    Sea cucumbers have high economic value, and in most forms of trade, their body wall is typically the only part that is harvested and sold. The organs of the sea cucumber, collectively known as the viscera, are frequently discarded, contributing to land and water pollution. However, discarded sea cucumber viscera contain various nutrients that can be used in many applications. Therefore, this review highlights the biological and economic aspects of sea cucumbers, followed by a critical discussion of the nutritional value of their internal organs and possible applications, including as functional feed additives in the aquaculture industry, sources of natural testosterone for application in sex reversal and production of monosex population, of neuroprotective agents against central nervous system disorders and of cosmetic ingredients, especially for skin whitening and anti-ageing products. The review further highlights the valorisation potential of viscera to maximize their economic potential, thus providing an enormous prospect for reusing sea cucumber waste, thereby reducing the negative impact of the sea cucumber fishery sector on the environment.

    Small-scale fisheries present challenges to management due to fishers' dependency on resources and the adaptability of management systems. We compared social-ecological processes in the sea cucumber fisheries of Zanzibar and Mayotte, Western Indian Ocean, to better understand the reasons for resource conservation or collapse. Commercial value of wild stocks was at least 30 times higher in Mayotte than in Zanzibar owing to lower fishing pressure. Zanzibar fishers were financially reliant on the fishery and increased fishing effort as stocks declined. This behavioral response occurred without adaptive management and reinforced an unsustainable fishery. In contrast, resource managers in Mayotte adapted to changing fishing effort and stock abundance by implementing a precautionary fishery closure before crossing critical thresholds. Fishery closure may be a necessary measure in small-scale fisheries to preserve vulnerable resources until reliable management systems are devised. Our comparison highlighted four poignant lessons for managing small-scale fisheries: (1) diagnose the fishery regularly, (2) enable an adaptive management system, (3) constrain exploitation within ecological limits, and (4) share management responsibility.

    Di Malaysia, timun laut lebih dikenali sebagai Gamat. Di kalangan kaum Melayu,
    gamat sering digunakan sebagai ubat tradisional untuk melegakan kesakitan,
    merawat luka dan kesan terbakar. Ianya juga digunakan sebagai tonik untuk
    memberi sumber tenaga tambahan. Stichopus chloronotus merupakan salah satu
    spesies timun laut yang boleh didapati di Malaysia. Kajian ini bertujuan untuk
    menentukan dos optimum ekstrak akues Stichopus chloronotus emulsi salap ke
    atas luka pada model tikus. Beberapa siri kepekatan iaitu 0.1%, 0.5% dan 1%
    ekstrak akues Stichopus chloronotus emulsi salap diberikan ke atas luka eksisi sekali
    sehari selama 10 hari. Perubahan pada kawasan luka diukur dengan menggunakan
    angkup dan gambar luka diambil pada hari pertama, ke-3, ke-6, ke-8 dan ke-10
    selepas pembentukan luka. Keputusan daripada peratusan pengurangan luka dan
    pemerhatian makroskopik akan menentukan dos optimum Stichopus chloronotus.
    Hasil kajian menunjukkan, kumpulan tikus kajian yang menerima rawatan
    Stichopus chloronotus 0.5% mempunyai peratusan pengurangan luka yang lebih
    tinggi dan pemerhatian makroskopik yang lebih baik bermula dari hari ke-6 selepas
    pembentukan luka berbanding kumpulan yang lain. Kesimpulannya, dos 0.5%
    merupakan kepekatan optimum bagi Stichopus chloronotus memberikan kesan
    kepada penyembuhan luka dan akan digunakan pada kajian sebenar

    Fatty acid profile from crude extracts of local sea cucumber Stichopus chloronotus was determined using gas chromatography (GC) technique. The extracts were prepared separately in methanol, ethanol, phosphate buffer saline (PBS), and distilled water as part of our study to look at the affinity of these solvents in extracting the lipid from sea cucumber. The PBS and distilled water extractions indicate water-soluble components, while the organic fractions are extracted in methanol and ethanol as organic solvents. Furthermore, water extraction is the conventional method practiced in Malaysia. In our analysis the C14:0 (myristic), C16:0 (palmitic), C18:0 (stearic), C18:2 (linoleic), C20:0 (arachidic), and C20:5 (eicosapentaenoic, EPA) were significantly different (p < 0.01) in the four solvent extractions. However, the PBS extraction contained a much higher percentage of EPA (25.69%) compared to 18.89% in ethanol, 7.84% in distilled water, and only 5.83% in methanol, and variances were significantly different (p < 0.01 ). On the other hand, C22:6 (docosahexaenoic acid or DHA) is much higher in water extraction (57.55%), in comparison to the others where only 3.63% in PBS and 1.20% in methanol, and this difference is significant at p < 0.01. No DHA was detected in ethanol extractions. Subsequently, C18:1 (oleic acid) was only detected in PBS (21.98%) and water extraction (7.50%). It is interesting that palmitic acid, C16:() was higher in methanol (20.82%) and ethanol (2.18%), while 12.55% was detected in PBS and only 2.20% in water extraction: and again this was significantly different at p < 0.01. Although our results have shown that all four solvents were different in terms of their ability to extract fatty acids, the major component for tissue repair was well preserved. Probably this is one of the important precocious steps when working with a delicate sea cucumber, in both experimental and/or at the preparative stages. Freshness of the sea cucumber samples is important when undertaking this type of experiment. Finally, we believe that the local sea cucumber S. chloronotus contains all the fatty acids required to play a potential active role in tissue repair.

    1. Three species of sea cucumbers found in the Sabah coastal areas were screened for the presence of antibacterial activity using three methods of extraction. Tests were conducted in vitro using the agar absorption method. 2. Both the lipid extract and the methanol-solvent extract from Holothuria atra, Holothuria scabra and Bohadshia argus were found to show no antibacterial activity. 3. Phosphate-buffered saline (PBS) from H. atra and B. argus, however, inhibited the growth of all gram-positive and gram-negative bacteria. 4. Comparisons were also made between extracts from the outer layer of H. atra and its inner part, and it was found that the extract from the outer layer showed less bacterial growth inhibition property. 5. The bacterial growth inhibition property of the PBS extract from H. atra, however, is dependent on the extract's concentration. Bacterial growth inhibition was apparent after 48 hr incubation.

    Five sea cucumber species including one new species of the genus Stichopus are reported from the shallow coral reefs of Straits of Malacca. The new species Stichopus fusiformiossa has unusual fusiform spicules in the tentacles, which are not found in the other species of the genus. Pseudo-tables and large perforated plates are newly recorded for Stichopus hermanni Semper, 1868 and Stichopus vastus Sluiter, 1887, respectively.

    Land reclamation in Penang began two decades ago and is still rampant with
    large reclamation projects planned to be executed in the near future. The present study
    provides the first information on effects of land reclamation in Penang towards the coastal
    macroinvertebrates. This study assessed the abundance, diversity, and evenness of
    coastal invertebrates assumed to be the foremost affected when land is reclaimed. Three
    kinds of areas were focused on: reclaimed, unclaimed (adjacent to reclaimed), and
    undisturbed. A total of 53 species of macroinvertebrates from 10 classes (Gastropoda,
    Bivalvia, Polychaeta, Malacostraca, Maxillopoda, Echinoidea, Polyplacophora,
    Branchiopoda, Scaphopoda, and Holothuroidea) were sampled. Reclaimed areas were
    moderately rich in species averaging 11 species compared to 7 species in adjacent and
    14 in undisturbed areas. Species richness was the highest in Teluk Aling (an undisturbed
    area) with 22 species, and was the lowest in Gurney Drive (an adjacent area) with 2
    species. The average species diversity and evenness on reclaimed land was the lowest
    with values of 1.9974 and 0.5787, respectively. The diversity was higher by 5.07% in
    adjacent areas and by 22.92% in undisturbed areas compared to reclaimed areas.
    Species evenness was 29.75% higher in unreclaimed areas and 17.87% higher in
    undisturbed areas compared to reclaimed areas. Land reclamation reduces species
    diversity and evenness, and to a lesser extent, species richness.

    Sea cucumbers, blind cylindrical marine invertebrates that live in the ocean intertidal beds have more than thousand species available of varying morphology and colours throughout the world. Sea cucumbers have long been exploited in traditional treatment as a source of natural medicinal compounds. Various nutritional and therapeutic values have been linked to this invertebrate. These creatures have been eaten since ancient times and purported as the most commonly consumed echinoderms. Some important biological activities of sea cucumbers including anti-hypertension, anti-inflammatory, anti-cancer, anti-asthmatic, anti-bacterial and wound healing. Thus, this short review comes with the principal aim to cover the profile, taxonomy, together with nutritional and medicinal properties of sea cucumbers.

    The unsustainable harvesting of sea cucumbers in the Straits of Malacca poses a danger of collapsing population of this marine resource. Recent survey revealed the absence of commercially important sea cucumber species like Stichopus horrens was alarming since there were a lot of taxonomical complications in identifying species from this genus. The knowledge of taxonomy and ecology is an integral part in determining resource management strategies and conservation of marine resources like sea cucumber.

    The calcareous rings of two species of local sea cucumbers, Stichopus hermanni Semper and Holothuria atra Jaeger, were located, dissected, exposed and recorded. The calcareous rings of both species each composed of ten plates knit together by connective tissues. The radial plate of H. atra was a square with a notch at the anterior part whereas in S. hermanni it was squarish consisting of 4 ridges in the anterior part with a notch at the posterior part. The interradial plates of both species were smaller than the radial plates. A ridge was present at the anterior part of the interradial plate in H. atra. In S. hermanii, the ridge at the anterior part of the interradial was thin and prominent. Under the scanning electron microscope the calcareous rings from the two species exhibited a mosaic of small, numerous spicules bound to each other.

    Atratoxin B1 which was extracted from a local sea cucumber, Holothuria atra is a potential antifungal agent against dermatophytes. Exposure of Microsporum canis mycelia inoculated into Sabouraud glucose agar medium to the 10 mg/ml atratoxin B1 solution, resulted in complete suppression of the fungal growth. Prolonged exposure to the atratoxin B] (72 hours) resulted with necrosis in a substantial portion of the existing hyphae. The main anomalies noted were the abnormally shaped hyphae and the alterations in its morphology and cytology.

Pendedahan yang lebih lama kepada larutan atratoksin B] (72 jam) mengakibatkan nekrosis berlaku pada sebahagian besar hifanya. Keganjilan yang jelas berlaku adalah ketidaknormalan pada bentuk hifanya dan juga pengubahsuaian yang terjadi pada morfologi dan sitologinya.

    The stone fish (Actinopyga lecanora) ethanolic and methanolic tissue extracts were investigated for total phenolic contents (TPCs) as well as antioxidant activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH(•)) radical scavenging activity and ferric reducing antioxidant power (FRAP) assays. Both extracts showed low amount of phenolics (20.33 to 17.03 mg of gallic acid equivalents/100 g dried sample) and moderate antioxidant activity (39% to 34% DPPH(•) radical scavenging activity and 23.95 to 22.30 mmol/100 mL FeSO4 FRAP value). Enzymatic proteolysis was carried out in order to improve the antioxidant activity using six commercially available proteases under their optimum conditions. The results revealed that the highest increase in antioxidant activity up to 85% was obtained for papain-generated proteolysate, followed by alcalase (77%), trypsin (75%), pepsin (68%), bromelain (68%), and flavourzyme (50%) as measured by DPPH(•) radical scavenging activity, whilst for the FRAP value, the highest increase in the antioxidant activity up to 39.2 mmol/100 mL FeSO4 was obtained for alcalase-generated proteolysate, followed by papain (29.5 mmol/100 mL FeSO4), trypsin (23.2 mmol/100 mL FeSO4), flavourzyme (24.7 mmol/100 mL FeSO4), bromelain (22.9 mmol/100 mL FeSO4), and pepsin (20.8 mmol/100 mL FeSO4). It is obvious that proteolysis of stone fish tissue by proteolytic enzymes can considerably enhance its antioxidant activity.

    Marine sources have been attracting the attention of scientists and manufacturers worldwide hoping to find new alternatives for biological active substances. Promising new research indicates that sea cucumber, which is slug-like in appearance and has been a staple in Japan, China and other parts of East Asia since ancient times, is beginning to gain popularity as a dietary supplement in western countries. The roles of sea cucumber extracts in various physiological functions have spurred researchers to investigate the ability of sea cucumber to be an alternative in neutraceutical and medical applications. This article provides a brief introduction to sea cucumber and reviews its numerous bioactive compounds, such as triterpene glycosides, glycosaminoglycans, gangliosides, collagen, branched-chain fatty acid and lectins, which serve as potential sources of neutraceutical, pharmaceutical and cosmetic agents, thus providing a new platform in biochemical research.

    Actinopyga lecanora (A. lecanora) is classified among the edible species of sea cucumber, known to be rich in protein. Its hydrolysates were reported to contain relatively high antioxidant activity. Antioxidants are one of the essential properties in cosmeceutical products especially to alleviate skin aging. In the present study, pH, reaction temperature, reaction time and enzyme/substrate ratio (E/S) have been identified as the parameters in the papain enzymatic hydrolysis of A. lecanora. The degree of hydrolysis (DH) with antioxidant activities of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric-reducing antioxidant power (FRAP) assays were used as the responses in the optimization. Analysis of variance (ANOVA), normal plot of residuals and 3D contour plots were evaluated to study the effects and interactions between parameters. The best conditions selected from the optimization were at pH 5.00, 70 °C of reaction temperature, 9 h of hydrolysis time and 1.00% enzyme/substrate (E/S) ratio, with the hydrolysates having 51.90% of DH, 42.70% of DPPH activity and 109.90 Fe2+μg/mL of FRAP activity. The A. lecanora hydrolysates (ALH) showed a high amount of hydrophobic amino acids (286.40 mg/g sample) that might be responsible for antioxidant and antityrosinase activities. Scanning electron microscopy (SEM) image of ALH shows smooth structures with pores. Antityrosinase activity of ALH exhibited inhibition of 31.50% for L-tyrosine substrate and 25.40% for L-DOPA substrate. This condition suggests that the optimized ALH acquired has the potential to be used as a bioactive ingredient for cosmeceutical applications.

    Collagen was extracted from the body wall of sea cucumber (Holothuria scabra) using the pepsin-solubilized collagen method followed by isolation using dialysis and the ultrafiltration membrane. The yield and physicochemical properties of the collagen obtained from both isolation methods, denoted as D-PSC and UF-PSC, were compared. The ultrafiltration method affords a higher yield of collagen (11.39%) than that of the dialysis (5.15%). The isolated collagens have almost the same amino acid composition, while their functional groups, referred to as amide A, B, I, II, and III bands, were in accordance with commercial collagen, as verified by Fourier Transform Infrared (FT-IR) spectroscopy. The UV-Vis absorption peaks at 240 nm and 220 nm, respectively, indicated that the collagens produced are type-I collagen. The D-PSC showed interconnecting sheet-like fibrils, while the UF-PSC exhibited a flaky structure with flat-sheets arranged very close to each other. The higher yield and comparable physicochemical properties of the collagen obtained by ultrafiltration as compared with dialysis indicate that the membrane process has high potential to be used in large-scale collagen production for food and pharmaceutical applications.

    Actinopyga lecanora, a type of sea cucumber commonly known as stone fish with relatively high protein content, was explored as raw material for bioactive peptides production. Six proteolytic enzymes, namely alcalase, papain, pepsin, trypsin, bromelain and flavourzyme were used to hydrolyze A. lecanora at different times and their respective degrees of hydrolysis (DH) were calculated. Subsequently, antibacterial activity of the A. lecanora hydrolysates, against some common pathogenic Gram positive bacteria (Bacillus subtilis and Staphylococcus aureus) and Gram negative bacteria (Escherichia coli, Pseudomonas aeruginosa, and Pseudomonas sp.) were evaluated. Papain hydrolysis showed the highest DH value (89.44%), followed by alcalase hydrolysis (83.35%). Bromelain hydrolysate after one and seven hours of hydrolysis exhibited the highest antibacterial activities against Pseudomonas sp., P. aeruginosa and E. coli at 51.85%, 30.07% and 30.45%, respectively compared to the other hydrolysates. Protein hydrolysate generated by papain after 8 h hydrolysis showed maximum antibacterial activity against S. aureus at 20.19%. The potent hydrolysates were further fractionated using RP-HPLC and antibacterial activity of the collected fractions from each hydrolysate were evaluated, wherein among them only three fractions from the bromelain hydrolysates exhibited inhibitory activities against Pseudomonas sp., P. aeruginosa and E. coli at 24%, 25.5% and 27.1%, respectively and one fraction of papain hydrolysate showed antibacterial activity of 33.1% against S. aureus. The evaluation of the relationship between DH and antibacterial activities of papain and bromelain hydrolysates revealed a meaningful correlation of four and six order functions.

    Despite its claimed therapeutic effects, the action of sea cucumber (known as gamat in the Malay language) on human osteoblast cells is still unknown. We performed in vitro studies utilising extract of Stichopus sp1 (gamat) to elucidate its effects on cell viability and functional activity. We found an inverse relationship between gamat concentration and its effect on osteoblast cell viability (p<0.001). Only gamat concentration at 1mg/ml significantly promoted cell viability at day 3 of incubation. There was a trend towards increased osteoblast cell function in the presence of gamat at 5mg/ml and 10mg/ml but this observation was not consistent at different incubation periods.

    Holothuria (Mertensiothuria) leucospilota (Brandt 1835), white threads fish or locally known as bat puntil is currently considered as the most abundant sea cucumber species in Malaysia. This study aimed to generate the genetic profile of H. leucospilota from Malaysia and then to determine the phylogenetic relationship between H. leucospilota and other members of genus Holothuria using partial sequences of cytochrome c oxidase I (COI) mitochondrial DNA (mtDNA) gene. In this study, specimens of H. leucospilota were collected from Intan Besar Island, Langkawi, Kedah Darul Aman in the west coast of Peninsular Malaysia. Three main methods namely neighbour joining, maximum parsimony and maximum likelihood were used for the phylogenetic tree reconstruction. Tree topologies showed that H. leucospilota has its own monophyletic clade clearly distinct from the other species. The pairwise genetic distance calculated further supported these findings. In addition, the results also should that the COI mtDNA gene is capable to unravel the phylogenetic relationship of H. leucospilota.

    This study aimed to resolve the taxonomic status of a morphologically undetermined sea cucumber species of order Apodida
    from Malaysia (GenBank accession no.: FJ223867) using partial 16S mitochondrial rRNA gene sequences and subsequently
    to determine the validity of morphological taxonomy of Holothuria species into its current subgenera. The undetermined
    species clustered with all taxa of Holothuria in previous study. Phylogenetic analyses using maximum parsimony and
    Bayesian methods suggest that the undetermined species was genetically closer to Holothuria (Lessonothuria) pardalis and
    Holothuria (Acanthotrapeza) coluber; and its position in both phylogenetic trees further suggests its status as a Holothuria
    taxon. Subgenera of Holothuria, Merthensiothuria and Metriatyla are monophyletic with strong bootstrap supports and
    posterior probabilities of clades, thus strengthening their morphological taxonomies. Nonetheless, the non-monophyly of
    subgenera of Halodeima, Microthele and Platyperona suggests a requirement for their taxonomic revisions using integrative
    taxonomy. The status of Holothuria (Halodeima) edulis subgroups in the maximum parsimony and Bayesian trees is
    indistinct and further taxonomic revisions are necessary. In terms of sister relationship, both phylogenetic trees suggest
    that subgenus Holothuria is a sister taxon of subgenus Roweothuria while the other sister relationships were unclear due
    to the undetermined species, paraphyly and polyphyly of a number of subgenera. Further studies with more specimens of
    genus Holothuria from broader geographical locations and various mtDNA genes along with morphological approaches
    may facilitate to provide better insights into the molecular phylogeny of subgenera of Holothuria.
