    Introduction: Colorectal cancer (CRC) is a major worldwide health problem owing to its high prevalence and mortality rate. Developments in screening, prevention, biomarker, personalized therapies and chemotherapy have improved detection and treatment. However, despite these advances, many patients with advanced metastatic tumors still succumb to the disease. New anticancer agents are needed for treating advanced stage CRC as most of the deaths occur due to cancer metastasis. A recently developed novel sulfonamide derivative 4-((2-(4-(dimethylamino) phenyl)quinazolin-4-yl)amino)benzenesulfonamide (3D) has shown potent antitumor effect; however, the mechanism underlying the antitumor effect remains unknown.

    Materials and methods: 3D-mediated inhibition on cell viability was evaluated by MTT and real-time cell proliferation was measured by xCelligence RTDP instrument. Western blotting was used to measure pro-apoptotic, anti-apoptotic proteins and JAK2-STAT3 phosphorylation. Flow cytometry was used to measure ROS production and apoptosis.

    Results: Our study revealed that 3D treatment significantly reduced the viability of human CRC cells HT-29 and SW620. Furthermore, 3D treatment induced the generation of reactive oxygen species (ROS) in human CRC cells. Confirming our observation, N-acetylcysteine significantly inhibited apoptosis. This is further evidenced by the induction of p53 and Bax; release of cytochrome c; activation of caspase-9, caspase-7 and caspase-3; and cleavage of PARP in 3D-treated cells. This compound was found to have a significant effect on the inhibition of antiapoptotic proteins Bcl2 and BclxL. The results further demonstrate that 3D inhibits JAK2-STAT3 pathway by decreasing the constitutive and IL-6-induced phosphorylation of STAT3. 3D also decreases STAT3 target genes such as cyclin D1 and survivin. Furthermore, a combination study of 3D with doxorubicin (Dox) also showed more potent effects than single treatment of Dox in the inhibition of cell viability.

    Conclusion: Taken together, these findings indicate that 3D induces ROS-mediated apoptosis and inhibits JAK2-STAT3 signaling in CRC.

    Landfill leachate is one of the most recalcitrant wastes for biotreatment and can be considered a potential source of contamination to surface and groundwater ecosystems. In the present study, Fenton oxidation was employed for degradation of stabilized landfill leachate. Response surface methodology was applied to analyze, model and optimize the process parameters, i.e. pH and reaction time as well as the initial concentrations of hydrogen peroxide and ferrous ion. Analysis of variance showed that good coefficients of determination were obtained (R2 > 0.99), thus ensuring satisfactory agreement of the second-order regression model with the experimental data. The results indicated that, pH and its quadratic effects were the main factors influencing Fenton oxidation. Furthermore, antagonistic effects between pH and other variables were observed. The optimum H2O2 concentration, Fe(II) concentration, pH and reaction time were 0.033 mol/L, 0.011 mol/L, 3 and 145 min, respectively, with 58.3% COD, 79.0% color and 82.1% iron removals.

    This study investigated the electrochemical oxidation of stabilized leachate from Pulau Burung semi-aerobic sanitary landfill by conducting laboratory experiments with sodium sulfate Na(2)SO(4) (as electrolyte) and graphite carbon electrodes. The control parameters were influent COD, current density and reaction time, while the responses were BOD removal, COD removal, BOD:COD ratio, color and pH. Na(2)SO(4) concentration was 1 g/L. Experiments were conducted based on a three-level factorial design and response surface methodology (RSM) was used to analyze the results. The optimum conditions were obtained as 1414 mg/L influent COD concentration, 79.9 mA/cm(2) current density and 4 h reaction time. This resulted in 70% BOD removal, 68% COD removal, 84% color removal, 0.04 BOD/COD ratio and 9.1 pH. Electrochemical treatment using graphite carbon electrode was found to be effective in BOD, COD and color removal but was not effective in increasing the BOD/COD ratio or enhancing biodegradability of the leachate. The color intensity of the treated samples increased at low influent COD and high current density due to corrosion of electrode material.

    This study proposed an oil palm by-product as a low-cost adsorbent for the removal of hexavalent chromium [Cr (VI)] from aqueous solution. Adsorption of Cr (VI) by sulphuric acid and heat-treated oil palm fibre was conducted using batch tests. The influence of pH, contact time, initial chromium concentration and adsorbent dosage on the removal of Cr (VI) from the solutions was investigated. The optimum initial pH for maximum uptake of Cr (VI) from aqueous solution was found to be 1.5. The removal efficiency was found to correlate with the initial Cr (VI) concentration, adsorbent dosage as well as the contact time between Cr (VI) and the adsorbent. The adsorption kinetics tested with pseudo first order and pseudo second order models yielded high R(2) values from 0.9254 to 0.9870 and from 0.9936 to 0.9998, respectively. The analysis of variance (ANOVA) showed significant difference between the R(2) values of the two models at 99% confidence level. The Freundlich isotherm (R(2)=0.8778) described Cr (VI) adsorption slightly better than the Langmuir isotherm (R(2)=0.8715). Difficulty in desorption of Cr (VI) suggests the suitability of treated oil palm fibre as a single-use adsorbent for Cr (VI) removal from aqueous solution.

    In order to develop a potent anti-tumor agent that can target both cancer stem cells and the bulk of tumor cells, a series of 2-((benzimidazol-2-yl)thio)-1-arylethan-1-ones 5a-o was synthesized. All compounds were evaluated for their anti-proliferative activity towards colon HT-29 cancer cell line. In addition, their inhibitory effect against cell surface expression of CD133, a potent cancer stem cells (CSCs) marker, in the same cells was evaluated by flow cytometry at 10 μM. Compound 5l emerged as the most active anti-proliferative analog against HT-29 (IC50 = 18.83 ± 1.37 μM), that almost equipotent as 5-fluorouracil (IC50 = 15.83 ± 1.63 μM) with 50.11 ± 4.05% inhibition effect on CD133 expression, suggested dual targeted effect. Also, compounds 5h, 5j, 5k and 5m-o inhibited the expression of CD133 with more than 50%. The SAR study pointed out the significance of substitution of the pendent phenyl group with lipophilic electron-donating groups or replacing it by 2-thienyl or 2-furyl groups.

    The emergence of colorectal cancer in developed nations can be attributed to dietary habits, smoking, a sedentary lifestyle and obesity. Several treatment regimens are available for primary and metastatic colorectal cancer; however, these treatment options have had limited impact on cure and disease‑free survival, and novel agents need to be developed for treating colorectal cancer. Thus, the objective of this study was to explore the anticancer mechanism of a benzo(1,3)dioxol‑based derivative of sulfonamide. The compound's inhibitory effect on cell proliferation was determined using the MTT assay and the xCelligence RTDP machine. Alternations in the expression of Bcl‑2 and inhibitor of apoptosis protein families were detected by western blotting. Apoptotic marker protein expression, including cytochrome c and cleaved poly(ADP‑ribose)polymerase was measured in the cytosolic extract of cells. Apoptosis and necrosis were detected by flow cytometry and immunofluorescence. Reactive oxygen species (ROS), and activation of caspase‑3 and caspase‑7 were measured using flow cytometry. Activation of the JNK pathway was detected by western blotting. We investigated the molecular mechanism of action of the sulfonamide derivative on colorectal cancer cells and found that the compound possesses a potent anticancer effect, which is primarily exerted by inducing apoptosis and necrosis. Interestingly, this compound exhibited little antiproliferative effect against the normal colonic epithelial cell line FHC. Furthermore, our results showed that the compound could significantly increase ROS production. Apoptosis induction could be attenuated by the free oxygen radical scavenger N‑acetyl cysteine (NAC), indicating that the antiproliferative effect of this compound on colorectal cancer cells is at least partially dependent on the redox balance. In addition, JNK signaling was activated by treatment with this derivative, which led to the induction of apoptosis. On the contrary, a JNK inhibitor could suppress the cell death induced by this compound. Our findings thus suggested a novel anticancer mechanism of a benzo(1,3)dioxol‑based derivative of sulfonamide for colorectal cancer cells and may have therapeutic potential for the treatment of colorectal cancer; however, further investigation is required.

    Wastewater treatment is a key challenge in the textile industry. The current treatment methods for textile wastewater are insufficient or ineffective for complex dyes generated from the textile industry. This study evaluated the performances of two novel inorganic coagulants with high cationic charges, namely, titanium tetrachloride (TiCl4) and zirconium tetrachloride (ZrCl4). They were utilised to treat textile industry wastewater. Both coagulation processes were performed under the same experimental operational conditions. Turbidity, suspended solids (SS), colour, chemical oxygen demand (COD) and ammonia were measured to assess the efficiencies of the coagulants. Results indicated that ZrCl4 and TiCl4 exhibited high potentials for textile wastewater treatment. ZrCl4 presented high removal efficiency in COD and SS, whereas TiCl4 showed excellent removal in ammonia.

    Sequencing batch reactor (SBR) is one of the various methods of biological treatments used for treating wastewater and landfill leachate. This study investigated the treatment of landfill leachate and domestic wastewater by adding a new adsorbent (powdered ZELIAC; PZ) to the SBR technique. ZELIAC consists of zeolite, activated carbon, lime stone, rice husk ash, and Portland cement. The response surface methodology and central composite design were used to elucidate the nature of the response surface in the experimental design and describe the optimum conditions of the independent variables, including aeration rate (L/min), contact time (h), and ratio of leachate to wastewater mixture (%; v/v), as well as their responses (dependent variables). Appropriate conditions of operating variables were also optimized to predict the best value of responses. To perform an adequate analysis of the aerobic process, four dependent parameters, namely, chemical oxygen demand (COD), color, ammonia-nitrogen (NH3-N), and phenols, were measured as responses. The results indicated that the PZ-SBR showed higher performance in removing certain pollutants compared with SBR. Given the optimal conditions of aeration rate (1.74 L/min), leachate to wastewater ratio (20%), and contact time (10.31 h) for the PZ-SBR, the removal efficiencies for color, NH3-N, COD, and phenols were 84.11%, 99.01%, 72.84%, and 61.32%, respectively.

    This study analyzes and compares the results of leachate composition at the semi-aerobic Pulau Burung Landfill Site (PBLS) (unaerated pond and intermittently aerated pond) and the anaerobic Kulim Sanitary Landfill in the northern region of Malaysia. The raw samples were collected and analyzed for twenty parameters. The average values of the parameters such as phenols (1.2, 6.7, and 2.6 mg/L), total nitrogen (448, 1200, and 300 mg/L N-TN), ammonia-N (542, 1568, and 538 mg/L NH(3)-N), nitrite (91, 49, and 52 mg/L NO(2)(-)-N), total phosphorus (21, 17, and 19 mg/L), BOD(5) (83, 243, and 326 mg/L), COD (935, 2345, and 1892 mg/L), BOD(5)/COD (0.096,0.1124,0.205%), pH (8.20, 8.28, and 7.76), turbidity (1546, 180, and 1936 Formazin attenuation units (FAU)), and color (3334, 3347, and 4041 Pt Co) for leachate at the semi-aerobic PBLS (unaerated and intermittently aerated) and the anaerobic Kulim Sanitary Landfill were recorded, respectively. The obtained results were compared with previously published data and data from the Malaysia Environmental Quality Act 1974. The results indicated that Pulau Burung leachate was more stabilized compared with Kulim leachate. Furthermore, the aeration process in PBLS has a considerable effect on reducing the concentration of several pollutants. The studied leachate requires treatment to minimize the pollutants to an acceptable level prior to discharge into water courses.

    Hydrocarbon pollution in marine ecosystems occurs mainly by accidental oil spills, deliberate discharge of ballast waters from oil tankers and bilge waste discharges; causing site pollution and serious adverse effects on aquatic environments as well as human health. A large number of petroleum hydrocarbons are biodegradable, thus bioremediation has become an important method for the restoration of oil polluted areas. In this research, a series of natural attenuation, crude oil (CO) and dispersed crude oil (DCO) bioremediation experiments of artificially crude oil contaminated seawater was carried out. Bacterial consortiums were identified as Acinetobacter, Alcaligenes, Bacillus, Pseudomonas and Vibrio. First order kinetics described the biodegradation of crude oil. Under abiotic conditions, oil removal was 19.9% while a maximum of 31.8% total petroleum hydrocarbons (TPH) removal was obtained in natural attenuation experiment. All DCO bioreactors demonstrated higher and faster removal than CO bioreactors. Half life times were 28, 32, 38 and 58 days for DCO and 31, 40, 50 and 75 days for CO with oil concentrations of 100, 500, 1000 and 2000 mg/L, respectively. The effectiveness of Corexit 9500 dispersant was monitored in the 45 day study; the results indicated that it improved the crude oil biodegradation rate.

    Elephantopus tomentosus is widely used in Asia, especially in Malaysia, for the treatment of pain and inflammation. In the present study, the analgesic and anti-inflammatory effects of a 95% ethanol extract of E. tomentosus were investigated in different experimental models. In the anti-inflammation study, 1000 mg/kg of extract significantly reduced carrageenan-induced hind paw edema (p < 0.05) and inhibited abdominal permeability compared with control (p < 0.01). The analgesic activity was assayed in several experimental models in mice: (1) hot plate, (2) tail flick, (3) writhing test; and rats: carrageenan-induced hyperalgesia pain threshold test. However, at the doses tested, no significant activity was found in the hot plate test and the tail flick test. E. tomentosus ethanol extract at 1000 mg/kg significantly (p < 0.05) increased hyperalgesia pain threshold and inhibited writhing activity. The results suggest that E. tomentosus ethanol extract at 1000 mg/kg dose is effective in anti-inflammatory and non-steroidal anti-inflammatory drug type anti-nociception activities.

    The treatability of stabilized sanitary landfill leachate via synthetic anion exchange resin (INDION FFIP MB) was investigated. An ideal experimental design was conducted based on central composite design using a response surface methodology to assess individual and interactive effects of critical operational variables (i.e., anionic dosage; contact time; shaking speed) and pH on treatment performance in terms of color, chemical oxygen demand (COD), suspended solid (SS), and turbidity removal efficiencies. Optimum operational conditions were established as 30.9 cm(3) anionic dosage, 90 min contact time, 150 rpm shaking speed, and pH 3.1. Under these conditions, the color, COD, SS, and turbidity removal efficiencies of 91.5, 70.3, 93.1, and 92.4% were experimentally attained and were found to fit well with the prediction model. According to these results, stabilized leachate treatment using INDION FFIP MB could be an effective alternative in the administration of color, COD, SS, and turbidity problems of landfill leachates.

    This study evaluated the effectiveness of Fenton's technique for the treatment of semi-aerobic landfill leachate collected from Pulau Burung Landfill Site (PBLS), Penang, Malaysia. The Fe2+ or Fe3+ as catalyst and H2O2 as oxidizing agent are commonly used for the classical Fenton's reaction. In present study, the effect of operating conditions such as pH, reaction time, molar ratio, agitation rate, feeding mode and Fenton reagent concentrations which are important parameters that affect the removal efficiencies of Fenton method were investigated. Under the most favorable conditions, the highest removals of 58.1 and 78.3% were observed for COD and color, respectively. In general, the best operating conditions were pH = 3, Fe = 560 mg L(-1), H2O2 = 1020 mg L(-1), H2O2/Fe2+ molar ratio = 3, agitation rate = 400 rpm and reaction time = 120 minutes. The results highlighted that stepwise addition of Fenton's reagent was more effective than adding the entire volume in a single step. Excessive hydrogen peroxide and iron have shown scavenging effects on hydroxyl radicals and reduced degradation of refractory organics in the landfill leachate.

    Response surface methodology (RSM) was employed to optimize nitrogen and phosphorus concentrations for removal of n-alkanes from crude oil contaminated seawater samples in batch reactors. Erlenmeyer flasks were used as bioreactors; each containing 250 mL dispersed crude oil contaminated seawater, indigenous acclimatized microorganism and different amounts of nitrogen and phosphorus based on central composite design (CCD). Samples were extracted and analyzed according to US-EPA protocols using a gas chromatograph. During 28 days of bioremediation, a maximum of 95% total aliphatic hydrocarbons removal was observed. The obtained Model F-value of 267.73 and probability F<0.0001 implied the model was significant. Numerical condition optimization via a quadratic model, predicted 98% n-alkanes removal for a 20-day laboratory bioremediation trial using nitrogen and phosphorus concentrations of 13.62 and 1.39 mg/L, respectively. In actual experiments, 95% removal was observed under these conditions.

    Landfill leachate is one of the major contamination sources. In this study, the ability of synthetic ion exchange resins which carry different mobile ion for removing color, chemical oxygen demand (COD), and ammonia nitrogen (NH(3)-N) from stabilized leachate was investigated. The synthetic resin INDION 225 Na as a cationic exchanger and INDION FFIP MB as an anionic exchanger were used in this study. INDION 225 Na was used in hydrogen form (H(+)) and in sodium form (Na(+)), while INDION FFIP MB resin was used in hydroxide form (OH(-)) and in calcium form (Cl(-)) form. The results indicated better removal of color, COD and NH(3)-N by using INDION 225 Na in H(+) as compared with Na(+) form, while no performance differences were observed by using INDION FFIP MB in OH(-) or Cl(-) form. Applying cationic resin followed by anionic resin achieved 97, 88 and 94, percent removal of color, COD and NH(3)-N. The residual amounts were 160 Pt-Co, 290 mg/L and 110 mg/L of color, COD and NH(3)-N respectively.

    Mature landfill leachate is typically non-biodegradable and contains high concentration of refractory organics. The aim of this research was to optimize operating parameters in electro-Fenton process, for the removal of recalcitrant organics from semi-aerobic landfill leachate using response surface methodology (RSM). Effectiveness of important process parameters H(2)O(2)/Fe(2+) molar ratio, current density, pH and reaction time were determined, optimized and modeled successfully. Significant quadratic polynomial models were obtained (R(2)=0.9972 and 0.9984 for COD and color removals, respectively). Numerical optimization based on desirability function were employed; in a 43 min trial 94.07% of COD and 95.83% of color were removed at pH 3 and H(2)O(2)/Fe(2+) molar ratio 1, while current density was 49 mA/cm(2). The results indicate that E-Fenton process was an effective technology for semi-aerobic landfill leachate treatment.

    The performance of a carbon-mineral composite adsorbent used in a fixed bed column for the removal of ammoniacal nitrogen and aggregate organic pollutant (COD), which are commonly found in landfill leachate, was evaluated. The breakthrough capacities for ammoniacal nitrogen and COD adsorption were 4.46 and 3.23 mg/g, respectively. Additionally, the optimum empty bed contact time (EBCT) was 75 min. The column efficiency for ammoniacal nitrogen and COD adsorption using fresh adsorbent was 86.4% and 92.6%, respectively, and these values increased to 90.0% and 93.7%, respectively, after the regeneration process.

    In the present study, Electrochemical Oxidation was used to remove COD and color from semi-aerobic landfill leachate collected from Pulau Burung Landfill Site (PBLS), Penang, Malaysia. Experiments were conducted in a batch laboratory-scale system in the presence of NaCl as electrolyte and aluminum electrodes. Central composite design (CCD) under Response surface methodology (RSM) was applied to optimize the electrochemical oxidation process conditions using chemical oxygen demand (COD) and color removals as responses, and the electrolyte concentrations, current density and reaction time as control factors. Analysis of variance (ANOVA) showed good coefficient of determination (R(2)) values of >0.98, thus ensuring satisfactory fitting of the second-order regression model with the experimental data. In un-optimized condition, maximum removals for COD (48.77%) and color (58.21%) were achieved at current density 80 mA/cm(2), electrolyte concentration 3,000 mg/L and reaction time 240 min. While after optimization at current density 75 mA/cm(2), electrolyte concentration 2,000 mg/L and reaction time 218 min a maximum of 49.33 and 59.24% removals were observed for COD and color respectively.

    A study was conducted to investigate the efficiency of coagulation and flocculation processes for removing colour from a semi-aerobic landfill leachate from one of the landfill sites in Malaysia. Four types of coagulant namely aluminium (III) sulphate (alum), ferric (III) chloride, ferrous (II) sulphate and ferric (III) sulphate were studied using standard jar test apparatus. Results indicated that ferric chloride was superior to the other coagulants and removed 94% of colour at an optimum dose of 800 mg/l at pH 4. The effect of coagulant dosages on colour removal showed similar trend as for COD, turbidity and suspended solids. This suggested that colour in landfill leachate was mainly contributed by organic matters with some insoluble forms that exhibited turbidity and suspended solids readings. The results from this study suggested that ferric chloride could be a viable coagulant in managing colour problems associated with landfill leachate.

    The purpose of the present study was to examine the removal of ammoniacal nitrogen (NH4-N) from synthetic wastewater using limestone (LS) and granular activated carbon (GAC) mixture as low cost adsorbent. In batch study, optimum shaking and settling times were 150 and 120 min, respectively. The LS-GAC mixture ratio of 25:15 removed about 58% NH4-N. The smaller particle size of medium yielded higher adsorption capacity. The equilibrium adsorption data followed the Freundlich isotherm (R2 > 0.98) but it showed weak bond. Adsorption kinetics were well described by the pseudo second-order rate model (R2 > 0.93). The upflow column showed that higher flow rate and initial concentration resulted in shorter column saturation time. The study showed that the usage of GAC could be reduced by combining GAC with LS for the removal of NH4-N from wastewater; thus reducing the cost of treatment.
