    BACKGROUND: Microalgae provide an excellent platform for the production of high-value-products and are increasingly being recognised as a promising production system for biomass, animal feeds and renewable fuels.

    RESULTS: Here, we describe an automated screen, to enable high-throughput optimisation of 12 nutrients for microalgae production. Its miniaturised 1,728 multiwell format allows multiple microalgae strains to be simultaneously screened using a two-step process. Step 1 optimises the primary elements nitrogen and phosphorous. Step 2 uses Box-Behnken analysis to define the highest growth rates within the large multidimensional space tested (Ca, Mg, Fe, Mn, Zn, Cu, B, Se, V, Si) at three levels (-1, 0, 1). The highest specific growth rates and maximum OD750 values provide a measure for continuous and batch culture.

    CONCLUSION: The screen identified the main nutrient effects on growth, pairwise nutrient interactions (for example, Ca-Mg) and the best production conditions of the sampled statistical space providing the basis for a targeted full factorial screen to assist with optimisation of algae production.

    In Peninsular Malaysia 'structural' factors are found to influence strongly people's persistent occupation of floodplains. Thus, despite a high level of flood hazard awareness, a high level of pessimism and a high level of expectation of future floods, poorer individuals seldom attempt to leave for more advantageous locations but are instead trapped in their present locations by structural factors such as poverty, low residential and occupational mobility, low educational attainment, traditional land inheritance, government aid, and government disaster preparedness, relief and rehabilitation programmes. These forces exert a strong influence upon individuals and largely control their choice of residential location in response to flood hazards, thereby reinforcing the persistent occupation of floodplains. Structural factors such as landlessness, rural-urban migration, floodplain encroachment and squatting are also highly influential in leading people to move. Even for those who move, structural factors have largely confined their choice of residential location to urban floodplains.

    Institutional aspects of flood hazards significantly affect their outcomes in Malaysia. Institutional arrangements to deal with floods include: legislative activity, organisational structures, attitudes and sub-culture, and policies and instruments. When assessed in terms of four specific criteria, institutional aspects of flood hazards are found to be largely inadequate. Disaster reduction programmes are over-dependent on a reactive approach based largely on technology and not even aimed at floods specifically. Structural flood reduction measures are the predominant management tool and, although the importance of non-structural measures is recognised, thus far they have been under-employed. Current laws and regulations with regard to flood management are also insufficient and both the financial and human resources of flood hazard organisations are generally found to be wanting. Finally, economic efficiency, equity and public accountability issues are not adequately addressed by institutional arrangements for flood hazards.

    Eleven Malaysian Plasmodium falciparum isolates were cultured in vitro and later subjected to antimalarial evaluations in 96-well microtiter plates. After cryopreservation, the IC50 (nM) for ST 195, ST 196, ST 197, ST 244 and ST 245 isolates were, respectively: 180.9, 198.7, 482.0, 580.0 and 690.1 for chloroquine; 3.4, 3.4, 9.2, 4.0 and 5.8 for mefloquine; 21.9, 10.5, 40.7, 40.1 and 48.7 for quinine; 136.7, 58.8, 116.4, 29.4 and 95.4 for cycloguanil, and 48.3, 57.5, 47.4, 61.5 and 37.8 for pyrimethamine. Before cryopreservation they were 172.5, 141.5, 453.2, 636.0 and 651.6 nM for chloroquine; 4.8, 2.6, 9.0, 6.9 and 5.8 nM for mefloquine; 21.3, 8.3, 41.9, 49.6 and 40.1 nM for quinine, 129.9, 47.3, 109.3, 30.6 and 95.4 nM for cycloguanil, and 45.4, 47.4, 40.2, 66.3 and 36.0 nM for pyrimethamine. IC50 (nM) for Gombak A, Gombak C, ST 9, ST 12, ST 85 and ST 148 isolates after 12 months of continuous in vitro culture were, respectively: 477.0, 492.3, 367.1, 809.4, 566.5 and 341.8 for chloroquine; 2.9, 11.1, 8.5, 16.9, 5.3 and 4.2 for mefloquine; 6.2, 58.3, 52.7, 36.7, 31.8 and 26.2 for quinine; 154.5, 57.2, 130.3, 94.2, 81.4 and 102.9 for cycloguanil, 26.9, 24.9, 43.8, 31.0, 14.1 and 56.7 for pyrimethamine. Before the 12-month culture they were 472.3, 452.9, 352.7, 773.7, 702.7 and 322.7 nM for chloroquine; 2.6, 13.2, 8.5, 17.2, 5.0 and 4.0 nM for mefloquine; 6.2, 85.4, 53.9, 38.5, 35.8 and 38.5 nM for quinine; 106.8, 74.3, 112.4, 89.8, 91.8 and 103.3 nM for cycloguanil, and 26.9, 31.4, 47.0, 28.1, 14.9 and 56.7 nM for pyrimethamine. Thus none of these isolates differed in their original susceptibilities after either of these procedures.

    Enucleation and evisceration are the most common surgical procedures that are performed to manage tumor, trauma, and infection. Given the consequences of surgical intervention, the conditions of the remaining eye socket may affect future prosthetic rehabilitation. A custom-made ocular prosthesis can be used to help restore the esthetics and functional defects and to improve the quality of life of patients with such conditions. An assessment must be performed on the prosthetic outcome before rehabilitation. The etiology of defect, type of surgery, condition of the remaining socket, and patient's age should all be considered. This report discusses three different etiological eye defects that have undergone enucleation and evisceration and describes the factors that have a significant role in the esthetic and functional outcome of the prosthesis. This report should serve as a helpful aid for maxillofacial prosthodontists to understand the primary objective of rehabilitating each eye defect and to meet patient expectations.

    We present a new set of subjective Age of Acquisition (AoA) ratings for 299 words (158 nouns, 141 verbs) in seven languages from various language families and cultural settings: American English, Czech, Scottish Gaelic, Lebanese Arabic, Malaysian Malay, Persian, and Western Armenian. The ratings were collected from a total of 173 participants and were highly reliable in each language. We applied the same method of data collection as used in a previous study on 25 languages which allowed us to create a database of fully comparable AoA ratings of 299 words in 32 languages. We found that in the seven languages not included in the previous study, the words are estimated to be acquired at roughly the same age as in the previously reported languages, i.e. mostly between the ages of 1 and 7 years. We also found that the order of word acquisition is moderately to highly correlated across all 32 languages, which extends our previous conclusion that early words are acquired in similar order across a wide range of languages and cultures.

    Three-point bending test is one of the main methods used in long bones to characterise bone material and determine the biomechanical properties. We have examined the mechanical competencies of the mouse bones at four-week-old by using a three-point bending jig so that the potential genotype-related deficiencies in mechanical properties of bones explored. The available bending jig was not suitable for small animal model and may cause slippage when applying the load. The tibial gross length measurements of the four-week-old mouse measured using the proximal anatomical point of the centre of the condyles to the distal anatomical significance of the medial malleolus (~16 mm). The mid tibia diameter measurement is taken at the middle tibia (~1 mm) and metaphyseal diameter (~3 mm). The bending jig was custom-made, where both ends support were cut in a v-shape to provide stability. The tibias were mechanically tested with the v-shape support under three-point bending using a Bose ElectroForce® 3200 until failure. The test revealed a significant result of flexural strength, work-to-fracture and strain to failure obtained from the load-displacement curves. The finding may be useful in the studies of quantitative assessments of the strength and toughness of small animal bones.

    Bacteriocin-like inhibitory substances (BLIS) produced by Lactococcus lactis Gh1 had shown antimicrobial activity against Listeria monocytogenes ATCC 15313. Brain Heart Infusion (BHI) broth is used for the cultivation and enumeration of lactic acid bacteria, but there is a need to improve the current medium composition for enhancement of BLIS production, and one of the approaches is to model the optimization process and identify the most appropriate medium formulation. Response surface methodology (RSM) and artificial neural network (ANN) models were employed in this study. In medium optimization, ANN (R2 = 0.98) methodology provided better estimation point and data fitting as compared to RSM (R2 = 0.79). In ANN, the optimal medium consisted of 35.38 g/L soytone, 16 g/L fructose, 3.25 g/L sodium chloride (NaCl) and 5.40 g/L disodium phosphate (Na2HPO4). BLIS production in optimal medium (717.13 ± 0.76 AU/mL) was about 1.40-fold higher than that obtained in nonoptimised (520.56 ± 3.37 AU/mL) medium. BLIS production was further improved by about 1.18 times higher in 2 L stirred tank bioreactor (787.40 ± 1.30 AU/mL) as compared to that obtained in 250 mL shake flask (665.28 ± 14.22 AU/mL) using the optimised medium.

    Introduction: Thymoquinone (TQ) is the main active compound extracted from Nigella sativa a traditional herb with wide therapeutic applications and recognizable anticancer properties. This study aimed to formulate and characterize TQ-nanoparticles using PLGA as a biocompatible coating material (TQ-PLGA NPs) with the evaluation of its therapeutic properties in human melanoma cancer cells.

    Methods: The TQ-PLGA NPs were prepared and characterized for size, zeta potential, encapsulation efficiency, and release profile.

    Results: The particle size was 147.2 nm, with 22.1 positive zeta potential and 96.8% encapsulation efficiency. The NPs released 45.6% of the encapsulated TQ within 3 h followed by characteristic sustained release over 7 days with a total of 69.7% cumulative release. TQ-PLGA NPs were taken up effectively by the cells in a time-dependent manner up to 24 h. Higher cell toxicity was determined within the first 24 h in melanoma cells due to the rapid release of TQ from the NPs and its low stability in the cell culture media.

    Conclusion: TQ-PLGA NPs is a potential anticancer agent taking advantage of the sustained release and tailored size that allows accumulation in the cancer tissue by the enhanced permeability and retention effect. However, stability problems of the active ingredient were address in this study and requires further investigation.

    Group B streptococcus (GBS) is a rare cause of meningitis in adults that commonly affects patients with multiple underlying comorbidities. Although it is uncommon, it typically progresses very rapidly and has a high mortality rate as compared to other causes of bacterial meningitis. Here, we report a patient with GBS meningitis who had no underlying medical illness and presented with multiple episodes of seizure within hours of developing fever. Cerebrospinal fluid analysis results were consistent with bacterial meningitis, and blood cultures grew GBS. She was treated with intravenous ceftriaxone for 2 weeks and made a great recovery without any sequalae. In conclusion, although GBS meningitis is uncommon in adults, it is a serious medical disease and associated with a high mortality rate. To the best of our knowledge, this patient represents one of the few reported cases of GBS meningitis in a previously healthy young adult.

    Treatment of osteoarthritis (OA) is still a major clinical challenge due to the limited inherent healing capacity of cartilage. Recent studies utilising stem cells suggest that the therapeutic benefits of these cells are mediated through the paracrine mechanism of bioactive molecules. The present study evaluates the regenerative effect of stem cells from human exfoliated deciduous teeth (SHED) conditioned medium (CM) on OA chondrocytes. The CM was collected after the SHED were cultured in serum-free medium (SFM) for 48 or 72 h and the cells were characterised by the expression of MSC and pluripotency markers. Chondrocytes were stimulated with interleukin-1β and treated with the CM. Subsequently, the expression of aggrecan, collagen type 2 (COL 2), matrix metalloproteinase-13 (MMP-13), nuclear factor-kB (NF-kB) and the level of inflammatory and anti-inflammatory markers were evaluated. SHED expressed mesenchymal stromal cell surface proteins but were negative for haematopoietic markers. SHED also showed protein expression of NANOG, OCT4 and SOX2 with differential subcellular localisation. Treatment of OA chondrocytes with CM enhanced anti-inflammation compared to control cells treated with SFM. Furthermore, the expression of MMP-13 and NF-kB was significantly downregulated in stimulated chondrocytes incubated in CM. The study also revealed that CM increased the expression of aggrecan and COL 2 in OA chondrocytes compared to SFM control. Both CM regenerate extracellular matrix proteins and mitigate increased MMP-13 expression through inhibition of NF-kB in OA chondrocytes due to the presence of bioactive molecules. The study underscores the potential of CM for OA treatment.

    In this work, we proposed a facile approach to fabricate a superhydrophobic surface for anti-icing performance in terms of adhesive strength and freezing time. A hierarchical structure was generated on as-received Al plates using a wet etching method and followed with a low energy chemical compound coating. Surfaces after treatment exhibited the great water repellent properties with a high contact angle and extremely low sliding angle. An anti-icing investigation was carried out by using a custom-built apparatus and demonstrated the expected low adhesion and freezing time for icephobic applications. In addition, we proposed a model for calculating the freezing time. The experimented results were compared with theoretical calculation and demonstrated the good agreement, illustrating the importance of theoretical contribution in design icephobic surfaces. Therefore, this study provides a guideline for the understanding of icing phenomena and designing of icephobic surfaces.

    Introduction: This study reported the concentration of bacterial and fungal bioaerosol at an animal house and hospi- tal laboratories with the aim to compare the concentration levels at library and administrative offices. The bioaerosol levels between mid-shift (afternoon) were also compared to the concentration measured during pre-shift (morning). Methods: The NIOSH 0800 method utilising microbiological air sampler collecting airborne bacterial and fungal samples via impaction technique on Nutrient agar (NA) and Sabouraud Dextrouse agar (SDA) as culture medium, respectively. Sampling was done twice daily; before (pre-shift) and during working (mid-shift) hour. Results: The highest bacteria and fungi concentration was recorded at the animal house with median concentration of 2477 CFU/ m3 (IQR=121-2477) and 791 CFU/m3 (IQR = 379-2081), respectively.     In this study, two native duckweeds (Lemna minor and Azolla pinnata) were cultivated in Palm Oil Mill Effluent (POME) to extract nutrients from the effluent. Five grams of A. pinnata and 2 g of L. minor were transferred to 2 L POME (Initial concentrations: 198 mg/L COD, 4.3 mg/L nitrates, pH 9.53, 4 mg/L phosphate, 2.98 mg/L ammonia) with four different dilutions (2.5%, 5%, 10%, 15%) under greenhouse conditions. Samples of POME were taken every two days up to 10 days. Growth parameter, phosphate, ammonia, nitrates, pH, and COD were monitored within 10 days to select the most suitable growth medium for both plants. Results showed that 2.5% POME dilution had positive effect on L. minor growth and A. pinnata (wet weight increased by 8.7 g and 9.8 g, respectively), with all plants able to survive until the final day of exposure. The highest removal of ammonia was accomplished in 5% POME dilution by A. pinnata (98%) and L. minor (95.5%). The maximum phosphate removal was obtained in 10% POME dilution with 93.3% removal by A. pinnata and 86.7% by L. minor. Significant COD removal in 15% POME was obtained by L. minor (78%) and A. pinnata (66%). Both plants responded positively to the phytoremediation process, especially for A. pinnata which showed significant decreases in all parameters. The nutrient extraction by both plants from POME showed a positive effect on growth parameter, which has further promising potential to be used as animal feedstock.

    A batch culture was enriched on phenol with trichloroethene-contaminated aquifer soil as an inoculum. Cupriavidus sp. strain P-10 was isolated from the culture using a diluted plating method. Here, we report the draft genome sequence and annotation of strain P-10, which provides insights into the metabolic processes of phenol degradation.

    Stem cells isolated from dental pulp possess the capacity for self-renewal and the potential for multi-lineage differentiation. However, dental pulp stem cells have different characteristics in terms of their culture conditions. The success of stem cells culture is governed by its micro-environmental niche. Therefore, we studied the effects of culture niche on long-term expansion of dental pulp stem cells in terms of cell morphology, growth kinetics, senescence pattern, cell surface marker expression differentiation capacity, and seeding plating density of dental pulp stem cells in four different, widely used media composition Among the various basal media tested, α-minimum essential media and knock out-minimum essential media supplemented with 10% fetal bovine serum were found to be the most optimal media composition in preserving the phenotypic characteristics and differentiation potential for prolonged periods as compared with DMEM-F12 and DMEM-LG. Plating density has been shown to affect overall yield. As a conclusion, the adoption of an appropriate culture system significantly improved cell yield, thus enabling the attainment of sufficient yields for therapeutic applications economizing in terms of cost of production and minimizing seeding cell density for maximum yield.

    The in vitro shoot proliferation of endemic Begonia pavonina in three culture conditions i.e semisolid medium (SM), liquid culture medium (LM) and in temporary immersion bioreactor system (RITA®) was analyzed in this study. To minimize contamination rates, seeds were surface sterilized and cultured on MS basal media. The clean raised shoots were then used as explants for inoculation onto the tested culture conditions. In this experiment, the explants were maintained in MS medium supplemented with 0.1mgL-1 BAP for shoot multiplication. After 4 weeks of incubation, higher regeneration rates were observed in TIM as compared to other medium conditions. The maximum shoot number was obtained from TIM system with a mean of 5.30 shoots per explant, followed by LM (2.47 shoots per explant) and SM (1.2 shoots per explant). Shoot hyperhydration was also lowest in a TIM system. Overall, TIM was shown to produce higher shoot multiplications combined with healthy morphological characteristics of plantlets. Shoot cultures from the all cultures were successfully rooted in vitro and acclimatized well in the greenhouse.
