    A structured weight management programme at a workplace may help in reducing the prevalence of overweight and obesity.
    Therefore, this intervention study was to determine the effectiveness of weight loss programmes including face-to-face,
    online and control group at workplace among employees who are overweight and obese. A total of 108 overweight and
    obese adults were recruited and randomly divided into three groups (face-to-face group (FT), n=38; online group (OG),
    n=31; control group (CG), n=39). In the FT group, the participants took part in health talks, interactive activities and
    counselling; the OG group was given access to an online weight management program and the CG group was provided
    with educational booklets on weight loss. All information given was related to nutrition, physical activity and motivation
    to reduce weight. Body weight, body mass index (BMI), waist circumference (WC), body fat percentage, dietary intake,
    fasting lipid profile and glucose levels were assessed at baseline and 4 months. The FT group showed greater reduction
    in body weight (-5.80 kg) compared to OG (-1.12 kg) and CG (-1.82 kg). Significant interaction effects were found for BMI,
    WC, fasting serum triglycerides, HDL-cholesterol and total cholesterol/HDL-cholesterol ratio (all p<0.05), with the FT
    group showing the biggest improvements, compared to the other groups. The face-to-face weight management program
    offered in the workplace showed to be the most effective at improving anthropometric profile, fasting serum triglycerides,
    HDL-C, total cholesterol/HDL-C ratio, and dietary intake among overweight and obese employees.

    Three single nucleotide polymorphisms (SNPs) in alcohol-metabolizing genes - ADH1B (Arg47His), ADH1C (Ile350Val) and ALDH2 (Glu504Lys) have been extensively associated with flush reaction and alcoholism. Therefore, we investigated the association of these three SNPs with alcohol-induced reactions (AIRs), alcoholism risk, personality and anthropometric traits among Malaysian university students. AIRs, Self-Rating of the Effects of Alcohol (SRE) and Ten-Item Personality were surveyed, anthropometric measurements and DNA samples were taken. Among 264 valid drinkers (111 males, 153 females; 229 ethnic Chinese, 35 ethnic Indians), the minor allele frequencies for ADH1B, ADH1C, ALDH2 among Chinese/Indians were .45/.07, .33/.40, .32/.41, respectively; distribution of ADH1B alleles significantly different between ethnicities. Current/former experiences of flushing, hives, heart palpitations after alcohol consumption; and sleepiness, headache reactions, early and overall SRE were significantly different between ethnicities and genders, respectively. Overall SRE score was associated with ADH1C and ALDH2 alleles. 'Openness to Experiences' was associated with ALDH2 genotypes and alleles; Glu/Glu or Glu carriers showed significantly higher means. ADH1B Arg/Arg and Arg carriers showed significantly higher total body and subcutaneous fats but association was abolished after controlling for ethnicity. In conclusion, gender and ethnicity, but not alcohol-metabolizing gene variants, play a role in influencing the manifestation of AIRs.

    Adipose tissue-derived stromal cells (ASCs) natively reside in a relatively low-oxygen tension (i.e., hypoxic) microenvironment in human body. Low oxygen tension (i.e., in situ normoxia), has been known to enhance the growth and survival rate of ASCs, which, however, may lead to the risk of tumourigenesis. Here, we investigated the tumourigenic potential of ASCs under their physiological condition to ensure their safe use in regenerative therapy. Human ASCs isolated from subcutaneous fat were cultured in atmospheric O2 concentration (21% O2) or in situ normoxia (2% O2). We found that ASCs retained their surface markers, tri-lineage differentiation potential, and self-renewal properties under in situ normoxia without altering their morphology. In situ normoxia displayed a higher proliferation and viability of ASCs with less DNA damage as compared to atmospheric O2 concentration. Moreover, low oxygen tension significantly up-regulated VEGF and bFGF mRNA expression and protein secretion while reducing the expression level of tumour suppressor genes p16, p21, p53, and pRb. However, there were no significant differences in ASCs telomere length and their relative telomerase activity when cultured at different oxygen concentrations. Collectively, even with high proliferation and survival rate, ASCs have a low tendency of developing tumour under in situ normoxia. These results suggest 2% O2 as an ideal culture condition for expanding ASCs efficiently while maintaining their characteristics.

    Obesity during pregnancy contributes to the development of metabolic disorders in offspring. Maternal exercise may limit gestational weight gain and ameliorate these programming effects. We previously showed benefits of post-weaning voluntary exercise in offspring from obese dams. Here we examined whether voluntary exercise during pregnancy influences lipid and glucose homeostasis in muscle and fat in offspring of both lean and obese dams. Female Sprague-Dawley rats were fed chow (C) or high fat (F) diet for 6 weeks before mating. Half underwent voluntary exercise (CE/FE) with a running wheel introduced 10 days prior to mating and available until the dams delivered; others remained sedentary (CS/FS). Male and female pups were killed at postnatal day (PND)19 and retroperitoneal fat and gastrocnemius muscle were collected for gene expression. Lean and obese dams achieved similar modest levels of exercise. At PND1, both male and female pups from exercised lean dams were significantly lighter (CE versus CS), with no effect in those from obese dams. At PND19, maternal obesity significantly increased offspring body weight and adiposity, with no effect of maternal exercise. Exercise significantly reduced insulin concentrations in males (CE/FE versus CS/FS), with reduced glucose in male FE pups. In males, maternal obesity significantly decreased muscle myogenic differentiation 1 (MYOD1) and glucose transporter type 4 (GLUT4) mRNA expressions (FS vs CS); these were normalized by exercise. Maternal exercise upregulated adipose GLUT4, interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-α), and peroxisome proliferator activated receptor gamma coactivator 1 alpha (PGC1α) mRNA expression in offspring of dams consuming chow. Modest voluntary exercise during pregnancy was associated with lower birth weight in pups from lean dams. Maternal exercise appeared to decrease the metabolic risk induced by maternal obesity, improving insulin/glucose metabolism, with greater effects in male than female offspring.

    INTRODUCTION: There is limited data on the effects of low carbohydrate diets on renal outcomes particularly in patients with underlying diabetic kidney disease. Therefore, this study determined the safety and effects of very low carbohydrate (VLCBD) in addition to low protein diet (LPD) on renal outcomes, anthropometric, metabolic and inflammatory parameters in patients with T2DM and underlying mild to moderate kidney disease (DKD).

    MATERIALS AND METHODS: This was an investigator-initiated, single-center, randomized, controlled, clinical trial in patients with T2DM and DKD, comparing 12-weeks of low carbohydrate diet (<20g daily intake) versus standard low protein (0.8g/kg/day) and low salt diet. Patients in the VLCBD group underwent 2-weekly monitoring including their 3-day food diaries. In addition, Dual-energy x-ray absorptiometry (DEXA) was performed to estimate body fat percentages.

    RESULTS: The study population (n = 30) had a median age of 57 years old and a BMI of 30.68kg/m2. Both groups showed similar total calorie intake, i.e. 739.33 (IQR288.48) vs 789.92 (IQR522.4) kcal, by the end of the study. The VLCBD group showed significantly lower daily carbohydrate intake 27 (IQR25) g vs 89.33 (IQR77.4) g, p<0.001, significantly higher protein intake per day 44.08 (IQR21.98) g vs 29.63 (IQR16.35) g, p<0.05 and no difference in in daily fat intake. Both groups showed no worsening of serum creatinine at study end, with consistent declines in HbA1c (1.3(1.1) vs 0.7(1.25) %) and fasting blood glucose (1.5(3.37) vs 1.3(5.7) mmol/L). The VLCBD group showed significant reductions in total daily insulin dose (39(22) vs 0 IU, p<0.001), increased LDL-C and HDL-C, decline in IL-6 levels; with contrasting results in the control group. This was associated with significant weight reduction (-4.0(3.9) vs 0.2(4.2) kg, p = <0.001) and improvements in body fat percentages. WC was significantly reduced in the VLCBD group, even after adjustments to age, HbA1c, weight and creatinine changes. Both dietary interventions were well received with no reported adverse events.

    CONCLUSION: This study demonstrated that dietary intervention of very low carbohydrate diet in patients with underlying diabetic kidney disease was safe and associated with significant improvements in glycemic control, anthropometric measurements including weight, abdominal adiposity and IL-6. Renal outcomes remained unchanged. These findings would strengthen the importance of this dietary intervention as part of the management of patients with diabetic kidney disease.

    TMP is gradually becoming a fundamental element for quality assurance and control in ionizing and non-ionizing radiation imaging modalities as well as in the development of different techniques. This study aimed to evaluate and obtain polyvinyl chloride tissue mimicking material for dual-modality breast phantoms in mammography and ultrasound. Breast tissue equivalence was evaluated based on X-ray attenuation properties, speed of sound, attenuation, and acoustic impedance. There are six samples of PVC-plasticizer material with variations of PVC concentration and additives. The evaluation of X-ray attenuation was carried out using mammography from 23 to 35 kV, while the acoustic properties were assessed with mode A ultrasound and a transducer frequency of 5 MHz. A breast phantom was created from TMP material with tissue equivalence and was then evaluated using mammography as well as ultrasound to analyze its image quality. The results showed that samples A (PVC 5%, DOP 95%), B (PVC 7%, DOP 93%), C (PVC 10%, DOP 90%), E (PVC 7%, DOP 90%, graphite 3%), and F (PVC 7%, DOP 90%, silicone oil 3%) have the closest equivalent to the ACR breast phantom material with a different range of 0.01-1.39 in the 23-35 kV range. Based on the evaluation of the acoustic properties of ultrasound, A had high similarity to fat tissue with a difference of 0.03 (dB cm- 1 MHz- 1) and 0.07 (106 kg m- 2 s- 1), while B was close to the glandular tissue with a difference of 9.2 m s- 1. Multilayer breast phantom images' results showed gray levels in mammography and ultrasound modalities. Therefore, this study succeeded in establishing TMP material for mammography and ultrasound. It can also be used for simple quality assurance and control programs.

    Diabesity is the combination of type 2 diabetes and obesity characterized by chronic low-grade inflammation. The Wnt signaling act as an evolutionary pathway playing crucial role in regulating cellular homeostasis and energy balance from hypothalamus to metabolic organs. Aberrant activity of certain appendages in the canonical and non-canonical Wnt system deregulates metabolism and leads to adipose tissue expansion, this key event initiates metabolic stress causing metaflammation and obesity. Metaflammation induced obesity initiates abnormal development of adipocytes mediating through the non-canonical Wnt signaling inhibition of canonical Wnt pathway to fan the flames of adipogenesis. Moreover, activation of toll like receptor (TLR)-4 signaling in metabolic stress invites immune cells to release pro-inflammatory cytokines for recruitment of macrophages in adipose tissues, further causes polarization of macrophages into M1(classically activated) and M2 (alternatively activated) subtypes. These events end with chronic low-grade inflammation which interferes with insulin signaling in metabolic tissues to develop type 2 diabetes. However, there is a dearth in understanding the exact mechanism of Wnt-TLR axis during diabesity. This review dissects the molecular facets of Wnt and TLRs that modulates cellular components during diabesity and provides current progress, challenges and alternative therapeutic strategies at preclinical and clinical level.

    The aim of this study was to compare physical activity prevalence estimates among the International Physical Activity Questionnaire (IPAQ), steps/day, and accelerometer in a sample of government employees in Kangar, Perlis, Malaysia. Ten government agencies in Kangar were randomly chosen, and all employees were invited to participate. A self-administered questionnaire was employed to obtain information on socio-demographic characteristics and a physical activity assessment using the IPAQ. Anthropometric measurements, which include measurements of weight, height, body mass index, percent body fat, waist and hip circumference, were carried out. An accelerometer was used to assess total daily energy expenditure and the number of steps/day. A total of 272 respondents were involved in this study with a response rate of 83.2%. According to IPAQ, accelerometer and steps/day, the majority of the respondents (22.0%, 55.1%, and 77.6%, respectively) were classified as sedentary. The agreement between physical activity level as determined by the accelerometer vs. the IPAQ (Kappa=-0.46 {95% CI -0.384,-0.536}, p=0.238) and the IPAQ vs. steps/day (Kappa =0.037 {95% CI 0.090,-0.016}, p=0.175) was not significant, but the agreement between physical activity level as determined by the accelerometer vs. steps/day was classified as fair (Kappa=0.296 (95% CI 0.392, 0.200}, p

    The stress and anxiety caused by the coronavirus (COVID-19) pandemic presents a serious threat to psychological well-being in populations worldwide and may also extend to body image outcomes. To test this hypothesis, we conducted a preliminary study in which an online sample of adults from the United Kingdom (N = 506, age M = 34.25 years) were asked to complete measures of perceived stress, stressful life events, trait anxiety, COVID-19-related stress and anxiety, and negative body image (body dissatisfaction and drive for thinness in women, body fat and muscularity dissatisfaction in men). The results of hierarchical regressions indicated that COVID-19-related stress and anxiety explained significant incremental variance in body image outcomes (Adj. ΔR2 = .02 to .10), over-and-above demographics (age and body mass index) and perceived stress, trait anxiety, and stressful life events. These findings suggest that COVID-19-related stress and anxiety may shape body image outcomes under conditions of physical and social distancing.

    INTRODUCTION: Although weight loss is common in Parkinson's disease (PD), longitudinal studies assessing weight and body composition changes are limited.

    METHODS: In this three-year longitudinal study, 125 subjects (77 PD patients and 48 spousal/sibling controls) underwent clinical, biochemical and body composition assessments using dual-energy X-ray absorptiometry.

    RESULTS: Patients were older than controls (65.6 ± 8.9 vs. 62.6 ± 7.1, P = 0.049), with no significant differences in gender, comorbidities, dietary intake and physical activity. Clinically significant weight loss (≥5% from baseline weight) was recorded in 41.6% of patients, with a doubling of cases (6.5 to 13.0%) classified as underweight at study end. Over three years, patients demonstrated greater reductions in BMI (mean -1.2 kg/m2, 95%CI-2.0 to -0.4), whole-body fat percentage (-2.5% points, 95%CI-3.9 to -1.0), fat mass index (FMI) (-0.9 kg/m2, 95%CI-1.4 to -0.4), visceral fat mass (-0.1 kg, 95%CI-0.2 to 0.0), and subcutaneous fat mass (-1.9 kg, 95%CI-3.4 to -0.5) than in controls, with significant group-by-time interactions after adjusting for age and gender. Notably, 31.2% and 53.3% of patients had FMI<3rd (severe fat deficit) and <10th centiles, respectively. Muscle mass indices decreased over time in both groups, without significant group-by-time interactions. Multiple linear regression models showed that loss of body weight and fat mass in patients were associated with age, dyskinesia, psychosis and constipation.

    CONCLUSIONS: We found progressive loss of weight in PD patients, with greater loss of both visceral and subcutaneous fat, but not muscle, compared to controls. Several associated factors (motor and non-motor disease features) were identified for these changes, providing insights on possible mechanisms and therapeutic targets.

    OBJECTIVES: Our previous studies on osteoarthritis (OA) revealed positive outcome after chondrogenically induced cells treatment. Presently, the functional improvements of these treated OA knee joints were quantified followed by evaluation of the mechanical properties of the engineered cartilages.
    METHODS: Baseline electromyogram (EMGs) were conducted at week 0 (pre-OA), on the locomotory muscles of nine un-castrated male sheep (Siamese long tail cross) divided into controls, adipose-derived stem cells (ADSCs) and bone marrow stem cells (BMSCs), before OA inductions. Subsequent recordings were performed at week 7 and week 31 which were post-OA and post-treatments. Afterwards, the compression tests of the regenerated cartilage were performed.
    RESULTS: Post-treatment EMG analysis revealed that the control sheep retained significant reductions in amplitudes at the right medial gluteus, vastus lateralis and bicep femoris, whereas BMSCs and ADSCs samples had no further significant reductions (P < 0.05). Grossly and histologically, the treated knee joints demonstrated the presence of regenerated neo cartilages evidenced by the fluorescence of PKH26 tracker. Based on the International Cartilage Repair Society scores (ICRS), they had significantly lower grades than the controls (P < 0.05). The compression moduli of the native cartilages and the engineered cartilages differed significantly at the tibia plateau, patella femoral groove and the patella; whereas at the medial femoral condyle, they had similar moduli of 0.69 MPa and 0.40-0.64 MPa respectively. Their compression strengths at all four regions were within ±10 MPa.
    CONCLUSION: The tissue engineered cartilages provided evidence of functional recoveries associated to the structural regenerations, and their mechanical properties were comparable with the native cartilage.

    BACKGROUND AND OBJECTIVES: Parental body mass index (BMI) is strongly linked with the development of offspring overweight and obesity. However, there are a limited number of studies focusing on the association of parental body mass index before pregnancy on offspring growth and body composition in early life, particularly in developing countries.

    METHODS: Data from the University Sains Malaysia (USM) Pregnancy Cohort which consists of 153 mother-offspring pairs were used. Data were collected using interview-administered questionnaires and anthropometric measurements were also obtained. Multiple linear regression and generalised equation estimation (GEE) were used to examine the direction and impact of the association between parental BMI and child growth and body composition (weight for age, height for age, body mass index for age, weight for height and fat mass at age 2m, 6m, and 12m). Potential confounders, including validated measures of maternal diets and physical activity during pregnancy, were considered.

    RESULTS: Of 153 parents, one-quarter of the mothers and 42.2% of the fathers, respectively, were overweight or obese before pregnancy. A significant association was found between maternal BMI and child's weight for height z-score (WHZ) and body mass index for age z-score (BAZ).

    CONCLUSIONS: Having high pre-pregnancy BMI may increase BMI and WAZ of offspring in early life. Findings from this study emphasise the importance of monitoring maternal weight status, particularly before and during pregnancy and early life of offspring among Malaysians.

    This paper describes body composition and dietary intakes of the three major ethnic groups residing in Singapore and how these are related to cardiovascular risk factors in these groups. When the relationship between body mass index (BMI, kg/m2) and body fat percentage was studied, Singaporeans were found to have higher percentage of body fat compared with Caucasians with the same BMI. At BMIs that are much lower than WHO-recommended cut-off values for obesity, both the absolute and relative risks of developing cardiovascular risk factors are markedly elevated for all three ethnic groups. The excessive fat accumulation and increased risks at lower BMIs signal a need to re-examine cut-off values for obesity among Chinese, Malays, and Indians.

    Cachexia is an irreversible condition that involves a significant loss of body weight, muscle mass, and adipose tissue. It is a complex condition that involves a variety of metabolic, hormonal, and immune-related factors, with the precise mechanisms not yet fully understood. In this review, the prevalence of cachexia in different types of cancer as well as the potential risk factors was evaluated from literature retrieved from databases such as ScienceDirect, PubMed and Scopus. Potential risk factors evaluated here include tumor-related factors such as location, and stage of the cancer, as well as patient-related factors such as age, gender, and comorbidities. Several findings were observed where cachexia is more prevalent in male cancer patients than females, with higher incidences of weight loss and poorer outcomes. This may be due to the different muscle compositions between gender. Additionally, cachexia is more prevalent at the later stages, which may be brought about by the late-stage diagnosis of certain cancers. The anatomical location of certain cancers such as the pancreas and stomach may play a significant factor in their high prevalence of cachexia. These are sites of the synthesis of digestive enzymes and hormones regulating appetite. Cachexia is an issue faced by cancer patients which could affect their recovery. However, it is poorly understood, which limit therapeutic options. Hence, understanding this disease from different perspectives (clinical and pre-clinical), and bridging those findings could further improve our comprehension and consequently improve therapeutic options.

    This study aims to examine the level of knowledge, attitude and practices (KAP) of adolescents towards sugar-sweetened beverages (SSB), together with the associated factors that determine their KAP. Data were collected using self-administered questionnaires that consisted of sociodemographic, the KAP for the SSB questionnaire, and the Beverage Intake Questionnaire (BEVQ). The respondents' heights, weights, waist circumferences and body fat percentages were measured. This study involved 439 adolescents aged between 13 and 17 years old, in public secondary schools in Selangor, Malaysia. The results reveal that 35% of the adolescents were overweight, 26% had a high waist circumference, and 45% had a high body fat percentage. Caffeinated drinks and full cream milk were the most frequently consumed SSBs. The KAP score revealed a good attitude (88.4%), a moderate knowledge (51.8%) and a poor practice (40.5%). Those with a higher body fat percentage showed significantly good attitude scores (p < 0.05). Low household income groups, females, adolescents aged 16-17 years old and being from an urban area demonstrated a significant (p < 0.05) positive determinant towards the KAP score. In conclusion, high awareness of negative health outcomes associated with SSBs among adolescents was not in accordance with the level of their lifestyle choices.

    Coxsackievirus A16 (CV-A16) is the leading cause of hand-foot-mouth disease (HFMD), which usually
    presents as mild and self-limiting symptoms in young children. Rarely, CV-A16 has been reported
    to cause severe and fatal neurological complications but little is known about these complications.
    In the present study, 1-day and 7-day old mouse models of CV-A16 were developed using a clinical
    strain via subcutaneous inoculation. All infected mice exhibited clinical signs of infection, including
    reduced mobility, limb weakness and paralysis between 3 to 6 days post-infection. Pathologically,
    the main organs involved were the central nervous system (CNS), skeletal muscles and brown fat. In
    the CNS, viral antigens as demonstrated by immunohistochemistry, were localized mainly to neurons
    in the brain stem and spinal cord, suggesting that CV-A16 is neurotropic although inflammation is
    very mild. The skeletal muscles showed necrosis and myositis due to viral infection as evidenced by
    the dense viral antigens. Focal viral antigens were also detected in the brown fat. These preliminary
    pathological findings indicate that our mouse models can be further developed to be useful models
    for pathogenesis studies, and vaccine and anti-viral drug evaluation.

    Background: Obesity is major contributor to diseases and mortality. Constant weight loss significantly reverses diseases related to obesity and helps prolong human life expectancy. To date, there has been limited scientific evidence comparing the effectiveness of the Aqua Zumba® Fitness program and conventional aqua jog among obese women. Thus, the purpose of this study is to compare the effects between 12 weeks of Aqua Zumba® Fitness and aqua jog on health parameters among middle-age women. Methods: A total of sixty sedentary obese women (BMI >30 kg/m2), aged 40-59 years old were randomized to either Aqua Zumba® Fitness (AZ, n=20), aqua jog (AJ, n=20) or control group (C, n=20). Exercise groups were involved in different 12-week aqua exercise programs, 3 times per week, 60 minutes per session, with an intensity of 50-75% of maximum heart rate.     The purpose of this study was to investigate the relationship between anthropometric and motor performance and to determine the contribution of combined anthropometric dimensions on motor performance. A total of 225 male (n = 138) and female (n = 87) Malaysian university athletes aged
    between 18 and 28 years (M = 22.1, SD = 1.8) from 18 different team related sports and individual sports participated in this study. The subjects underwent anthropometric measurements (height, weight, BMI,% body fat, waist-hip ratio) and motor performance tests (grip strength - GS, back
    strength - BS, 7 level sit-up - SU7, 10m sprint - S10, 30m sprint - S30, sit and reach - SR, trunk extension test - TE, SEMO agility test - SEMO, vertical jump - VJ, standing long jump - SLJ, reaction time (audio) - RTa, reaction time (visual) - RTv, bleep test - Bleep, leg strength - LS, stork test– ST, and push-ups - PU). Descriptive analysis showed that anthropometric and performance difference between sports reflected the needs and requirements of the sports. Multivariate Analysis of Covariance (MANCOVA) and multiple regression analysis showed that height, weight, BMI, % body fat, waist-hip ratio contributed positively or negatively on specific components of motor performance. Among male athletes, all anthropometric were found to be a significant contributor to strength, vertical jump, cardiovascular endurance, while among females, only % body fat and WHR contributed significantly to abdominal strength, vertical jump and trunk extension. Findings of the study suggested that anthropometric characteristics that contributed to motor performance should provide a scientific rationale in selecting and training of athletes.
