    Telomeres shorten with physiological aging but undergo substantial restoration during cancer immortalization. Increasingly, cancer studies utilize the archive of formalin-fixed, paraffin-embedded (FFPE) tissues in diagnostic pathology departments. Conceptually, such studies would be confounded by physiological telomere attrition and loss of DNA integrity from prolonged tissue storage. Our study aimed to investigate these two confounding factors. 145 FFPE tissues of surgically-resected, non-diseased appendixes were retrieved from our pathology archive, from years 2008 to 2014. Cases from 2013 to 2014 were categorized by patient chronological age (0-20 years, 21-40 years, 41-60 years, > 60 years). Telomere lengths of age categories were depicted by telomere/chromosome 2 centromere intensity ratio (TCR) revealed by quantitative fluorescence in situ hybridization. Material from individuals aged 0-20 years from years 2013/2014, 2011/2012, 2009/2010, and 2008 were compared for storage effect. Telomere integrity was assessed by telomere fluorescence intensity (TFI). Epithelial TCRs (mean ± SD) for the respective age groups were 4.84 ± 2.08, 3.64 ± 1.21, 2.03 ± 0.37, and 1.93 ± 0.45, whereas corresponding stromal TCRs were 5.16 ± 2.55, 3.84 ± 1.36, 2.49 ± 1.20, and 2.93 ± 1.24. A trend of inverse correlation with age in both epithelial and stromal tissues is supported by r = -0.69, p < 0.001 and r = -0.42, p < 0.001 respectively. Epithelial TFIs (mean ± SD) of years 2013/2014, 2011/2012, 2009/2010 and 2008 were 852.60 ± 432.46, 353.04 ± 127.12, 209.24 ± 55.57 and 429.22 ± 188.75 respectively. Generally, TFIs reduced with storage duration (r = -0.42, p < 0.001). Our findings agree that age-related telomere attrition occurs in normal somatic tissues, and suggest that an age-based reference can be established for telomere studies on FFPE tissues. We also showed that FFPE tissues archived beyond 2 years are suboptimal for telomere analysis.

    Non-alcoholic fatty liver disease (NAFLD) is a clinicopathological syndrome strongly associated with
    hyperlipidemia, hypertriglyceridemia, insulin resistance and obesity. The objective of this study is to evaluate the
    potential health benefits of Baccaurea angulata fruit in preventing liver damage due to hypercholesterolemia.
    Twenty-five healthy adult New Zealand White rabbits were assigned to five groups for 90 days diet and
    intervention plan. While four groups (atherogenic groups) were fed 1% cholesterol diet and 0, 0.5, 1.0, and 1.5 mL
    of B. angulata juice kg/per day, the fifth group (negative control) was fed with commercial rabbit pellet only.
    After overnight fasting and sacrifice, the thoracic cavity was opened and the liver was carefully removed,
    specifically observing for any presence of a gross lesion and then immediately fixing in 4% paraformaldehyde for
    at least 48 hours for histopathological analysis. The hepatoprotective effect of B. angulata fruit juice was evident
    in the histological examination of liver tissues. The results indicate the potential health benefits of the antioxidantrich
    B. angulata fruit juice as a functional food with a therapeutic effect against hypercholesterolemia-induced
    liver injury.

    Blastocystis hominis merupakan antara protozoa yang paling biasa ditemui di dalam sampel feses manusia di seluruh dunia. Prevalens infeksi protozoa ini adalah lebih tinggi di kalangan mereka yang tinggal di negara membangun berbanding negara maju. Seramai 71 orang kanak-kanak Orang Asli dari Pos Lenjang, Pahang telah menjadi subjek dalam kajian ini. Bagi kajian yang lebih terperinci, kumpulan kanak-kanak ini telah dibahagikan menurut jantina dan umur. Sampel feses dikumpul dan setiap sampel diperiksa dengan menggunakan 3 teknik diagnostik iaitu teknik apusan langsung, konsentrasi formalin-eter dan perwarnaan trikrom bagi tujuan pengesanan dan pengenalpastian Blastocystis hominis. Prevalens infeksi Blastocystis hominis di kalangan kanak-kanak Orang Asli adalah sangat tinggi iaitu 93%. Kanak-kanak perempuan didapati lebih ramai terinfeksi (97.5%) berbanding kanak-kanak lelaki (87.1%) walaupun secara statistiknya tidak signifikan (p>0.05). Protozoa ini juga telah menginfeksi kesemua kanak-kanak prasekolah (100%) manakala kanak-kanak yang bersekolah turut menunjukkan prevalens infeksi yang tinggi iaitu 86.5%. Daripada segi diagnosis, teknik perwarnaan trikrom didapati paling sensitif dan ia dapat mengenalpasti kesemua (66) sampel feses yang positif dengan Blastocystis hominis. Ini diikuti dengan teknik konsentrasi formalin-eter (43 sampel) dan teknik apusan langsung (18 sampel) (p<0.05). Prevalens infeksi Blastocystis hominis yang tinggi di kalangan kanak-kanak Orang Asli adalah berhubungkait dengan pelbagai faktor termasuk status sosioekonomi yang rendah, budaya, kekurangan kemudahan asas dan tahap pengetahuan mengenai penjagaan kesihatan serta kebersihan diri yang rendah. Selain itu, peningkatan prevalens infeksi dalam kajian ini menunjukkan pentingnya penggunaan teknik diagnostik yang lebih berkesan di dalam pemeriksaan rutin bagi memperolehi hasil diagnosis yang lebih tepat.

    Cryptococcus neoformans is an encapsulated fungal pathogen that causes severe disease primarily in
    immunocompromised patients. Adherence and internalisation of microbial pathogens into host cells often
    begin with engagement of microbes to the surface receptors of host. However, the mechanisms involved
    remain poorly understood. In this study, we investigated the association of cell surface determinants of C.
    neoformans with mammalian cells. Our results showed that treatment with trypsin, but not paraformaldehyde
    or heat killing, could reduce host-cryptococci interaction, suggesting the involvement of cell surface proteins
    (CSPs) of C. neoformans in the interaction. We extended our investigations to determine the roles of CSPs
    during cryptococci-host cells interaction by extracting and conjugating CSPs of C. neoformans to latex beads.
    Conjugation of CSPs with both encapsulated and acapsular C. neoformans increased the association of latex
    beads with mammalian alveolar epithelial cells, alveolar macrophages and monocyte-derived macrophages.
    Further examination on the actin organisation of the host cells implied the involvement of actin-dependent
    phagocytosis in the internalisation of C. neoformans in CSP-conjugated latex beads. We hypothesised that
    CSPs present on the cell wall of C. neoformans mediate the adherence and actin-dependent phagocytosis
    of cryptococci by mammalian cells. Our results warrant further studies on the exact role of CSPs in the
    pathogenesis of cryptococcosis.

    A study to determine the immunoglobulin and cellular responses in the respiratory tract of goats following intranasal exposures to formalin-killed Pasteurella haemolytica A2 was carried out. Forty-two goats were divided into two groups. Goats in Group 1 were subjected to double intranasal exposures to formalin-killed P. haemolytica A2 while goats in Group 2 were the unexposed control. Prior to and at weekly intervals post-exposure, three goats from each group were killed, serum samples were collected while the lungs were flushed with 50 ml normal saline before the right apical lobes were fixed in 10% buffered formalin. Both serum and lung lavage fluid were subjected to enzyme-linked immunosorbent assay (ELISA) to determine the levels of IgA, IgM and IgG while the formalin-fixed tissues were examined histologically. IgA levels in the lung lavage fluid increased rapidly to reach a significantly (p < 0.05) high level as early as Week 2 post-exposure and remained significantly (p < 0.05) high throughout the study period. The IgM levels increased at an intermediate rate to reach a significantly (p < 0.05) high level at Week 3 post-exposure before they decreased to an insignificant (p > 0.05) level the following week and the weeks thereafter. IgG levels increased gradually and only reached a significantly (p < 0.01) high level at Weeks 5 and 6 of the study. The size of the bronchus-associated lymphoid tissue (BALT) and the number of lymphocytes in BALT increased significantly from Week 2 and remained high thereafter. However, differences in the numbers of BALT were insignificant (p > 0.05) initially before becoming significantly (p < 0.05) high at Weeks 5 and 6. The BALT responses were parallel to those of imunoglobulins in the lung lavage fluid.

    A survey was conducted to determine the diversity of fish in Pergau Lake, Jeli, Kelantan, Malaysia from 2 September to 18 October of 2016. Fish samples were collected at seven random sampling stations around the lake by using setting trap method. The collected samples were preserved in 10% formalin solution and delivered to the laboratory for further identification process. Fourteen species, namely as Hemibagrus nemurus, Hampala macrolepidota, Clarias batrachus, Channa striata, Cyprinus carpio, Poropuntius smedleyi, Pangasius sp., Oreochromis niloticus, Oreochromis mossambicus, Leptobarbus hoevenii, Neolissochilus hexagonolepis, Tor tambroides, Osteochilus hasselti and Neolissochilus soroides, comprising of six families were collected during this study. From this study, family of Cyprinidae was the dominant species in Pergau Lake with exactly 50% of catch percentage, followed by family of Bagridae (29%), Cichlidae (10%), Clariidae (5%), Channidae (3%) and Pangasiidae (3%). This study shows that there is still a diversity of fish species in Pergau Lake, showing that the water and its environmental condition is very good.

    INTRODUCTION: In recent years, "double hit" and "double protein" involving gene rearrangement and protein expression of c-MYC and BCL2 and/or BCL6 are the most used terms to describe poor prognostic factors in diffuse large B-cell lymphoma (DLBCL). This study was to determine the frequency of double or triple protein expression by using immunohistochemistry (IHC) and comparing the result with clinicopathological features and cell of origin (COO) classification.

    METHODS: We conducted a cross-sectional study by using 29 archived formalin-fixed paraffin embedded tissue blocks of DLBCL. All the samples were evaluated for the subgrouping of COO DLBCL was determined by expression of CD10, BCL6 and MUM1 based on Hans classification. In addition, expressions of c-MYC, BCL2 and BCL6 were detected by IHC.

    RESULTS: Among the 29 cases, MYC, BCL2 and BCL6 proteins were detected in 72.4%, 62.1% and 62.1% of patients, respectively. Concurrent expression (c-MYC positive/BCL2 positive and/or BCL6 positive) was present in 58.6% of patients. 34.5% were categorised as germinal centre like (GCB) subgroup and 65.5% were categorised as nongerminal centre like (non-GCB) subgroup. Among the clinicopathological features, the double/triple protein expression lymphoma was significantly associated with elevated LDH level (p=0.018), IPI score (p=0.003), Ann Arbor stage (p=0.011) and complete response rate (p=0.011).

    CONCLUSION: Double/triple protein lymphoma was strongly associated more adverse clinical risk factors. Thus, analyses of MYC, BCL2 and BCL6 expression by IHC represents a rapid and inexpensive approach to risk-stratify patients with DLBCL at diagnosis.

    This study has been conducted in a new constructed building of NIOSH Malaysia located at Bandar Baru Bangi, Selangor. The goal of the case study is focusing on the level of Indoor Air Contaminants (IAC) including chemical contaminants within three consequent stages which are before furniture install, after furniture install and during one month occupancy. This study was divided the sampling area into two main facilities which are training and office setting. The contaminants has been measured consist of sixparameters such as Carbon Dioxide (CO2), Carbon Monoxide (CO), Total Volatile Organic Compounds (TVOC), Formaldehyde, Respirable Particulates (PM10) and Ozone. The result of Carbon Monoxide (CO), Total Volatile Organic Compound (TVOC), Respirable Particulates (PM10) and Ozone show an increasing trend across the three sampling stages. The Formaldehyde show an increasing trend in the first and second stages but were reduced significantly the last stage of sampling. These finding indicates that furniture and fittings installed might be a potential sources of indoor air contaminants. The management should be aware to their indoor air status to protect the occupant from the risk of unwanted exposure especially during the early stage of building occupancy.

    Dentin morphology and the lesion found in dental caries have been studied for many years. It was first observed under optical microscopy, and later using electron microscopy. Confocal laser scanning microscopy (CLSM) applied with several fluorescent dyes such as alizarin red to see normal dentinal tubules. However, as far as authors aware, the CLSM studies of dentinal tubules in human caries using alizarin red is rare. The aim of this study is to examine histopathological and morphological changes in dentinal tubules of dentin caries stained with alizarin red using CLSM. Fifteen extracted carious teeth (premolar or molar) was collected and fixed in neutral formalin solution buffered with phosphate buffer, rinsed and stored in calcium free phosphate buffer saline (PBS) at 4°C. The specimens were dehydrated and embedded in resin. Longitudinal or cross sections were cut and polished and then stained with alizarin red S (100 μg/ml) in 0.5 M HCl solution for 24-48 hour at 37°C. After dehydration specimens were mounted on glass slide and examined under CLSM using epi-flourescent mode or transmission light mode with wave length of 512 nm. The images of dentinal tubules were taken serially and optimum images of three-dimensional structures were reconstructed using software of CLSM. Histopathological changes of dentinal tubules in human caries showed area of demineralized dentin, translucent zone, and normal area. The dentinal tubules were thin and had numerous branches. In conclusion, confocal microscopy revealed Study shows that confocal microscopy revealed histopathological changes in dentinal tubules affected by carious lesions.

    Food safety issues associated with aquatic food products become more important with the increasing consumption and followed by its ongoing challenges. The objective of this paper is to review the food safety hazards and health risks related to aquatic food products for the Southeast Asian region. These hazards can be categorized as microplastics (MPs) hazard, biological hazards (pathogenic bacteria, biogenic amines, viruses, parasites), and chemical hazards (antimicrobial, formaldehyde, heavy metal). In different Southeast Asian countries, the potential health risks of aquatic food products brought by food hazards to consumers were at different intensity and classes. Among all these hazards, pathogenic bacteria, antimicrobials, and heavy metal were a particular concern in the Southeast Asian region. With environmental changes, evolving consumption patterns, and the globalization of trade, new food safety challenges are created, which put forward higher requirements on food technologies, food safety regulations, and international cooperation.

    Background: Except for a few studies with contradictory observations, information is lacking on the possibility of association between DNA mismatch repair (MMR) status and the presence of cancer stem cells in colorectal carcinoma (CRC), two important aspects in colorectal carcinogenesis.

    Methods: Eighty (40 right-sided and 40 left-sided) formalin-fixed, paraffin-embedded primary CRC were immunohistochemically studied for CD133, a putative CRC stem cell marker, and MMR proteins MLH1, MSH2, MSH6 and PMS2. CD133 expression was semi-quantitated for proportion of tumor immunopositivity on a scale of 0-5 and staining intensity on a scale of 0-3 with a final score (units) being the product of proportion and intensity of tumor staining. The tumor was considered immunopositive only when the tumor demonstrated moderate to strong intensity of CD133 staining (a decision made after analysis of CD133 expression in normal colon). Deficient MMR (dMMR) was interpreted as unequivocal loss of tumor nuclear staining for any MMR protein despite immunoreactivity in the internal positive controls.

    Results: CD133 was expressed in 36 (90.0%) left-sided and 28 (70.0%) right-sided tumors (p  0.05).

    Conclusion: Proficient MMR correlated with high levels of CD133-marked putative cancer stem cells in both right- and left-sided tumors, whereas significantly lower levels of CD133-marked putative cancer stem cells were associated with deficient MMR status in colorectal carcinomas found on the right.

    A diesel engine has been a desirable machine due to its better fuel efficiency, reliability, and higher power output. It is widely used in transportations, locomotives, power generation, and industrial applications. The combustion of diesel fuel emits harmful emissions such as unburned hydrocarbons (HC), particulate matter (PM), nitrogen oxides (NOx), and carbon monoxides (CO). This article presents data on the efficiency, combustion, and emission of a 4-stroke diesel engine. The engine is a 6.8 L turbocharged 6-cylinder Tier II diesel engine fitted with a common rail injection system. The test was carried out at the Powerhouse Energy Campus, Colorado State University Engines and Energy Conversion facility. The ISO Standard 8178:4 Cycle D2 cycle was adopted for this study consists of five test runs at 1800 rpm. During the testing, CO, carbon dioxide (CO2), oxygen (O2), NOx, PM, unburned HC as a total HC (THC), methane (CH4), formaldehyde (CH2O), and volatile organic compound (VOC) emissions were measured. At the same time, the data acquisition system recorded the combustion data. The engine's performance is characterized by the brake specific fuel combustion (BSFC) and thermal efficiency. A dataset of correlations among the parameters was also presented in this article.

    Although primary localised tumours of renal cell carcinoma (RCC) can be treated relatively successfully with surgery, metastatic RCC has poor prognosis because of late diagnosis and resistance to therapies. In the present study, we were interested in profiling the protein expression of "inhibitor of caspase-activated DNase" (ICAD), an apoptosis inhibitor, in kidney cancer and its paired normal kidney. Immunohistochemistry with automated batch staining and morphometry using digital pathology were used to compare ICAD in 121 RCC specimens with their paired normal kidney tissue. Tissue microarray of formalin-fixed, paraffin-embedded archival tissue was used. Intensity and localisation of ICAD were compared between normal and cancer samples, and against grading within the cancers. The results demonstrated that, in this cohort, ICAD was highly expressed in the proximal tubular epithelium of normal kidney, and significantly decreased in clear cell RCC tissue (p < 0.05) as well as other subtypes of RCC (p < 0.01) compared with normal kidney. There was a tendency towards nuclear localisation of ICAD in clear cell RCC, but not in other subtypes of RCC. No significant association was found between ICAD intensity and grade of RCC. In summary, down-regulation of ICAD occurs in RCC. ICAD normally inhibits DNA fragmentation and apoptosis; thus, its down-regulation was unexpected in a cancer known for its resistance to apoptosis. However, these RCC samples were from primary, not metastatic, RCC sites, and down-regulated ICAD may be part of a progressive pathway that promotes RCC metastasis.

    BACKGROUND: Somatic mutations of the epidermal growth factor receptor (EGFR) are reportedly associated with various responses in non-small cell lung cancer (NSCLC) patients receiving the anti-EGFR agents. Detection of the mutation therefore plays an important role in therapeutic decision making. The aim of this study was to detect EGFR mutations in formalin fixed paraffin embedded (FFPE) samples using both Scorpion ARMS and high resolution melt (HRM) assay, and to compare the sensitivity of these methods.

    RESULTS: All of the mutations were found in adenocarcinoma, except one that was in squamous cell carcinoma. The mutation rate was 45.7% (221/484). Complex mutations were also observed, wherein 8 tumours carried 2 mutations and 1 tumour carried 3 mutations.

    CONCLUSIONS: Both methods detected EGFR mutations in FFPE samples. HRM assays gave more EGFR positive results compared to Scorpion ARMS.

    The effect of morphology on the plasmonic sensing of the presence of formaldehyde in water by gold nanostructures has been investigated. The gold nanostructures with two different morphologies, namely spherical and rod, were prepared using a seed-mediated method. In typical results, it was found that the plasmonic properties of gold nanostructures were very sensitive to the presence of formaldehyde in their surrounding medium by showing the change in both the plasmonic peaks position and the intensity. Spherical nanoparticles (GNS), for example, indicated an increase in the sensitivity when the size was increased from 25 to 35 nm and dramatically decreased when the size was further increased. An m value, the ratio between plasmonic peak shift and refractive index change, as high as 36.5 nm/RIU (refractive index unit) was obtained so far. An expanded sensing mode to FD was obtained when gold nanostructures with nanorods morphology (GNR) were used because of the presence of two plasmonic modes for response probing. However, in the present study, effective plasmonic peak shift was not observed due to the intense plasmonic coupling of closely packed nanorod structures on the surface. Nevertheless, the present results at least provide a potential strategy for response enhancement via shape-effects. High performance plasmonic sensors could be obtained if controlled arrays of nanorods can be prepared on the surface.

     The association of human papillomavirus (HPV) with juvenile laryngeal papillomatosis has been well documented. We report two cases of juvenile laryngeal papillomatosis and correlated these cases with presence of HPV, p53 and c-erbB-2 proteins. The first case was a one-year-old male patient and the second a six-year-old female patient. Formalin-fixed paraffin-embedded biopsy specimens were tested for the presence of HPV genome by the technique of in situ hybridisation using wide spectrum and type specific biotinylated probes while the immunohistochemical expression of p53 (D07, 1:50) and c-erbB-2 (DAKO A0485, 1:300) proteins were evaluated with commercially available antibodies. Histologically the tumours in both cases showed papillary configuration of squamous papilloma. The first case detected HPV type 6, HPV type 11 and p53 protein expression while the second case showed only HPV type 6. Both cases of HPV showed positive signals confined to the nuclei in the superficial squamous epithelium. The first case showed p53 positivity seen from the basal region up to one third of the epithelium of laryngeal papillomas and the subsequent recent repeat biopsy showed the positivity of p53 had extended throughout the upper layers of the epithelium. Expression of c-erbB-2 protein was not detected in both cases. These findings were similar as in other studies where follow-up of the cases was recommended since they tend to recur.

    Cervical carcinoma, the second most common malignancy in Malaysian females, is aetiologically linked to the human papillomavirus (HPV). A study was conducted at the Department of Pathology, University of Malaya Medical Centre to compare the identification of HPV 6, 11, 16 and 18 in 40 archived formalin-fixed, paraffin-embedded cervical carcinoma by non-isotopic in-situ hybridisation (NISH) and polymerase chain reaction (PCR). HPV L1 ORF consensus PCR was also carried in cases which were negative on HPV type-specific PCR. NISH detected HPV 16 in 13 (32.5%) cases with one case demonstrating a concomitant HPV 18. beta-globin DNA PCR was carried out on the same paraffin block as for NISH in 27 cases and on a different paraffin block in 13, with amplification in 9 of the former and 3 of the latter. Thus only 12 cases were subjected to further HPV PCR. HPV was detected in 10 (83.3%) with HPV 16 in 9 cases and HPV L1 ORF in one. When using the same paraffin block for both methods of HPV detection, NISH detected HPV in 6 and PCR in 7. NISH failed to detect HPV in a case detected by PCR. 2 cases were negative for HPV using both methods. Hence, HPV detection results by NISH and PCR were concordant in 88.9%. Interestingly, NISH detected HPV in 2 cases with non-amplifiable beta-globin DNA. Using an alternative paraffin block for HPV PCR from NISH, HPV DNA was detected in 3 cases, two of which also showed type-specific positivity on NISH. The third case did not reveal type-specific positivity with NISH or PCR but demonstrated HPV DNA on L1 ORF consensus PCR. It thus appears that PCR and NISH can be successfully used to detect HPV in formalin-fixed, paraffin-embedded tissue and in the presence of intact DNA NISH may be as sensitive as PCR.

    Synthetic adhesives in the plywood industry are usually volatile compounds such as formaldehyde-based chemical which are costly and hazardous to health and the environment. This phenomenon promotes an interest in developing bio-boards without synthetic adhesives. This study proposed a novel application of natural mycelium produced during mushroom cultivation as natural bio-adhesive material that convert spent mushroom substrate (SMS) into high-performance bio-board material. Different types of spent mushroom substrates were compressed with specific designed mould with optimal temperature at 160 °C and 10 mPa for 20 min. The bio-board made from Ganoderma lucidum SMS had the highest internal bonding strength up to 2.51 mPa. This is far above the 0.4-0.8 range of China and US national standards. In addition, the material had high water and fire resistance, high bonding and densified structures despite free of any adhesive chemicals. These properties and the low cost one step procedure show the potential as a zero-waste economy chain for sustainable agricultural practice for waste and remediation.

    This review summarizes the results of epidemiological studies focusing on the detrimental effects of home environmental factors on asthma morbidity in adults. We reviewed the literature on indoor air quality (IAQ), physical and sociodemographic factors, and asthma morbidity in homes, and identified commonly reported asthma, allergic, and respiratory symptoms involving the home environment. Reported IAQ and asthma morbidity data strongly indicated positive associations between indoor air pollution and adverse health effects in most studies. Indoor factors most consistently associated with asthma and asthma-related symptoms in adults included fuel combustion, mold growth, and environmental tobacco smoke. Environmental exposure may increase an adult's risk of developing asthma and also may increase the risk of asthma exacerbations. Evaluation of present IAQ levels, exposure characteristics, and the role of exposure to these factors in relation to asthma morbidity is important for improving our understanding, identifying the burden, and for developing and implementing interventions aimed at reducing asthma morbidity.
