Macrofungi of the order Polyporales are among the most important wood decomposers and caused economic losses by decaying the wood in standing trees, logs and in sawn timber. Diversity and distribution of Polyporales in Peninsular Malaysia was investigated by collecting basidiocarps from trunks, branches, exposed roots and soil from six states (Johor, Kedah, Kelantan, Negeri Sembilan, Pahang and Selangor) in Peninsular Malaysia and Federal Territory Kuala Lumpur. This study showed that the diversity of Polyporales were less diverse than previously reported. The study identified 60 species from five families; Fomitopsidaceae, Ganodermataceae, Meruliaceae, Meripilaceae, and Polyporaceae. The common species of Polyporales collected were Fomitopsis feei, Amauroderma subrugosum, Ganoderma australe, Earliella scabrosa, Lentinus squarrosulus, Microporus xanthopus, Pycnoporus sanguineus and Trametes menziesii.
The water chemistry of selected rivers in Kota Marudu, Sabah was studied based on the major ion chemistry and its suitability for drinking and irrigation purposes. Ten sampling stations were selected and water samples were collected from each station to assess its chemical properties. The physico-chemical variables including temperature, electrical conductivity (EC), total dissolved solids (TDS), salinity, dissolved oxygen (DO), pH, turbidity, ammoniacal-nitrogen (NH3-N), biological oxygen demand (BOD), chemical oxygen demand (COD) and total suspended solid (TSS) were measured. The cations (K, Mg, Ca, Na) were analyzed by ICP-MS. Most of the variables were within the drinking water quality standards stipulated by the World Health Organization (WHO) and the Ministry of Health (MOH), Malaysia except for turbidity. Sodium adsorption ratio (SAR) and salinity hazard were calculated to identify the suitability of the water as irrigation water. The Wilcox diagram classifies that only 10% of samples are not suitable for the purpose of irrigation. The overall results showed that most of the rivers in Kota Marudu are still in a clean condition and suitable for drinking and irrigation purposes except for Sumbilingan River, which is considered as slightly polluted. The results are supported by the hierarchical cluster analysis as the stations were grouped into two groups; low and high pollution intensities. This preliminary result can update the baseline data of selected water quality parameters in the Kota Marudu and could serve as tool for assisting relevant government bodies in regulating the water resources policies in the future.
The effect of composition on nickel cobaltites prepared by co-precipitation was investigated. Various compositions were synthesised under different heat treatment conditions. The formation mechanism, lattice parameter and crystallite size were determined by various techniques. X-ray diffraction (XRD) analysis revealed that the optimum condition for the formation of single phase nickel-cobalt oxide (NiCo2O4) can be obtained from solution containing Ni: Co ratio of 33.3 mol%: 66.7 mol% with heat treatment at 400 °C. The presence of pure phase NiCo2O4 with the particle size of approximately 34.10 nm was also observed in field emission scanning electron microscopy (FESEM). The crystallinity of the synthesized oxides was improved with the increment of Ni content.
The study of medicine is often regarded by students as a stressful environment particularly during examination period. Studies found a high percentage of medical students experience significant psychological distress during the examination period. This study compared percentage and level of psychological distress between two batches of first year medical students who underwent different selection admission processes during a stressful examination period. A comparative
cross-sectional study was done on two batches of first year medical students; one group selected based on academic merit (2008/2009 batch) and the other selected based on academic merit, psychometric tests and interview (2009/2010 batch). The psychological distress was measured by the 12-item general health questionnaire (GHQ-12). The data were collected right after the final examinations. A total of 99 (46.05%) medical students of the 2008/2009 batch and 196 (100%) medical students of the 2009/2010 batch participated. The percentage of medical students who had psychological distress of the 2008/2009 and the 2009/2010 batches were 58.59% and 42.3%, respectively. The mean GHQ-12 score and percentage of psychological distress were significantly different between the two batches (p< 0.01). The older batch had 2.01 times higher risk for developing psychological distress compared with the newer batch (p< 0.01). The newer batch
of medical students had better psychological health status and was less likely to develop psychological distress during the stressful period compared with older batch.
In this study, the asymmetry of the main effects of action, background and tonal frequency during a pitch memory processing
were investigated by means of brain activation. Eighteen participants (mean age 27.6 years) were presented with low and
high frequency tones in quiet and in noise. They listen, discriminate and recognize the target tone against the final tone
in a series of four distracting tones. The main effects were studied using the analysis of variance (ANOVA) with action (to
wring (rubber bulb) vs. not to wring), background (in quiet vs. in noise) and frequency (low vs. high) as the factors (and
levels respectively). The main effect of action is in the right pre-central gyrus (PCG), in conformation with its contralateral
behavior. The main effect of background indicated the bilateral primary auditory cortices (PAC) and is right lateralized,
attributable to white noise. The main effect of frequency is also observed in PAC but bilaterally equal and attributable to
low frequency tones. Despite the argument that the temporo-spectral lateralization dichotomy is not especially rigid as
revealed by the main effect of frequency, right lateralization of PAC for the respective main effect of background clearly
demonstrates its functional asymmetry suggesting different perceptual functionality of the right and left PAC.
The relationship between heavy metal and trophic properties in polymictic lake at Sembrong Lake, Peninsular Malaysia was assessed. Sixteen parameters, including heavy metals and trophic parameters were monitored. pH, temperature, dissolved oxygen and heavy metals level changes significantly influenced by the dynamic of polymictic mixing pattern. The mean concentrations of heavy metals in the reservoir decreased in the following order: Fe > Mn > Zn > Cu > As > Pb. The result showed that this polymictic lake is being threatened by cultural eutrophication with TSI value range from 72.40 to 80.41 and classified as a hypereutrophic lake. The levels of heavy metal pollution in the reservoir range from slightly polluted to polluted. Factor analysis was performed to determine the relationship between heavy metals and trophic parameters. Five factors were responsible for data structure and explained the 83% of total variance. These factors differentiate each group of parameters according to their common characteristics. Photosynthesis, respiration and redox processes were main factors contributing to the variability of both properties.
High sensitivity signal detection for a sparse temporal sampling (STS) functional magnetic resonance imaging (fMRI) is compensated by the increase in the number of scans (Ns) and consequently the scan time. A long scan time would result in fatigue and restlessness in participants, while a short scan time is undesirable for an STS-fMRI due to insufficient Ns for averaging. The purpose of this study was to determine the Ns practically sufficient for a sparse fMRI study. Eighteen participants were presented with white noise during a sparse fMRI scan. The height extent of activation was determined via t statistics and region of interest (ROI) based percentage of signal change (PSC). The t statistics and PSC for Heschl’s gyrus (HG) and superior temporal gyrus (STG) during which the participants listened to the white noise were calculated for different number of scans which were 6, 12, 18, 24, 30 and 36. The t statistics and PSC values calculated for the bilateral HG and STG qualitatively indicated a minimal change over Ns = 12 to 36. Both ROIs showed a consistent common right lateralization of activation for all Ns, indicating the right-hemispheric dominance of auditory cortex in processing white noise stimulus. It was proposed that for a sparse fMRI study, Ns may practically fall between 12 and 36
A 31-year-old lady with normal physical characteristics was found to have persistent high FSH and LH and was suspected possible premature ovarian failure after reported to have not normal menstrual cycle. Leucocytes were collected from patient’s fresh peripheral blood sample and Giemsa banding (G-banding) was done. All metaphases were captured and analysed using Cytovision software 4.5 and the final analysis show 47,XXX.
Advanced parental age is a risk factor for chromosomal abnormalities in their offspring. Trisomy X or Triple X syn- drome has previously been reported with advanced maternal age. Here we report two (2) cases of Trisomy X with paternal age as risk factor. Generally, Trisomy X individuals show variable physical and psychological manifesta- tions. However, both cases reported here have advanced paternal age as a risk factor; 55 years old (46 years old at conception) for Case 1 with patient having right eye squint, beaked nose, Posterior Misalignment Type Ventricular Septal Defect (PMVSD) and small Patent Ductus Arteriosus (PDA) with failure to thrive and 49 years old (45 years old at conception) for Case 2 with speech delay and protruding tongue. In view of that, advanced paternal age could possibly contribute the accumulation of de novo mutations in germ line mosaicism.
Clinacanthus nutans (Acanthaceae) is a local plant consumed as tisane in Indonesia and ‘ulam’ in Malaysia. This plant has been claimed for its ability to prevent many diseases including diabetes. However, the scientific proof on this claim is still lacking. Therefore, the present work study was designed to evaluate the antidiabetic potential and antioxidant capacity of C. nutans leaves extracts using in vitro bioassay tests. The 80% methanolic crude extract of this plant was further partitioned using different polarity solvents namely hexane, hexane:ethyl acetate (1:1, v/v), ethyl acetate, ethyl acetate:methanol (1:1, v/v), and methanol. All the sub-fractions were analysed for antioxidant effect via 2, 2-diphenyl-2-picrylhydrazil (DPPH) scavenging activity, ferric reducing power (FRAP) and xanthine oxidase (XO) assays followed by antidiabetic evaluation via α-glucosidase and dipeptidyl peptidase-IV (DPP-IV) inhibitory assays and glucose uptake experiment. The ethyl acetate fraction showed a good antioxidant potential while the hexane fraction exhibited high α-glucosidase and DPP-IV enzyme inhibition. The hexane fraction also improved glucose uptake in a dose-dependent manner. The present work thus provides an informative data on the potential of C. nutans to be developed as a functional food in preventing diabetes.
Introduction: Transfusion Medicine is an evolving filed which integrates multidisciplinary science in providing safe blood and blood products for patients. With an increasing demand for Transfusion Medicine training in Malaysia, a formal survey is needed to evaluate the postgraduate Transfusion Medicine programme offered by Advanced Med- ical and Dental Institute, Universiti Sains Malaysia to identify areas of deficiency based on the alumni experiences. Methods: An English language survey form was developed specifically to assess the programme contents (overall contents, learning experience in each year, and support in research), the alumni perception on soft-skills gained during the study, and the outcome of the programme. The survey forms were distributed to all alumni between March 2018 and October 2018 via e-mail or hand-delivered. Results: The survey response rate was 79% (37 of 47). A ma- jority (97%) of the transfusion medicine specialists (TMS) in this study reported that the course offered in the program was relevant to their current job. The learning experience which includes course content, student’s placement, facili- ties, and support in research) were rated between satisfactory and good. Communicating effectively through speaking was the highest reported soft-skill gained during the programme, whereas communicating effectively in writing was the lowest soft-skill gained. On the programme outcome, all TMS agreed that this programme will produce qualified and well trained specialists for current working market. Conclusion: Some improvement in the programme contents and teaching activities are needed to equip the future TMS for the nation.
Background: Coronary artery disease (CAD) is the leading cause of mortality globally, primarily attributed by atherosclerosis, of which dyslipidaemia is one of the main risk factors. There is limited data in Malaysia on the prevalence and awareness of having dyslipidaemia, and the proportion who are treated.
Objectives: To investigate the prevalence of (1)dyslipidaemia; (2)awareness of dyslipidaemia and (3)subjects with dyslipidaemia who are treated.
Methodology: This was a cross-sectional study involving 11,525 Malaysian subjects from various rural and urban populations, with representations from the three major ethnic groups (age mean + SD: 52.6 + 11.3years; 6487 females, 5038 males). Clinical history and physical examinations were performed and fasting blood samples were collected for the measurement of lipid profiles. Dyslipidaemia was defined by mild, moderate or severe hypercholesterolaemia(HC):TC > 5.2, 6.5 and 7.8mmol/L respectively, or hypertriglyceridaemia (HTG):TG > 1.7 mmol/L, or low HDL-c:females < 1.3, males < 1.0mmol/L. Questionnaires were completed for data on awareness and treatment of dyslipidaemia.
Results: Subjects with HC and HTG were 66.9% and 40.9% respectively. Low HDL-c were found in 15.2% and 74.9% in females and males respectively. Among those with HC, 42.6%, 19.3% and 5.0% had mild, moderate and severe HC respectively. Awareness of dyslipidaemia was only found in 13.9% of the population, of whom only 8.0% were on treatment.
Conclusion: There is a high prevalence of dyslipidaemias in Malaysia, majority of whom are unaware of having the major risk factor for atherosclerosis-related complications such as CAD. Hence, there is an urgent need for coronary risk identification, prevention and intervention to combat the global epidemic of CAD
Introduction The prevalence of Type 2 diabetes mellitus (T2DM) is increasing worldwide
and many of these affected individuals remain unidentified. Undiagnosed
T2DM may impose substantial public health implications because these
individuals remain untreated and at risk for complications. The objective of
this study was to determine the national prevalence of undiagnosed T2DM
and to identify the associated risk factors.
Methods A nationwide cross-sectional study was conducted involving 17,783
respondents. Two-stage stratified sampling design was used to select a
representative sample of the Malaysian adult population. Structured
validated questionnaires with face to face interviews were used to obtain
data. Respondents, who claimed that they were not having diabetes, were
then asked to perform a fasting blood glucose finger-prick test by Accutrend
GC machine.
Results The prevalence of undiagnosed T2DM was 8.9% (n=1587). The highest
percentage of undiagnosed T2DM was found among males (10.2%), 55-59
years old (13.4%), highest education attainers of primary school (11.1%),
Indians (10.3%), married (10.3%), working (8.9%) and living in the urban
areas (9.2%). Multivariate analyses showed that factors associated with
undiagnosed T2DM were gender, age group, ethnicity, marital status, obesity
and hypertension.
Conclusion This study found an increasing trend of undiagnosed T2DM in Malaysia
compared to 2006. This finding is alarming as risk factors associated with
undiagnosed diabetes were related to most of the socio-demographic factors
studied. Therefore, early diabetic screening is crucial especially among adults
aged 30 and above to prevent more serious complications of this disease.
A new series of liquid crystals decorated gold nanoparticles is synthesized whose molecular architecture has azobenzenes moieties as the peripheral units connected to gold nanoparticles (Au NPs) via alkyl groups. The morphology and mesomorphic properties were investigated by field emission scanning electron microscope, high-resolution transmission electron microscopy, differential scanning calorimetry and polarizing optical microscopy. The thiolated ligand molecules (3a-c) showed enantiotropic smectic A phase, whereas gold nanoparticles (5a-c) exhibit nematic and smectic A phase with monotropic nature. HR-TEM measurement showed that the functionalized Au NPs are of the average size of 2nm and they are well dispersed without any aggregation. The trans-form of azo compounds showed a strong band in the UV region at ∼378nm for the π-π(∗) transition, and a weak band in the visible region at ∼472nm due to the n-π(∗) transition. These molecules exhibit attractive photoisomerization behaviour in which trans-cis transition takes about 15s whereas the cis-trans transition requires about 45min for compound 5c. The extent of reversible isomerization did not decay after 10 cycles, which proved that the photo-responsive properties of 5c were stable and repeatable. Therefore, these materials may be suitably exploited in the field of molecular switches and the optical storage devices.
There remains a need for newer therapeutic approaches to combat HIV/AIDS. Viral capsid protein p24 plays important roles in HIV pathogenesis. Peptides and small molecule inhibitors targeting p24 have shown to inhibit virus replication in treated cell. High specificity and biological stability of monoclonal antibodies (mAbs) make them an attractive contender for in vivo treatments. However, mAbs do not enter into cells, thus are restricted to target surface molecules. This also makes targeting intracellular HIV-1 p24 a challenge. A mAb specific to p24 that can internalize into the HIV-infected cells is hypothesized to inhibit the virus replication. We selected a mAb that has previously shown to inhibit p24 polymerization in an in vitro assay and chemically conjugated it with cell penetrating peptides (CPP) to generate cell internalizing anti-p24 mAbs. Out of 8 CPPs tested, κFGF-MTS -conjugated mAbs internalized T cells most efficiently. At nontoxic concentration, the κFGF-MTS-anti-p24-mAbs reduced the HIV-1 replication up to 73 and 49% in T-lymphocyte and PBMCs respectively. Marked inhibition of HIV-1 replication in relevant cells by κFGF-MTS-anti-p24-mAbs represents a viable strategy to target HIV proteins present inside the cells.
Numerous studies have shown the importance of physical activity in reducing the morbidity and mortality rates caused by cardiovascular disease (CVD). However, most of these studies emphasise little on the cumulative effect of CVD risk factors. Hence, this study investigates the association between physical exercise and cumulative CVD risk factors among adults in three different age groups.
In this work, the synthesis of silver nanoparticles from a pigment produced by a recently-discovered bacterium, Chryseobacterium artocarpi CECT 8497, was achieved, followed by an investigation of its anticancer properties. The bacterial pigment was identified as flexirubin following NMR ((1)H NMR and (13)C NMR), UV-Vis, and LC-MS analysis. An aqueous silver nitrate solution was treated with isolated flexirubin to produce silver nanoparticles. The synthesised silver nanoparticles were subsequently characterised by UV-Vis spectroscopy, Scanning Electron Microscopy (SEM), Energy Dispersive X-ray Spectroscopy (EDX), X-Ray Diffraction (XRD), and Fourier Transform Infrared (FTIR) Spectroscopy methodologies. Furthermore, the anticancer effects of synthesised silver nanoparticles in a human breast cancer cell line (MCF-7) were evaluated. The tests showed significant cytotoxicity activity of the silver nanoparticles in the cultured cells, with an IC50 value of 36μgmL(-1). This study demonstrates that silver nanoparticles, synthesised from flexirubin from C. artocarpi CECT 8497, may have potential as a novel chemotherapeutic agent.
The study aimed to evaluate nutraceutical potential of three commercially significant edible jellyfish species (Acromitus hardenbergi, Rhopilema hispidum and Rhopilema esculentum). The bell and oral arms of these jellyfishes were analyzed for their proximate composition, calorific value, collagen content, amino acid profile, chemical score and elemental constituent. In general, all jellyfish possessed low calorific values (1.0-4.9 kcal/g D.W.) and negligible fat contents (0.4-1.8 g/100 g D.W.), while protein (20.0-53.9 g/100 g D.W.) and minerals (15.9-57.2g/100g D.W.) were found to be the richest components. Total collagen content of edible jellyfish varied from 122.64 to 693.92 mg/g D.W., accounting for approximately half its total protein content. The dominant amino acids in both bell and oral arms of all jellyfish studied includes glycine, glutamate, threonine, proline, aspartate and arginine, while the major elements were sodium, potassium, chlorine, magnesium, sulfur, zinc and silicon. Among the jellyfish, A. hardenbergi exhibited significantly higher total amino acids, chemical scores and collagen content (p<0.05) compared to R. hispidum and R. esculentum. Having good protein quality and low calories, edible jellyfish is an appealing source of nutritive ingredients for the development of oral formulations, nutricosmetics and functional food.
In this study, the zinc oxide (ZnO) layer was synthesised on the surface of Zn plates by three different techniques, i.e. electrolysis, hydrogen peroxide and heat treatment. The synthesised ZnO layers were characterised using scanning electron microscopy, X-ray diffraction, UV-visible diffuse reflectance and photoluminescence spectroscopy. The photocatalytic activity of the ZnO layer was further assessed against methylene blue (MB) degradation under UV irradiation. The photocatalytic degradation of MB was achieved up to 84%, 79% and 65% within 1 h for ZnO layers synthesised by electrolysis, heat and hydrogen peroxide treatment, respectively. The reusability results show that electrolysis and heat-treated ZnO layers have considerable photocatalytic stability. Furthermore, the results confirmed that the photocatalytic efficiency of ZnO was directly associated with the thickness and enlarged surface area of the layer. Finally, this study proved that the ZnO layers synthesised by electrolysis and heat treatment had shown better operational stability and reusability.
Genetic structure can be altered by chemical mutagenesis, which is a common method applied in molecular biology and genetics. Second-generation sequencing provides a platform to reveal base alterations occurring in the whole genome due to mutagenesis. A model legume, Lotus japonicus ecotype Miyakojima, was chemically mutated with alkylating ethyl methanesulfonate (EMS) for the scanning of DNA lesions throughout the genome. Using second-generation sequencing, two individually mutated third-generation progeny (M3, named AM and AS) were sequenced and analyzed to identify single nucleotide polymorphisms and reveal the effects of EMS on nucleotide sequences in these mutant genomes. Single-nucleotide polymorphisms were found in every 208 kb (AS) and 202 kb (AM) with a bias mutation of G/C-to-A/T changes at low percentage. Most mutations were intergenic. The mutation spectrum of the genomes was comparable in their individual chromosomes; however, each mutated genome has unique alterations, which are useful to identify causal mutations for their phenotypic changes. The data obtained demonstrate that whole genomic sequencing is applicable as a high-throughput tool to investigate genomic changes due to mutagenesis. The identification of these single-point mutations will facilitate the identification of phenotypically causative mutations in EMS-mutated germplasm.
