    This study examined the physicochemical properties of rice flours with five different particle sizes (≤63, 80, 100, 125, and 140 μm) prepared by dry milling and their effects on textural properties of laksa noodles. Rice flour with the smallest particle size had the highest water absorption index, peak viscosity, hot paste viscosity, breakdown, final or cold paste viscosity and gel hardness, but the lowest gelatinization temperature. Reduction of rice flour particle size improved textural properties of laksa noodle. Laksa noodle produced from rice flour with the smallest particle size had the best textural properties.

    Changes in nitrate and nitrite contents (leaves and stem) of Amaranthus gangeticus (AG) and Amaranthus paniculatus (AP), resulting from blanching, storage time (0-4 days), storage temperature (0 and 4ºC), and reheating were analysed. Results showed that fresh AG (1859 ± 7.07 mg/kg) had higher nitrite content than AP (1262 ± 2.12 mg/kg). Nitrites content was 506 ± 2 and 825 ± 3.5 mg/kg for AG and AP, respectively. Reheating and storage times significantly increased the conversion of nitrate to nitrite in AG and AP. Storage at 0 and 4oC exhibited a significant change (P < 0.05) in nitrate and nitrite contents for both samples. Higher nitrite content was found in AP when stored at 4oC and 0oC. The present study indicated that storage time and temperature affected the nitrite contents in blanched AG and AP when stored in low temperatures. Apart from that reheating was also found to increase the formation of nitrite.

    The temperature profile of a cryogenic system for cooling of beryllium filter of a small-angle neutron scattering (SANS) instrument of TRIGA MARK II PUSPATI research reactor was investigated using computational fluid dynamics (CFD) modeling and simulation. The efficient cooling of beryllium filter is important for obtaining higher cold neutron transmission for the SANS instrument. This paper presents the transient CFD results of temperature distributions via the thermal link to the beryllium and simulation of heat
    flux. The temperature simulation data are also compared with the experimental results for the cooling time and distribution to the beryllium.

    The structure of psychrophilic chitinase (CHI II) from Glaciozyma antarctica PI12 has yet to be studied in detail. Due to its low sequence identity (<30 %), the structural prediction of CHI II is a challenge. A 3D model of CHI II was built by first using a threading approach to search for a suitable template and to generate an optimum target-template alignment, followed by model building using MODELLER9v7. Analysis of the catalytic insertion domain structure in CHI II revealed an increase in the number of aromatic residues and longer loops compared to mesophilic and thermophilic chitinases. A molecular dynamics simulation was used to examine the stability of the CHI II structure at 273, 288 and 300 K. Structural analysis of the substrate-binding cleft revealed a few exposed aromatic residues. Substitutions of certain amino acids in the surface and loop regions of CHI II conferred an increased flexibility to the enzyme, allowing for an adaptation to cold temperatures. A substrate binding comparison of CHI II with the mesophilic chitinase from Coccidioides immitis, 1D2K, suggested that the psychrophilic adaptation and catalytic activity at low temperatures were achieved through a reduction in the number of salt bridges, fewer hydrogen bonds and an increase in the exposure of the hydrophobic side chains to the solvent.

    RNA-binding proteins (RBPs) have been implicated as regulatory proteins involved in the post-transcriptional processes of gene expression in plants under various stress conditions. In this study, we report the cloning and characterization of a gene, designated as EgRBP42, encoding a member of the plant heterogeneous nuclear ribonucleoprotein (hnRNP)-like RBP family from oil palm (Elaeis guineensis Jacq.). EgRBP42 consists of two N-terminal RNA recognition motifs and a glycine-rich domain at the C-terminus. The upstream region of EgRBP42 has multiple light-responsive, stress-responsive regulatory elements and regulatory elements associated with flower development. Real-time RT-PCR analysis of EgRBP42 showed that EgRBP42 was expressed in oil palm tissues tested, including leaf, shoot apical meristem, root, female inflorescence, male inflorescence and mesocarp with the lowest transcript level in the roots. EgRBP42 protein interacted with transcripts associated with transcription, translation and stress responses using pull-down assay and electrophoretic mobility shift assay. The accumulation of EgRBP42 and its interacting transcripts were induced by abiotic stresses, including salinity, drought, submergence, cold and heat stresses in leaf discs. Collectively, the data suggested that EgRBP42 is a RBP, which responds to various abiotic stresses and could be advantageous for oil palm under stress conditions. Key message EgRBP42 may be involved in the post-transcriptional regulation of stress-related genes important for plant stress response and adaptation.

    The viability and activity of Bifidobacterium pseudocatenulatum G4, B. longum BB 536 and yoghurt cultures (Lactobacillus delbrueckii ssp. bulgaricus and Streptococcus thermophilus) were studied in yoghurt containing 0.75% Mangefira pajang fibrous polysaccharides (MPFP) and inulin. Growth of probiotic organisms, their proteolytic activities, the production of short chain fatty acids (lactic, acetic and propionic) and the pH of the yoghurt samples were determined during refrigerated storage at 4 °C for 28 d. B. pseudocatenulatum G4 and B. longum BB 536 showed better growth and activity in the presence of MPFP and inulin, which significantly increased the production of short chain fatty acids as well as the proteolytic activity of these organisms.

    The Toba supereruption in Sumatra, ∼74 thousand years (ka) ago, was the largest terrestrial volcanic event of the Quaternary. Ash and sulfate aerosols were deposited in both hemispheres, forming a time-marker horizon that can be used to synchronize late Quaternary records globally. A precise numerical age for this event has proved elusive, with dating uncertainties larger than the millennial-scale climate cycles that characterized this period. We report an astronomically calibrated (40)Ar/(39)Ar age of 73.88 ± 0.32 ka (1σ, full external errors) for sanidine crystals extracted from Toba deposits in the Lenggong Valley, Malaysia, 350 km from the eruption source and 6 km from an archaeological site with stone artifacts buried by ash. If these artifacts were made by Homo sapiens, as has been suggested, then our age indicates that modern humans had reached Southeast Asia by ∼74 ka ago. Our (40)Ar/(39)Ar age is an order-of-magnitude more precise than previous estimates, resolving the timing of the eruption to the middle of the cold interval between Dansgaard-Oeschger events 20 and 19, when a peak in sulfate concentration occurred as registered by Greenland ice cores. This peak is followed by a ∼10 °C drop in the Greenland surface temperature over ∼150 y, revealing the possible climatic impact of the eruption. Our (40)Ar/(39)Ar age also provides a high-precision calibration point for other ice, marine, and terrestrial archives containing Toba sulfates and ash, facilitating their global synchronization at unprecedented resolution for a critical period in Earth and human history beyond the range of (14)C dating.

    Polypyrrole (PPy) and polypyrrole-carboxylic functionalized multi wall carbon nanotube composites (PPy/f-MWCNT) were synthesized by in situ chemical oxidative polymerization of pyrrole on the carbon nanotubes (CNTs). The structure of the resulting complex nanotubes was characterized by transmission electron microscopy (TEM) and X-ray diffraction (XRD). The effects of f-MWCNT concentration on the electrical properties of the resulting composites were studied at temperatures between 100 K and 300 K. The Hall mobility and Hall coefficient of PPy and PPy/f-MWCNT composite samples with different concentrations of f-MWCNT were measured using the van der Pauw technique. The mobility decreased slightly with increasing temperature, while the conductivity was dominated by the gradually increasing carrier density.

    α-Mangostin was extracted from the pericarp of the Malaysian local Garcinia mangostana linn., The structure was characterised by Infrared red, UV-Visible and Nuclear Magnetic Resonance spectroscopic data. The fluorescence peak at 500nm in ethanol was not observed in PNIPAM microgel solution. The increase of colloidal size of the gel in the presence of α-mangostin was studied by Dynamic Light Scattering and Transmission Electron Microscope. The size of the particle also increases with increasing temperature up to 45⁰C after which it began to shrink. The TEM micrograph at 45°C showed a uniformly structured pattern of the gel occurs in the range of the lowest solution critical temperature.

    Iran is situated in a very diverse environmental area. The climate of the region is varied and influencedby different patterns. In order to best describe the expected climate change impacts for the region,climate change scenarios and climate variables must be developed on a regional, or even site-specific,scale. The weather generator is one of the valid downscaling methods. In the current study, LARSWG(a weather generator) and the outputs from ECHO-G for present climate, as well as future timeslice of 2010-2039 based on A1 scenario, were used to evaluate LARS-WG as a tool at 13 synopticstations located in the north and northeast parts of Iran. The results obtained in this study illustratethat LARS-WG has a reasonable capability of simulating the minimum and maximum temperaturesand precipitation. In addition, the results showed that the mean precipitation decreased in Semnan, thesouth of Khorasan and Golestan. Meanwhile, the mean temperature during 2010-2039 would increaseby 0.5°C, especially in the cold season.

    The objective of the present study was to simultaneously evaluate the effect of a postharvest treatment on the pepper's antioxidant content and its ability to retain its economical value during the postharvest period. The fruits were pretreated by modified atmosphere packaging (MAP) with or without treatment with 1-methylcyclopropene (1-MCP) before cold storage at 10°C. Changes in the levels of non-enzymatic antioxidants, including the total phenolic, ascorbic acid levels and the total glutathione level, as well as enzymatic antioxidants, including ascorbate peroxidase (APX), glutathione reductase (GR), and catalase (CAT), were determined. Both treatments successfully extended the shelf life of the fruit for up to 25 days, and a high level of antioxidant capacity was maintained throughout the storage period. However, 1-MCP treatment maintained the high antioxidant capacity for a longer period of time. The 1-MCP-treated peppers maintained high levels of phenolic content, a high reduced glutathione (GSH)/oxidised glutathione (GSSG) ratio, decreased levels of ascorbic acid and CAT activity, and increased levels of APX and GR compared with the peppers that were not treated with 1-MCP. The overall results suggested that a combination of 1-MCP and MAP was the most effective treatment for extending shelf life while retaining the nutritional benefits.

    The psychrophilic yeast Glaciozyma antarctica demonstrated high antifreeze activity in its culture filtrate. The culture filtrate exhibited both thermal hysteresis (TH) and ice recrystallization inhibition (RI) properties. The TH of 0.1 °C was comparable to that previously reported for bacteria and fungi. A genome sequence survey of the G. antarctica genome identified a novel antifreeze protein gene. The cDNA encoded a 177 amino acid protein with 30 % similarity to a fungal antifreeze protein from Typhula ishikariensis. The expression levels of AFP1 were quantified via real time-quantitative polymerase chain reaction (RT-qPCR), and the highest expression levels were detected within 6 h of growth at -12 °C. The cDNA of the antifreeze protein was cloned into an Escherichia coli expression system. Expression of recombinant Afp1 in E. coli resulted in the formation of inclusion bodies that were subsequently denatured by treatment with urea and allowed to refold in vitro. Activity assays of the recombinant Afp1 confirmed the antifreeze protein properties with a high TH value of 0.08 °C.

    A new strain of psychrophilic bacteria (designated strain AMS8) from Antarctic soil was screened for extracellular lipolytic activity and further analyzed using molecular approach. Analysis of 16S rDNA showed that strain AMS8 was similar to Pseudomonas sp. A lipase gene named lipAMS8 was successfully isolated from strain AMS8, cloned, sequenced and overexpressed in Escherichia coli. Sequence analysis revealed that lipAMS8 consist of 1,431 bp nucleotides that encoded a polypeptide consisting of 476 amino acids. It lacked an N-terminal signal peptide and contained a glycine- and aspartate-rich nonapeptide sequence at the C-terminus, which are known to be the characteristics of repeats-in-toxin bacterial lipases. Furthermore, the substrate binding site of lipAMS8 was identified as S(207), D(255) and H(313), based on homology modeling and multiple sequence alignment. Crude lipase exhibited maximum activity at 20 °C and retained almost 50 % of its activity at 10 °C. The molecular weight of lipAMS8 was estimated to be 50 kDa via sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The optimal expression level was attained using the recombinant plasmid pET32b/BL21(DE3) expressed at 15 °C for 8 h, induced by 0.1 mM isopropyl β-D thiogalactoside (IPTG) at E. coli growth optimal density of 0.5.

    A novel α-amylase was isolated successfully from Glaciozyma antarctica PI12 using DNA walking and reverse transcription-polymerase chain reaction (RT-PCR) methods. The structure of this psychrophilic α-amylase (AmyPI12) from G. antarctica PI12 has yet to be studied in detail. A 3D model of AmyPI12 was built using a homology modelling approach to search for a suitable template and to generate an optimum target-template alignment, followed by model building using MODELLER9.9. Analysis of the AmyPI12 model revealed the presence of binding sites for a conserved calcium ion (CaI), non-conserved calcium ions (CaII and CaIII) and a sodium ion (Na). Compared with its template-the thermostable α-amylase from Bacillus stearothermophilus (BSTA)-the binding of CaII, CaIII and Na ions in AmyPI12 was observed to be looser, which suggests that the low stability of AmyPI12 allows the protein to work at different temperature scales. The AmyPI12 amino acid sequence and model were compared with thermophilic α-amylases from Bacillus species that provided the highest structural similarities with AmyPI12. These comparative studies will enable identification of possible determinants of cold adaptation.

    This paper presents a numerical study of the thermal performance of fins mounted on the bottom wall of a horizontal channel and cooled with either pure water or an Al2O3-water nanofluid. The bottom wall of the channel is heated at a constant temperature and cooled by mixed convection of laminar flow at a relatively low temperature. The results of the numerical simulation indicate that the heat transfer rate of fins is significantly affected by the Reynolds number (Re) and the thermal conductivity of the fins. The influence of the solid volume fraction on the increase of heat transfer is more noticeable at higher values of the Re.

    The psychrophilic enzyme is an interesting subject to study due to its special ability to adapt to extreme temperatures, unlike typical enzymes. Utilizing computer-aided software, the predicted structure and function of the enzyme lipase AMS8 (LipAMS8) (isolated from the psychrophilic Pseudomonas sp., obtained from the Antarctic soil) are studied. The enzyme shows significant sequence similarities with lipases from Pseudomonas sp. MIS38 and Serratia marcescens. These similarities aid in the prediction of the 3D molecular structure of the enzyme. In this study, 12 ns MD simulation is performed at different temperatures for structural flexibility and stability analysis. The results show that the enzyme is most stable at 0°C and 5°C. In terms of stability and flexibility, the catalytic domain (N-terminus) maintained its stability more than the noncatalytic domain (C-terminus), but the non-catalytic domain showed higher flexibility than the catalytic domain. The analysis of the structure and function of LipAMS8 provides new insights into the structural adaptation of this protein at low temperatures. The information obtained could be a useful tool for low temperature industrial applications and molecular engineering purposes, in the near future.

    The glow curve in TLD-100 was compared by applying long preheat time, short preheat time
    techniques and without preheat technique before the TLD readout. Fading effect of the TLD signal
    upon certain storage time with long preheat time (100°C, 10 minutes using the oven) and short
    preheat time techniques (100°C, 10 seconds using the reader) were also studied. 15 TLD-100 chips
    were used with 3 of the TLD chips were used for measuring background radiation. 12 TLD chips
    were annealed, irradiated, preheated long and short preheat time techniques) and analyzed. The TL
    signals output from TLD chips of without preheated were used as the control. Two sets of data were
    taken using TLD chips irradiated with 6 MV and 10 MV photon beams. TL signal output was
    recorded the highest for short preheat time, followed by long preheat time and no preheating. The
    TL signal loss upon certain storage time was also reduced when short preheat time technique was
    applied. By applying long preheat time technique the low temperature peak in the glow curve was
    completely removed for both energies. Whereas, TLD chips exposed to 6 MV and with short preheat
    time technique the low temperature peak did not disappear completely but decreased in intensity as
    compared to the control data by 19.80%, 37.69%, 48.19% and 100% at 24, 48, 72 and 96 hours
    after exposure prior to readout, respectively. Meanwhile, for 10 MV photon beam with short
    preheat time, the small peak intensity was reduced by 19.58% for readout at 24 hours after
    irradiation and 100% for 48,72 and 96 hours delayed time prior to readout. It was observed that
    the TLD-100 was highly dependent on preheat heating time before readout. Short preheat time
    technique was able to reduce post irradiation fading of TLD-100 dosimeters
