Synthesis of layered double hydroxides (LDHs) of Zn/Al-NO3- hydrotalcite (HIZAN) and Zn/Al-diocytyl sodium sulfosuccinate (DSS) nanocomposite (NAZAD) with a molar ratio of Zn/Al of 4:1 were carried out by coprecipitation through continuous agitation. Their structures were determined using X-ray diffractometer spectra, which showed that basal spacing for LDH synthesized by both methods was about 8.89 A. An expansion of layered structure of about 27.9 A was observed to accommodate the surfactant anion between the interlayer. This phenomenon showed that the intercalation process took place between the LDH interlayer. Lipase from Candida rugosa was immobilized onto these materials by physical adsorption method. It was found that the protein loading onto NAZAD is higher than HIZAN. The activity of immobilized lipase was investigated through esterification of oleic acid and 1-butanol in hexane. The effects of pore size, surface area, reaction temperature, thermostability of the immobilized lipases, storage stability in organic solvent, and leaching studies were investigated. Stability was found to be the highest in the nanocomposite NAZAD.
Lipase from Candida rugosa was immobilized by entrapment on poly(N-vinyl- 2-pyrrolidone-co-2-hydroxyethyl methacrylate) (poly[VP-co-HEMA]) hydrogel, and divinylbenzene was the crosslinking agent. The immobilized enzymes were used in the esterification reaction of oleic acid and butanol in hexane. The activities of the immobilized enzymes and the leaching ability of the enzyme from the support with respect to the different compositions of the hydrogels were investigated. The thermal, solvent, and storage stability of the immobilized lipases was also determined. Increasing the percentage of composition of VP from 0 to 90, which corresponds to the increase in the hydrophilicity of the hydrogels, increased the activity of the immobilized enzyme. Lipase immobilized on VP(%):HEMA(%) 90:10 exhibited the highest activity. Lipase immobilized on VP(%):HEMA(%) 50:50 showed the highest thermal, solvent, storage, and operational stability compared to lipase immobilized on other compositions of hydrogels as well as the native lipase.
The feasibility of using palm oil fractions as cheap and abundant sources of raw material for the synthesis of amino acid surfactants was investigated. Of a number of enzymes screened, the best results were obtained with the immobilized enzyme, Lipozyme. The effects of temperature, solvent, incubation period, fatty substrate/amino acid molar ratio, enzyme amount, and water removal on the reactions were analyzed and compared to those on reactions with free fatty acids and pure triglycerides as fatty substrates. All reactions were most efficient when carried out at high temperatures (70-80 degrees C) in hexane as a solvent. However, while reactions with free fatty acids proceeded better when a slight excess of the free fatty acids over the amino acids was used, reactions with triglycerides and palm oil fractions were best performed at equimolar ratios. Also, the addition of molecular sieves slightly enhanced reactions with free fatty acids but adversely affected reactions with triglycerides and palm oil fractions. Although reactions with palm oil fractions took longer (6 d) to reach equilibrium compared to reactions with free fatty acids (4 d) and pure triglycerides (4 d), better yields were obtained. Such lipase-catalyzed transacylation of palm oil fractions with amino acids is potentially useful in the production of mixed medium- to long-chain surfactants for specific applications.
The study explored on the commonly available weed plant Commelina nudiflora which has potential in-vitro antioxidant and antimicrobial activity. The different polar solvents such as ethanol, chloroform, dichloromethane, hexane and aqueous were used for the soxhlet extraction. The extracts were identified pharmacologically as important bioactive compounds and their potential free radical scavenging activities, and antimicrobial properties were studied. C. nudiflora extracts were monitored on their in-vitro antioxidant ability by DPPH and ABTS radical scavenging assay. Aqueous extract shows significant free radical scavenging activity of 63.4 mg/GAE and 49.10 mg/g in DPPH and ABTS respectively. Furthermore, the aqueous crude extract was used in antibacterial studies, which shows the highest inhibitory activity against Pseudomonas aeruginosa, Escherichia coli and Salmonella typhi. Among all the extracts, aqueous extract of C. nudiflora has significant control over free radical scavenging activity and inhibition of the growth of food pathogenic bacteria. Also, the aqueous extract contains abundance of phenolics and flavonoids higher than other extracts. This study explored weed plant C. nudiflora as a potential source of antioxidant and antibacterial efficacy and identified various therapeutic value bioactive compounds from GC-MS analysis.
Introduction: This study examined the antioxidant activity and phenolic, flavonoid and saponin contents from mul-tiple solvent extracts of Albizia myriophylla (ABZ) bark. Antioxidant activity of the methanol extract and its derived fractions namely hexane, chloroform, ethyl acetate, butanol and a residual aqueous fraction of the bark of ABZ was assessed. Methods: All the extracts showed a significant presence of phenolic, flavonoids and saponins. In DPPH (1, 1-diphenyl-2-picrylhidrazyl) radical scavenging test, ABTS (2-2’-azinobis 3-ethyl-6-sulfonic acid) radical scavenging test and reducing activity on ferrous iron (FRAP) test, the total antioxidant capacity was found to be varied in different fractions. Results: The IC50 calculated value of the three assays showed that the methanolic extract of ABZ bark had the lowest IC50 value for each assay, compared to the other extracts signifying highest anti-oxidant activity. Methan-olic extract of ABZ was tested in high-fat diet induced mice, which showed reduce adipocyte cellularity and reduces the cholesterol, triglyceride, LDL level while increasing the HDL level. Conclusion: The antioxidant capacity with probable free radical scavenging activity of the methanolic extract of ABZ may be useful for the treatment of chronic inflammatory-related metabolic diseases such as obesity.
Introduction:Spilanthes acmella, also known as “subang nenek’, has been used traditionally in Malaysia to treat toothache. A previous study has shown Spilanthes acmella leaves extracts (SALE) inhibit Streptococcus mutans growth. Streptococcus mutans is commonly found in the human oral cavity and is the main contributor to tooth de-cay. There is no study on the antibacterial effects of Spilanthes acmella flower extracts (SAFE) against Streptococcus mutans reported to date. Therefore, the objective of this study is to investigate antibacterial properties of SAFE against S. mutans. Methods:S. mutans was subcultured in Muller Hinton (MH) broth and agar. Sequential extractions of S. acmella flowers were conducted using four different solvents with increasing polarity, [n- hexane, dichloromethane (DCM), acetone, methanol (MeoH)] and tested with different concentrations against S. mutans via the disc diffusion assay, minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC). Sodium fluoride (NaF) was used as a positive control while DMSO was used as a negative control. Results: The disc diffusion assay shows SAFE inhibited Streptococcus mutans growth. SAFE-DCM shows the greatest inhibition properties (12.33±2.30 mm) followed by SAFE-n-hexane (11.33±0.57 mm). Meanwhile, SAFE-Meoh and SAFE-acetone show no inhibition zone (6.00±0.001 mm). MIC value for SAFE-DCM and SAFE-n-hexane is 12.5 mg/mL respectively. Whereas, MBC value SAFE-DCM and SAFE-n-hexane is 50.0 mg/mL respectively. Conclusion: It can be concluded SAFE-DCM and SAFE-n-hexane possesses bactericidal properties against Streptococcus mutans.
Processing of soybeans to other products and consumption of soy products is increasing worldwide mainly due to acclaimed health benefits. Processing can alter soybean sensory appeal, nutritive value and potentially affect consumer health. Rhizopus oligosporus was used to ferment soybean for 3 days. The tempeh flour (TF) was produced form tempeh while defatted tempeh flour (DTF) was then produced from TF by immersing in hexane solvent while soy protein isolate (SPI) was prepared from DTF by using alkali and acid followed by neutralization treatment. In this study, nutritional properties and amino acid content of tempeh, TF, DTF and SPI were determined. Therefore, the objective of this study is was to evaluate the effect of each treatment on the chemical composition and amino acid content for all the samples. The results showed that the nutritional properties (total ash, moisture, crude fat, total carbohydrate and crude fibre) were reduced significantly (p < 0.05) except for protein content. Protein content was significantly (p < 0.05) increased by 50.5% in SPI. For amino acid content, the results obtained showed that SPI contain highest amount of essential and non-essential amino acid followed by DTF, Tempeh and TF. Glutamic acid was found to be the highest amino acid component in all samples. The evaluation from the results showed that SPI can be considered as potential functional food ingredients.
Mushrooms have been highly regarded as possessing enormous nutritive and medicinal values. In the present study, we evaluated the anti-oxidative and anti-atherosclerotic potential of shiitake mushroom (Lentinula edodes) using its solvent-solvent partitioned fractions that consisted of methanol:dichloromethane (M:DCM), hexane (HEX), dichloromethane (DCM), ethyl acetate (EA) and aqueous residue (AQ). The hexane fraction (1 mg/mL) mostly scavenged (67.38%, IC50 0.55 mg/mL) the 2,2-diphenyl-1-picryl hydrazyl (DPPH) free radical, contained the highest reducing capacity (60.16 mg gallic acid equivalents/g fraction), and most potently inhibited lipid peroxidation (67.07%), low density lipo-protein oxidation and the activity of 3-hydroxy 3-methyl glutaryl co-enzyme A reductase (HMGR). GC-MS analyses of the hexane fraction identified α-tocopherol (vitamin E), oleic acid, linoleic acid, ergosterol and butyric acid as the bio-functional components present in L. edodes. Our findings suggest that L. edodes possesses anti-atherosclerotic bio-functionality that can be applied as functional food-based therapeutics against cardiovascular diseases.
Oil palm or Elaeis guineens is a rich natural source of phenolic with flavonoid as the main constituents. These phenolics are potent antioxidants that can be used in the food industry, cosmetics and others. Therefore, the study was aimed to determine the effect of solvents which were methanol, ethyl acetate and hexane also different plant parts which were leaves, frond and fresh fruit bunch toward antioxidant activity (AOA), total phenolic content (TPC) and total flavonoid
content (TFC). The antioxidant was analysed using the DPPH method, TPC by Ciocalteu assay and TFC by aluminium chloride colorimetric assay. The result from ANOVA indicated that there was a difference (P < 0.05) in the extracting ability of each solvent and different plant parts for AOA, TPC and TFC. Generally, the result suggested that methanol give the highest antioxidant
activity, TPC and TFC compared to ethyl acetate and hexane. Therefore, the solvent used should be selected properly to allow for a high level of extraction efficiency.
We have determined the protective effects of Thymus serpyllum (TS) extract and nanoparticle-loaded TS on hydrogen peroxide-induced cell death of mesenchymal stromal cells (MSCs) in vitro. Gas chromatography-mass spectroscopy confirmed the spectrum of active components in the extract. Out of the three different extracts, the hexane extract showed significant free radical scavenging activity. Treatment of MSCs with H2O2 (hydrogen peroxide) significantly increased intracellular cell death; however, pretreatment with TS extract and nanoparticle-loaded TS (200 μg/ml) suppressed H2O2-induced elevation of Cyt-c and MMP13 and increased the survival rates of MSCs. H2O2-induced (0.1 mM) changes in cytokines were attenuated in the extract and nanoparticles by pretreatment and cotreatment at two time points (p < 0.05). H2O2 increased cell apoptosis. In contrast, treatment with nanoparticle-loaded TS suppressed the percentage of apoptosis considerably (p < 0.05). Therefore, TS may be considered as a potential candidate for enhancing the effectiveness of MSC transplantation in cell therapy.
Strobilanthes crispus and Clinacanthus nutans are popular herbal plants in the Southeast
Asian region. The present work was aimed at determining the antioxidant activities and the
associated components in the leaf extracts of both species using polar and non-polar solvents
namely water, methanol, ethyl acetate, and hexane. The total phenolic content (TPC) and total
flavonoid content (TFC) were higher in the leaf extracts of S. crispus as compared to C.
nutans. Among the solvents, methanol was the best solvent in extracting the antioxidant
components for S. crispus (TPC: 159.85 ± 0.89 mg GAE/g extract and TFC: 955.47 ± 2.66 mg
RE/g extract). However, for C. nutans, its methanolic extract yielded the highest TPC (36.39
± 0.17 mg GAE/g extract), whereas ethyl acetate yielded the highest TFC (229.61 ± 7.81 mg
RE/g extract). The high levels of both TPC and TFC contributed to the antioxidant activities
of S. crispus extract as reflected in the methanolic extract attaining the highest level of
antioxidant activities, measured by ferric reducing antioxidant power (FRAP) (6.84 ± 1.12
mmol Fe2+/g extract), DPPH radical scavenging (IC50: 203.60 ± 7.28 μg/mL), and Trolox
equivalent antioxidant capacity (TEAC) (1.01 ± 0.01 mmol TE/g extract) assays. This
contrasted with C. nutans which showed lower antioxidant activities owing to its lower TPC
and TFC. Correlation analysis revealed significant correlations (p < 0.05, r = 0.915 - 0.985)
between both TPC and TFC in S. crispus and antioxidant activities. However, only TPC of C.
nutans showed a significant correlation with FRAP values (r = 0.934). Further tentative
identification of the constituents in the extracts using HPLC-ESI-QToF-MS/MS revealed the
existence of 20 polyphenolic compounds in both S. crispus and C. nutans, which were likely
responsible for their antioxidant activities. In addition, 15 polyphenolic compounds classified
as chalcones, isoflavanoids, flavones, and flavonols have not been previously reported in both
species. The methanolic extracts of both species yielded a higher content of antioxidants, with
S. crispus offering a richer source of dietary antioxidants as compared to C. nutans. However,
further study is needed to identify their bioactivities in relation to their bioactive components.
A study was conducted on the chemical and biological properties of three different species of Mangifera i.e. Mangifera pajang, M. indica L., and M. kemanga leaves obtained from Pitas, Sabah. The aims of this study were to determine the presence of secondary metabolites as well as the antioxidative activities especially the catalase (CAT) and guiacol peroxidase specific activities (gPOD) in the leaves part of these three species. The extraction of these samples was carried out using three different polarities of solvents: hexane, ethyl acetate, and methanol. The total percentage of the crude extract of is 7.30% for M. pajang, 12.87% for M. indica and 7.98% for M. kemanga. Phytochemical screening was performed with various tests for each of the crude extracts. The results showed that these three species gave positive results for alkaloids, saponins, flavonoids, phenols, carbohydrates, phytosterols, and tannins metabolites. Based on the tests, CAT specific activities were significantly higher in the leaves of M. pajang with 4.35 ± 1.18 units/mg protein compared to M. indica L. and M. kemanga. The guaiacol peroxidase (gPOD) specific activities showed that M. indica L. has the highest activity with the value of 0.0047 ± 0.0004 units/mg protein.
Introduction: Hygrocybe conica (HC), a wild mushroom commonly consumed by the indigenous people (Orang Asli) in Peninsular Malaysia, was assessed for its antioxidant content. Methods: The HC mushroom was extracted using distilled water and the crude extract partitioned using different solvents and open column chromatography to evaluate its potential antioxidant properties. The mushroom extract was partitioned using liquid-liquid extraction into the hexane (Fl), chloroform (F2), butanol (F3) and formic acid (F4) fractions. Based on solvent polarity, the water extract of the mushroom was fractionated into non-polar (FI), semi-polar (Fii), and polar fractions (Fiii) using open column chromato graphy. Antioxidant capacities were determined using DPPH, ABTS, and ferric reducing antioxidant power (FRAP) assays while Folin-Ciocalteu reagent assay was used to determine total phenolic content (TPC). Results: The HC extract had the highest TPC and DPPH scavenging capacity compared to its extract fractions. TE values (ABTS assay) of F2 and F4 were not significantly higher than the HC extract. Among the extract fractions of different polarities, Fiii had the highest antioxidant capacities (DPPH and FRAP) compared to FI and Fii while FRAP values of these fractions were not significantly lower than the FRAP value of HC extract. The HC extract had significantly lower antioxidant capacity than antioxidant standards (ascorbic acid and BHA). Tannie acid as the main bioactive component in HC mushroom was detected using HPLC method. The presence of phenolics in HC extract was also confirmed using TLC. Conclusion: Due to the presence of potent phenolic components, the mycelia of HC could be consumed for potential antioxidative benefits.
This study examined the tunneling activity and the behaviour of two subterranean termites, Globitermes sulphurues and Coptotermes gestroi treated with four plant extracts, namely Pyllanthus niruri, Azadirachta indica, Leucaena leucocephala and Andrographis paniculata. All plants were extracted with three different solvents (methanol, hexane and water) and were diluted into three concentrations (500, 5,000 and 10,000 ppm). A group of 50 workers and 2 soldiers were tested and observed daily during the exposure in order to determine their survivorship and behaviour. Both sections were scored to determine their tunneling activities after seven days. There was a significant difference of tunneling activities of C. gestroi on sand treated with plant extracts (X2(2) = 31.790, p < 0.0001) with a mean rank of 8.50 for methanolic extracts and 32.50 for both hexane and water extracts. Meanwhile, no significant difference was observed on tunneling activity of G. sulphureus on treated sand (X2(2) = 2.200, p = 0.333) with a mean rank of 20.72 for methanolic extracts, 26.31 for water extracts and 26.47 for hexane extracts. Results showed that plants extracted with methanol demonstrated strong repellent properties with 0 tunneling activity on the treated sand and low survivorship of both termites. Moreover, both termites did not survive (0%) when they were treated with methanolic extracts at 10,000 ppm. They also displayed a different behaviour post-exposure such as avoidance, gradually losing the ability to walk and low feeding consumption. These results indicate that there is a strong termiticidal activity of plants extracted with methanol against C. gestroi and G. sulphureus.
The adulticidal activity of methanol extracts from three Malaysian plants namely Acorus calamus Linn., Litsea elliptica Blume and Piper aduncum Linn. against adult of Aedes aegypti (L.) were studied. Standard WHO bioassay tests were used to evaluate the effectiveness of these plant extracts. The hexane fraction from methanol extract of Acorus calamus rhizome was the most effective, exhibiting LC50 and LC90 values of 0.04 mgcm(-2) and 0.09 mgcm(-2) respectively. For L. elliptica, the methanol fraction also displayed good adulticidal property with the LC50 and LC90 values of 0.11 mgcm(-2) and 6.08 mgcm(-2) respectively. It is found that hexane fraction of the P. aduncum crude extract was the least effective among the three plants showing LC50 and LC90 values of 0.20 mgcm(-2) and 5.32 mgcm(-2), respectively. However, although A. calamus showed lowest LC values, the LT50 results indicated that the methanol fraction of L. elliptica was most potent extract among the extracts tested.
Palm pressed fiber (PPF) is a by-product from oil extraction of oil palm fruits. It has unique characteristics resulting from the combination of palm mesocarp fiber, kernel shell and crushed kernel. The present study on different extraction methods for PPF indicated that conventional hexane cold extraction would be the more preferable method compared to soxhlet and reflux method due to the yield recovered (4.35%) and diacylglycerol (19.93% ± 0.07) obtained. Acylglycerol composition using cold extraction gave high Diaclyglycerol and Triacylglycerol (67.04% ± 0.05) with reasonable amount of Monoaclyglycerol / Free Fatty Acid (13.02% ± 0.02). Lauric acid which was not present in crude palm oil were found to be significant in PPF (5.89 to 9.09%), thus making this oil suitable for application in the food industry in products such as shortening and margarine.
Food-borne diseases have always been a major concern to the consumers, food safety authorities and food industries. Hylocereus spp or Pitaya peels are normally treated as wastes and will be discarded during processing. In this study, the antibacterial activity of ethanol, chloroform and hexane extracts from Hylocereus polyrhizus (red flesh pitaya) and Hylocereus undatus (white flesh pitaya) peels against nine pathogens was evaluated using disc diffusion method and broth micro-dilution method. Result from disc diffusion method showed that chloroform extracts from H. polyrhizus and H. undatus peel were found to exhibit good antibacterial activity where almost all the pathogens studied were successfully inhibited. Result of the minimum inhibitory concentration (MIC) showed that all extracts inhibit the growth of bacteria in the range of 1.25-10.00 mg/mL for all bacteria while their minimum bacteriacidal concentrations (MBC) indicated double of the MICs concentration except for B. cereus, L. monocytogenes and C. jejuni. Even though there is no clear trend indicating which bacteria were sensitive most to the extract, it can be concluded that chloroform extract of both H. polyrhizusand H. undatus peel showed the most potent antibacterial activity. Thus, these findings could be used further to understand the antibacterial property of the peel of pitaya fruits.
Lansium domesticum Corr. is a fruit tree of the Meliaceae family, which is commonly found in SouthEast Asia with a wide range of varieties. This study investigated three varieties of L. domesticum; Duku, Langsat and Dokong for the phytochemical screening and antimicrobial activity. Seeds from the matured fruits were extracted using hexane, methanol and water. The crude extracts were screened for antimicrobial activities toward three bacteria, namely, Pseudomonas aeruginosa, Bacillus subtilis, and Staphylococcus aureus. The findings showed that Langsat seed extracts contained more groups of compounds compared with the other two varieties, and its methanol extract demonstrated the highest inhibition zones against the three bacteria. The crude methanol extract of Duku seeds showed inhibition zones only towards Bacillus subtilis at a high concentration (1.0 mgL-1), whilst the seed extracts of Dokong showed no inhibition zones towards any of the tested bacteria.
This study aimed at investigating the presence of alkaloids and other chemical constituents in Datura stramonium (Saikaran, Jimson weed). All parts of the plant were dried, crushed and then underwent extraction by soxhlet and maceration methods. The solvents used in these methods were normal hexane (nonpolar) and ethanol (polar). Thin Layer Chromatography (TLC) and FTIR techniques were used to analyse the chemical components of jimson weed. The results showed the presence of hyoscine in all plant parts while atropine in the seeds only. The best separation was found to be when the solvent system was acetone: water: ammonia (90:07:03). Maceration method is the best and cost effective procedure for extraction.
This study investigated the recovery of phytochemical antioxidants in Dacryodes rostrata fruit using different extraction solvents. The effects of solvent of varying polarities with sequential extraction method on the recovery of phenolics, flavonoids, carotenoids and anthocyanins from different parts of the fruit (seed, pulp and peel) were determined. Their antioxidant activities were further determined using DPPH radical, ferric reducing antioxidant power (FRAP), hydroxyl radical scavenging, superoxide anion radical scavenging and phosphomolybdenum method. Dacryodes Rostrata seed had the highest total phenolic content with 50% ethanol as the most efficient extraction solvent. The highest total flavonoid content was obtained in ethyl acetate extract of fruit pulp, whereas peel extracted with hexane and 50% ethanol was the highest in total carotenoid content and total anthocyanin content, respectively. The seed extracted with 50% ethanol exhibited the strongest DPPH radical scavenging activity. Iron chelating activity measured by FRAP assay was the best in seed extracts, particularly in those polar extracts derived from water and 50% ethanol. Antioxidant activities of 50% ethanol extract of D. rostrata seed was the highest when determined by FRAP and phosphomolydenum assays. However, the influence of extraction solvents is not distinctly shown by hydroxyl radical and superoxide anion radical scavenging activities. This is the first report on the effect of various extraction solvents on the recovery of phytochemicals in D. rostrata fruit parts and the seed of D. rostrata is a potential source of polar antioxidants.
