Processing of soybeans to other products and consumption of soy products is increasing worldwide mainly due to acclaimed health benefits. Processing can alter soybean sensory appeal, nutritive value and potentially affect consumer health. Rhizopus oligosporus was used to ferment soybean for 3 days. The tempeh flour (TF) was produced form tempeh while defatted tempeh flour (DTF) was then produced from TF by immersing in hexane solvent while soy protein isolate (SPI) was prepared from DTF by using alkali and acid followed by neutralization treatment. In this study, nutritional properties and amino acid content of tempeh, TF, DTF and SPI were determined. Therefore, the objective of this study is was to evaluate the effect of each treatment on the chemical composition and amino acid content for all the samples. The results showed that the nutritional properties (total ash, moisture, crude fat, total carbohydrate and crude fibre) were reduced significantly (p < 0.05) except for protein content. Protein content was significantly (p < 0.05) increased by 50.5% in SPI. For amino acid content, the results obtained showed that SPI contain highest amount of essential and non-essential amino acid followed by DTF, Tempeh and TF. Glutamic acid was found to be the highest amino acid component in all samples. The evaluation from the results showed that SPI can be considered as potential functional food ingredients.

The global trend in increasing plant-based protein diets due to health and ideological reasons, has created an increased demand for food legumes that exceeds current production. To meet this demand, it is timely to reduce relying solely on soybean, and explore the potential of the underutilised legumes that are cultivated regionally. Underutilised legumes are rich in protein, carbohydrates and other nutrients that are essential for consumer. However, relatively little is known about their anti-nutritional properties and processing methods. Anti-nutritional factors (ANFs) such as enzyme inhibitors are prevalent in legumes and may interfere with digestibility and nutrient absorption. Nevertheless, an optimised food processing method will overcome this challenge and warrant a safe inclusion of legume in plant-based protein diets. Hence current study aimed to optimise the food processing methods (soaking, wet heating, autoclaving and freezing) and evaluate their efficiency in eliminating the enzyme inhibitors [trypsin, chymotrypsin (CIA) and α-amylase (AIA) inhibitors] present in seven underutilised legumes. Current study showed that autoclaving at 121 °C for 15 min reduced the AIA in all underutilised legumes tested. The AIA and CIA of bambara groundnut were successfully inactivated by wet heating at 50 °C for 60 min, and by autoclaving at 121 °C for 15 min. While the CIA of chickpea was successfully inactivated by freezing at - 80 °C for 24 h.

At least nine Colletotrichum species, particularly Colletotrichum truncatum, have been recorded on legumes worldwide (1). In June 2010, samples of chickpea leaflets showing leaf spot disease symptoms were collected from experimental farms in Ladang Dua, Selangor state of Malaysia. Tan lesions with darker brown borders were observed on leaflets and were associated with premature leaf drop. Stem lesions initially appeared on the lower parts of stems and later progressed higher in the plant. Lesions often girdled the stem and caused severe dieback. Abundant acervuli developed in the lesions visible as black dots. Foliar lesions were removed, surface sterilized in 1% sodium hypochlorite for 2 min, rinsed twice with distilled water, dried on sterilized tissue paper, plated on PDA plates, and incubated at 25°C (3). Three isolates of the fungus were obtained and identified as C. truncatum on the basis of morphological characteristics (2). The isolates were deposited in the University Putra of Malaysia Culture Collection (UPMCC). Colony characteristics on PDA varied from greyish white to dark in color and exhibited mycelial growth with sparse acervuli. The isolates produced both sclerotia and setae in culture. Conidia (mean ± SD = 22 ± 0.83 × 3.6 ± 0.08 μm, L/W ratio = 6.1) produced in acervuli were falcate, hyaline, and aseptate, with tapering towards the acute and greatly curved apex. The conidial mass color varied from pale buff to saffron. Isolates produced simple to slightly lobed, mainly short clavate appressoria (mean ± SD = 9.60 ± 0.36 × 6.67 ± 0.29 μm, L/W ratio = 1.45). Amplification and sequence analysis of coding and none-coding regions of the ITS-rDNA (GenBank Accession JX971160), actin (JX975392), β-tubulin (KC109495), histone (KC109535), chitin synthase (KC109575), and glyceraldehyde-3-phosphate dehydrogenase (KC109615) obtained from the representative isolate, CTM37, aligned with deposited sequences from GenBank and revealed 99 to 100% sequence identity with C. truncatum strains (AJ301945, KC110827, GQ849442, GU228081, GU228359, and HM131501 from GenBank). Isolate CTM37 was used to test pathogenicity in the greenhouse. Five chickpea seeds of cultivar ILC-1929 were sown per pot in four replications. Ten days after seedling emergence, plants were inoculated with a spore suspension (concentration = 106 conidia ml-1) and check pots were sprayed with distilled water. After inoculation, the plants were covered with plastic bags for 48 h and kept at 28 to 33°C and >90% RH. After incubation, the plastic bags were removed and the plants were placed on greenhouse benches and monitored daily for symptom development (3). One week after inoculation, typical anthracnose symptoms developed on the leaves and stems of inoculated plants including acervuli formation, but not on the checks. A fungus with the same colony and conidial morphology as CTM37 was recovered from the lesions on the inoculated plants. The experiment was repeated twice. The ability to accurately diagnose Colletotrichum species is vital for the implementation of effective disease control and quarantine measures. We believe this is the first report of C. truncatum causing anthracnose on chickpea in Malaysia. References: (1) B. D. Gossen et al. Can. J. Plant Pathol. 31:65, 2009. (2) B. C. Sutton. Public perceptions, understanding and acceptance of modern biotechnology can both promote and hamper their commercial introduction and adoption. Various studies have shown that consumer acceptance of modern biotechnology tend to be conditional and dependent on several factors. Public perceptions of biotechnology have received extensive attention in recent years in most Western countries such as Europe, USA and Canada but there have been limited similar surveys in developing countries. Most of the earlier studies used uni-dimensional or bi-dimensional instrument with multi-items or the most is four dimensions with single item. In this study, public attitude towards genetically modified (GM) soybean that is already available in the Malaysian market. A survey was carried out on 577 general public respondents in the Klang Valley region. In order to detect the structure of attitude amongst the expert group in the Klang Valley region, structural equation modeling (SEM) using AMOS version 5.1 was carried out. Result of the survey has confirmed that attitude towards complex issues such as biotechnology should be seen as multi-faceted/ multidimensional process. The most important factors predicting encouragement of GM soybean are the specific application-linked perceptions about the benefits and acceptance of risk while moral concern, risk and familiarity are significant predictors of intermediate factors. Researchers, policy makers and industries interested in developing and marketing GM products in Malaysia should consider the various factors mentioned in this in order to gain public approval.

Yield performance in wheat (Triticum aestivum) was compared under crop residue, tillage system and nitrogen rate treatments in cereal based cropping system. The experiments were conducted at Agronomy Research Farm, The University of Agriculture, Peshawar, in 2009-2010 and 2010-2011. Chopped crop residue on dry matter basis (5 t ha1-) of legume (Vigna unguicuata, var. Ebney) and cereal (Zea mays, var. Azam) was applied in main plots with no residue treatments and plowed with Mould Board (MB) and Cultivator as deep and shallow treatments, respectively. A month after the crop residue and tillage system treatments, field was uniformly plowed with cultivator and wheat was sown with drill in rows 25 cm apart in the month of November on both years. Both P2O55 and K2O (80 and 40 kg ha , respectively) were applied uniformly to all fields before sowing. Nitrogen as subplot treatment (0, 40, 80, 120 and 160 kg ha-1) was applied in two splits, half at 15 and the other half at 45 days after sowing with uniform cultural practices for crop growth and development. Compared to year 1, crop of year 2 showed better phenology with extended life cycle (LC). On two years average across tillage and N treatments, biological yield did not change (p<0.05) under the residue but did report lower at no-residue treatment. Nonetheless, grain yield showed a significant (p<0.05) change with the highest in legume followed by cereal and the lowest in no-residue treatments. A non-significant tiller number and significant variations in grain weight and spike m-2 were observed that influenced the grain and biological yield differently. Deep than shallow tillage resulted in better traits, which returned better biomass and grain yield. Nitrogen application from control to every increment showed a significant (p<0.05) improvement in all observations contributing in yield. The study confirms the significance of legume vs. cereal over no-crop residue incorporated through deep tillage system with optimum N (120 kg ha-1) in cereal-based cropping system for sustainable performance to sustain soil C:N for future production.

This study examined the tunneling activity and the behaviour of two subterranean termites, Globitermes sulphurues and Coptotermes gestroi treated with four plant extracts, namely Pyllanthus niruri, Azadirachta indica, Leucaena leucocephala and Andrographis paniculata. All plants were extracted with three different solvents (methanol, hexane and water) and were diluted into three concentrations (500, 5,000 and 10,000 ppm). A group of 50 workers and 2 soldiers were tested and observed daily during the exposure in order to determine their survivorship and behaviour. Both sections were scored to determine their tunneling activities after seven days. There was a significant difference of tunneling activities of C. gestroi on sand treated with plant extracts (X2(2) = 31.790, p < 0.0001) with a mean rank of 8.50 for methanolic extracts and 32.50 for both hexane and water extracts. Meanwhile, no significant difference was observed on tunneling activity of G. sulphureus on treated sand (X2(2) = 2.200, p = 0.333) with a mean rank of 20.72 for methanolic extracts, 26.31 for water extracts and 26.47 for hexane extracts. Results showed that plants extracted with methanol demonstrated strong repellent properties with 0 tunneling activity on the treated sand and low survivorship of both termites. Moreover, both termites did not survive (0%) when they were treated with methanolic extracts at 10,000 ppm. They also displayed a different behaviour post-exposure such as avoidance, gradually losing the ability to walk and low feeding consumption. These results indicate that there is a strong termiticidal activity of plants extracted with methanol against C. gestroi and G. sulphureus.

soflavonoids are the main compound in White Kwao Krua (Pueraria mirifica), which is an effective folk medicinal plant endemic to Thailand. It has been widely used for improving human physical and treating diseases. There are substances with estrogenic activities have been isolated from P. mirifica, such as puerarin, daidzein and genistein. Isoflavone synthase (IFS) is one of the key enzymes in Leguminous plants to convert liquiritigenin, liquiritigenin C-glucoside and naringenin chalcone to isoflavonoids. The aim of this research was to enhance the production of isoflavonoids by metabolic engineering. Transgenic plants were constructed by introducing P450 gene (EgP450) which is similar to IFS from oil palm (Elaeis guineensis), into P. mirifica by a biolistic method. After the transgenic plants had proved successfully, isoflavonoids of each group plants were determined by HPLC. The contents of daidzein and genistein in transgenic plants were higher than the control plants

Phytase activity and growth of anaerobic rumen bacterium, Mitsuokella jalaludinii were investigated by semi-solid
state fermentation. Carbon source (rice bran, yam and cassava), nitrogen sources (soya bean, offal meal, fish meal and
feather meal) and growth factors (hemin, L-cysteine hydrochloride and minerals) were evaluated in a one-factor-at-atime
approach. Rice bran and fish meal produced better growth and phytase enzyme activity. The removal of L-cysteine
hydrochloride and minerals significantly decreased (p<0.05) phytase activity from 1178.72 U to 446.99 U and 902.54
U, respectively. The response surface methods (RSM) was conducted to optimize the phytase production and the results
showed the combination of 7.7% of rice bran and 3.7% of fish meal in semi-solid state fermentation gave the highest
phytase activity. Maximum phytase production and optimum growth of bacteria were detected at 12 h incubation in both
MF medium (control) and agro-medium. In this agro-medium, M. jalaludinii produced 2.5 fold higher phytase activity
    compared to MF medium.

The last decade has witnessed dramatic changes in global food consumption patterns mainly because of population growth and economic development. Food substitutions for healthier eating, such as swapping regular servings of meat for protein-rich crops, is an emerging diet trend that may shape the future of food systems and the environment worldwide. To meet the erratic consumer demand in a rapidly changing world where resources become increasingly scarce due largely to anthropogenic activity, the need to develop crops that benefit both human health and the environment has become urgent. Legumes are often considered to be affordable plant-based sources of dietary proteins. Growing legumes provides significant benefits to cropping systems and the environment because of their natural ability to perform symbiotic nitrogen fixation, which enhances both soil fertility and water-use efficiency. In recent years, the focus in legume research has seen a transition from merely improving economically important species such as soybeans to increasingly turning attention to some promising underutilized species whose genetic resources hold the potential to address global challenges such as food security and climate change. Pulse crops have gained in popularity as an affordable source of food or feed; in fact, the United Nations designated 2016 as the International Year of Pulses, proclaiming their critical role in enhancing global food security. Given that many studies have been conducted on numerous underutilized pulse crops across the world, we provide a systematic review of the related literature to identify gaps and opportunities in pulse crop genetics research. We then discuss plausible strategies for developing and using pulse crops to strengthen food and nutrition security in the face of climate and anthropogenic changes.

Introduction:Tropical herbs are used as a medicine long before the introduction of modern medicine, especially by traditional healers. However, its’ scientific benefits are not widely investigated. There is a belief that the use of tradi-tional herbs potentially causing health deterioration due to its unknown mechanism of actions and interactions, and the nature of its unregulated processing as a health supplement. Parkia speciosa (local: petai) and Pithecellobium jir-inga (local: jering) are two tropical herbs from the Leguminosae family that are widely used raw as salads with many perceived health benefits and harms. We reviewed both species, looking into scientific pieces of evidence supporting the belief. Methods: The databases of MEDLINE, PubMed, EBSCOhost, and Google Scholars were systematically searched for both in vitro and in vivo effects of both species with no language restriction. Data were extracted using standardised fashioned. Results:P speciosa has pharmacological properties including antioxidant and anti-inflam-mation. It has shown to have antibacterial properties and glucose-lowering activity. P jiringa contains known an-tioxidant metabolites such as flavonoids, phenolics and terpenoids. It also contains antiangiogenic properties with potential use in retinopathy, cancer, and Rheumatoid arthritis. However, both herbs have potentially harmful effects which include liver injuries. Large consumption of P jiringa leads to acute kidney injury. Conclusion: Consumption of both P speciosa and P jiringa has medicinal properties as well as health harm. If taken as a supplement, rigorous clinical trials are needed to delineate benefit vs harm effect of both herbs.

Understanding dietary diversity is a fundamental task in the study of stump-tailed macaque, Macaca arctoides in its natural habitat. However, direct feeding observation and morphological identification using fecal samples are not effective and nearly impossible to obtain in natural habitats because this species is sensitive to human presence. As ecological methods are challenging and time-consuming, DNA metabarcoding offers a more powerful assessment of the diet. We used a chloroplast tRNL DNA metabarcoding approach to identify the diversity of plants consumed by free-ranging M. arctoides in the Malaysia-Thailand border region located in Perlis State Park, Peninsular Malaysia. DNA was extracted from three fecal samples, and chloroplast tRNL DNA was amplified and sequenced using the Illumina MiniSeq platform. Sequences were analyzed using the CLC Genomic Workbench software. A total of 145 plant species from 46 families were successfully identified as being consumed by M. arctoides. The most abundant species were yellow saraca, Saraca thaipingensis (11.70%), common fig, Ficus carica (9.33%), aramata, Clathrotropis brachypetala (5.90%), sea fig, Ficus superba (5.44%), and envireira, Malmea dielsiana (1.70%). However, Clathrotropis and Malmea are not considered Malaysian trees because of limited data available from Malaysian plant DNA. Our study is the first to identify plant taxa up to the species level consumed by stump-tailed macaques based on a DNA metabarcoding approach. This result provides an important understanding on diet of wild M. arctoides that only reside in Perlis State Park, Malaysia.

Parkia speciosa is a food plant that grows indigenously in Southeast Asia. A great deal of interest has been paid to this plant due to its traditional uses in the treatment of several diseases. The pods contain many beneficial secondary metabolites with potential applications in medicine and cosmetics. However, studies on their phytochemical properties are still lacking. Therefore, the present study was undertaken to profile the bioactive compounds of P. speciosa pods collected from six different regions of Malaysia through ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC-QTOF-MS) and α-glucosidase inhibitory potential. This study applied metabolomics to elucidate the differences between P. speciosa populations found naturally in the different locations and to characterize potential α-glucosidase inhibitors from P. speciosa pods. P. speciosa collected from different regions of Malaysia showed good α-glucosidase inhibitory activity, with a median inhibitory concentration (IC50) of 0.45-0.76 μg/mL. The samples from the northern and northeastern parts of Peninsular Malaysia showed the highest activity. Using UHPLC-QTOF-MS/MS analysis, 25 metabolites were identified in the pods of P. speciosa. The findings unveiled that the pods of P. speciosa collected from different locations exhibit different levels of α-glucosidase inhibitory activity. The pods are a natural source of potent antidiabetic bioactive compounds.

Milk is a physiological fluid which has high nutritional value and soybean has strong antioxidant characteristics which is believed to inhibit carcinogenesis. The objective of this study was to investigate the effects of administration of soybean and goat's milk on hepatocarcinogenesis in rats (fed with diethylnitrosamine; DEN and acetylaminofluorene; AAF) by determining the activities of plasma gamma-glutamyl transpeptidase (GGT) and alkaline phosphatase (ALP). Thirty-six rats from the species Sprague-Dawley were divided into 6 groups : control, DEN/AAF, soybean, DEN/AAF with soybean treatment, goat's milk and DEN/AAF with goat's milk treatment. Soybean and goat's milk administrations were given 5 ml/day. The rats were sacrificed after 8 weeks and the blood was collected. Treatment with DEN/AAF caused an increase in ALP and GGT levels and a decrease in weight significantly (p<0.05). ALP and GGT activities decreased significantly after administration of soybean and goat's milk (p<0.05). Administration of goat's milk and soybean alone did not cause any changes in the enzyme activities. Comparison between the effect of soybean and goat's milk in reducing the enzyme activities (ALP and GGT) did not give significant values (p>0.05). However, a decrease in weight was observed in the rats given soybean as well as goat's milk. The results obtained suggested that soybean and goat's milk may work as anti cancer agents in hepatocarcinogenesis although further studies are required to further elucidate this aspect.

The Clitoria ternatea (Fabaceae) root, seed, and leaf are commonly used in Ayurvedic medicine in Malaysia and Indonesia. The methanol leaf extracts of C. ternatea was tested for toxicity by means of brine shrimp lethality test and acute oral toxicity assay. In the brine shrimp lethality test, the leaf extract were non-toxic or showed weak lethality (LC50 > 1 mg/ml) at the 6 h, 12 h and 24 h incubation period. Nevertheless, at the 48 h incubation time, the leaf extract exhibited moderate toxicity activity with LC50 values of 0.49 mg/ml. In the acute toxicity study using mice, the median lethal dose (LD50) of the extract was found greater than 2000 mg/kg, and we found no pathological changes by means of macroscopic examination of tissues of mice treated with the extract. We conclude that the C. ternatea leaf extract is not toxic in mice and brine shrimp.

Starch blend films made from sago and mung bean were prepared by casting with glycerol as the plasticizer and subsequently exposed to ultraviolet (UV) irradiation for 2 h. The films were characterized by thickness, moisture sorption isotherms, X-ray diffraction and Fourier transform infrared (FTIR) spectroscopy. All films produced were colorless while incorporation of glycerol resulted in more flexible and manageable films. Moisture sorption isotherms for all films showed sigmoidal shape and the control films showed slightly higher curve than treated films. While for X-ray analysis, the control and treated films for all formulations showed similar pattern, however for treated films showed more crystalline character. UV radiation showed affect on X-ray diffraction and sorption isotherms; however the UV radiation did not affect the spectra pattern of FTIR.

Proximate content and fatty acid composition of germinated and non-germinated legumes (kidney, mung, soy bean and peanut) and rice varieties (red, black, Barrio, brown and milled) were evaluated. In germinated samples, moisture content increased significantly while carbohydrate, protein and fat were decreased significantly. Total dietary fibre was increased in germinated samples except germinated kidney and mung bean. Germination also increased saturated fatty acids (SFA) in legumes, black, red and brown rice. Monounsaturated fatty acids (MUFA) decreased in all samples except germinated kidney, soy and Barrio rice. Polyunsaturated fatty acids (PUFA) increased in some germinated samples (mung bean, peanut, red, brown, Barrio and white rice) but decreased in other legume and rice samples. Generally, palmitic acid increased while stearic, oleic and linoleic acids decreased after germination. Overall, the proximate content and fatty acids of legume and rice varieties changed after germination and may be used as alternate resources for individuals with lifestyle diseases.

Legume is a plant in the family of Fabaceae (or Leguminosae) that is cultivated and consumed
throughout the world. Legume’s role in human health appears to be limited because of several
limiting factors such as low protein and starch digestibility, poor mineral bioavailability and
high antinutritional factors. Germination is defined as a process that occurs during seed growth
that starts with uptake of water until the emergence of radicle through the surrounding structure.

Bambara groundnut (Vigna subterranea (L.) Verdc.) is an underutilised legume crop, which has long been recognised as a protein-rich and drought-tolerant crop, used extensively in Sub-Saharan Africa. The aim of the study was to identify quantitative trait loci (QTL) involved in agronomic and drought-related traits using an expression marker-based genetic map based on major crop resources developed in soybean. The gene expression markers (GEMs) were generated at the (unmasked) probe-pair level after cross-hybridisation of bambara groundnut leaf RNA to the Affymetrix Soybean Genome GeneChip. A total of 753 markers grouped at an LOD (Logarithm of odds) of three, with 527 markers mapped into linkage groups. From this initial map, a spaced expression marker-based genetic map consisting of 13 linkage groups containing 218 GEMs, spanning 982.7 cM (centimorgan) of the bambara groundnut genome, was developed. Of the QTL detected, 46% were detected in both control and drought treatment populations, suggesting that they are the result of intrinsic trait differences between the parental lines used to construct the cross, with 31% detected in only one of the conditions. The present GEM map in bambara groundnut provides one technically feasible route for the translation of information and resources from major and model plant species to underutilised and resource-poor crops.
