    Sphingosine kinases (SPHKs) are conserved lipid enzymes that catalyze the formation of sphingosine-1-phosphate (S1P) through ATP-dependent phosphorylation of sphingosine. Two distinct SPHK isoforms, namely SPHK1 and SPHK2, have been identified to date, and the former has been implicated for its oncogenic roles in cancer development and progression. While SPHK1 signaling axis has been extensively studied in non-stem breast cancer cells, recent evidence has emerged to suggest a role of SPHK1 in regulating cancer stem cells (CSCs). With the clinical implications of CSCs in disease relapse and metastasis, it is believed that therapeutic approaches that can eradicate both non-stem cancer cells and CSCs could be a key to cancer cure. In this review, we first explore the oncogenic functions of sphingosine kinase 1 in human cancers and summarize current research findings of SPHK1 signaling with a focus on breast cancer. We also discuss the therapeutic potentials and perspectives of targeting SPHK1 signaling in breast cancer and cancer stem cells. We aim to offer new insights and inspire future studies looking further into the regulatory functions of SPHK1 in CSC-driven tumorigenesis, uncovering novel therapeutic avenues of using SPHK1-targeted therapy in the treatment of CSC-enriched refractory cancers.

    The 1998 Nipah virus outbreak in Malaysia resulted in major financial losses to the multi-million-dollar swine industry. While productivity and biosecurity of pig farms have improved since, biosecurity in some farms remains substandard with farmers struggling to adapt to current national pig farming policies. Farm viability and animal health depends on farmers' role as decision-makers in managing disease threats and other aspects of farm management. This study aimed to describe the mental model of farmers in making decisions about disease prevention and control measures during the 1998 Nipah virus outbreak, and in 2019, 20 years after the last reported Nipah case. Using a qualitative approach, in-depth, semi-structured interviews with 12 pig farmers (mostly small-scale or medium-scale farms) were conducted in three states in Malaysia. Data were analyzed via inductive content analysis. Thirty-six unique dimensions in the mental model were identified, representing six interrelated themes corresponding to participants' decision making related to disease prevention and control: drivers of action to prevent and control disease; perception of practice options; individual determinants of familiar practices; external social factors; external economic factors; and additional external factors. Key drivers of disease control and prevention responses during the Nipah outbreak included heightened perception of risk, emotions, perceived economic loss, and subjective norms whereas key drivers in 2019 included perception of risk, perceived effectiveness, perceived benefits, and other dimensions such as perception of the future, perceived economic cost, barriers, and loss. An unfavorable future outlook, perceived economic factors, and socio-political and personal factors currently hinders farm improvement and adoption of Pig Farming Areas (PFAs) and Modern Pig Farming (MPF) systems. Private sector service providers and veterinarians are highly influential in advocating for good biosecurity, herd health, and animal health intervention practices. Insights gained can inform the development of strategic policies and interventions.

    OBJECTIVE: To investigate the prevalence and incidence of TB by focusing on its environmental risk factor in Malaysia.

    CONTENT: Databases search of Scopus, ScienceDirect, PubMed, Directory of Open Access Journals (DOAJ), Cumulative Index to Nursing and Allied Health Literature (CINAHL) Plus, MyJournal, Biblioteca Regional de Medicina (BIREME), BioMed Central (BMC) Public Health, Medline, Commonwealth Agricultural Bureaux (CAB), EMBASE (Excerpta Medica dataBASE) OVID, and Web of Science (WoS) was performed, which include the article from 1st January 2008 until 31st August 2018 using medical subject heading (MeSH). Articles initially identified were screened for relevance.

    SUMMARY: Out of 744 papers screened, nine eligible studies did meet our inclusion criteria. Prison and housing environments were evaluated for TB transmission in living environment, while the other factor was urbanization. However, not all association for these factors were statistically significant, thus assumed to be conflicting or weak to end up with a strong conclusion.

    OUTLOOK: Unsustainable indoor environment in high congregate setting and overcrowding remained as a challenge for TB infection in Malaysia. Risk factors for transmission of TB, specifically in high risk areas, should focus on the implementation of specialized program. Further research on health care environment, weather variability, and air pollution are urgently needed to improve the management of TB transmission.

    OBJECTIVES: Glucosamine, chondroitin and diacerein are natural compounds commonly used in treating osteoarthritis. Their concomitant intake may trigger drug-natural product interactions. Cytochrome P450 (CYP) has been implicated in such interactions. Cytochrome P450 2D6 (CYP2D6) is a major hepatic CYP involved in metabolism of 25% of the clinical drugs. This study aimed to investigate the inhibitory effect of these antiarthritic compounds on CYP2D6.

    METHODS: CYP2D6 was heterologously expressed in Escherichia coli. CYP2D6-antiarthritic compound interactions were studied using in vitro enzyme kinetics assay and molecular docking.

    RESULTS: The high-performance liquid chromatography (HPLC)-based dextromethorphan O-demethylase assay was established as CYP2D6 marker. All glucosamines and chondroitins weakly inhibited CYP2D6 (IC50 values >300 µM). Diacerein exhibited moderate inhibition with IC50 and K i values of 34.99 and 38.27 µM, respectively. Its major metabolite, rhein displayed stronger inhibition potencies (IC50=26.22 μM and K i =32.27 μM). Both compounds exhibited mixed-mode of inhibition. In silico molecular dockings further supported data from the in vitro study. From in vitro-in vivo extrapolation, rhein presented an area under the plasma concentration-time curve (AUC) ratio of 1.5, indicating low potential to cause in vivo inhibition.

    CONCLUSIONS: Glucosamine, chondroitin and diacerein unlikely cause clinical interaction with the drug substrates of CYP2D6. Rhein, exhibits only low potential to cause in vivo inhibition.

    As the world tries to cope with the devastating effects of the COVID-19 pandemic and emerging variants of the virus, COVID-19 vaccination has become an even more critical tool toward normalcy. The effectiveness of the vaccination program and specifically vaccine uptake and coverage, however, is a function of an individual's knowledge and individual opinion about the disease and available vaccines. This study investigated the knowledge, attitudes, and resulting community practice(s) associated with the new COVID-19 variants and vaccines in Bangladesh, Colombia, India, Malaysia, Zimbabwe, and the USA. A cross-sectional web-based Knowledge, Attitudes, and Practices (KAP) survey was administered to respondents living in six different countries using a structured and multi-item questionnaire. Survey questions were translated into English, Spanish, and Malay to accommodate the local language in each country. Associations between KAP and a range of explanatory variables were assessed using univariate and multiple logistic regression. A total of 781 responses were included in the final analysis. The Knowledge score mean was 24 (out of 46), Attitude score 28.9 (out of 55), and Practice score 7.3 (out of 11). Almost 65% of the respondents reported being knowledgeable about COVID-19 variants and vaccination, 55% reported a positive attitude toward available COVID-19 vaccines, and 85% reported engaging in practices that supported COVID-19 vaccination. From the multiple logistic models, we found post-graduate education (AOR = 1.83, 95% CI: 1.23-2.74) and an age range 45-54 years (AOR = 5.81, 95% CI: 2.30-14.69) to be significantly associated with reported COVID-19 knowledge. In addition, positive Attitude scores were associated with respondents living in Zimbabwe (AOR = 4.49, 95% CI: 2.04-9.90) and positive Practice scores were found to be associated with people from India (AOR = 3.68, 95% CI: 1.15-11.74) and high school education (AOR = 2.16, 95% CI: 1.07-4.38). This study contributes to the identification of socio-demographic factors associated with poor knowledge, attitudes, and practices relating to COVID-19 variants and vaccines. It presents an opportunity for collaboration with diverse communities to address COVID-19 misinformation and common sources of vaccine hesitancy (i.e., knowledge, attitudes, and practices).

    Wax is an organic compound found on the surface of lignocellulose biomass to protect plants from physical and biological stresses in nature. With its small mass fraction in biomass, wax has been neglected from inclusion in the design of the biorefinery process. This study investigated the interfering effect of wax in three types of lignocellulosic biomass, including rice straw (RS), Napier grass (NG), and sugarcane bagasse (SB). In this study, although small fractions of wax were extracted from RS, NG, and SB at 0.57%, 0.61%, and 1.69%, respectively, dewaxing causes changes in the plant compositions and their functional groups and promotes dissociations of lignocellulose fibrils. Additionally, dewaxing of biomass samples increased reducing sugar by 1.17-, 1.04-, and 1.35-fold in RS, NG, and SB, respectively. The ethanol yield increased by 1.11-, 1.05-, and 1.23-fold after wax removal from RS, NG, and SB, respectively. The chemical composition profiles of the waxes obtained from RS, NG, and SB showed FAME, alcohol, and alkane as the major groups. According to the conversion rate of the dewaxing process and ethanol fermentation, the wax outputs of RS, NG, and SB are 5.64, 17.00, and 6.00 kg/ton, respectively. The current gasoline price is around USD 0.903 per liter, making ethanol more expensive than gasoline. Therefore, in order to reduce the cost of ethanol in the biorefinery industry, other valuable products (such as wax) should be considered for commercialization. The cost of natural wax ranges from USD 2 to 22 per kilogram, depending on the source of the extracted wax. The wax yields obtained from RS, SB, and NG have the potential to increase profits in the biorefining process and could provide an opportunity for application in a wider range of downstream industries than just biofuels.

    JPEG is the most commonly utilized image coding standard for storage and transmission purposes. It achieves a good rate-distortion trade-off, and it has been adopted by many, if not all, handheld devices. However, often information loss occurs due to transmission error or damage to the storage device. To address this problem, various coefficient recovery methods have been proposed in the past, including a divide-and-conquer approach to speed up the recovery process. However, the segmentation technique considered in the existing method operates with the assumption of a bi-modal distribution for the pixel values, but most images do not satisfy this condition. Therefore, in this work, an adaptive method was employed to perform more accurate segmentation, so that the real potential of the previous coefficient recovery methods can be unleashed. In addition, an improved rewritable adaptive data embedding method is also proposed that exploits the recoverability of coefficients. Discrete cosine transformation (DCT) patches and blocks for data hiding are judiciously selected based on the predetermined precision to control the embedding capacity and image distortion. Our results suggest that the adaptive coefficient recovery method is able to improve on the conventional method up to 27% in terms of CPU time, and it also achieved better image quality with most considered images. Furthermore, the proposed rewritable data embedding method is able to embed 20,146 bits into an image of dimensions 512×512.

    Sponge-derived fungi have recently attracted attention as an important source of interesting bioactive compounds. Aspergillus nomius NC06 was isolated from the marine sponge Neopetrosia chaliniformis. This fungus was cultured on rice medium and yielded four compounds including three new oxisterigmatocystins, namely, J, K, and L (1, 2, and 3), and one known compound, aspergillicin A (4). Structures of the compounds were elucidated by 1D and 2D NMR spectroscopy and by high-resolution mass spectrometry. The isolated compounds were tested for cytotoxic activity against HT 29 colon cancer cells, where compounds 1, 2, and 4 exhibited IC50 values of 6.28, 15.14, and 1.63 µM, respectively. Under the fluorescence microscope by using a double staining method, HT 29 cells were observed to be viable, apoptotic, and necrotic after treatment with the cytotoxic compounds 1, 2, and 4. The result shows that compounds 1 and 2 were able to induce apoptosis and cell death in HT 29 cells.

    Bee venom (BV) is a typical toxin secreted by stingers of honeybee workers. BV and BV therapy have long been attractive to different cultures, with extensive studies during recent decades. Nowadays, BV is applied to combat several skin diseases, such as atopic dermatitis, acne vulgaris, alopecia, vitiligo, and psoriasis. BV is used extensively in topical preparations as cosmetics and used as dressing for wound healing, as well as in facemasks. Nevertheless, the safety of BV as a therapeutic choice has always been a concern due to the immune system reaction in some people due to BV use. The documented unfavorable impact is explained by the fact that the skin reactions to BV might expand to excessive immunological responses, including anaphylaxis, that typically resolve over numerous days. This review aims to address bee venom therapeutic uses in skin cosmetics.

    Infectious bronchitis virus (IBV) poses significant financial and biosecurity challenges to the commercial poultry farming industry. IBV is the causative agent of multi-systemic infection in the respiratory, reproductive and renal systems, which is similar to the symptoms of various viral and bacterial diseases reported in chickens. The avian immune system manifests the ability to respond to subsequent exposure with an antigen by stimulating mucosal, humoral and cell-mediated immunity. However, the immune response against IBV presents a dilemma due to the similarities between the different serotypes that infect poultry. Currently, the live attenuated and killed vaccines are applied for the control of IBV infection; however, the continual emergence of IB variants with rapidly evolving genetic variants increases the risk of outbreaks in intensive poultry farms. This review aims to focus on IBV challenge-infection, route and delivery of vaccines and vaccine-induced immune responses to IBV. Various commercial vaccines currently have been developed against IBV protection for accurate evaluation depending on the local situation. This review also highlights and updates the limitations in controlling IBV infection in poultry with issues pertaining to antiviral therapy and good biosecurity practices, which may aid in establishing good biorisk management protocols for its control and which will, in turn, result in a reduction in economic losses attributed to IBV infection.

    Metabolic dysfunction-associated fatty liver disease (MAFLD) is an excessive buildup of liver lipids closely associated with various kinds of undesirable metabolic effects and oxidative stress. We aimed to investigate the protective and therapeutic effects of orlistat on metabolic syndrome and oxidative stress parameters in high-fat diet (HFD) induced-MAFLD rats. Twenty-four male Sprague-Dawley rats were randomly divided into four groups (n = 6/group), i.e., Normal control (N), HFD, HFD + orlistat (HFD + O) (10 mg/kg/day administered concomitantly for 12 weeks as a protective model), and obese+orlistat (OB + O) (10 mg/kg/day administered 6 weeks after induction of obesity as a therapeutic model) groups. After 12 weeks, the HFD group had significantly increased Lee obesity index, serum alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, total cholesterol, triglyceride, low-density lipoprotein levels, liver total cholesterol and triglyceride levels, insulin resistance and non-alcoholic steatohepatitis (NASH) together with decreased serum high-density lipoprotein level. Additionally, the HFD group also showed increased Nrf2 translocation to the nucleus with high Keap1 expression and increased liver oxidative stress parameters. Orlistat significantly improved all these alterations in HFD rats. We demonstrated that orlistat might have protective and therapeutic effects against HFD-induced MAFLD rats by its activation on Nrf2 signaling pathway, which subsequently improved metabolic syndrome and oxidative stress parameters.

    Non-point source (NPS) pollution has become a vital contaminant source affecting the water environment because of its wide distribution, hydrodynamic complexity, and difficulty in prevention and control. In this study, the identification and evaluation of NPS pollution risk based on landscape pattern were carried out in the Hanjiang River basin above Ankang hydrological section, Shaanxi province, China. Landscape distribution information was obtained through land use data, analyzing the contribution of "source-sink" landscape to NPS pollution through the location-weighted landscape contrast index. Using the NPS pollution risk index to identify and evaluate the regional NPS pollution risk considering the slope, cost distance, soil erosion, and precipitation erosion affect migration of pollutants. The results showed that (i) the pollution risk was generally high in the whole watershed, and the sub-watersheds dominated by "source" landscapes account for 74.61% of the whole basin; (ii) the high-risk areas were distributed in the central, eastern, and western regions of the river basin; the extremely high-risk areas accounted for 12.7% of the whole watershed; and the southern and northern regions were dominated by forestland and grassland with little pollution risk; (iii) "source" landscapes were mostly distributed in areas close to the river course, which had a great impact on environment, and the landscape pattern units near the water body needed to be further adjusted to reduce the influence of NPS pollution.

    Inconsistencies have been reported on the effect of sex on aldosterone (ALDO) levels leading to clinical confusion. The reasons for these inconsistencies are uncertain but include estrogen and/or its receptor modulating target gene responses to mineralocorticoid receptor activation and ALDO secretagogues' levels. This study's goal was to determine whether ALDO's biosynthesis also differed by sex. Two approaches were used. First, plasma renin activity and aldosterone were measured in rats. Both were significantly higher in males. Secondly, using rat zona glomerulosa (ZG) cells, we assessed three ex vivo areas: (1) activity/levels of early steps in ALDO's biosynthesis (StAR and CYP11A1); (2) activity/levels of a late step (CYP11B2); and (3) the status of the mineralocorticoid receptor (MR)-mediated, ultrashort feedback loop. Females had higher expression of CYP11A1 and StAR and increased CYP11A1 activity (increased pregnenolone/corticosterone levels) but did not differ in CYP11B2 expression or activity (ALDO levels). Activating the ZG's MR (thereby activating the ultrashort feedback loop) reduced CYP11B2's activity similarly in both sexes. Exvivo, these molecular effects were accompanied, in females, by lower ALDO basally but higher ALDO with angiotensin II stimulation. In conclusion, we documented that not only was there a sex-mediated difference in the activity of ALDO's biosynthesis but also these differences at the molecular level help explain the variable reports on ALDO's circulating levels. Basally, both in vivo and ex vivo, males had higher ALDO levels, likely secondary to higher ALDO secretagogue levels. However, in response to acute stimulation, ALDO levels are higher in females because of the greater levels and/or activity of their StAR/CYP11A1.

    Circular RNAs (circRNAs) are abundantly expressed in cancer. Their resistance to exonucleases enables them to have potentially stable interactions with different types of biomolecules. Alternative splicing can create different circRNA isoforms that have different sequences and unequal interaction potentials. The study of circRNA function thus requires knowledge of complete circRNA sequences. Here we describe psirc, a method that can identify full-length circRNA isoforms and quantify their expression levels from RNA sequencing data. We confirm the effectiveness and computational efficiency of psirc using both simulated and actual experimental data. Applying psirc on transcriptome profiles from nasopharyngeal carcinoma and normal nasopharynx samples, we discover and validate circRNA isoforms differentially expressed between the two groups. Compared with the assumed circular isoforms derived from linear transcript annotations, some of the alternatively spliced circular isoforms have 100 times higher expression and contain substantially fewer microRNA response elements, showing the importance of quantifying full-length circRNA isoforms.

    Today's intelligent computing environments, including the Internet of Things (IoT), Cloud Computing (CC), Fog Computing (FC), and Edge Computing (EC), allow many organizations worldwide to optimize their resource allocation regarding the quality of service and energy consumption. Due to the acute conditions of utilizing resources by users and the real-time nature of the data, a comprehensive and integrated computing environment has not yet provided a robust and reliable capability for proper resource allocation. Although traditional resource allocation approaches in a low-capacity hardware resource system are efficient for small-scale resource providers, for a complex system in the conditions of dynamic computing resources and fierce competition in obtaining resources, they cannot develop and adaptively manage the conditions optimally. To optimize the resource allocation with minimal delay, low energy consumption, minimum computational complexity, high scalability, and better resource utilization efficiency, CC/FC/EC/IoT-based computing architectures should be designed intelligently. Therefore, the objective of this research is a comprehensive survey on resource allocation problems using computational intelligence-based evolutionary optimization and mathematical game theory approaches in different computing environments according to the latest scientific research achievements.

    The COVID-19 pandemic has inspired unprecedented data collection and computer vision modelling efforts worldwide, focused on the diagnosis of COVID-19 from medical images. However, these models have found limited, if any, clinical application due in part to unproven generalization to data sets beyond their source training corpus. This study investigates the generalizability of deep learning models using publicly available COVID-19 Computed Tomography data through cross dataset validation. The predictive ability of these models for COVID-19 severity is assessed using an independent dataset that is stratified for COVID-19 lung involvement. Each inter-dataset study is performed using histogram equalization, and contrast limited adaptive histogram equalization with and without a learning Gabor filter. We show that under certain conditions, deep learning models can generalize well to an external dataset with F1 scores up to 86%. The best performing model shows predictive accuracy of between 75% and 96% for lung involvement scoring against an external expertly stratified dataset. From these results we identify key factors promoting deep learning generalization, being primarily the uniform acquisition of training images, and secondly diversity in CT slice position.

    Map-based neuronal models have received much attention due to their high speed, efficiency, flexibility, and simplicity. Therefore, they are suitable for investigating different dynamical behaviors in neuronal networks, which is one of the recent hottest topics. Recently, the memristive version of the Rulkov model, known as the m-Rulkov model, has been introduced. This paper investigates the network of the memristive version of the Rulkov neuron map to study the effect of the memristor on collective behaviors. Firstly, two m-Rulkov neuronal models are coupled in different cases, through electrical synapses, chemical synapses, and both electrical and chemical synapses. The results show that two electrically coupled memristive neurons can become synchronous, while the previous studies have shown that two non-memristive Rulkov neurons do not synchronize when they are coupled electrically. In contrast, chemical coupling does not lead to synchronization; instead, two neurons reach the same resting state. However, the presence of both types of couplings results in synchronization. The same investigations are carried out for a network of 100 m-Rulkov models locating in a ring topology. Different firing patterns, such as synchronization, lagged-phase synchronization, amplitude death, non-stationary chimera state, and traveling chimera state, are observed for various electrical and chemical coupling strengths. Furthermore, the synchronization of neurons in the electrical coupling relies on the network's size and disappears with increasing the nodes number.

    BACKGROUND: Although various studies have been conducted on the effects of seasonal climate changes or emotional variables on the risk of AMI, many of them have limitations to determine the predictable model. The currents study is conducted to assess the effects of meteorological and emotional variables on the incidence and epidemiological occurrence of acute myocardial infarction (AMI) in Sari (capital of Mazandaran, Iran) during 2011-2018.

    METHODS: In this study, a time series analysis was used to determine the variation of variables over time. All series were seasonally adjusted and Poisson regression analysis was performed. In the analysis of meteorological data and emotional distress due to religious mourning events, the best results were obtained by autoregressive moving average (ARMA) (5,5) model.

    RESULTS: It was determined that average temperature, sunshine, and rain variables had a significant effect on death. A total of 2375 AMI's were enrolled. Average temperate (°C) and sunshine hours a day (h/day) had a statistically significant relationship with the number of AMI's (β = 0.011, P = 0.014). For every extra degree of temperature increase, the risk of AMI rose [OR = 1.011 (95%CI 1.00, 1.02)]. For every extra hour of sunshine, a day a statistically significant increase [OR = 1.02 (95% CI 1.01, 1.04)] in AMI risk occurred (β = 0.025, P = 0.001). Religious mourning events increase the risk of AMI 1.05 times more. The other independent variables have no significant effects on AMI's (P > 0.05).

    CONCLUSION: Results demonstrate that sunshine hours and the average temperature had a significant effect on the risk of AMI. Moreover, emotional distress due to religious morning events increases AMI. More specific research on this topic is recommended.

    The stability and high yielding of Vigna subterranea L. Verdc. genotype is an important factor for long-term development and food security. The effects of G × E interaction on yield stability in 30 Bambara groundnut genotypes in four different Malaysian environments were investigated in this research. The experiment used a randomized complete block design with three replications in each environment. Over multiple harvests, yield component traits such as the total number of pods per plant, fresh pods weight (g), hundred seeds weight (g), and yield per hectare were evaluated in the main and off-season in 2020 and 2021.
