    The usage of natural products in pharmaceuticals has steadily seen improvements over the last decade, and this study focuses on the utilization of palm oil in formulating liposomal doxorubicin (Dox). The liposomal form of Dox generally minimizes toxicity and enhances target delivery actions. Taking into account the antiproliferative and antioxidant properties of palm oil, the aim of this study is to design and characterize a new liposomal Dox by replacing phosphatidylcholine with 5% and 10% palm oil content. Liposomes were formed using the freeze_thaw method, and Dox was loaded through pH gradient technique and characterized through in vitro and ex vivo terms. Based on TEM images, large lamellar vesicles (LUV) were formed, with sizes of 438 and 453 nm, having polydispersity index of 0.21 ± 0.8 and 0.22 ± 1.3 and zeta potentials of about -31 and -32 mV, respectively. In both formulations, the entrapment efficiency was about 99%, and whole Dox was released through 96 hours in PBS (pH = 7.4) at 37°C. Comparing cytotoxicity and cellular uptake of LUV with Caelyx(R) on MCF7 and MDA-MBA 231 breast cancer cell lines indicated suitable uptake and lower IC50 of the prepared liposomes.

    This work investigates a new interrogation method of a fiber Bragg grating (FBG) sensor based on longer and shorter wavelengths to distinguish between transversal forces and temperature variations. Calibration experiments were carried out to examine the sensor's repeatability in response to the transversal forces and temperature changes. An automated calibration system was developed for the sensor's characterization, calibration, and repeatability testing. Experimental results showed that the FBG sensor can provide sensor repeatability of 13.21 pm and 17.015 pm for longer and shorter wavelengths, respectively. The obtained calibration coefficients expressed in the linear model using the matrix enabled the sensor to provide accurate predictions for both measurements. Analysis of the calibration and experiment results implied improvements for future work. Overall, the new interrogation method demonstrated the potential to employ the FBG sensing technique where discrimination between two/three measurands is needed.

    In this study, it focused on empty fruit brunch (EFB) fibres reinforcement in polybutylene succinate (PBS) with modified tapioca starch by using hot press technique for the use of agricultural mulch film. Mechanical, morphological and thermal properties were studied. Mechanical analysis showed decreased in values of modulus strength for both tensile and flexural testing for fibres insertion. Higher EFB fibre contents in films resulted lower mechanical properties due to poor fibre wetting from insufficient matrix. This has also found evident in SEM micrograph, showing poor interfacial bonding. Water vapour permeability (WVP) shows as higher hydrophilic EFB fibre reinforcement contents, the rate of WVP also increase. Besides this, little or no significant changes on thermal properties for composite films. This is because high thermal stability PBS polymer show its superior thermal properties dominantly. Even though EFB fibres insertion into PBS/tapioca starch biocomposite films have found lower mechanical properties. It successfully reduced the cost of mulch film production without significant changes of thermal performances.

    Aim: This study was aimed at evaluating the anticancer potential of curcumin-loaded poly(lactic-co-glycolic acid) (PLGA) based nanoparticles (NPs) in MDA-MB231 human breast cancer cells. Methods: Curcumin-loaded PLGA NPs were developed using a modified solvent evaporation technique. Physical characterization was performed on the formulated NPs. Furthermore, in vitro experiments were conducted to study the biological activity of the curcumin-loaded NPs. Results: Curcumin-loaded PLGA NPs demonstrated high encapsulation efficiency and sustained payload release. Moreover, the NPs exhibited a significant reduction in cell viability, cell migration and cell invasion in the MDA-MB231 cells. Conclusion: The study revealed that the formulated curcumin-loaded PLGA NPs possessed significant anti-metastatic properties. The findings showcased the possible potential of curcumin-loaded NPs in the management of debilitating conditions such as cancer. In addition, this study could form the basis for further research and advancements in this area.

    This paper reports the performance of two different artificial neural networks (ANN), Multi Layer Perceptron (MLP) and Radial Basis Function (RBF) compared to conventional software for prediction of the pore size of the asymmetric polyethersulfone (PES) ultrafiltration membranes. ANN has advantages such as incredible approximation, generalization and good learning ability. The MLP are well suited for multiple inputs and multiple outputs while RBF are powerful techniques for interpolation in multidimensional space. Three experimental data sets were used to train the ANN using polyethylene glycol (PEG) of different molecular weights as additives namely as PEG 200, PEG 400 and PEG 600. The values of the pore size can be determined manually from the graph and solve it using mathematical equation. However, the mathematical solution used to determine the pore size and pore size distribution involve complicated equations and tedious. Thus, in this study, MLP and RBF are applied as an alternative method to estimate the pore size of polyethersulfone (PES) ultrafiltration membranes. The raw data needed for the training are solute separation and solute diameter. Values of solute separation were obtained from the ultrafiltration experiments and solute diameters ware calculated using mathematical equation. With the development of this ANN model, the process to estimate membrane pore size could be made easier and faster compared to mathematical solutions.

    Granulocyte-colony stimulating factor (G-CSF) mobilizes and increases the amount of hematopoietic stem cells in peripheral blood, enabling its harvest by few apheresis procedures. The pegylated G-CSF has longer half-life and is given once only, which is more comfortable for patients, whereas the non-pegylated requires multiple daily injection because of its short half-life. We summarized results of randomized trials comparing the efficacy and safety of pegylated and non-pegylated G-CSF for peripheral blood stem cell mobilization. We searched the Cochrane CENTRAL, MEDLINE, EMBASE, and two conference proceedings. Two authors made the selection, extracted data and evaluated methodological quality using GRADE independently. We used random-effects model for meta-analysis. We found 3956 records and retrieved 47 full texts. We included eight randomized trials with a total number of 554 randomized and 532 analyzed subjects. The meta-analysis included five trials because not all trials reported the same outcomes. Pooling data from two studies shows no evidence for a difference in the successful mobilization rate (CD34+ cell ≥ 2 × 106 /kg collected) between pegfilgrastim 6 mg (early administration) and filgrastim 5 µg/kg/day (147 participants; risk ratio (RR) 0.87, 95% confidence interval (95%CI) 0.67-1.11; P = .26). Pooling data from three studies shows no difference in the incidence of adverse events between pegylated and non-pegylated G-CSF (170 participants; RR 0.86, 95%CI 0.34-2.17; P = .75). No difference found on the quantity of CD34+ cells collected, number of apheresis procedure in successful mobilization, level of peak PB CD34+ cells achieved, and day of neutrophil and platelet engraftment.

    Calcium carbonate was evaluated as a replacement for the base during the fermentation of glycerol by a highly productive strain of 1,3-propanediol (PDO), viz., Clostridium butyricum JKT37. Due to its high specific growth rate (µmax=0.53h(-1)), 40g/L of glycerol was completely converted into 19.6g/L of PDO in merely 7h of batch fermentation, leaving only acetate and butyrate as the by-products. The accumulation of these volatile fatty acids was circumvented with the addition of calcium carbonate as the pH neutraliser before the fermentation was inoculated. An optimal amount of 15g/L of calcium carbonate was statistically determined from screening with various glycerol concentrations (20-120g/L). By substituting potassium hydroxide with calcium carbonate as the pH neutraliser for fermentation in a bioreactor, a similar yield (YPDO/glycerol=0.6mol/mol) with a constant pH was achieved at the end of the fermentation.

    A single wall single image x-ray radiographic technique was adopted to investigate thickness variation of steel specimens caused by uniform corrosion. The ability of the 100 kV-160 kV x-rays to produce a meaningful film density for steel was also investigated. The thickness contour maps of corroded steel plates were found matching with the x-ray film density contour maps of the plates. The results confirm that x-ray radiography can be used to detect the thickness reduction of steel caused by uniform corrosion.
    Bagi mengesan perubahan ketebalan keluli akibat kakisan seragam maka radiografi sinar-x teknik imej tunggal dinding tunggal telah digunakan. Kemampuan sinar-x 100 kV-160 kV menghasilkan ketumpatan filem yang sesuai untuk keluli telah juga dikaji. Peta kontur ketebalan bagi spesimen kepingan keluli yang telah mengalami kakisan seragam didapati sepadan dengan peta kontur ketumpatan filem radiografi sinar-x bagi spesimen tersebut. Kajian ini menunjukkan radiografi sinar-x boleh digunakan bagi mengesan penipisan keluli akibat kakisan seragam.

    Micro powder injection molding (vim) is a promising process that may satisfy the demand on miniaturization parts to micro domain in mass production with low manufacturing cost. Three mol% yttria stabilized zirconia (Ysz) with nano-sized powder and binder system consists of polyethylene glycol (PEG), polymethyl methacrylate (PMMA) and stearic acid (sA) were used. Nano-size powders with higher surface area generally require more binder to form a feedstock. As such, determination of the optimum powder loading of the feedstock for 1UPIM process is important. The rheological characteristics of different YSZ feedstocks with powder loading of 52 53 and 54 vol.% were investigated in terms of flow behavior as a function of viscosity and shear rate. Fairly low values of flow behavior exponent ranging from 025 to 0.39 (n<1) resulted in pseudoplastic flow behavior of the examined Yszfeedstock. The 52 vol.% feedstock exhibited the lowest viscosity resulting in highest activation energy and lowest moldability index of 1.862x10-6, while the 54 vol.% feedstock regardless to its high viscosity, yielded a low activation energy of 4.14 kJImol and high moldability index of 4.59x10-6. Based on rheological properties obtained, a powder loading of 54 vol.% has desirable feedstock characteristics for iumm process and exhibited molding ability for micro detail filling. The relationship between the optimum rheological properties obtained and the actual injection process was also determined. The results showed that the green parts were able to be injected without defects such as short shot or flashing.

    Most of the polymers and their blends, utilized in carbon capture membranes, are costly, but cellulose acetate (CA) being inexpensive is a lucrative choice. In this research, pure and mixed matrix membranes (MMMs) have been fabricated to capture carbon from natural gas. Polyethylene glycol (PEG) has been utilized in the fabrication of membranes to modify the chain flexibility of polymers. Multi-walled carbon nanotubes (MWCNTs) provide mechanical strength, thermal stability, an extra free path for CO2 molecules and augment CO2/CH4 selectivity. Membranes of pure CA, CA/PEG blend of different PEG concentrations (5%, 10%, 15%) and CA/PEG/MWCNTs blend of 10% PEG with different MWCNTs concentrations (5%, 10%, 15%) were prepared in acetone using solution casting techniques. Fabricated membranes were characterized using SEM, TGA and tensile testing. Permeation results revealed remarkable improvement in CO2/CH4 selectivity. In single gas experiments, CO2/CH4 selectivity is enhanced 8 times for pure membranes containing 10% PEG and 14 times for MMMs containing 10% MWCNTs. In mix gas experiments, the CO2/CH4 selectivity is increased 13 times for 10% PEG and 18 times for MMMs with 10% MWCNTs. Fabricated MMMs have a tensile strength of 13 MPa and are more thermally stable than CA membranes.

    Temozolomide (TMZ) is one of the most effective chemotherapeutic agents for glioblastoma multiforme, but the required high administration dose is accompanied by side effects. To overcome this problem and to further improve TMZ's efficacy, targeted delivery of TMZ by using polymeric nanoparticles has been explored. We synthesised the PLGA-PEG-FOL copolymer and attempted encapsulation of TMZ into PLGA-PEG-FOL nanoparticles using the emulsion solvent evaporation method and the nanoprecipitation method. Conjugation of PEG and FOL to PLGA has been reported to be able to increase the delivery of TMZ to the brain as well as targeting the glioma cells. However, despite making numerous modifications to these methods, the loading of TMZ in the nanoparticles only ranged between 0.2% and 2%, and the nanoparticles were between 400 nm and 600 nm in size after freeze-drying. We proceed with determining the release profile of TMZ in phosphate buffered saline (PBS). Our initial data indicated that TMZ was slowly released from the nanoparticles. The metabolite of TMZ rather than the parent compound was detected in PBS. Our study suggests that while PLGA-PEG-FOL can be used as a polymeric or encapsulation material for central delivery of TMZ, a practical and cost effective formulation method is still far from reach.

    A fast melt tablet (FMT) is well regarded as an alternative delivery system that might help resolve a patient's non-compliance issue. The main objective of this study was to develop a cocoa butter-based FMT. Additives, namely 5-15% of PEG 6000, beeswax, paraffin wax, and corn starch, were incorporated into the cocoa butter-based FMT to study the effects of these additives with the physical characteristic of a cocoa butter FMT. An optimum-based formulation was chosen according to the desired hardness and disintegration time and the taste masking property achieved with the model drug-dapoxetine. The analysis demonstrated that incorporating beeswax (15%) and paraffin wax (15%) could prolong the disintegration time by at least two-fold. On the contrary, the presence of corn starch was found to cause an increase in the hardness and reduction of the disintegration time. The disintegration mechanism might be presumed due to the synergistic effect of starch swelling and cocoa butter melting. The hardness value and in vitro disintegration time of the optimum formulation were recorded at 2.93 ± 0.22 kg and 151.67 ± 6.98 s. In terms of dissolution, 80% of dapoxetine was released within 30 min and the dissolution profile was comparable to the innovator product. The formulation was palatable and stable for at least 1 year. The exposure of the FMT formulation at 30 °C for 12 months was reported to be stable. Along with the sound palatability profile and high drug load capacity, the current formulation possesses the desired characteristics to be scaled up and marketed.

    The carbon dioxide (CO2) separation technology has become a focus recently, and a developed example is the membrane technology. It is an alternative form of enhanced gas separation performance above the Robeson upper bound line resulting in the idea of mixed matrix membranes (MMMs). With attention given to membrane technologies, the MMMs were fabricated to have the most desirable gas separation performance. In this work, blend MMMs were synthesised by using two polymers, namely, poly(ether sulfone) (PES) and poly (ethylene glycol) (PEG). These polymers were dissolved in blend N-methyl-2-pyrrolidone (NMP) and dimethylformamide (DMF) solvents with the functionalised multi-walled carbon nanotubes (MWCNTs-F) fillers by using the mixing solution method. The embedding of the pristine MWCNTs and MWCNTs-F within the new synthesised MMM was then studied towards CO2/N2 separation. In addition, the optimisation of the loading of MWCNTs-F for blend MMM for CO2/N2 separation was also studied. The experimental results showed that the functionalised MWCNTs (MWCNTs-F) were a better choice at enhancing gas separation compared to the pristine MWCNTs (MWCNTs-P). Additionally, the effects of MWCNTs-F at loadings 0.01 to 0.05% were studied along with the polymer compositions for PES:PEG of 10:20, 20:20 and 30:10. Both these parameters of study affect the manner of gas separation performance in the blend MMMs. Overall, the best performing membrane showed a selectivity value of 1.01 + 0.05 for a blend MMM (MMM-0.03F) fabricated with 20 wt% of PES, 20 wt% of PEG and 0.03 wt% of MWCNTs-F. The MMM-0.03F was able to withstand a pressure of 2 bar, illustrating its mechanical strength and ability to be used in the post combustion carbon capture application industries where the flue gas pressure is at 1.01 bar.

    On account of heterogeneity, intrinsic ability of drug resistance, and the potential to invade to other parts of the body (malignancy), the development of a rational anticancer regimen is dynamically challenging. Chemotherapy is considered the gold standard for eradication of malignancy and mitigation of its reoccurrence; nevertheless, it has also been associated with detrimental effects to normal tissues owing to its nonselectivity and nominal penetration into the tumor tissues. In recent decades, nanotechnology-guided interventions have been well-acclaimed due to their ability to facilitate target-specific delivery of drugs, avoidance of nontarget distribution, alleviated systemic toxicity, and maximized drug internalization into cancer cells. Despite their numerous biomedical advantages, clinical translation of nanotechnology-mediated regimens is challenging due to their short plasma half-life and early clearance. PEGylation of nanomedicines has been adapted as an efficient strategy to extend plasma half-life and diminished early plasma clearance via alleviating the opsonization (uptake by monocytes and macrophages) of drug nanocarriers. PEGylation provides "stealth" properties to nanocarrier's surfaces which diminished their recognition or uptake by cellular immune system, leading to longer circulation time, reduced dosage and frequency, and superior site-selective delivery of drugs. Therefore, this review aims to present a comprehensive overview of the pharmaceutical advantages and therapeutic feasibility of PEGylation of nanocarriers in improving tumor-specific targetability, reversing drug resistance, and improving pharmacokinetic profile of drugs and anticancer efficacy. Challenges to PEGylated cancer nanomedicines, possible adaptations to resolve those challenges, and pivotal requirement for interdisciplinary research for development of rational anticancer regimen have also been pondered.

    BACKGROUND: Doxorubicin (DOX), an anthracycline anticancer antibiotic, is used for treating various types of cancers. However, its use is associated with toxicity to normal cells and development of resistance due to overexpression of drug efflux pumps. Poloxamer 407 (P407) and vitamin E TPGS (D-α-tocopheryl polyethylene glycol succinate, TPGS) are widely used polymers as drug delivery carriers and excipients for enhancing the drug retention times and stability. TPGS reduces multidrug resistance, induces apoptosis, and shows selective anticancer activity against tumor cells. Keeping in view the problems, we designed a mixed micelle system encapsulating DOX comprising TPGS for its selective anticancer activity and P407 conjugated with folic acid (FA) for folate-mediated receptor targeting to cancer cells.

    METHODS: FA-functionalized P407 was prepared by carbodiimide crosslinker chemistry. P407-TPGS/FA-P407-TPGS-mixed micelles were prepared by thin-film hydration method. Cytotoxicity of blank micelles, DOX, and DOX-loaded micelles was determined by alamarBlue(®) assay.

    RESULTS: The size of micelles was less than 200 nm with encapsulation efficiency of 85% and 73% for P407-TPGS and FA-P407-TPGS micelles, respectively. Intracellular trafficking study using nile red-loaded micelles indicated improved drug uptake and perinuclear drug localization. The micelles show minimal toxicity to normal human cell line WRL-68, enhanced cellular uptake of DOX, reduced drug efflux, increased DOX-DNA binding in SKOV3 and DOX-resistant SKOV3 human ovarian carcinoma cell lines, and enhanced in vitro cytotoxicity as compared to free DOX.

    CONCLUSION: FA-P407-TPGS-DOX micelles show potential as a targeted nano-drug delivery system for DOX due to their multiple synergistic factors of selective anticancer activity, inhibition of multidrug resistance, and folate-mediated selective uptake.

    The toxicities of ROUNDUP and its component chemicals, glyphosate (N-phosphonomethylglycine) and polyoxyethyleneamine (POEA), were determined at 0, 1, 3, 6 and 24 h following administration to rats. The intratracheal administration of glyphosate (0.2 g/kg), POEA (0.1 g/kg), a mixture of glyphosate (0.2 g/kg) + POEA (0.1 g/kg), or ROUNDUP (containing 0.2 g/kg glyphosate and 0.1 g/kg POEA) elicited immediate respiratory effects which were more severe and which lasted longer in the groups receiving the POEA-containing preparations than in the glyphosate alone group. By 1 h, all test preparations had caused deaths, but more occurred from the POEA-containing preparations than from glyphosate. The po administration of POEA (1 g/kg), the mixture of glyphosate (2 g/kg) +POEA (1 g/kg), or ROUNDUP (containing 2 g/kg glyphosate and 1 g/kg POEA) produced diarrhea and blood-stained weeping from noses. Death was only seen from POEA at 24 h. Glyphosate (2 g/kg po) produced transient diarrhea without nose bleeds; POEA caused diarrhea at 1 h; and the mixture of POEA + glyphosate produced diarrhea later that increased in severity with time. Bloody nose secretions were seen only with the preparations that contained POEA. No deaths, respiratory effects or bloody nose secretions occurred in controls given saline. Both POEA and glyphosate caused lung hemorrhages and lung epithelial cell damage with po or intratracheal exposures. These results indicate POEA and preparations that contained POEA were more toxic than glyphosate.

    INTRODUCTION: Nonacog beta pegol (N9-GP) is a glycoPEGylated recombinant factor IX (FIX) with an extended half-life developed for routine prophylaxis and the prevention and treatment of bleeding episodes in patients with haemophilia B.

    AIM: The aim of this study was to evaluate the pharmacokinetics (PK) of N9-GP.

    METHODS: Data from 41 previously treated haemophilia B patients, enrolled globally (16 adolescents/adults and 25 children; FIX activity ≤0.02 IU mL-1) with no history of FIX inhibitors, were included. N9-GP was administered once-weekly as 10 IU kg-1or 40 IU kg-1in adolescents/adults and 40 IU kg-1in children. Blood was sampled up to 168 h (1 week) post dose. Standard PK was estimated on the basis of plasma FIX activity vs. time (PK profiles) using non-compartmental methods. Furthermore, a population PK analysis and FIX activity predictions were performed.

    RESULTS: Incremental recoveries were 0.02 (IU mL-1)/(IU kg-1) in both adolescents/adults and children. The extended half-life resulted in mean trough levels of 0.27 IU mL-1for adolescents/adults and 0.17 IU mL-1for children at steady-state after weekly dosing at 40 IU kg-1. The population PK analysis confirmed a mono-exponential decay in FIX activity and allowed for predictions of FIX activity for adolescents/adults above 0.15 IU mL-1at all times and 6.4 days week-1in children.

    CONCLUSION: N9-GP has the potential to shift previously treated haemophilia B patients from a severe/moderate disease state into a mild- or non-haemophilic range for most of the dosing interval, which is expected to reduce the number of bleeding episodes.

    SUMMARY: Background Nonacog beta pegol is a recombinant glycoPEGylated factor IX with an extended half-life, developed to improve care for patients with hemophilia B. Objectives To investigate the safety, efficacy and pharmacokinetics of nonacog beta pegol for the prophylaxis and treatment of bleeds in previously treated children with hemophilia B. Patients/Methods This phase 3 trial, paradigm(™) 5, enrolled and treated 25 children (aged ≤ 12 years) with hemophilia B (FIX ≤ 2%). Patients were stratified by age (0-6 years and 7-12 years), and received once-weekly prophylaxis with 40 IU kg(-1) nonacog beta pegol for 50 exposure days. Results No patient developed inhibitors, and no safety concerns were identified. Forty-two bleeds in 15 patients were reported to have been treated; the overall success rate was 92.9%, and most bleeds (85.7%) resolved after one dose. The median annualized bleeding rates (ABRs; bleeds per patient per year) were 1.0 in the total population, 0.0 in the 0-6-year group, and 2.0 in the 7-12-year group; the estimated mean ABRs were 1.44 in the total population, 0.87 in the 0-6-year group, and 1.88 in the 7-12-year group. For 22 patients who had previously been receiving prophylaxis, the estimated mean ABR was 1.38 versus a historical ABR of 2.51. Estimated mean steady-state FIX trough levels were 0.153 IU mL(-1) (0-6 years) and 0.190 IU mL(-1) (7-12 years). Conclusion Nonacog beta pegol was well tolerated in previously treated children with hemophilia B; a 40 IU kg(-1) dose provided effective once-weekly prophylaxis and hemostasis when bleeds were treated.
