    Three short sediment cores from inner continental shelf of the southern South China Sea (5-50 km) off Terengganu were analyzed for lipid contents (i.e., homologous aliphatic compounds and sterols) using gas chromatography-mass spectrometry. The concentrations of the total aliphatic hydrocarbons (TAHs) ranged from 0.152 to 6.91 μg/g dry weight. The n-alkane distribution was from nC₁₃ to nC₃₆, with a carbon preference index (CPI₁₃₋₃₅) from 1.08 to 4.28 and a carbon number maximum (Cmax), depending on a sample, at 31 or 18. In addition, a strong odd-to-even carbon number predominance was observed in nC₂₅-nC₃₅ range. The distribution of the n-alkanoic acids and n-alkanols in all samples exhibited an even-to-odd carbon number predominance and ranged from C₁₀ to C₂₆ and from C₁₂ to C₃₄, respectively. The n-alkanols were dominated by the long-chain homologs with Cmax at 22; on the other hand, the n-alkanoic acid distributions showed a predominance of short-chain homologs with a Cmax at 16. The total sterol concentrations ranged from 0.41 to 3.57 μg/g dry weight. Cholesterol was most abundant at the offshore stations, whereas sitosterol was dominant at near-shore station. Pentacyclic triterpenoids such as friedelin and taraxerol α- and β-amyrins, which are known biomarkers for higher plants, were detected at all stations with a dilution trend offshore. In conclusion, the marine sediments off southern Terengganu can still be considered uncontaminated, where the compound sources are biogenic from terrestrial plants superimposed with a marine productivity input.

    Two different procedures for the methylation of fatty acids (FAs) and trans fatty acids (TFAs) in food fats were compared using gas chromatography (GC-FID). The base-catalyzed followed by an acid-catalyzed method (KOCH3/HCl) and the base-catalyzed followed by (trimethylsilyl)diazomethane (TMS-DM) method were used to prepare FA methyl esters (FAMEs) from lipids extracted from food products. In general, both methods were suitable for the determination of cis/trans FAs. The correlation coefficients (r) between the methods were relatively small (ranging from 0.86 to 0.99) and had a high level of agreement for the most abundant FAs. The significant differences (P = 0.05) can be observed for unsaturated FAs (UFAs), specifically for TFAs. The results from the KOCH3/HCl method showed the lowest recovery values (%R) and higher variation (from 84% to 112%), especially for UFAs. The TMS-DM method had higher R values, less variation (from 90% to 106%), and more balance between variation and %RSD values in intraday and interday measurements (less than 4% and 6%, resp.) than the KOCH3/HCl method, except for C12:0, C14:0, and C18:0. Nevertheless, the KOCH3/HCl method required shorter time and was less expensive than the TMS-DM method which is more convenient for an accurate and thorough analysis of rich cis/trans UFA samples.

    BACKGROUND: Sugars and triglycerides are common carbon sources for microorganisms. Nonetheless, a systematic comparative interpretation of metabolic changes upon vegetable oil or glucose as sole carbon source is still lacking. Selected fungi that can grow in acidic mineral salt media (MSM) with vegetable oil had been identified recently. Hence, this study aimed to investigate the overall metabolite changes of an omnipotent fungus and to reveal changes at central carbon metabolism corresponding to both carbon sources.

    RESULTS: Targeted and non-targeted metabolomics for both polar and semi-polar metabolites of Phialemonium curvatum AWO2 (DSM 23903) cultivated in MSM with palm oil (MSM-P) or glucose (MSM-G) as carbon sources were obtained. Targeted metabolomics on central carbon metabolism of tricarboxylic acid (TCA) cycle and glyoxylate cycle were analysed using LC-MS/MS-TripleQ and GC-MS, while untargeted metabolite profiling was performed using LC-MS/MS-QTOF followed by multivariate analysis. Targeted metabolomics analysis showed that glyoxylate pathway and TCA cycle were recruited at central carbon metabolism for triglyceride and glucose catabolism, respectively. Significant differences in organic acids concentration of about 4- to 8-fold were observed for citric acid, succinic acid, malic acid, and oxaloacetic acid. Correlation of organic acids concentration and key enzymes involved in the central carbon metabolism was further determined by enzymatic assays. On the other hand, the untargeted profiling revealed seven metabolites undergoing significant changes between MSM-P and MSM-G cultures.

    CONCLUSIONS: Overall, this study has provided insights on the understanding on the effect of triglycerides and sugar as carbon source in fungi global metabolic pathway, which might become important for future optimization of carbon flux engineering in fungi to improve organic acids production when vegetable oil is applied as the sole carbon source.

    Diabetes mellitus in Asia accounts for more than half of the global prevalence. There is a high prevalence of cardiovascular disease (CVD) in the region among people with type 2 diabetes mellitus (T2DM) and it is often associated with multiple risk factors including hypertension, renal disease and obesity. The early onset of T2DM and the eventual long disease duration portends an increasing proportion of the population to premature CVD. In addition to lowering blood glucose, sodium-glucose co-transporter-2 (SGLT-2) inhibitors exert favourable effects on multiple risk factors (including blood pressure, body weight and renal function) and provide an opportunity to reduce the risk of CVD in patients with T2DM. In this article, we consolidated the existing literature on SGLT-2 inhibitor use in Asian patients with T2DM and established contemporary guidance for clinicians. We extensively reviewed recommendations from international and regional guidelines, published data from clinical trials in the Asian population (dapagliflozin, canagliflozin, empagliflozin, ipragliflozin, luseogliflozin and tofogliflozin), CVD outcomes trials (EMPAREG-OUTCOME, CANVAS and DECLARE-TIMI 58) and real-world evidence studies (CVD-REAL, EASEL, CVD-REAL 2 and OBSERVE-4D). A series of clinical recommendations on the use of SGLT-2 inhibitors in Asian patients with T2DM was deliberated among experts with multiple rounds of review and voting. Based on the available evidence, we conclude that SGLT-2 inhibitors represent an evidence-based therapeutic option for the primary prevention of heart failure hospitalization and secondary prevention of CVD in patients with T2DM, and should be considered early on in the treatment algorithm for patients with multiple risk factors, or those with established CVD.

    The effects of γ-aminobutyric (GABA) on enzymatic browning, storage quality, membrane and reactive oxygen species (ROS) metabolism in fresh-cut stem lettuce were investigated. The results illustrated that GABA treatment delayed browning degree, polyphenol oxidase (PPO) activity and the expression of LsPPO. Meanwhile, higher chlorophyll and ascorbic acid contents were exhibited in GABA-treated stem lettuce, as well as the slower microbial propagation. Further investigation revealed that exogenous GABA application declined malondialdehyde content, electrolyte leakage and the enzyme activities of membrane metabolism, and the expression levels of related genes were also downregulated. In addition, GABA treatment scavenged ROS and strengthened the enzyme activities of ROS metabolism, as well as the expression levels of corresponding genes. Taken together, these findings implied that the repressed enzymatic browning and microbial propagation in GABA-treated stem lettuce were due to the inhibition of ROS accumulation, enhancement of membrane stability and increased resistance to oxidation.

    We report a case of a 54-year-old man with severe HTG which did not respond to conventional anti lipid therapies. He was treated with intravenous insulin and concurrent dextrose infusions which led to a dramatic reduction in serum triglyceride levels.

    The microalgal-bacterial co-cultivation was adopted as an alternative in making microbial-based biofuel production to be more feasible in considering the economic and environmental prospects. Accordingly, the microalgal-bacterial symbiotic relationship was exploited to enhance the microbial biomass yield, while bioremediating the nitrogen-rich municipal wastewater. An optimized inoculation ratio of microalgae and activated sludge (AS:MA) was predetermined and further optimization was performed in terms of different increment ratios to enhance the bioremediation process. The nitrogen removal was found accelerating with the increase of the increment ratios of inoculated AS:MA, though all the increment ratios had recorded a near complete total nitrogen removal (94-95%). In light of treatment efficiency and lipid production, the increment ratio of 0.5 was hailed as the best microbial population size in accounting the total nitrogen removal efficiency of 94.45%, while not compromising the lipid production of 0.241 g/L. Moreover, the cultures in municipal wastewater had attained higher biomass and lipid productions of 1.42 g/L and 0.242 g/L, respectively, as compared with the synthetic wastewater which were only 1.12 g/L (biomass yield) and 0.175 g/L (lipid yield). This was possibly due to the presence of trace elements which had contributed to the increase of biomass yield; thus, higher lipid attainability from the microalgal-bacterial culture. This synergistic microalgal-bacterial approach had been proven to be effective in treating wastewater, while also producing useful biomass for eventual lipid production with comparable net energy ratio (NER) value of 0.27, obtained from the life-cycle analysis (LCA) studies. Thereby, contributing towards long-term sustainability and possible commercialization of microbial-based biofuel production.

    In this research investigation, three microalgal species were screened (Pleurosigma sp., Amphora sp., and Amphiprora sp.) for lipid content before choosing the potential microalgae for biodiesel production. It was found that the lipid content of Amphiprora sp. was 41.48 ± 0.18%, which was higher than the Pleurosigma sp. (27.3 ± 0.8%) and Amphora sp. (22.49 ± 0.21%). The diatom microalga, Amphiprora sp. was isolated and exposed to a controlled environment. Two different media were prepared, and the main research was on the SiO2-NP medium as the cell wall of diatom was made up of silica. Essential growth parameters were studied such as dry cell weight and chlorophyll a content. The results revealed that Amphiprora sp. cultured in the modified medium showed a higher biomass yield and growth rate in all the analyses. In Soxhlet extraction method, biodiesel yield of Amphiprora sp. in modified medium under 24 μmol m-2 s-1 of light intensity was 81.47 ± 1.59% when using 2% of catalyst amount with 1.5:1 volume ratio of methanol/oil in 3 h reaction time at 65 °C. Results reveled that Amphiprora sp. diatom has a higher yield of oil 52.94 ± 0.42% and can be efficiently optimized with further studies with modified nanomaterial culture medium. The present research revealed the series of experiments on microalgal lipid transesterification and in future investigation different types of nanomaterials should be used in culture medium to identify the lipid production in microalgal cells.

    In many ways, cancer cells are different from healthy cells. A lot of tactical nano-based drug delivery systems are based on the difference between cancer and healthy cells. Currently, nanotechnology-based delivery systems are the most promising tool to deliver DNA-based products to cancer cells. This review aims to highlight the latest development in the lipids and polymeric nanocarrier for siRNA delivery to the cancer cells. It also provides the necessary information about siRNA development and its mechanism of action. Overall, this review gives us a clear picture of lipid and polymer-based drug delivery systems, which in the future could form the base to translate the basic siRNA biology into siRNA-based cancer therapies.

    In this study the possibility of replacing current surfactants in chocolate formulations with natural lipids extracted from spinach leaf (SPLIP) or spinach chloroplast (CH.SPLIP) was evaluated. SPLIP and CH.SPLIP were extracted with chloroform/methanol following enzyme deactivation with hot isopropanol. Results showed a higher extraction yield for SPLIP while glycolipids were more concentrated in CH.SPLIP. Sugar/oil suspensions with dispersed volume fractions of 0.28, 0.33 and 0.37 containing 0.1% to 0.7% (w/w) surfactant (SPLIP, CH.SPLIP, lecithin and PGPR as commercial references) based on oil phase were prepared and analyzed in shear rheology. Apparent viscosity at 40 s-1 was significantly lower for the natural surfactants compared to lecithin at 0.5-0.7% (w/w) addition. With regard to yield stress, taken as the shear stress at 5 s-1, both natural surfactants showed comparable performance to PGPR at 0.3% to 0.7% addition. As SPLIP and CH.SPLIP behaved similar (p > 0.05), SPLIP, due to higher extraction yield, would be the preferred choice for application in chocolate matrices.

    The present study investigates the prospective of substituting inorganic medium with organic food waste compost medium as a nutrient supplement for the cultivation of Chlorella vulgaris FSP-E. Various percentages of compost mixtures were replaced in the inorganic medium to compare the algal growth and biochemical composition. The use of 25% compost mixture combination was found to yield higher biomass concentration (11.1%) and better lipid (10.1%) and protein (2.0%) content compared with microalgae cultivation in fully inorganic medium. These results exhibited the potential of combining the inorganic medium with organic food waste compost medium as an effective way to reduce the cultivation cost of microalgae and to increase the biochemical content in the cultivated microalgae.

    This study explores the role of sterols as lipid biomarkers to indicate their input which originates from various sources in the marine environment. Sterols and their ratios were investigated in sediments taken from sixteen sampling stations at Pulau Tinggi, Johor in order to assess the sources of organic matter. The compounds extracted from the sediments were quantified using a gas chromatography-mass spectrometry (GC-MS). The distributions of sterols indicated that organic matter at all sampling stations originated from a mixture of marine source and terrestrial origins at different proportions. A total of eleven sterols were quantified, with the major compounds being phytosterols (44% of total sterols), cholesterol (11%), brassicasterol (11%) and fecal sterols (12%).

    PURPOSE: The aim of current study is to formulate, optimize and characterize the developed formulation of Mesalamine-Curcumin Nanostructured Lipid Carriers (Mes-Cur NLCs).

    METHODS: It was formulated using high pressure homogenization followed by probe sonication and formulation variables were optimized using Central Composite Design. The particle size (PS), zeta potential (ZP), entrapment efficiency (EE), drug release, cytotoxicity on NIH 3T3 fibroblasts cells and HaCaT keratinocytes cells and efficacy on RAW264.7 cells for optimized formulation was determined.

    RESULTS: The PS, ZP and EE were found to be 85.26 nm, -23.7 ± 7.45 mV, 99.2 ± 2.62 % (Mes) and 84 ± 1.51 % (Cur), respectively. The good correlation between predicted and obtained value indicated suitability and reproducibility of experimental design. NLCs showed spherical shape as confirmed by TEM. In vitro drug release profile of prepared formulation showed that Mes exhibited 100 % release at 48 h, whereas Cur exhibited 82.23 ± 2.97% release at 120 h. Both the drugs exhibited sustained release upon incorporation into the NLCs. The absence of any significant cell death during MTT assay performed on NIH 3T3 fibroblasts cells and HaCaT keratinocytes cells indicated that NLCs' were safe for use. Furthermore, significant reduction in nitric oxide level during anti-inflammatory evaluation of formulation on RAW264.7 cells showed excellent potential for the formulation to treat inflammation. The formulation was found stable as no significant difference between the PS, ZP and EE of the fresh and aged NLCs was observed.

    CONCLUSION: The outcomes of study deciphered successful formulation of Mes-Cur NLCs.

    The immobilisation of Chlorella vulgaris 211/11B entrapped in combinations of natural matrices to simplify the harvesting process was demonstrated in this study. Three combinations of matrices composed of calcium alginate (CA) and sodium alginate (SA), sodium carboxymethyl cellulose (CMC) and SA, and mixed matrices (SA, CA, and CMC) were investigated. The number of cells grown for each immobilised matrix to microalgae volume ratios (0.2:1-1:1) were explored and compared with using SA solely as a control. The optimum volume ratios obtained were 1:1 for SA, 0.3:1 for CA and SA, 1:1 for CMC and SA, and 0.3:1 for mixed matrices. The immobilised microalgae of mixed matrices exhibited the highest number of cells with 1.72 × 109 cells/mL at day 10 and 30.43% of oil extraction yield followed by CA and SA (24.29%), CMC and SA (13.00%), and SA (6.71%). Combining SA, CA, and CMC had formed a suitable structure which improved the growth of C. vulgaris and increased the lipid production compared to the immobilisation using single matrix. Besides, the fatty acids profile of the oil extracted indicates a high potential for biodiesel production.

    The single cell oil (SCO) production by the mono and mixed culture of microalgae Chlorella pyrenoidosa and red yeast Rhodotorula glutinis was investigated using non-detoxified cassava bagasse hydrolysate (CBH) as carbon source. The results suggested that the two strains were able to tolerate and even degrade some byproducts presented in the CBH, and the mixed culture approach enhanced the degradation of certain byproducts.     Cancer mortality and morbidity is projected to increase significantly over the next few decades. Current chemotherapeutic strategies have significant limitations, and there is great interest in seeking novel therapies which are capable of specifically targeting cancer cells. Given that fundamental differences exist between the cellular membranes of healthy cells and tumor cells, novel therapies based on targeting membrane lipids in cancer cells is a promising approach that deserves attention in the field of anticancer drug development. Phosphatidylethanolamine (PE), a lipid membrane component which exists only in the inner leaflet of cell membrane under normal circumstances, has increased surface representation on the outer membrane of tumor cells with disrupted membrane asymmetry. PE thus represents a potential chemotherapeutic target as the higher exposure of PE on the membrane surface of cancer cells. This feature as well as a high degree of expression of PE on endothelial cells in tumor vasculature, makes PE an attractive molecular target for future cancer interventions. There have already been several small molecules and membrane-active peptides identified which bind specifically to the PE molecules on the cancer cell membrane, subsequently inducing membrane disruption leading to cell lysis. This approach opens up a new front in the battle against cancer, and is of particular interest as it may be a strategy that may be prove effective against tumors that respond poorly to current chemotherapeutic agents. We aim to highlight the evidence suggesting that PE is a strong candidate to be explored as a potential molecular target for membrane targeted novel anticancer therapy.

    Innovative solutions are needed to limit environmental effect and optimise resource use as food waste generation rises worldwide. This study investigates the potential of upcycling food waste from fresh markets using Black Soldier Fly (Hermetia illucens) larvae (BSFL) as a sustainable approach. This study explored four fresh market food waste substrates for BSFL bioconversion: discarded fish waste (FI), slaughtered chicken waste (CHI), vegetable waste (VEG), and a 1:1:1 combination of all three (MIX). Soybean curd residue (SCR) was treated as the control substrate. The effects on larval growth, nutritional content, and waste bioconversion rates were examined. The larvae growth rate was strongly impacted by waste type, with BSF-fed CHI and MIX gaining 18.0 and 16.7 mg/d, respectively, followed by BSF-fed with SCR (12.2 mg/d), FI (8.9 mg/d) and VEG (7.6 mg/d). The waste type did not substantially alter BSFL length. The survival rate of the BSFL fed with the food waste studied ranges from 95 to 98.47%, with SCR being the highest. Our findings indicated that BSFL can effectively convert a variety of fresh market food waste into valuable biomass. CHI waste produced the highest larval biomass and bioconversion rate followed by MIX, SCR, FI and VEG. The different waste stream has a major influence on BSFL biomass nutrition. BSFL nutritional composition is independent of the substrate's nutritional content, indicating no direct correlation between substrate and BSFL biomass nutritional composition. SCR waste produced the highest protein content of BSFL (50.49%), followed by VEG (32.61%), MIX (32.57%), FI (31.03%) and CHI (29.06%). SCR waste also produced BSFL biomass with lowest lipid content (26.55%) compared to other waste which resulted into BSFL with lipid levels ranging from 42.92% to 53.72%. BSFL-fed with SCR is the most suitable to be used as an alternative animal's feed based on the protein and lipid levels, while defatting procedure is necessary for the other waste-fed BSFL to render it suitability as animal feed alternatives. Based on bioconversion rate, BSFL growth, and lipid content, the MIX and CHI waste might be viable substrates for future research.

In this paper, Nannochloropsis sp. and Tetraselmis sp. were chosen as the strains and four different methods were used to extract the lipids: Hara and Radin, Folch, Chen and Bligh and Dyer. These methods were performed by using conventional heating and microwave irradiation methods. Results revealed that highest lipid yield from the different species was obtained using different extraction methods; both under microwave irradiation. The lipid yield for Tetraselmis sp. and Nannochloropsis sp. was highest when Hara and Radin (8.19%), and Folch (8.47%) methods were used respectively under microwave irradiation. The lipids extracted were then transesterified to biodiesel and the quality of the biodiesel was analyzed using the gas chromatography.

    Different solvent extracts of Pleurotus giganteus fruiting bodies were tested for antifungal activities against Candida species responsible for human infections. The lipids extracted from the ethyl acetate fraction significantly inhibited the growth of all the Candida species tested. Analysis by GC/MS revealed lipid components such as fatty acids, fatty acid methyl esters, ergosterol, and ergosterol derivatives. The sample with high amounts of fatty acid methyl esters was the most effective antifungal agent. The samples were not cytotoxic to a mammalian cell line, mouse embryonic fibroblasts BALB/c 3T3 clone A31. To our knowledge, this is the first report of antifungal activity of the lipid components of Pleurotus giganteus against Candida species.
