanoma (A-375). The cell-based assays were extended to conduct apoptotic and caspase-3 activities, to determine the mechanism of action of sinigrin in the treatment of cancer. MTT assay showed IC50 values of 15.88, 21.42, and 24.58 µg/mL for DU-145, HCT-15, and A-375 cell lines, respectively. Increased cellular apoptosis and caspase-3 expression were observed with sinigrin-rich fraction, indicating significant increase in overexpression of caspase-3 in DU-145 cells. In conclusion, a simple, sensitive, fast, and accurate RP-HPLC method was developed for the estimation of sinigrin in the prepared fraction. The data observed here indicate that sinigrin can be beneficial in treating prostate cancer possibly by inducing apoptosis.

    The present study investigates heavy metal uptake and protein expression by different vegetables collected from various districts of Khyber Pakhtunkhawa province of Pakistan. Statistical analysis of the data showed that maximum concentration of Cd, Cr, Ni, Zn were found in radish and spinach, respectively, collected from Peshawar. Maximum Pb and Mg accumulation were found in cauliflower and pea at Swat followed by coriander at Haripur and minimum Pb uptake was noticed in radish taken from Nowshehra. Highest Cu uptake was detected in spinach at Nowshehra. Data regarding Cd, Cr, Pb, Cu, Ni, Mg and Zn concentration in water samples gathered from different sites of KPK indicated that maximum concentration of Cd was observed in Swat. Maximum Cr and Cu concentration were measured in water samples from Peshawar while maximum concentration of Pb and Ni were detected in water samples from Haripur. In case of soil samples, maximum Cd, Cr, Mg and Zn uptake was observed in soil sample at Nowshehra. Maximum Pb and Ni concentration was found in soil samples collected from Peshawar. Cu concentration was observed to be the highest in soil at Swat. Protein profile of different vegetables i.e. cauliflower, radish, carrot, turnip, pea, spinach, coriander and garlic sampled across five different sites showed that uptake of Cd, Cr, Pb, Cu, Ni, Mg and Zn by these vegetables caused the expression of numerous polypeptides.

    In this study, the antimicrobial, antioxidant, phytotoxic and phytochemical properties of defatted seeds of Jatropha curcas were evaluated. A crude methanolic extract of defatted seeds was tested against three fungal strains - Aspergillus niger, Aspergillus flavus and Aspergillus fumigatus - and five bacteria: Escherichia coli and Klebsiella pneumoniae (Gram negative) and Micrococcus luteus, Bacillus subtilis and Staphylococcus aureus (Gram positive). The methanolic extract was diluted in dimethylsulfoxide to final concentrations of 1, 2, 3, 4 and 5 mg/10 mL. The largest zones of inhibition against K. pneumoniae, M. luteus and B. subtilis were achieved using the concentration of 5 mg/10 mL. The concentration of 1 mg/10 mL was most effective against S. aureus and E. coli. In a 1, 1-diphenyl-2-picrylahydrazyl (DPPH) radical scavenging assay, the 5 mg/10 mL concentration of the Jatropha seed extract showed the strongest activity. Higher concentrations of the Jatropha seed extract (10 mg/50 mL and 5 mg/50 mL) significantly inhibited the germination of radish seeds and had negative effects on radish seedling relative water content, shoot length, root length, seedling fresh weight and seedling dry weight (p<0.05). Phytochemical analyses of the defatted seeds detected alkaloids (7.3%), flavonoids (0.39%) and soluble phenolics (mg gallic acid equivalents/g extract). Based on these results, it was inferred that J. curcas seeds contain active ingredients that are effective against pathogenic microbes and therefore could be used to formulate drugs to treat various diseases.

    The organic foods’ market is becoming one of the rapidly growing sections in agricultural economies in the world. During the last two decades, food-borne outbreaks associated with fresh produce have rapidly increased. E. coli O57:H7, the caustic agent of acute hemorrhagic diarrhea and abdominal cramps, is mainly associated with meat and poultry product outbreaks but frequent outbreaks linked to the consumption of vegetables have been reported. The aim of this study was to investigate prevalence of E. coli O157:H7 in some organic foods. A total of 230 organic food samples including four-winged bean, tomato, white radish, red cabbage, chinese cabbage, lettuce, cucumber and chicken form retailed groceries and supermarkets in Malaysia were investigated. Low prevalence of E. coli O157:H7 was detected in organic vegetables and chickens. The estimated quantity of E. coli O157:H7 in all samples ranged from 2400 MPN/g. The overall MPN/g estimate of E. coli O157:H7 in the samples from organic groceries was higher than supermarket with the maximum of >2400 MPN/g.
