Trace and minor elements play crucial roles in a variety of biological processes, including amyloid fibrils formation. Mechanisms include activation or inhibition of enzymatic reactions, competition between elements and metal proteins for binding positions, also changes to the permeability of cellular membranes. These may influence carcinogenic processes, with trace and minor element concentrations in normal and amyloid tissues potentially aiding in cancer diagnosis and etiology. With the analytical capability of the spectroscopic technique X-ray fluorescence (XRF), this can be used to detect and quantify the presence of elements in amyloid characterization, two of the trace elements known to be associated with amyloid fibrils. In present work, involving samples from a total of 22 subjects, samples of normal and amyloid-containing tissues of heart, kidney, thyroid, and other tissue organs were obtained, analyzed via energy-dispersive X-ray fluorescence (EDXRF). The elemental distribution of potassium (K), calcium (Ca), arsenic (As), and iron (Fe) was examined in both normal and amyloidogenic tissues using perpetual thin slices. In amyloidogenic tissues the levels of K, Ca, and Fe were found to be less than in corresponding normal tissues. Moreover, the presence of As was only observed in amyloidogenic samples; in a few cases in which there was an absence of As, amyloid samples were found to contain Fe. Analysis of arsenic in amyloid plaques has previously been difficult, often producing contradictory results. Using the present EDXRF facility we could distinguish between amyloidogenic and normal samples, with potential correlations in respect of the presence or concentration of specific elements.

Small islands are susceptible to anthropogenic and natural activities, especially in respect of their freshwater supply. The freshwater supply in small islands may be threatened by the encroachment of seawater into freshwater aquifers, usually caused by over pumping. This study focused on the hydrochemistry of the Kapas Island aquifer, which controls the groundwater composition. Groundwater samples were taken from six constructed boreholes for the analysis and measurement of its in-situ and major ions. The experimental results show a positive and significant correlation between Na-Cl (r=0.907; p<0.01), which can be defined as the effect of salinization. The mechanisms involved in groundwater chemistry changes were ion exchange and mineralization. These processes can be demonstrated using Piper's diagram in which the water type has shifted into a Na-HCO(3) water type from a Ca-HCO(3) water type. Saturation indices have been calculated in order to determine the saturation condition related to dissolution or the precipitation state of the aquifer bedrock. About 76% of collected data (n=108) were found to be in the dissolution process of carbonate minerals. Moreover, the correlation between total CEC and Ca shows a positive and strong relationship (r=0.995; p<0.01). This indicates that the major mineral component in Kapas Island is Ca ion, which contributes to the groundwater chemical composition. The output of this research explains the chemical mechanism attributed to the groundwater condition of the Kapas Island aquifer.

A water extraction method has been used to extract plant proteins from the roots of Eurycoma longifolia harvested from Perak and Pahang, Malaysia. On the basis of the spectroscopic Bradford assay, Tongkat Ali Perak and Pahang contained 0.3868 and 0.9573 mg mL(-1) of crude protein, respectively. The crude proteins were separated by one dimensional 15% sodium dodecyl sulphate polyacrylamide gel electrophoresis into two (49.8 and 5.5 kD) and four (49.8, 24.7, 21.1 and 5.5 kD) protein spots for Tongkat Ali Perak and Pahang, respectively. Isoleucine was present in the highest concentration significantly. Both plant samples showed differences in the mineral and trace element profiles, but the minerals calcium, magnesium and potassium were present in the highest concentration. The highly concerned toxic metals such as arsenic and lead were not detected.

A study on elemental composition in the otolith of giant mudskipper, Periophthalmodon schlosseri, was done from June to October 2003. Specimens were obtained from the mangrove areas of Kuala Selangor, Sepang and Melaka in the west coast of Peninsular Malaysia. A total of 70 sagitta otoliths were analyzed to detect variation of Sr, Ba and Mg, replacing the natural chemical composition of the otolith, which is the calcium carbonate (CaCO3). The average ratio of Sr:Ca was 0.11 x 10(-4), Ba:Ca was 5.7 x 10(-3) and Mg:Ca was 0.2 x 10(-3). Strong correlation (R > 0.8) between fish body size and otolith weight ofmudskipper (p < 0.01) also found during this study.

Vitamin E is found to reverse the effects of nicotine on bone and this study aimed to determine its mechanism. Male Sprague Dawley rats were divided into four groups and treated for 3 months: Group 1 was the control group (RC). Groups 2 (N), 3 (N+TT) and 4 (N+ATF) received nicotine 7 mg/kg throughout the treatment period. In addition, groups 3 and 4 received tocotrienol 60 mg/kg and alpha-tocopherol 60 mg/kg respectively during months 2 and 3. Parameters measured were serum osteoprotegerin (OPG), serum receptor activator of nuclear factor kappa B ligand (RANKL), femoral and lumbar bone calcium content and body weight. Nicotine did not affect OPG or RANKL levels but reduced bone calcium content suggesting the calcium loss is not due to increase osteoclastogenesis. OPG was increased in N+ATF while RANKL was slightly increased in N+TT. Both vitamin E supplements restored bone calcium loss induced by nicotine. Nicotine impaired weight gain in all treatment groups starting week 4 however, N+TT group was comparable to RC from week 6 onwards. Bone protective effects of ATF, but not TT, may be partly due to inhibition of osteoclastogenesis.

BACKGROUND: There have been cononcerns about the dissolution of conventional glass ionomer cement (GIC) and its possible degradation when exposed to an acidic environment over time. The objective of this study was to investigate the effects of exposure of Fuji IX Fast to the simulated acidic aspects of the oral environment in terms of any change in the elemental composition of strontium (Sr), phosphorus (P), calcium (Ca) and fluorine (F) which resulted at the surface of this material.

METHODS: Sixty-five cylindrical block of Fuji IX Fast were prepared using split moulds. The demineralizing solution was an acetate buffered demineralizing solution at pH 403. The remineralizing solution was a buffered solution containing 1.5 mM Ca, 0.9 mM P and 10 ppm F at pH 7. The blocks of Fuji IX Fast were subjected either to two-day alternating cycles of remineralization and demineralization for up to 24 days (test); 6 two-day cycles of demineralizing or remineralizing solution separately, or deionized distilled water alone (controls) or were left untreated (base line control). Mineral profiles of Ca, P, Sr and F within 100 microm of the material surface were assessed following 8, 16 and 24 days of treatment (test); 4, 8 or 12 days (controls) or for baseline control samples, using electron probe microanalysis (EPMA).

RESULTS: There were significant changes in mineral profile in the test specimens in terms of Sr and Ca concentrations. A molecule for molecule exchange of these elements resulted between GIC and eluant solutions. Fluoride loss from the GIC occurredto the level comparable with uptake levels recorded in eluant solutions from previous studies. The ionic exchanges appeared to be the result of dissolution followed by an equilibrium-driven diffusion. These exchanges were superficial though substantial.

CONCLUSIONS: Simulated exposure of Fuji IX to the oral environment resulted in an exchange of Ca from the bathing solutions into Fuji IX to replace any Sr which was lost to the GIC. Fluorine loss from the GIC followed previously described patterns. The possible clinical significance of this exchange was discussed.

The concentrations of calcium, phosphate, protein and nitrite in whole unstimulated saliva, and the salivary flow rate under fasting conditions (saliva collected at least after 6 h without food and water) were compared with those under control conditions (saliva collected within 30 min to 1 h after food). The flow rate of fasting saliva was half that of control (0.098 ml/min vs 0.208 ml/min) and no significant differences in the flow rate were observed between sexes. The concentration of nitrite under fasting conditions was 50% higher than that in control saliva (p less than 0.05). The protein concentration was decreased, but not significantly, under fasting conditions. The composition of fasting saliva with regard to calcium and phosphate concentrations was comparable to that of the control. No significant variations in these components between sexes were observed under either condition.

Calcium contents of demineralised human cortical bone determined by titrimetric assay and atomic absorption spectrophotometry technique were verified by comparing to neutron activation analysis which has high recovery of more than 90%. Conversion factors determined from the comparison is necessary to correct the calcium content for each technique. Femurs from cadaveric donors were cut into cortical rings and demineralised in 0.5 M hydrochloric acid for varying immersion times. Initial calcium content in the cortical bone measured by titration was 4.57%, only 21% of the measurement by neutron activation analysis; while measured by atomic absorption spectrophotometer was 13.4%, only 61% of neutron activation analysis. By comparing more readings with the measurements by neutron activation analysis with 93% recovery, a conversion factor of 4.83 was verified and applied for the readings by titration and 1.45 for atomic absorption spectrophotometer in calculating the correct calcium contents. The residual calcium content started to reduce after the cortical bone was demineralised in hydrochloric acid for 8 h and reduced to 13% after 24 h. Using the linear relationship, the residual calcium content could be reduced to less than 8% after immersion in hydrochloric acid for 40 h. Atomic absorption spectrophotometry technique is the method of choice for calcium content determination as it is more reliable compared to titrimetric assay.

It is important to assess indoor air quality in school classrooms where the air quality may significantly influence school children's health and performance. This study aims to determine the concentrations of PM2.5 and dust chemical compositions in indoor and outdoor school classroom located in Kuala Lumpur City Centre. The PM2.5 concentration was measured from 19th September 2017-16th February 2018 using an optical PM2.5 sensor. Indoor and outdoor dust was also collected from the school classrooms and ion and trace metal concentrations were analysed using ion chromatography (IC) and inductively couple plasma-mass spectrometry (ICP-MS) respectively. This study showed that the average indoor and outdoor 24 h PM2.5 was 11.2 ± 0.45 µg m-3 and 11.4 ± 0.44 µg m-3 respectively. The 8 h PM2.5 concentration ranged between 3.2 and 28 µg m-3 for indoor and 3.2 and 19 µg m-3 for outdoor classrooms. The highest ion concentration in indoor dust was Ca2+ with an average concentration of 38.5 ± 35.0 µg g-1 while for outdoor dust SO42- recorded the highest ion concentration with an average concentration of 30.6 ± 9.37 µg g-1. Dominant trace metals in both indoor and outdoor dust were Al, Fe and Zn. Principle component analysis-multiple linear regression (PCA-MLR) demonstrated that the major source of indoor dust was road dust (69%), while soil dominated the outdoor dust (74%). Health risk assessment showed that the hazard quotient (HQ) value for non-carcinogenic trace metals was

This article reports a degradation study that was done on stent prototypes made of biodegradable Fe35Mn alloy in a simulated human coronary arterial condition. The stent degradation was observed for a short-term period from 0.5 to 168 h, which simulates the early period of stenting procedure. Potentiodynamic polarization and electrochemical impedance spectroscopy were used to quantify degradation rate and surface property of the stents. Results showed that signs of degradation were visible on both crimped and expanded stents after 1 h of test, mostly located on the stent's curvatures. The degradation rate of stent was higher compared to that of the original alloy, indicating the surface altering effect of stent fabrication processing to degradation. A single oxide layer was formed and detected as a porous structure with capacitive behavior. Expanded stents exhibited lower polarization resistance compared to the nonexpanded ones, indicating the cold work effect of expansion procedure to degradation.

Dentine erosion is an increasingly recognised problem, especially in aging population, and various methods have been utilised for its assessment. This narrative review was planned to summarise the methods for the assessment of the early stages of dentine erosion. Relevant original articles published in the English language from 2013 to 2017 were reviewed. Laboratory techniques and methods with in vivo potential were separately studied. It is evident that the assessment of early dentine erosion is complex and requires a combination of methods. For clinical evaluation, chemical analysis and optical methods show great potential but are in need of more validation.

Fish movements between aquatic habitats of different salinity ranges (fresh, estuarine, marine) by the tropical catadromous eels Anguilla bicolor bicolor and A. bicolor pacifica were examined by analysing the otolith strontium and calcium concentrations of yellow (immature) and silver (mature) stage eels collected in south-east Asian (Indonesia, Malaysia and Vietnam) waters. The ratios suggest that all migratory-type eels, including freshwater, brackish water and marine residents, pass the river mouth. However, the habitat preference was different among the sites (countries). In Indonesia and Vietnam, most A. bicolor bicolor and A. bicolor pacifica were either marine or brackish water residents in this study. Alternatively, most A. bicolor bicolor were freshwater residents in Malaysia; such a typical catadromous migration pattern in these eels has not been found in previous studies. The wide range of otolith Sr:Ca in both subspecies indicates that the habitat use of these tropical eels was opportunistic among fresh, brackish and marine waters during their growth phases following recruitment to coastal areas. The geographical variability of migratory histories suggests that habitat use might be determined by the inter and intraspecific competition and environmental conditions at each site.

Present research concerns the TL signal stored in chalk of the variety commercially available for writing on blackboards. Samples of this have been subjected to x-ray irradiation, the key dosimetric parameters investigated including dose and energy response, sensitivity, fading and glow curve analysis. Three types of chalk have been investigated, each in five different colours. The samples were annealed at 323 K prior to irradiation. For all three chalk types and all five colours, the dose response has been found linear over the investigated dose range, 0-9 Gy. Regardless of type or colour, photoelectric energy dependency is apparent at the low energy end down to the lowest investigated accelerating potential of 30 kV. Crayola (Yellow) has shown the greatest TL sensitivity, thus selection has been made to limit further analysis to this medium alone, specifically in respect of glow curve and fading study. In addition, elemental compositional and structural change characterizations were made for the same medium, utilizing Energy Dispersive X-Ray (EDX) and Raman spectroscopy, respectively.

BACKGROUND: The extent of calcium volume in the carotid arteries of contrast-based computer tomography (CT) is a valuable indicator of stroke risk. This study presents an automated, simple and fast calcium volume computation system. Since the high contrast agent can sometimes obscure the presence of calcium in the CT slices, it is therefore necessary to identify these slices before the corrected volume can be estimated.

METHODS: The system typically consists of segmenting the calcium region from the CT scan into slices based on Hounsfield Unit-based threshold, and subsequently computing the summation of the calcium areas in each slice. However, when the carotid volume has intermittently higher concentration of contrast agent, a dependable approach is adapted to correct the calcium region using the neighboring slices, thereby estimating the correct volume. Furthermore, mitigation is provided following the regulatory constraints by changing the system to semi-automated criteria as a fall back solution. We evaluate the automated and semi-automated techniques using completely manual calcium volumes computed based on the manual tracings by the neuroradiologist.

RESULTS: A total of 64 patients with calcified plaque in the internal carotid artery were analyzed. Using the above algorithm, our automated and semi-automated system yields correlation coefficients (CC) of 0.89 and 0.96 against first manual readings and 0.90 and 0.96 against second manual readings, respectively. Using the t-test, there was no significant difference between the automated and semi-automated methods against manual. The intra-observer reliability was excellent with CC 0.98.

CONCLUSIONS: Compared to automated method, the semi-automated method for calcium volume is acceptable and closer to manual strategy for calcium volume. Further work evaluating and confirming the performance of our semi-automated protocol is now warranted.

Discriminant analysis of six trace element concentrations measured by instrumental neutron activation analysis (INAA) in 26 paired-samples of malignant and histologically normal human breast tissues shows the technique to be a potentially valuable clinical tool for making malignant-normal classification. Nonparametric discriminant analysis is performed for the data obtained. Linear and quadratic discriminant analyses are also carried out for comparison. For this data set a formal analysis shows that the elements which may be useful in distinguishing between malignant and normal tissues are Ca, Rb and Br, providing correct classification for 24 out of 26 normal samples and 22 out of 26 malignant samples.

Tannic acid (TA) is a phenolic compound that might act directly on osteoblast metabolism. The study was performed to investigate the effects of TA on the proliferation, mineralization, and morphology of human fetal osteoblast cells (hFOB 1.19). The cells were divided into TA-treated, untreated, and pamidronate-treated (control drug) groups. Half maximal effective concentration (EC50 ) values for TA and pamidronate were measured using MTT assay. The EC50 of hFOB 1.19 cells treated with TA was 2.94 M. This concentration was more effective compared to the pamidronate (15.27 M). Cell proliferation assay was performed to compare cell viability from Day 1 until Day 14. The morphology of hFOB 1.19 was observed via inverted microscope and scanning electron microscope. Calcium (Ca) and phosphate (P) were assessed using energy-dispersive X-ray (EDX) analysis. Furthermore, the mineralization of hFOB 1.19 was determined by von Kossa staining (P depositions) and Alizarin Red S staining (Ca depositions). The number of cells treated with TA was significantly higher than the two control groups at Day 10 and Day 14. The morphology of cells treated with TA was uniformly fusiform-shaped with filopodia extensions. Besides, globular-like structures of deposited minerals were observed in the TA-treated group. In line with other findings, EDX spectrum analysis confirmed the presence of Ca and P. The cells treated with TA had significantly higher percentage of both minerals at Day 3 and Day 10 compared to the two control groups. In conclusion, TA enhances cell proliferation and causes cell morphology changes, as well as improved mineralization.

This study was undertaken to determine the effects of varied salinity regimes on the morphological traits (plant height, number of leaves, number of flowers, fresh and dry weight) and major mineral composition of 13 selected purslane accessions. Most of the morphological traits measured were reduced at varied salinity levels (0.0, 8, 16, 24 and 32 dS m(-1)), but plant height was found to increase in Ac1 at 16 dS m(-1) salinity, and Ac13 was the most affected accession. The highest reductions in the number of leaves and number of flowers were recorded in Ac13 at 32 dS m(-1) salinity compared to the control. The highest fresh and dry weight reductions were noted in Ac8 and Ac6, respectively, at 32 dS m(-1) salinity, whereas the highest increase in both fresh and dry weight was recorded in Ac9 at 24 dS m(-1) salinity compared to the control. In contrast, at lower salinity levels, all of the measured mineral levels were found to increase and later decrease with increasing salinity, but the performance of different accessions was different depending on the salinity level. A dendrogram was also constructed by UPGMA based on the morphological traits and mineral compositions, in which the 13 accessions were grouped into 5 clusters, indicating greater diversity among them. A three-dimensional principal component analysis also confirmed the output of grouping from cluster analysis.

Over the past decade, rice (Oryza sativa or Oryza glaberrima) cultivation has increased in many rice-growing countries due to the increasing export demand and population growth and led to a copious amount of rice residues, consisting mainly of rice straw (RS) and rice husk (RH), being generated during and after harvesting. In this study, Eudrilus eugeniae was used to decompose rice residues alone and rice residues amended with cow dung (CD) for bio-transformation of wastes into organic fertilizer. Generally, the final vermicomposts showed increases in macronutrients, namely, calcium (11.4-34.2%), magnesium (1.3-40.8%), phosphorus (1.2-57.3%), and potassium (1.1-345.6%) and a decrease in C/N ratio (26.8-80.0%) as well as increases in heavy metal content for iron (17-108%), copper (14-120%), and manganese (6-60%) after 60 days of vermicomposting. RS as a feedstock was observed to support healthier growth and reproduction of earthworms as compared to RH, with maximum adult worm biomass of 0.66 g/worm (RS) at 60 days, 31 cocoons (1RS:2CD), and 23 hatchlings (1RS:1CD). Vermicomposting of RS yielded better results than RH among all of the treatments investigated. RS that was mixed with two parts of CD (1RS:2CD) showed the best combination of nutrient results as well as the growth of E. eugeniae. In conclusion, vermicomposting could be used as a green technology to bio-convert rice residues into nutrient-rich organic fertilizers if the residues are mixed with CD in the appropriate ratio.

INTRODUCTION: Gynura procumbens has been shown to decrease blood pressure via inhibition of the angiotensinconverting enzyme. However, other mechanisms that may contribute to the hypotensive effect have not been studied.

OBJECTIVES: To investigate the cardiovascular effects of a butanolic fraction of Gynura procumbens in rats.

METHODS: Anaesthetized rats were given intravenous bolus injections of butanolic fraction at doses of 2.5-20 mg/kg in vivo. The effect of butanolic fraction on vascular reactivity was recorded in isolated rat aortic rings in vitro.

    RESULTS: Intravenous administrations of butanolic fraction elicited significant (p < 0.001) and dose-dependent decreases in the mean arterial pressure. However, a significant (p < 0.05) decrease in the heart rate was observed only at the higher doses (10 and 20 mg/kg). In isolated preparations of rat aortic rings, phenylephrine (1 × 10⁻⁶ M)- or potassium chloride (8 × 10⁻² M)-precontracted endothelium-intact and -denuded tissue; butanolic fraction (1 × 10⁻⁶ - 1 × 10⁻¹ g/ml) induced similar concentration-dependent relaxation of the vessels. In the presence of 2.5 × 10⁻³ and 5.0 × 10⁻³ g/ml butanolic fraction, the contractions induced by phenylephrine (1 × 10⁻⁹-3 × 10⁻⁵ M) and potassium chloride (1 × 10⁻² - 8 × 10⁻² M) were significantly antagonized. The calcium-induced vasocontractions (1 × 10⁻⁴-1 × 10⁻²M) were antagonized by butanolic fraction concentration-dependently in calcium-free and high potassium (6×10⁻² M) medium, as well as in calcium- and potassium-free medium containing 1×10⁻⁶ M phenylephrine. However, the contractions induced by noradrenaline (1 × 10⁻⁶ M) and caffeine (4.5 × 10⁻² M) were not affected by butanolic fraction.

CONCLUSION: Butanolic fraction contains putative hypotensive compounds that appear to inhibit calcium influx via receptor-operated and/or voltage-dependent calcium channels to cause vasodilation and a consequent fall in blood pressure.
