    Fusarium keratoplasticum is arguably the most common Fusarium solani species complex (FSSC) species associated with human infections. Invasive fusariosis is a life-threatening fungal infection that is difficult to treat with conventional azole antifungals. Azole drug resistance is often caused by the increased expression of pleiotropic drug resistance (PDR) ATP-binding cassette (ABC) transporters of the ABCG sub-family. Most investigations of Fusarium ABC transporters associated with azole antifungal drug resistance are limited to plant pathogens. Through the manual curation of the entire ABCG protein family of four FSSC species including the fully annotated genome of the plant pathogen Nectria haematococca we identified PDR transporters ABC1 and ABC2 as the efflux pump candidates most likely to be associated with the innate azole resistance phenotype of Fusarium keratoplasticum. An initial investigation of the transcriptional response of logarithmic phase F. keratoplasticum cells to 16 mg/L voriconazole confirmed strong upregulation (372-fold) of ABC1 while ABC2 mRNA levels were unaffected by voriconazole exposure over a 4 h time-period. Overexpression of F. keratoplasticum ABC1 and ABC2 in the genetically modified Saccharomyces cerevisiae host ADΔΔ caused up to ∼1,024-fold increased resistance to a number of xenobiotics, including azole antifungals. Although ABC1 and ABC2 were only moderately (20% and 10%, respectively) expressed compared to the Candida albicans multidrug efflux pump CDR1, overexpression of F. keratoplasticum ABC1 caused even higher resistance levels to certain xenobiotics (e.g., rhodamine 6G and nigericin) than CDR1. Our investigations suggest an important role for ABC1 orthologues in the innate azole resistance phenotype of FSSC species.

    The receptors for IL-35, IL-12Rβ2 and gp130, have been implicated in the inflammatory pathophysiology of autoimmune diseases. In this study, we set out to investigate the serum IL-35 levels and the surface levels of IL-12Rβ2 and gp130 in CD3+CD4+, CD3+CD4─ and CD3─CD4─ lymphocyte subpopulations in systemic lupus erythematosus (SLE) patients (n=50) versus healthy controls (n=50). The potential T cell subsets associated with gp130 transcript (i.e. IL6ST) expression in CD4+ T cells of SLE patients was also examined in publicly-available gene expression profiling (GEP) datasets. Here, we report that serum IL-35 levels were significantly higher in SLE patients than healthy controls (p=0.038) but it was not associated with SLEDAI-2K scores. The proportions of IL-12Rβ2+ and gp130+ cells in SLE patients did not differ significantly with those of healthy controls in all lymphocyte subpopulations investigated. Essentially, higher SLEDAI-2K scores were positively correlated with increased proportion of gp130+ cells, but not IL-12Rβ2+ cells, on CD3+CD4+ T cells (r=0.425, p=0.002, q=0.016). Gene Set Enrichment Analysis (GSEA) of a GEP dataset of CD4+ T cells isolated from SLE patients (n=8; GSE4588) showed that IL6ST expression was positively associated with genes upregulated in CD4+ T cells vs myeloid or B cells (q<0.001). In an independent GEP dataset of CD4+ T cells isolated from SLE patients (n=9; GSE1057), IL6ST expression was induced upon anti-CD3 stimulation, and that Treg, TCM and CCR7+ T cells gene sets were significantly enriched (q<0.05) by genes highly correlated with IL6ST expression (n=92 genes; r>0.75 with IL6ST expression) upon anti-CD3 stimulation in these SLE patients. In conclusion, gp130 signaling in CD3+CD4+ T cell subsets may contribute to increased disease activity in SLE patients, and it represents a promising therapeutic target for inhibition in the disease.

    Pluronic F-68 (PF-68) is a non-ionic surfactant used in plant tissue culture as a growth additive. Despite its usage as a plant growth enhancer, the mechanism underlying the growth-promoting effects of PF-68 remains largely unknown. Hence, this study was undertaken to elucidate the growth-promoting mechanism of PF-68 using recalcitrant MR 219 callus as a model. Supplementation of 0.04% PF-68 (optimum concentration) was shown to enhance callus proliferation. The treated callus recorded enhanced sugar content, protein content, and glutamate synthase activity as exemplified in the comparative proteome analysis, showing protein abundance involved in carbohydrate metabolism (alpha amylase), protein biosynthesis (ribosomal proteins), and nitrogen metabolism (glutamate synthase), which are crucial to plant growth and development. Moreover, an increase in nutrients uptake was also noted with potassium topping the list, suggesting a vital role of K in governing plant growth. In contrast, 0.10% PF-68 (high concentration) induced stress response in the callus, revealing an increment in phenylalanine ammonia lyase activity, malondialdehyde content, and peroxidase activity, which were consistent with high abundance of phenylalanine ammonia lyase, peroxidase, and peroxiredoxin proteins detected and concomitant with a reduced level of esterase activity. The data highlighted that incorporation of PF-68 at optimum concentration improved callus proliferation of recalcitrant MR 219 through enhanced carbohydrate metabolism, nitrogen metabolism, and nutrient uptake. However, growth-promoting effects of PF-68 are concentration dependent.

    Air pollution is a matter of concern among the public, especially for those living in urban and industrial areas. Markov chain modeling is often used to model the underlying dynamics of air pollution, which involves describing the transition probability of going from one air pollution state to another. Thus, estimating the transition probability matrix for the data of the air pollution index (API) is an essential process in the modeling. However, one may observe many zero probabilities in the transition probability matrix, especially when faced with a small sample, interpreting the results with respect to the climate condition less realistic. This study proposes a robust empirical Bayes method, which incorporates a method of smoothing the zero frequencies in the count matrix, contributing to an improved estimation of the transition probability matrix. The robustness of the empirical Bayesian estimation is investigated based on Bayes risk. The transition probability matrices estimated based on the robust empirical Bayes method for the hourly API data collected from seven monitoring stations in Malaysia for the period 2012 to 2014 are used for determining the air pollution characteristics such as the mean residence time, the steady-state probability and the mean recurrence time. Furthermore, the proposed method has been evaluated by Monte Carlo simulations. Results suggest that it is quite effective in producing non-zero transition probability estimates, and superior to the maximum likelihood method in terms of minimizing the mean squared error for individual and entire transition probabilities. Therefore, the robust empirical Bayes method proves to be an improved approach to the estimation of the Markov chain. When applied to API data, it could provide important information on air pollution dynamics that may help guiding the development of proper strategies for managing the impact of air quality.

    The functionalized graphene oxide by silica and chitosan helped to prepared an adsorbent with high adsorption potential for removing cadmium(II). In this study, the adsorbent was synthesized and the batch system of adsorption method was examined to find the potential of the new adsorbent with the various factors of the concentration, pH, time and temperature. The characterization of adsorbent was analyzed by FT-IR, TEM, Zeta potential and XRD analysis. Regards to the analysis it can be understood that the adsorbent was synthesized successfully. The investigational results were validated and analyzed by applying the 5 models of isotherm and 4 models of kinetic. The Langmuir, Freundlich, Temkin, Harkins-Jura and Dubinin-radushkevich models were used which the Langmuir, Freundlich and Temkin fitted well for removing cadmium(II). The Qmax value was achieved 126.58 mg/g by using the Langmuir model for removing Cd(II) respectively. The pseudo-first-order, pseudo-second-order, Elovich and Intra-particle models were used to validate the kinetic models of the process. The pseudo-second-order and Elovich models were the best fitted kinetic model in this investigation. Thermodynamic parameters of the energy of gibes, the enthalpy, and the entropy were calculated. Generally, the adsorption process was distinguished as an exothermic and spontaneous.

    Chordomas are rare and slow-growing locally destructive bone tumors that can develop in the craniospinal axis. It is commonly found in the sacrococcygeal region whereas only 25-35% are found in the clival region. Headache with neurological deficits are the most common clinical presentations. Complete surgical resection either via open or endoscopic endonasal approaches are the main mode of treatment. Here, we report a series of 5 cases of clival chordomas which was managed via endoscopic endonasal approaches in our center. A retrospective analysis of patients who had undergone endoscopic endonasal resection of clival chordoma in Sarawak General Hospital from 2014 to 2018. A total of 5 cases were operated on endoscopically via a combine effort of both the otorhinolaryngology team and the neurosurgical team during the study period from year 2013 to 2018. From our patient, 2 were female and 3 were male patients. The main clinical presentation was headache, squinting of eye and nasopharyngeal fullness. All our patient had endoscopic endonasal debulking of clival tumor done, with average of hospital stay from 9 - 23 days. Pos-operatively, patients were discharged back well. Endoscopic endonasal resection of clival chordomas gives good surgical resection results with low morbidity rates and therefore can be considered as a surgical option in centers where the surgical specialties are available.

    This article presents data on digital adoption by enterprises in Malaysian industrial sectors during the COVID-19 pandemic. The data were collected during the periods of Conditional Movement Control Order (CMCO) and Recovery Movement Control Order (RMCO) from October 11 to December 31, 2020. Data collection was completed through an online questionnaire survey conducted among a sample of 432 enterprises from four industrial sectors, namely services, retail, manufacturing, and tourism, in all states in Malaysia. The sample was selected using cluster and systematic random sampling. The questionnaire asked respondents to state whether they used the Internet, computers, phones, web sites, e-payment, and e-commerce to complete their activities relating to finance, production and operations, human resource management, and marketing. The data were analysed using descriptive statistics and cross-tabulation. The data show the extent of digital adoption by Malaysian enterprises during the pandemic in comparison to the situation before the pandemic. The data may be of use to other similar researchers as comparison and to policy makers as guides in devising related policies.

    The formation of foam due to the injection of surfactant foam in FAWAG causes significant problems in the oil well production and separation facilities. The excessive foam can lead to the reduction of the separator capacity as well as its efficiency. A defoamer is needed to break and destroy the foam in the separator. There are many commercially available defoamer agents in the market, but not all defoamers are suitable for every application. For this reason, four modified silicone-based defoamers were successfully synthesized and characterized based on the data obtained from the screening process using various commercial defoamers. The performance of modified defoamers was evaluated using TECLIS FoamScan that imitate real conditions of treatment. The results show that all four of the modified silicone-based defoamers, especially amide-terminated-modified defoamers (S2) showed excellent performance as a defoaming agent to mitigate foam in specific conditions. The best-case condition for the modified defoamer to perform was at a high temperature (60 °C), gas flow rate of 1.0 L/min, and low ration concentration of the surfactant to brine (30:70). The study on the bubble count and distribution using a KRÜSS Dynamic Foam Analyzer revealed that S2 excellently contributes to the formation of unstable foam that can fasten foam destruction in the foaming system.

    Current phylogenetic analysis of the flavivirus genus has identified a group of mosquito-borne viruses for which the vertebrate hosts are currently unknown. Here we report the identification of a novel member of this group from a peridomestic rodent species (Sundamys muelleri) collected in Sarawak, Malaysia in 2016. We propose to name this novel flavivirus Batu Kawa virus after the location in which it was identified, with the abbreviation BKWV. Characterization of the BKWV genome allowed identification of putative mature peptides, potential enzyme motifs and conserved structural elements. Phylogenetic analysis found BKWV to be most closely related to Nhumirim virus (from Brazil) and Barkedji virus (from Senegal and Israel). Both of these viruses have been identified in Culex mosquitoes and belong to a group of viruses with unknown vertebrate hosts. This is the first known report of a member of this group of viruses from a potential mammalian host.

    The carbon dioxide emissions from Portland cement production have increased significantly, and Portland cement is the main binder used in self-compacting concrete, so there is an urgent need to find environmentally friendly materials as alternative resources. In most developing countries, the availability of huge amounts of agricultural waste has paved the way for studying how these materials can be processed into self-compacting concrete as binders and aggregate compositions. Therefore, this experimental program was carried out to study the properties of self-compacting concrete (SCC) made with local metakaolin and coal bottom ash separately and combined. Total 25 mixes were prepared with four mixes as 5, 10, 15, and 20% replacement of cement with metakaolin; four mixes as 10, 20, 30, and 40% of coal bottom ash as partial replacement of fine aggregates separately; and 16 mixes prepared combined with metakaolin and coal bottom ash. The fresh properties were explored by slump flow, T50 flow, V-funnel, L-box, and J-ring sieve segregation test. Moreover, the hardened properties of concrete were performed for compressive, splitting tensile and flexural strength and permeability of SCC mixtures. Fresh concrete test results show that even if no viscosity modifier is required, satisfactory fresh concrete properties of SCC can be obtained by replacing the fine aggregate with coal bottom ash content. At 15% replacement of cement with local metakaolin is optimum and gave better results as compared to control SCC. At 30% replacement of fine aggregate is optimum and gave better results as compared to control SCC. In the combined mix, 10% replacement of cement with metakaolin combined with 30% replacement of fine aggregate with coal bottom ash is optimum and gave better results as compared to control SCC.

    Bee orchids have long been an excellent example of how dishonest signal works in plant-animal interaction. Many studies compared the flower structures that resemble female bees, leading toward pseudo-copulation of the male bees on the flower. Using Machine Learning, we tested whether nature is capable of besting artificial intelligence. A total of 2000 images of related bees, wasps, and Ophrys sp. were collected from the Google Image Repository. Unsuitable images were later filtered out manually, leaving a total of 995 images in the final selection. 80% of these images were used to build a supervised model using Logistic Regression, while the model accuracy was tested using 20% of the remaining images. Based on our results using Wolfram Mathematica, the Ophrys is not capable of fooling artificial intelligence. The accuracy, accuracy baseline, mean cross-entropy, Area Under ROC (receiver operating characteristic curve) curve (AUC) and the confusion matrix gave excellent image classification. However, we can now show the key points and highlights of the images and how the structures closely resemble actual bees using the SURF method. Rather than just a descriptive method, ML learning has enabled a more quantitative approach. Since this is a simple test, we encourage other scientists to adopt our approach using a larger dataset and better database samples.

    BACKGROUND: The health belief model explains that individual intentions and motivation of health behaviors are mostly subject to external cues to action, such as from interpersonal communications and media consumptions. The concept of mobilizing information (MI) refers to a type of mediated information that could call individuals to carry out particular health actions. Different media channels, especially digital media outlets, play an essential role as a health educator to disseminate cancer health information and persuade and mobilize cancer prevention in the community. However, little is known about calls to action (or MI) in online cancer news, especially from Asian media outlets.

    OBJECTIVE: This study aimed at analyzing cancer news articles that contain MI and their news components on the selected Malaysian English and Chinese newspapers with online versions.

    METHODS: The Star Online and Sin Chew Online were selected for analysis because the two newspaper websites enjoy the highest circulation and readership in the English language and the Chinese language streams, respectively. Two bilingual coders searched the cancer news articles based on sampling keywords and then read and coded each news article accordingly. Five coding variables were conceptualized from previous studies (ie, cancer type, news source, news focus, cancer risk factors, and MI), and a good consistency using Cohen kappa was built between coders. Descriptive analysis was used to examine the frequency and percentage of each coding item; chi-square test (confidence level at 95%) was applied to analyze the differences between two newspaper websites, and the associations between variables and the presence of MI were examined through binary logistic regression.

    RESULTS: Among 841 analyzed news articles, 69.6% (585/841) presented MI. News distributions were unbalanced throughout the year in both English and Chinese newspaper websites; some months occupied peaks (ie, February and October), but cancer issues and MI for cancer prevention received minimal attention in other months. The news articles from The Star Online and Sin Chew Online were significantly different in several news components, such as the MI present rates (χ2=9.25, P=.003), providing different types of MI (interactive MI: χ2=12.08, P=.001), interviewing different news sources (government agency: χ2=12.05, P=.001), concerning different news focus (primary cancer prevention: χ2=10.98, P=.001), and mentioning different cancer risks (lifestyle risks: χ2=7.43, P=.007). Binary logistic regression results reported that online cancer news articles were more likely to provide MI when interviewing nongovernmental organizations, focusing on topics related to primary cancer prevention, and highlighting lifestyle risks (odds ratio [OR] 2.77, 95% CI 1.89-4.05; OR 97.70, 95% CI 46.97-203.24; OR 186.28; 95% CI 44.83-773.96; P=.001, respectively).

    CONCLUSIONS: This study provided new understandings regarding MI in cancer news coverage. This could wake and trigger individuals' preexisting attitudes and intentions on cancer prevention. Thus, health professionals, health journalists, and health campaign designers should concentrate on MI when distributing health information to the community.

    BACKGROUND: Recently, Papatheodoridi et al proposed to refine the Baveno VI elastography dual-cutoffs and introduce an algorithm for the detection of compensated advanced chronic liver disease (cACLD) in asymptomatic European patients with chronic liver diseases.

    AIMS: To validate the performance of the dual-cutoffs (8/12 kPa) and the proposed algorithm to identify patients with cACLD in three well-characterised Asian nonalcoholic fatty liver disease (NAFLD) cohorts.

    METHODS: We included 830 patients with biopsy-proven NAFLD. Liver stiffness was measured using transient elastography (FibroScan).

    RESULTS: cACLD was found in 21.8% of patients. Compared with the original Baveno VI elastography criteria (10/15 kPa), the new cutoffs showed a comparable specificity and a higher sensitivity for identifying cACLD. We developed a simplified risk model incorporating age, liver stiffness value, and platelet count, which outperformed liver stiffness measurement alone in two Chinese cohorts (P = 0.001), and was further validated in a Malaysian cohort (P = 0.04). Overall, the "two-step" screening of cACLD improved classification rates from 73.5% by the original dual-cutoffs to 86.7%. Notably, usage of our simplified risk model resulted in significantly lower false-negative rate than the refined screening approach by Papatheodoridi et al (27.1% vs 41.4%; P = 0.01).

    CONCLUSIONS: The dual elastography cutoffs of 8 and 12 kPa are more appropriate to identify cACLD in Asian patients with NAFLD. In combination with a simplified risk model in unclassified patients, the two-step approach showed a classification rate of about 85%.

    In the current study, a phase inversion scheme was employed to fabricate hydroxyapatite (HA)/polysulfone (PSF)-based asymmetric membranes using a film applicator with water as a solvent and nonsolvent exchanging medium. Fourier Transform Infrared (FTIR) and X-ray diffraction (XRD) spectroscopic studies were conducted to confirm the bonding chemistry and purity of filler. The inherent thick nature of PSF generated sponge-like shape while the instantaneous demixing process produced finger-like pore networks in HA/PSF-based asymmetric membranes as exhibited by scanning electron microscope (SEM) micrographs. The FTIR spectra confirmed noncovalent weak attractions toward the polymer surface. The leaching ratio was evaluated to observe the dispersion behavior of HA filler in membrane composition. Hydrophilicity, pore profile, pure water permeation (PWP) flux, and molecular weight cutoff (MWCO) values of all formulated membranes were also calculated. Antifouling results revealed that HA modified PSF membranes exhibited 43% less adhesion of bovine serum albumin (BSA) together with >86% recovery of flux. Membrane composition showed 74% total resistance, out of which 60% was reversible resistance. Biocompatibility evaluation revealed that the modified membranes exhibited prothrombin time (PT), and thrombin time (TT) comparable with typical blood plasma, whereas proliferation of living cells over membrane surface proved its nontoxic behavior toward biomedical application. The urea and creatinine showed effective adsorption aptitude toward HA loaded PSF membranes.

    Pemphigus vulgaris is an autoimmune blistering disease with an increased potential for mortality. The epithelium is key in understanding the pathobiology as it is specialized to perform functions like mechanical protection, immunological defense, and proprioception. In order to perform these array of functions, epithelial integrity is important. This integrity is maintained by a host of molecules which orchestrate the ability of the keratinocytes to function as a single unit. Desmoglein 3 antibodies formed in genetically susceptible individuals are known to cause the disruption of the intact oral mucosa leading to the formation of blisters in pemphigus vulgaris patients. However, there are underlying complex triggering pathways leading to the clinical disease. The aim of the review is to congregate and critically appraise the various triggering pathways which contribute toward the pathobiology of pemphigus vulgaris. Articles relevant to the pathobiology of pemphigus vulgaris were identified from various search databases till the year 2020. The pathogenesis of pemphigus vulgaris is complex, and it involves an in-depth understanding of the various predisposing factors, provoking factors, and progression mechanisms. Congregation of the various triggering pathways will open our minds to understand pemphigus vulgaris better and in turn develop a reliable treatment in the near future.

    Patients with rheumatoid arthritis (RA) experience a higher prevalence of periodontitis. This study aimed to examine the variation of periodontitis experienced with different serotypes suffered by RA patients and to examine the relationship between the different medications taken for RA that may influence this relationship. Two hundred and sixty RA and control participants underwent standardized periodontal examinations. Medical, serological and radiological (Sharp/van der Heijde) records were assessed. Functional status was assessed using the administered Health Assessment Questionnaire. Moreover, disease parameters, including disease activity (DAS28-ESR) and anti-citrullinated protein antibodies (ACPA) and rheumatoid factor (RF) seropositivity were evaluated. Periodontitis was higher in RA (71.54%) compared with controls (54.62%). The stage of periodontitis experienced by ACPA-positive participants were higher than APCA-negative participants. The probing pocket depth and recession experienced by RF-positive participants were higher than those who were RF-negative. RA participants on methotrexate had lower clinical attachment loss and lower periodontal probing depth compared with participants on a combination methotrexate and other disease-modifying antirheumatic drugs. Participants taking corticosteroids had lower gingival index scores. The association between seropositivity and the type of medications taken with periodontal health parameters in this group of patients suggests that both seropositivity and medications taken are important modifiers in the relationship between periodontitis and RA.

    This study examines the effects of varying the concentrations of sorbitol (S) and glycerol (G) on the physical, morphological, thermal, and mechanical properties of Dioscorea hispida, starch-based films. In this context, the films of Dioscorea hispida starch were developed using solution casting technique with glycerol (G), sorbitol (S), and a mixture of sorbitol-glycerol (SG) as plasticizers at the ratios of 0, 30, 45, and 60 wt%. The films' moisture contents were increased when increasing the plasticizer contents. The tensile strengths were decreased, but elongations at break were increased; 7.38%-11.54% for G-plasticized films, 10.17%-15.76% for S-plasticized films, and 14.41%- 16.10% for SG-plasticized films with increasing plasticizer concentrations of the film samples. Varying plasticizer concentrations exhibited a minor effect on the S-plasticized film's thermal properties. Significant decrement in the glass transition temperatures of Dioscorea hispida starch films was observed when the plasticizer contents were raised from 30% to 60%. Significantly, the present work has shown that plasticized Dioscorea hispida starch can be considered a promising biopolymer for the applications of biodegradable films.

    OBJECTIVE: The purpose of the present study was bleached enamel reversal using conventional ascorbic acid (AA) and contemporary methods including Photodynamic therapy (PDT), Er, Cr: YSGG (ECL) on bond integrity to composite resin.

    MATERIAL AND METHOD: Forty sound permanent mandibular premolars were collected from a dental clinical setting and disinfected. All forty samples were mounted vertically in a rubber mold exposing only the clinical crown. All samples were bleached using Opalescence Boost Professional Teeth Whitening. After the bleaching procedure, each sample was randomly allocated into four groups according to surface treatment. Samples in group 1 were treated with methylene blue photosensitizer (MBP). Samples in group 2 were exposed to 10% sodium ascorbate. Samples in group 3 were treated with Er, Cr: YSGG laser (ECL). Samples in group 4 were not treated (control). All Samples were treated with 37% phosphoric acid and a bonding agent was applied. A bulk-fill composite was cured to all specimens and all samples were treated in a thermocycler. Specimens were placed in a universal testing machine for shear bond strength (SBS) testing. Descriptive statistics were associated by analysis of variance (ANOVA) and Tukey's post hoc test maintaining level of significance (p<0.05) RESULTS: The lowest SBS scores were achieved in the bleached enamel (BE) group (15.25±1.745 MPa). Whereas, the highest bond integrity was attained by AA group (32.23±1.854 MPa). Samples treated with ECL (31.87±1.659 MPa) and AA (32.23±1.854) were comparable (p>0.05). Samples treated with PDT exhibited significantly different SBS (22.41±1.258) compared to other experimental groups CONCLUSION: ECL showed a reversal effect of BE compared to AA and has the potential to be used in clinical settings. BE reversal using MBP needs further investigation.
