    Roasting of whole-kernels is an important step in the production of pistachio paste. The effect of hot air roasting temperatures (90-190°C) and times (5-65 min) on the hardness, moisture content and colour attributes (‘L’, ‘a’ and ‘b’ values and yellowness index) of both whole-kernel and ground-state were investigated using response surface methodology (RSM). Increases in roasting temperature and time caused a decrease in all the responses except for ‘a’ value of ground-state. The interaction and quadratic models sufficiently described the changes in the hardness and colour values, respectively. The result of RSM analysis showed that hardness and colour attributes (‘L’ and ‘b’ values, yellowness index) of kernels and ‘a’ value of ground-state could be used to monitor the roasting quality of whole-kernels. This study showed that the recommended range of roasting temperature and time of whole-kernel for the production of pistachio paste were 130-140°C and 30-40 min, respectively.

    Proximate content and fatty acid composition of germinated and non-germinated legumes (kidney, mung, soy bean and peanut) and rice varieties (red, black, Barrio, brown and milled) were evaluated. In germinated samples, moisture content increased significantly while carbohydrate, protein and fat were decreased significantly. Total dietary fibre was increased in germinated samples except germinated kidney and mung bean. Germination also increased saturated fatty acids (SFA) in legumes, black, red and brown rice. Monounsaturated fatty acids (MUFA) decreased in all samples except germinated kidney, soy and Barrio rice. Polyunsaturated fatty acids (PUFA) increased in some germinated samples (mung bean, peanut, red, brown, Barrio and white rice) but decreased in other legume and rice samples. Generally, palmitic acid increased while stearic, oleic and linoleic acids decreased after germination. Overall, the proximate content and fatty acids of legume and rice varieties changed after germination and may be used as alternate resources for individuals with lifestyle diseases.

    Little is known on the biosafety level of Vibrio spp. in freshwater fish in Malaysia. The purpose of this study was to investigate the prevalence and concentration of Vibrio spp. and V. parahaemolyticus in
    freshwater fish using the Most Probable Number-Polymerase Chain Reaction (MPN-PCR) method. The study was conducted on 150 samples from two types of freshwater fish commonly sold at hypermarkets, i.e. Pangasius hypophthalmus (catfish) and Oreochromis sp. (red tilapia). Sampling was done on the flesh, intestinal tract and gills of each fish. The prevalence of Vibrio spp. and V. parahaemolyticus was found to be 98.67% and 24% respectively with higher percentages detected in samples from the gills followed by the intestinal tract and flesh. Vibrio spp. was detected in almost all red tilapia and catfish samples. V. parahaemolyticus was detected in 25% of the catfish samples compared to 22.6% of red tilapia fish. The density of Vibrio spp. and V. parahaemolyticus in the samples ranged from 0 to 1.1x107 MPN/g. Although the maximum value was 1.1x107 MPN/g, most samples had microbial loads ranging from 0 to >104 MPN/g. The outcome on the biosafety assessment of Vibrio spp. and V. parahaemolyticus in freshwater fish indicates another potential source of food safety issues to consumers.

    Several Norovirus cases due to consumption of green onions have been reported during recent years but reports on red onions are not found. Onions are one of the major tastes in Malaysian food which are sometimes consuming raw especially the green onion. Viral contamination in onions can occur due to planting condition and not properly prepared food. This situation can pose the human health risk. A method was developed to detect the Norovirus that might present on different type of onions. In this study, 60 samples were collected from local market. Elution by Tryptose Phosphate Glycine broth and concentration steps using negatively charge filter were applied to enhance the detection of virus in food due to low copies of virus on food surface. The viral RNA was extracted using Qiagen Rneasy Mini kit before further detection using One-step RT-PCR. The total incidence of Norovirus in green onion and red onion was 13.33% (4/30) and 3.33 % (1/30) respectively. This is the first report of the detection of Norovirus in red and green onions in Malaysia. Based on the results, it is concluded that this method is reliable to detect Norovirus on onions and vegetables surface and hence can be applied in the laboratories for routine or food borne outbreak investigation.

    This research was to determine nutritional composition, essential and toxic elemental content, and major phenolic acid with antioxidant activity in Kadsura coccinea fruit. The results indicated that Kadsura coccinea fruit exhibited the high contents of total protein, total fat, ash and essential elements such as calcium (Ca), ferrum (Fe) and phosphorus (P). The levels of four common toxic elements, i.e. cadmium (Cd), mercury (Hg), arsenic (As) and lead (Pb), were lower than legal limits. By high-performance liquid chromatography (HPLC) analysis, gallic acid was identified as major phenolic acid in peel and pulp tissues. Its contents were no significant difference in both tissues. In comparison with two commercial antioxidants, the major phenolic acid extracted from Kadsura coccinea exhibited stronger 1,1-diphenyl-2-picrylhydrazyl radical-scavenging activity and reducing power. Kadsura coccinea fruit is a good source of nutrition and natural antioxidant. It is worthwhile to popularize this exotic fruit around the world.

    The optical and textural properties of chicken patty formulated with different level of grey oyster mushroom (Pleurotus sajor-caju) at 0, 25 or 50% to replace chicken meat were investigated. The addition of up to 50% oyster mushroom to chicken patty formulations did not change colour a*(redness),compared with the control patty. Chicken patties containing oyster mushroom had lower L* value ranging from 51.02 – 52.65 compared to that of the control patty (57.86). All oyster mushroom-based patties had lower colour b*(yellowness) value compared to chicken patty without mushroom. The hardness of chicken patty decreased proportionally with the level of oyster mushroom. On the other hand, oyster mushroom-based patties were springier than the control patty. Chicken which was replaced with 25% of fresh mushroom, recorded the highest moisture retention (77.19%) and cooking yield (80.71%), respectively. However, replacement of 25% of oyster mushroom with chicken breast in chicken patty formulation was not change the moisture retention, fat retention and cooking yield compared to control patty. Chicken patty added with 50% ground oyster mushroom the highest concentration of total dietary fibre (TDF) at 4.90 g/100 g compared to chicken patty containing 25% of mushroom (3.40 g/100 g) and control (1.90 g/100 g). In summary, the addition of oyster mushroom in chicken patties has decreased the lightness, yellowness, hardness and chewiness while no changes were noted in the redness of the patties.

    Genetically modified organisms (GMO) are increased remarkably from year to year and the estimated global area cultivated with genetically modified (GM) crops reached 125 million hectares in year 2008. However, insect resistance maize based on Bacillus thuringienses (Bt) is of the most cultivated GM crop in worldwide. Bacillus thuringiensis (Bt) is an aerobic, gram-positive bacterium that synthesize one or more Cry protein that are toxic to various types crop and forestry insects pests. To date, several cry genes have been introduced into GM plant to combat with various type of insect. Worldwide commercialization of GM crops has raised the customers’ concern about the Biosafety issues, and thus, many countries have implemented the labeling legislations for GM food and their derivatives. In this study, we introduced the quantitative analysis method based on the recombinant plasmid DNA as calibrators that can be used to determine the percentage of GMO content in various types of food and feed samples. Therefore, we have reported 7.5% (6/80) of the samples were contained StarLink maize and 1.25% (1/80) samples were contained Bt176 maize. Additionally, the percentage of GM content in each positive sample were further determined with the developed quantitative method. The percentage of the StarLink corns that present in the positive samples were varies from 0.09% to 2.53% and Bt176 corn that present in the positive sample was 16.90%. The present study demonstrated that the recombinant plasmid DNA that used in quantitative real-time method as good alternative quantitative analysis of GM content.

    This study was conducted to evaluate antimicrobial properties of ethanolic extracts of the leaves of Nephelium lappaceum, Curcuma longa, Cinnamomun cassia, Durio zibethinus, Vitex trifolia, Amaranthus tricolor, Syzygium samarangense and Manihot esculenta. Antibacterial properties of the extracts were studied against fifteen strains of different gram positive and gram negative pathogenic bacteria, including Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, Vibrio para, and Escherichia coli using the agar disk diffusion method. Among the tested extracts, only Amaranthus tricolor exhibited specific inhibition of one of the tested bacteria; Bacillus cereus. Using the microdilution method, its minimum inhibitory concentration (MIC) value was determined to be 20 mg/mL.

    A total of sixty V. parahaemolyticus strains isolated from local cockles (Anadara granosa) were investigated by their antibiotic resistance patterns and plasmid profiles. The isolates showed multiple resistances towards most of the antibiotics tested. All strains of V. parahaemolyticus isolated harbored 1-3 plasmids, with sizes ranging from 2.7 to 54 kb. All V. parahaemolyticus strains showed high multiple antibiotics in frequencies of 0.58 – 0.94 indicating that the strains were derived from high-risk sources. In addition, no particular plasmid profile was predictive of a particular pattern of antibiotic susceptibility. These findings are essential because of the suggested involvement of seafood especially shellfish and environment in transmission of this pathogen to human. Thus, indicating that seafood may be a source of food- acquired antibiotic resistant bacteria to consumer.

    Efficacy of some Malaysian herbal aqueous extracts, BHA/BHT (synthetic antioxidants) and ascorbic acid in retarding oxidative rancidity was tested with cakes. The development of lipid oxidation products during 15 days at room temperature was evaluated by means of Peroxide Value (PV) and Thiobarbituric acidreactive substances (TBARS) value. The six formulations consists of control sample (cake without addition of antioxidant) (F1), cake added with curry leaves extract (F2), cake incorporated with kesum leaves extract (F3), cake added with tenggek burung leaves extract (F4), cake incorporated with ascorbic acid (F5) and cake added with BHA/BHT (F6). Formulation with the incorporation of tenggek burung leaves extract showed powerful oxidative stability effect compared to the formulations with othe plant extracts and control sample. However, cakes with BHA/BHT showed the strongest oxidative stability throughout the storage period. Therefore, it is suggested that tenggek burung leaves extract can be added into the food system for effectiveness as antioxidant to prolong the shelf life of the product.

    The study investigated socio-demographic factors and product attributes affecting purchase decision of special rice by Malaysian consumer. The primary data were analyzed by using binary logit model.
    Demographic factors and consumer preference for special rice (with reference to basmati rice) attributes were identified to affect purchasing behavior for special rice. Size of household, marital status, number of children, household income and gender of consumers are the main socio-demographic factors that significantly influence households’ choices of special rice for home consumption in the Klang Valley area. The findings also suggest that product attributes such as flavor and aroma, availability, brand name and quality also influence the frequent purchasing of Basmati rice among the Malaysian consumers. However price and easy preparation are not significant in influencing the frequent purchasing of Basmati rice since most consumers are aware that special rice such as Basmati is expensive and all rice has to be prepared in a usual way.

    The fatty acid composition and trans fatty acid (TFA) contents of biscuits products were determined by gas chromatography, using a highly polar 100m capillary column (HP-88) and flame ionization detection. Total TFA ranged from 0.00 – 0.52 g/100 g total fatty acids and 0.12 – 0.68 g/100 g total fatty acids for local packed and unpacked biscuits, respectively. In imported biscuits, total TFA was higher ranging from 0.03 – 3.09 g/100 g of total fatty acids. Trans 16:1 was the most abundant, with values ranging from 0.01% to 38% followed by trans 18:1 Δ11 (0.01% - 13.11%), trans 18:1 Δ9 (0.01% - 4.68%), trans 18:2 (0.23% - 2.77%) and small quantities of trans 18:1 Δ6.     Pineapple (Ananas comosus), Bromeliaceae family, is a fruit grows in tropical countries including Malaysia. This fruit has several pharmacological benefits due to the presence of high concentration of bromelain (cysteine proteases). Condition of elevated temperature will induce deformation of enzyme and result in loss of activity. Sulfhydryl groups in cysteine proteases are readily to be oxidized and might account for the denaturation of bromelain at elevated temperature. Polyphenol from ethanolic cashew leave extract could be complexed with bromelain to stabilize the enzymatic activity. In thermal stability test, the heat damage effect on bromelain was ten times reduced after complexing with cashew extract. The enzymatic activity of free bromelain decreased gradually from 25 o C to 95 o C. Complexed bromelain was stable in activity to heating up to 85 o C. Bromelainpolyphenol complex showed a good heat resistance. The result revealed that polyphenol could protect bromelain in pineapple juice from heat denaturation.

    This study was conducted to determine the proximate composition and energetic value of selected marine fish and shellfish from the West Coast of Peninsular Malaysia. This study has included 20 species of fish (10 pelagic fish and 10 demersal fish) and 4 species of shellfish. For pelagic fish, Longtail shad (terubuk) was found to contain significantly lower moisture (59.31+0.00%), but significantly higher fat content (23.15+0.00%) compared to other fish species. For demersal fish, moisture and ash contents ranged between 74-82%, and 0.9-2.1%, respectively. Long-tailed butterfly ray contained the highest protein (22.22+1.24%) compared to other fish studied. The highest fat content of demersal fish was in Moonfish at 6.89+2.76%. For shellfish, prawn contained the highest protein (19.12+1.44%). The fat content of shellfish ranged between 1-2%. Oyster contained significantly higher carbohydrate at 6.45+0.00%, compared to other shellfish. Longtail shad contained the highest energetic value of 13.34 kJ g-1 of all samples. These values are useful references for consumers in order to choose fish and shellfish based on their nutritional contents.

    Keropok Lekor or frankfurter-like snack has long existed in Malaysia and has been sold in commercial retail shops as well as on the streets. The aims of this study were to analyze consumer’s perception and attitude towards keropok lekor and to determine consumer’s behavior on the product. Survey questionnaire was employed as a data collection tool. A total of 212 respondents from a university participated in this study. Results showed that more than 70% of the respondents chose to eat keropok lekor because of the taste. Half of the respondents chose black pepper as an innovative supplementary flavor of keropok lekor. This study also explores and analyses preliminary consumer behavior about keropok lekor such as satisfaction level, factors that drive consumption, product’s image from consumer perspectives and etc.

    This study evaluates the sanitary and physico-chemical quality of Sg. Jarum Mas shellfish waters in order to establish its suitability as a model farm. Seawater and shellfish from nine stations (4 shellfish harvesting waters, 4 surrounding waters and 1 control site) were collected and analyzed monthly from September 2004 - September 2005. The results show that shellfish harvesting waters in Sg. Jarum Mas can be classified as ‘approved’ and ‘conditionally approved’. Hepatitis A virus was not detected in any of the shellfish examined. Dinophysis caudata and Pseuodonitzshia spp. were the most common harmful alga species observed. Harmful species that are known to produce toxins and cause shellfish poisoning such as Alexandrium spp., Gymnodinum spp., Pyrodinium sp. and Prorocentrum spp. were not detected. The physico-chemical characteristics of shellfish waters in Sg. Jarum Mas imply that they are suitable for aquaculture activity of moderately tolerant species such as shellfish.

    This study aims to determine the frequency and density of potentially pathogenic Vibrio parahaemolyticus, defined as those possessing thermostable-direct hemolysin (tdh) and/or tdh-related hemolysin (trh) genes, in raw salad vegetables at retail level in Selangor, Malaysia. A combination of Most Probable Number - Polymerase Chain Reaction (MPN-PCR) method was applied to detect the presence of tdh and/or trh gene-possessing V. parahaemolyticus and to enumerate their density in the samples. A total of 276 samples of vegetables commonly eaten raw in Malaysia (Cabbage = 30; Carrot = 31; Cucumber = 28; Four winged bean = 26; Indian pennywort = 17; Japanese parsley = 21; Lettuce = 16; Long bean = 32; Sweet potato = 29; Tomato = 38; Wild cosmos = 8) were analyzed. The samples were purchased from two supermarkets (A and B) and two wet markets (C and D). With the MPN-PCR technique, about 12.0% of the samples were positive for the presence of V. parahaemolyticus tdh-positive, with maximum densities of up to 39 MPN/g. The total frequency of V. parahaemolyticus trh-positive in the samples was 10.1%, with maximum concentration 15 MPN/g. V. parahaemolyticus tdh-positive was most prevalent in samples from Wet Market C (20.78%) and also in vegetable type Oenanthe stolonifera (Japanese parsley) with 19.0%, while V. parahaemolyticus trhpositive was predominant in samples from Wet Market D (16.7%) and was most frequent in both Oenanthe stolonifera (Japanese parsley) and Cucumis sativus (Cucumber) with 14.3% prevalence for each type. The results highlighted the fact that raw vegetables could be contaminated with virulent V. parahaemolyticus and could act as a transmission route, thus poses risk to consumers from the consumption of raw vegetables. To the author’s knowledge, this is the first assessment of V. parahaemolyticus carrying tdh and trh genes in raw
    vegetables from retail outlets in Malaysia.

    A total of 49 isolates of V. parahaemolyticus and 8 isolates of V. cholerae isolated from freshwater fish of patin (Pangasius hypopthalmus) and red tilapia (Oreochromis sp.) were purchased from different retail level in Selangor, Malaysia. All of the isolates showed a multiple resistances towards all 15 antibiotics tested. Some of the isolates show a high resistance to different antibiotics including bacitracin, vancomycin, tetracycline, furazididone, cephalothin and erythromycin. However, both species was susceptible towards imipenem. Overall antibiotics resistance patterns of all isolates were resistant from 2 to 14 resistance patterns with multiple antibiotic resistance (MAR) index ranging from 0.13 to 0.93 respectively. As the results obtained in the dendrogram produced from both species had indicates that these antibiotics were intensively used whether in the aquaculture farm through feeds during culture or at the hatchery production of seed. Thus, this study will provides an essential information of the MAR index and also the clustering analysis in order to determine the biosafety of Vibrio spp. in freshwater aquaculture fish sold at different retail level in Malaysia.

    This study was performed to enumerate the total viable cell count of probiotic in five brands (A to E) of commercially cultured milk drinks that are available in the Malaysian market as well as to test their tolerance to various pH and bile concentrations by simulating the human gastrointestinal pH and bile concentration. The acid tolerance test was studied under pH 1.5 and 3.0 with 7.2 as control. The cell count for the acid tolerance test was obtained at an interval of 0, 1.5 and 3 hours respectively and was plated onto duplicate MRS agars to be incubated at 37°C for 48 hours. All cells recovered after 3 hours of pH treatment were selected for bile tolerance test in MRS broth containing bile concentrations of 0% (control), 0.3% and 2.0% and cell counts were recorded after 24 hours of incubation. The probiotic strains in products A, B, C & D met the suggested initial count of 106 CFU/ml with brand C recording the highest at 9.19 ± 0.14 log CFU/ml. Strains in product A, B & C showed good tolerance to pH 3.0 and 7.2 recording a count of >106 CFU/ml after 3 hours with a range of 6.60 – 9.04 log CFU/ml. The higher bile concentrations resulted in lower growth of strains in all the brands. Upon pH 1.5 treatment, only brand C recorded growth in all bile concentrations. After pH 3.0 treatment, all brands except brand E met the requirement of survival at 0.3% bile concentration. Results showed probiotics in product A, B & C met the initial count requirement, and exhibited good acid and bile tolerance therefore being a potentially good source of probiotic.
