    Therapeutic application of metallic nanoparticles such as gold nanoparticles have been extensively investigated and intriguing finding have been reported. Superparamagnetic iron oxide nanoparticles (SPION) could also potentially have therapeutic properties that can be exploited to enhance radiotherapy outcome. In this study, investigations on the dose enhancement effects inflicted by SPIONs under irradiation with megavoltage photon beam radiotherapy were conducted. T24 human bladder cancer cell lines were pretreated with 1 mMol/L of SPION and irradiated with 6 MV and 10 MV photon beam at different doses.The non-treated cells irradiation was used as a control. Clonogenic assay was performed to determine the cell survival. Linear quadratic (LQ) model are used as fitting curve and does enhancement factors (DEF) were extrapolated from the curves. The cytotoxicity indicated cell growth normally after 72 hours and no long term cytotoxicity effects of SPIONs towards the cells were observed. The dose enhancement effects were observed for both 6 MV and 10 MV photon beam with DEF obtained 1.71 and 2.50, respectively. This reduction of cell colonies growth could be resulted from the interaction that induced free radical and reactive oxygen species (ROS) by megavoltage photon beams. The SPIONs were therefore act as multifunction nanoparticle both in diagnostic agent and radiotherapy as radiation dose enhancer, thus clearly qualified as future theranostic agents.

    The purpose of the study was to determine the effect of out-of-field photon beams radiotherapy to the cancer cell survival. In this study, HeLa and T24 cancer cells were irradiated with 6 MV and 10 MV photon beams in two different conditions, one with intercellular communication with the in-field cell and one without the communication. Cells survival was determined by clonogenic assay. In the presence of intercellular communication, the cell death was increased which indicate the presence of radiation induced bystander effects (RIBE). The effects were also dependent on the cell types and photon energy where the HeLa cells exhibit less survival compares to T24 cells and the effects were prominent at higher photon energy. This study demonstrates that the out-of-field cells in conjunction with RIBE plays important roles in the cells response towards megavoltage photon beam radiation therapy.

    The transwell migration assay is commonly used for assessing cell migration. It involves the enumeration of cells that
    have migrated across a pore-containing membrane. We describe a randomised approach to quantifying migrated
    cells and compare it to a conventional full cell count. We used ATP as a chemoattractant and automatic cell quantification performed on all fields (Full count; FC) or 10 randomly selected fields (Randomised count; RC). The two
    methods were compared by evaluating standard deviations (SD), coefficient of variation (CV) and using the Bland-Altman analysis. The dispersion of data is higher with the RC approach (3.77-6.66% CV for control; 3.89-4.48% CV
    for ATP-treated wells) compared to FC (0.27-0.46% CV for control; 0.05-0.09% CV for ATP-treated wells), but are
    acceptable considering that the number of migrated cells are in the thousands. Both methods verified that an ATP
    migration assay for BV2 microglia was established, demonstrating that the RC approach is reliable and comparable
    to a full count.

    Introduction: Virgin coconut oil (VCO) is known for its health and therapeutic benefits. However, the immunomodulatory effects of VCO have not been extensively investigated. Objective: The present study was devoted to examining the effects of VCO on cyclophosphamide (CY)-induced toxicity of lymphoid tissues. Methods: Thirty healthy male Wistar rats were sorted into 5 groups of 6 animals. The first control (NC) group was given distilled water via gavage at 5 ml/kg once daily. The second (CY) group received CY orally at 10 mg/kg/day for 4 weeks. Rats in the other three groups (CV5, CV10, and CV15) were given 10 mg/kg/day CY for 4 weeks, 5 m/kg/day, 10 ml/kg/day and 15 ml/kg/day VCO for 6 weeks, respectively. Rats were sacrificed at the end of 6th week; blood sample from the animals was collected for full blood count and biochemical analysis. The thymus and spleen of each animal was processed for histological examination. Results: The thymus and spleen showed marked reduction in lymphoid cellularity following daily administration of CY. The thymus also showed a marked reduction in the size of the medulla, and the white pulp areas of spleen had reduction in the follicle number and size. Supplementation with 10 ml/kg and 15 ml/kg VCO showed evidence of restoration of both the thymus and splenic lymphoid architecture. The total white cell counts, absolute lymphocyte counts and plasma globulin levels of the VCO groups were significantly increased compared to CY group. Conclusion: VCO displayed potential protective effects on CYinduced histological changes in lymphoid tissues.

    ABSTRACT
    Clinacanthus nutans (C. nutans) leaf extracts have been widely used by cancer patients in Malaysia and local practice claims a cure to cancer. There were several studies done to determine the cytotoxicity potency of C. nutans extracts on various types of cells. However, there is still lacking on the knowledge regarding the combination effect of C. nutans with anticancer drugs. Thus, the study was carried out to determine the cytotoxicity potency of C. nutans extracts and paclitaxel (PTX) alone and, in combination on MDA-MB-231 cells. The cells were treated with 100% ethanol extract of C. nutans (CNE) and water extract of C. nutans (CNA), PTX and combination of both extracts and PTX for 72 hours and the cytotoxic activity was determined using SRB assay. Result showed that CNE had little cytotoxic activity, whereas CNA showed no cytotoxic activity on MDA-MB-231 cells. For combination treatment of C. nutans extracts and PTX, only CNE showed significant enhanced PTX-induced cytotoxicity (p < 0.05), meanwhile CNA inhibited PTX-induced cytotoxicity significantly (p < 0.05). As a conclusion, CNE was able to increase PTX potency to inhibit the viability of MDA-MB-231 cells.

    Purple urine bag syndrome (PUBS) is a rare presentation of urinary tract infections (UTIs). It is commonly seen in constipated patients. There is a deep purple discoloration of contents of urine bag due to presence of indigo and indirubin pigments which are metabolites of tryptophan. We would like to describe an interesting case of purple urine bag syndrome of 88-year-old woman who presented with catheter-related urinary tract infection. She had low-grade fever and suprapubic discomfort for three days duration. She had increased white cell count and C-reactive peptide (CRP). Urinalysis showed protein 2+, nitrite and leucocyte esterase positive. Urine culture grew Escherichia coli and Klebsiella pneumoniae. She was treated with oral cefuroxime and recovered. This case report may be the first case of PUBS reported in this region.

    The aim of this study to determine the numbers of CD34+ cells and total nucleated cell (TNC) in umbilical cord blood (UCB)
    collected from pregnant mothers with gestational diabetes mellitus (GDM) and preeclampsia (PE), following statistical
    analysis of both maternal and perinatal factors which affect UCB parameters. Most of studies explored the influence of
    obstetric factors on the number of UCB cell collection and only a few looked at the effects on UCB haematopoietic stem
    cell (UCB-HSC) of common disorders complicating pregnancy. A total of 112 UCB samples (32 PE, 42 GDM and 38 nondiseased) were collected. CD34+ cell and NC count were enumerated using FACS Calibur. The TNC and CD34+ cells were
    significantly reduced in both PE and GDM groups as compared to the control group. The PE group shows significantly
    lower birth weight and higher BP which led to a lower UCB volume and CD34+ count. Gestational age shows significant
    correlation with nucleated cell count (NCC) and TNC. GDM group shows significantly lower systolic BP, NCC and TNC count,
    including low placental weight and birth weight. Conclusively, some obstetrics factors have significant influences to the
    numbers and quality of UCB-HSC in both PE and GDM groups, which could guide in the selection criteria for CB banking.

    Persistent neutrophilic meningitis is an atypical form of chronic meningitis defined as clinical meningitis with a cerebrospinal fluid (CSF) analysis suggestive of neutrophilic pleocytosis that persists for more than one week despite giving empirical antimicrobial therapy. The predominant presentation of tubercular meningitis is with CSF lymphocytic pleocytosis and even if early phase of the disease presents with neutrophilic pleocytosis, no persistence is usually observed for more than one week. We hereby report the case of an immunocompetent man aged 25 years with tubercular meningitis manifesting as persistent neutrophilic meningitis which is very rare as an entity in tubercular meningitis (TBM). Patient was started on anti-tubercular therapy based on the granulomas in magnetic resonance imaging (MRI) of the brain after which there was a reduction in total cell count in CSF but persisting neutrophilic predominance. Patient improved clinically and was given a full course of anti-tubercular treatment (ATT) thereafter.

    Conventional mammalian models of neurodegeneration are often limited by futile axonogenesis with minimal functional recuperation of severed neurons. The emergence of zebrafish, a non-mammalian model with excellent neuroregenerative properties, may address these limitations. This study aimed to establish an adult zebrafish-based, neurotoxin-induced Parkinson's disease (PD) model and subsequently validate the regenerative capability of dopaminergic neurons (DpN). The DpN of adult male zebrafish (Danio rerio) were lesioned by microinjecting 6-hydroxydopamine (6-OHDA) neurotoxin (6.25, 12.5, 18.75, 25, 37.5, 50 and 100 mg/kg) into the ventral diencephalon (Dn). This was facilitated by an optimised protocol that utilised 1,1'-dioctadecyl-3,3,3',3'-tetramethyl-indocarbocyanineperchlorate (DiI) dye to precisely identify the injection site. Immunostaining was utilised to identify the number of tyrosine hydroxylase immunoreactive (TH-ir) DpN in brain regions of interest (i.e. olfactory bulb, telencephalon, preoptic area, posterior tuberculum and hypothalamus). Open tank video recordings were performed for locomotor studies. The Dn was accessed by setting the injection angle of the microinjection capillary to 60° and injection depth to 1200 μm (from the exposed brain surface). 6-OHDA (25 mg/kg) successfully ablated >85% of the Dn DpN (preoptic area, posterior tuberculum and hypothalamus) whilst maintaining a 100% survival.     AIM: This study was conducted to analyze the changes in blood profile of goats inoculated with Corynebacterium pseudotuberculosis and its immunogen mycolic acid (MA) extract.

    MATERIALS AND METHODS: A total of 12 clinically healthy crossbred Boer female goats were divided into three groups; A, B and C (4 goats each per group). Group A was inoculated with 2 ml sterile phosphate buffered saline via intradermal route as the negative control group whilst Group B was inoculated with 2 ml of MA extract (1 g/ml) intradermally and Group C was then inoculated with 2 ml (1×10(9)) colony forming unit of active C. pseudotuberculosis intradermally. Blood sample was collected aseptically from the jugular vein periodically for complete blood count (CBC) analysis throughout the experimental period (3 months).

    RESULT: A significant decrease (p<0.05) was observed in red blood cells, hemoglobin (Hb), packed cell volume, mean corpuscular volume and mean corpuscular Hb concentration in Groups B and C as compared to the control while WBCs, neutrophil, lymphocyte and basophil showed a significant increase (p<0.05) as compared to the control.

    CONCLUSION: The inoculation of C. pseudotuberculosis and MA resulted in a significant change in the CBC, thereby, indicating that MA has a role in caseous lymphadenitis pathogenesis.

    The growth of several biological tissues is known to be controlled in part by local geometrical features, such as the curvature of the tissue interface. This control leads to changes in tissue shape that in turn can affect the tissue's evolution. Understanding the cellular basis of this control is highly significant for bioscaffold tissue engineering, the evolution of bone microarchitecture, wound healing, and tumor growth. Although previous models have proposed geometrical relationships between tissue growth and curvature, the role of cell density and cell vigor remains poorly understood. We propose a cell-based mathematical model of tissue growth to investigate the systematic influence of curvature on the collective crowding or spreading of tissue-synthesizing cells induced by changes in local tissue surface area during the motion of the interface. Depending on the strength of diffusive damping, the model exhibits complex growth patterns such as undulating motion, efficient smoothing of irregularities, and the generation of cusps. We compare this model with in vitro experiments of tissue deposition in bioscaffolds of different geometries. By including the depletion of active cells, the model is able to capture both smoothing of initial substrate geometry and tissue deposition slowdown as observed experimentally.

    The location model proposed in the past is a predictive discriminant rule that can classify new observations into one
    of two predefined groups based on mixtures of continuous and categorical variables. The ability of location model to
    discriminate new observation correctly is highly dependent on the number of multinomial cells created by the number
    of categorical variables. This study conducts a preliminary investigation to show the location model that uses maximum
    likelihood estimation has high misclassification rate up to 45% on average in dealing with more than six categorical
    variables for all 36 data tested. Such model indicated highly incorrect prediction as this model performed badly for
    large categorical variables even with large sample size. To alleviate the high rate of misclassification, a new strategy
    is embedded in the discriminant rule by introducing nonlinear principal component analysis (NPCA) into the classical
    location model (cLM), mainly to handle the large number of categorical variables. This new strategy is investigated
    on some simulation and real datasets through the estimation of misclassification rate using leave-one-out method. The
    results from numerical investigations manifest the feasibility of the proposed model as the misclassification rate is
    dramatically decreased compared to the cLM for all 18 different data settings. A practical application using real dataset
    demonstrates a significant improvement and obtains comparable result among the best methods that are compared. The
    overall findings reveal that the proposed model extended the applicability range of the location model as previously it
    was limited to only six categorical variables to achieve acceptable performance. This study proved that the proposed
    model with new discrimination procedure can be used as an alternative to the problems of mixed variables classification,
    primarily when facing with large categorical variables.

    Cancer is one of the global health problems that has a detrimental effect
    to a person's life. However, chemotherapeutic agents success are subject to the side
    effects due to lack of specificity in the drug delivery system to cancer cells and an
    increase risk of systemic toxicity to the normal cells. Polyamine transport system (PTS)
    is one of the potential pathways for transporting anticancer agent into specific cancer
    cells. This is due to the upregulation of PTS in cancer cells compared to normal cells for
    the proliferation activity. The aim of this study was to investigate the cytotoxicity effect
    of putrescine-sulphur analogues type 1 (PSA-1) and type 2 (PSA-2) on human lung
    adenocarcinoma cells (A549), human colorectal adenocarcinoma cells (HCT-8) and
    human breast adenocarcinoma cell (MCF-7).

    Liver perfusion has been the standard method to digest and isolate liver
    cells including liver sinusoidal endothelial cells (LSEC). Poor cannulating skills through
    portal vein results in a waste of animal resource. Familiarization of both liver perfusion
    technique and adhering strictly to aseptic technique during cell handling ensure high
    cell yield, minimum morphology disruption and cell contamination. We aimed to present

    This study describes the adaptations of diatoms, Cylindrotheca fusiformis and other marine diatoms, in a new formulated enriched medium Tris-phosphate seawater (TP-SW). The medium was designed to maintain long-term cultures of wide-range marine diatoms in laboratory that produces high biomass of cultures. The diatoms were adapted and cultivated in the medium for 15 days and the number of cells was recorded daily. It was found that the number of cells declined after two weeks indicating death phase of the cells. This indicates that the TP-SW medium has supported the growth of diatoms during the period and can be used to cultivate diatoms in vitro. Studies on the TP-SW medium must be done to obtain optimal medium that can provide not only a conducive environment for the survival of diatoms but also high biomass production.

    Lung cancer is the most common cancer worldwide, accounting for 1.8 million new cases and 1.6 million deaths in 2012. Non-small cell lung cancer (NSCLC), which is one of two types of lung cancer, accounts for 85-90% of all lung cancers. Despite advances in therapy, lung cancer still remains a leading cause of death. Cancer relapse and dissemination after treatment indicates the existence of a niche of cancer cells that are not fully eradicated by current therapies. These chemoresistant populations of cancer cells are called cancer stem cells (CSCs) because they possess the self-renewal and differentiation capabilities similar to those of normal stem cells. Targeting the niche of CSCs in combination with chemotherapy might provide a promising strategy to eradicate these cells. Thus, understanding the characteristics of CSCs has become a focus of studies of NSCLC therapies.

    The process of intercellular communication among bacteria, termed quorum sensing (QS), is mediated by small diffusible molecules known as the autoinducers. QS allows the population to react to the change of cell density in unison, in processes such as biofilm formation, plasmid conjugation, virulence, motility and root nodulation. In Gram-negative proteobacteria, N-acyl homoserine lactone (AHL) is the common "language" to coordinate gene expression. This signaling molecule is usually synthesized by LuxI-type proteins. We have previously discovered that a rare bacterium, Cedecea neteri, exhibits AHL-type QS activity. With information generated from genome sequencing, we have identified the luxIR gene pair responsible for AHL-type QS and named it cneIR. In this study, we have cloned and expressed the 636 bp luxI homolog in an Escherichia coli host for further characterization. Our findings show that E. coli harboring cneI produced the same AHL profile as the wild type C. neteri, with the synthesis of AHL known as N-butyryl-homoserine lactone. This 25 kDa LuxI homolog shares high similarity with other AHL synthases from closely related species. This work is the first documentation of molecular cloning and characterization of luxI homolog from C. neteri.

    PURPOSE: To study the effects of intracameral phenylephrine 1.5% on corneal endothelial cell loss and morphological changes in patients who had uneventful phacoemulsification surgery.

    DESIGN: A double-blind randomized controlled trial.

    METHODS: This study comprised 295 patients who were randomized into the intracameral (ICM) mydriatic group or topical mydriatic group. Central corneal endothelial cell density (ECD), coefficient of variation (CV), and percentage of hexagonal cells were measured preoperatively and postoperatively at 1 week, 6 weeks, and 3 months with specular microscope.

    RESULTS: There was no significant difference in endothelial cell density and endothelial cell loss between the topical and ICM mydriatic groups. At 3 months, the mean endothelial cell density in the ICM group was 2129.76 ± 423.53 cells/mm2 and 2100.54 ± 393.00 cells/mm2 in the topical group (P = 0.539). The endothelial cell loss was 18.60 ± 12.79% in the IC M group and 19.44 ± 11.24% in the topical group (P = 0.550). No significant difference was seen in the percentage of hexagonal cells and coefficient of variation of patients between the 2 groups.

    CONCLUSIONS: Intracameral phenylephrine was not associated with increased risk of postoperative endothelial cell loss or morphological changes. It can be safely injected into the anterior chamber for pupil dilatation before phacoemulsification cataract surgery.

      The effect of physical shearing on the attachment of six Pseudomonas aeruginosa strains and six Burkholderia cepacia strains to glass, stainless steel, polystyrene and Teflon® was determined. A significant (p < 0.05) decrease in hydrophobicity was apparent for all P. aeruginosa strains (17-36%) and B. cepacia, MS 5 (20%) after shearing. A significant (p < 0.05) decrease in attachment of some P. aeruginosa (0.2-0.5 log CFU/cm2) and B. cepacia (0.2-0.4 log CFU/cm2) strains to some surface types was apparent after shearing. Significant (p < 0.05) correlation was observed for both numbers of flagellated cells and hydrophobicity against attachment to glass, stainless steel and polystyrene for P. aeruginosa while only hydrophobicity showed significant correlation against the same surfaces for B. cepacia. Scanning electron microscopy and protein analysis showed that shearing removed surface proteins from the cells and may have led to the observed changes in hydrophobicity and attachment to abiotic surfaces.

    OBJECTIVES: The aim of this study was to determine the changes in cell density and morphology of selected cells of the ageing human dental pulp.

    BACKGROUND: Changes in cell density and morphology of dental pulp cells over time may affect their capability to respond to tooth injury.

    MATERIALS AND METHODS: One hundred thirty-one extracted teeth were obtained from individuals between the ages of 6 and 80 years. The apical 1/3 of the root region was removed from all teeth prior to routine processing for producing histological slides. The histology slides were used to study the changes in cell density and morphology of selected pulp cells; odontoblasts, subodontoblasts and fibroblasts in the crown and root regions of the dental pulp. Student's t-test and one-way anova were used for statistical analyses.

    RESULTS: In all age groups, the cell density for all types of cells was found to be higher in the crown than in the root (p cell density was found to decrease with age in both the crown and root regions. However, it was noted that the reduction of coronal odontoblasts occurred later in life (40-49 years) when compared to that of subodontoblasts or fibroblasts (30-39 years).

    CONCLUSIONS: The density of the coronal pulp cells reduces and these cells undergo morphological changes with ageing of individuals and this may affect the pulp's ability to resist tooth injury.
