Enzymatic saccharification of corn stover using Phanerochaete chrysosporium and Gloeophyllum trabeum and subsequent fermentation of the saccharification products to ethanol by Saccharomyces cerevisiae and Escherichia coli K011 were achieved. Prior to simultaneous saccharification and fermentation (SSF) for ethanol production, solid-state fermentation was performed for four days on ground corn stover using either P. chrysosporium or G. trabeum to induce in situ cellulase production. During SSF with S. cerevisiae or E. coli, ethanol production was the highest on day 4 for all samples. For corn stover treated with P. chrysosporium, the conversion to ethanol was 2.29 g/100 g corn stover with S. cerevisiae as the fermenting organism, whereas for the sample inoculated with E. coli K011, the ethanol production was 4.14 g/100 g corn stover. Corn stover treated with G. trabeum showed a conversion 1.90 and 4.79 g/100 g corn stover with S. cerevisiae and E. coli K011 as the fermenting organisms, respectively. Other fermentation co-products, such as acetic acid and lactic acid, were also monitored. Acetic acid production ranged between 0.45 and 0.78 g/100 g corn stover, while no lactic acid production was detected throughout the 5 days of SSF. The results of our experiment suggest that it is possible to perform SSF of corn stover using P. chrysosporium, G. trabeum, S. cerevisiae and E. coli K011 for the production of fuel ethanol.

Antioxidant and gastroprotective activities of aqueous and ethanolic extract of Andrographis paniculata leaves in rats have been reported. Sprague Dawley rats, 6 per group were used and rats in groups 1 to 6 were pretreated with (0.25% w/v) carboxymethyl cellulose (negative control, 5 ml/kg), 20 mg/kg omeprazole (positive control), (250 mg/kg and 500 mg/kg) of aqueous leaf extracts (APLAE) and (250 and 500 mg/kg) of ethanol leaf extracts (APLEE) respectively. Animals were orally administered with 95% ethanol (5 ml/kg) 60 min after their pretreatments. Rats were sacrificed 1 h after treatment and gastric contents were collected to measure pH and mucous weight. Stomach was analyzed for gross and histological changes. Ulcer control group showed extensive lesions of gastric mucosal layer, whereas rats pretreated with omeprazole, 250 and 500 mg/kg of APLAE showed significant and dose dependent reduction in gastric lesions with increased pH and mucus content of stomach. Rats pretreated with 250 or 500 mg/kg of APLEE showed significantly better inhibition of gastric mucosal lesions. Further, the in vitro antioxidant studies using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay showed that ethanol extracts have superior free radical scavenging activity with IC50 value = 10.9 than aqueous extracts with IC50 value = 24.65. Results of this study showed that pretreatment with ethonolic extract of A. paniculata ethanolic provided significant protection against gastric ulcer by regulating of pH, mucous production and antioxidant property.

ETHNOPHARMACOLOGICAL RELEVANCE: Sargassum polycystum, a type of brown seaweed, has been used for the treatment of skin-related disorders in traditional medicine.

AIM OF THE STUDY: The aim of the present study is to investigate the antimelanogenesis effect of Sargassum polycystum extracts by cell-free mushroom tyrosinase assay followed by cell viability assay, cellular tyrosinase assay and melanin content assay using B16F10 murine melanoma cells.

MATERIALS AND METHODS: Sargassum polycystum was extracted with 95% ethanol and further fractionated with hexane, ethyl acetate and water. The ethanolic crude extract and its fractionated extracts were tested for their potential to act as antimelanogenesis or skin-whitening agents by their abilities to inhibit tyrosinase activity in the cell-free mushroom tyrosinase assay and cellular tyrosinase derived from melanin-forming B16F10 murine melanoma cells. The tyrosinase inhibitory activity was correlated to the inhibition of melanin production in α-MSH-stimulated and unstimulated B16F10 cells.

RESULTS: Sargassum polycystum ethanolic extract and its fractions had little or no inhibitory effect on mushroom tyrosinase activity. However, when tested on cellular tyrosinase, the ethanolic extract and its non-polar fraction, hexane fraction (SPHF), showed significant inhibition of cellular tyrosinase activity. In parallel to its cellular tyrosinase inhibitory activity, SPHF was also able to inhibit basal and α-MSH-stimulated melanin production in B16F10 cells.

CONCLUSIONS: Our findings showed that (i) cellular tyrosinase assay is more reliable than mushroom tyrosinase assay in the initial testing of potential antimelanogenesis agents and, (ii) SPHF inhibited melanogenesis by inhibiting cellular tyrosinase activity. SPHF may be useful for treating hyperpigmentation and as a skin-whitening agent in cosmetics industry.

In the organosolv pulping of the oil palm fronds, the influence of the operational variables of the pulping reactor (viz. cooking temperature and time, ethanol and NaOH concentration) on the properties of the resulting pulp (yield and kappa number) and paper sheets (tensile index and tear index) was investigated using a wavelet neural network model. The experimental results with error less than 0.0965 (in terms of MSE) were produced, and were then compared with those obtained from the response surface methodology. Performance assessment indicated that the neural network model possessed superior predictive ability than the polynomial model, since a very close agreement between the experimental and the predicted values was obtained.

Pineapple (Ananas comosus), Bromeliaceae family, is a fruit grows in tropical countries including Malaysia. This fruit has several pharmacological benefits due to the presence of high concentration of bromelain (cysteine proteases). Condition of elevated temperature will induce deformation of enzyme and result in loss of activity. Sulfhydryl groups in cysteine proteases are readily to be oxidized and might account for the denaturation of bromelain at elevated temperature. Polyphenol from ethanolic cashew leave extract could be complexed with bromelain to stabilize the enzymatic activity. In thermal stability test, the heat damage effect on bromelain was ten times reduced after complexing with cashew extract. The enzymatic activity of free bromelain decreased gradually from 25 o C to 95 o C. Complexed bromelain was stable in activity to heating up to 85 o C. Bromelainpolyphenol complex showed a good heat resistance. The result revealed that polyphenol could protect bromelain in pineapple juice from heat denaturation.

One of the compounds present in Pluchea indica extracts is antioxidant which plays an important role in inhibiting free radicals and thus protects humans against infections and degenerative diseases, such as cancer, arthritis, and ageing process. The main objective of this study was to investigate and determine the total phenolic compounds of Pluchea indica in different concentrations of ethanolic extracts. This species was chosen because of its high phytonutrient compounds with potential medicinal properties. There was a significant difference (P ≤ 0.05) in the total phenolic among the different parts of the tested plant. 50% of the ethanolic extract produced the highest total phenolic compounds (1775.00±86.00 to 658.95±5.00 µmol/g), followed by water extract (759.79±1.53 µmol/g) and 100% ethanol extract (352.72±22.30 to 249.29±5.37 µmol/g), respectively. In terms of the plant parts, the leaves contained the highest phenolic compounds (1775.00±86.00 µmol/g in 50% ethanol extract, 759.79±1.53 µmol/g in 100% aqueous extract and 352.72±22.30 µmol/g in 100% ethanol extract), followed by the stems (990.22±24.00 µmol/g in 50% ethanol extract, 990.22±24.59 µmol/g in 100% aqueous extract and 293.48±0.00 µmol/g in 100% ethanol extract). Meanwhile, lower total phenolic compounds were detected in the flowers (727.71±11.00 µmol/g in 50% ethanol extract, 603.81±8.46 µmol/g in 100% aqueous extract and 249.29±5.37 µmol/g in 100% ethanol extract) and roots (658.95±5.00 µmol/g in 50% ethanol extract, 450.00±10.76 µmol/g in 100% aqueous extract and 272.28±0.53 µmol/g in 100% ethanol extract). Based on these findings, Pluchea indica has potential medicinal properties that can be further developed to produce neutraceutical products, diet supplements or cosmetic products. However, further research should first be conducted on the effects of these compounds on laboratory animals.

Single-walled carbon nanotubes (SWCNTs) were synthesized by catalytic chemical vapor deposition (CCVD) of ethanol (C2H5OH) over Fe-Mo-MgO catalyst by using argon as a carrier gas. The reaction conditions are important factors that influence the yield and quality of carbon nanotubes. The effects of temperature and flow rate of carrier gas were investigated to increase the yield of carbon nanotubes. The synthesized carbon nanotubes were characterized by scanning electron microscopy, transmission electron microscopy, X-Ray diffraction and thermo-gravimetric analysis. The results showed that the growth of carbon nanotubes was effectively influenced by the reaction ambience and the synthesis condition. The temperature and flow rate of carrier gas played a key role in the yield and quality of synthesized CNTs. The estimated yield of synthesized carbon nanotubes was almost over 70%.

Natural organic and abundant resources biopolymers received more attention due to their low cost, availability and degradability after usage. Cassava skin was used as natural fillers to the polyvinyl alcohol (PVA). Cassava skin/poly vinyl alcohol blends were compounded using melt extrusion twin screw extruder and test samples were prepared using the compression method. Various ratios of cassava skin and glycerol were investigated to identify suitable composition based on the water absorption and tensile properties. The water absorption of the cassava skins/PVA samples increased at higher composition of cassava skin due to their hydrophilic properties but decrease with glycerol content. The strength of the cassava skins/PVA samples increased with the higher composition of cassava skin up to 70 wt% while gradually decreased with the increasing composition of glycerol. The Young modulus increased with glycerol content but decreased with fibre loading up to 70 wt%. Elongation at break decreased with fibre loading and glycerol up to 70 wt% and 30 phr, respectively.

Several methods of incorporating sago pith waste (SPW) into poly(vinyl alcohol) (PVA) had been conducted: (i) dry blending (PVA/SPW/G), (ii) blending of SPW and pre-plasticized PVA (pPVA/SPW/G) and (iii) blending of pre-plasticized of both PVA and SPW (pPVA/pSPW). The effect of the compounding method on the mechanical and water absorption properties were investigated. The addition of SPW into PVA greatly reduced the tensile strength and elongation at break. The tensile strength and elongation at break of PVA/SPW composites with identical geometry during compounding stage (powder/powder and pellet/pellet), which were PVA/SPW/G and pPVA/pSPW yielded the highest value. The percentage of water absorbed by PVA/SPW/G (without pre-plasticization) was the highest, followed by pPVA/pSPW and pPVA/SPW/G.

In this study, two nickel based electrodes were prepared; nickel foil and nickel-polyvinylchloride (Ni-PVC), in order to study their electrochemical behavior using cyclic voltammetry, CV and chronocoulometry, CC. Ni electrode was prepared from Ni metal foil while Ni-PVC electrode was prepared by mixing a weighed portion of Ni powder and PVC in THF solvent, swirled until the suspension was homogeneous and drying the suspension in an oven at 50oC for 3 h. The dry sample was then placed in a 1 cm diameter stainless steel mould and pressed at 10 ton/cm2. From CV data, Ni-PVC electrode showed a better electrochemical behavior compared to Ni metal foil electrode. The use of Ni-PVC electrode at higher concentration of supporting electrolyte (1.0 M KOH) was better than at lower concentration of the same supporting electrolyte in electroxidation of ethanol. In addition to acetic acid, the oxidation of ethanol also produced ethyl acetate and acetaldehyde.

BACKGROUND: Recently there has been a remarkable surge of interest about natural products and their applications in the cosmetic industry. Topical delivery of antioxidants from natural sources is one of the approaches used to reverse signs of skin aging. The aim of this research was to develop a nanoemulsion cream for topical delivery of 30% ethanolic extract derived from local Phyllanthus urinaria (P. urinaria) for skin antiaging.

METHODS: Palm kernel oil esters (PKOEs)-based nanoemulsions were loaded with P. urinaria extract using a spontaneous method and characterized with respect to particle size, zeta potential, and rheological properties. The release profile of the extract was evaluated using in vitro Franz diffusion cells from an artificial membrane and the antioxidant activity of the extract released was evaluated using the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) method.

RESULTS: Formulation F12 consisted of wt/wt, 0.05% P. urinaria extract, 1% cetyl alcohol, 0.5% glyceryl monostearate, 12% PKOEs, and 27% Tween 80/Span 80 (9/1) with a hydrophilic lipophilic balance of 13.9, and a 59.5% phosphate buffer system at pH 7.4. Formulation F36 was comprised of 0.05% P. urinaria extract, 1% cetyl alcohol, 1% glyceryl monostearate, 14% PKOEs, 28% Tween 80/Span 80 (9/1) with a hydrophilic lipophilic balance of 13.9, and 56% phosphate buffer system at pH 7.4 with shear thinning and thixotropy. The droplet size of F12 and F36 was 30.74 nm and 35.71 nm, respectively, and their nanosizes were confirmed by transmission electron microscopy images. Thereafter, 51.30% and 51.02% of the loaded extract was released from F12 and F36 through an artificial cellulose membrane, scavenging 29.89% and 30.05% of DPPH radical activity, respectively.

CONCLUSION: The P. urinaria extract was successfully incorporated into a PKOEs-based nanoemulsion delivery system. In vitro release of the extract from the formulations showed DPPH radical scavenging activity. These formulations can neutralize reactive oxygen species and counteract oxidative injury induced by ultraviolet radiation and thereby ameliorate skin aging.

In the present work, we prepared silver nanoparticles by laser ablation of pure silver plate in ethanol and then irradiated the silver nanoparticles using a 532 nm Q-switched Nd:YAG pulsed laser. Transmission electron microscopic images of the sample after irradiation clearly showed formation of big structures, such as microrods and microbelts in ethanol. The obtained microbelts had a width of about 0.166 μm and a length of 1.472 μm. The reason for the formation of such a big structure is the tendency of the nanoparticles to aggregate in ethanol before irradiation, which causes fusion of the nanoparticles.

Orthosiphon stamineus as medicinal plant is commonly used in Malaysia for treatment of hepatitis and jaundice; in this study, the ethanol extracts were applied to evaluate the hepatoprotective effects in a thioacetamide-induced hepatotoxic model in Sprague Dawley rats. Five groups of adult rats were arranged as follows: Group 1 (normal control group), Group 2 Thioacetamide (TAA) as positive control (hepatotoxic group), Group 3 Silymarin as a well-known standard drug (hepatoprotective group), and Groups 4 and 5 as high and low dose (treatment groups). After 60-day treatment, all rats were sacrificed. The hepatotoxic group showed a coarse granulation on the liver surface when compared to the smooth aspect observed on the liver surface of the other groups. Histopathological study confirmed the result; moreover, there was a significant increase in serum liver biochemical parameters (ALT, AST, ALP, and Bilirubin) and the level of liver Malondialdehyde (MDA), accompanied by a significant decrease in the level of total protein and Albumin in the TAA control group when compared with that of the normal group. The high-dose treatment group (200 mg/kg) significantly restored the elevated liver function enzymes near to normal. This study revealed that 200 mg/kg extracts of O. stamineus exerted a hepatoprotective effect.
