    Raffaelea variabilis is described as a new species in culture from Lannea grandis. It is distinguished by turbinate to cuneiform conidia measuring 4-16 times 2.5-7.5 mum, and compared and contrasted with established species. Trichocladium lobatum is described as a new species in culture with 1-2 euseptate spherical conidia, 19-22 mum diam., ornamented with flabelliform, spathulate or petaloid lobes 7 mum long. It is compared and contrasted with established Trichocladium species and representatives of Chlamydomyces, Histoplasma, Mycogone, Sepedonium and Thermomyces.

    A study was carried out to isolate and identify filamentous fungi for the treatment of domestic wastewater sludge by enhancing biodegradability, settleability and dewaterability of treated sludge using liquid state bioconversion process. A total of 70 strains of filamentous fungi were isolated from three different sources (wastewater, sewage sludge and leachate) of IWK's (Indah Water Konsortium) sewage treatment plant, Malaysia. The isolated strains were purified by conventional techniques and identified by microscopic examination. The strains isolated belonged to the genera of Penicillium, Aspergillus, Trichoderma, Spicaria and Hyaloflorae The distribution of observed isolated fungi were 41% in sewage sludge followed by 39% in wastewater and 20% in leachate. The predominant fungus was Penicillium (39 strains). The second and third most common isolates were Aspergillus (14 strains) and Trichoderma (12 strains). The other isolates were Spicaria (3 strains) and Hyaloflorae (2 strains). Three strains (WWZP1003, LZP3001, LZP3005) of Penicillium (P. corylophilum, P. waksmanii, and P. citrinum respectively), 2 strains (WWZA1006 and SS2017) of Aspergillus (A. terrues and A. flavus respectively) and one strain (SSZT2008) of Trichoderma (T. harzianum) were tentatively identified up to species level and finally verified by CABI Bioscience Identification Services, UK.

    The study aimed to determine the fungal diversity in clinical waste samples from a healthcare facility in Penang Malaysia. Different fungi species were detected in 83.75 % of the 92 clinical waste samples that were screened from different sections of the healthcare facility. One hundred fifty fungal isolates comprising of 8 genera and 36 species were obtained. They were purified by using single spore isolation technique. Subsequently, the isolates were identified by phenotypic method based on morphological and culture characteristics on different culture media. Among all fungal isolates, Aspergillus spp. in section Nigri 10.2 %, Aspergillus niger 9.5 %, Aspergillus fumigatus 8.8 %, Penicillium. simplicissium 8 %, Aspergillus tubingensis 7.3 %, Aspergillus terreus var. terreus 6.6 %, Penicillium waksmanii 5.9 % and Curvularia lunata 6.5 % were the most frequent. Among five sections of the Wellness Centre, the clinical wastes collected from the diagnostic labs of haematology section had the highest numbers of fungal species (29 species). Glove wastes had the highest numbers of fungal species (19 species) among 17 types of clinical wastes screened. Among all fungal species, Aspergillus spp. exhibited higher growth at 37 °C than at 28 °C, indicating the potential of these opportunistic fungi to cause diseases in human. These results indicated the potential of hospital wastes as reservoirs for fungal species.

    Roselle, or Jamaica sorrel (Hibiscus sabdariffa) is a popular vegetable in many tropical regions, cultivated for its leaves, seeds, stems and calyces which, the dried calyces are used to prepare tea, syrup, jams and jellies and as beverages. The main objectives of this study were to identify and characterise fungal pathogens associated with Roselle diseases based on their morphological and cultural characteristics and to determine the pathogenicity of four fungi infecting Roselle seedlings, namely Phoma exigua, Fusarium nygamai, Fusarium tgcq and Rhizoctonia solani in Penang. A total of 200 fungal isolates were obtained from 90 samples of symptomatic Roselle tissues. The isolates were identified based on cultural and morphological characteristics, as well as their pathogenicity. The fungal pathogen most frequently isolated was P. exigua (present in 45% of the samples), followed by F. nygamai (25%), Rhizoctonia solani (19%) and F. camptoceras (11%). Pathogenicity tests showed that P. exigua, F. nygamai, F. camptoceras and R. solani were able to infect both wounded and unwounded seedlings with different degrees of severity as indicated by the Disease severity (DS). R. solani was the most pathogenic fungus affecting both wounded and unwounded Roselle seedlings, followed by P. exigua that was highly pathogenic on wounded seedlings. F. nygamai was less pathogenic while the least pathogenic fungus was F. camptoceras, infecting only the unwounded seedlings but, surprisingly, not the wounded plants.

    The spatial distribution of basidiocarps provides much information on the dispersal abilities, habitat preferences, and inter- and intraspecific interactions of aphyllophoraceous fungi. To reveal the spatial distribution and resource utilization patterns of aphyllophoraceous fungi in Malaysia, we conducted field observations in a primary forest in 2006 and analyzed the relationships between the abundance of eight dominant fungal species and various environmental factors. The topographical characteristics were significantly patchily distributed at the 100-m scale, whereas woody debris and most fungal species were distributed randomly. Although the dominant fungal species differed among the decay classes and diameters of the woody debris, the abundance of a few dominant species was significantly correlated with environmental factors. Although the latter factors might affect the spatial distribution of these fungi, the effects appear to be so small that they would not create an aggregated distribution at a few 100-m scales.

    Ten filamentous fungi adapted to domestic wastewater sludge (DWS) were further studied to evaluate their potential in terms of adaptation to higher sludge supplemented growing media and phytopathogenicity (induction of diseases to plants) to three germinating crop (Corn: Zea mays, Mung bean: Phaseolus aureus and Mustard: Brassica napus) seeds. The performances of the fungi in seed germination were evaluated based on percent germination index (GI) and infected/spotted seeds on direct fungal biomass (FBM) and fungal metabolite (FM). Significantly the highest biomass production was achieved with RW-P1 512 and Penicillium corylophilum (WW-P1003) at the highest (25%) sludge supplemented growing media that implied its excellent potentiality of adaptation and multiplication to domestic wastewater sludge. Significantly encouraging results of percent GI and spotted/infected seedlings were observed in FM than FBM by all fungi except the strain Aspergillus niger. A. niger gave the poorest percent of GI (24.30, 26.98 and 00.00%) and the highest percent of infected/spotted seeds (70, 100, and 100%) using FBM for corn, mung bean and mustard, respectively. On the other hand, comparatively the highest percent of GI (107.99, 106.25 and 117.67%) and the lowest percent of spotted/infected seedlings (3.3, 3.3 and 3.3%) were achieved with the isolate RW-P1 512 using FM. In FBM, the superior results of percent GI (86.61, 95.92 and 83.87%) and spotted/infected seedlings (3.3, 63.3 and 43.3%) were obtained by A. versicolor. Several crop seeds were responded differently for different fungal treatments. Hundred percent infected/spotted seeds in FM were recorded only for mustard with Trichoderma family that implied its strong sensitiveness to its metabolites.

    In a survey of starch-based foods sampled from retail outlets in Malaysia, fungal colonies were mostly detected in wheat flour (100%), followed by rice flour (74%), glutinous rice grains (72%), ordinary rice grains (60%), glutinous rice flour (48%) and corn flour (26%). All positive samples of ordinary rice and glutinous rice grains had total fungal counts below 10(3) cfu/g sample, while among the positive rice flour, glutinous rice flour and corn flour samples, the highest total fungal count was more than 10(3) but less than 10(4) cfu/g sample respectively. However, in wheat flour samples total fungal count ranged from 10(2) cfu/g sample to slightly more than 10(4) cfu/g sample. Aflatoxigenic colonies were mostly detected in wheat flour (20%), followed by ordinary rice grains (4%), glutinous rice grains (4%) and glutinous rice flour (2%). No aflatoxigenic colonies were isolated from rice flour and corn flour samples. Screening of aflatoxin B1, aflatoxin B2, aflatoxin G1 and aflatoxin G2 using reversed-phase HPLC were carried out on 84 samples of ordinary rice grains and 83 samples of wheat flour. Two point four percent (2.4%) of ordinary rice grains were positive for aflatoxin G1 and 3.6% were positive for aflatoxin G2. All the positive samples were collected from private homes at concentrations ranging from 3.69-77.50 micrograms/kg. One point two percent (1.2%) of wheat flour samples were positive for aflatoxin B1 at a concentration of 25.62 micrograms/kg, 4.8% were positive for aflatoxin B2 at concentrations ranging from 11.25-252.50 micrograms/kg, 3.6% were positive for aflatoxin G1 at concentrations ranging from 25.00-289.38 micrograms/kg and 13.25% were positive for aflatoxin G2 at concentrations ranging from 16.25-436.25 micrograms/kg. Similarly, positive wheat flour samples were mostly collected from private homes.

    Endophytic fungi are widely known as fungi that infect internal tissues of host plants for all or part of their life cycles, without causing visible symptoms of disease. The present study was carried out to identify and investigate the pathogenicity of endophytic fungi residing in husks, silks, and kernels of corn. Endophytic fungi were isolated from surface-sterilised silks, kernels, and husks of healthy corn plants and identified using sequencing of multiple markers comprising TEF-1α, β-tubulin, calmodulin, ITS, LSU, and ACT. A total of 56 isolates of endophytic fungi belonging to 17 species, namely Fusarium pseudocircinatum (n = 8), F. verticillioides (n = 2), F. andiyazi (n = 4), F. sacchari (n = 1), F. mangiferae (n = 1), F. fujikuroi (n = 1), F. proliferatum (n = 3), F. incarnatum (n = 2), Penicillium oxalicum (n = 2), P. polonicum (n = 2), P. citrinum (n = 11), Aspergillus flavus (n = 10), A. tubingensis (n = 1), Cladosporium tenuissimum (n = 3), Aureobasidium pullulans (n = 3), Curvularia lunata (n = 1), and Epicoccum sorghinum (n = 1) were identified. Pathogenicity test showed that all endophytic fungi induced varying severities of disease symptoms on corn plants such as leaf chlorosis and necrosis, stem malformation, wilt, and stunted growth with F. verticillioides being the most virulent. The study revealed that corn tissues harbour diverse genera of endophytic fungi that can infect corn plants and may cause harmful effects to the host plants.

    During an investigation of submerged leaves and twigs sampled from tropical peat swamp forests located in Peninsular Malaysia, an anamorphic fungus not attributable to a described genus was detected and isolated in pure culture. Conidial ontogeny was thoroughly studied and illustrated using both light and SEM, which revealed a unique conidial morphology. Analysis of partial nuLSU rDNA and ITS data revealed a phylogenetic position within the Xylariales (Ascomycota), but family affiliation remained unclear.

    Red rice is a fermented product of Monascus spp. It is widely consumed by Malaysian Chinese who believe in its pharmacological properties. The traditional method of red rice preparation disregards safety regulation and renders red rice susceptible to fungal infestation and mycotoxin contamination. A preliminary study was undertaken aiming to determine the occurrence of mycotoxigenic fungi and mycotoxins contamination on red rice at consumer level in Selangor, Malaysia. Fifty red rice samples were obtained and subjected to fungal isolation, enumeration, and identification. Citrinin, aflatoxin, and ochratoxin-A were quantitated by ELISA based on the presence of predominant causal fungi. Fungal loads of 1.4 × 10(4) to 2.1 × 10(6) CFU/g exceeded Malaysian limits. Monascus spp. as starter fungi were present in 50 samples (100%), followed by Penicillium chrysogenum (62%), Aspergillus niger (54%), and Aspergillus flavus (44%). Citrinin was present in 100% samples (0.23-20.65 mg/kg), aflatoxin in 92% samples (0.61-77.33 μg/kg) and Ochratoxin-A in 100% samples (0.23-2.48 μg/kg); 100% citrinin and 76.09% aflatoxin exceeded Malaysian limits. The presence of mycotoxigenic fungi served as an indicator of mycotoxins contamination and might imply improper production, handling, transportation, and storage of red rice. Further confirmatory analysis (e.g., HPLC) is required to verify the mycotoxins level in red rice samples and to validate the safety status of red rice.

    Isolates of anaerobic fungi obtained from the rumen, duodenum and faeces of sheep were identified as Piromyces mae based on their morphological characteristics observed using light microscopy. There was no significant morphological variation among the isolates of P. mae from the rumen, duodenum and faeces. Isozymes of 12 isolates of P. mae (one each from the rumen, duodenum and faeces from 4 different sheep) were analysed by PAGE. A total of 12 isozymes were studied and 5 isozyme loci were successfully typed. They were malic enzyme, malate dehydrogenase, shikimate dehydrogenase, alpha-esterase and beta-esterase. All the isolates of P. mae regardless of whether they were from the rumen, duodenum or faeces or from different animals produced very similar isozyme banding patterns for each of the enzyme systems. The similar isozyme profiles of the isolates indicate that they are of the same species although they exist in different regions of the alimentary tract.

    Hands of Health Care Personnel (HCP) are one of the most common vehicles for the transmission of infection. Microorganisms can survive well on the hands of HCP for a certain duration. Therefore, the purpose of this study is to bring awareness to HCP that their hands can actually be contaminated with many microorganisms. These microbes on the hands of HCP can potentially infect their patients if they do not comply with the proper hand hygiene practice. This cross-sectional study was conducted at a randomly selected Intensive Care Unit (ICU) and general ward in a hospital. Twenty five HCP from each ward were randomly selected and their hands were imprinted on blood culture plates. Microorganism growth were quantified and identified. Data were analyzed and presented as descriptive analysis. One hundred blood agar plates were processed and analyzed. Majority (71%) of the samples had more than 50 colony-forming units (CFU) and only 17% of the samples had less than 25 CFU. Microorganisms identified include Staphylococcus spp., Acinetobacter spp., Enterobacteriaceae, Pseudomonas spp., Moraxella, Delftiaacidovorans and fungi. All isolated microorganisms were antibiotic sensitive strain. This study showed that the hands of HCP were contaminated with many microorganisms. Therefore, it is imperative that HCP must practice proper hand hygiene when taking care of their patients in the wards.

    Edible bird nests (EBNs) are consumed worldwide for various health benefits. EBNs are nests built from the saliva of swiftlets of Aerodramus species. The global market for EBNs is on the rise, especially from Hong Kong and mainland China. In the past, EBNs were harvested mainly from natural caves; however in the recent years, there has been a rapid growth of swiftlet farming. Little is known about the actual composition of EBNs except for protein, carbohydrate, ash and lipid contents, amino acids, vitamins and macro/ micronutrients. Besides the biochemical components of EBNs, are there any other structures that are associated with EBNs? This paper reports on the structural analysis of raw unprocessed farm and processed commercial EBNs. The raw EBNs were purchased from swiftlet farms in five locations in Peninsula Malaysia: Kuala Sanglang (Perlis; 6° 16' 0"N, 100° 12' 0"E), Pantai Remis (Perak; 4º 27' 0" N, 100º 38' 0" E), Kluang (Johor; 02º 012 303N 103º 192 583E), Kajang (Selangor; 2º 59' 0"N, 101º 47' 0"E) and Kota Bharu (Kelantan; 6º 8' 0"N, 102º 15' 0"E). The commercial nests were purchased from five different Chinese traditional medicinal shops (Companies A-E). A portion of each EBN was randomly broken into small fragments, attached to carbon tape and coated with gold and palladium particles for examination and photography under a scanning electron microscope. Structural analysis revealed the presence of mites, fungi, bacteria and feather strands on both the raw and commercial nests. Mite eggshells and faecal pellets, and body parts of other arthropods were seen only in the raw nests. The commercial nests had a variety of unidentified structures and substances coated on the nests' surfaces that were not found on the raw nests. The presence of these contaminants may jeopardise the quality of EBNs and pose health risks to consumers. Further identification of the mites and their allergens, fungi and bacteria are on-going and will be reported separately.

    An investigation was undertaken for screening and isolating nematophagous-fungi from the faecal samples of various grazing animals and soils in Malaysia. Total of 111 faeces and 50 soil samples were collected and the samples were cultured on 2% water agar plates. The growth of nematophagous-fungi was stimulated by sprinkling-baiting technique. The conidia of suspected nematophagous-fungi were inoculated on 2% water agar plates. All isolated were maintained on 2% cornmeal agar plates. Verticillium spp., Fusarium spp. and Arthrobotrys spp. were identified from the faecal and soil samples. 62.5% of the faecal samples and 100% of the soil samples were shown to be positive with nematophagous-fungi. This study highlights the present of nematophagous-fungi population in faecal and soil samples. Much study remains to be done to better understanding some fungi especially their mode of action and their predatory behaviour against parasitic nematodes.
