    The aim of this study was to investigate the genotoxicity effect of Cd and Zn and their binary mixtures in tilapia fish Oreochromis niloticus using the micronucleus test. Two cytogenetic end points were considered; the frequencies of micronucleated cells and nuclear abnormalities. Fishes were exposed to 4.63 mg/L Cd, 7.50 mg/L Zn and 4.63 mg/L Cd + 7.50 mg/L Zn mixture for the period of 24, 48, 72 and 96 h. The results showed that the frequencies of micronuclei and nuclear abnormalities in the erythrocyte were significantly increased in all groups of treatments when compared with the control group (0 exposures). In addition, the highest frequencies of micronucleated and nuclear abnormalities were obtained after 48 h exposure in almost all cases (except in the mixture of Cd+Zn) and decreased after 72 and 96 h exposure. Frequencies of micronuclei and erythrocytes with nuclear abnormalities exposed to a mixture of Cd+Zn in O. niloticus were always lower at all-time points (after 24, 48, 72 and 96 h) than that of a single Cd and Zn exposure. Therefore, the study demonstrated that the genotoxic potential of these metal compounds and the simultaneous treatment of Cd and Zn suggest the presence of antagonistic interactions.

    A comprehensive survey and descriptions on the petiole anatomy was undertaken on 16 species belonging to four selected genera (Anisoptera, Cotylelobium, Vatica and Upuna) in the tribe Dipterocarpeae to investigate variations in vascular bundle structure. Methods used in this study were petiole sectioning using a sliding microtome, staining, mounting and observation under light microscope. Findings had shown that all species studied have complex vascular bundle structures, consisting of outer and medullary vascular bundles. Four different types of vascular bundle arrangements were found and described as Types 1, 2, 3 and 4. The vascular bundle types were determined based on the arrangement and system of vascular bundle strands present in the petiole transverse sections. These vascular bundle types were determined and suggested for easy reference. The results showed that vascular bundles can be used for identification of certain species and can definitely be used for classification of different genera. The presence of sclerenchyma cells is useful in differentiating genera. As a conclusion, the petiole vascular bundle anatomical characteristics have taxonomic value, especially in the identification of some species and possibly applicable in the classification of the tribe Dipterocarpeae.

    Introduction: Virgin coconut oil (VCO) is known for its health and therapeutic benefits. However, the immunomodulatory effects of VCO have not been extensively investigated. Objective: The present study was devoted to examining the effects of VCO on cyclophosphamide (CY)-induced toxicity of lymphoid tissues. Methods: Thirty healthy male Wistar rats were sorted into 5 groups of 6 animals. The first control (NC) group was given distilled water via gavage at 5 ml/kg once daily. The second (CY) group received CY orally at 10 mg/kg/day for 4 weeks. Rats in the other three groups (CV5, CV10, and CV15) were given 10 mg/kg/day CY for 4 weeks, 5 m/kg/day, 10 ml/kg/day and 15 ml/kg/day VCO for 6 weeks, respectively. Rats were sacrificed at the end of 6th week; blood sample from the animals was collected for full blood count and biochemical analysis. The thymus and spleen of each animal was processed for histological examination. Results: The thymus and spleen showed marked reduction in lymphoid cellularity following daily administration of CY. The thymus also showed a marked reduction in the size of the medulla, and the white pulp areas of spleen had reduction in the follicle number and size. Supplementation with 10 ml/kg and 15 ml/kg VCO showed evidence of restoration of both the thymus and splenic lymphoid architecture. The total white cell counts, absolute lymphocyte counts and plasma globulin levels of the VCO groups were significantly increased compared to CY group. Conclusion: VCO displayed potential protective effects on CYinduced histological changes in lymphoid tissues.

    Segmentation and counting of blood cells are considered as an important step that helps to extract features to diagnose some specific diseases like malaria or leukemia. The manual counting of white blood cells (WBCs) and red blood cells (RBCs) in microscopic images is an extremely tedious, time consuming, and inaccurate process. Automatic analysis will allow hematologist experts to perform faster and more accurately. The proposed method uses an iterative structured circle detection algorithm for the segmentation and counting of WBCs and RBCs. The separation of WBCs from RBCs was achieved by thresholding, and specific preprocessing steps were developed for each cell type. Counting was performed for each image using the proposed method based on modified circle detection, which automatically counted the cells. Several modifications were made to the basic (RCD) algorithm to solve the initialization problem, detecting irregular circles (cells), selecting the optimal circle from the candidate circles, determining the number of iterations in a fully dynamic way to enhance algorithm detection, and running time. The validation method used to determine segmentation accuracy was a quantitative analysis that included Precision, Recall, and F-measurement tests. The average accuracy of the proposed method was 95.3% for RBCs and 98.4% for WBCs.

    The distribution of immunoregulatory cells in the peripheral blood of an individual has now been established as an important tool in helping the management of several diseases. It is necessary to set the normal ranges of these cells for the laboratory. We have undertaken in this study to establish the reference ranges for normal Malaysian adults. We found that the mean percentages of T cells, B cells, T Helper cells (CD4), T suppressor cells (CD8), NK cells and the ratio of CD4/CD8 were 70.91%, 11.38%, 38.15%, 37.76%, 17.45%, and 1.00 respectively. There was no significant difference between the sexes. In certain parameters, there was significant differences between Malay, Chinese and Indians. The Chinese and Indians were significantly different in the distribution of B cells and in the CD4/CD8 ratio. In the case of CD4 and NK cells, the Indians were different from the other two groups.

    OBJECTIVES: The aim of this study was to determine the changes in cell density and morphology of selected cells of the ageing human dental pulp.

    BACKGROUND: Changes in cell density and morphology of dental pulp cells over time may affect their capability to respond to tooth injury.

    MATERIALS AND METHODS: One hundred thirty-one extracted teeth were obtained from individuals between the ages of 6 and 80 years. The apical 1/3 of the root region was removed from all teeth prior to routine processing for producing histological slides. The histology slides were used to study the changes in cell density and morphology of selected pulp cells; odontoblasts, subodontoblasts and fibroblasts in the crown and root regions of the dental pulp. Student's t-test and one-way anova were used for statistical analyses.

    RESULTS: In all age groups, the cell density for all types of cells was found to be higher in the crown than in the root (p cell density was found to decrease with age in both the crown and root regions. However, it was noted that the reduction of coronal odontoblasts occurred later in life (40-49 years) when compared to that of subodontoblasts or fibroblasts (30-39 years).

    CONCLUSIONS: The density of the coronal pulp cells reduces and these cells undergo morphological changes with ageing of individuals and this may affect the pulp's ability to resist tooth injury.

    This study was conducted to describe corneal endothelial cell density and morphology in Malay eyes. Non-contact specular microscopy was performed in 125 eyes of 125 Malay volunteers, aged 20-87 years. Studied parameters included endothelial cell density (CD), mean cell area (MCA), coefficient of variation (CV) in cell area, as well as hexagonal appearance of the cells. Mean endothelial cell density in the study population was 2648 +/- 310 cell/mm(2). Mean CA, CV and percentage of hexagonal cells were 382.8 +/- 47.7microm(2), 58.1 +/- 22.6, 44.3% +/- 11.5% respectively. There was a statistically significant decrease in endothelial cell density (correlation - 0.300, P = 0.001) and CV in cell size (correlation - 0.208, P = 0.02) with age. There was a statistically significant increase in mean cell area (correlation 0.300, P = 0.001) with increasing age. The correlation between age and percentage of hexagonal cells was insignificant (correlation 0.074, P = 0.41). In conclusion, a consistent decrease was noted in the endothelial cell density with increasing age. The differences in endothelial cell density between genders were statistically insignificant.

    Complete blood counts were analyzed for 30 samples of human blood with radiation dose rate ranging between 10 and 41 μSv/h using a Radium-226 source with different time of exposure. A new technique involving a nuclear track detector type CR-39(CR-39 NTDs) was used to estimate the alpha particle density incident on the blood samples. The results show that the ranges of alpha particle in blood samples and on the surface of CR-39NTDs vary exponentially with energy of alpha particles. This depends on the restricted energy loss and target density. Changes in the blood components due to irradiation occurred for different durations of irradiation, and the duration of irradiation that influenced the blood samples in this study was 6 min. The change in red blood cell (RBC) was negligible, so it is less affected than other blood components. In addition, most changes in the blood contents began at a low radiation dose (10.38-13.41 μSv/h). For the doses 13.41-21.77 μSv/h, platelet (PLT) counts increased rapidly and adversely with the RBC and white blood cell (WBC) due to chromosomal aberration. Besides, rapid PLT count reduction rapidly at high dose (42.1 μSvh) causes thrombocytopenia; in contrast, WBC increased, which is an indication of cancer caused due to increase in alpha particle dose. Generally, our results are in agreement with the essentials of blood content and the principles of biological radiation interaction.

    Health and disease can only be distinguished by accurate and reliable reference values of a particular laboratory test. It is now a proven fact that there is considerable variation in hematology reference intervals depending on the demographic and preanalytical variables. There are evidences that values provided by manufacturers do not have appropriate application for all populations. Moreover, reference ranges provided by different laboratory manuals and books also do not solve this problem. We are presenting here normal reference ranges of Malaysian population. These values were determined by using Sysmex XE-2100 and ACL 9000 hematology and coagulation analyzers. Results from this study showed that there were considerable differences in the reference values from manufacturers, western population or laboratory manuals compared with those from the local population.

    OBJECTIVE: This study was conducted to investigate the effects of low-level laser (LLL) doses on human red blood cell volume. The effects of exposure to a diode pump solid state (DPSS) (λ = 405 nm) laser were observed.

    BACKGROUND DATA: The response of human blood to LLL irradiation gives important information about the mechanism of interaction of laser light with living organisms. Materials and methods Blood samples were collected into ethylenediaminetetraacetic acid (EDTA)-containing tubes, and each sample was divided into two equal aliquots, one to serve as control and the other for irradiation. The aliquot was subjected to laser irradiation for 20, 30, 40, or 50 min at a fixed power density of 0.03 W/cm(2).     In order to assess the feasibility of various carbon sources and plant materials in increasing the growth rate and biomass productivity of Monoraphidium griffithii, ten carbon sources as well as six plant materials were tested in mixotrophic cultures with or without aeration. It was found that glucose, fructose, maltose, sodium acetate and mannitol were potential carbon sources for growth enhancement of M. griffithii. Supplementation of culture medium with these carbon sources resulted in approximately 1-4-fold increase in cell density compared to control in a small scale culture. In a larger scale mixotrophic culture with aeration, 0.05% mannitol and 0.1% fructose resulted in a decent 1-1.5-fold increase in final cell density, approximately 2-fold increase in growth rate and 0.5-1-fold increase in dry biomass weight. Findings from this study suggests that glucose, fructose, maltose and mannitol were potential organic carbon sources for mixotrophic culture of M. griffithii.

    There are few randomized trials comparing filgrastim and pegfilgrastim in peripheral blood stem cell mobilization (PBSCM). None of the trials studied the effects of the timing of pegfilgrastim administration on the outcomes of mobilization. We conducted a randomized triple blind control trial comparing the outcomes of filgrastim 5 µg/kg daily from day 3 onwards, 'early' pegfilgrastim 6 mg on day 3 and 'delayed' pegfilgrastim 6 mg on day 7 in cyclophosphamide PBSCM in patients with no previous history of mobilization. Peripheral blood (PB) CD34+ cell count was checked on day 8 and day 11 onward. Apheresis was started when PB CD34+ ≥ 10/µl from day 11 onward. The primary outcome was the successful mobilization rate, defined as cumulative collection of ≥2 × 10(6)/kg CD34+ cells in three or less apheresis. The secondary outcomes were the day of neutrophil and platelet engraftment post transplantation. There were 156 patients randomized and 134 patients' data analyzed. Pegfilgrastim 6 mg day 7 produced highest percentage of successful mobilization, 34 out of 48 (70.8%) analyzed patients, followed by daily filgrastim, 28 out of 44 (63.6%) and day 3 pegfilgrastim, 20 out of 42 (47.6%) (p = 0.075). Pegfilgrastim day 7 and daily filgrastim reported 1.48 (p = 0.014) and 1.49 (p = 0.013) times higher successful mobilization rate respectively as compared to pegfilgrastim day 3 after adjusting for disease, gender and exposure to myelotoxic agent. Multiple myeloma patients were three times more likely to achieve successful mobilization as compared to acute leukemia or lymphoma patients. Pegfilgrastim avoided the overshoot of white cells compared to filgrastim. There was no difference in the duration of both white cells and platelet recovery post transplantation between the three interventional arms.

    Red tide of dinoflagellate was observed in brackish water fish ponds of Terengganu along the coast of the South China Sea during the study period between January 1992 to December 1992. The nearby coastal moat water facing the South China Sea is the source of water for fish pond culture activities of sea bass during the study period. An examination of water quality in fish ponds during the study period indicated that both the organic nutrients were high during the pre-wet monsoon period. The source of the nutrients in coastal water was believed to be derived from the agro-based industrial effluents, fertilizers from paddy fields and untreated animal wastes. This coincided with the peak production of dinoflagellate in the water column in October 1992. The cell count ranges from 8.3 to 60.4×10.4×10(4)/l during the bloom peak period and the bloom species were compared entirely of non-toxic dinoflagellates with Protoperidinium quinquecorne occurring >90% of the total cell count. However, both cultured and indigenous fish species were seen to suffer from oxygen asphyxiation (suffocation due to lack of oxygen). The bloom lasted for a short period (4-5 days) with a massive cell collapse from subsurface to bottom water on the sixth day. The productivity values ranged from 5-25 C g/ l / h with a subsurface maximum value in October 1992. Two species of Ciliophora, Tintinnopsis and Favella, were observed to graze on these dinoflagellates at the end of the bloom period.

    We studied by scanning electron microscopy the effects of the repeated use of a newly-designed, disposable, suction trephine (Microsurgical, Ltd, Bedfordshire, England) on the nature of the cut of successive corneal buttons from sheep eyes. After the trephine was used more than nine times, the cut edge became more irregular as successive buttons were produced. Also, progressively greater degrees of endothelial cells adjacent to the cut edge were lost. Although the corneal buttons initially were circular, those produced after the 20th cut became more oval. These results suggest that a disposable corneal trephine should be reused no more than nine times.

    Cancer is one of the global health problems that has a detrimental effect
    to a person's life. However, chemotherapeutic agents success are subject to the side
    effects due to lack of specificity in the drug delivery system to cancer cells and an
    increase risk of systemic toxicity to the normal cells. Polyamine transport system (PTS)
    is one of the potential pathways for transporting anticancer agent into specific cancer
    cells. This is due to the upregulation of PTS in cancer cells compared to normal cells for
    the proliferation activity. The aim of this study was to investigate the cytotoxicity effect
    of putrescine-sulphur analogues type 1 (PSA-1) and type 2 (PSA-2) on human lung
    adenocarcinoma cells (A549), human colorectal adenocarcinoma cells (HCT-8) and
    human breast adenocarcinoma cell (MCF-7).

    Drug resistance remains a severe problem in most chemotherapy regimes. Recently, it has been suggested that cancer cell-derived extracellular vesicles (EVs) could mediate drug resistance. In this study, the role of EVs in mediating the response of oral squamous cell carcinoma (OSCC) cells to cisplatin was investigated. We isolated and characterized EVs from OSCC cell lines showing differential sensitivities to cisplatin. Increased EV production was observed in both de novo (H314) and adaptive (H103/cisD2) resistant lines compared to sensitive H103 cells. The protein profiles of these EVs were then analyzed. Differences in the proteome of EVs secreted by H103 and H103/cisD2 indicated that adaptation to cisplatin treatment caused significant changes in the secreted nanovesicles. Intriguingly, both resistant H103/cisD2 and H314 cells shared a highly similar EV protein profile including downregulation of the metal ion transporter, ATP1B3, in the EVs implicating altered drug delivery. ICP-MS analysis revealed that less cisplatin accumulated in the resistant cells, but higher levels were detected in their EVs. Therefore, we inhibited EV secretion from the cells using a proton pump inhibitor and observed an increased drug sensitivity in cisplatin-resistant H314 cells. This finding suggests that control of EV secretion could be a potential strategy to enhance the efficacy of cancer treatment.

    Marudu Bay, north coast of Sabah is characterized with mesotrophic water body and typical environmental parameters
    throughout the year. The current study was undertaken to evaluate the effect of environmental parameters and nutrients
    in mesotrophic water on the occurrence and distribution of potentially harmful phytoplankton species. The samplings
    were conducted over a period of thirteen months, covering southwest monsoon (SWM), inter-monsoon (IM), and northeast
    monsoon (NEM), at ten stations throughout the bay. Physical parameters (temperature, salinity, pH, dissolved oxygen,
    current speed and secchi depth), biological parameters (cell densities of phytoplankton) and chemical parameters
    (phosphate, nitrate, silicate and ammonia) were examined. The results indicated at least eight potentially harmful
    phytoplankton species (Dinophysis caudata, D. miles, Ceratium furca, C. fursus, Prorocentrum micans, P. sigmoides, P.
    triestinum and Pseudo-nitzschia sp.) were detected in north coast of Sabah. However, the potentially harmful phytoplankton
    species contributed only about 1.3% of the total phytoplankton community. Under nutrient deprivation conditions, the
    potentially harmful phytoplankton species distribution was mainly influenced by the ability to utilize other nitrogen
    sources, cell mobility and toleration to low nutrients environments.

    Chitosan nanoparticles (CSNPs) have potential applications in stem cell research. In this study, ex vivo cytotoxicity of CSNPs on mouse bone marrow-derived (MBMCs) hematopoietic stem and progenitor cells (HSPCs) was determined. MBMCs were exposed to CSNPs of different particle sizes at various concentrations for up to 72 h. Cytotoxicity effect of CSNPs on MBMCs was determined using MTT, Live/Dead Viability/Cytotoxicity assays and flow cytometry analysis of surface antigens on HSCs (Sca-1(+)), myeloid-committed progenitors (CD11b(+), Gr-1(+)), and lymphoid-committed progenitors (CD45(+), CD3e(+)). At 24 h incubation, MBMCs' viability was not affected by CSNPs. At 48 and 72 h, significant reduction was detected at higher CSNPs concentrations. Small CSNPs (200 nm) significantly reduced MBMCs' viability while medium-sized particle (∼400 nm) selectively promoted MBMCs growth. Surface antigen assessment demonstrated lineage-dependent effect. Significant decrease in Sca-1(+) cells percentage was observed for medium-sized particle at the lowest CSNPs concentration. Meanwhile, reduction of CD11b(+) and Gr-1(+) cells percentage was detected at high and intermediate concentrations of medium-sized and large CSNPs. Percentage of CD45(+) and CD3e(+) cells along with ROS levels were not significantly affected by CSNPs. In conclusion, medium-sized and large CSNPs were relatively non-toxic at lower concentrations. However, further investigations are necessary for therapeutic applications.
