    The resistance status of Selangor Aedes aegypti (Linnaeus) larvae against four major groups of insecticides (i.e., organochlorines, carbamates, organophosphates and pyrethroids) was investigated. Aedes aegypti were susceptible against temephos (organophosphate), although resistance (RR50 = 0.21-2.64) may be developing. The insecticides susceptibility status of Ae. aegypti larvae were found heterogeneous among the different study sites. Results showed that Ae. aegypti larvae from Klang, Sabak Bernam and Sepang were susceptible against all insecticides tested. However, other study sites exhibited low to high resistance against all pyrethroids (RR50 = 1.19-32.16). Overall, the application of synergists ethacrynic acid, S.S.S.- tributylphosphorotrithioate and piperonyl butoxide increased the toxicity of insecticides investigated. However, the application failed to increase the mortality to susceptible level (>97%) for certain populations, therefore there are chances of alteration of target site resistance involved. Biochemical assays revealed that α-esterase, (Gombak, Kuala Langat, Kuala Selangor and Sabak Bernam strains) β-esterase (Klang and Sabak Bernam strains), acetylcholinesterase (Kuala Selangor and Sabak Bernam strains), glutathione-S-transferase (Kuala Selangor and Sabak Bernam strains) and mono-oxygenases (Gombak, Hulu Langat, Hulu Selangor and Kuala Langat strains) were elevated. Spearman rank-order correlation indicated a significant correlation between resistance ratios of: DDT and deltamethrin (r = 0.683, P = 0.042), cyfluthrin and deltamethrin (r = 0.867, P =0.002), cyflyuthrin and lambdacyhalothrin (r = 0.800, P =0.010), cyfluthrin and permethrin (r = 0.770, P =0.015) deltamethrin and permethrin (r = 0.803, P =0.088), propoxur and malathion (r = 0.867, P = 0.002), malathion and temephos (r = 0.800, P = 0.010), etofenprox and MFO enzyme (r = 0.667, P =0.050). The current study provides baseline information for vector control programs conducted by local authorities. The susceptibility status of Ae. aegypti should be monitored sporadically to ensure the effectiveness of current vector control strategy in Selangor.

    In Malaysia, rivers are the main source of public water supplies. This study was conducted from 2002 to 2003 to determine the levels of selected organochlorine and organophosphate pesticides in the Selangor River in Malaysia. Surface water samples have been collected seasonally from nine sites along the river. A liquid-liquid extraction followed by gas chromatography-mass spectrometry technique was used to determine the trace levels of these pesticide residues. The organochlorine pesticides detected were lindane, heptachlor, endosulfan, dieldrin, endosulfan sulfate, o,p'-DDT, p,p'-DDT, o,p'-DDE and p,p'-DDE whereas for organophosphate pesticides, they were chlorpyrifos and diazinon. At the river upstream where a dam is located for public water supply, incidents of pesticide levels exceeding the European Economic Community Directive of water quality standards have occurred. Furthermore, the wetland ecosystems located at the downstream of the river which houses the fireflies community is being threatened by occasional pesticide levels above EPA limits for freshwater aquatic organisms. The occurrence of these residual pesticides in the Selangor River can be attributed to the intense agriculture and urban activity.

    Magnetic solid phase extraction (MSPE) employing oil-palm fiber activated carbon (OPAC) modified with magnetite (Fe3O4) and polypyrrole (OPAC-Fe3O4-PPy) was successfully used for the determination of two organochlorine pesticides (OCPs), namely endosulfan and dieldrin in environmental water samples. Analysis was performed using gas chromatography with micro-electron capture detection (GC-μECD). The effects of three preparation variables, namely Fe3O4:OPAC ratio, amount of pyrrole monomer, and amount of FeCl3 oxidant were optimized using Box-Behnken design (BBD) (R2 < 0.99, p-value < 0.001%). The optimum conditions were as follows: Fe3O4:OPAC ratio of 2:1 w/w, 1 g of FeCl3 and 100 μL of pyrrole monomer. The experimental results obtained agreed satisfactorily with the model prediction (> 90% agreement). Optimized OPAC-Fe3O4-PPy composite was characterized using field emission scanning electron microscope, vibrating sample magnetometer and Fourier transform infrared spectroscopy. Four numerical parameters of MSPE procedure was optimized using BBD. The significance of the MSPE parameters were salt addition > sample solution pH > extraction time and desorption time. Under the optimized conditions (extraction time: 90 s, desorption time: 10 min, salt: 0%, and pH: 5.8), the method demonstrated good linearity (25-1000 ng L-1) with coefficients of determination, R2 > 0.991, and low detection limits for both endosulfan (7.3 ng L-1) and dieldrin (8.6 ng L-1). The method showed high analyte recoveries in the range of 98.6-103.5% for environmental water samples. The proposed OPAC-Fe3O4-PPy MSPE method offered good features such as sustainability, simplicity, and rapid extraction.

    Suatu kajian untuk menilai kesan gabungan aktiviti guna tanah dan perubahan musim terhadap kualiti sumber air telah dijalankan di Terusan Utara, Kedah. Kajian ini dijalankan di lima stesen terpilih selama 12 bulan berdasarkan Indeks Kualiti Air (WQI) dan Piawaian Interim Kualiti Air Kebangsaan (INWQS). Kajian mendapati kualiti air di Terusan Utara berada pada Kelas III dan berlaku peningkatan kualiti air dari hulu ke hilir. Hampir semua stesen berpotensi sebagai punca pencemaran berdasarkan aktiviti guna tanah terutamanya di Pelubang, Jitra serta Tunjang. Kajian menunjukkan terdapat perbezaan yang signifikan ketika perubahan musim pada DO, TSS serta BOD, namun tidak pada ammonia, pH dan COD. Secara amnya, kombinasi aktiviti guna tanah dan perubahan musim boleh mempengaruhi atau memberi kesan terhadap kualiti sumber air. Oleh itu, tindakan segera perlu dilakukan untuk mengawal punca pencemaran bagi memastikan kualiti sumber air di Terusan Utara kekal terpelihara.

    Kajian ini bertujuan untuk mengenal pasti punca berlakunya banjir di Segamat pada 17 hingga 21 Disember 2006. Kegiatan termasuklah mengumpul, menilai dan mengalisis data taburan hujan dari Januari 1998 hingga Disember 2006 serta menganalisis data telemetri harian bagi aras dan luahan air sungai pada Disember 2006. Satu set data hujan harian, bulanan dan tahunan dari Stesen Felda Medoi, Segamat telah dianalisis dan dihubungkait dengan data telemetri. Data taburan hujan selama 8 tahun tersebut menunjukkan kawasan Bandar Segamat menerima hujan bulanan yang berjulat daripada 2.5 mm (Februari 2004) sehingga 536.8 mm (Januari 2003). Taburan hujan bulanan Oktober sehingga Disember 2006 menunjukkan kenaikan secara positif, iaitu daripada 118 mm sehingga 435 mm/bulan tetapi berlaku penurunan (392 mm) pada Januari 2007. Hasil kajian menunjukkan bahawa hujan lebat yang luar biasa dan keadaan bentuk topografi yang rendah dan beralun di kawasan kajian adalah penyebab utama banjir di Segamat.

    Artikel ini bertujuan mengkaji keberkesanan program SHIELDS daripada perspektif pelajar dan urusetia yang telah turut terlibat. Program SHIELDS adalah satu bentuk program campbased yang diperkenalkan hasil kerjasama antara Kementerian Pendidikan dan Pihak Agensi Anti Dadah Kebangsaan (AADK). Nama SHEILDS ini adalah akronim yang bermaksud Sayangi Hidup, Elak Derita Selamanya. Sehubungan itu, program ini bertujuan meningkatkan kesedaran dan mengupayakan ketahanan diri murid (lingkungan umur 13 hingga 18 tahun) yang dikenalpasti berisiko dalam penyalahgunaan dadah. Kajian kuantitatif ini melibatkan dua fasa iaitu fasa pertama adalah mengedarkan soal selidik sebelum dan selepas program SHIELDS dijalankan. Manakala, fasa kedua pula adalah sesi follow up selepas 3 bulan terhadap 50 pelajar yang telah terlibat dengan program SHIELDS terlebih dahulu. Hasil kajian menunjukkan secara keseluruhan perspektif pelajar seramai 559 orang yang terlibat dengan program SHIELDS ini adalah baik dengan min=4.48. Kesimpulannya, dapatan ini menunjukkan bahawa program SHIELDS ini sewajarnya diteruskan dengan beberapa penambahbaikan untuk kelangsungan program dan kebaikan semua pihak yang terlibat.

    Penuaan ovari telah dikaitkan dengan tekanan oksidatif dan kehilangan fungsi ovari. Tokotrienol telah dibuktikan dapat memberi kesan yang baik terhadap sistem pembiakan wanita. Walau bagaimanapun, peranan tokotrienol ke atas metabolisma ovari dan seterusnya peningkatan kualiti oosit dalam mencit tua masih tidak diketahui. Oleh itu, hubungan antara perubahan aktiviti metabolik dalam ovari dan kualiti oosit dalam mencit tua selepas suplementasi fraksi kaya tokotrienol (TRF) telah dikaji. Mencit betina berusia enam minggu digunakan sebagai kumpulan Muda. Mencit betina berusia enam bulan dibahagikan kepada empat kumpulan iaitu kumpulan pertama yang diberikan minyak jagung-bebas tokoferol (kawalan) manakala tiga kumpulan yang lain diberi suplimen TRF pada dos 90, 120, dan 150 mg/kg. Rawatan diberikan secara oral selama dua bulan. Pada akhir rawatan, mencit dari semua kumpulan disuperovulasi dan kemudian dikorbankan.     Terumbu karang adalah ekosistem yang paling mempunyai kepelbagaian biologi di planet ini. Terumbu karang hanya dijumpai dalam lingkungan pengecilan parameter julat yang sempit, yang meletakkan terumbu karang berisiko di seluruh dunia. Terumbu karang dipengaruhi oleh kesan antropogenik dan alam sekitar yang mengubah taburan genera karang. Memahami di mana jenis karang yang berbeza terletak dan bagaimana setiap genus terjejas oleh tekanan tempatan dan global adalah penting untuk pemuliharaan. Teknik coral video transect mula digunakan pada bulan Mei 2016 untuk membandingkan taburan genera karang di tapak yang dipengaruhi oleh faktor-faktor yang berbeza. Tapak yang mempunyai impak paling sedikit mencatatkan pelitupan karang yang paling tinggi, tapak yang berkaitan dengan sedimen mencatatkan pelitupan yang paling rendah, dan tapak yang mempunyai aktiviti pelancongan yang tinggi mempunyai pelitupan alga yang paling besar.     The main goal of this research work is to measure the concentration levels of organochlorine residue in soil. The potential health risk of this pollutant on human was also determined. 10 samples were taken from a lowland paddy field situated in Kelantan, Malaysia. Physical parameters namely soil pH, organic carbon content, water content and particle size were identified to evaluate the quality of soil from the agriculture site. Soxhlet extraction and florisil clean-up process were applied to isolate 10 targeted organochlorine compounds prior to the final determination using a gas chromatography-electron capture detector. Soil from the lowland has characteristics such as slightly acidic, low organic carbon content, high water content and texture dominated by the sandy type. Concentration levels of six detected organochlorine pesticides were calculated in µg/kg. Hazard quotient value in all samples was less than the acceptable risk level HQ ≤ 1, thus reflecting the status of soil in the subjected area as unlikely to pose any adverse health effects.

    Mussel samples were used in this study to measure the levels of polybrominated diphenyl ethers (PBDEs) and organochlorines (OCs) in the coastal waters of Asian countries like Cambodia, China, Hong Kong, India, Indonesia, Japan, Korea, Malaysia, the Philippines, and Vietnam. PBDEs were detected in all the samples analyzed, and the concentrations ranged from 0.66 to 440 ng/g lipid wt. Apparently higher concentrations of PBDEs were found in mussels from the coastal waters of Korea, Hong Kong, China, and the Philippines, which suggests that significant sources of these chemicals exist in and around this region. With regard to the composition of PBDE congeners, BDE-47, BDE-99, and BDE-100 were the dominant congeners in most of the samples. Among the OCs analyzed, concentrations of DDTs were the highest followed by PCBs > CHLs > HCHs > HCB. Total concentrations of DDTs, PCBs, CHLs, and HCHs in mussel samples ranged from 21 to 58 000, 3.8 to 2000, 0.93 to 900, and 0.90 to 230 ng/g lipid wt., respectively. High levels of DDTs were found in mussels from Hong Kong, Vietnam, and China; PCBs were found in Japan, Hong Kong, and industrialized/urbanized locations in Korea, Indonesia, the Philippines, and India; CHLs were found in Japan and Hong Kong; HCHs were found in India and China. These countries seem to play a role as probable emission sources of corresponding contaminants in Asia and, in turn, may influence their global distribution.

    Pendidikan kemahiran resusitasi kardiopulmonari (CPR) kepada orang awam sangat penting dalam merendahkan kadar kematian serangan jantung. American Heart Association mencadangkan kemahiran tersebut harus dimasukkan ke dalam kurikulum sekolah. Pelatih rakan sebaya adalah kaedah yang berkos rendah dan berkesan dalam mencapai objektif ini. Objektif kajian terkawal 3 bulan ini adalah untuk membandingkan keberkesanan pengajaran kompresi CPR antara rakan sebaya dan jurulatih Basic Life Support (BLS) kepada pelajar sekolah menengah. Peningkatan pengetahuan dan kemahiran psikomotor pemampatan CPR adalah hasil utama yang dinilai. Dua belas pelatih rakan sebaya berusia 16 tahun dan dua belas pelatih BLS telah direkrut dalam kajian ini. Kompresi CPR diajarkan kepada 36 pelajar sekolah menengah secara rawak oleh pelatih rakan sebaya (Kumpulan P) atau pelatih BLS (Kumpulan B). Pra-ujian, pasca ujian serta ujian pengekalan 3 bulan mengenai pengetahuan dan kemahiran psikomotor telah dijalankan. Tidak terdapat perbezaan yang signifikan secara statistik dalam skor min pengetahuan dan psikomotor pada pra-ujian, pasca ujian serta ujian pengekalan 3 bulan antara Kumpulan P dan Kumpulan B.     A study on the quality of water abstracted for potable use was conducted in the Selangor River basin from November 2008 to July 2009. Seven sampling sites representing the intake points of water treatment plants in the basin were selected to determine the occurrence and level of 15 organochlorine pesticides (OCPs), six phthalate esters (PAEs) and bisphenol A (BPA). Results indicated OCPs were still detected regularly in 66.1 % of the samples with the Σ(15)OCPs ranging from 0.6-25.2 ng/L. The first data on PAEs contamination in the basin revealed Σ(6)PAEs concentrations were between 39.0 and 1,096.6 ng/L with a median concentration of 186.0 ng/L while BPA concentration ranged from <1.2 to 120.0 ng/L. Although di-n-butyl phthalate was detected in all the samples, concentrations of di-ethyl(hexyl)phthalate were higher. Sampling sites located downstream recorded the highest concentrations, together with samples collected during the dry season. Comparison of the detected contaminants with the Department of Environment Water Quality Index (DOE-WQI) showed some agreement between the concentration and the current classification of stream water. While the results suggest that the sites were only slightly polluted and suitable to be used as drinking water source, its presence is cause for concern especially to the fragile firefly "Pteroptyx tener" ecosystem located further downstream.

    A study to assess the level of organochlorine pesticides (OCPs) and bisphenol A (BPA) in edible marine biota collected from coastal waters of Malaysia was conducted using GC-MS and SPE extraction. An analytical method was developed and validated to measure the level of 15 OCPs and BPA simultaneously from five selected marine species. It was observed that some samples had low levels of p,p'-DDE, p,p'-DDT and p,p'- DDD ranging from 0.50 ng g(-1) to 22.49 ng g(-1) dry weight (d.w) but significantly elevated level of endosulfan I was detected in a stingray sample at 2880 ng g(-1) d.w. BPA was detected in 31 out of 57 samples with concentration ranging from below quantification level (LOQ: 3 ng g(-1)) to 729 ng g(-1) d.w. The presence of OCPs is most likely from past use although there is also indication of illegal use in recent times. The study also reveals that BPA is more widely distributed in coastal species caught off the coast of the most developed state. The potential health risk from dietary intakes of OCPs and BPA from the analysed fish species was negligible.

    Dalam kajian ini, pendekatan pengelompokan kabur Gustafson-Kessel (Gx) telah digunakan untuk mengelaskan 35 stesen hujan di Semenanjung Malaysia ke dalam rantau homogen. Pertama, algoritma pengelompokan kabur GK digunakan untuk mengenal pasti rantau awal. Kemudian, diuji keserasian dan kehomogenan rantau berkenaan. Akhir sekali, penyesuaian rantau dilakukan untuk mendapatkan rantau homogen. Hasil kajian mendapati 35 stesen hujan kajian boleh dibahagikan kepada enam rantau yang homogen. Rantau 1 meliputi bahagian barat laut dan utara Semenanjung Malaysia, rantau 2, 3 dan 4 meliputi bahagian barat, rantau 5 meliputi bahagian barat daya dan rantau 6 meliputi bahagian timur. Hasil kajian ini juga memperlihatkan bahawa berdasarkan nilai purata Indeks Kerpasan Piawai (sPi) skala masa satu bulan, rantau 2 lebih sering mengalami keadaan kemarau melampau. Walau bagaimanapun, berdasarkan sPi skala masa satu bulan, peristiwa kemarau terjadi secara rawak dalam semua rantau yang dianalisis , bahkan semua rantau tersebut pernah mengalami kejadian kemarau melampau dalam tempoh masa setahun. Hasil kajian ini turut menunjukkan bahawa pendekatan pengelompokan kabur Gustafson-Kessel boleh digunakan untuk membina rantau homogen.

    Pengurusan kendiri asma menggunakan pelan tindakan asma secara bertulis (WAAP) penting dalam memastikan pengurusan asma yang berkesan kerana ia mengurangkan kadar kehadiran ke hospital. Kajian ini menilai peningkatan pengetahuan dan kawalan asma menggunakan WAAP di kalangan pesakit asma di Jabatan Kecemasan. Kajian cubaan kawalan rawak ini dijalankan selama 19 bulan di Jabatan Perubatan Kecemasan sebuah hospital universiti. Kajian ini melibatkan pesakit dewasa serangan asma ringan dan sederhana. Pesakit dibahagikan secara rawak kepada dua kumpulan iaitu kumpulan intervensi dan kawalan. Soal selidik pengetahuan penyakit asma diberikan pada pesakit. Pesakit dari kedua kumpulan menerima kaunseling tentang penyakit asma berdasarkan risalah asma yang telah disediakan. Kumpulan intervensi, kemudiannya menerima kaunseling WAAP mengikut keperluan individu tersebut. Kumpulan kawalan menerima kaunseling lisan berdasarkan perawatan. Susulan kajian dijalankan pada 1 dan 3 bulan. Pengetahuan penyakit asma dan kawalan penyakit asma dinilai melalui tinjauan telefon. Lima puluh pesakit telah dibahagikan secara rawak kepada kumpulan intervensi dan kawalan. Setiap kumpulan menerima 25 pesakit. Skor pengetahuan kumpulan intervensi jauh lebih tinggi daripada kawalan (16.88+0.44 vs 13.36+3.72; p19) pada 1 bulan berbanding kumpulan kawalan (20.64+3.26 vs 17.72+4.17). Kedua-dua kumpulan mencapai kawalan yang baik pada 3 bulan. Pengurusan kendiri asma dengan menggunakan WAAP di Jabatan Kecemasan meningkatkan pengetahuan dan kawalan asma di kalangan pesakit yang mendapat serangan asma ringan dan sederhana.

    A method to determine six organochlorine and three pyrethroid pesticides in grape, orange, tomato, carrot and green mustard based on solvent extraction followed by solid phase extraction (SPE) clean-up is described. The pesticides were spiked into the sample prior to analysis, extracted with ethyl acetate, evaporated and reconstituted with a solvent mixture of acetone:n-hexane (3:7). Three different sorbents (Strong Anion Exchanger/Primary Secondary Amine (SAX/PSA), Florisil and C18) were used for the clean-up step. Pesticides were eluted with 5mL of acetone:n-hexane (3:7, v/v) and determined by gas chromatography and electron-capture detection (GC-ECD). SAX/PSA was the sorbent, which provided chromatograms with less interference and the mean recoveries obtained were within 70-120% except for captafol. The captafol recoveries for grape were within acceptable range with C18 clean-up column.

    This study investigates the presence and distribution of organochlorine pesticides in streams and the lake in the Sembrong Lake Basin in Malaysia. The catchment of Sembrong Lake has been converted to agricultural areas over the past 30 years, with oil palm plantations and modern agricultural farming being the main land use. Surface water samples were collected from eight sites comprising the stream and lake and analysed for 19 organochlorine pesticides (OCPs). In situ measurement of temperature, dissolved oxygen, pH and conductivity were also undertaken at each site. Aldrin, endrin, δ-BHC, 4,4-DDT, methoxychlor and endosulfan were the main OCPs detected in the lake basin. The total OCP concentration ranged between 5.42 and 349.2 ng/L. The most frequently detected OCPs were δ-BHC, heptachlor and aldrin. The maximum values detected were 23.0, 43.2 and 50.4 ng/L respectively. The highest concentration of OCPs was attributed to 4,4-DDT, but such high residue was rare and only detected once. Other OCP residues were low.     This study determined the concentrations of polychlorinated dibenzo-p-dioxins/dibenzofurans (PCDD/Fs), polychlorinated biphenyls (PCBs), organochlorine (OC) pesticides, and tris(4-chlorophenyl) methane (TCPMe) in human breast milk samples collected in 2003 from primipara mothers living in Penang, Malaysia. OCs were detected in all the samples analyzed with DDTs, hexachlorocyclohexane isomers (HCHs), and PCBs as the major contaminants followed by chlordane compounds (CHLs), hexachlorobenzene (HCB), and TCPMe. The residue levels of DDTs, HCHs, and CHLs were comparable to or higher than those in general populations of other countries, whereas PCBs and HCB were relatively low. In addition, dioxins and related compounds were also detected with a range of dioxin equivalent concentrations from 3.4 to 24 pg-TEQs/g lipid wt. Levels of toxic equivalents (TEQs) were slightly higher than those in other developing countries but still much lower than those of industrialized nations. One donor mother contained a high TEQs level, equal to the mean value in human breast milk from Japan, implying that some of the residents in Malaysia may be exposed to specific pollution sources of dioxins and related compounds. No association was observed between OCs concentrations and maternal characteristics, which might be related to a limited number of samples, narrow range of age of the donor mothers, and/or other external factors. The recently identified endocrine disrupter, TCPMe, was also detected in all human breast milk samples of this study. A significant positive correlation was observed between TCPMe and DDTs, suggesting that technical DDT might be a source of TCPMe in Malaysia. The present study provides a useful baseline for future studies on the accumulations of OCs in the general population of Malaysia.

    Halogenated compounds are recalcitrant environmental pollutants prevalent in agricultural fields, waste waters and industrial by-products, but they can be degraded by dehalogenase-containing microbes. Notably, 2-haloalkanoic acid dehalogenases are employed to resolve optically active chloropropionates, as exemplified by the d-specific dehalogenase from Rhizobium sp. RCI (DehD), which acts on d-2-chloropropionate but not on its l-enantiomer. The catalytic residues of this dehalogenase responsible for its affinity toward d-2-chloropropionate have not been experimentally determined, although its three-dimensional crystal structure has been solved. For this study, we performed in silico docking and molecular dynamic simulations of complexes formed by this dehalogenase and d- or l-2-chloropropionate. Arg134 of the enzyme plays the key role in the stereospecific binding and Arg16 is in a position that would allow it to activate a water molecule for hydrolytic attack on the d-2-chloropropionate chiral carbon for release of the halide ion to yield l-2-hydroxypropionate. We propose that within the DehD active site, the NH group of Arg134 can form a hydrogen bond with the carboxylate of d-2-chloropropionate with a strength of ∼4 kcal/mol that may act as an acid-base catalyst, whereas, when l-2-chloropropionate is present, this bond cannot be formed. The significance of the present work is vital for rational design of this dehalogenase in order to confirm the involvement of Arg16 and Arg134 residues implicated in hydrolysis and binding of d-2-chloropropionate in the active site of d-specific dehalogenase from Rhizobium sp. RC1.
