    The extraction of catfish (Clarias gariepinus) bone gelatin was optimized by using Response Surface Methodology (RSM) involving 4-factors, 5-levels Central Composite Design (CCD). The optimum conditions for extraction were produced by a pre-treatment of 3.35% HCl for 14.5 h along with hot water extraction at 67.23°C for 5.2 h. Results showed that the predicted yield by RSM (61.81%) was closely matched the experimental yield of 60.54%. The results also indicated that the extracted bone gelatin possessed high protein content (81.75%) and imino acid (proline and hydroxyproline) (144 residues per 1000 residues), with gel strength (230.25 g), viscosity (4.64 mPa.s) and isoionic point (5.35) comparable to that of bovine gelatin. The results suggested that RSM is a great optimizing tool for extraction of gelatin from clarias catfish bone and values of the physicochemical properties of gelatin are higher or comparable than those from other fish species and bovine gelatin.

    Antibiotic susceptibility and genetic diversity of E. coli isolated from cultured catfish and their surrounding environment were determined. The levels of resistance of the E. coli isolates towards six different antibiotics tested differed considerably. Though the isolates displayed resistance towards some of the antibiotics tested, none of the isolates showed resistant towards norfloxacin, sulphametoxazole/trimethoprim and chloramphenicol. RAPD-PCR analysis using single primer and primers combination clustered the E. coli isolates into 3 and 5 groups, respectively. The results of this study suggest that the E. coli isolates from the catfish and their surrounding environment derived from a mixture of sensitive and resistant strains with diverse genetic contents. The use of the RAPD analysis is sufficiently discriminatory for the typing of the E. coli isolates.

    Background. Fish species may be identified based on their unique otolith shape or contour. Several pattern recognition methods have been proposed to classify fish species through morphological features of the otolith contours. However, there has been no fully-automated species identification model with the accuracy higher than 80%. The purpose of the current study is to develop a fully-automated model, based on the otolith contours, to identify the fish species with the high classification accuracy. Methods. Images of the right sagittal otoliths of 14 fish species from three families namely Sciaenidae, Ariidae, and Engraulidae were used to develop the proposed identification model. Short-time Fourier transform (STFT) was used, for the first time in the area of otolith shape analysis, to extract important features of the otolith contours. Discriminant Analysis (DA), as a classification technique, was used to train and test the model based on the extracted features. Results. Performance of the model was demonstrated using species from three families separately, as well as all species combined. Overall classification accuracy of the model was greater than 90% for all cases. In addition, effects of STFT variables on the performance of the identification model were explored in this study. Conclusions. Short-time Fourier transform could determine important features of the otolith outlines. The fully-automated model proposed in this study (STFT-DA) could predict species of an unknown specimen with acceptable identification accuracy.     Genomic DNA of 13 fish (n=13) species consist of four freshwater which were catfish (Clarias gariepinus), shark catfish (Pangasius larnaudii), tilapia (Oreochromis mossambicus), perch (Lates calcarifer) and nine marine species which were black pomfret (Parastromateus niger), anchovy (Stolephorus commersonii), mabong (Rastrelliger kanagurta), red snapper (Lutjanus erythropterus), herring (Chirocentrus dorab), ray fish (Himantura gerrardii), sardine (Decapterus macrosoma), mackerel (Euthynnus affinis) and tuna (Thunnus tuna) were differentiated using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Seven endonucleases of AluI, BsaJI, HaeIII, HindIII, HinfI, MboI and MboII were examined for the ability to digest cyt b amplicon from each species. Genomic DNA of pork (Sus scrofa domestica) were differentiated from fishes by comparing the digestion patterns produced by similar amplified region and enzymes used. In the present study, it was demonstrated that fishes and pork DNA genome were successfully differentiated using all endonucleases except for HindIII. Thus, PCR-RFLP analysis was found useful for future pork DNA detection in fish products.

    This study was conducted to determine the morphology, prevalence and mean intensity of monogeneans on gill filaments of striped catfish Pangasianodon hypophthalmus (Pangasiidae).P. hypophthalmus are important economic fish in Southeast Asia region and predominantly produced in Asian countries such as Malaysia, Bangladesh, Indonesia, Vietnam, Laos, China and Cambodia through aquaculture. Thirty fish were sampled from an earthen pond located in Kuala Besut, Terengganu, in the period from January to February, 2018. The monogenean parasites were removed from the gill filaments and counted. For the morphological study, the extracted monogeneanswere mounted on a slide by using drop of ammonium picrate-glycerin (APG) and then were observed under the Compound Advanced Research Microscope. The drawing was done with the aid of lucida camera attached to compound microscope. The monogenean parasite was identified as Thaparocleidussp based on the characteristics of the morphology and morphometrics of the parasite, which did not differ significantly from the previous descriptions of the same species discovered in other geographic locations. The prevalence and mean intensity levels were 100% and 106.07 parasites per fish, respectively. The monogenean parasites discovered throughout this examination were recorded and the data obtained was summarized.

    Scanty information exists pertaining to the length-weight relationship (LWR) and length-length relationship (LLR) parameters of longsnouted catfish, Plicofollis argyropleuron in lotic systems throughout the northern part of Peninsular Malaysia. It is vital to reveal these biological properties of P. argyropleuron in Kuala Muda and Merbok estuary for future management and to increase knowledge about this fish stocks. The fish samples were randomly collected in the estuary area of Kuala Muda and Merbok, Kedah for 10 months from March 2009 to December 2009. The values of the exponent b in the LWR equations (W = aL(b) ) were approximately 3, indicating an isometric growth with high correlation coefficient (r(2)). The value of LLR (r(2)>0.9) indicated that they are highly significant and highly correlated. These parameters are essential for evaluating the relative condition of fish and species managements as well as their fisheries and stock assessment.

    A total of 17 species of the genus Bifurcohaptor Jain, 1958 have been reported from two fish families namely Bagridae Bleeker, 1858 (Mystus vittatus (Bloch, 1794), M. tengara (Hamilton, 1822), M. keletius (Valenciennes, 1840), Hemibagrus nemurus (Valenciennes, 1840), Rita rita (Hamilton, 1822) and Sperata seenghala (Sykes, 1839)) and Sisoridae Bleeker, 1858 (Bagarius bagarius (Hamilton, 1822)). Out of these, only two species viz. B. indicus and B. giganticus are found valid in India, parasitizing gills of Mystus spp. and Bagarius sp. Taxonomic studies suggest, present specimen of B. indicus and B. giganticus, both are morphologically close to species described by Jain (1958), except morphometric variations and posses 7 pairs of marginal hooks instead of 6 pairs. Present manuscript delves with the characterization of B. indicus and B. giganticus reported from India, using molecular techniques. Partial mt COI nucleotide sequence based insilico protein analysis and partial 28S and ITS-1 rDNA based phylogenetic analysis, estimated by Neighbour-joining (NJ) and Minimum Evolution (ME) methods revealed that the species of the genus Bifurcohaptor are genetically distinct and valid. The grouping of Bifurcohaptor spp. with other representatives of family Dactylogyridae supports morphology based placement into family Dactylogyridae. Present and previous host-parasite information suggests both Bifurcohaptor spp. are species specialist however, the genus Bifurcohaptor is generalist at generic level.

     The prevalence of antibiotic resistant bacteria in aquaculture has reached alarming proportions and intensified the search for microbe derived antimicrobial compounds. This study isolated bacteria from the intestine of Sagor catfish (Hexanematichthys sagor) and screened it for antagonistic properties. Five out of 334 bacterial isolates inhibited growth of fish pathogens. The 5 bacterial strains included relatives of Shewanella haliotis, Myroides odoratimimus, Vibrio harveyi, Vibrio alginolyticus and Alcaligenes faecalis. The growth profiles and probiotic properties of these bacteria were examined. The results showed that the isolate 9 (3) 7.5.2.1, whose closest relative was S. haliotis exhibited growth and probiotic advantage compared to the other bacterial strains, such as highest doubling time and the ability to survive at all experimental temperatures (18 to 60℃) , and bile concentrations (0.01 to 1.00%) and pH (pH2 to 9) . While the bacteria with probiotic properties were successfully isolated. Further study is necessary to examine the efficiency of the probiotic candidate bacteria in boosting fish immunity against pathogens.

    Discrimination of different fishes can be done through different means which includes morphological appearance. When two fishes are successfully hybridized, they produce progenies that have shared morphology between their pure parent, hence, making morphometric characterization an important aspect of hybrid discrimination. However, erythrocyte characterization is also a simpler method for characterization. The dataset presented in this article represents the traditional morphological data, truss network data and erythrocyte data of pure and novel hybrids from reciprocal crosses of African catfish Clarias gariepinus and Asian catfish Pangasianodon hypophthalmus. Breeding of the broodstocks was done to produce pure and hybrid progenies which were maintained for a period of four to six months. Based on the cross combinations and morphotypes, traditional measurement of twenty-five morphological characters and five meristic counts were recorded. Thereafter pictures of the different fish groups were used to determine values of thirty-six distances between ten landmark points. The morphological abnormality of the hybrids at market size is also presented in this data article for the very first time. Blood was then collected from the caudal peduncle of ten fish per group and smeared on a slide for observation under a compound microscope (at 100 × magnification). Data gotten included erythrocytes parameters such as cell major axis, cell minor axis, nucleus major axis, nucleus minor axis cell area, nucleus area, cell volume, and nucleus volume. Data recording was through the Microsoft excel spreadsheet; which was also used to process the data to get the exclusive ranges of values for paired progenies. The data as presented is associated with the research article "Morphological characterization of the progenies of pure and reciprocal crosses of Pangasianodon hypophthalmus (Sauvage, 1878) and Clarias gariepinus (Burchell, 1822)" [1]. The dataset presented in this article can be used for easy identification of the novel hybrid progenies of the African Catfish and Asian Catfish.

    This study investigated the use of electric-shock in inducing triploidy in African catfish Clarias gariepinus. To achieve this, three voltages (9, 12, 21 V) were applied for different durations (3, 5, 10 min). The shock was initiated approximately three minutes after fertilization followed by incubation in ambient temperature. After incubation, hatchability and survival rates were determined while ploidy status of the treatment fishes was confirmed in one-month-old fingerlings using the exclusive triploid range of the erythrocyte major axis previously reported for the same species (11.9-14.9 μm) and by cytogenetic analysis of the chromosome. The results showed triploidy were achieved in 10 to 85% of the treatment groups. A consistent trend of decrease in hatchability and an increase in triploidy rate was observed with increased electroporation voltages and shock durations. The mean erythrocyte major axis length of triploid progenies (3n = 84) was observed to be between 11.3-14.6 μm and was higher than the range of 7.0-10.5 μm recorded for diploid progenies (2n = 56). It was concluded that electric shock can be used to induce triploidy in African catfish C. gariepinus.

    The optimum distance and duration of ultraviolet (UV) irradiation for the complete inactivation of African catfish Clarias gariepinus egg nucleus was investigated in this study. The UV light was suspended above the unfertilized eggs at four distances (5, 10, 20 and 30 cm) and for five durations (1, 2, 3, 4 and 5 min). Then, the irradiated eggs were activated with sperm from diploid C. gariepinus and cold shocked at 5°C for 5 min just moments before cell cleavage. Ploidy analysis was performed using karyotype chromosome counting. The results obtained suggested that the further the distance, the better the hatchability rate, however prolonged duration seemed to significantly reduced hatchability. All treatments with surviving progenies at the end of the study showed evidence of successfully diploid gynogen (2n = 56) induction at different percentages. However, the optimal protocol that gave a moderately high hatchability/survival rate and completely induced gynogens was exposure of the eggs to UV irradiation at 20 cm for 1 min.     Breeding and larval performance of novel hybrids from reciprocal crosses of Asian catfish Pangasianodon hypophthalmus (Sauvage, 1878) and African catfish Clarias gariepinus (Burchell, 1822) were investigated in this study. Spawning was by hormonal injection of brood fish, artificial fertilization, and incubation in triplicate aquarium tanks (0.5 × 0.5 × 0.5 m3) with continuous aeration. Reciprocal crosses (♀C. gariepinus × ♂P. hypophthalmus and ♀P. hypophthalmus × ♂C. gariepinus) had lower hatchability (≤50%) than their pure siblings (≥75%). Fish from all crosses survived until the juvenile stage but survival at 35 days post hatching (dph) was higher for pure C. gariepinus sib. ♀C. gariepinus × ♂P. hypophthalmus was observed to be less resistant to degradation of water quality than the other crosses, however it had higher body weight compared with the other crosses that showed similar performance. Morphological comparison of surviving juvenile at 35 dph, showed that all ♀P. hypophthalmus × ♂C. gariepinus and 13% of the ♀C. gariepinus × ♂P. hypophthalmus exhibited the very same morphology as that of their maternal parent species, while the other portion of the ♀C. gariepinus × ♂P. hypophthalmus cross exhibited morphological traits that were intermediate between those of both parent species. This study been the first successful attempt to hybridize both species and therefore, laid the groundwork for further studies on the aquaculture potentials of the novel hybrids.

    To obtain well spread chromosomes, the cytogenetic protocol for Pangasianodon hypophthalmus and Clarias gariepinus were optimized. This includes, the colchicine concentration (0.01%, 0.025%, 0.05%)/exposure duration (1, 3, and 5 h), hypotonic solution (distilled water or 0.075M KCl solution)/exposure duration (30 min, 1, and 2 h), the time of cell suspension preparation (at hypotonic treatment or before slide preparation) and chromosome aging period (0, 3, and 7 days in Carnoy's fixative). In addition, the type (i.e., fin, gill or kidney) and the amount of tissue (10, 50, 100 or 150 mg) were also investigated. Regardless of the species, the result obtained showed that well-spread chromosomes could be obtained using the following optimized protocol: Juveniles are injected with 0.05% colchicine (at one ml kg-1) and allowed to swim for 3 h. Then, 50 mg of gill tissue is made into cell suspension in 0.075M KCl for 1 h. The cell suspension is treated in Carnoy's fixative (changed three times at 20 min interval) and then aged for 3 days. Finally, chromosome slides are made and stained with 10% Giemsa for 1 h.

    Twenty-five traditional and thirty-four geometric morphometric comparisons were carried out on pure and reciprocal crosses of Pangasianodon hypophthalmus (Sauvage, 1878) and Clarias gariepinus (Burchell, 1822). Thirty fish samples each of the C. gariepinus (CH), P. hypophthalmus (PH), Pangapinus (♀PH × ♂CG) and the two distinct morphotypes of the Clariothalmus (♀CG × ♂PH) (Clarias-like and Panga-like) between the ages of four and six months were used for this study. Phenotypically, the Clarias-like Clariothalmus and the Pangapinus progenies were indistinguishable from their maternal parents while the Panga-like Clariothalmus was a phenotypic intermediary of the putative parents but looks more closely to the paternal parent. Hence, both univariate proportion and multivariate analysis of the collected data successfully separated the different fishes into three multivariate spaces. The analysis of the dendrogram with complete linkage and Euclidean distance further showed the close relationship of the isolated Panga-like Clariothalmus progenies to the paternal parent, however, Clarias-like Clariothalmus and the Pangapinus were completely intermingled with their maternal parents. The most important index of discrimination of these fishes into different multivariate spaces was the fin characteristic which showed 100% exclusive ranges for the individual groups in many cases.

    This study aimed to document the fish diversity and water quality at Semariang mangrove area, Kuching, Sarawak, which is located at the eastern part of Kuching Wetland National Park. Field samplings were carried out in 2009 during the construction of the flood mitigation channel at the eastern part of the park. A total of 21 families represented by 37 species of fish were caught from the area. The six dominant families in terms of the number of individuals caught were Mugilidae (16%), Leiognathidae (16%), Ambassidae (11%), Ariidae (9%), Lutjanidae (8%) and Plotosidae (6%). In terms of the percentage of six dominant genera based on the number of individuals caught, 16% was represented by Valamugil, 11% by Ambassis, 10% by Gazza, 9% by Arius, 8% by Lutjanus and 6% by Plotosus. The values of diversity and richness indices were lower at stations located close to the flood mitigation channel. Similarly, the concentrations of dissolved oxygen were lower and total suspended solids were significantly higher at stations close to the channel and sand mining area. Therefore, fish fauna and water quality at Semariang mangrove area were affected during the construction of the flood mitigation channel.

    Gelatin from sutchi catfish (Pangasius hypophthalmus) skin was extracted and applied in the preparation of gummy in order to determine the suitability of sutchi catfish gelatin in gummy production. The skin was subjected to pre-treatment in the following sequence; 0.8M NaCl, 0.19 N NaOH followed by 0.12 N acetic acid prior to 12 hours extraction in distilled water at 50oC. The physicochemical characteristics of sutchi catfish gelatin was analysed and compared with the commercial bovine gelatin. Gummy added with sutchi catfish gelatin was also compared with gummy added with commercial gelatin. Analysis comprises of yield, gel strength, setting point and setting time, amino acid composition, texture profile analysis and sensory evaluation. The extraction resulted in 14.47% yield of gelatin. Sutchi catfish gelatin showed higher gel strength value (360.86 g) compared to the commercial gelatin (217.37 g) which is in accordance with proline content.     Collagen was extracted from catfish (Clarias gariepnus) waste using 0.5M acetic acid and its subsequent precipitation in 2.6M NaCl. The resultant collagen was analysed with respect to its moisture content and physicochemical properties including yield, pH, protein content, colour, odour and thermal stability. A yield of 16.4% and positive collagen attributes indicate that catfish waste has potential as a collagen source. The snowy white, crystal-like and light textured collagen comprises of 5.97% protein and 0.46% moisture, and exhibits a pH of 4.75. Sensory evaluation indicates that the collagen has a slight fishy odour. Viscosity analysis indicates a steady decrease with increasing temperature over the range considered (20-50°C). The pale colour exhibited and limited odour emitted by the extracted collagen indicate that catfish waste collagen could be applied in the food industry without resulting in any undesirable food products attributes. Differential Scanning Calorimetry (DSC) analysis indicated that the collagen exhibits good thermal stability and denatures at a high temperature in a similar manner to mammalian collagen.

    Silver catfish (Pangasius sutchi) skin gelatin was extracted to determine the effects of extraction time on the functional properties of the gelatin in terms of solubility, protein solubility as a function of pH and sodium chloride concentration, emulsifying capacity and stability, water holding capacity, fat binding capacities and foaming properties. Silver catfish skins were washed in sodium chloride (NaCl) solution prior to pre-treatment in sodium hydroxide (NaOH) and acetic acid solution. Gelatin was extracted at 50ºC for 6, 8, 10 and 12 hours extraction time followed by freeze drying. The extraction of silver catfish skin gelatin at 50 ºC for 12 hours was more effective than extraction at 6, 8 and 10 hours where the gelatin was characterized by higher emulsifying capacity (52.63%), emulsifying stability (47.83%), water holding capacity (31.78 mL/g), fat binding capacities (54.76%), foaming capacity (41.47 mL) and foaming stability (56.42%) than gelatins extracted at other extraction time. The longer the extraction time, the better the functional properties of the gelatin. Based on its good functional properties, silver catfish skin gelatin may be useful in various food applications such as soups, sauces and gravies.

    Little is known on the biosafety level of Vibrio spp. in freshwater fish in Malaysia. The purpose of this study was to investigate the prevalence and concentration of Vibrio spp. and V. parahaemolyticus in
    freshwater fish using the Most Probable Number-Polymerase Chain Reaction (MPN-PCR) method. The study was conducted on 150 samples from two types of freshwater fish commonly sold at hypermarkets, i.e. Pangasius hypophthalmus (catfish) and Oreochromis sp. (red tilapia). Sampling was done on the flesh, intestinal tract and gills of each fish. The prevalence of Vibrio spp. and V. parahaemolyticus was found to be 98.67% and 24% respectively with higher percentages detected in samples from the gills followed by the intestinal tract and flesh. Vibrio spp. was detected in almost all red tilapia and catfish samples. V. parahaemolyticus was detected in 25% of the catfish samples compared to 22.6% of red tilapia fish. The density of Vibrio spp. and V. parahaemolyticus in the samples ranged from 0 to 1.1x107 MPN/g. Although the maximum value was 1.1x107 MPN/g, most samples had microbial loads ranging from 0 to >104 MPN/g. The outcome on the biosafety assessment of Vibrio spp. and V. parahaemolyticus in freshwater fish indicates another potential source of food safety issues to consumers.
