    Fluorescence in situ hybridization (FISH) is increasingly gaining importance in clinical diagnostics settings. Due to the ability of the technique to detect chromosomal abnormalities in samples with low cellularity or containing a mixed population of cells even at a single-cell level, it has become more popular in cancer research and diagnosis. Here, we describe the FISH technique for detection of PAX8-PPARγ translocation in follicular thyroid neoplasms, and the optimal protocol for the detection of this fusion gene using in archival formalin-fixed paraffin-embedded (FFPE) thyroid tissue sections.

    The aim of the study was to determine the frequency, size and location of apical and lateral foramina on anterior teeth. A total of 100 anterior teeth consisting of maxillary and mandibular incisors and canines were fixed in 10% formalin. Periodontal tissue remnants were mechanically removed and teeth were stained in 2% aqueous silver nitrate. The teeth were dried and examined using a Leica MZ 7.5 zoom stereomicroscope. The size of apical and lateral foramina and their distance from the anatomical apex of the tooth were measured directly using a calibrated eyepiece scale. Accessory foramina more than 1.8 mm from the apex were regarded as lateral foramina. Eighteen percent of teeth possessed more than one apical foramen. Seven teeth (three maxillary centrals, three maxillary canines, one mandibular lateral) had 11 lateral foramina each. The mean diameter of the lateral foramina was 0.14 mm (SD = 0.08) and their mean distance from the apex was 4.49 mm (SD = 2.63, range 1.9-10.5 mm). Multiple foramina were most common on maxillary canines and least common on maxillary laterals. The mean diameter of apical foramina for all teeth possessing a single foramen was 0.35 mm (SD = 0.10) and the mean apical foramen diameter for all teeth with multiple apical foramina was 0.22 mm (SD = 0.08). Most anterior teeth possess one apical foramen and no lateral foramina; about 20% possessed more than one apical foramen and about 10% possessed one or more lateral foramina. These findings should be considered when root-treating anterior teeth.

    This study was carried out to evaluate the anti-inflammatory effects of three concentrations of Labisia pumila (Blume) F. Vill-Naves aqueous leaf extract in rats. The effects of these extracts as anti-inflammatory agents were determined using two experiments namely formalin-induced paw licking and carrageenan-induced paw oedema test. The exposure of inflammation to various treatments resulted in significant differences between treatments in formalin-induced paw licking in rats experiment whereas in phase 2, 50 mg kg-1 of L. pumila extract showed the most significant inhibition of 82.12%, followed by 10 mg kg-1 with 76.00% and 25 mg kg-1 with 57.80%. Similarly, different treatments showed significant effects at p<0.05 in the carrageenan inducing paw oedema experiment. All treatments were able to suppress the oedema formation induced by carrageenan as compared with the control. It is evident that the anti-inflammtory effect of every concentration of L. pumila extract started as early as the first hour of carrageenan injection and showed the maximum inhibition during the fifth hour. Again, 50 mg kg-1 of L. pumila extract was found to be the best treatment that could reduce inflammation with highest inhibition of 64.59% followed by 25 mg kg-1 with 56.99% and 10 mg kg-1 with 5.55%. The result of this study has shown that these extracts of L. pumila can be effective for anti-inflammation purposes which supports and justifies traditional uses of this plant.

    Nonepithelial ovarian cancer (NEOC) is a rare cancer that is often misdiagnosed as other malignant tumors. Research on this cancer using fresh tissues is nearly impossible because of its limited number of samples within a limited time provided. The study is to identify potential genes and their molecular pathways related to NEOC using formalin-fixed paraffin embedded samples. Total RNA was extracted from eight archived NEOCs and seven normal ovaries. The RNA samples with RNA integrity number >2.0, purity >1.7 and cycle count value <28 cycles were hybridized to the Illumina Whole-Genome DASL assay (cDNA-mediated annealing, selection, extension, and ligation). We analyzed the results using the GeneSpring GX11.0 and FlexArray software to determine the differentially expressed genes. Microarray results were validated using an immunohistochemistry method. Statistical analysis identified 804 differentially expressed genes with 443 and 361 genes as overexpressed and underexpressed in cancer, respectively. Consistent findings were documented for the overexpression of eukaryotic translation elongation factor 1 alpha 1, E2F transcription factor 2, and fibroblast growth factor receptor 3, except for the down-regulated gene, early growth response 1 (EGR1). The immunopositivity staining for EGR1 was found in the majority of cancer tissues. This finding suggested that the mRNA level of a transcript did not always match with the protein expression in tissues. The current gene profile can be the platform for further exploration of the molecular mechanism of NEOC.

    Non-melanoma skin cancer (NMSC) is classified among the ten most frequent cancers in Malaysia. A common polymorphism at codon 72 of the p53 tumor suppressor gene and its influence on cancer risk has been studied for different types of cancer with mixed and inconsistent results with limited published data on the Malaysian population so far. In the present study, the frequency of p53 codon 72 polymorphism in 60 patients with NMSC was investigated from archival formalin-fixed paraffin-embedded (FFPE) tissue obtained from Hospital Universiti Kebangsaan Malaysia (HUKM). Additionally, random amplified polymorhic DNA -polymorphic chain reaction (RAPD-PCR) was employed for preliminary biomarker development. NMSC FFPE samples (70%) possess Arg/Arg, 20% with Pro/Pro and 10% with Arg/Pro. In total, there was no significant difference in the p53 codon 72 genotypes between histological types of NMSC, gender, race, tumor location and age group. However, there was an apparent age-associated increase in the Arg/Arg genotype but did not reach statistical significance (P=0.235). NMSC types and demographic characteristics did not influence genotype distribution. On the other hand, BCC and SCC distributions are influenced by age group, race and tumor location.

    The effect of morphology on the plasmonic sensing of the presence of formaldehyde in water by gold nanostructures has been investigated. The gold nanostructures with two different morphologies, namely spherical and rod, were prepared using a seed-mediated method. In typical results, it was found that the plasmonic properties of gold nanostructures were very sensitive to the presence of formaldehyde in their surrounding medium by showing the change in both the plasmonic peaks position and the intensity. Spherical nanoparticles (GNS), for example, indicated an increase in the sensitivity when the size was increased from 25 to 35 nm and dramatically decreased when the size was further increased. An m value, the ratio between plasmonic peak shift and refractive index change, as high as 36.5 nm/RIU (refractive index unit) was obtained so far. An expanded sensing mode to FD was obtained when gold nanostructures with nanorods morphology (GNR) were used because of the presence of two plasmonic modes for response probing. However, in the present study, effective plasmonic peak shift was not observed due to the intense plasmonic coupling of closely packed nanorod structures on the surface. Nevertheless, the present results at least provide a potential strategy for response enhancement via shape-effects. High performance plasmonic sensors could be obtained if controlled arrays of nanorods can be prepared on the surface.

    A cross-sectional study was conducted in February 2006 to determine the prevalence of Trichuris trichiura infection among Orang Asli (Aborigine) children at Pos Lenjang, Pahang. A total of 71 faecal samples were collected from the children (40 girls and 31 boys) aged between 1-12 years. The samples were examined for the presence of Trichuris trichiura ova using direct smear and formalin-ether concentration techniques. The result revealed that the overall prevalence of Trichuris trichiura infection was 43.7%. The infection was higher in males (51.6%) compared to females (37.5%), though not statistically significant (p > 0.05). According to age group, the school-aged children had higher prevalence of infection (56.8%) than preschool children (29.4%) (p < 0.05). Low socioeconomic status, large family size, poor environmental sanitation and poor personal hygiene are possible contributing factors that increase the prevalence of infection among the Orang Asli children at Pos Lenjang. In 31 samples positive for Trichuris trichiura, a detection rate of 100% was obtained using formalin-ether concentration, compared to 25.8% with direct smear technique. Thus, it is recommended that both techniques be performed in routine faecal examination for a more accurate diagnosis.

    This study has been conducted in a new constructed building of NIOSH Malaysia located at Bandar Baru Bangi, Selangor. The goal of the case study is focusing on the level of Indoor Air Contaminants (IAC) including chemical contaminants within three consequent stages which are before furniture install, after furniture install and during one month occupancy. This study was divided the sampling area into two main facilities which are training and office setting. The contaminants has been measured consist of sixparameters such as Carbon Dioxide (CO2), Carbon Monoxide (CO), Total Volatile Organic Compounds (TVOC), Formaldehyde, Respirable Particulates (PM10) and Ozone. The result of Carbon Monoxide (CO), Total Volatile Organic Compound (TVOC), Respirable Particulates (PM10) and Ozone show an increasing trend across the three sampling stages. The Formaldehyde show an increasing trend in the first and second stages but were reduced significantly the last stage of sampling. These finding indicates that furniture and fittings installed might be a potential sources of indoor air contaminants. The management should be aware to their indoor air status to protect the occupant from the risk of unwanted exposure especially during the early stage of building occupancy.

    entamoeba histolytica dan/atau entamoeba dispar merupakan protozoa usus yang mempunyai prevalens infeksi yang tinggi dalam kalangan masyarakat pedalaman terutamanya masyarakat Orang Asli. Ia tersebar secara meluas di kawasan tropika dan subtropika serta di negara membangun berbanding negara maju. Sebanyak 111 sampel feses kanak-kanak Orang Asli daripada suku kaum Jahai telah diterima dan disaring untuk entamoeba histolytica dan/atau entamoeba dispar menggunakan kaedah apusan langsung yang memberi hasil positif terhadap 43 sampel atau 38.7%. Oleh sebab amaun sampel yang diterima adalah sedikit, hanya 66 sampel feses sahaja yang dapat diperiksa menggunakan tiga jenis teknik diagnostik berbeza iaitu apusan langsung, kepekatan formalin-eter dan pewarnaan trikrom. Hasil kajian mendapati, prevalens yang tinggi bagi infeksi entamoeba histolytica dan/atau entamoeba dispar iaitu 50% menggunakan
    ketiga-tiga teknik diagnosis. Prevalens infeksi yang tinggi juga turut ditunjukkan pada kanak-kanak perempuan iaitu 62.5% berbanding kanak-kanak lelaki 30.8% (p<0.05). Selain itu, daripada segi umur, kanak-kanak yang berumur 7-9 tahun adalah lebih terdedah kepada infeksi entamoeba histolytica dan/atau entamoeba dispar dengan prevalens 60.7% (p>0.05). Teknik pewarnaan trikrom menunjukkan pengesanan 100% ke atas infeksi entamoeba histolytica dan/atau entamoeba dispar, diikuti teknik kepekatan formalin-eter 78.8% dan apusan langsung 72.7%. Prevalens infeksi entamoeba histolytica dan/atau entamoeba dispar yang tinggi dalam kalangan kanak-kanak Orang Asli di Pos Sungai Rual ini berhubungkait dengan pelbagai faktor iaitu status sosioekonomi yang rendah, kekurangan pengetahuan tentang penjagaan kesihatan serta kebersihan diri yang rendah. Peningkatan prevalens infeksi dalam kajian ini juga menunjukkan pentingnya penggunaan teknik diagnostik yang lebih berkesan dalam pemeriksaan rutin bagi mendapatkan hasil diagnosis yang lebih tepat.

    Muntingia calabura L. (family Elaeocarpaceae) has been traditionally used to relieve various pain-related ailments. The present study aimed to determine the antinociceptive activity of methanol extract of M. calabura leaves (MEMC) and to elucidate the possible mechanism of antinociception involved. The in vivo chemicals (acetic acid-induced abdominal constriction and formalin-, capsaicin-, glutamate-, serotonin-induced paw licking test) and thermal (hot plate test) models of nociception were used to evaluate the extract antinociceptive activity. The extract (100, 250, and 500 mg/kg) was administered orally 60 min prior to subjection to the respective test. The results obtained demonstrated that MEMC produced significant (P < 0.05) antinociceptive response in all the chemical- and thermal-induced nociception models, which was reversed after pretreatment with 5 mg/kg naloxone, a non-selective opioid antagonist. Furthermore, pretreatment with L-arginine (a nitric oxide (NO) donor), N(G)-nitro-L-arginine methyl esters (L-NAME; an inhibitor of NO synthase (NOS)), methylene blue (MB; an inhibitor of cyclic-guanosine monophosphate (cGMP) pathway), or their combination also caused significant (P < 0.05) change in the intensity of the MEMC antinociception. In conclusion, the MEMC antinociceptive activity involves activation of the peripheral and central mechanisms, and modulation via, partly, the opioid receptors and NO/cGMP pathway.

    We present results from the OP3 campaign in Sabah during 2008 that allow us to study the impact of local emission changes over Borneo on atmospheric composition at the regional and wider scale. OP3 constituent data provide an important constraint on model performance. Treatment of boundary layer processes is highlighted as an important area of model uncertainty. Model studies of land-use change confirm earlier work, indicating that further changes to intensive oil palm agriculture in South East Asia, and the tropics in general, could have important impacts on air quality, with the biggest factor being the concomitant changes in NO(x) emissions. With the model scenarios used here, local increases in ozone of around 50 per cent could occur. We also report measurements of short-lived brominated compounds around Sabah suggesting that oceanic (and, especially, coastal) emission sources dominate locally. The concentration of bromine in short-lived halocarbons measured at the surface during OP3 amounted to about 7 ppt, setting an upper limit on the amount of these species that can reach the lower stratosphere.

    This review summarizes the results of epidemiological studies focusing on the detrimental effects of home environmental factors on asthma morbidity in adults. We reviewed the literature on indoor air quality (IAQ), physical and sociodemographic factors, and asthma morbidity in homes, and identified commonly reported asthma, allergic, and respiratory symptoms involving the home environment. Reported IAQ and asthma morbidity data strongly indicated positive associations between indoor air pollution and adverse health effects in most studies. Indoor factors most consistently associated with asthma and asthma-related symptoms in adults included fuel combustion, mold growth, and environmental tobacco smoke. Environmental exposure may increase an adult's risk of developing asthma and also may increase the risk of asthma exacerbations. Evaluation of present IAQ levels, exposure characteristics, and the role of exposure to these factors in relation to asthma morbidity is important for improving our understanding, identifying the burden, and for developing and implementing interventions aimed at reducing asthma morbidity.

    In Malaysia, maggot debridement therapy (MDT) utilizes maggots of Lucilia cuprina (Wiedemann) to debride necrotic tissue from wound surface, reduce bacterial infection and therefore, enhance wound healing process. To evaluate the sterility of the sterile maggots produced after sterilization process before delivering onto patient wounds. Sterility of sterile maggots is crucial in ensuring the safe usage of MDT and patient's health. Eggs of L. cuprina collected from a laboratory colony were divided into treated group (sterilized) and control group (non-sterilized). Treated group underwent sterilization while eggs from control group were allowed to hatch without sterilization. Sodium hypochlorite and formaldehyde were the main disinfectants used in this sterilization process. Scanning electron microscope (SEM) was used to examine and ascertain the sterility of sterile maggots. SEM results showed that all sterilized L. cuprina eggs and maggots achieved sterility and all were cleared from bacterial contamination. In contrast, all non-sterilized eggs and maggots were found to be colonized by microorganisms. Sterilization method employed to sterilize eggs and maggots used in Malaysia MDT was proven successful and MDT is safe to be used as wound management tools.

    Painful neuropathy, a common complication of diabetes mellitus is characterized by allodynia and hyperalgesia. Recent studies emphasized on the role of non-neuronal cells, particularly microglia in the development of neuronal hypersensitivity. The purpose of the present study is to evaluate the effect of minocyline, a selective inhibitor of microglial activation to define the role of neuroimmune activation in experimental diabetic neuropathy. Cold allodynia and thermal and chemical hyperalgesia were assessed and the markers of inflammation and oxidative and nitrosative stress were estimated in streptozotocin-induced diabetic rats. Chronic administration of minocycline (40 and 80 mg/kg, i.p.) for 2 weeks started 2 weeks after diabetes induction attenuated the development of diabetic neuropathy as compared to diabetic control animals. In addition, minocyline treatment reduced the levels of interleukin-1β and tumor necrosis factor-α, lipid peroxidation, nitrite and also improved antioxidant defense in spinal cords of diabetic rats as compared to diabetic control animals. In contrast, minocycline (80 mg/kg, per se) had no effect on any of these behavioral and biochemical parameters assessed in age-matched control animals. The results of the present study strongly suggest that activated microglia are involved in the development of experimental diabetic neuropathy and minocycline exerted its effect probably by inhibition of neuroimmune activation of microglia. In addition, the beneficial effects of minocycline are partly mediated by its anti-inflammatory effect by reducing the levels of proinflammatory cytokines and in part by modulating oxidative and nitrosative stress in the spinal cord that might be involved in attenuating the development of behavioral hypersensitivity in diabetic rats.

    Downstream Regulatory Element Antagonist Modulator (DREAM) protein modulates pain by regulating prodynorphin gene transcription. Therefore, we investigate the changes of mRNA and DREAM protein in relation to the mRNA and prodynorphin protein expression on the ipsilateral side of the rat spinal cord after formalin injection (acute pain model). DREAM like immunoreactivity (DLI) was not significantly different between C and F groups. However, we detected the upregulation of mean relative DREAM protein level in the nuclear but not in the cytoplasmic extract in the F group. These effects were consistent with the upregulation of the relative DREAM mRNA level. Prodynorphin like immunoreactivity (PLI) expression increased but the relative prodynorphin mRNA level remained unchanged. In conclusion, we suggest that upregulation of DREAM mRNA and protein expression in the nuclear compartment probably has functional consequences other than just the repression of prodynorphin gene. It is likely that these mechanisms are important in the modulation of pain.

    Introduction: The aim of this study was to determine the effects of gamat extract on pain behaviour and Fos like immunoreactivity (FLI) expression in the ventral posterolateral thalamus using the acute pain model. Materials & Methods: Fourteen Sprague-Dawley male rats (220-300 gram) were given intraplantar injection of 0.05ml formalin (1%) followed by intraperitoneal administration of either 4 mg/kg gamat extracts (Holothuria spp.) or saline (control). Behavioural changes were observed and rats were sacrificed 2 hours post-formalin injection. Immunohistochemistry testing was done on the brain sections.     A preliminary study has been conducted in a new constructed 8 stories building (2 level of Hostels facility, 3 level of Training Room, 2 Level of Offices and 1 level of Exhibition Halls and Rooms) in Bandar Baru Bangi, Selangor. The Hostels facility is a floor tile and furnished with build in locker and use split air conditioning system while the Training Room and Exhibition Rooms used floor tile.
