    Recurrent boils are troublesome enough to children and their parents to be finally referred to the paediatricians. This study attempts to provide local data on epidemiology, pattern of infections of offending organism especially Staphylococcus aureus and underlying immunological abnormality. This condition does not appear to be related to socio economic status nor is staphylococcal nasal cariage a direct contributing factor. Defect in neutrophil function especially the respiratory burst assayed via chemiluminescence response to Staphylococcus aureus is closely associated with this condition. It is hoped that this data would be a further impetus for further research into this exciting field.

    Chemiluminescence assay was used to assess the respiratory burst activities of polymorpho-nuclear leukocytes from paediatric patients reported to manifest with several episodes of recurrent bacterial infections. From this group of patients evaluated, only 10 cases of severe recurrent bacterial abscess exhibited more than 80% depression in the phagocytic chemilumi-nescence responses. The assay, being a sensitive method, was able to provide further useful laboratory investigation in diagnosing 4 patients with chronic granulomatous disease.

    Chemiluminescence was evaluated as a diagnostic aid in the detection of oral cancer and potentially malignant epithelial lesions (PMELs) by comparing it against 1% tolonium chloride mouth rinse. Forty-six clinically identified lesions [14 primary squamous cell carcinoma (SCC), 26 PMELs and 6 benign lesions] and five cases of normal oral mucosa from 40 subjects (inclusive of 10 previously treated SCC cases) were examined with a commercial chemiluminescent kit (Vizilite) and tolonium chloride. Biopsy and histological verification of 31 lesions disclosed 14 SCC (45.2%), 10 epithelial dysplasias (32.3%), 5 lichen planus (16.1%) and 2 benign lesions (6.4%). For the remaining 15 lesions, a biopsy was not performed owing to patient's lack of consent or ill-health. The five cases of normal oral mucosa which tested negative for both tools were also not biopsied for ethical reasons. Sensitivity for Vizilite and tolonium chloride was 100% and 70.3%, respectively; and specificity was 14.2% for Vizilite and 25% for tolonium chloride. Their accuracy was 80.6% and 64.5%, respectively. Current findings suggest that chemiluminescence is a more reliable diagnostic tool than tolonium chloride in the detection of oral cancer and PMELs, and for follow-up of patients treated for the same.

    This research explores the possibility of using fluorescence technique to detect the presence of volatile organic compounds based on a single sensing material. The material used was TiO2 nanoparticles coated with porphyrin dye. The TiO2 nanoparticles colloid is in a sol-gel form synthesized from titanium (IV) ethoxide in ethanol with addition of kalium chloride (KCl) as stabilizer. TiO2 nanoparticles were then coated with porphyrin dye, Manganase (III) 5,10,15,20 tetra (4-pyridyl)-21H, 23H porphine chloride tetrakis (metachloride). The coated nanoparticles were deposited on quartz substrate using self-assembly through dip coating technique. The sensing properties of the thin film toward volatile organic compounds; ethanol, acetone, cyclohexane and 2-propanol were studied using luminescence spectrometer. It was found that the thin film produced different emission spectra peaks for different volatile organic compounds (VOCs). Hence, it eases chemical identification process and potentially be use as fluorescence gas sensor.

    Seven species of Mycena are reported as luminescent, representing specimens collected in Belize, Brazil, Dominican Republic, Jamaica, Japan (Bonin Islands), Malaysia (Borneo) and Puerto Rico. Four of them represent new species (Mycena luxaeterna, M. luxarboricola, M. luxperpetua, M. silvaelucens) and three represent new reports of luminescence in previously described species (M. aff. abieticola, M. aspratilis, M. margarita). Mycena subepipterygia is synonymized with M. margarita, and M. chlorinosma is proposed as a possible synonym. Comprehensive descriptions, illustrations, photographs and comparisons with phenetically similar species are provided. A redescription of M. chlorophos, based on analyses of type specimens and recently collected topotypical material, is provided. The addition of these seven new or newly reported luminescent species of Mycena brings the total to 71 known bioluminescent species of fungi.

    A series of Zn(II)-Schiff bases I, II and III complexes were synthesized by reaction of o-phenylenediamine with 3-methylsalicylaldehyde, 4-methylsalicylaldehyde and 5-methylsalicylaldehyde. These complexes were characterized using FT-IR, UV-Vis, Diffuse reflectance UV-Vis, elemental analysis and conductivity. Complex III was characterized by XRD single crystal, which crystallizes in the triclinic system, space group P-1, with lattice parameters a=9.5444(2) Å, b=11.9407(2) Å, c=21.1732(3) Å, V=2390.24(7) Å(3), D ( c )=1.408 Mg m(-3), Z=4, F(000)=1050, GOF=0.981, R1=0.0502, wR2=0.1205. Luminescence property of these complexes was investigated in DMF solution and in the solid state. Computational study of the electronic properties of complex III showed good agreement with the experimental data.

    Quorum sensing controls the virulence determinants in most proteobacteria. In this work, the hexane, chloroform and methanol extracts of an Ayurveda spice, namely clove (Syzygium aromaticum), shown anti-quorum sensing activity. Hexane and methanol extracts of clove inhibited the response of C. violaceum CV026 to exogenously supplied N-hexanoylhomoserine lactone, in turn preventing violacein production. Chloroform and methanol extracts of clove significantly reduced bioluminescence production by E. coli [pSB1075] grown in the presence of N-(3-oxododecanoyl)-L-homoserine lactone. We demonstrated that clove extract inhibited quorum sensing-regulated phenotypes in Pseudomonas aeruginosa PA01, including expression of lecA::lux (by hexane extract), swarming (maximum inhibition by methanol extract), pyocyanin (maximum inhibition by hexane extract). This study shows that the presence of natural compounds that exhibit anti-quorum sensing activity in the clove extracts may be useful as the lead of anti-infective drugs.

    Quorum sensing regulates bacterial virulence determinants, therefore making it an interesting target to attenuate pathogens. In this work, we screened edible, endemic plants in Malaysia for anti-quorum sensing properties. Extracts from Melicope lunu-ankenda (Gaertn.) T. G. Hartley, a Malay garden salad, inhibited response of Chromobacterium violaceum CV026 to N-hexanoylhomoserine lactone, thus interfering with violacein production; reduced bioluminescence expression of E. coli [pSB401], disrupted pyocyanin synthesis, swarming motility and expression of lecA::lux of Pseudomonas aeruginosa PAO1. Although the chemical nature of the anti-QS compounds from M. lunu-ankenda is currently unknown, this study proves that endemic Malaysian plants could serve as leads in the search for anti-quorum sensing compounds.

    The ethanol extract of Moringa oleifera Lam. leaves and its major constituents, crypto-chlorogenic acid, quercetin 3-O-glucoside and kaempferol 3-O-glucoside, were investigated on the respiratory burst of human whole blood and isolated human polymorphonuclear leukocytes (PMNs) using a luminol-based chemiluminescence assay. The chemotactic migration of PMNs was also investigated using the Boyden chamber technique. The ethanol extract demonstrated inhibitory activities on the oxidative burst and the chemotactic migration of PMNs. Quercetin 3-O-glucoside, crypto-chlorogenic acid, and kaempferol 3-O-glucoside, isolated from the extract, expressed relatively strong inhibitory activity on the oxidative burst of PMNs with IC50 values of 4.1, 6.7 and 7.0 microM, respectively, comparable with that of aspirin. They also demonstrated strong inhibition of chemotatic migration of PMNs with IC50 values of 9.5, 15.9 and 18.2 microM, respectively. The results suggest that M. oleifera leaves could modulate the immune response of human phagocytes, linking to its ethnopharmacological use as an anti-inflammatory agent. The immunomodulating activity of the plant was mainly due to its major components.

    Understanding the influence of co-dopants in the luminescence enhancement of carbonate glasses is the key issue in dosimetry. A series of borate glasses modified by lithium and potassium carbonate were synthesized by the melt-quenching method. The glass mixture activated with various concentrations of TiO2 and MgO was subjected to various doses of gamma-rays ((60)Co). The amorphous nature of the samples was confirmed by x-ray diffraction (XRD) spectra. The simple glowing curve of the glass doped with TiO2 features a peak at 230°C, whose intensity is maximal at 0.5 mol% of the dopant. The intensity of the glowing curve increases with the concentration of MgO added as a co-dopant up to 0.25 mol%, where it is two times higher than for the material without MgO thermoluminescence properties, including dose response, reproducibility, and fading were studied. The effective atomic number of the material was also determined. Kinetic parameters, such as kinetics order, activation energy, and frequency factor are estimated. The photoluminescence spectra of the titanium-doped glass consist of a prominent peaks at 480 nm when laser excitation at 650 nm is used. A three-fold photoluminescence enhancement and a blue shift of the peak were observed when 0.1% MgO was introduced. In addition, various physical parameters, such as ion concentration, polaron radius and internuclear distances were calculated. The mechanism for the thermoluminescence and photoluminescence enhancements are discussed.

    A series of novel isoxazole and pyrazoline derivatives has been synthesized and evaluated for their effects on the chemiluminescence and chemotactic activity of human phagocytes. Their effects on the chemotactic migration of isolated polymorphonuclear leukocytes (PMNs) and on the release of reactive oxygen species (ROS) during respiratory burst of human whole blood and PMNs were carried out using the Boyden chamber technique and luminol-based chemiluminescence assay, respectively. Of the compounds tested, compounds 8, 9, 11 and 12 exhibited higher inhibitory activity on the release of ROS (with IC50 values ranging from 5.6 to 8.4 μM) than acetylsalicylic acid (IC50 = 9.5 μ M). These compounds also showed strong inhibitory activity on the migration of PMNs with compound 8 exhibiting an IC50 value lower than that of ibuprofen. The results suggest that some of these isoxazole and pyrazoline derivatives have ability to modulate the innate immune response of phagocytes at different steps, indicating their potential as a source of new immunomodulatory agents.

    Polymers and organic materials that are exposed to sunlight undergo photooxidation, which leads to deterioration of their physical properties. To allow adequate performance under outdoor conditions, synthetic polymers require additives such as antioxidants and UV absorbers. A major problem with optimising polymer formulations to maximise their working life span is that accelerated weathering tests are empirical. The conditions differ significantly from real weathering situations, and samples require lengthy irradiation period. Degradation may not be apparent in the early stages of exposure, although this is when products such as hydroperoxides are formed which later cause acceleration of oxidation. A simple way of quantifying the number of free radicals presents in organic materials following exposure to light or heat is by measuring chemiluminescence (CL) emission. Most polymers emit CL when they undergo oxidative degradation, and it originates from the bimolecular reaction of macroperoxy radicals which creates an excited carbonyl.

    Two new synthesized and characterized quinazoline Schiff bases 1 and 2 were investigated for anticancer activity against MCF-7 human breast cancer cell line. Compounds 1 and 2 demonstrated a remarkable antiproliferative effect, with an IC50 value of 6.246×10(-6) mol/L and 5.910×10(-6) mol/L, respectively, after 72 hours of treatment. Most apoptosis morphological features in treated MCF-7 cells were observed by AO/PI staining. The results of cell cycle analysis indicate that compounds did not induce S and M phase arrest in cell after 24 hours of treatment. Furthermore, MCF-7 cells treated with 1 and 2 subjected to apoptosis death, as exhibited by perturbation of mitochondrial membrane potential and cytochrome c release as well as increase in ROS formation. We also found activation of caspases-3/7, -8, and -9 in compounds 1 and 2. Moreover, inhibition of NF-κB translocation in MCF-7 cells treated by compound 1 significantly exhibited the association of extrinsic apoptosis pathway. Acute toxicity results demonstrated the nontoxic nature of the compounds in mice. Our results showed significant activity towards MCF-7 cells via either intrinsic or extrinsic mitochondrial pathway and are potential candidate for further in vivo and clinical breast cancer studies.

    Structural and luminescence properties of borophosphate glasses with different modifier doped with transition metal ions have been investigated in this study. The glass sample from the series of xPb 30 4:0 .2Sb20 3:0.3B 20 3:(0 .5-x)P 20 5 and ySb 20 3:0 .2Pb 30 4:(0 .5-y)B 20 3:0 .3P 20 5where x 20 mol% and 0 y 0 .1 5mol% , respectively, were doped with 0.01 mol% of Fe 20 3 system have been prepared using the melt-quenching technique. The structural properties of samples had been studied using fourier transform infrared (FT-m) spectroscopy. The FT-IR study showed the network structure of the studied glasses based on the B 20 3-P 20 5host with the 5b203 and Pb203 modifiers. The results of FT-IR showed traces of B03 and B04 with the introduction of 5b203 and Pb203 modifiers. With the increasing content of B203 which replacing content of P205 in the glass network, the intensity of the borate band decreases and shifted to lower frequency. uv-Vis spectroscopy analysed the transition of Fe3+ from ground state to excited state in the ultraviolet spectral region. The photoluminescence of samples were studied using photoluminescence spectroscopy. The result of photoluminescence spectroscopy showed the effect of photoluminescence enhancement by doping Fe3+ as activator.

    Study of a series of lead lithium borophosphate glass samples was performed to determine the structural and luminescence properties. The glass samples containing the composition of 20Pb0-xLi20-30B 20 3-(50-x)P 20 5-2TiO 2 (where x = 0, 5, 10, 15, 20) system were prepared using melt-quenching technique. The Pb0-Li20-B 20 3-P20 5-Ti0 2 samples were investigated for structural properties using Fourier transform infrared and photoluminescence spectroscopy for studies of luminescence properties. The results from FTIR showed the presence of trigonal and tetrahedral PO4' PO3, BO4 and B03 groups within the host network structure. The samples exhibit luminescence emission centered at 420, 482 and 496 nm when excited at 300 nm wavelength. The emission peak at 420 nm were assigned to F center emission and charge-transfer transition of Ti4+ ions. The results showed that emission intensity was dependent on lithium contents in Ti-doped lead lithium borophosphate glass composition.

    With interest in the potential of optical fibres as the basis of next-generation thermoluminescence dosimeters (TLDs), the development of suitable forms of material and their fabrication has become a fast-growing endeavour. Present study focuses on three types of Ge-doped optical fibres with different structural arrangements and/or shapes, namely conventional cylindrical fibre, capillary fibre, and flat fibre, all fabricated using the same optical fibre preform. For doses from 0.5 to 8 Gy, obtained at electron and photon energies, standard thermoluminescence (TL) characteristics of the optical fibres have been the subject of detailed investigation. The results show that in collapsing the capillary fibre into a flat shape, the TL yield is increased by a factor of 5.5, the yield being also some 3.2 times greater than that of the conventional cylindrical fibre fabricated from the same perform. This suggests a means of production of suitably sensitive TLD for in-vivo dosimeter applications. Addressing the associated defects generating luminescence from each of the optical fibres, the study encompasses analysis of the TL glow curves, with computerized glow curve deconvolution (CGCD) and 2(nd) order kinetics.

    Study has been undertaken of the thermoluminescence (TL) yield of various tailor-made flat cross-section 6 mol% Ge-doped silica fibers, differing only in respect of external dimensions. Key TL dosimetric characteristics have been investigated, including glow curves, dose response, sensitivity, fading and reproducibility. Using a (60)Co source, the samples were irradiated to doses within the range 1 to 10 Gy. Prior to irradiation, the flat fibers were sectioned into 6 mm lengths, weighed, and annealed at 400 °C for 1 h. TL readout was by means of a Harshaw Model 3500 TLD reader, with TLD-100 chips (LiF:Mg, Ti) used as a reference dosimeter to allow the relative response of the fibers to be evaluated. The fibers have been found to provide highly linear dose response and excellent reproducibility over the range of doses investigated, demonstrating high potential as TL-mode detectors in radiation medicine applications. Mass for mass, the results show the greatest TL yield to be provided by fibers of the smallest cross-section, analysis indicating this to be due to minimal light loss in transport of the TL through the bulk of the silica medium.

    This study was carried out to investigate the suitability of using the optically stimulated luminescence dosimeter (OSLD) in measuring surface dose during radiotherapy. The water equivalent depth (WED) of the OSLD was first determined by comparing the surface dose measured using the OSLD with the percentage depth dose at the buildup region measured using a Markus ionization chamber. Surface doses were measured on a solid water phantom using the OSLD and compared against the Markus ionization chamber and Gafchromic EBT3 film measurements. The effect of incident beam angles on surface dose was also studied. The OSLD was subsequently used to measure surface dose during tangential breast radiotherapy treatments in a phantom study and in the clinical measurement of 10 patients. Surface dose to the treated breast or chest wall, and on the contralateral breast were measured. The WED of the OSLD was found to be at 0.4 mm. For surface dose measurement on a solid water phantom, the Markus ionization chamber measured 15.95% for 6 MV photon beam and 12.64% for 10 MV photon beam followed by EBT3 film (23.79% and 17.14%) and OSLD (37.77% and 25.38%). Surface dose increased with the increase of the incident beam angle. For phantom and patient breast surface dose measurement, the response of the OSLD was higher than EBT3 film. The in-vivo measurements were also compared with the treatment planning system predicted dose. The OSLD measured higher dose values compared to dose at the surface (Hp(0.0)) by a factor of 2.37 for 6 MV and 2.01 for 10 MV photon beams, respectively. The measurement of absorbed dose at the skin depth of 0.4 mm by the OSLD can still be a useful tool to assess radiation effects on the skin dermis layer. This knowledge can be used to prevent and manage potential acute skin reaction and late skin toxicity from radiotherapy treatments.

    The assessment of surface dose is essential in radiotherapy to avoid deterministic effect or to
    reduce the severity of side effects from radiation treatment. In this study, the surface dose for breast
    cancer radiotherapy was measured using two types of dosimeter; Thermoluminescent Dosimeter
    (TLD) and Optically Stimulated Luminescent Dosimeter (OSLD). The study was performed on the
    left breast of female Alderson Radiation Therapy (ART) phantom. The treatment planning was
    carried out on the ART phantom to determine the homogeneity of dose distribution within the target
    organ is complied with the tolerance limits of 95% to 107% as recommended by the International
    Commission on Radiation Units and Measurements (ICRU)’s Report No. 50. From the treatment
    planning result, the phantom then was irradiated with 267 cGy dose per fraction for two beam
    fields; medial tangential and lateral tangential fields using a 6 MV photon beam produced from
    three-dimensional (3D) conformal radiotherapy. Result shows that the OSLD provides 25.7% and
    23.5% higher surface dose compared to TLD for medial tangential and lateral tangential fields,
    respectively. This condition may be due to higher effective point of measurement and angular
    dependence of the OSLD compared to TLD. As a conclusion, suitable dosimeter should be selected
    to ensure accurate estimation of surface dose could be made thus reduction of skin reaction to
    patient could be achieved.
