    Transforming growth factor-beta 1 (TGF-β1) has been reported to promote chondrogenic differentiation and proliferation in the multipotent stromal cell (MSCs), and the transforming growth factor-beta 3 (TGF-β3) tends to be exclusively in promoting cell differentiation alone. The objective of this study was to determine the effect of TGF-β1 and -β3 on the MSCs chondrogenic differentiation on the poly (vinyl alcohol)-chitosan-poly (ethylene glycol) (PVA-NOCC-PEG) scaffold, compared with that of monolayer and pellet cultures. In this study, P2 rabbit bone marrow-derived MSCs were seeded either on the untreated six-well plate (for monolayer culture) or onto the PVA-NOCC-PEG scaffold or cultured as a pellet culture. The cultures were maintained in a chemically defined serum-free medium supplemented with 10 ng/mL of either TGF-β1 or TGF-β3. Cell viability assay, biochemical assay, and real-time polymerase chain reaction were performed to determine the net effect of cell proliferation and chondrogenic differentiation of each of the growth factors. The results showed that the PVA-NOCC-PEG scaffold enhanced MSCs cell proliferation from day 12 to 30 (p  0.05). In terms of chondrogenic differentiation, the PVA-NOCC-PEG scaffold augmented the GAGs secretion in MSCs and the mRNA expression levels of Sox9, Col2a1, Acan, and Comp were elevated (p  0.05). In conclusion, TGF-β1 and TGF-β3 enhanced the chondrogenic differentiation of MSCs seeded on the PVA-NOCC-PEG scaffold; however, there was no significant difference between the effect of TGF-β1 and TGF-β3. Impact statement Transforming growth factor-beta (TGF-β) superfamily members is a key requirement for the in vitro chondrogenic differentiation of mesenchymal stem cells (MSCs). In this study, the effects of TGF-β1 and -β3 on MSC chondrogenic differentiation and proliferation on a novel three-dimensional scaffold, the poly(vinyl alcohol)-chitosan-poly(ethylene glycol) (PVA-NOCC-PEG) scaffold, was evaluated. In this study, the results showed both TGF-β1 and TGF-β3 can enhance the chondrogenic differentiation of MSCs seeded on the PVA-NOCC-PEG scaffold.

    A fast melt tablet (FMT) is well regarded as an alternative delivery system that might help resolve a patient's non-compliance issue. The main objective of this study was to develop a cocoa butter-based FMT. Additives, namely 5-15% of PEG 6000, beeswax, paraffin wax, and corn starch, were incorporated into the cocoa butter-based FMT to study the effects of these additives with the physical characteristic of a cocoa butter FMT. An optimum-based formulation was chosen according to the desired hardness and disintegration time and the taste masking property achieved with the model drug-dapoxetine. The analysis demonstrated that incorporating beeswax (15%) and paraffin wax (15%) could prolong the disintegration time by at least two-fold. On the contrary, the presence of corn starch was found to cause an increase in the hardness and reduction of the disintegration time. The disintegration mechanism might be presumed due to the synergistic effect of starch swelling and cocoa butter melting. The hardness value and in vitro disintegration time of the optimum formulation were recorded at 2.93 ± 0.22 kg and 151.67 ± 6.98 s. In terms of dissolution, 80% of dapoxetine was released within 30 min and the dissolution profile was comparable to the innovator product. The formulation was palatable and stable for at least 1 year. The exposure of the FMT formulation at 30 °C for 12 months was reported to be stable. Along with the sound palatability profile and high drug load capacity, the current formulation possesses the desired characteristics to be scaled up and marketed.

    In-situ bioremediation of mangrove soil contaminated with polyethylene terephthalate (PET) and polystyrene (PS) microplastics was investigated using indigenous microbial consortium with adequate capacity to degrade the plastics. Eight (8) bacteria were isolated from plastic/microplastic-inundated mangrove soil and screened for the ability to degrade PET and PS microplastics. Optical density at 600 nm and colony forming unit counts were measured to evaluate the growth response of the microbes in the presence of PS and PET microplastics at different times of exposure. Structural and surface changes that occurred post biodegradation on the microplastics were determined through EDS and SEM analysis. The obtained results demonstrated the elongation and disappearance of peaks, suggesting that the microbial consortium could modify both types of microplastics. The overall results of the microplastic degradation showed varied degrees of weight loss after 90 experimental days, with the treated plot recorded 18% weight loss. The augmented soil was increased in the concentrations of Si S, and Fe and decreased in the concentrations of C, O, Na, Mg, Al, Cl, and K after bioremediation.

    Postconsumer polyethylene terephthalate (PET) has potential applications in many areas of manufacturing, but contamination by hazardous polyvinyl chloride (PVC) in common waste streams can reduce its recyclable value. Separating collected PET-PVC mixtures before recycling remains very challenging because of the similar physicochemical properties of PET and PVC. Herein, we describe a novel flotation process with corona modification pretreatment to facilitate the separation of PET-PVC mixtures. Through water contact angle, surface free energy, X-ray photoelectron and FT-IR characterization, we found that polar hydroxyl groups can be more easily introduced on the PVC surface than on the PET surface induced by corona modification. This selective wetting can suppress the floatability of PVC, leading to the separation of PET as floating product. A reliable mechanism including two different hydrogen-abstraction pathways was established. Response surface methodology consisting of Plackett-Burman and Box-Behnken designs was adopted for optimization of the combined process, and control parameters were solved based on high-quality prediction models, with fitting from significant variables and interactions. For physical or chemical circulation strategies with PET purity prioritization, the validated purity of the product reached 96.05% at a 626 W corona power, 5.42 m/min passing speed, 24.78 mg/L frother concentration and 286 L/h air flow rate. For the energy recuperation strategy with PET recovery prioritization, the factual recovery reached 98.08% under a 601 W corona power, 6.04 m/min passing speed, 27.55 mg/L frother concentration and 184 L/h air flow rate. The current work provides technological insights into the cleaner disposal of waste plastics.

    Towards optimizing the growth of extracellular matrix to produce repair cartilage for healing articular cartilage (AC) defects in joints, scaffold-based tissue engineering approaches have recently become a focus of clinical research. Scaffold-based approaches by electrospinning aim to support the differentiation of chondrocytes by providing an ultrastructure similar to the fibrillar meshwork in native cartilage. In a first step, we demonstrate how the blending of chitosan with poly(ethylene oxide) (PEO) allows concentrated chitosan solution to become electrospinnable. The chitosan-based scaffolds share the chemical structure and characteristics of glycosaminoglycans, which are important structural components of the cartilage extracellular matrix. Electrospinning produced nanofibrils of ∼100 nm thickness that are closely mimicking the size of collagen fibrils in human AC. The polymer scaffolds were stabilized in physiological conditions and their stiffness was tuned by introducing the biocompatible natural crosslinker genipin. We produced scaffolds that were crosslinked with 1.0% genipin to obtain values of stiffness that were in between the stiffness of the superficial zone human AC of 600 ± 150 kPa and deep zone AC of 1854 ± 483 kPa, whereas the stiffness of 1.5% genipin crosslinked scaffold was similar to the stiffness of deep zone AC. The scaffolds were degradable, which was indicated by changes in the fibril structure and a decrease in the scaffold stiffness after seven months. Histological and immunohistochemical analysis after three weeks of culture with human articular chondrocytes (HACs) showed a cell viability of over 90% on the scaffolds and new extracellular matrix deposited on the scaffolds.

    Poly lactic-co-glycolic acid (PLGA) nanoparticles are intensively studied nanocarriers in drug delivery because of their biodegradability and biochemical characteristics. Polyethylene glycol (PEG) coating for nanocarriers gives them long circulation time in blood and makes them invisible to the reticuloendothelial system. Breast cancer cells have greater uptake of hyaluronic acid compared to normal cells as it binds to their overexpressed CD44 receptors. Since hypoxia plays an important role in cancer metastasis; we formulated PEG-PLGA nanoparticles coated with hyaluronic acid as targeted delivery system for doxorubicin (DOX) using nanoprecipitation method, and characterized them for chemical composition, size, surface charge, shape, and encapsulation efficiency. Then we tested them in vitro on hypoxia-optimized metastatic breast cancer cells. The nanoparticles were spherical with an average size of about 106 ± 53 nm, a negative surface charge (-15 ± 3 mV), and high encapsulation efficiency (73.3 ± 4.1%). In vitro investigation with hypoxia-elevated CD44 MDA-MB-231 cells showed that hyaluronic acid-targeted nanoparticles maintained their efficacy despite hypoxia-induced drug resistance unlike free DOX and nontargeted nanoparticles. In conclusion, this study revealed a simple third generation nanoparticle formulation for targeted treatment of hypoxia-induced drug resistance in breast cancer metastatic cells. Further, optimization is needed including In vivo efficacy and nanoparticle-specific pharmacokinetic studies.

    An innovative nano-base polymer that scavenges radicals and reactive oxygen species exhibits potential antibacterial properties, which are crucial in the biomedical field, particularly in reducing nosocomial infections. However, the safety of this nano-based polymer, which has direct contact with the human system, has not been fully understood. The present study investigated the cytocompatibility and hemocompatibility responses of linear low-density polyethylene polymer (LLDPE) embedded with difference ratios of heterogeneous TiO2/ZnO nanocomposites. Exposure of the blood and fibroblast cells to LLDPE/100Z and LLDPE/25T75Z/10% nanocomposite films for 48 and 72 h decreased their viability by less than 40%, compared with LLDPE, LLDPE/100T and LLDPE/25T75Z/5% nanocomposite films. It also presented possible cellular damage and cytotoxicity, which was supported by the findings from the significant release of extracellular lactate dehydrogenase profiles and cell survival assay Further observation using an electron microscope revealed that LLDPE films with heterogeneous 25T75Z/5% promoted cell adhesion. Moreover, no hemolysis was detected in all ratios of heterogeneous TiO2/ZnO nanocomposite in LLDPE film as it was less than 0.2%, suggesting that these materials were hemocompatible. This study on LLDPE film with heterogeneous TiO2/ZnO nanocomposites demonstrated favorable biocompatible properties that were significant for advanced biomedical polymer application in a hospital setting.

    Gallic acid (GA) is a hydrophilic polyphenol which is noteworthy for strong antioxidant capacity. The drawbacks of conventional extraction approaches such as time-consuming and high processing cost are often viewed as a hurdle to extract GA from plant sources in industrial scale. Aqueous two-phase system (ATPS) is a separation approach which can be employed as an alternative to the conventional approaches. The partition behaviour of GA in an alcohol/salt ATPS was investigated in this study to aid the development of industrial scale ATPS to extract GA from natural sources. The separation of GA was characterized by determining the types of alcohol and salt, phase composition, sample load, pH of the system and addition of adjuvants applied in the alcohol/salt ATPS construction. The hydrophilic GA was targeted to the salt-rich phase of the alcohol/salt ATPS with a partition coefficient (KGA) of 25.00 ± 0.00. The optimum condition of ATPS for the maximum partition of GA was achieved in ATPS comprised of 24% (w/w) 1-propanol and 22% (w/w) phosphate salt at pH 8 with 5% (w/w) of 1 mg/mL sample loading and 2% (w/w) NaCl addition. The findings suggest that ATPS can be applied for separation of GA from various natural sources.

    In this work, mesoporous TiO2-modified ZnO quantum dots (QDs) were immobilised on a linear low-density polyethylene (LLDPE) polymer using a solution casting method for the photodegradation of tetracycline (TC) antibiotics under fluorescent light irradiation. Various spectroscopic and microscopic techniques were used to investigate the physicochemical properties of the floating hybrid polymer film catalyst (8%-ZT@LLDPE). The highest removal (89.5%) of TC (40 mg/L) was achieved within 90 min at pH 9 due to enhanced water uptake by the LDDPE film and the surface roughness of the hybrid film. The formation of heterojunctions increased the separation of photogenerated electron-hole pairs. The QDs size-dependent quantum confinement effect leads to the displacement of the conduction band potential of ZnO QDs to more negative energy values than TiO2. The displacement generates more reactive species with higher oxidation ability. The highly stable film photocatalyst can be separated easily and can be repeatedly used up to 8 cycles without significant loss in the photocatalytic ability. The scavenging test indicates that the main species responsible for the photodegradation was O2●-. The proposed photodegradation mechanism of TC was demonstrated in further detail based on the intermediates detected by LC-time-of-flight/mass spectrometry (LC/TOF-MS).

    This work investigates a new interrogation method of a fiber Bragg grating (FBG) sensor based on longer and shorter wavelengths to distinguish between transversal forces and temperature variations. Calibration experiments were carried out to examine the sensor's repeatability in response to the transversal forces and temperature changes. An automated calibration system was developed for the sensor's characterization, calibration, and repeatability testing. Experimental results showed that the FBG sensor can provide sensor repeatability of 13.21 pm and 17.015 pm for longer and shorter wavelengths, respectively. The obtained calibration coefficients expressed in the linear model using the matrix enabled the sensor to provide accurate predictions for both measurements. Analysis of the calibration and experiment results implied improvements for future work. Overall, the new interrogation method demonstrated the potential to employ the FBG sensing technique where discrimination between two/three measurands is needed.

    Membrane distillation (MD) is an attractive technology for the separation of highly saline water used with a polytetrafluoroethylene (PTFE) hollow fiber (HF) membrane. A hydrophobic coating of low-density polyethylene (LDPE) coats the outer surface of the PTFE membrane to resolve membrane wetting as well as increase membrane permeability flux and salt rejection, a critical problem regarding the MD process. LDPE concentrations in coating solution have been studied and optimized. Consequently, the LDPE layer altered membrane morphology by forming a fine nanostructure on the membrane surface that created a hydrophobic layer, a high roughness of membrane, and a uniform LDPE network. The membrane coated with different concentrations of LDPE exhibited high water contact angles of 135.14 ± 0.24 and 138.08 ± 0.01° for membranes M-3 and M-4, respectively, compared to the pristine membrane. In addition, the liquid entry pressure values of LDPE-incorporated PTFE HF membranes (M-1 to M-5) were higher than that of the uncoated membrane (M-0) with a small decrease in the percentage of porosity. The M-3 and M-4 membranes demonstrated higher flux values of 4.12 and 3.3 L m-2 h-1 at 70 °C, respectively. On the other hand, the water permeation flux of 1.95 L m-2 h-1 for M-5 further decreased when LDPE concentration is increased.

    Polymer composites are favorite materials for sensing applications due to their low cost and easy fabrication. In the current study, composite nanofibers consisting of polyethylene oxide (PEO), oxidized multi-walled carbon nanotubes (MWCNT) and copper oxide (CuO) nanoparticles with 1% and 3% of fillers (i.e., PEO-CuO-MWCNT: 1%, and PEO-CuO-MWCNT: 3%) were successfully developed through electrospinning for humidity sensing applications. The composite nanofibers were characterized by FTIR, XRD, SEM and EDX analysis. Firstly, they were loaded on an interdigitated electrode (IDE), and then the humidity sensing efficiency was investigated through a digital LCR meter (E4980) at different frequencies (100 Hz-1 MHz), as well as the percentage of relative humidity (RH). The results indicated that the composite nanofibers containing 1% and 3% MWCNT, combined with CuO in PEO polymer matrix, showed potent resistive and capacitive response along with high sensitivity to humidity at room temperature in an RH range of 30-90%. More specifically, the PEO-CuO-MWCNT: 1% nanocomposite displayed a resistive rapid response time within 3 s and a long recovery time of 22 s, while the PEO-CuO-MWCNT: 3% one exhibited 20 s and 11 s between the same RH range, respectively.

    Vitexin of Ficus deltoidea exhibits intestinal α-glucosidase inhibitory and blood glucose lowering effects. This study designs oral intestinal-specific alginate nanoparticulate system of vitexin. Nanospray-dried alginate, alginate/stearic acid and alginate-C18 conjugate nanoparticles were prepared. Stearic acid was adopted to hydrophobize the matrix and minimize premature vitexin release in stomach, whereas C-18 conjugate as immobilized fatty acid to sustain hydrophobic effect and drug release. Nanoparticles were compacted with polyethylene glycol (PEG 3000, 10,000 and 20,000). The physicochemical, drug release, in vivo blood glucose lowering and intestinal vitexin content of nanoparticles and compact were determined. Hydrophobization of alginate nanoparticles promoted premature vitexin release. Compaction of nanoparticles with PEG minimized vitexin release in the stomach, with stearic acid loaded nanoparticles exhibiting a higher vitexin release in the intestine. The introduction of stearic acid reduced vitexin-alginate interaction, conferred alginate-stearic acid mismatch, and dispersive stearic acid-induced particle breakdown with intestinal vitexin release. Use of PEG 10,000 in compaction brought about PEG-nanoparticles interaction that negated initial vitexin release. The PEG dissolution in intestinal phase subsequently enabled particle breakdown and vitexin release. The PEG compacted nanoparticles exhibited oral intestinal-specific vitexin release, with positive blood glucose lowering and enhanced intestinal vitexin content in vivo.

    The main purpose of this research is to design a high-fatigue performance hoop wrapped compressed natural gas (CNG) composite cylinder. To this end, an optimization algorithm was presented as a combination of finite element simulation (FES) and response surface analysis (RSA). The geometrical model was prepared as a variable wall-thickness following the experimental measurements. Next, transient dynamic analysis was performed subjected to the refueling process, including the minimum and maximum internal pressures of 20 and 200 bar, respectively. The time histories of stress tensor components were extracted in the critical region. Furthermore, RSA was utilized to investigate the interaction effects of various polymer composite shell manufacturing process parameters (thickness and fiber angle) on the fatigue life of polymer composite CNG pressure tank (type-4). In the optimization procedure, four parameters including wall-thickness of the composite shell in three different sections of the CNG tank and fiber angle were considered as input variables. In addition, the maximum principal stress of the component was considered as the objective function. Eventually, the fatigue life of the polymer composite tank was calculated using stress-based failure criterion. The results indicated that the proposed new design (applying optimal parameters) leads to improve the fatigue life of the polymer composite tank with polyethylene liner about 2.4 times in comparison with the initial design.

    Aim: To synthesize niosomes co-encapsulating gemcitabine (GEM) and tocotrienols, and physicochemically characterize and evaluate the antipancreatic effects of the nanoformulation on Panc 10.05, SW 1990, AsPC-1 and BxPC-3 cells. Materials & methods: Niosomes-entrapping GEM and tocotrienols composed of Span 60, cholesterol and D-α-tocopheryl polyethylene glycol 1000 succinate were produced by Handjani-Vila and film hydration methods. Results: The film hydration produced vesicles measuring 161.9 ± 0.5 nm, approximately 50% smaller in size than Handjani-Vila method, with maximum entrapment efficiencies of 20.07 ± 0.22% for GEM and 34.52 ± 0.10% for tocotrienols. In Panc 10.05 cells, GEM's antiproliferative effect was enhanced 2.78-fold in combination with tocotrienols. Niosomes produced a significant ninefold enhancement in cytotoxicity of the combination, supported by significantly higher cellular uptake of GEM in the cells. Conclusion: This study is a proof of concept on the synthesis of dual-drug niosomes and their efficacy on pancreatic cancer cells in vitro.

    Frictional and nanomechanical properties of nanostructured polymer surfaces are important to their technological and biomedical applications. In this work, poly(ethylene terephthalate) (PET) surfaces with a periodic distribution of well-defined nanopillars were fabricated through an anodization/embossing process. The apparent surface energy of the nanopillared surfaces was evaluated using the Fowkes acid-base approach, and the surface morphology was characterized using scanning electron microscope (SEM) and atomic force microscope (AFM). The normal and lateral forces between a silica microparticle and these surfaces were quantified using colloidal probe atomic force microscopy (CP-AFM). The friction-load relationship followed Amonton's first law, and the friction coefficient appeared to scale linearly with the nanopillar height. Furthermore, all the nanopillared surfaces showed pronounced frictional instabilities compared to the smooth sliding friction loop on the flat control. Performing the stick-slip amplitude coefficient (SSAC) analysis, we found a correlation between the frictional instabilities and the nanopillars density, pull-off force and work of adhesion. We have summarised the dependence of the nanotribological properties on such nanopillared surfaces on five relevant parameters, i.e. pull-off force fp, Amontons' friction coefficient μ, RMS roughness Rq, stick-slip amplitude friction coefficient SSAC, and work of adhesion between the substrate and water Wadh in a radar chart. Whilst demonstrating the complexity of the frictional behaviour of nanopillared polymer surfaces, our results show that analyses of multiparametric nanotribological properties of nanostructured surfaces should go beyond classic Amontons' laws, with the SSAC more representative of the frictional properties compared to the friction coefficient.

    Carboxymethyl starch (CMS) was produced from sago starch via carboxymethylation. The CMS with different degree of substitution (DS) ranges from 0.4 to 0.8 were mixed with polyethylene glycol (PEG) of different molecular weight and distilled water and the hydrogel was cured by electron beam irradiation with doses ranging from 25 to 35 kGy. The results revealed that CMS-PEG hydrogels with DS 0.4 give the optimum gel content when radiated at 30 kGy and with PEG 600. Thermogravimetric analysis (TGA) revealed that there are two phases exist in CMS with DS 0.4 in contrast to the three steps decomposition occurs in DS 0.6 and 0.8. It shows that the CMS with DS 0.4 is more thermally stable. Surface morphology revealed crosslinking among the blends when subjected into the radiation dose. The study shows both radiation and PEG addition improved most of the properties of CMS irrespective of the DS value.

    Enzyme immobilization has been known to be one of the methods to improve the stability and reusability of enzyme. In this study, a strategy to optimize laccase immobilization on polyethylene terephthalate grafted with maleic anhydride electrospun nanofiber mat (PET-g-MAH ENM) was developed. The development involves the screening and optimization processes of the crucial factors that influence the immobilization yield such as enzyme concentration, pH values, covalent bonding (CV) time, CV temperature, crosslinking (CL) time, CL temperature and glutaraldehyde concentration using two-level factorial design and Box-Behnken design (BBD), respectively. It was found that laccase concentration, pH values and glutaraldehyde concentration play important role in enhancing the immobilization yield of laccase on PET-g-MAH ENM in the screening process. Subsequently, the optimization result showed at 0.28 mg/ml laccase concentration, pH 3 and 0.45% (v/v) glutaraldehyde concentrations gave the highest immobilization yield at 87.64% which was 81.2% increment from the immobilization yield before optimization. Under the optimum condition, the immobilized laccase was able to oxidize 2, 2-azino-bis 3-ethylbenzothiazoline-6- sulfonic acid (ABTS) in a broad range of pH (pH 3-6) and temperature (20- 70 °C). Meanwhile, the kinetic parameters for Km and Vmax were 1.331 mM and 0.041 mM/min, respectively. It was concluded that the optimization of immobilized laccase on PET-g-MAH ENM enhance the performance of this biocatalyst.

    Microplastics receive global attention due to its strong presence in marine and freshwater organisms. Yet, there are few studies on estuarine organisms. This baseline study evaluates the abundance and distribution of microplastics in the gastropods of a tropical estuary in Selangor, Malaysia. The abundance of microplastics ranged from 0.50 to 1.75 particles/g or from 0.25 to 0.88 particles/individual. The variation in microplastic distribution between the upper and lower estuary indicates that the microplastics originated from the urbanised area of the Klang River estuary. Microplastic sizes varied from 30 to 1850 μm, with the majority being between 300 and 1000 μm (57%). Characteristics of microplastics were dominant for fibres (91%) with black colour (50%). Polyethylene-propylene-diene (PE-PDM) and polyester were the main polymer materials. Assessing the contamination of gastropods by microplastics provides insight into the possibility of utilising gastropods as bioindicators that could be used for monitoring and baseline studies.

    Drought is one of the severe environmental stresses threatening agriculture around the globe. Nitric oxide plays diverse roles in plant growth and defensive responses. Despite a few studies supporting the role of nitric oxide in plants under drought responses, little is known about its pivotal molecular amendment in the regulation of stress signaling. In this study, a label-free nano-liquid chromatography-mass spectrometry approach was used to determine the effects of sodium nitroprusside (SNP) on polyethylene glycol (PEG)-induced osmotic stress in banana roots. Plant treatment with SNP improved plant growth and reduced the percentage of yellow leaves. A total of 30 and 90 proteins were differentially identified in PEG+SNP against PEG and PEG+SNP against the control, respectively. The majority of proteins differing between them were related to carbohydrate and energy metabolisms. Antioxidant enzyme activities, such as superoxide dismutase and ascorbate peroxidase, decreased in SNP-treated banana roots compared to PEG-treated banana. These results suggest that the nitric oxide-induced osmotic stress tolerance could be associated with improved carbohydrate and energy metabolism capability in higher plants.
