    A 25-week feeding trial was conducted to assess the growth performance, organoleptic quality, and to estimate the viability of nourishing hybrid grouper (Epinephelus fuscoguttatus x Epinephelus lanceolatus) with low-cost fish (LCF) and commercially compound feed (CCF). A group of 3600 juvenile fish (182g) were released in four sea cages and fed with either LCF or CCF in duplicate. At the end of the trial, the hybrid grouper provided LCF attained a significantly higher (P0.05). Although technicalities of fish fed with LCF suggest that LCF is more efficient than CCF, feeding LCF to high-value fish is an unsustainable practice as LCF is usually obtained through trawling – a destructive fishing method for the marine ecosystem. Therefore, feeding with CCF without the use of LCF as the source of protein for its fishmeal will contribute to sustainable aquaculture. In order to convince the local farmers in Sabah to adopt the practice of feeding CCF, future research should focus on completing the species-specific diet formulation to promote optimum growth, and find ways to reduce the CCF local selling price.

    The study compares the bacteriological quality on Asian seabass (Lates calcarifer) between ice and salt storage methods. The main objectives of the study were to identify different bacteria constituents and quantitative bacterial load in Asian seabass when preserved with ice and sea salt. For the purpose of this study, Asian seabass was stored in two different conditions of ice-chilled and salted for 2 days. All fish samples were analyzed by performing bacteriological analysis and the isolated bacteria were identified by using API identification system. In case of the quantity of bacteria in the flesh, Chilling and salting had no significant difference to the quantity of bacteria on fish flesh. As for the skin, salt-preserved fish showed higher quantity of bacteria than ice-preserved fish. Acinetobacter baumannii and Pseudomonas fluorescens had been identified from skin sample of ice-chilled fish. Besides P. fluorescens and A. baumannii other isolates identified include Vibrio and Myxobacteria. All bacteria were cocci-shaped except a few bacilli. In term of bacteria number and morphological characteristics, ice-chilled preserved fish was better than salt preserved fish. Overall, less number of bacteria was observed in both ice-chilled and sea salt preserved fish. The result of this study indicated that the quick preservation is a very important factor to control bacterial load in the preserved fish.

    A new sanguinicolid blood fluke, Parasanguinicola vastispina, is described from sea bass Lates calcarifer cultured in Malaysia. It is distinguished by its massive armature and widely spaced genital pores, the female pore being pre-ovarian. P. vastispina inhabits the branchial arteries, dorsal aorta, mesenteric venules and renal artery of its host. No pathological effect was observed in infected fish.

    Microsatellites are the most popular markers for parentage assignment and population genetic studies. To meet the demand for international comparability for genetic studies of Asian seabass, a standard panel of 28 microsatellites has been selected and characterized using the DNA of 24 individuals from Thailand, Malaysia, Indonesia and Australia. The average allele number of these markers was 10.82 +/- 0.71 (range: 6-19), and the expected heterozygosity averaged 0.76 +/- 0.02 (range: 0.63-1.00). All microsatellites showed Mendelian inheritance. In addition, eight standard size controls have been developed by cloning a set of microsatellite alleles into a pGEM-T vector to calibrate allele sizes determined by different laboratories, and are available upon request. Seven multiplex PCRs, each amplifying 3-5 markers, were optimized to accurately and rapidly genotype microsatellites. Parentage assignment using 10 microsatellites in two crosses (10 x 10 and 20 x 20) demonstrated a high power of these markers for revealing parent-sibling connections. This standard set of microsatellites will standardize genetic diversity studies of Asian seabass, and the multiplex PCR sets will facilitate parentage assignment.

    Feed efficiency (FE) is the amount of body weight gain for a given feed intake. Improving FE through selective breeding is key for sustainable finfish aquaculture but its evaluation at individual level is technically challenging. We therefore investigated whether individual routine metabolic rate (RMR) was a predictor of individual FE in the European sea bass Dicentrarchus labrax, a major species in European mariculture. The European sea bass has three genetically distinct populations across its geographical range, namely Atlantic (AT), West Mediterranean (WM), and East Mediterranean (EM). We compared FE and RMR of fish from these three populations at 18 or 24 °C. We held 200 fish (62 AT, 66 WM, and 72 EM) in individual aquaria and fed them from ad libitum down to fasting. FI was assessed for an ad libitum feeding rate and for a fixed restricted ration (1% of metabolic body weight·day-1, with metabolic body weight = body weight0.8). After being refed 12 wk in a common tank, individual RMR was measured over 36 h by intermittent flow respirometry. There was a significant effect of temperature whereby fish at 18 °C had greater mean FE (P < 0.05) and lower RMR (P < 0.001). There was also a significant effect of population, where AT fish had lower FE (P < 0.05) and greater RMR (P < 0.001) than WM and EM, at both temperatures. Despite these differences in temperature and population means, individual FE and RMR were not significantly correlated (P > 0.05). Therefore, although the results provide evidence of an association between metabolic rate and FE, RMR was not a predictor of individual FE, for reasons that require further investigation.

    The Asian seabass is euryhaline, therefore it is interesting to describe the infestation and survival of caligids at varying salinity on the host. In this study, two different brackish water culture systems with monoculture and polyculture practices were investigated for the occurrence of Caligus spp. on Lates calcarifer. Polyculture practices mainly consisted of snapper (Lutjanus spp.), grouper (Epinephelus spp.) and seabass (L. calcarifer), while the monoculture was stocked with only seabass. A total of 777 Caligus spp. specimens were isolated from the sampling in 2009, consisting of three species; Caligus chiastos, Caligus epidemicus and Caligus rotundigenitalis. In 2011, the total specimen was increased to 3110 and two additional species were found; Caligus punctatus and one unknown species (Caligus sp.). A 98.6% of the total examination was represented by C. epidemicus. Constant presence of C. epidemicus was observed throughout the study, regardless the differences in between culturing practices and systems. This species was able to survive within wide salinity range, from 5 to 28 ppt. The other isolated species (C. chiastos, C. punctatus, C. rotundigenitalis and Caligus sp.) were only found infesting in polyculture cages with the salinity ranging from 25 to 28 ppt. Despite accounts for less than 2% of the total specimens, these species may able to produce a challenge for L. calcarifer polyculture farming activity due to their capability for host switching. The present study revealed the potential risk for cross-species transmission in polyculture practices.

    In this study, we aimed to isolate, identify and characterize lactic acid bacteria (LAB) from the intestine of juvenile seabass (Lates calcarifer) as a new potential probiotic. Four strains of LABs were isolated from the intestines of ten healthy seabass juveniles. In the in vitro screening process using spot lawn assay, one isolate labeled as LAB3 showed inhibitory activity against Vibrio harveyi (ATCC 35,084). Strain LAB3 was determined to belong to the gram positive bacteria group with cocci shape and was identified as Enterococcus hirae using 16S rDNA analysis. This bacterium was able to grow at pH ranging from pH 2 to 10 with the best growth at pH 7. This strain was also able to grow at 0-4% NaCl after 24 h incubation and grew best at 1.5% NaCl. Enterococcus hirae strain LAB3 of the present study is worthy to be further characterized as a potential probiotic for use in seabass culture.

    Background: The Xisha Islands are composed of the Yongle Islands and the Xuande Islands in Hainan Province, China. It has one of the highest species diversity in the world and is also a typical oceanic distribution area of coral reefs globally. The ichthyofauna of the Xisha Islands were recorded by underwater visual census in May 2019 and July 2020. The survey data were combined with previous records of species into the checklist of the Xisha Islands presented herein. A total of 691 species, belonging to 24 orders and 97 families, was recorded. The major families were Labridae, Pomacentridae, Serranidae, Chaetodontidae, Hexanchidae, Lutjanidae, Scaridae, Gobiidae, Scorpaenidae and Carangidae. In this study, the Coral Fish iversity Index (CFDI) of six families (Chaetodontidae, Pomacanthidae, Pomacentridae, Labridae, Scaridae and Acanthuridae) was 229, indicating 756 coral fishes. In terms of the IUCN Red List, one species is Critically Endangered (Glyphis gangeticus), six species are Endangered (Stegostoma fasciatum, Aetomylaeus maculatus, Aetomylaeus vespertilio, Epinephelus akaara, Cheilinusundulatus sp. and Xiphias gladius), 16 species are Vulnerable, and 13 species are Near Threatened in the Xisha Archipelago, so conservation should be strengthened in this area in the future.

    New information: One species is a new record for China (Dischistodus pseudochrysopoecilus) and 23 species are newly found in the Xisha Islands.

    Fish epidermal mucus is an important reservoir of antipathogenic compounds which serves as the first line of the immune defence. Despite its significant role in the physiology and health of fish, detailed profiling of fish epidermal mucus has yet to be explored. Therefore, this study investigates a label-free colloidal surface-enhanced Raman spectroscopic (SERS) method for profiling grouper mucus. Gold nanoparticles were first synthesised using the standard citrate reduction and characterised using ultraviolet-visible spectroscopy, transmission electron microscopy and dynamic light scattering. The influence of acidified sodium sulphate (Na2SO4) at pH 3 as the aggregating agent on the enhancement of the SERS spectrum of different analyte samples including rhodamine 6G (R6G) dye, lysozyme solution and hybrid grouper (Epinephelus fuscoguttatus × Epinephelus lanceolatus) mucus was observed. Based on the results, an optimal Na2SO4 concentration of 1 M was recorded to achieve the highest enhancement of the SERS signal for R6G and grouper mucus, while the optimal concentration for lysozyme was 0.1 M. The results indicated a higher degree of aggregation induced by lysozyme than R6G and grouper mucus. A few overlapping peaks of the SERS spectra of lysozyme and grouper mucus made it possible to confirm the presence of lysozyme as potential biomarkers.

    Metallic contamination in seafood, especially fish, has been of increasing concern to human health. Moreover, with increasing dependency on farmed fish for fish resources, the metallic contamination in them is still questionable. This study aimed to investigate the effects of cooking (steaming) on heavy metal concentration in farmed fish and to estimate its potential human health risk. Farmed sea basses (Lates calcarifer) from Setiu Lagoon were used to study the difference in metal uptake through human consumption of raw and cooked (steamed) fish samples. Selected heavy metals, namely copper (Cu), zinc (Zn), arsenic (As), cadmium (Cd), mercury (Hg) and lead (Pb), were measured using ICP-MS following Teflon bomb closed digestion of the fish samples. Cooking of the fish muscle by steaming was applied to investigate if cooking changes the concentration of heavy metals. Mercury and As were found accumulated more notably in the fish muscle, though only Hg was found to show significant (p > 0.05) increase when L. calcarifer is cooked. The amount of As in the fish muscle throughout its growth can be potentially harmful to humans, with the highest averaged concentration at 3.29 ± 0.65 mg/kg dw. above the standard set by the Malaysian Food Regulation (1985) of >1 mg/kg. All the other heavy metals were at relatively safe concentrations well below the standard set by both national and international guidelines. The PTWI per individual of L. calcarifer for As was at 0.84 mg/kg bw., which indicated that the amount of this fish safe for consumption without any adverse effect is 170 g/week. Therefore, long-term intake of these fish may pose a risk to human health due to the relatively higher Hg and As concentration found in these fishes.

    In general, demand feeding devices are equipped with a mechanical trigger switch. Such a switch is not suitable for
    juvenile fish with a small body size, because the body weight is insufficient to trigger the feeder. An infrared light
    sensor that does not require the fish to push a feeder switch is more suitable for small fish. The brown-marbled grouper
    Epinephelus fuscoguttatus is an important fish species in Southeast Asia. The purpose of this study was to compare the
    growth rates (GRs) of brown-marbled grouper juveniles reared using customised demand feeding devices with an infrared
    light sensor (the infrared light demand feeder (IRDF) group) and automatic feeding devices (the automatic feeder (AF)
    group). The results indicated that GRs of standard lengths and body weights showed no significant differences using
    one-way analysis of variance; however, the standard length of the IRDF group showed a tendency of a higher GR than
    the AF group. Although the feed conversion ratio (FCR) also showed no significant difference, the FCR of the IRDF group
    was more efficient, indicating that the IRDF group yielded a more desirable FCR. These results indicate that IRDF can be
    used in the culture of brown-marbled grouper juveniles. In view of the working schedule of the fish farm staff, IRDF are
    superior to other feeding devices, because they are less labour-intensive than usual tasks. We conclude that IRDF is a
    useful feeding system for aquaculture.

    Previous studies have found positive correlations between mangrove forest extent and fisheries yield but none of these univariate relationships provide a reliable estimate of yield from mangrove area. This study tests the hypothesis that the nursery ground value or natural production of fish and shrimps is related to the hydrogeomorphology settings of mangrove forests by using multivariate redundancy analysis (RDA). The hydrogeomorphological metrics of five mangrove forests imaged by satellite were measured using Geographical Information System (GIS). The RDA indicated that the metrics, including mangrove area, multiple waterways and creeks, mangrove-river interface, waterway surface area and sediment organic matter, influenced the diversity and abundance of fish and shrimps. Larger values of these metrics increase the abundance of economically important fish species of the families Lutjanidae, Haemulidae, Serranidae and economically-important penaeid shrimps. Sediment organic matter also significantly correlates with the distribution and abundance of fish that feed off the bottom such as the Leiognathidae, Clupeidae and Mullidae. Mangrove forests with combinations of large mangrove area, river surface area, high stream ordering and longest mangrove-river interface will provide greater role as nursery grounds for fish and shrimps.

    Some of Vibrio species is well known as pathogenic bacteria in aquaculture and the marine industry. Its infection is able to generate a massive outbreak and affect the fish population, especially for net caged fish such as seabass. This study was conducted to investigate the prevalence of Vibrio spp. isolated from seabass (Lates calcarifer) in Sri Tujuh Lagoon, Tumpat, Kelantan. Then, to determine the antibiotic resistance in Vibrio isolates. Polymerase chain reaction (PCR) was used to detect Vibrio species using specific primer VR169 and VR744 with estimation base pair size band, 597 bp and further identified by sequencing. On the other hand, antibiotic susceptibility tests were continued by using 13 types of antibiotics; kanamycin (K30), chloramphenicol (C30), neomycin (N10), ampicillin (AMP10), nitrofurantoin (F300), tetracycline (TE30), streptomycin (S10), norfloxacin (NOR10), ciprofloxacin (CIP5), nalidixic acid (NA30), gentamicin (CN10), doxycycline (DO30) and sulfamethoxazole (SXT100). As a result, 14 Vibrio isolates were identified, including Vibrio fluvialis (n=6), Vibrio parahaemolyticus (n=3), Vibrio harveyi (n=2) and each isolate for Vibrio vulnificus, Vibrio alginolyticus and Vibrio spp. The results showed that all isolates were sensitive to most antibiotics except ampicillin, neomycin and streptomycin. The MAR index value was ranging from 0 to 0.31. This study demonstrates the prevalence of Vibrio spp. in seabass and the report on multidrug resistance strains that could be of concern to the fish farmers. In addition, data from this study can be further used in fish disease management plans.

    A new cell line, Asian sea bass brain (ASBB), was derived from the brain tissue of Asian sea bass Lates calcarifer. This cell line was maintained in Leibovitz L-15 media supplemented with 10% fetal bovine serum (FBS). The ASBB cell line was subcultured more than 60 times over a period of 15 mo. The ASBB cell line consists predominantly of fibroblastic-like cells and was able to grow at temperatures between 20°C and 30°C with an optimum temperature of 25°C. The growth rate of these cells increased as the proportion of FBS increased from 2% to 20% at 25°C with optimum growth at the concentrations of 10% or 15% FBS. Polymerase chain reaction products were obtained from ASBB cells and tissues of sea bass with primer sets of microsatellite markers of sea bass. An isolate of piscine nodavirus from juveniles of marine fish species tested positive by IQ2000 kit for viral nervous necrosis detection and was examined for its infectivity to a fish cell line of ASBB. A marine fish betanodavirus was tested to determine the susceptibility of this new cell line in comparison with commercial highly permissive SSN-1 cells. The ASBB cell line was found to be susceptible to nodavirus (RGNNV genotype), and the infection was confirmed by comparison cytopathic effect (CPE) with commercial SSN-1 and reverse transcriptase-polymerase chain reaction. A nodavirus was further elucidated by electron microscopy, and the virus tested was shown to induce CPE on ASBB cells with significant high titer. This suggests that the ASBB cell line has good potential for the isolation of fish viruses.

    Red tide of dinoflagellate was observed in brackish water fish ponds of Terengganu along the coast of the South China Sea during the study period between January 1992 to December 1992. The nearby coastal moat water facing the South China Sea is the source of water for fish pond culture activities of sea bass during the study period. An examination of water quality in fish ponds during the study period indicated that both the organic nutrients were high during the pre-wet monsoon period. The source of the nutrients in coastal water was believed to be derived from the agro-based industrial effluents, fertilizers from paddy fields and untreated animal wastes. This coincided with the peak production of dinoflagellate in the water column in October 1992. The cell count ranges from 8.3 to 60.4×10.4×10(4)/l during the bloom peak period and the bloom species were compared entirely of non-toxic dinoflagellates with Protoperidinium quinquecorne occurring >90% of the total cell count. However, both cultured and indigenous fish species were seen to suffer from oxygen asphyxiation (suffocation due to lack of oxygen). The bloom lasted for a short period (4-5 days) with a massive cell collapse from subsurface to bottom water on the sixth day. The productivity values ranged from 5-25 C g/ l / h with a subsurface maximum value in October 1992. Two species of Ciliophora, Tintinnopsis and Favella, were observed to graze on these dinoflagellates at the end of the bloom period.

    This study was conducted to investigate the effects of dietary inclusion of Spirulina platensis on growth performance and hematological parameters of juvenile Asian sea bass (Lates calcarifer) reared in a freshwater culture system. Five experimental diets were prepared by replacing fish meal protein with the microalga at replacement levels of 5% (SP5), 10% (SP10), 20% (SP20) and 30% (SP30), and the substitution effect was compared with a control diet (Con) in which fish meal was the sole protein. Fish were stocked in net cages placed in a 150-ton tank with a stocking density of 20 fish per cage. After 8 weeks of feeding trial, the fish did not show any significant differences in growth performance but numerically higher weight gain and specific growth rate were achieved in the fish stock fed diet SP10 compared to other treatments. Feed conversion ratio and survival rate of fish were significantly affected by the inclusion of Spirulina in the diets. Except for crude ash content, whole-body proximate composition of the experimental fish was also significantly influenced by the diets. Regarding the effects of diets on blood parameters, only hematocrit, hemaglobin, HDL-c and AST were significantly affected by the inclusion of Spirulina in the diets. This study demonstrated that Spirulina could replace up to 10% of FM protein in practical diets of juvenile Asian sea bass without negative effects on growth performance. However, replacement of fish meal with Spirulina meal at 5% might be considered for commercial use considering a significant decrease in survival above this value.

    The effects of dietary protein level on the growth performance and ammonia excretion of the leopard coral grouper,
    Plectropomus leopardus were investigated for eight weeks. Fish were fed diets with 40, 45, 50, 55 and 60% crude protein
    levels in separate recirculating systems. Fish fed with the 50% crude protein containing diet showed the best ingestion
    rate, which was significantly higher than that found in the other groups. As the dietary protein level increased, the
    specific growth rate increased significantly and it reached the highest level at 50% crude protein containing diet. Based
    on the results of all measured parameters 50% protein containing diet was the best among all test diets. The regression
    equation for dietary protein level versus ammonia excretion indicated that the optimal dietary protein level with the
    least ammonia excretion was 53.14%. More research is still needed to elucidate the effects of 53.14% crude protein
    containing diet on the specific growth rate, feed conversion ratio, protein efficiency ratio and ingestion rate of leopard
    coral grouper before recommending this level. Until then, 50% protein containing diet can be recommended for leopard
    coral grouper culture in the recirculation system.

    Skin abrasions often occur in farmed fish following handling by labourers, injury by farm facilities, cannibalism and ectoparasites. Vibrio spp. are opportunistic pathogens that can invade host fish through damaged tissues and cause outbreaks of vibriosis. This study describes the effect of skin abrasions on the infectivity of V. harveyi using Asian seabass Lates calcarifer (Bloch, 1790) fingerlings as a case example and compares bacterial load and fish survival following immersion challenge with different doses. In total, 315 fish (6.67 ± 1.8 g) were divided into 3 treatments: skin abrasion followed by immersion infection, immersion infection only and an uninfected, uninjured control. Fish in the infection treatments were divided into 3 subgroups and exposed in triplicate to a 7 d immersion challenge with 106, 107 and 108 CFU ml-1 of live V. harveyi. No mortalities were observed in the control and immersion infection groups. However, fish in the skin abrasion treatment group that were infected with 108 CFU ml-1 of live V. harveyi showed signs of progressing disease throughout the experiment, which resulted in mortalities. Significantly higher bacterial loads (p < 0.05) were recorded in the intestine, liver and gills of the fish in this group. Fish in the skin abrasion treatment that were exposed to 107 and 108 CFU ml-1 of V. harveyi showed 100% mortality by Days 5 and 4, respectively. These findings confirm that skin injuries increase the susceptibility of seabass fingerlings to V. harveyi infection.
