    Andrographolide (AGP) and 14-deoxy-11,12-didehydroandrographolide (DDAG), two main diterpenoid constituents of Andrographis paniculata were previously shown to ameliorate asthmatic symptoms in a mouse model. However, due to inadequacies of both compounds in terms of drug-likeness, DDAG analogues were semisynthesised for assessment of their anti-asthma activity. A selected analogue, 3,19-diacetyl-14-deoxy-11,12-didehydroandrographolide (SRS27), was tested for inhibitory activity of NF-κB activation in TNF-α-induced A549 cells and was subsequently evaluated in a mouse model of ovalbumin (OVA)-induced asthma. Female BALB/c mice, 6-8weeks old were sensitized on days 0 and 14, and challenged on days 22, 23 and 24 with OVA. Compound or vehicle (3% dimethyl sulfoxide) was administered intraperitoneally 1h before and 11h after each OVA aerosol challenge. On day 25, pulmonary eosinophilia, airway hyperresponsiveness, mucus hypersecretion, inflammatory cytokines such as IL-4, -5 and -13 in BAL fluid, gene expression of inflammatory mediators such as 5-LOX, E-selectin, VCAM-1, CCL5, TNF-α, AMCase, Ym2, YKL-40, Muc5ac, CCL2 and iNOS in animal lung tissues, and serum IgE were determined. SRS27 at 30μM was found to suppress NF-κB nuclear translocation in A549 cells. In the ovalbumin-induced mouse asthma model, SRS27 at 3mg/kg displayed a substantial decrease in pulmonary eosinophilia, BAL fluid inflammatory cytokines level, serum IgE production, mucus hypersecretion and gene expression of inflammatory mediators in lung tissues. SRS27 is the first known DDAG analogue effective in ameliorating inflammation and airway hyperresponsiveness in the ovalbumin-induced mouse asthma model.

    Apoptosis is a tightly programmed cell suicide which occurs in multiple physiologic and pathological conditions where it plays an important role in tissue development and homeostasis by eliminating unwanted and damaged cells. Appropriate apoptosis signalling is crucial in maintaining the fine balance between cell death and cell survival in cancer. In response to death stimuli the morphology of the cell undergoes unique changes. The aim of this study was to examine and compare the changes in the cell surface morphology using scanning electron microscopy in HCS-2 cells, following 24 hour treatment with components of highly active antiretroviral therapy (HAART) at their clinical plasma concentrations. The cells were fixed in 2.5% Glutaraldehyde and post-fixed in 1% osmium tetroxide. The cells were then dehydrated through a graded series of alcohol and treated with hexamethyl-disilazane, then coated with a double layer of carbon. The cells were viewed under a Zeiss Ultra FEG Scanning Electron Microscope and a one way ANOVA and Tukey Kramer Post Hoc test was conducted based on the scoring of surface morphology of the cells using JMP 11 statistical software. The drugs used in this study induced morphological features which are known to be characteristic of apoptotic cell death. The drug combinations (ATP and LPV/r) were seemingly more effective than individual treatments in inducing cell death because morphological features observed were more advanced than those observed in individual treatments. However, LPV/r was more potent than ATP. In conclusion, HAART showed anticancer properties by inducing cell death through apoptosis.

    Combination between tissue engineering and other fields has brought an innovation in the area of regenerative medicine which ultimate aims are to repair, improve, and produce a good tissue construct. The availability of many types of scaffold, both synthetically and naturally have developed into many outstanding end products that have achieved the general objective in tissue engineering. Interestingly, most of this scaffold emulates extracellular matrix (ECM) characteristics. Therefore, ECM component sparks an interest to be explored and manipulated. The ECM featured in human amniotic membrane (HAM) provides a suitable niche for the cells to adhere, grow, proliferate, migrate and differentiate, and could possibly contribute to the production of angiogenic micro-environment indirectly. Previously, HAM scaffold has been widely used to accelerate wound healing, treat bone related and ocular diseases, and involved in cardiovascular repair. Also, it has been used in the angiogenicity study, but with a different technical approach. In addition, both side of HAM could be used in cellularised and decellularised conditions depending on the objectives of a particular research. Therefore, it is of paramount importance to investigate the behavior of ECM components especially on the stromal side of HAM and further explore the angiogenic potential exhibited by this scaffold.

    OBJECTIVE: Thrombocytopenia in dengue fever is a common and serious complication. However, no specific treatment is available for dengue fever induced thrombocytopenia. In few countries (Pakistan, Malaysia, Sri Lanka and other Asian countries) the leaf extract of Carica papaya has been effectively used for thrombocytopenia. So, the study is planned to access effect of Carica papaya leaf extract on platelet count in dengue fever patients.

    METHODS: All participants were randomised into two groups, study group and control group; the study group was given papaya leaf extract capsule of 500 mg once daily and routine supportive treatment for consecutive five days. The controls were given only routine supportive treatment. Daily complete blood counts, platelet counts and haematocrit level, liver function test, renal function test of both groups were observed.

    RESULTS: On the first day platelet count of study group and control group was (59.82±18.63, 61.06±20.03 thousands, p value 0.36). On the 2nd day platelet count of both study and control groups was not significantly different (61.67±19.46 and 59.93±19.52 thousands, p value 0.20) but on 3rd day platelet count of study group was significantly higher than control group (82.96±16.72, 66.45±17.36 thousands, p value < 0.01). On 4th and 5th day platelet count of study group (122.43±19.36 and 112.47±17.49 thousands respectively) was also significantly higher than the control group (88.75±21.65 and 102.59±19.35 thousands) (p value < 0.01). On 7th day platelet count of study group and control group were not significantly different (124.47±12.35 and 122.46±19.76 thousands respectively, p value 0.08). Average hospitalization period of study group v/s control group was 3.65±0.97 v/s 5.42±0.98 days (p value < 0.01). Average platelet transfusion requirement in study group was significantly less than control group (0.685 units per patient v/s 1.19 units per patient) (p value <0.01).

    CONCLUSIONS: It is concluded that Carica papaya leaf extract increases the platelet count in dengue fever without any side effect and prevents the complication of thrombocytopenia. So, it can be used in dengue fever with thrombocytopenia patients.

    Acute myeloid leukemia (AML) is one of the most frequent types of leukemia which mostly affects adult people. Resistance to therapeutic drugs is considered as a major clinical concern resulting in a weaker response to chemotherapy, disease relapse and decreased survival rate. Survivin, a member of Inhibitor of Apoptosis Proteins (IAPs), is associated with drug resistance and inhibition of apoptotic mechanisms in numerous hematological malignancies. In the present study, we examined the combined effect of etoposide and siRNA-mediated silencing of survivin on U-937 acute myeloid leukemia cells. The AML cells were transfected with survivin specific siRNA and gene knockdown was confirmed by quantitative real time PCR and western blotting. Subsequently, U-937 cells were assessed for response to etoposide treatment and apoptosis rate was measured with flowcytometery. The cytotoxic effects in siRNA-etoposide group were measured and compared to etoposide single therapy group. Survivin siRNA effectively knocked down the mRNA and protein levels of survivin, which led to lower cell proliferation and enhanced apoptosis. Furthermore, combined treatment of etoposide and survivin siRNA synergistically increased the cell toxic effects of etoposide and its ability to induce apoptosis.

    OBJECTIVE: This study was conducted to investigate the effects of low-level laser (LLL) doses on human red blood cell volume. The effects of exposure to a diode pump solid state (DPSS) (λ = 405 nm) laser were observed.

    BACKGROUND DATA: The response of human blood to LLL irradiation gives important information about the mechanism of interaction of laser light with living organisms. Materials and methods Blood samples were collected into ethylenediaminetetraacetic acid (EDTA)-containing tubes, and each sample was divided into two equal aliquots, one to serve as control and the other for irradiation. The aliquot was subjected to laser irradiation for 20, 30, 40, or 50 min at a fixed power density of 0.03 W/cm(2). Mean cell volume (MCV) and red blood cell (RBC) counts were measured immediately after irradiation using a computerized hemtoanalyzer.

    Steroid-induced hypertension and glaucoma is associated with increased extracellular meshwork (ECM) deposition in trabecular meshwork (TM). Previous studies have shown that single drop application of trans-resveratrol lowers IOP in steroid-induced ocular hypertensive (SIOH) rats. This IOP lowering is attributed to activation of adenosine A1 receptors, which may lead to increased matrix metalloproteinase (MMP)-2 activity. This study evaluated the effect of repeated topical application of trans-resveratrol for 21 days in SIOH animals on IOP, changes in MMP-2 level in aqueous humor, trabecular meshwork and retinal morphology and retinal redox status. We observed that treatment with trans-resveratrol results in significant and sustained IOP reduction in SIOH rats. This IOP reduction is associated with significantly higher aqueous humor total MMP-2 level; significantly reduced TM thickness and increased number of TM cells. Treatment with trans-resveratrol also significantly increased ganglion cell layer (GCL) thickness, the linear cell density in the GCL and inner retina thickness; and significantly reduced retinal oxidative stress compared to the SIOH vehicle-treated group. In conclusion, repeated dose topical application of trans-resveratrol produces sustained IOP lowering effect, which is associated with increased level of aqueous humor MMP-2, normalization of TM and retinal morphology and restoration of retinal redox status.

    Marudu Bay, north coast of Sabah is characterized with mesotrophic water body and typical environmental parameters
    throughout the year. The current study was undertaken to evaluate the effect of environmental parameters and nutrients
    in mesotrophic water on the occurrence and distribution of potentially harmful phytoplankton species. The samplings
    were conducted over a period of thirteen months, covering southwest monsoon (SWM), inter-monsoon (IM), and northeast
    monsoon (NEM), at ten stations throughout the bay. Physical parameters (temperature, salinity, pH, dissolved oxygen,
    current speed and secchi depth), biological parameters (cell densities of phytoplankton) and chemical parameters
    (phosphate, nitrate, silicate and ammonia) were examined. The results indicated at least eight potentially harmful
    phytoplankton species (Dinophysis caudata, D. miles, Ceratium furca, C. fursus, Prorocentrum micans, P. sigmoides, P.
    triestinum and Pseudo-nitzschia sp.) were detected in north coast of Sabah. However, the potentially harmful phytoplankton
    species contributed only about 1.3% of the total phytoplankton community. Under nutrient deprivation conditions, the
    potentially harmful phytoplankton species distribution was mainly influenced by the ability to utilize other nitrogen
    sources, cell mobility and toleration to low nutrients environments.

    The terrestrial Ludisia discolor, also referred to as the jewel orchid is prized for
    the quality of its leaves. L. discolor is known as a medicinal herb and is touted for its heatand
    pathogen-resisting qualities. L. discolor is valuable in the production of both flavonoids
    and anthocyanins, antioxidants that are exalted in the health industry. Plant cell cultures
    have emerged as alternative sources of anthocyanin production. Plant protoplast cultures
    are used frequently in transient gene expression studies and in the establishment of callus
    and cell suspension cultures. Benefits of plant protoplast system include similarity to cells
    found in plant tissues, reproduction under controlled conditions, and prevention of masking
    of stress responses to previous handling techniques. A study was conducted to assess the
    amenability of the stem and leaves of L. discolor to protoplast isolation. The stem and leaf
    segments were weighed, sliced into thin layers, immersed in a digestion medium, washed
    and then cultured onto a recovery medium. Results indicated that the production of plant
    protoplasts from L. discolor may be viewed as an alternative in the generation of cell
    cultures and ultimately in the production of anthocyanins from the cell cultures.

    Haemophagocytic lymphohistiocytosis (HLH) is a clinico-pathologic entity caused by increased proliferation
    and activation of benign macrophages with haemophagocytosis throughout the reticulo-endothelial system.
    Virus-associated HLH is a well-recognised entity. Although majority of parvovirus B19 associated HLH does not
    require any specific treatment and carries good prognosis, outcome of children is worse than adults. We report
    here a case of HLH associated with acute parvovirus B19 infection in a young healthy patient with underlying
    hereditary spherocytosis, with bone marrow findings typical of parvovirus infection. Although this patient
    had spontaneous recovery of cell counts, he succumbed due to complication from prolonged ventilation.
    Unexpectedly, his immunoglobulin levels were inappropriately normal despite on-going ventilator associated
    pneumonia, which reflects inadequate humoral immune response towards infection.

    A 22-year-old Indian female was referred to Sg Buloh hospital with the diagnosis of bilateral keratoconus. On examination, slit lamp biomicroscopy and corneal topography revealed stage 3 keratoconus in the right eye and stage 2 keratoconus in the left eye. Corneal cell morphology in both eyes was evaluated using confocal microscope. In qualitative observation, almost all corneal layers in right eye except endothelium were partially or completely obscured by haze. Additionally, morphological alterations, such as elongation of keratocyte nuclei and cluster of cells, and dark bands in the anterior stroma were observed in right eye. In the left eye, the amount of haze was less, allowing better visibility of the corneal layers compared with the right eye. The dark bands were evident in the posterior stroma. Quantitative analysis showed that anterior and posterior stromal keratocyte density and endothelium cell density were relatively low in the right eye (834.0, 700.5, and 2,133 cells/mm2, respectively) compared with the left eye (934.1, 750.6, and 2,361 cells/mm2, respectively). In this case, the right eye, exhibiting stage 3 keratoconus, showed more morphological alteration, particularly in the anterior stroma compared with the left eye with stage 2 keratoconus. Increased severity of the disease can explain these differences in corneal cell morphology.

    Two experiments were conducted to develop Acartia erythraea and Oithona brevicornis cultures: The performance of five microalgal diets to produce nauplii, copepodites and adults of A. erythraea; and the performance of the same diets to produce nauplii, copepodites and adults of O. brevicornis. The five different microalgal diets were Isochrysis galbana (IG), Chaetoceros affinis (CA), Chlorella marina (CM), Nannochloropsis oculata (NO) and mixed algae (mixture of IG, CA, CM and NO at an equal abundance to provide the exact cell density). The results indicated that the abundance of both A. erythraea and O. brevicornis was higher in tanks supplied with IG and mixed algae than the tanks supplied with CA, CM and NO. IG and mixed algal diets were statistically similar on the mean abundance for both A. erythraea and O. brevicornis. The maximum production of A. erythraea nauplii was observed on day 12 of culture period and the nauplii production decreased from day 13 onwards. The mean abundance of A. erythraea copepodites and adults increased along with time up to the end of the culture period. In the case of O. brevicornis nauplii, the maximum abundance was observed on day 9 day of culture period and the nauplii production decreased from day 10 onwards. The mean abundance of O. brevicornis copepodites and adults increased gradually from the beginning to the end of the culture period. Under the experimental conditions of this study, both IG and mixed algal diets can be recommended for the best growth performance of A. erythraea and O. brevicornis.

    A comprehensive survey and descriptions on the petiole anatomy was undertaken on 16 species belonging to four selected genera (Anisoptera, Cotylelobium, Vatica and Upuna) in the tribe Dipterocarpeae to investigate variations in vascular bundle structure. Methods used in this study were petiole sectioning using a sliding microtome, staining, mounting and observation under light microscope. Findings had shown that all species studied have complex vascular bundle structures, consisting of outer and medullary vascular bundles. Four different types of vascular bundle arrangements were found and described as Types 1, 2, 3 and 4. The vascular bundle types were determined based on the arrangement and system of vascular bundle strands present in the petiole transverse sections. These vascular bundle types were determined and suggested for easy reference. The results showed that vascular bundles can be used for identification of certain species and can definitely be used for classification of different genera. The presence of sclerenchyma cells is useful in differentiating genera. As a conclusion, the petiole vascular bundle anatomical characteristics have taxonomic value, especially in the identification of some species and possibly applicable in the classification of the tribe Dipterocarpeae.

    The effect of addition of avocado (Persea americana) puree on the physical and microstructure of butter cake was studied.
    Butter cakes were made by replacing butter with 10, 30 and 50% of avocado puree. Physical properties including batter
    specific gravity, volume, colour and image analysis of cellular structure of the crumb were analyzed. Texture profile
    analysis was determined using texture analyzer. The results showed that with the increased amount of avocado puree,
    the batter specific gravity increased while volume of the cakes reduced. The texture profile analysis showed that the cakes
    became harder as the amount of avocado puree increased, while cohesiveness was not affected. The cellular structure of
    the crumb exhibited a decrease in the number of air cells while the average cell size increased with addition of avocado
    puree. The colour analysis showed that the cake crumb became darker as the aavocado puree was increased.

The quasi-independence model, which is a generalization of the independence model, is most commonly model used to analyze incomplete contingency tables. Goodness of fit tests of the quasi-independence model are usually based on Pearson chi square test statistic and likelihood ratio test statistic. In power divergence statistics family, the selection of power divergence parameter is of interest in multivariate discrete data. In this study, a simulation study is conducted to evaluate the performance of the power divergence statistics under quasi independence model for particular power divergence parameters in terms of power values.

    As a natural anti-oxidant source, Tualang honey, produced by wild bees nesting on the Tualang tree (Koompassia excelsa) is expected to have positive influence on health, including memory. This study investigated the effect of Tualang honey on the cell count of memory formation related hippocampal pyramidal neuron and on spatial memory performance (SMP) of rats using the radial arm maze (RAM). Sprague Dawley male rats (n=24), 7-8 weeks old were divided into two groups; experimental group group force-fed 1 mL/100 g body weight with 70% honey (HG); and the control group with 0.9% saline (CG) for 12 weeks. Nissl staining technique (with cresyl violet) was employed for neurohistological analysis of the hippocampal tissue. Six randomly selected rats from each group were used for the neuronal soma counting of pyramidal cell layer CA1, CA3a and CA3c regions. Two-way ANOVA analysis showed positively significant differences between treatment and control groups for SMP comparison of working memory and reference memory components, as well as the number of pyramidal neurons. Hence, this positive effects of Tualang honey, as demonstrated behaviorally and neurohistologically, supported report that Tualang honey could improve memory and deter hippocampal morphological impairments; possibly due to its high anti-oxidant properties.

    Multidrug resistance poses a great challenge to cancer treatment. In order to improve the targeting and codelivery of small interfering RNA (siRNA) and doxorubicin, and to overcome multidrug resistance, we conjugated a cholic acid-polyethylenimine polymer with folic acid, forming CA-PEI-FA micelles. CA-PEI-FA exhibited a low critical micelle concentration (80 μM), small average particle size (150 nm), and positive zeta potential (+ 12 mV). They showed high entrapment efficiency for doxorubicin (61.2 ± 1.7%, w/w), forming D-CA-PEI-FA, and for siRNA, forming D-CA-PEI-FA-S. X-ray photoelectron spectroscopic analysis revealed the presence of external FA on D-CA-PEI-FA micelles. About 25% doxorubicin was released within 24 h at pH 7.4, while more than 30% release was observed at pH 5. The presence of FA enhanced micelle antitumor activity. The D-CA-PEI-FA and D-CA-PEI-FA-S micelles inhibited tumor growth in vivo. No significant differences between their in vitro cytotoxic activities or their in vivo antitumor effects were observed, indicating that the siRNA coloading did not significantly increase the antitumor activity. Histological analysis revealed that tumor tissues from mice treated with D-CA-PEI-FA or D-CA-PEI-FA-S showed the lowest cancer cell density and the highest levels of apoptosis and necrosis. Similarly, the livers of these mice exhibited the lowest level of dihydropyrimidine dehydrogenase among all treated groups. The lowest serum vascular endothelial growth factor level (VEGF) (24.4 pg/mL) was observed in mice treated with D-CA-PEI-FA-S micelles using siRNA targeting VEGF. These findings indicated that the developed CA-PEI-FA nanoconjugate has the potential to achieve targeted codelivery of drugs and siRNA.

    A vaccine against human enterovirus 71 (EV-A71) is urgently needed to combat outbreaks of EV-A71 and in particular, the serious neurological complications that manifest during these outbreaks. In this study, an EV-A71 virus-like-particle (VLP) based on a B5 subgenogroup (EV-A71-B5 VLP) was generated using an insect cell/baculovirus platform. Biochemical analysis demonstrated that the purified VLP had a highly native procapsid structure and initial studies in vivo demonstrated that the VLPs were immunogenic in mice. The impact of VLP immunization on infection was examined in non-human primates using a VLP prime-boost strategy prior to EV-A71 challenge. Rhesus macaques were immunized on day 0 and day 21 with VLPs (100 μg/dose) containing adjuvant or with adjuvant alone (controls), and were challenged with EV-A71 on day 42. Complete blood counts, serum chemistry, magnetic resonance imaging (MRI) scans, and histopathology results were mostly normal in vaccinated and control animals after virus challenge demonstrating that the fatal EV-A71-B3 clinical isolate used in this study was not highly virulent in rhesus macaques. Viral genome and/or infectious virus were detected in blood, spleen or brain of two of three control animals, but not in any specimens from the vaccinated animals, indicating that VLP immunization prevented systemic spread of EV-A71 in rhesus macaques. High levels of IgM and IgG were detected in VLP-vaccinated animals and these responses were highly specific for EV-A71 particles and capsid proteins. Serum from vaccinated animals also exhibited similar neutralizing activity against different subgenogroups of EV-A71 demonstrating that the VLPs induced cross-neutralizing antibodies. In conclusion, our EV-A71-B5 VLP is safe, highly immunogenic, and prevents systemic EV-A71-B3 infection in nonhuman primates making it a viable attractive vaccine candidate for EV-A71.

    In order to assess the feasibility of various carbon sources and plant materials in increasing the growth rate and biomass productivity of Monoraphidium griffithii, ten carbon sources as well as six plant materials were tested in mixotrophic cultures with or without aeration. It was found that glucose, fructose, maltose, sodium acetate and mannitol were potential carbon sources for growth enhancement of M. griffithii. Supplementation of culture medium with these carbon sources resulted in approximately 1-4-fold increase in cell density compared to control in a small scale culture. In a larger scale mixotrophic culture with aeration, 0.05% mannitol and 0.1% fructose resulted in a decent 1-1.5-fold increase in final cell density, approximately 2-fold increase in growth rate and 0.5-1-fold increase in dry biomass weight. Findings from this study suggests that glucose, fructose, maltose and mannitol were potential organic carbon sources for mixotrophic culture of M. griffithii.

    Chitosan nanoparticles (CSNPs) have potential applications in stem cell research. In this study, ex vivo cytotoxicity of CSNPs on mouse bone marrow-derived (MBMCs) hematopoietic stem and progenitor cells (HSPCs) was determined. MBMCs were exposed to CSNPs of different particle sizes at various concentrations for up to 72 h. Cytotoxicity effect of CSNPs on MBMCs was determined using MTT, Live/Dead Viability/Cytotoxicity assays and flow cytometry analysis of surface antigens on HSCs (Sca-1(+)), myeloid-committed progenitors (CD11b(+), Gr-1(+)), and lymphoid-committed progenitors (CD45(+), CD3e(+)). At 24 h incubation, MBMCs' viability was not affected by CSNPs. At 48 and 72 h, significant reduction was detected at higher CSNPs concentrations. Small CSNPs (200 nm) significantly reduced MBMCs' viability while medium-sized particle (∼400 nm) selectively promoted MBMCs growth. Surface antigen assessment demonstrated lineage-dependent effect. Significant decrease in Sca-1(+) cells percentage was observed for medium-sized particle at the lowest CSNPs concentration. Meanwhile, reduction of CD11b(+) and Gr-1(+) cells percentage was detected at high and intermediate concentrations of medium-sized and large CSNPs. Percentage of CD45(+) and CD3e(+) cells along with ROS levels were not significantly affected by CSNPs. In conclusion, medium-sized and large CSNPs were relatively non-toxic at lower concentrations. However, further investigations are necessary for therapeutic applications.
