Electrophoresis is a crucial step for the studies of proteins, allozymes, DNAs and RNAs. Two commonly used electrophoresis systems are agarose gel and polyacrylmide gel. Agarose gel is frequently used for DNAs and RNAs studies whereas polyacrylmide gel is widely used for the studies of other macromolecules such as proteins, allozymes (isozymes), DNAs and RNAs. The banding patterns of the gels, rather than the numbers of bands appearing on the gels are important for scoring in fingerprinting, footprinting and in population genetic studies.
Objective: Several cases of suicide attempt by charcoal burning producing CO have been reported in Malaysia. This case report highlights a case of delayed delayed neuropsychiatry sequelae (DNS) in CO poisoning.
Method: We report a young Chinese homemaker who presented with DNS who was detained in a Malaysian forensic psychiatric ward.
Results: After approximately two weeks of admission, in a familicide attempt, she had started to exhibit subtle changes in her behaviour, including social withdrawal, impaired memory, and there was lack of emotional distress.
Conclusion: We report an interesting DNS as one of the two forms of chronic sequelae of CO poisoning, whereas the other type of neuropsychiatric sequelae is the persistently progressive type.
There are a number of validated questionnaires available for the screening of Post-Traumatic Stress Disorder (PTSD), and the PTSD Checklist for Civilians is one of them. However, none was translated into the Malay Language and validated for use in the Malaysian population. The aim of this study is to translate and validate the Malay PTSD Checklist for Civilians (MPCL-C). Methods: The PCL-C was translated into the Malay Language and back-translated. The reliability and validity of the MPCL-C were then determined by administering them to those who presented at the emergency department for motor vehicle accident at least one month before. Results: The MPCL-C has good face and content validity. In terms of reliability, it is also good, with Chronbach’s alpha values of 0.90, 0.77, 0.75 and 0.74 for the full scale, re-experiencing, avoidance and arousal domains respectively. Conclusions: The MPCL-C is a valid and reliable instrument to screen for PTSD in motor vehicle accident victims for the studied population.
Triyttrium Pentairon (iii) Oxide (Y3Fe501,2) or familiar called as Yttrium Iron Garnet (YIG) is successfully prepared using a conventional mixed-oxide method of 5:3 Fe to Y ratios. YIG prepared from conventional mixed-oxide usually produced some associated phase which surely will affect electrical properties. In this study, various temperature used in the sintering process to induce associated phases (YIP) to be fully reacting to form single phase of YIG and the effect on resonance frequency is studied for resonator applications. The mixtures of oxide powders are calcined at 1100 "C and sintered at various temperatures of (1350, 1380, 1400, 1420 1450 QC, respectively). Cubic phase is detected from the formation of YIG. Some other phases such as YIP and hematite also present as secondary phase. However, it is seen that, based on the Rietvield refinement method, the total amount of secondary phase simulated are inversely proportional with sintering temperature. The powder was pressed into cylindrical pellet and tested as a microwave resonator in antenna application. It was found that, on the actual antenna circuit the operating frequencies measured are in the range of 10-12 GHz for all samples which suitable for X-band. At the end, overall radiation pattern for all samples exhibit an omnidirectional behavior.
Recently, research and development in the field of drug delivery systems (DDS) facilitating site-specific therapy has reached significant progression. DDS based on polymer micelles, coated micro- and nanoparticles, and various prodrug systems including water-soluble polymer have been prepared and extensively studied as novel drugs designed for cancer chemotherapy and brain delivery. Since polymers are going to be used in human, this study has the interest of testing two types of polymer, polyimides (PI) and polyphenylenevinylene (PPV) on neuronal cells. The objective of this study was to determine the possible neurotoxicity and potential neuroprotective effects of PI and PPV towards SH-SY5Y neuronal cells challenged by hydrogen peroxide (1120) as an oxidant. Cells were pretreated with either PI or PPV for 1 hour followed by incubation for 24 hour with 100 ,uM of 11201. MTS • assay was used to assess cell viability. Results show that PI and PPV are not harmful within the concentration up to 10 pM and 100 pM, respectively. However, PI and PPV do not protect neuronal cells against toxicity induced by H2O, or further up the cell death.
Sexual dysfunction is highly prevalent in gynaecological cancer patients. Most of the time, sexual dysfunction in gynaecological cancer is underdiagnosed as there is overlapping of symptoms with other psychological morbidities, interplaying of multiple risks, patients’ reluctance to complain or incompetence of health care provider to assess. Determining the risk factors of sexual dysfunction in cancer patients enables us to pay more attention to those who are vulnerable and to device strategies for early detection, prevention and treatment of sexual dysfunction in them. The main aim of the study was to determine the prevalence of sexual dysfunction and its risk factors in gynaecological cancer patients in Hospital Sultanah Bahiyah, Alor Star, Malaysia. Sexual function of eighty-three gynaecological cancer patients who were married were assessed with self-rated MVFSFI (Malay version Female Sexual Function Index). Self-rated WHOQOL-BREF (World Health Organization- Quality of Life- 26) which assessed the domains of quality of life was used while MINI (Mini International Neuropsychiatry Interview) was used for diagnosis of major depressive disorder. The prevalence of sexual dysfunction among the married gynaecological cancer patients was 65% (54/83). Sexual dysfunction was significantly associated with low education level (OR 3.055, CI 1.009-9.250), shorter duration of cancer (OR 0.966, CI 0.966- 0.998), ongoing chemotherapy (OR 3.045, CI 1.149-8.067), pain perception (OR 3.230, CI 1.257-8.303), absence of sexual intercourse for more than one month (OR 1.862) and three domains of quality of life such as physical health, psychological health and social relationship (OR 0.942, CI 0.908-0.978; OR 0.955, CI 0.916-0.995; OR 0.933, CI 0.894-0.973, respectively). However, sexual dysfunction was not associated with major depressive disorder (χ2 ² = 1.224, p = 0.268). The prevalence of sexual dysfunction in gynaecological cancer patients was comparable to other studies of similar population. Since, the risk factors of sexual dysfunction in gynaecological cancer patients are multidimensional, the process of assessment and management need to be holistic and patient-oriented.
Knowledge about the hemodynamic model that mediates synaptic activity and measured magnetic resonance signal is essential in understanding brain activation. Neural efficacy is a hemodynamic parameter that would change the evoked hemodynamic responses. In this work, brain activation and neural efficacy of the activated brain areas during simple addition task in two different backgrounds were studied using fMRI. The objectives were to determine the activated areas during the performance of arithmetic addition in quiet (AIQ) and noisy (AIN) background and to investigate the relationship between neural efficacy and height extent of activation for the respective areas. Eighteen healthy male participants performed simple arithmetic addition in quiet and in noise. Bilateral cerebellum, superior temporal gyrus (STG), temporal pole (TP) and supplementary motor area (SMA) were significantly (p < 0.05) activated during AIQ and AIN. Left middle frontal gyrus (L-MFG), right superior frontal gyrus (R-SFG), right superior orbital gyrus (R-SOG) and bilateral insula were more active in quiet as compared to in noise while the left middle cingulate cortex (L-MCC), left amygdala (L-AMG), right temporal pole (R-TP) and left cerebellum (L-CER) were more active in noise as compared to in quiet. The t value for most of the activated regions was found to be inversely proportional to the neural efficacy. Significant (p < 0.05) negative relationship between t value and neural efficacy were found for R-STG and bilateral cerebellum during AIQ, while for AIN, similar relationships were found in R-CER, R-STG and R-TP. This study suggests that while being significantly activated, the hemodynamic responses of these brain regions could have been suppressed by the stimulus resulting in an intensity decrease with increasing neural efficacy.
In this study polymer electrolytes composed of poly(methyl methacrylate) (PMMA) as a host polymer and ethylene carbonate (EC) as a plasticizer complexed with different lithium salts, i.e. lithium tetrafluoroborate (LiBF4) and lithium triflate (LiCF3SO3) were prepared by the solution casting technique. The conductivities of the films were characterized by impedance spectroscopy. At room temperature, the highest conductivities were 4.07 × 10–7S cm–1 and 3.40 × 10–5 S cm–1 achieved, respectively from the films containing 30 wt% LiBF4 in the PMMA-EC-LiBF4 system and 35 wt% LiCF3SO3 in the PMMA-EC-LiCF3SO3 system. The conductivity-temperature dependence of the films seemed to obey the Arrhenius equation in which the ion transport in these materials was thermally assisted. Scanning electron microscopy analysis showed that the surface of PMMA-EC-LiCF3SO3 film was smooth and homogeneous, hence lithium ions could traverse through the PMMA-EC-LiCF3SO3 film more easily compared to the PMMA-EC-LiBF4 film. X-Ray diffraction studies revealed that complexation had occurred and the complexes formed were amorphous.
Muslim doctors are those qualified doctors who practise their professional knowledge and skills in line with Islam and upholds the highest standards of ethical and professional behaviour. The medical curriculum of the Faculty of Medicine and Health Sciences of Universiti Sains Islam Malaysia (USIM) was designed with the integration of Islamic input which aims at producing doctors who are able to practise medicine that is integrated with Islamic, moral and ethical values. Halaqah Studies and Fundamental Islamic Knowledge (FIK) courses such as History of Medicine in Islam, Science and Medicine in Quran and Sunnah, Akhlak and Tasawuf, Islamic Jurisprudence and Medical Ethics and Fiqh Issues are taught to students during the pre-clinical and clinical phases. Memorization of selected Quranic verses throughout the programme aim to get the students to apply the verses of the al-Quran into practice in everyday life and especially in their clinical practice. Islamic values are emphasised during doctor-patient interactions in all clinical postings. Islamic knowledge and values integrated in the curriculum are assessed in written and clinical examinations. The outcome of the integration of the Naqli component in the medical curriculum has been demonstrated positively by the students in the patient management problems and clinical consultations. Studies on the outcome of the integrated Islamic input in the medical curriculum among the clinical students and graduates are being carried out.
Generally, the karyotype profile of Down Syndrome has been reported to be full trisomy 21 in 92% of patients, mosaic trisomy 21 in 4% of patients and translocation involving chromosome 21 in 4% of patients in most of the population groups worldwide. But, karyotype analysis of 149 DS patients at the Human Genome Center, USM, during the past five years revealed that free trisomy accounted for 94.6%, mosaic trisomy 21 for 4.7% and translocation involving chromosome 21 in 0.7% of the Down Syndrome etiology in North East Malaysian population, indicating a low frequency of translocation DS in this region. Here, we report one case of translocation Down Syndrome encountered during karyotype analysis of 149 DS cases. Karyotype showed a robertsonian translocation where an entire extra chromosome 21 was attached to the centromere of one of the chromosome 14, resulting in a derivative chromosome 14 with attached chromosome 21. Karyotype analysis of the parents revealed a normal 46,XY pattern for father and 46,XX pattern for mother indicating that this robertsonian translocation had arisen de novo either prior to or at conception.
Two isolates of brown root disease fungi were obtained from diseased roots of sentang (Azadirachta excelsa). Morphological characters from macroscopic and microscopic studies suggested that both isolates were from the same genus namely Phellinus noxius and Phellinus sp. Cloning and sequencing of ITS region were conducted to investigate further the variation between the two species at
molecular level. PCR-amplified ITS regions were cloned in pCR2.1 and sequenced. DNA sequences sized 723bp and 710bp were obtained for Phellinus noxius and Phellinus sp respectively. Comparison between the two sequences showed 98% similarity where three nucleotide substitutions and three insertion/deletion regions were found sized 8bp, 2bp and 3bp respectively.
Objective: The family caregiver has a pivotal role in the management of HIV/AIDS patients and their well-being is consequently crucial as it could impact negatively on the quality of caregiving. This preliminary qualitative investigation intended to explore and describe the challenges and experiences of HIV/AIDS family caregivers in Terengganu, Malaysia. Methods: A convenient sample of family caregivers of HIV/AIDS patients who were aware of the diagnosis was enrolled. Recruitment was conducted in Hospital Sultanah Nur Zahirah, Kuala Terengganu, Malaysia and semi-structured interviews were used. Results: Results were transcribed into verbatim before being subjected to analysis. Twelve Muslim caregivers consented participation (age range = 18.0 - 81.0; female = 75.0%, mother/wife = 50.0%; married = 83.3%; ≤ primary school
= 50.0%; and self-employed = 66.7%). The four major themes that emerged were challenges of caregiving, financial issues, stigma and discrimination, and support for caregivers. Additionally, caregivers did mention several positive aspects of their caregiving role including satisfaction from helping family member and improved family relationships. Conclusion: A variety of life aspects were negatively affected by caring for HIV patients, thus requiring a multidisciplinary approach to address such issues. Context: Question vetting is important to ensure validity, reliability, and other quality indicators of assessment tools, including the MCQ. Faculty members invest a substantial amount of time and effort into the MCQ vetting process. However, there is shortage of scientific evidence showing its effectiveness and at which level it needs to be focused on. This study aimed to provide scientific evidence regarding the effects of question vetting process on students’ examination performance by looking at their scores and pass-fail outcomes.
Method: A parallel randomized control trial was conducted on third year medical students in a medical school. They were randomly assigned into two equal groups (i.e. control and experimental). Two mock examinations were conducted (i.e. time I and time II). At time I, non-vetted MCQs were administered to both groups as a baseline measurement. At time II, vetted MCQs were administered to the experimental group, while the same non-vetted MCQs were administered to the control group.
Results: Out of 203 students, 129 (63.5%) participated in both mock examinations. 65 students were in the control group and 64 students were in the experimental group. Statistical analysis showed no significant differences (p > 0.05) in mean examination scores and pass-fail outcomes between or within the control and experimental groups.
Conclusion: This study indicated that the MCQ vetting process did not influence examination performance. Despite these findings, the MCQ vetting process should still be considered an important activity to ensure that test items are developed at the highest quality and standards. However, it can be suggested that such activity can be done at the departmental level rather than at the central level.
This paper describes a study on the design, fabrication and testing of a prototype digging device for sweet potato tubers in bris soil. The soil texture was sandy soil (fine sand 94.53%), with mean moisture content of 9.16% and mean bulk density of 1.44 g-cm-3. The soil was prepared in a soil bin. Three types of soil digging tools were designed and fabricated to determine the optimum draft force. These were Flat or plane, V-shaped and Hoe type blades. Plane and V-shaped blades were 30 cm long, and 13 cm wide, while the Hoe type had three rods, 25 mm in diameter, 30 cm long and 6.5 cm wide with sharp cutting edge. The digging tools were tested in a soil bin filled with bris soil to determine the optimum draft force and area of soil disturbance. The results were analysed using statistical analysis of variance (ANOVA). Comparison between all blade types and blade depths to measured draft force and the area of soil disturbed showed that the highest draft of 0.54 kN-m-2 was caused by a flat or plane blade at the optimum depth of 20 cm when the area of soil disturbed was 0.180 m2 . The V-shaped blade had the mean draft of 0.51 kN-m-2, with area of soil disturbance of 0.185 m2 . Thebest choice was V-shaped blade with a rake angle of 30o at 20 cm. depth. The selected blade was fixed onto the sweet potato harvester and tested on bris soil planted with sweet potato of Telong and VitAto varieties. The harvesting efficiency of the machine in bris soil was 93.64% and 90.49% for Telong (Plot A) and VitAto (Plot B) varieties, respectively. The average ground speed and turning time during operation for plots A and B was 0.56 km-hr-1 and 102.7 s and 0.99 km-hr-1 and 81.22 s, respectively. The harvesting efficiencies for both plots showed no significant difference. The total productive time (harvesting time) and unproductive time (turning time) in plot A, at a tractor speed of 0.56 km.hr-1, was 14.8 hours for harvesting a hectare of sweet potato ( 0.068 ha.hr-1). In plot B, the total time for harvesting a hectare of sweet potato was 8.35 hours (0.12 ha.hr-1) at a tractor speed of 0.99 km.hr-1. The average harvesting time for both plots was 11.47 hr.ha-1. The average field work rate was 0.087 ha.hr-1 or 34 man-hr.ha-1 compared to manual harvesting of 150 man-hrs.ha-1.
Mitochondrial DNA (mtDNA) is a hereditary material located in mitochondria and is normally maternally inherited. Mutational analysis performed on mtDNA proved that the mutations are closely related with a number of genetic illnesses, besides being exploitable for forensic identification. Those findings imply the importance of mtDNA in the scientific field. MtDNA can be found in abundance in tooth dentin where it is kept protected by the enamel, the hardest outer part of the tooth. In this study, two techniques of mtDNA extraction were compared to determine the efficacy between the two techniques. Teeth used for the study was collected from Dental Clinic, Hospital Universiti Sains Malaysia. After the removal of tooth from the tooth socket of the patient, the tooth was kept at -20C until use. Later, pulp tissue and enamel was excised using dental bur and only the root dentin was utilized for the isolation of mtDNA by crushing it mechanically into powdered form. MtDNA was extracted using the two published methods, Pfeifer and Budowle and then subjected to spectrophotometry DNA quantification and purity, Polymerase chain reaction (PCR) amplification of hypervariable-two region of mtDNA, followed by DNA sequencing to analyze the reliability of the extraction techniques. In conclusion, both techniques proved to be efficient and capable for the extraction of mtDNA from tooth dentin.
This case report highlights the success of community services and supported employment program that may be of help to a woman with chronic inhalant use to achieve a complete abstinence. Methods: We report the case of a 45 year old lady who chronically abused glue, presented after 10 years with substance induced psychosis and comorbid mood disorder. Results: Abstinent from inhalant dependence was achieved after series of home treatment services and supported employment program. Conclusion: Provision of acute community service and incorporating this patient in supported employment program contribute to her successful journey in achieving abstinence from inhalant dependence.
This research describes a comparative study using convenience sampling on management of intravenous lines in Maternity Neonatal Intensive Care Unit (MNICU), HKL. Questionnaires were answered by nurses and the techniques of anchoring, splinting and maintaining of intravenous lines by nurses in MNICU were observed by the researchers. The study showed that nurses with less than two years experience working in MNICU had significantly less knowledge and skills in the management of intravenous lines and are likely to encounter more difficulties and complications, compared to nurses with more experience.
β-tricalcium phosphate (β-TCP) powders were synthesized by using various particles sizes (40 nm – 780 μm) calcium carbonate (CaCO3) and phosphoric acid (H3PO4) at room temperature (25 ˚C). The synthesized powders were characterized by using X-Ray Diffraction (XRD) method. The purity of β-TCP powders were determined from XRD pattern while the crystallite size of β-TCP powders were calculated by using Scherrer equation. Results shows that the purity of β-TCP powders were ranged from 20.33 % to 81.94 % while the crystallite size of β- TCP powders were ranged from 0.04391 μm to 0.06751 μm. From this work, particle size of CaCO3 will influenced the purity but not the mean crystallite size of synthesized β-TCP.
A study was carried out to determine short term diurnal radon concentration at five locations in Malaysia. Two locations (KG & AP) are former tin mining areas that has been converted to housing area and training centre respectively, one a relatively new city (SA), that was formerly a rubber and oil palm plantation area, one older cities (KB) and one housing complex by the sea (LP). The study was carried out in 2005, 2006 and 2007 using a diffused-junction photodiode sensor continuous radon monitor. The monitor is recognized by the USEPA. In each location, measurements were carried out on at least ten sites. Former tin mining areas of KG and AP shows up to seven times higher indoor average than the average in the other three locations. However the indoor average in all locations is still below the action level of 4 pCiL-1. For outdoor, the former tin mining areas average concentration was higher than the global average of 0.4 pCiL-1. For the twenty four hours temporal variation the trend indicated that former tin mining areas concentration are always higher, and at time up to six fold higher. The hourly variation of all locations follows an identical trend of high concentration during early to late morning and drop in the afternoon till evening. The outdoor twenty four hour temporal average of former tin mining areas is consistently higher than the outdoor global average of 0.4 pCiL-1. The strong correlation between indoor and outdoor concentration at AP, indicates that indoor radon might originates from outdoor environment. The study was also extended to estimate the effective dose (mSvyr-1) of Rn-222 to the public.
Analyses activities of 226 Ra and 228 Ra were conducted at ten stations of Pulau Redang, Malaysia. Dissolved radium isotopes such as 226 Ra and 228 Ra had shown enrichment at coastal area stations. Meanwhile, activities of both nuclides in the suspended particle matters were slightly in equilibrium with the activity ratio ranging from 0.88 – 1.86. The calculated distribution coefficient values (Kd) of 226 Ra and 228 Ra were in the range of 0.78 x 10 5 L g -1 to 5.56 x 10 5 L g -1 and 0.21 x 10 5 L g -1 to 1.86 x 10 5 L g -1 , respectively, indicate that most of the radium nuclides in the study area are strongly absorbed into the particulate phases. Therefore, low concentrations of suspended particles matter in the water column (< 10 mg L -1 ) have insignificant effects on the Kd values.
