    An N-acylhomoserine lactone (AHL)-degrading bacterial strain, L62, was isolated from a sample of fermentation brine of Chinese soya sauce by using rich medium agar supplemented with soya sauce (10% v/v). L62, a rod-shaped Gram positive bacterium with amylolytic activity, was phylogentically related to Bacillus sonorensis by 16S ribosomal DNA and rpoB sequence analyses. B. sonorensis L62 efficiently degraded N-3-oxohexanoyl homoserine lactone and N-octanoylhomoserine lactone. However, the aiiA homologue, encoding an autoinducer inactivation enzyme catalyzing the degradation of AHLs, was not detected in L62, suggesting the presence of a different AHL-degrading gene in L62. To the best of our knowledge, this is the first report of AHL-degrading B. sonorensis from soya sauce liquid state fermentation.

    BACKGROUND: To examine the effects of soy [in the form of textured soy protein (TSP) and soy-nut] on body composition in elderly women with metabolic syndrome (MetS).
    METHODS: A 12-week randomized clinical trial was conducted on 75 women between 60-70 years of age with MetS in rural health clinics around Babol, Iran in 2009. The participants were randomly assigned to one of the three groups of soy-nut (35g/d), TSP (35g/d) and control. Body fat, lean mass and anthropometric indicators were measured before and after intervention, too.
    RESULTS: Participants were classified as overweight and showing android fat distribution. After 12 weeks of intervention, both soy-nut and TSP groups showed an increase of non-significant in lean mass (0.9 and 0.7 kg), hip circumference (0.45 and 0.28 cm), triceps skinfold (TSF) thickness (0.87 and 0.67mm) and reduction in BMI (-0.15 and -0.33), waist circumference (-0.83 and -1.2) and body fat (-1.5% and -1.7%). Significant increase in the mean change of TSF and lean mass was observed in the users of soy-nut compared to the control group (P<0.01, P<0.05).
    CONCLUSION: 12-week intervention of soy had a mild favorable effect on body composition in elderly women with MetS.

    The advantage of cooking cannot be summarized just as the better food digestion. Some investigations showed the effect of cooking on reduction of food anti-nutrients such as oxalate. This study was aimed to determine the effect of cooking on oxalate content and its negative effects on calcium availability in eight Malaysian soy-based dishes. Since there is few data which examined the effects of cooking on food oxalate content globally, thus this study was designed as the first in Malaysia. Oxalate in this research was analyzed by using enzymatic methods, while calcium content was determined by using Atomic Absorption Spectrophotometer. The oxalate concentration was in the range of 6.43-19.40 mg/100 g for whole cooked samples, 9.03-11.90 mg/100 g for raw soy products, and 4.36-7.99 mg/100 g for cooked ones. There were 5 out of 12 samples containing oxalate, which was significantly lower (p < 0.05) in cooked products compared to the raw ones. The rest of the samples were also lower in oxalate but not significantly different (p > 0.05). Oxalate in raw/cooked fermented soy products (tempeh) was slightly lower compared to the non-fermented ones. However, there was no significant difference (p > 0.05) in oxalate amount between fermented and non-fermented soy products. As Oxalate/Calcium ratio was below 1, oxalate did not have an effect on availability of calcium in the studied samples. Optimal cooking and food processing might be effective in reducing oxalate content in soy products. There is a need for more investigations about the effect of cooking on soy products to confirm the present results.

    Makanan berasaskan kacang soya terkenal di Asia sejak beribu tahun dahulu dan ia tidak hanya diiktiraf sebagai makanan tradisi tetapi mempunyai potensi untuk kesihatan. Kajian ini dijalankan untuk menentukan penerimaan, kesukaan dan kadar penggunaan produk soya dalam kalangan pengguna di Semenanjung Malaysia. Seramai 330 responden telah terlibat dalam kajian ini. Mereka terdiri daripada pelajar universiti dan masyarakat umum yang berumur 18 tahun ke atas. Setiap responden diminta untuk mengisi borang soal selidik dengan seliaan penyelidik. Kajian menunjukkan bahawa 56% responden lebih menyukai susu soya berbanding dengan produk soya yang lain seperti taufufa (16%), kicap (9%), tempeh (8%), tauhu telur (7%) dan tauhu soya (4%). Sebahagian besar responden (>50%) percaya produk soya boleh mengurangkan risiko kanser dan osteoporosis. Sebanyak 89.4% responden makan tauhu soya. Sementara itu, 63.9% responden makan tauhu telur dan 42.2% suka tauhu telur yang digoreng. Lebih daripada 80% responden meminati warna, aroma, rasa dan tekstur tauhu telur.

    In this study, four selected commercial strains of Aspergillus oryzae were collected from soy sauce koji. These A. oryzae strains designated as NSK, NSZ, NSJ and NST shared similar morphological characteristics with the reference strain (A. oryzae FRR 1675) which confirmed them as A. oryzae species. They were further evaluated for their ability to produce γ-aminobutyric acid (GABA) by cultivating the spore suspension in a broth medium containing 0.4 % (w/v) of glutamic acid as a substrate for GABA production.     Year by year, huge quantities of by-products are generated during the manufacturing process of soybean-based products. Okara is one of the by-products, and it is an insoluble portion of the soybean. It consists of high moisture (8.4-22.9%); on dry matter basis, it contains high metabolizable energy (9.0-14.2 MJ/kg) and other components that include crude protein (20.9-39.1%), crude fiber (12.2-61.3%), crude fat (4.9-21.5%), and ash (3.4-5.3%). Fermentation of okara improves its nutritional quality and reduces its anti-nutrient contents. Due to animals' palatability, okara can be used to replace the soybean meal/concentrate feed partially or completely in ruminant's diet and partially in nonruminant's diet. Okara feeding does not depress the intake, digestibility, growth, milk production, blood metabolic profiles, and meat quality of animals. However, this by-product decays quickly due to its high moisture content, and its heavy weight and sticky nature make it difficult to process and expensive to dry using conventional methods. This paper thoroughly summarizes the utilization of okara as animal feed in the cause of developing a general guideline with favorable levels of inclusion in the diets of animals for its exploitation and valorization. This review will encourage further research to develop eco-friendly and value added feed for animals.

    The objective of the study was to determine the effect of different types of soy sauce and marinating time on the formation of heterocyclic amines (HCAs) in roasted chicken. Chicken breast samples were marinated with sweet, salty, light and dark soy sauce at 0, 3, 6 and 12 h (control treatment was the chicken without marinade). The concentrations of free amino acids, sugars and creatinine were determined before roasting while HCA concentrations were determined after roasting. All types of soy sauce significantly increased (p ≤ 0.05) the concentration of HCAs in roasted chicken with increasing marinating time. The highest increment of total concentration of HCAs was found in samples marinated with light soy sauce (887%) followed by dark (375%), salty (193%) and sweet (169%) at 12 h. PhIP (2-amino-1-methyl-6-phenylimidazo(4,5-b)pyridine) showed a substantial reduction in samples only momentarily marinated with sweet, salty and dark soy sauce (0 h). Free amino acids were found to be more strongly correlated with the formation of HCAs than reducing sugars or creatinine.

    The soya-breast cancer risk relationship remains controversial in Asia due to limited and inconsistent research findings and is exacerbated by difficulties in recruiting and retaining participants in intervention trials. Understanding public perceptions towards soya is important for designing effective intervention trials. Here, we administered a close-ended, quantitative survey to healthy, peri- and post-menopausal Asian women in the Malaysian Soy and Mammographic Density (MiSo) Study to assess perception towards soya and explore motivators and barriers that affect study adherence using the Capability, Opportunity, Motivation and Belief (COM-B) Model and Theoretical Domains Framework (TDF). Of 118 participants, the majority reported the belief that soya promotes good health (Supplement = 85⋅7 %, Diet = 90⋅0 %, Control = 87⋅9 %). Most participants reported obtaining information about soya from the internet (Supplement = 61⋅0 %, Diet = 55⋅3 %, Control = 35⋅9 %), while health professionals were least reported (Supplement = 9⋅8 %, Diet = 7⋅9 %, Control = 5⋅1 %). Stratified analyses by study completion and adherence status yielded comparable findings. By the end of the study, dietary arm participants reported a strong belief that soya has no impact on their health (Supplement = 7⋅1 % v. Diet = 20⋅0 % v. Control = 0⋅0 %, P = 0⋅012). Motivation and opportunity strongly facilitated soya consumption, while psychological capability was the most common barrier to consumption though less evident among dietary arm participants. While most Asian women have a positive perception towards soya, theory-based intervention trials are warranted to understand the perception-study adherence relationship and to accurately inform the public of the health effects of soya.

    Soy-based products are one of famous raw ingredients used in the preparation of Asian cuisines. These soy-based products are good source of isoflavones. This study was carried out to observe the effects of different cooking methods on isoflavone content in soy-based products. A total of eight Malaysian dishes prepared using soy-based products with different cooking methods was selected as samples for this study. Daidzein and genestein contents in raw and cooked soy-based products were both quantified using high performance liquid chromatography (HPLC). The results showed that dishes containing tempe as ingredients had significantly higher (p<0.05) amount of isoflavone content, in both raw and cooked forms, as compared to those prepared from other types of soy-based products when based on dry and wet. In conclusion, the isoflavone content was significantly reduced (p<0.05) in some cooked soy-based products (tempe (TSS and ML), fu jook (ML) and firm tofu (FTC)) based on dry weight as compared with the raw ones. However, the correlations between isoflavone content with cooking methods, durations and temperatures were not significant (p>0.05) in this study.

    Kajian ini dijalankan untuk menentukan kesan perencatan fi tat terhadap bioavailabiliti kalsium (Ca), ferum (Fe) dan zink (Zn) dalam produk kacang soya tempatan terpilih. Fitat menyebabkan kesan perencatan terhadap penyerapan mineral seperti Ca, Fe dan Zn dengan membentuk kompleks yang tidak larut dan tidak dapat dicernakan. Kesan perencatan fi tat terhadap bioavailabiliti mineral seperti Ca, Fe dan Zn telah dianggarkan dengan nisbah molar fi tat kepada mineral. Kandungan fi tat ditentukan dengan menggunakan kaedah pertukaran anion dan kandungan mineral ditentukan dengan Spektrofotometer Serapan Atom (AAS). Produk kacang soya dibeli dari pasar malam dan gerai makanan yang terletak di Cheras dan Sri Rampai, Kuala Lumpur. Setiap sampel yang dibeli dari kedua- dua lokasi dianalisis secara duplikat. Kajian ini mendapati kek soya dan kesemua masakan yang mengandungi tempe tidak mempunyai kandungan fi tat. Ini mungkin disebabkan oleh kehadiran R. oligosporus yang menghasilkan fi tase lalu menghidrolisiskan kandungan fi tat semasa proses fermentasi tempe. Terdapat lima jenis sampel makanan yang mempunyai nisbah molar fi tat/Ca > 0.24, sepuluh jenis sampel makanan dengan nisbah molar fi tat/ Fe > 1, lapan jenis sampel makanan mempunyai nisbah molar fi tat/ Zn > 15 dan satu jenis sampel makanan mempunyai nisbah molar [fi tat x Ca]/ Zn > 200. Keputusan nisbah molar fi tat/ mineral dari kajian ini menganggarkan bioavailabiliti Ca, Fe dan Zn yang rendah. Kesimpulannya, kek soya dan masakan mengandungi tempe mempunyai bioavailabiliti mineral yang tinggi manakala tau-hoo pok bakar mempunyai bioavailabiliti mineral yang rendah.

    The present study investigated the efficiency of aerobic granular sludge (AGS) technology in treating effluent from soy sauce industry which is categorized as a high strength wastewater. The combination of anaerobic and aerobic granulation technology in SBR system was used in this study which was efficiently treated COD from the soy sauce wastewater where 87% of removal was achieved. Ammonia and colour was removed at a maximum of 87 and 76%, respectively, in the SBR system. Matured, dense and compact granules with 2.5 mm in diameters were developed with a good settling velocity (45 m/h) and 28 mL/gSS of sludge volume index (SVI). Hence, AGS technology was proven as an excellent treatment for soy sauce wastewater for being discharge into the environment, as the effluent was treated in one biological reactor with high hydraulic and organic loadings besides less production of sludge. In this study, the capabilities of AGS technology in treating relatively higher concentration of organic impurities present in the soy sauce wastewater were demonstrated.

    In the present work, the biogenic amines tryptamine (TRP), putrescine (PUT), histamine (HIS), tyramine (TYR) and spermidine (SPD) were determined in 32 various types of tofu that were obtained from different states in Malaysia. Three main types of tofu; soft tofu, firm tofu and processed tofu, were analysed in the present work. The biogenic amine contents in the respective types of tofu were analysed by a reversed-phase HPLC with a DAD detector after the aqueous extraction and derivatisation with dansyl chloride. The LOD values ranged from 0.019 mg/L for PUT to 0.028 mg/L for TYR. While, the LOQ values ranged from 0.063 mg/L (PUT) to 0.096 mg/L (TYR). The recovery values for all the five amines ranged from 80.3% to 120.5% with RSD ≤ 3.1%. The total levels of biogenic amines found varied, ranging from 1.5 mg/kg to 687.9 mg/kg, with mean values (p < 0.05) in descending order of 44.6, 12.6, 9.1, 4.8 and 4.7 mg/kg for PUT, TYR, SPD, HIS and TRP, respectively. PUT and TRP were the most prevailing biogenic amines and they were found respectively in 90.62% of the tofu analysed. Significant positive correlations (r = 0.266 to 0.874, p < 0.05) were found between some individual biogenic amines and protein content in all the three types of tofu. However, negative correlations (r = -0.246 to -0.832, p < 0.05) were observed between biogenic amines and moisture content, and between biogenic amines and water activity in all the three types of tofu. Significant and strong correlations (r = 0.525 to 0.999, p < 0.05) were found between most of the individual biogenic amines and the total biogenic amines. Those tofu exceeding the legal limits may affect the health of sensitive individuals.

    In this study, a selected γ-aminobutyric acid (GABA)-rich Malaysian strain Aspergillus oryzae NSK was collected from soy sauce koji. The strain was used to explore the effect of using renewable native sugar syrup, sugarcane, nipa, and molasses as fermentable substrates for developing a novel functional GABA soy sauce. We evaluated the strain using the chosen native sugars for 7 days using shake flask fermentation at 30 °C. The results showed optimum GABA concentration was achieved using cane molasses as the fermentable substrate (354.08 mg/L), followed by sugarcane syrup (320.7 mg/L) and nipa syrup (232.07 mg/L). Cane molasses was subsequently utilized as a substrate to determine the most suitable concentration for A. oryzae NSK to enhance GABA production and was determined as 50% g/L of glucose standard cane molasses. Our findings indicate that cane molasses can be used as a GABA-rich ingredient to develop a new starter culture for A. oryzae NSK soy sauce production.

    In submerged-liquid fermentation, seven key parameters were assessed using one-factor-at-a-time to obtain the highest GABA yield using an industrial soy sauce koji Aspergillus oryzae strain NSK (AOSNSK). AOSNSK generated maximum GABA at 30 °C (194 mg/L) and initial pH 5 (231 mg/L), thus was able to utilize sucrose (327 mg/L of GABA) for carbon source. Sucrose at 100 g/L, improved GABA production at 646 mg/L. Single nitrogen sources failed to improve GABA production, however a combination of yeast extract (YE) and glutamic acid (GA) improved GABA at 646.78 mg/L. Carbon-to-nitrogen ratio (C8:N3) produced the highest cell (24.01 g/L) and GABA at a minimal time of 216 h. The key parameters of 30 °C, initial pH 5, 100 g/L of sucrose, combination YE and GA, and C8:N3 generated the highest GABA (3278.31 mg/L) in a koji fermentation. AOSNSK promisingly showed for the development of a new GABA-rich soy sauce.

    Protein efficiency ratio (PER) and protein digestibility are important parameters used in protein quality determination. Protein nutritive values of selected protein sources: buffalo meat, casein, soy protein isolate, and tempeh, with sodium caseinate as a reference formulation, were evaluated. Determination of proximate analysis, protein quality and protein digestibility were monitored. Procedures for evaluation of protein quality and digestibility included PER using the rat bioassay and in vivo Apparent Protein Digestibility (APD). The rats fed with buffalo meat had the highest mean increase in body weight (102.73g±8.95) while rats fed with tempeh had the lowest mean for increase in body weight (16.34g±9.11). Although the mean for body weight gained showed significant differences between all treatments (P0.05) found between casein and soy protein isolate for total food intake. For the PER value, buffalo meat had the highest value (2.99), followed by sodium caseinate (2.41), casein (1.93), soy protein isolate (1.52) and tempeh (1.10). The PER value for buffalo meat (2.99) was higher than sodium caseinate (2.41) while the rest of the treatment were comparatively lower than sodium caseinate. For the in vivo apparent protein digestibility, tempeh had the highest value (91.41%±3.76), followed by casein (91.34%±3.15), buffalo meat (90.79%±1.44), soy protein isolate (89.52%±2.96) and sodium caseinate (89.47%±2.31).

    Quantitation of isoflavones in humans is important to establish the benefits of these compounds to the populations. Urinary isoflavones are frequently used as a biomarker of isoflavone bioavailability from food or supplement since urine contains 100-fold higher concentrations of isoflavones. The objective of the present study was to determine and compare the urinary excretions of daidzein (DA), genistein (GE) and equol (EQ) in postmenopausal Malay women following the consumption of tempeh and milk in a calcium absorption study and to test the hypothesis that the excretion of isoflavones following consumption of tempeh maybe higher compared with milk. The amounts of DA (47.06 ± 4.18 μmol/h), GE (33.27 ± 3.71 μmol/h) and EQ (24.35 ± 4.34 μmol/h) excreted in urine following tempeh consumption were significantly higher (P < 0.05) compared with those in milk (3.51 ± 0.62 μmol/h DA, 2.79 ± 0.35 μmol/h GE and 0 μmol/h EQ). Almost all studied postmenopausal Malay women were able to excrete EQ following consumption of 240 g tempeh but only one subject can be classified as an equol producer. We concluded that most postmenopausal Malay women excreted DA, GE and EQ in their urine following tempeh consumption and the amount of the excreted isoflavones were higher compared with those in milk. However, further studies are needed to determine whether longer periods of time are required to capture EQ producers.

    Acceptance of healthful foods by consumers is not yet well understood. In this study, 3 formulations of frozen dessert bars were prepared containing both soy and wild blueberries. Soy content was controlled to provide an amount of soy protein that qualified for the health claim for soy and reduced risks for cardiovascular disease. Consumers were asked to complete the Health and Taste Attitude Scales (HTAS) and then evaluate the acceptability of the 3 frozen bar types using a 9-point hedonic scale. One week after the 1st session, the participants returned. Approximately half were given information to read regarding the health benefits of soy protein, the other participants were given no information. The samples were then presented a 2nd time and labeled with their soy protein content. Changes in hedonic scores between sessions were compared and correlated with HTAS ratings. Nutrition information generally did not affect acceptability scores.

    Quantifiable levels of 3-chloropropane-1,2-diol (3-MCPD) and 1,3-dichloro-2-propanol (1,3-DCP) were found in domestically manufactured soy-based sauces. Selected commercial foods in the Malaysian market (n = 43) were analyzed for their 3-MCPD and 1,3-DCP contents using a validated gas chromatography-mass spectrometry technique. The 3-MCPD and 1,3-DCP contents of the analyzed food samples varied from not detectable levels to 0.1223 ± 0.0419 mg kg-1 and not detectable levels to 0.025 ± 0.0041 mg kg-1, respectively. High concentrations of 3-MCPD, exceeding Malaysia's maximum tolerable limit of 0.02 mg kg-1, were found in chicken seasoning cubes (mean = 0.0898 ± 0.0378 mg kg-1). Monte Carlo simulation-based health risk assessment revealed that 3-MCPD and 1,3-DCP intakes in the 50th, 95th, and 99th percentiles were lower than 4 µg kg-1 bw day-1, the limit recommended by JECFA in 2016. Hence, it was concluded that the exposure of Malaysian citizens to chloropropanols through soy sauce consumption does not present a health risk.

    BACKGROUND: Tempeh is a widely known fermented soybean that contains elevated level of bioactive contents. Our previous study has shown that anaerobic fermented Nutrient Enriched Soybean Tempeh (NESTE) with increase amino acid and antioxidant levels possessed better hepatoprotective effect than raw soybean.

    METHODS: In this study, the anti-inflammatory effect of the NESTE aqueous extract and raw soybean aqueous extract (SBE) were evaluated by quantifying the inhibition of IL-1β, TNF-α and nitric oxide (NO) secretion in LPS treated RAW 264.7 cell in vitro. On the other hand, in vivo oral acute toxicity effect of the extract was tested on mice at the dose of 5000 mg/kg body weight. In vivo oral analgesic effect of both aqueous extracts at 200 and 1000 mg/kg body weight was evaluated by the hot plate test.

    RESULTS: In the in vitro anti-inflammatory study, 5 mg/mL NESTE was able to inhibit 25.50 ± 2.20%, 35.88 ± 3.20% and 28.50 ± 3.50% of NO, IL-1β and TNF-α production in LPS treated RAW 264.7 cells without inducing cytotoxic effect on the cells. However, this effect was lower than 4 μg/mL of curcumin, which inhibited NO, IL-1β and TNF-α production by 89.50 ± 5.00%, 78.80 ± 6.20% and 87.30 ± 4.00%, respectively. In addition, 1.5 to 2.5-fold increase of latency period up to 120 min for mice in the hot plate test was achieved by 1000 mg/kg NESTE. The analgesic effect of NESTE was better than 400 mg/kg of acetyl salicylic acid, which only increased ~ 1.7-fold of latency period up to 90 min. Moreover, NESTE did not show acute toxicity (no LD50) up to 5000 mg/kg body weight.

    CONCLUSION: NESTE is a nutritious food ingredient with potential anti-inflammatory and analgesic effects.

    Processing of soybeans to other products and consumption of soy products is increasing worldwide mainly due to acclaimed health benefits. Processing can alter soybean sensory appeal, nutritive value and potentially affect consumer health. Rhizopus oligosporus was used to ferment soybean for 3 days. The tempeh flour (TF) was produced form tempeh while defatted tempeh flour (DTF) was then produced from TF by immersing in hexane solvent while soy protein isolate (SPI) was prepared from DTF by using alkali and acid followed by neutralization treatment. In this study, nutritional properties and amino acid content of tempeh, TF, DTF and SPI were determined. Therefore, the objective of this study is was to evaluate the effect of each treatment on the chemical composition and amino acid content for all the samples. The results showed that the nutritional properties (total ash, moisture, crude fat, total carbohydrate and crude fibre) were reduced significantly (p < 0.05) except for protein content. Protein content was significantly (p < 0.05) increased by 50.5% in SPI. For amino acid content, the results obtained showed that SPI contain highest amount of essential and non-essential amino acid followed by DTF, Tempeh and TF. Glutamic acid was found to be the highest amino acid component in all samples. The evaluation from the results showed that SPI can be considered as potential functional food ingredients.
