    Results from the present study have shown that the ionic species of buffers, pH values and reaction temperature can affect the enzyme unit activities and product specificity of Toruzyme (Novo Nordisk A/S Bagsvaerd, Denmark) CGTase (cyclodextrin glucanotransferase). Applying a similar reaction environment (acetate buffer, pH 6.0; temperature, 60 degrees C), the CGTase was found to be capable of producing pre dominantly beta-cyclodextrin from either raw or gelatinized sago (Cycas revoluta) starch. Changing the buffer from acetate to phosphate reduced the yield of beta-cyclodextrin from 2.48 to 1.42 mg/ml and also affected the product specificity, where production of both alpha- and beta-cyclodextrins were more pronounced. The decrease in the production of cyclodextrins in phosphate buffer was significant at both pH 6.0 and 7.0. However, changing the buffer to Tris/HCl (pH 7.0) showed a significant increase in beta-cyclodextrin production. Increasing the ionic strength of sodium acetate and Tris/HCl buffers at pH 6.0 and 7.0 to equivalent ionic strength of phosphate buffers showed no significant effects on cyclodextrin production. Higher yield of cyclodextrins at pH 7.0 when Tris/HCl was used might be due to the binding of chloride ions at the calcium-binding sites of the CGTase, resulting in the shift of the optimum pH close to physiological environment, leading to an increase in the activities and specificity.

    Telangiectatic osteosarcoma is a rare variant of osteosarcoma and can be easily misdiagnosed as aneurysmal bone cyst. We report an atypical case of iliac telangiectatic osteosarcoma in a young healthy female, who presents with painful slow growing expansile lytic septate lesion in the left hemipelvis, which is initially treated as aneurysmal bone cyst. The diagnosis of aneurysmal bone cyst is made after histopathological examination of core needle biopsy. Her condition became unstable and massive bleeding is noted at the lesion site after sclerosant injection. She undergoes emergency hemipelvectomy and eventually the biopsy turns up to be telangiectatic osteosarcoma. Our case highlights that core needle biopsy is not useful in making diagnosis for iliac telangiectatic carcinoma. Hence, an open biopsy should be carried out in our case. This case also emphasises on careful evaluation for malignancy which is mandatory because bleeding from pelvis after an unsuitable treatment can be grave, to the extent that major amputation hemipelvectomy is an option. Even though telangiectatic osteosarcoma has the same prognosis and treatment with conventional osteosarcoma, the outcome of delayed treatment for telangiectatic osteosarcoma is not good due to the dilemma in establishing an early correct diagnosis.

    BACKGROUND: The bite-sized jelly sphere with a gelatinous exterior and fruit puree interior is a type of innovative fruit-based dessert. This study aimed to produce jelly spheres with a gelatinous exterior and mangopineapple puree interior by using frozen reverse spherification.

    METHODS: A full factorial design (23) was applied to study the effects of mango-pineapple ratio (x1), immersion time in sugar solution (x2), and concentration of sugar solution (x3) in the production  of mango-pineapple jelly spheres using frozen reverse spherification. The responses studied were the physicochemical properties (color, total soluble solids, and texture) and sensory evaluation of mango-pineapple jelly spheres.

    RESULTS: Mango-pineapple ratio had a positive effect on a* and b* while having a negative effect L* value on the jelly sphere. Total soluble solids of jelly spheres were influenced by both immersion time in sugar solution and concentration of sugar solution. Immersion time in sugar solution had a positive effect on the peak force of the compression cycle and deformation at peak load while having a negative effect on the total soluble solid of jelly spheres. On the other hand, the concentration of sugar solution had a positive effect on the sensory evaluation in terms of flavor, texture, and overall acceptability. The desirability function approach was used to optimize the factors, and an overall desirability of 0.89 for all responses was achieved with 1.28:1 mango-pineapple ratio, 30 mins immersion time in sugar solution, and 22°Brix sugar solution. A proximate analysis of the optimized mango-pineapple jelly spheres had an energy content of 73.18 kcal/100 g and showed nutrient values of 81.11% moisture, 0.10% ash, 0.46% protein, 0% fat, 0.97% total dietary fiber, and 17.35% digestible carbohydrate.

    CONCLUSIONS: The development of the optimal mango-pineapple jelly sphere allows food producers to produce a dessert that is low in calories, with a good appearance and consumer acceptability.

    The composition of ophthalmic preparation is administered topically to the eye in the form of a solution, suspension, ointment, gel or foam for the purpose of treating eye disease. Virgin Coconut Oil (VCO) has been one of the desired ingredients of choice, as its benefits as functional food oil is known among the public. The uniqueness of coconut oil is its fats and oils that contain the highest percentage of medium-chain fatty acids (MCFA), which have antimicrobial properties, such as lauric acid and capric acid. This study aimed to evaluate the antimicrobial effects of eye drop containing a different VCO concentration using the Kirby-Bauer test. The formulation of eye drop had 1.5%, 2% and 3% amount of virgin coconut oil, which was later added with all basic materials needed for eye drop. The samples were evaluated for its zone of inhibition (ZOI). The antimicrobial effects of eye drop formulation that contains 3% of VCO (F3) against Streptococcus pneumonia were similar with all control products (Eye Glo, Pred Forte, Cationorm), and antimicrobial effects of F3 against Staphylococcus aureus are better than Cationorm. It is noticeable that the higher the VCO content in the formulation, the better the antimicrobial effects of the eye drop. In conclusion, VCO possesses moisture, anti-inflammation, better anti-microbial properties, and it could be further formulated as a stable eye drop emulsion.

    The incidence of Acanthamoeba keratitis has been increasing since the previous decades, especially among contact lens users. This infection is majorly caused by the use of ineffective contact lens disinfecting solution. Thus, this study was conducted to evaluate the in vitro effects of multi-purpose disinfecting solutions (MPDS) against Acanthamoeba trophozoites and cysts. Acanthamoeba genotype T4 isolated from contact lens paraphernalia and an environmental strains were propagated for trophozoite or cyst-containing culture and adjusted in final concentration of 1 × 105 cells/ml. Amoebicidal and cysticidal assays were conducted by incubating trophozoites and cysts with OPTI-FREE® Express®, ReNu® Fresh™, Complete® Multi-Purpose Solution and AVIZOR Unica® Sensitive according to the manufacturer's minimum recommended disinfectant time (MMRDT) for up to 12 h at 30 ⁰C. Trypan blue hemocytometer-based microscopic counts determined amoebicidal and cysticidal effects. The viability of Acanthamoeba trophozoites and cysts was confirmed by re-inoculated them in the 1.5% non-nutrient agar plates. It was found that none of the MPDS showed amoebicidal and cysticidal effects during the MMRDT. However, OPTI-FREE® Express® demonstrated a significant differences in average cell reduction for both stages within MMRDT. When subjected to 12 h exposure, both OPTI-FREE® Express® and ReNu® Fresh™ led to significant reduction in the number of trophozoite and cyst cells. Notably, Complete® Multi-Purpose Solution and AVIZOR Unica® Sensitive did appreciably improve the solution effectiveness towards trophozoite cells when incubated for 12 h. All MPDS were largely ineffective, with 100% survival of all isolates at MMRDT, while OPTI-FREE® Express® showed limited amoebicidal activity against the contact lens paraphernalia isolate, however, it was more against the environmental strains after 12 h incubation time. The commercially available MPDS employed in this research offered minimal effectiveness against the protozoa despite the contact time. Improvement or development of new solution should consider the adjustment of the appropriate disinfectant concentration, adequate exposure time or the incorporation of novel chemical elements, which are effective against Acanthamoeba for accelerated disinfecting and more reduction of potential exposure of contact lens users to Acanthamoeba keratitis.

    Poly (vinyl chloride) membrane electrodes that responded selectively towards the antimalarial drug chloroquine are described. The electrodes were based on the use of the lipophilic potassium tetrakis(4-chlorophenyl)borate as ion-exchanger and bis(2-ethylhexyl)adipate (BEHA), or trioctylphosphate (TOP) or dioctylphenylphosphonate (DOPP) as plasticizing solvent mediator. All electrodes produced good quality characteristics such as Nernstian- and rapid responses, and are minimally interfered with by the alkali and alkaline earth metal ions tested. The membranes were next applied to a flow-through device, enabling it to function as flow-injection analysis (FIA) detector. The performance of the sensor after undergoing the FIA optimization was further evaluated for its selectivity characteristics and lifetime. Results for the determination of chloroquine in synthetic samples that contained common tablet excipients such as glucose, starch, and cellulose, and other foreign species such as cations, citric acid or lactic acid were generally satisfactory. The sensor was also successfully used for the determination of the active ingredients in mock tablets, synthetic fluids and biological fluids. The sensor was applied for the determination of active ingredients and the dissolution profile of commercial tablets was also established.

    Marination directly affects the texture and flavor of meat and is important for product yield. This research investigated the effects of various marinating solutions on the physicochemical, microstructural and sensory properties of golek chicken, a popular dish in Malaysia, Indonesia and southern Thailand. Non-marinated meat (Tr1-control) was compared with five marinating treatments: distilled water (Tr2); sodium chloride (Tr3); a mixture of sodium chloride and sodium tripolyphosphate (STPP) (Tr4); a mixture of sodium chloride, STPP and citric acid (Tr5); and a mixture of sodium chloride, STPP and sodium bicarbonate (Tr6).     The impact of good assessment in medical education depends on how appropriately the tools measure the clinical performance and how reliable, valid and feasible they are to achieve the logical decision. The traditional methods of clinical examination using long and short cases and orals are often argued for its subjectivity, low reliability and inadequate context specificity. Oral test though comparatively more valid due to its face-to-face questions are considered less reliable for problems of unstandardized questions, inconsistent marking and lack of sufficient testing time. Development of an “objective structured clinical examination” (OSCE) was sought as a solution to these problems. But the fragmented representation of the context in a number of stations in OSCE makes it less authentic for an integrated judgment of performance. Yet another method to thought of, was the workplace-based assessment (WPBA) but it takes a snapshot as a predefined attribute of a more complex integrated assessment such as long case. However due to the problem of feasibility it is less likely that high stakes examination as summative assessment will ever be able to attain workplace-based assessment such as Mini-CEX and DOPS. A TOACS (task oriented assessment of clinical skills) format currently used in high stakes fellowship examination in one of the center and claimed to have more active role for examiners was analyzed and compared with OSCE. Author however, did not find a difference except the difference of acronyms of the two formats. Both have multiple, fragmented static or interactive stations of 5-10 minutes duration with or without examiners, patients or exhibits and a marking scheme comprising of checklist or global rating. In the backdrop of this context a new assessment format named the ‘task integrated objective structured clinical examination” or TIOSCE modified from OSCE is currently developed in School of Medical Sciences (SMS) at USM. However, it is a different version of OSCE in which though the principle concept is the same as that of an OSCE, the continuum of clinical skill’s work up of the same patient’s is followed through to test multiple short attributes of clinical competences. As it retains most of the favorable features, TIOSCE also addresses some of the odds features of OSCE.

    The effect of chemical reaction and variable viscosity on mixed convection heat and mass transfer for Hiemenz flow over a porous wedge plate was studied in the presence of heat radiation. The wall of the wedge was embedded in a uniform Darcian porous medium to allow for possible fluid wall suction or injection and had a power-law variation of both the wall temperature and concentration. The fluid was assumed to be viscous and incompressible. Numerical calculations were carried out for different values of dimensionless parameters and an analysis of the results obtained showed that the flow field was influenced appreciably by the buoyancy ratio between species, thermal diffusion and suction/injection at wall surface. The effects of these major parameters on the transport behaviours were investigated methodically and typical results illustrated to reveal the tendency of the solutions. Representative results are presented for the velocity, temperature, and concentration distributions. Comparisons with previously published works were performed and excellent agreement between the results were obtained. It is predicted that this research might prove to be useful for study of the movement of oil or gas and water through the reservoir of an oil or gas field, in the migration of underground water, in filtration, and water purification processes.

    Xiang Sha Yang Wei Wan (XSYWW) is a Chinese traditional medicine that is used for gastrointestinal disorders, specifically gastric ulcer in many countries of South-East Asia. The aim of the study was to evaluate the potential effects of XSYWW on ethanol-induced gastric ulcer in rats by means of histological Study. On a similar basis of treatment, ranitidine, a conventional medication was used as gold standard. Methods: Fifty five male Sprague-Dawley rats (250-300 gm) were divided into four groups. Group I (ethanol treated group) was the control group and gastric ulcers were induced by administering 100% ethanol (1 ml/200 g). Group II (Pre-treatment group) was divided into two subgroups; they were orally fed with 1.0 gm/kg and 2.0 gm/kg respectively of XSYWW solution. Thirty minutes later they were administered with absolute ethanol as in group I. Group III, was given an oral dose of 2gm/kg of XSYWW solution after one hour of ethanol administration. Group IV was given an oral dose of 200mg/kg ranitidine solution after one hour of ethanol administration. Five rats from groups I, III and IV were sacrificed on day 1, 2 and 3 while the animals of group II were sacrificed one hour after ethanol administration. Results: Histological study of the stomachs from ethanol treated rats showed multiple ulcers of various depths that reached the muscularis and the serosa. Conclusion: Pre or post-treated rats with XSYWW showed that XSYWW has protective effect against ethanol-induced gastric mucosal lesion. However, there was a faster and more complete healing process in the ranitidine treated group when compared to the XSYWW treated subjects.

    α-Mangostin was extracted from the pericarp of the Malaysian local Garcinia mangostana linn., The structure was characterised by Infrared red, UV-Visible and Nuclear Magnetic Resonance spectroscopic data. The fluorescence peak at 500nm in ethanol was not observed in PNIPAM microgel solution. The increase of colloidal size of the gel in the presence of α-mangostin was studied by Dynamic Light Scattering and Transmission Electron Microscope. The size of the particle also increases with increasing temperature up to 45⁰C after which it began to shrink. The TEM micrograph at 45°C showed a uniformly structured pattern of the gel occurs in the range of the lowest solution critical temperature.

    Gelatin is widely used in food and pharmaceutical products. However, the addition of gelatin especially in food products becomes a controversial issue among Muslims due to its animal origin. Thus, the present study was aimed to detect and differentiate the origin of gelatin added in processed foods using a combination method of sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and Principal Component Analysis (PCA). Porcine gelatin had exhibited 11 prominent polypeptides compared to bovine gelatin with 2 prominent polypeptides. Polypeptides of both gelatin sources at molecular weight ranged from 53 to 220 kDa can be used to differentiate between porcine and bovine gelatins using PCA. The efficiency in extracting gelatin from processed foods by different solutions was also evaluated. Extraction of gelatin in processed foods by cold acetone and deionised water had exhibited a similar polypeptide patterns, suggesting both solutions are suitable. The study indicated that approach of a simple gelatin extraction combined with SDS-PAGE and PCA, may provide robust information for gelatin species differentiation of processed foods.

    The accumulation of tear film proteins as well as microbes colonization onto worn contact lenses can be eliminated conventionally by mechanical rubbing during the cleaning process. Lens2® functions in rotation manner to loosen the deposits on the contact lens and has antimicrobial coating to keep lenses away from contamination. The objective of this study was to determine the efficiency of Lens2® to remove deposited protein and reduce microbial contamination compared to conventional method. Twenty-eight subjects each wore a pair of contact lens FDA Group 1 (Polymacon, SoftLens® 38, Bausch & Lomb) for one month and cleaned them using multipurpose solution (COMPLETE® MoisturePLUSTM, Advanced Medical Optics) separately using two different methods. The right lens was cleaned conventionally while the left lens were cleaned using the Lens2®. The control group of thirteen subjects each wore a pair of contact lens for the same period and cleaned both conventionally. These lenses and its cases were then analyzed for protein deposition using Bichinchoninic Acid Assay (BCA) Kit (Sigma, USA) in 96-well plate. Microbial contamination was determined by culturing the samples on nutrient agar for bacteria and fungi and non-nutrient agar for amoeba isolation. The mean of total protein on control lenses (17.014 ± 13.246 µg/mL) was not significantly different from those on the Lens2® (21.623 ± 19.127 µg/mL). There were also low growth numbers of amoeba in each group of samples. Interestingly, there were no growths of amoeba from all Lens2® samples collected. There was also low growth numbers of bacteria in each sample group whereby Lens2® had the lowest growth of bacteria. No growth of fungi was obtained from all samples. The automatic lens cleaner, Lens2® was found to be as efficient as the conventional cleaning method. However, the Lens2® has additional advantage because of its antimicrobial material and need shorter time in the cleaning process as well as easy and effective.

    Melaleuca cajuputi and Acacia auriculiformis trees are major sources of nectar and pollen for Apis dorsata and the colonies are a major source of honey to the rural poor, honey hunters. Honey is a supplementary income to many of these people (including school children) in the Marang district, Terengganu. In this study, Marang area with 270 square kilometers was chosen as pilot study area in Terengganu state for mapping M. cajuputi and A. auriculiformis as two dominant species in low land secondary forest in Terengganu state.To inventory and produce land use map of Melaleuca forest in Marang area, in this study SPOT-5 satellite image in multispectral mode with 10 meter resolution which is acquired in 2007 as optical satellite was utilized. Most images from optical satellites have some null data from ground because of clouds and shadow of clouds. To solve this problem, Hue, Saturation and value (HSV) and Principal Component Analysis (PCA) were used as fusion techniques to replace null data with microwave data which taken from Radarsat-1 image in C-band with 25 meter resolution image. Accordingly, fusion technique which was used in this research not only was a technique to improve information but also caused the accuracy increasing than land use map by just only SPOT-5 image. Also between two different fusion techniques, PCA shows the better result than HSV as two different fusion techniques.

    In this study polymer electrolytes composed of poly(methyl methacrylate) (PMMA) as a host polymer and ethylene carbonate (EC) as a plasticizer complexed with different lithium salts, i.e. lithium tetrafluoroborate (LiBF4) and lithium triflate (LiCF3SO3) were prepared by the solution casting technique. The conductivities of the films were characterized by impedance spectroscopy. At room temperature, the highest conductivities were 4.07 × 10–7S cm–1 and 3.40 × 10–5 S cm–1 achieved, respectively from the films containing 30 wt% LiBF4 in the PMMA-EC-LiBF4 system and 35 wt% LiCF3SO3 in the PMMA-EC-LiCF3SO3 system. The conductivity-temperature dependence of the films seemed to obey the Arrhenius equation in which the ion transport in these materials was thermally assisted. Scanning electron microscopy analysis showed that the surface of PMMA-EC-LiCF3SO3 film was smooth and homogeneous, hence lithium ions could traverse through the PMMA-EC-LiCF3SO3 film more easily compared to the PMMA-EC-LiBF4 film. X-Ray diffraction studies revealed that complexation had occurred and the complexes formed were amorphous.

    Introduction: The purpose of the present study was to investigate exploratory factor analysis of the
    Beck Anxiety Inventory (BAI) and reports its reliability and validity in Malaysia. Method: One thousand
    and ninety participants from four different samples (namely students, general population, medical and
    psychiatric patients) completed the Malay version of Beck Anxiety Inventory, as well as additional
    questionnaires such as Fear Questionnaire, Anxiety Sensitivity Index, Depression Anxiety Stress Scale, and Catastrophic Cognition Questionnaire. Results: Exploratory factor analysis revealed a three factor solution and accounted for 48.01% of the total variance. The three-factor structure appeared to be; subjective anxiety, autonomic, and neurophysiology. The Cronbach alpha coefficients (α) ranged from 0.66 to 0.89 with satisfactory overall alpha value (.91). Evidence was also found acceptable concurrent validity of the BAI-Malay (range between r=.22 to r=.67). Conclusion: This study shows that the BAIMalay is a reliable and valid instrument to measure symptoms of anxiety in the Malay population
    and can be used in research and clinical service in Malaysia. However, replication of study by using
    confirmatory factor analysis and application of the instrument among anxiety patients worth of further
    investigation.

    HCFCs, in addition to destroying the ozone layer, have been recognized as a contributing factor that increases global warming. It is widely used as working fluid in window air-conditioning system, where capillary tube serves as an expansion device. Literature reports have shown that no single refrigerant can solve the problem of ozone layer depletion and global warming. Refrigerant HC290/HC600a/HFC407C mixture, an eco-friendly refrigerant, has been recognized as an alternative to HCFC22. The objective of this study is to, for cost effectiveness, develop an empirical correlation to predict the refrigerant HC290/ HC600a/HFC407C mixture mass flow rate using statistical experimental design approach. A review of relevant literature shows that refrigerant’s mass flow rate depends on condensing temperature, degree of subcooling, inner diameter and length of capillary tube. The relationship between the mass flow rate and the four independent variables was established as an empirical mathematical correlation using central composite design (CCD), a response surface methodology (RSM). This empirical correlation was examined using analysis of variance (ANOVA) of 5% level of significance. The results of these analysis showed that the correlation fitted well with the experimental data yielding an average and standard deviation of 1.05% and 2.62%, respectively. The validity of the present correlation was further assessed by comparing it with published empirical correlation in literature and the result showed that the present correlation is consistent.

    Enzyme Linked Immunosorbent Assay (ELISA) is a standard assay that has been used widely to validate the presence of analyte in the solution. With the advancement of ELISA, different strategies have shown and became a suitable immunoassay for a wide range of analytes. Herein, we attempted to provide additional evidence with ELISA, to show its suitability for multi-analyte detection. To demonstrate, three clinically relevant targets have been chosen, which include 16kDa protein from Mycobacterium tuberculosis, human blood clotting Factor IXa and a tumour marker Squamous Cell Carcinoma antigen. Indeed, we adapted the routine steps from the conventional ELISA to validate the occurrence of analytes both in homogeneous and heterogeneous solutions. With the homogeneous and heterogeneous solutions, we could attain the sensitivity of 2, 8 and 1nM for the targets 16kDa protein, FIXa and SSC antigen, respectively. Further, the specific multi-analyte validations were evidenced with the similar sensitivities in the presence of human serum. ELISA assay in this study has proven its applicability for the genuine multiple target validation in the heterogeneous solution, can be followed for other target validations.

    A technique to isolate DNA from ghee was developed for the authentication of beef fat product. The method was based on pre-mixed ghee with phosphate buffer solution (PBS) prior to DNA extraction using Epicentre extraction method. The recovery of beef DNA was then analysed by polymerase chain reaction (PCR) using beef species-specific oligonucleotide primers which targeted the mitochondria DNA (mtDNA) of cytochrome b (cyt b) gene. The amplicon was 274 bp in size. The developed ghee extraction method offers a high yield of DNA providing 100 ng per μl and useful for validating beef fat product.

    This study aimed to develop and validate whole grain KAP questionnaire among children. A
    guided self-administered questionnaire was developed. In this cross sectional study, a total
    of 207 children aged 9 to 11 years in Kuala Lumpur, Malaysia were recruited. Knowledge
    domain was considered as optimal level of difficulty and able to discriminate performance of
    good and poor children. Construct validity was assessed using exploratory factor analysis with
    principle components method and varimax rotation. Four factor-solutions emerged for attitude
    domain whereas 3 factor-solutions were constructed for practice domain. Internal consistency
    was acceptable for knowledge (KR20=0.70), attitude (CA=0.72) and practice (CA=0.73).
